Abstract:The dependence of "finite ground CPW on silicon" properties on light illumination is presented. It is shown that the losses of FGCPW transmission lines increase with the illumination. By removing the passivation layer from the slot areas, the losses of FGCPWs are reduced. The strong dependence of the substrate conductivity on illumination suggests that FGCPWs can be used as devices with available loss and constant insertion phase.

Safety assessment of drug impurities for patient safety: A comprehensive review

Abstract:Drug impurities are undesirable but unavoidable chemicals which can occur throughout the drug life cycle. The safety implications of drug impurities can be significant given that they can impact safety, quality, and efficacy of drug products and that certain drug impurities are mutagenic, carcinogenic, or teratogenic. The characteristics of drug impurities could be specific to drug modalities (e.g., small molecules vs. biologics). The commonly encountered drug impurities include elemental impurity, residual solvent, organic impurity, host cell protein and DNA, residual viral vector, extractable and leachable, and particle. They can cause various adverse effects such as immunogenicity, infection, genotoxicity, and carcinogenicity upon significant exposure. Therefore, the effective control of these drug impurities is central for patient safety. Regulations and guidelines are available for drug developers to manage them. Their qualification is obtained based on authoritative qualification thresholds or safety assessment following the classic toxicological risk assessment. The current review focuses on the safety assessment science and methodology used for diverse types of drug impurities. Due to the different nature of diverse drug impurities, their safety assessment represents a significant challenge for drug developers.
Development and Qualification of Visible Particle Load Analysis Methods for Injectable Drug Product Primary Packaging Components

Abstract:In the biopharmaceutical industry, the observation of a single particle in a vial or syringe may result in entire lots of drug product recalls. U.S. Pharmacopeia <787> and <788> describe light obscuration methods and particle collection (membrane filtration) followed by light microscopy for particle counting of filled drug products. However, there are no corresponding pharmacopeial methods for determining the particle levels of unfilled primary packaging components or their packaging materials (tubs, nests, bags, etc.). This article describes a quantification method to accurately assess the number of particles in primary containers and corresponding closures. As a microscopic method, the size ranges can be set by the user and are limited only by the optical properties of the microscope and analysis time. Particle load is a critical quality attribute that has a direct impact on product safety. Applying a standardized method to compare the effect of process changes on particle load can aid manufacturers in refining their processes to minimize particulates. Described herein are the critical parameters to develop physical rinse methods and the subsequent qualification results to measure the visible particle load of nonsiliconized and siliconized primary packaging systems.

Abstract:Capsule-based dry powder inhalers (cDPIs) are widely utilized in the delivery of pharmaceutical powders to the lungs. In these systems, the fundamental nature of the interactions between the drug/formulation powder, the capsules, the inhaler device, and the patient must be fully elucidated in order to develop robust manufacturing procedures and provide reproducible lung deposition of the drug payload. Though many commercially available DPIs utilize a capsule-based dose metering system, an in-depth analysis of the critical factors associated with the use of the capsule component has not yet been performed. This review is intended to provide information on critical factors to be considered for the application of a quality by design (QbD) approach for cDPI development. The quality target product profile (QTPP) defines the critical quality attributes (CQAs) which need to be understood to define the critical material attributes (CMA) and critical process parameters (CPP) for cDPI development as well as manufacturing and control.

Abstract:An inductively coupled plasma optical emission spectroscopy (ICP‐OES) method is developed and validated for quantifying trace elements from a high‐density polyethylene (HDPE) packaging material. The method found to be sensitive, accurate and reproducible for total of 16 metal elements and can be employed to assess quality of a packaging material. The safety and effectiveness of pharmaceutical medicines depend heavily on extractables and leachables. Patients may be adversely affected by contaminants that enter drug products through packing materials. As a result, regulatory organizations like the European Medicines Agency (EMA) and the US Food and Drug Administration (FDA) have set standards and regulations for evaluating and managing extractables and leachables. Characterizing the packing materials, carrying out extraction experiments and checking the extracted solutions for contaminants are all parts of this research. As part of the process for approving new drugs, regulatory agencies make use of these data. In present work, an inductively coupled plasma optical emission spectroscopy (ICP‐OES) method is developed and validated for quantifying trace elements from a high‐density polyethylene (HDPE) packaging material. The approach delineated in accordance with United States Pharmacopoeia (USP) and for the comprehensive analysis of 16 metallic elements including aluminium, arsenic, cadmium, chromium, cobalt, lead, mercury, nickel, titanium, vanadium, zinc, zirconium, barium, manganese, calcium and tin is applicable for evaluating the efficacy of packaging materials. The method detection limits (MDLs) complied as per USP 39‐NF 34 along with relative standard deviations (RSDs) obtained were ranging from 0.3% to 3.1% and recoveries of all elements spiked at two different trace levels in HDPE samples were within 97%–110%, providing sensitive, accurate and reproducible methodology.

Abstract:Sensitivity of drugs to one or more elements of the primary packaging is a serious concern for the pharmaceutical industry. Biologics in particular are highly sensitive, leading to a higher risk of incompatibility and stability test failure as worst-case scenario.This potential incompatibility-and the consequent formulation instability due to the interactions between the drug and the primary container surface-may have multiple causes: the intrinsic nature of the container surface, leachables coming from the materials used, substances coming from the production process, or silicone oil droplets or other particles.The Alba primary packaging platform was designed to have the same interface between the drug and the glass container surface on the different primary packaging containers in order to minimize the emergence of instabilities at later stages of formulation development. Alba containers are internally treated with an innovative cross-linked coating based on silicone oil lubricant, and the additional rubber components have been selected to minimize the possible differences between the container typologies.This paper shows in great detail the reduction of the inorganic extractables released and the comparability of the performances of the different containers obtained using Alba technology.The improvement has been demonstrated by stressing the containers with different extract solutions; Alba-coated containers show a strong reduction of inorganic extractables and of corrosion degree compared to spray-on siliconized and bulk products. Assessment of Patient Exposure to Leachables From Lyophilized Drug Formulations Following Reconstitution, Storage, and Administration via Polymeric Packaging/Delivery Systems

Abstract:It has been demonstrated that lyophilized drug formulations have an increased propensity to leach substances from the rubber stoppers comprising their primary packaging system when compared to aqueous liquid formulations stored in the same manner. Unfortunately, patient exposure to leachables originating in lyophilized drug products is not known. To that end, the goal of this study was to assess patient exposure to these leachables after reconstitution, storage, and administration of the lyophilized drug. To achieve this goal, several leachables present in 2 commercial lyophilized drug products were quantified after contact with polyvinyl chloride and non-polyvinyl chloride medication bags as well as an infusion set for durations of 15 min to 7 days at refrigerated and ambient temperature. The results obtained from this study showed that the bag's material of construction and the drugs formulation did not impact the mass of the leachables administered. Conversely, the mass of each leachable administered to the patient was reduced or eliminated as the contact duration with the intravenous bag and the temperature increased. However, for shorter contact durations, refrigerated storage, and higher molecular weight compounds, the patient would be exposed to a majority of the leachables originating from the vial.

Abstract:A strategy for the risk assessment of potentially genotoxic impurities is described that utilizes Quality by Design in an effort to furnish greater process and analytical understanding, ultimately leading to a determination of impurity criticality. By identifying the risks and parameters that most influence those risks, an enhancement of both product and process control is attained that mitigates the potential impact of these impurities. This approach calls for the use of toxicological testing where necessary, chemical fate arguments when possible, multivariate analyses to develop design space, and use of spiking data to support specifications. Strong analytical support, especially with the development of low-level detection methods, is critical. We believe that this strategy not only aids in the development of a robust API process but also delivers on the identification and subsequent mitigation of risks to a class of impurities that are of high interest in the field.

Lipid Peroxidation and Cancer: A Critical Reconsideration

Abstract:The author reviews the problem of the pattern of lipid peroxidation in cancer cells with special reference to a comparison between normal liver cells and hepatomas both transplanted and induced by diethylnitrosamine. It is stated that the loss of lipid peroxidation is proportional to the degree of dedifferentiation of hepatoma cells. During carcinogenesis, however, the loss is already evident at the stage of preneoplastic nodules. A common feature of all tumors, independently of the extent of the loss of peroxidation in basal conditions, is the lack of further stimulation by ADP/iron or by ascorbate/iron. As regards the reasons for the decline in lipid peroxidation, they are certainly not unique. An important cause is the low activity of the enzymes of the monooxygenase microsomal chain. Another very important one is the change in lipid composition of membranes, with a marked decrease in polyunsaturated fatty acids, which are the main substrate for lipid peroxidation. It has been shown that enrichment of membranes of hepatomas with arachidonic acid results in restoration of stimulation of peroxidation by ascorbate/iron, but not with ADP/iron. The last type of stimulation mostly reflects the behaviour of the monooxygenase chain, whereas ascorbate/ iron-induced stimulation does not require the presence of an efficient cytochrome P450-chain. Another cause for decreased lipid peroxidation in tumors is the increased rigidity of membranes, due to the large increase in cholesterol content: this prevents to some extent the influx of oxygen inside the membranes. Yet another cause is the presence of increased amounts of antioxidants in both cytosol and membranes. The main toxic product of lipid peroxidation, 4-hydroxynonenal, has been found to elicit several actions at extremely low concentrations. In fact, 4-hydroxynonenal stimulates Chemotaxis of polymorphonuclear leukocytes, stimulates plasma membrane adenylate cyclase, stimulates plasma membrane guanylate cyclase, and stimulates phospholipase C. The last three enzymes involve the action of G-proteins. The effect of the aldehyde is present at less than micromolar concentrations, which may occur inside the cells in certain conditions. Morever, at concentrations from 10–6 to 10–7 M, the aldehyde is able to block oncogene c-myc expression in the human erythroleukemic K562 cell line, which at the same time becomes able to express the gamma-globin gene. These facts are discussed with reference to a possible biological meaning of the loss of lipid peroxidation in tumors.

Abstract:The packaging concept has been adopted from plants’ seed as it is guarded in a closed shell till it germinates in a suitable season. Therefore, a type of natural packaging results in protection, preservation, and production. In the modern era, packaging aims to secure the product's safety until usage. Delivering a product to the customers requires a suitable container and closure with non-interaction with the product by any external influence. The primary container protection is strengthened by the secondary ones, resulting in safe storage and enhanced stability. The display of essential information on the label with elegance ensures the integrity of the product. The label displays the name, strength and quality, identification number, storage instructions, usage directions, the product's expiry date, and the manufacturer's name and address. This information provides compliance aid to the patients. According to the regulatory body’s guidelines, all forms of counterfeiting must be guarded against. Packaging processes and related equipment need validation, and quality control of the equivalent is to be followed. The article discusses the packaging components' role and various types (materials, measurement, etc.), technology, design, regulatory guidelines and tests applied, types of hazards, labelling procedures with guidelines, and quality assurance and control. This review paper highlights structural features of various parts of containers, strip packaging, blister packaging, bubble packaging, and various types of injection vials that were not addressed in a previous publication on the packaging system.Graphical abstract

Abstract:Elemental impurities determination in pharmaceuticals has become an emerging topic in the last years due to the revision of the Chapter 231 from the United States Pharmacopeia by three guidelines recently implemented: Chapters 232, 233 and 2232. Chapter 233 describes sample preparation and two analytical procedures, i.e. using inductively coupled plasma optical emission spectrometry (ICP-OES) or using inductively coupled plasma mass spectrometry (ICP-MS) for determination of elemental impurities. The main advantages and limitations of these instrumental methods have been well described, however, inherent in performing any analysis is the sample preparation step, which can be considered the most critical part of the analytical procedure. Generally, pharmaceutical samples present a wide range of different compositions, such as excipients, flavoring agents, stabilizers, and active ingredients. Consequently, these complex matrices make sample preparation a challenging step. In this sense, this review intends to discuss the trends, advantages and challenges of proposed approaches for preparation of pharmaceutical samples (active pharmaceutical ingredients, raw materials, pharmaceutical dosage forms, and dietary supplements) for determination of elemental impurities based on published works since 2000. Challenges and suitable alternatives observed during the determination of elemental impurities in pharmaceuticals using ICP-based methods are also discussed.

Thus it is necessary to check impurity level is present at acceptable amount. The present review gives an account of updated information about metal impurities and reviews the regulatory aspects for such metal impurities in drug substance/drug product. In addition the aim of this article is to discuss the currently used different analytical techniques for detection of metals from drug product like spectrophotometry, X – Ray florescence spectrometry, AAS, INAA, ICP – AES, ICP – MS, MP – AES, Laser Ablation – ICP – MS etc which is used for quality control of metal impurities in pharmaceuticals.

Abstract:Gamma-ray irradiation has been widely applied in large-scale industrial pharmaceutical packages sterilization however, high-energy rays may result in degradation of packaging materials comprising polymers and additives. These degradation products (DPs) can migrate into drugs, reducing their efficacy and safety. Therefore, a holistic study about structure elucidation and toxicity assessment of DPs is necessary. In this work, the effect of γ irradiation on rubber closures and additives was investigated by supercritical fluid extraction (SFE) coupled with UHPLC-QTOF MS and GC-MS. Comprehensive DPs were screened and identified by high-resolution mass spectrum and the potential degradation way was proposed. However, as there is no standard present for these DPs, Density-functional theory (DFT) was incorporated to verify degradation way and their structures by analyzing energy barriers and thermodynamic parameters. The initial step of the degradation mechanism was determined and structures and intensity difference of isomers were validated to be consistent experimentally and theoretically. Then based on toxicity prediction consistency by three (Q)SAR tools, the risk of DPs was evaluated and most DPs were negative for carcinogenicity and mutagenicity but positive for developmental toxicity. 7,9-di-tert-butyl-1-oxaspiro [4,5] deca-6,9-dien-2,8-dione was predicted to be carcinogenic in consensus which should be focused on in the further leachable study. Finally, risk assessment in the simulated lyophilized drug showed that exposure of all DPs was within the threshold. The present work addresses the structure elucidation of DPs, especially Irganox 1076, and helps to access a deeper insight into the safety evaluation of rubber closures with more confidence.

Abstract:Wind power is a growing sector of the United States electricity generation system. The integration of wind energy into the electrical grid presents capacity-related challenges to policy makers. The introduction of large-scale storage capabilities for wind power could lead to cost and reliability benefits if the interface locations are chosen wisely. The location of storage capability in the electrical grid relative to sources of demand and supply becomes a key research question. This paper describes the development of a conceptual framework using marginal pricing and associated modeling requirements necessary to analyze siting issues of storage capacity into the electricity grid.

Abstract:Although the secondary packing materials do not directly contact the finished drug products, compound migration may still happen between them. To ensure drug quality and safety, extractables and leachables of the packing materials should be analyzed. In this study, 2,6-di-tert-butyl-4-methylphenol (BHT) was first found in the labels for pharmaceutical packaging. For the identification of the compound, a strategy combining high performance liquid chromatography (HPLC), ultra-performance liquid chromatography-quadrupole time-of-flight mass (UPLC-Q-TOF-MS) and nuclear magnetic resonance (NMR) spectroscopy was utilized. Afterwards, a effective and sensitive HPLC method for quantification of BHT was developed and validated. Finally, a toxicological risk assessment of BHT was performed to ensure the safety of drugs.

Abstract:The objective of this chapter is to give guidance with degradation and impurity structure elucidation for pharmaceutical drug candidates. Stress testing knowledge is provided as guidance in developing and executing forced degradation experiments. Impurity and degradant structure elucidation is a collaborative effort involving the degradation chemist, analytical chemist, process chemist, and/or formulator, as well as the isolation chemist, mass spectrometry and NMR experts. Updates in this second addition include: (1) the use of quality by design (QbD) model as applied to stress testing practices, expanded oxidative experimental analysis, and expanded information on predictive programs. (2) The impact of supercritical fluid chromatography as a tool for impurity isolations, along with expanded information on process workflow. (3) Mass spectrometry: additional examples and a section on the use of accurate mass information in the role of structure elucidation. (4) Expansion on the use of NMR as applied to structure characterization and elucidation. (5) In‐depth case studies that illustrate collaborative efforts made between disciplines, as well as the impact that new technologies have on the structure elucidation process.

Abstract:The presence of impurities is the primary cause of safety related drug product recall. Clinical data generated during the drug discovery process may not be able to conclude the long-term safety of a drug. The best way to ensure drug safety is to control the associated impurities. It can be achieved by a clear understanding of the route and factors for impurity formation. Tight control on manufacturing, storage, and transport process can provide a beneficial effect on minimization of impurity generation. Control strategies employing quality by design and risk assessment-based approaches potentially enhance drug safety profile and, thereby, minimize product recalls. Prior understanding of key aspects of the safety profile of impurities, post-marketing surveillance criticalities, and establishment of constructive preventive strategies is the best way to minimize unwanted product recall due to associated safety issues. Effective preventive strategies should be designed and executed during the product manufacturing and supply chain process. The novelty of this article can be justified by the unavailability of any similar type of reports discussing the functional pathway of regulatory bodies aiming for effective control of drug impurities. This article contains novel, critical, and constructive prevention strategies to establish a robust safety profile of pharmaceutical products.
