Abstract:RationaleThis study developed a method for the simultaneous determination of 12 element impurities in lomefloxacin hydrochloride ear drops using inductively coupled plasma-mass spectrometry (ICP-MS) in accordance with the requirements of the International Council for Harmonisation of Technical Requirements for Pharmaceuticals for Human Use Q3D. However, this product contains high concentrations of organic compounds such as glycerol and ethanol, and soluble carbon interference exists when it is determined by direct injection method. Therefore, we applied ICP-MS using oxygen reaction mode to reduce the effect of high organic matter on plasma body. The catalytic effect of trace metal ions on the photodegradation of lomefloxacin hydrochloride was also investigated. MethodsThe sample was dissolved, diluted, and directly determined by direct injection using oxygen reaction mode of ICP-MS with Sc-45, Ge-73, and In-115 as internal standards. Direct injection using oxygen reaction mode was equipped with argon-oxygen mixer. To investigate the catalytic effect of metal ions on photodegradation, a series of combinations of lomefloxacin hydrochloride mixed with different metal ions were designed. After being irradiated under ultraviolet light for 10 days, the photodegraded impurities in combinations were determined using high-performance liquid chromatography. ResultsThe linear relationships in the corresponding concentration range for 12 element impurities were good (r > 0.9993). Limits of detection were between 0.002 and 1.294 ng ml(-1). The average recoveries were 93.7%-108.2%, and the precision relative standard deviation was 0.04%-0.55% (n = 6). The experimental results showed that metal ions had a certain catalytic effect on photodegradation of lomefloxacin when EDTA, Mg2+, and Cu2+ coexisted. ConclusionsICP-MS using oxygen reaction mode is suitable for the direct determination of the sample rich in organic matter without digestion and can effectively eliminate the interference of high organic matter in this product. The established method was rapid, sensitive, and accurate and can be used for the quality control of elemental impurities in lomefloxacin hydrochloride ear drops.

Abstract:An inductively coupled plasma optical emission spectroscopy (ICP‐OES) method is developed and validated for quantifying trace elements from a high‐density polyethylene (HDPE) packaging material. The method found to be sensitive, accurate and reproducible for total of 16 metal elements and can be employed to assess quality of a packaging material. The safety and effectiveness of pharmaceutical medicines depend heavily on extractables and leachables. Patients may be adversely affected by contaminants that enter drug products through packing materials. As a result, regulatory organizations like the European Medicines Agency (EMA) and the US Food and Drug Administration (FDA) have set standards and regulations for evaluating and managing extractables and leachables. Characterizing the packing materials, carrying out extraction experiments and checking the extracted solutions for contaminants are all parts of this research. As part of the process for approving new drugs, regulatory agencies make use of these data. In present work, an inductively coupled plasma optical emission spectroscopy (ICP‐OES) method is developed and validated for quantifying trace elements from a high‐density polyethylene (HDPE) packaging material. The approach delineated in accordance with United States Pharmacopoeia (USP) and for the comprehensive analysis of 16 metallic elements including aluminium, arsenic, cadmium, chromium, cobalt, lead, mercury, nickel, titanium, vanadium, zinc, zirconium, barium, manganese, calcium and tin is applicable for evaluating the efficacy of packaging materials. The method detection limits (MDLs) complied as per USP 39‐NF 34 along with relative standard deviations (RSDs) obtained were ranging from 0.3% to 3.1% and recoveries of all elements spiked at two different trace levels in HDPE samples were within 97%–110%, providing sensitive, accurate and reproducible methodology.

A Case Study Demonstrating the Importance of Glass Vial Selection for Parenteral Pharmaceutical Products

Abstract:There are many factors to consider when selecting a container closure system for parenteral drug products to maintain their quality, efficacy, and safety. One aspect to consider for products stored in glass vials is the glass type. Although the glass vials in which most parenteral products are stored are classified as Type I by the United States Pharmacopoeia, Chapter <660>, not all glass vials that meet the glass performance characteristics of Type I are equivalent. In the study presented here, Type I glass vials from three suppliers of three different Type I glass vials (standard, delamination control, and coated) were investigated to evaluate the impact that each Type I glass vial had on the stability of a drug product under development. To evaluate this impact, a three-phase study was conducted in which the compatibility between the drug product and each vial was assessed through the measurement of the critical quality attributes of the product, extractable and leachable inorganic elements were analyzed for each vial, and finally a stability study under accelerated conditions was conducted for the drug product in the most compatible vial based on the aforementioned experiments. Results from this study demonstrated that there are, in fact, significant differences in glass vials regardless of their classification as Type I. In the study conducted here, delamination control Type I glass vials were found to be superior to both Standard Type I and coated Type I vials for the drug product under investigation.

Abstract:The dependence of "finite ground CPW on silicon" properties on light illumination is presented. It is shown that the losses of FGCPW transmission lines increase with the illumination. By removing the passivation layer from the slot areas, the losses of FGCPWs are reduced. The strong dependence of the substrate conductivity on illumination suggests that FGCPWs can be used as devices with available loss and constant insertion phase.
Stomatocytosis in a colony of captive squirrel monkeys (Saimiri sciureus).

Abstract:Chronic stomatocytosis, which increased in severity as a function of the time that animals were held in captivity, was observed in a small colony of squirrel monkeys. The stomatocytosis was associated with increasing hematocrits possibly due to a change in packing characteristics of the erythrocytes rather than any overt changes in the erythrocyte mass. Reticulocytes remained at control levels throughout development of the stomatocytosis. The most probable cause of this alteration in erythrocyte shape distribution was dietary, but the exact etiology was not determined.
Safety assessment of drug impurities for patient safety: A comprehensive review

Abstract:Drug impurities are undesirable but unavoidable chemicals which can occur throughout the drug life cycle. The safety implications of drug impurities can be significant given that they can impact safety, quality, and efficacy of drug products and that certain drug impurities are mutagenic, carcinogenic, or teratogenic. The characteristics of drug impurities could be specific to drug modalities (e.g., small molecules vs. biologics). The commonly encountered drug impurities include elemental impurity, residual solvent, organic impurity, host cell protein and DNA, residual viral vector, extractable and leachable, and particle. They can cause various adverse effects such as immunogenicity, infection, genotoxicity, and carcinogenicity upon significant exposure. Therefore, the effective control of these drug impurities is central for patient safety. Regulations and guidelines are available for drug developers to manage them. Their qualification is obtained based on authoritative qualification thresholds or safety assessment following the classic toxicological risk assessment. The current review focuses on the safety assessment science and methodology used for diverse types of drug impurities. Due to the different nature of diverse drug impurities, their safety assessment represents a significant challenge for drug developers.
Integrating ICP-MS and Chemometrics for Profiling Inorganic Elements in Lianhua Qingwen Capsules and Evaluating Health Risk

Abstract:Lianhua Qingwen capsule (LHQWC) is composed of 13 traditional Chinese herbs. In this study, we employed inductively coupled plasma mass spectrometry (ICP-MS) to quantify the concentrations of 26 inorganic elements (Na, Mg, Al, K, Ca, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Ga, As, Se, Rb, Sr, Ag, Cd, Cs, Ba, Hg, Tl, Pb, U) across 22 batches of LHQWC. These results were complemented with Chemometrics analysis and health risk assessment of selected hazardous elements. Chemometric analysis revealed significant quality variations among the 22 batches of LHQWC, identifying U, Cs, Tl, Rb, Mn, As, Mg, and Al as characteristic elements influencing formulation consistency. Moreover, the health risk assessment indicated that while levels of Cu, As, Cd, Pb, Cr, and Hg in LHQWC were within acceptable limits, concerns arose regarding vanadium levels in certain batches. These findings underscore the necessity of comprehensive elemental analysis and health risk assessment to ensure the safety and quality of LHQWC. Our study provides valuable insights for both quality evaluation and regulatory considerations in the production of LHQWC and similar herbal formulations.
Development and Qualification of Visible Particle Load Analysis Methods for Injectable Drug Product Primary Packaging Components

Abstract:In the biopharmaceutical industry, the observation of a single particle in a vial or syringe may result in entire lots of drug product recalls. U.S. Pharmacopeia <787> and <788> describe light obscuration methods and particle collection (membrane filtration) followed by light microscopy for particle counting of filled drug products. However, there are no corresponding pharmacopeial methods for determining the particle levels of unfilled primary packaging components or their packaging materials (tubs, nests, bags, etc.). This article describes a quantification method to accurately assess the number of particles in primary containers and corresponding closures. As a microscopic method, the size ranges can be set by the user and are limited only by the optical properties of the microscope and analysis time. Particle load is a critical quality attribute that has a direct impact on product safety. Applying a standardized method to compare the effect of process changes on particle load can aid manufacturers in refining their processes to minimize particulates. Described herein are the critical parameters to develop physical rinse methods and the subsequent qualification results to measure the visible particle load of nonsiliconized and siliconized primary packaging systems.

Abstract:Elemental impurities are substances present in drug products, excipients, or drug formulations. They may be formed by the presence of catalysts and environmental contaminants. Elemental impurities can be detected by a sophisticated method such as Inductively Coupled Plasma Mass Spectrometry (ICP–MS). ICP-MS is an advanced method to detect elemental impurities in drug substances. In this study Propafenone hydrochloride drug was used, Propafenone Hydrochloride is an antiarrhythmic medication belonging to class 1C used to prevent supraventricular and ventricular arrhythmias. The present study was aimed to develop and validate inductively coupled plasma mass spectroscopic (ICP–MS) method for detection of elemental contaminants, i.e., Class 1, Cd, Pb, As, Class 2A, Hg, Co, V, and Class 2B impurities such as Ni, T1, Se, Ag, Au, Pd, Ir, Os, Rh, Ru, and Pt. Total 17 elemental impurities were detected in Propafenone Hydrochloride and this method was employed for the regular sample analysis of 17 elemental impurities in Propafenone Hydrochloride for pharmaceutical use. The instrument conditions were set using RF power of 1550 W, auxiliary gas of 0.5 L/min, and nebulizer flow of 1.01 L/min nebulizer pump pressure was 0.10 rps, spray chamber temperature was 2°C, and mode used was He, He flow rate was 4.3 mL/min and the energy discrimination rate was 3.0 V. The technique is sensitive and may identify desirable elemental impurities within permissible regulatory limits when additional elements are present. The proposed ICP-MS approach has been found to be accurate, precise, linear, rugged, robust, and convenient for the quality control of the drug substance propafenone hydrochloride. The linearity results for each impurity were 0.9990. The methods were validated according to USP requirements and International Council for Harmonization ICH guidelines. The suggested approach is an excellent quality control tool for the concurrent quantitative assessment and detection of elemental contaminants at low levels in the drug substance propafenone hydrochloride.
Assessment of Patient Exposure to Leachables From Lyophilized Drug Formulations Following Reconstitution, Storage, and Administration via Polymeric Packaging/Delivery Systems

Abstract:It has been demonstrated that lyophilized drug formulations have an increased propensity to leach substances from the rubber stoppers comprising their primary packaging system when compared to aqueous liquid formulations stored in the same manner. Unfortunately, patient exposure to leachables originating in lyophilized drug products is not known. To that end, the goal of this study was to assess patient exposure to these leachables after reconstitution, storage, and administration of the lyophilized drug. To achieve this goal, several leachables present in 2 commercial lyophilized drug products were quantified after contact with polyvinyl chloride and non-polyvinyl chloride medication bags as well as an infusion set for durations of 15 min to 7 days at refrigerated and ambient temperature. The results obtained from this study showed that the bag's material of construction and the drugs formulation did not impact the mass of the leachables administered. Conversely, the mass of each leachable administered to the patient was reduced or eliminated as the contact duration with the intravenous bag and the temperature increased. However, for shorter contact durations, refrigerated storage, and higher molecular weight compounds, the patient would be exposed to a majority of the leachables originating from the vial.

Abstract:Starting materials, intermediates and by-products are often found as impurities in active pharmaceutical ingredients (APIs). Some of these known impurities are potential mutagens or carcinogens but can be difficult or impossible to eliminate completely from the synthetic scheme. Based on current regulatory guidances for genotoxic impurities, analytical methods should be developed to meet the required limit of 1.5 µg/day daily intake of each individual impurity for new drug substances. During the early clinical development stages the Pharma Task group proposed a staged TTC concept, where greater daily intake can be allowed. The allowable daily intake thus calculated would then provide a basis for estimating the quantitation limit (QL) required for the development of an analytical method for determining genotoxic impurity levels. The approach for choosing an analytical technique will depend on many factors including availability of the instrumentation and physical and chemical properties of the analyte. Although UV absorbance is normally the first choice detection technique for HPLC analysis, many genotoxic impurities lack a UV chromophore and consequently are not suitable for quantitation via UV detection. The examples described in this report show different approaches for quantitation on a case-by-case basis using various detection techniques such as UV, ELSD, CAD and MS. Although in one case the required alert level of 30 ppm was easily achieved using standard UV detection, other examples highlight the need for using alternatives such as an LC-MS/MS method. The latter methodology was necessary to achieve a required QL level of 57 ppm for one particular impurity. Validation of this method as per ICH guidelines with respect to specificity, linearity, accuracy and QL is also discussed.

Abstract:Elemental impurities determination in pharmaceuticals has become an emerging topic in the last years due to the revision of the Chapter 231 from the United States Pharmacopeia by three guidelines recently implemented: Chapters 232, 233 and 2232. Chapter 233 describes sample preparation and two analytical procedures, i.e. using inductively coupled plasma optical emission spectrometry (ICP-OES) or using inductively coupled plasma mass spectrometry (ICP-MS) for determination of elemental impurities. The main advantages and limitations of these instrumental methods have been well described, however, inherent in performing any analysis is the sample preparation step, which can be considered the most critical part of the analytical procedure. Generally, pharmaceutical samples present a wide range of different compositions, such as excipients, flavoring agents, stabilizers, and active ingredients. Consequently, these complex matrices make sample preparation a challenging step. In this sense, this review intends to discuss the trends, advantages and challenges of proposed approaches for preparation of pharmaceutical samples (active pharmaceutical ingredients, raw materials, pharmaceutical dosage forms, and dietary supplements) for determination of elemental impurities based on published works since 2000. Challenges and suitable alternatives observed during the determination of elemental impurities in pharmaceuticals using ICP-based methods are also discussed.

Abstract:Single-use systems in biopharmaceutical manufacturing can potentially release chemical constituents (leachables) into drug products. Prior to conducting toxicological risk assessments, it is crucial to establish the qualitative and quantitative methods for these leachables. In this study, we conducted a comprehensive screening and structure elucidation of 23 leachables (nonvolatile organic compounds, NVOCs) in two antibody drugs using multiple (self-built and public) databases and mass spectral simulation. We identified 7 compounds that have not been previously reported in medical or medicinal extractables and leachables. The confidence levels for identified compounds were classified based on analytical standards, literature references, and fragment assignments. Most of the identified leachables were found to be plasticizers, antioxidants, slip agents or polymer degradants. Polysorbate (namely Tween) is commonly used as an excipient for protein stabilization in biopharmaceutical formulations, but its ionization in liquid chromatography-electrospray ionization mass spectrometry can interfere with compound quantification. To address this, we employed a complexation-precipitation extraction method to reduce polysorbate content and quantify the analytes. The developed quantitative method for target NVOCs demonstrated high sensitivity (limit of quantification: 20 or 50 μg/L), accuracy (recoveries: 77.2 to 109.5%) and precision (RSD ≤ 8.2%). Overall, this established method will facilitate the evaluation of NVOC safety in drug products.

Telematics and sentinel health information system with general practitioners in Aquitaine, southwest France.

Abstract:A sentinel health information system using a telematics system to collect epidemiological data from a network of general practitioners (GPs) making up 5% of the GP population was set up in October 1986 in Aquitaine, France. In the first year, four topics were under surveillance. The GPs reported data for each case diagnosed by filling out a questionnaire displayed on their home terminal (Minitel), which transmitted the data by standard telephone lines to a central minicomputer located in the Bordeaux University Medical Centre. Regular feedback was provided to the participating GPs, public health officials and the Bordeaux Medical School. A detailed report of GP participation is presented, the reliability of our findings is discussed in terms of the motivational factors of participants, and the usefulness of the system is evaluated both for immediate teaching implications and for future research possibilities.
Computer simulation of the epitaxy of surfactant-templated inorganic nanomaterials on patterned surfaces.

Abstract:For a surfactant-inorganic-water system, we study systemically the epitaxy process of the surfactant-templated inorganic nanomaterial on a patterned surface with a lattice Monte Carlo method. It is found that by using ring-patterned substrate as a template, vertically oriented inorganic material may be formed through the nanometer-scale epitaxy. For all the cases studied in this work, a hemicylinder initially formed on the ring pattern behaves as nucleation sites for the following growth process. Different parameters, such as surfactant architecture, bulk surfactant concentration, fraction of inorganic component, and pattern size, are found to affect the epitaxial growth process of the inorganic nanomaterial. The change of surfactant architecture alters the structure of formed aggregates significantly, thus affecting the epitaxial growth. For the effects of surfactant concentration, it is found that there exists a critical value. If and only if the bulk surfactant concentration is higher than the critical value is the epitaxial growth of inorganic material nucleated from the patterned substrate possible. For the effects of the fraction of the inorganic component, simulation results indicate that there also exists a volume fraction above which the nanomaterial growth is dominated by macrophase separation but not templated by the substrate pattern. The geometry of the chemical modification of the surface also shows significant effects on the nanometer-scale epitaxy, depending on whether its sizes are commensurate with the morphology of the formed inorganic nanomaterials or not.
Application of ICH Guidelines for the Assessment and Control of Elemental Impurities in Parecoxib Sodium by Graphite-Digestion and ICP-MS

Abstract:Parecoxib sodium is a widely used parenteral cyclooxygenase 2 selective inhibitor to relieve acute postoperative pain following gynecologic laparotomy surgery. To ensure the quality of the drug, a detailed quality specification is indispensable. Nevertheless, it is unavoidable to introduce inorganic impurities during the drug preparation process and how to assess and control them matters. This study proposed an analytical procedure for the determination of elemental impurities (Cd, Pb, As, Hg, Co, V, Ni, Li, Sb, and Cu) in parecoxib sodium, where an easier and safer digestion protocol, graphite digestion, combined with an inductively coupled plasma-mass spectrometer (ICP-MS) was adopted when compared with microwave digestion. Moreover, the study also discussed whether should they be listed in specification to comply with ICH Q3D guidelines after test of process validation batches. Limit of quantitation (LOQ) of the above elemental impurities reached to 0.05, 0.125, 0.375, 0.075, 0.125, 0.25, 0.5, 6.25, 2.25 and 7.5 ppm, respectively, and recovery in accuracy item ranged from 90.2% to 129.9%, reflecting a sensitive and accurate method.

Abstract:Performant sample preparation is mandatory in any leachable study to clean and preconcentrate analytes within the sample to offer the best possible extraction recovery as well the best precision for any given substance. The aim consists in building a sample preparation of hospital pharmacy-prepared drug products such as long-term storage prefilled syringes, vials and IV bags for the screening of leachable compounds. The Quality Control Laboratory of the Pharmacy of the Lausanne University Hospital (Switzerland) has developed a time- and cost-effective, highly sensitive, robust, fast method using liquid chromatography coupled to a high-resolution mass spectrometer (LC-HRMS) for the analysis of 205 plastic additives. An innovative setup, based on postcolumn infusion (PCI) using 2% ammonium hydroxide in methanol was used to boost the signal intensity of the analytes in MS detection. A database for extractable and leachable trace assessment (DELTA) was built to assist in the screening process of 205 plastic packaging-related compounds. The development of the sample preparation was based on 33 plastic additive candidates in different hospital pharmacy compounding solutions, of which their extraction recovery rates as well as their relative standard deviation were taken into consideration. As a result, the developed DLLME was assigned with ultrasound assistance and triple extraction, which brought about extraction recovery rates between 67% and 92%, a good RSD <10%, and a preconcentration factor of 50X. Therefore, DLLME could be considered suitable for the semiquantitative screening of leachable additives in simple hospital pharmacy-prepared prefilled drug products.

In-silico Toxicity Assessment and Trace Level Quantification of Veratryl Chloride a Potential Genotoxic Impurity in Ivabradine Hydrochloride using LC-MS/MS

Abstract:A sensitive and selective liquid chromatography-mass spectrometry (LC-MS/MS) method using multiple reaction monitoring (MRM) mode was developed and validated for trace analysis of 4-(chloromethyl)-1, 2-dimethoxybenzene (Veratryl chloride) a potential genotoxic impurity in Ivabradine hydrochloride (IVB). Chromatographic separation was performed on a Poroshell 120EC C18 (50 × 3.0 mm, 2.7 μm) column using a mixture of 10 mM ammonium formate and acetonitrile in isocratic elution mode at a 0.25 mL/min flow rate. A simple pre-column derivatization with di-ethylamine was employed for the derivatization of the veratryl chloride. The developed LC-MS/MS method was linear and accurate in the 1.5–10.0 ppm concentration range with r2 ˃ 0.999 and percent recoveries greater than 90%. The developed method was precise with RSD (%) of not more than 4.5%. In-silico genotoxicity and carcinogenicity potential of veratryl chloride was assessed using ICH M7 principles found to be positive. The developed method can identify and quantify veratryl chloride in IVB, hence can be applied by quality control labs of pharmaceutical industries for trace quantification.
