Abstract:Enzymatic biosensor (EBS) is the most promising substitute for traditional analytical methods on determination of organophosphorus pesticides (OPs). The merits of simple, inexpensive and miniature make it potential for environmental monitoring. Enzymatic determination of OPs is often based on cholinesterase (acetylcholineaterase and butyrylcholinesterase) and organophosphorus hydrolase. Various methods have been applied for enzyme immobilization in the construction of EBS for determination of OPs. Since the chemical and physical properties of the polymers may be tailored for particular needs, they gained importance as the substrate for enzyme immobilization. Application of single-use screen-printed enzymatic electrodes are most intensively developed procedures.

Abstract:A novel electrochemical microbial biosensor for the rapid monitoring of p-nitrophenyl-substituted organophosphates (OPs) compounds based on glass carbon electrode (GCE) modified with both ordered mesopore carbons (OMCs) and cell surface-expressed organophosphorus hydrolase (OPH) (OPH-bacteria/OMCs/GCE) was described in this paper. The genetically engineered Escherichia coli strain surface displayed mutant OPH (S5) with improved enzyme activity and favorable stability was constructed using a newly identified N-terminal of ice nucleation protein as an anchoring motif, which can be used directly without further time-consuming enzyme-extraction and purification, thereafter greatly improved the stability of the enzyme. Compared to OPH-bacteria modified GCE (OPH-bacteria/GCE), the OPH-bacteria/OMCs/GCE not only significantly enhanced the current response but also reduced the oxidation overpotential towards oxidizable p-nitrophenol (p-NP), which was the hydrolysate of p-nitrophenyl-substituted OPs. Under the optimized experimental conditions, at +0.84V (vs. SCE), the current–time curve was performed with varying OPs concentration. The current response was linear with paraoxon concentration within 0.05–25μM. Similarly, linear range of 0.05–25μM was found for parathion, and 0.08–30μM for methyl parathion. The low limits of detection were evaluated to be 9.0nM for paraoxon, 10nM for parathion and 15nM for methyl parathion (S/N=3). Thus, a highly specific, sensitive and rapid microbial biosensor was established, which holds great promise for on-site detection of trace p-nitrophenyl-substituted OPs.

Abstract:The excessive or illegal use of organophosphorus pesticides (OPs) in agriculture has led to potential residue problems, making it essential to develop an effective analytical method. As reliable and rapid analytical methods, biosensors open an avenue for OP detection. However, traditional biosensors mainly depend on bio-recognition elements, consequently limiting their practical application. To address these issues, nanozymes have been widely applied because of their low cost, excellent stability, and favorable catalytic activity. This review summarizes the related research progress of nanozyme-based biosensors for OP detection in the last six years (2018–2023). The structural classification, catalytic activity, and biosensing strategy are discussed. Emphatically, this paper introduces the practical applications of nanozyme-enabled biosensors in the direction of simple operation, low cost, and on-site detection, such as optical and electrochemical biosensors. Finally, the main challenges and future trends are prospected, which could provide theoretical references for nanozyme-based biosensors in OP detection.

Abstract:Organophosphorus pesticides (OPPs) are some of the most popular pesticides that, once used, inevitably affect the ecological chains and food chains of water, soil, and foodstuff sources, which can severely damage the ecosystem, and adversely affect human health. The shortcoming of enzymatic sensors, being the way in which enzymes are susceptible and vulnerable to environmental factors, could be compensated for, by using non-enzymatic nanosensors. Thus, it is important to develop effective non-enzymatic methods for the simple and fast analysis of the residues of OPPs. In order to develop new detection methods, a range of nanomaterials are being innovatively used to construct non-enzymatic electrochemical sensors endowed with specificity, selectivity and cost-effectiveness. Herein, we review the latest research progress in the field of nano-materials and related composites used for the electrochemical non-enzymatic sensing of OPPs. The design method, detection limit, and sensing performances of both non-enzymatic nanomaterials and enzymatic ones were highlighted and compared. The challenges and prospects in this field are found and discussed. Undoubtedly, non-enzymatic sensors for OPPs detection will facilitate the increasing demands of food and ecosystem monitoring and safeguarding.

Abstract:Organophosphorus pesticides were easily remained in fruits and vegetables which would be harm to the environmental safety and human health. In recent years, due to the simple preparation process, fast response and high sensitivity, the electrochemical (bio)sensors have received increasing attention, which were extensively used as the sensing platform for the detection of OPPs. The mechanisms for the determination of OPPs mainly included redox of nitrophenyl OPPs, enzyme hydrolysis and inhibition, immunosensor, aptasensor. Nowadays, the mainly explored electrode material has focused on metal-organic frameworks, metal and metal derivatives, carbon materials (carbon nanotube, graphene, g-C3N4), MXene, etc. These nanomaterials played important roles in the electrochemical (bio)sensors, which included: (a) as an electrocatalyst to promote the redox reaction, (b) as a carrier to load the enzyme or aptamer, (c) as a recognizer to identify the targets. The nanomaterials-based electrochemical (bio)sensor was a rapid, cost-effective methods to detect OPPs with high sensitivity. Besides, this review compared the analytical performance of different nanomaterials-based electrochemical (bio)sensors, and also identified the key challenges in the future. It would provide new ideas and insights to the further development and application of electrochemical (bio)sensors and the detection of pesticides in real samples.

Abstract:The enzymatic electrochemical biosensor has the advantages of simple operation, speed, and integration in the detection of organophosphorus pesticide (OPs) residues. It has the potential to become the best alternative to the traditional OP detection technology. This article introduces the OP identification principle of different enzymes, the OP detection mechanism of several common sensors, and the enzyme assembly method. In addition, the article discusses application of nanomaterials in sensor preparation and sensor performance parameters in the past decade. The related content of early sensors is outside the scope of this article.

Abstract:There has been a rise in pesticide use as a result of the growing industrialization of agriculture. Organophosphorus pesticides have been widely applied as agricultural and domestic pest control agents for nearly five decades, and they remain as health and environmental hazards in water supplies, vegetables, fruits, and processed foods causing serious foodborne illness. Thus, the rapid and reliable detection of these harmful organophosphorus toxins with excellent sensitivity and selectivity is of utmost importance. Aptasensors are biosensors based on aptamers, which exhibit exceptional recognition capability for a variety of targets. Aptasensors offer numerous advantages over conventional approaches, including increased sensitivity, selectivity, design flexibility, and cost-effectiveness. As a result, interest in developing aptasensors continues to expand. This paper discusses the historical and modern advancements of aptasensors through the use of nanotechnology to enhance the signal, resulting in high sensitivity and detection accuracy. More importantly, this review summarizes the principles and strategies underlying different organophosphorus aptasensors, including electrochemical, electrochemiluminescent, fluorescent, and colorimetric ones.

Abstract:Over recent decades, the widespread use of pesticides has been instrumental in securing a global food supply. However, this has led to significant environmental pollution concerns. Given the prevalent use of organophosphorus pesticides (OPs), there is an urgent need to develop sensors that can detect OPs with high selectivity and sensitivity. Electrochemical sensors offer an affordable and highly efficient solution for this challenge. Particularly, there have been remarkable advancements in creating metal–organic framework (MOF)-based electrochemical sensors. This review delves into the current advancements of MOF-based electrochemical sensors, emphasizing their use in detecting OPs. We investigate prevalent MOF modifications, detail methods for affixing MOF materials to various electrodes, and compare the electrochemical performance and stability of these methods. We also provide insights into the design principles of MOFs used in the electrochemical detection of OPs, breaking down the core detection mechanism. We hope our insights will guide researchers in developing sustainable, effective, and robust MOF-based portable electrochemical sensors.

Abstract:Ochratoxins are the major components of mycotoxin that exist extensively in plant origin food. In the ochratoxins group, ochratoxin A (OTA) as the most toxic compound, which possesses a high chemical and thermal stability in foodstuff and would do harm to human beings. Hence, it is imperative to develop a simple, rapid, sensitive, robust, and accurate way to determine OTA. Nanomaterial-based biosensors seem to be ideal analysis tools for identifying OTA on account of their tremendous merits, such as high sensitivity, excellent selectivity, simplicity of operation, and rapidity. This review focuses on the application and development of nanomaterial-enabled biosensors in OTA detection in the past years (2007–2018). Three dominant signal transduction mechanisms, i.e. optical, electrochemical, and piezoelectric, are discussed in details. Moreover, the major challenges and perspectives for future developments of OTA biosensors are also contained to provide an overview for the forthcoming research orientation.

Abstract:Pesticides pose a serious risk to public health, even in trace amounts. The sensitive, rapid, visual, and on-site detection of pesticides in a real sample remains a challenge. Herein, we report a visual and rapid organophosphorus pesticide (OP) sensing device with excellent sensitivity, ease of application, and specificity based on a photothermal effect-based peroxidase (POD) mimetic. The ultrathin MIL-101-NH 2 (Fe) (Fe-MOF) shell-coated Au nanobipyramide (NBP) not only possesses POD-like activity, but also exhibits excellent photothermal effect under near-infrared (NIR) light. The results reveal that the excellent photo-to-thermal conversion of the Au NBPs can rapidly heat the MIL-101-NH 2 (Fe) shell, which subsequently induces a markedly enhanced POD-like activity. Using paraoxon-ethyl as model OP, the combination of the OP inhibition effect on the biocatalytic activity of acetylcholinesterase with the inhibition effect of thiocholine on the colorimetric assays of the POD-mimicking Fe-MOF leads to a significant improved selectivity for OP sensing. In particular, the NIR light irradiation increased the OP detection sensitivity by 100 times. Importantly, based on this detection strategy, a visual test strip with high sensitivity was developed and enabled real-time detection through a simple color analysis software on a smartphone. The successful detection of OP residues in fruit peel using this test strip further confirmed its application potential in real sample analysis.

Abstract:The widespread application of organophosphorus pesticides (OPs) has inflicted significant damage on human well-being and food security. Hence, it is imperative to develop a friendly and accessible biosensor for the detection of OPs. Herein, apple polysaccharide (AP) stabilized palladium nanoparticles (AP-PdnNPs) with a particle size of 2.75-5.95 nm were prepared using AP as a stabilizer and reducing agent. AP-Pd30NPs exhibited good peroxidase-like activity and effectively decomposed H2O2 to ·OH, which catalyzed the 3,3',5,5'-tetramethylbenzidine system to become blue. The catalytic kinetics of AP-Pd30NPs conformed to the typical Michelis-Menten equation. Furthermore, OPs directly inhibited the peroxidase-like activity of AP-Pd30NPs. Thus, a highly effective colorimetric biosensor was developed for the detection of OPs. The detection range of the biosensor was 0.050 μg/L - 200 mg/L, and the limit of detection was extremely low to 0.010 μg/L. Compared with other nanomaterials, the detection platform based on AP-Pd30NPs can effectively detect organophosphorus pesticides without coupling natural enzymes；this method is more economical and practical. Therefore, this established method explores good perspective for the detection of OPs.
A Multi-Enzyme Cascade Response for the Colorimetric Recognition of Organophosphorus Pesticides Utilizing Core-Shell Pd@Pt Nanoparticles with High Peroxidase-like Activity

Abstract:In modern agricultural practices, organophosphorus pesticides or insecticides (OPs) are regularly used to restrain pests. Their limits are closely monitored since their residual hinders the capability of acetylcholinesterase (AChE) and brings out a threatening accumulation of the neurotransmitter acetylcholine (ACh), which affects human well-being. Therefore, spotting OPs in food and the environment is compulsory to prevent human health. Several techniques are available to identify OPs but encounter shortcomings like time-consuming, operating costs, and slow results achievement, which calls for further solutions. Herein, we present a rapid colorimetric sensor for quantifying OPs in foods using TMB as a substrate, a multi-enzyme cascade system, and the synergistic property of core-shell Palladinum@Platinum (Pd@Pt) nanoparticles. The multi-enzyme cascade response framework is a straightforward and effective strategy for OPs recognition and can resolve the previously mentioned concerns. Numerous OPs, including Carbofuran, Malathion, Parathion, Phoxim, Rojor, and Phosmet, were successfully quantified at different concentrations. The cascade method established using Pd@Pt had a simple and easy operation, a lower detection limit range of (1–2.5 ng/mL), and a short detection time of about 50 min. With an R2 value of over 0.93, OPs showed a linear range of 10–200 ng/mL, portraying its achievement in quantifying pesticide residue. Lastly, the approach was utilized in food samples and recovered more than 80% of the residual OPs.

Abstract:The residues of organophosphorus pesticides (OPs) are increasing environmental pollution and public health concerns. Thus, the development of simple, convenient and sensitive method for detection of OPs is crucial. Herein, a multifunctional Fe-based MOF with fluorescence, catalytic and adsorption, is synthesized by a simple one-pot hydrothermal method. The ratiometric fluorescence sensor for detection of OPs is constructed by using only one multifunctional sensing material. The NH2-MIL-101(Fe) is able catalyze the o-phenylenediamine (OPD) into 2,3-diaminophenazine (DAP) in the presence of H2O2. The generated DAP can significantly quench the intrinsic fluorescence of NH2-MIL-101(Fe) by the fluorescence resonance energy transfer (FRET) and internal filtration effect (IFE), while producing a new measurable fluorescence. Without immobilization or molecular imprinting, pyrophosphate ion (PPi) can inhibit the peroxidase-like activity of the NH2-MIL-101(Fe) by chelating with Fe3+/Fe2+ redox couple. Moreover, PPi can also be hydrolyzed by alkaline phosphatase (ALP), the presence of OPs inhibits the activity of ALP, resulting in the increase of extra PPi preservation and signal changes of ratiometric fluorescence, the interactions of ALP with different OPs are explored by molecular docking, the OPs (e.g., glyphosate) interact with crucial amino acid residues (Asp, Ser, Ala, Lys and Arg) are indicated. The proposed sensor exhibits excellent detection performance for OPs with the detection limit of 18.7 nM, which provides a promising strategy for detection of OPs.

Abstract:The reliable detection of organophosphorus pesticides (OPs) in complex matrices remains an enormous challenge due to inevitable interference of sample matrices and testing factors. To address this issue, we designed a nanozyme-coated mesoporous COF with guest molecule loading, and successfully used it to construct a dual-ratio dual-mode sensor through target-regulated signal generation. The multifunctional COF-based composite (MB/COF@MnO2, MCM) featured high loading of methylene blue (MB), oxidase-like MnO2 coatings as gatekeepers, and specific recognition of thiocholine (TCh). TCh, a regulator produced from acetylcholinesterase (AChE)-catalyzed hydrolysis of acetylthiocholine, could decompose MnO2 coatings, triggering the release of abundant MB and oxidation of few o-phenylenediamine (OPD). OPs, strong inhibitors of AChE, could restrain TCh production and MnO2 decomposition, thereby controlling the release of less MB and oxidation of more OPD. This regulation boosted the dual-ratio dual-mode assay of OPs by using the released MB and oxidized OPD in the solution as testing signals, measured by both fluorescent and electrochemical methods. Experimental results demonstrated the sensitive detection of dichlorvos with LODs of 0.083 and 0.026 ng/mL via the fluorescent/electrochemical mode, respectively. This study represented a creative endeavor to develop dual-ratio dual-mode sensors for OPs detection in complex samples, offering high sensitivity, excellent selectivity, and good reliability.

Abstract:This review examines recent advancements in electrochemical immunosensors for the detection of organophosphate pesticides, focusing on strategies to enhance sensitivity and selectivity. The widespread use of these pesticides has necessitated the development of rapid, accurate, and field-deployable detection methods. We discuss the fundamental principles of electrochemical immunosensors and explore innovative approaches to improve their performance. These include the utilization of nanomaterials such as metal nanoparticles, carbon nanotubes, and graphene for signal amplification; enzyme-based amplification strategies; and the design of three-dimensional electrode architectures. The integration of these sensors into microfluidic and lab-on-a-chip devices has enabled miniaturization and automation, while screen-printed and disposable electrodes have facilitated on-site testing. We analyze the challenges faced in real sample analysis, including matrix effects and the stability of biological recognition elements. Emerging trends such as the application of artificial intelligence for data interpretation and the development of aptamer-based sensors are highlighted. The review also considers the potential for commercialization and the hurdles that must be overcome for widespread adoption. Future research directions are identified, including the development of multi-analyte detection platforms and the integration of sensors with emerging technologies like the Internet of Things. This comprehensive overview provides insights into the current state of the field and outlines promising avenues for future development in organophosphate pesticide detection.

Abstract: Organophosphorous pesticides (OPs) are widely used in agriculture due to their high effectiveness and low toxicity for pest control and protecting crops and seeds [1, 2]. Their residuals in crops, livestock, and poultry products are clearly dangerous to human health. The related clinical signs include negative effects on the visual system, sensory function, cognitive function, and nervous system [3, 4]. Specifically, exposure to OPs has been shown to cause headache, dizziness, profuse sweating, blurred vision, nausea, vomiting, reduced heartbeat, diarrhea, loss of coordination, slow and weak breathing, fever, coma, and even death [5]. The toxicity of OPs mainly arises from their irreversible inhibition on acetylcholinesterase (AChE), which is essential for the central nervous system to function, often causing respiratory paralysis and death [6]. Because of the high toxicities, rapid detection of these agents in the environment, public places, or workplaces and the monitoring of individual exposures to OPs have become increasingly important for homeland security and health protection [7].
Advancements in nanomaterial-based aptasensors for the detection of emerging organic pollutants in environmental and biological samples

Abstract:The combination of nanomaterials (NMs) and aptamers into aptasensors enables highly specific and sensitive detection of diverse pollutants. The great potential of aptasensors is recognized for the detection of diverse emerging organic pollutants (EOPs) in different environmental and biological matrices. In addition to high sensitivity and selectivity, NM-based aptasensors have many other advantages such as portability, miniaturization, facile use, and affordability. This work showcases the recent advances achieved in the design and fabrication of NM-based aptasensors for monitoring EOPs (e.g., hormones, phenolic contaminants, pesticides, and pharmaceuticals). On the basis of their sensing mechanisms, the aptasensing systems can be classified as electrochemical, colorimetric, PEC, fluorescence, SERS, and ECL aptasensors. Special attention has been paid to the fabrication processes, analytical reliability, and sensing mechanisms of NM-based aptasensors. Further, the practical utility of aptasensing approaches has also been assessed based on their basic performance metrics (e.g., detection limits, sensing ranges, and response times).

Abstract:A multicolor enzyme-linked immunosorbent sensor was constructed to detect organophosphates pesticides (OPs) on base of TMB2+-mediated AuNRs etching. As the etching progresses, a rainbow-like color responses was generated. The naked eye limit of detection (LOD) of the sensing system is 0.25 ng mL-1. In particular, the longitudinal localized surface plasmon resonances (LSPR) peak shift of AuNRs is linearly correlated with the logarithm of OPs concentration in range of 104 to 5×106 pg mL-1 for parathion-methyl and fenitrothion, 5×104 to 107 pg mL-1 for fenthion, respectively. Compared with reported methods, this sensor displayed satisfactory linear ranges and detection limits. Average recoveries of this method ranged from 73.28% to 98.08% and relative standard deviations (RSDs) were below 8.0% as for food samples analysis without complicated pretreatments. Remarkably, the developed multicolor enzyme-linked immunosorbent sensor is capable of providing accurate and reliable assay for OPs in complex food samples.

Abstract:Traditional methods for detecting organophosphorus pesticides (OPs) are costly and complex, and also require expensive instruments. Although many colorimetric methods have been established to detect OPs, unfortunately, these methods are based on the inhibition of the activity of natural enzymes by pesticides, which is prone to false positive signals. In addition, a variety of OPs often co-exist in the actual detection, which will interfere with each other, resulting in unsatisfactory detection results. To avoid this occasion, we developed a nanozyme sensor array based on Ag 2 O nanospheres to achieve simultaneous discrimination and efficient detection of a variety of OPs without the participation of natural enzymes. The nanozyme sensor array was constructed for detecting six OPs simultaneously, which took advantage of Ag 2 O with oxidase-like activity to catalyze the TMB, OPD and ABTS reaction system. Additionally, the colorimetric results of the sensor array for OPs detection were recorded and analyzed using a smartphone with a camera. This work not only provides a convenient and high-efficiency method to identify OPs, but it also advances food safety and environmental monitoring.
