Abstract:The culture of microalgae is important for the production and maintenance of bivalves. One of the major challenges is to maintain the reliability of microalgae forages over the long term. The aim of this work is to use Ultrafiltered (UF) seawater to cultivate them. Thus, cultures in a volume of 300 L of 2 species of microalgae Tetraselmis and T-isochrysis, were monitored in UF water (membrane pore size: 20 nm) and in sea water usually used on the Ifremer mollusk experimental platform of Bouin (France) (Prefiltration, 3 filtrations and 2 UV). The major result is the securing of microlagae cultures with the absence of parasites in all cultures supplied with ultrafiltered water, unlike analyses of the various control cultures. In the case of T-isochrysis, 3 cultures out of 4 resulted in higher microalgae concentrations, up to 30%, in ultrafiltered water thus bringing a benefit on the algal density. These conclusions and the ease of recovering water (linked to the reduction in treatment stages) allowed a transfer of technology. In fact the 300 L cultures hitherto carried out on the experimental platform are now produced in ultrafiltered water since early 2019.

Abstract:The release of inadequately treated urban wastewater is the main cause of environmental pollution of aquatic ecosystems. Among efficient and environmentally friendly technologies to improve the remediation process, those based on microalgae represent an attractive alternative due to the potential of microalgae to remove nitrogen (N) and phosphorus (P) from wastewaters. In this work, microalgae were isolated from the centrate stream of an urban wastewater treatment plant and a native Chlorella-like species was selected for studies on nutrient removal from centrate streams. Comparative experiments were set up using 100% centrate and BG11 synthetic medium, modified with the same N and P as the effluent. Since microalgal growth in 100% effluent was inhibited, cultivation of microalgae was performed by mixing tap-freshwater with centrate at increasing percentages (50%, 60%, 70%, and 80%). While algal biomass and nutrient removal was little affected by the differently diluted effluent, morpho-physiological parameters (FV/FM ratio, carotenoids, chloroplast ultrastructure) showed that cell stress increased with increasing amounts of centrate. However, the production of an algal biomass enriched in carotenoids and P, together with N and P abatement in the effluent, supports promising microalgae applications that combine centrate remediation with the production of compounds of biotechnological interest; for example, for organic agriculture.

Abstract:Due to the increasing production of wastewater from human activities, the use of algal consortia for phytoremediation has become well‐established over the past decade. Understanding how interspecific interactions and cultivation modes (monocultures vs. polyculture) influence algal growth and behaviour is a cutting‐edge topic in both fundamental and applied science. Ammonium‐rich growth media were used to challenge the monocultures of Auxenochlorella protothecoides, Chlamydomonas reinhardtii and Tetradesmus obliquus, as well as their polyculture; NO3− was also used as the sole nitrogen chemical form in control cultures. The study primarily compared the growth, carbon and nitrogen metabolisms, and protein content of the green microalgae monocultures to those of their consortium. Overall, the cultivation mode significantly affected all the measured parameters. Notably, at 50 mM NH4+, the assimilation rates of carbon and nitrogen were at least twice as high as those in the monoculture counterparts, and the protein content was three times more abundant.Additionally, the consortium's response to NH4+ toxicity was investigated by observing a linear relationship between the indicator of tolerance to NH4+ nutrition and the N isotopic signature. The study highlighted a high degree of acclimation through metabolic flexibility and diversity, as well as species abundance plasticity in the consortium, resulting in a functional resilience that would otherwise have been unattainable by the respective monocultures.

Abstract:This paper focuses on the evaluation of the biomass production of a novel native microalgal strain Chlorella sorokiniana using centrate from municipal wastewater as the sole source of nutrients and six different temperatures. The experiments were performed in semi-continuous cultures using Bold's Basal Medium and centrate separately, modifying the temperature (25–50 °C). The study aimed to elucidate whether a thermotolerant strain can be used for both the production of biomass and the removal of nutrients from wastewater. It was observed that C. sorokiniana biomass can be produced using centrate as the sole nutrient source. The highest biomass yield, for both culture media, was achieved at 35 °C showing maximum values for centrate (1.0 gbiomass·L−1, 0.20 gbiomass·L−1·day−1 dry weight), surpassing the productivity obtained when using Bold's Basal Medium (BBM) (0.60 gbiomass·L−1; 0.13 gbiomass·L−1·day−1 dry weight). Variable chlorophyll fluorescence confirmed that the excess of ammonium (601 mgNH4·L−1) did not inhibit the growth of C. sorokiniana, and thus, the biomass productivity was not affected. The maximum nutrient removals reached for BBM and centrate were 96% and 61.5% for nitrogen, 53.2% and 61% for phosphorus, and 93% and 0.18% for total carbon, respectively. Fatty acids accumulating in the microalgal biomass were mainly composed of arachidonic, oleic, and eicosatetraenoic acids. Consequently, it is demonstrated that microalgae native to desertic areas can be used to produce microalgal biomass using centrate from wastewater treatment plants as the exclusive nutrient source, reaching high productivities.

Abstract:The piggery digestate of high ammonia was mixed with the anoxic aerated effluent of high nitrate and phosphorus, to cultivate a microalgal-bacterial consortium for simultaneous pollution removal and resource recovery. The highest removal of total inorganic nitrogen was achieved at 324.77 mg/L in 40% piggery digestate mixed with 60% anoxic aerated effluent, along with the most microalgae biomass production. The crude protein and fatty acids of C14-C20 in microalgae cells were 21.80% and 69.78%, indicating that this mixing strategy could produce abundant microalgal biomass suitable for biofuel generation and animal feed. High-throughput sequencing showed that microbial diversity increased and Paenibacillus, Thiopseudomonas and Pseudomonas were the dominant species promoting microalgal growth. Overall, these results provided a new insight of mixing two types of wastewaters for cultivating microalgal-bacterial consortia, to remove contamination and recover nutrients simultaneously.
Optimal influent N-to-P ratio for stable microalgal cultivation in water treatment and nutrient recovery

Abstract:Species specific nitrogen-to-phosphorus molar ratio (NPR) has been suggested for green microalgae. Algae can store nitrogen and phosphorus, suggesting that the optimum feed concentration dynamically changes as function of the nutrient storage. We assessed the effect of varying influent NPR on microalgal cultivation in terms of microbial community stability, effluent quality and biokinetics. Mixed green microalgae (Chlorella sorokiniana and Scenedesmus sp.) and a monoculture of Chlorella sp. were cultivated in continuous laboratory-scale reactors treating used water. An innovative image analysis tool, developed in this study, was used to track microbial community changes. Diatoms proliferated as influent NPR decreased, and were outcompeted once cultivation conditions were restored to the optimal NPR range. Low NPR operation resulted in decrease in phosphorus removal, biomass concentration and effluent nitrogen concentration. ASM-A kinetic model simulation results agreed well with operational data in the absence of diatoms. The failure to predict operational data in the presence of diatoms suggest differences in microbial activity that can significantly influence nutrient recovery in photobioreactors (PBR). No contamination occurred during Chlorella sp. monoculture cultivation with varying NPRs. Low NPR operation resulted in decrease in biomass concentration, effluent nitrogen concentration and nitrogen quota. The ASM-A model was calibrated for the monoculture and the simulations could predict the experimental data in continuous operation using a single parameter subset, suggesting stable biokinetics under the different NPR conditions. Results show that controlling the influent NPR is effective to maintain the algal community composition in PBR, thereby ensuring effective nutrients uptake.

Abstract:The goal of this research was to assess the efficiency of the liquid digestate treatment conducted with algal, environmental isolates illuminated entirely with sunlight. The photobioreactor was exposed to natural conditions and evaluated based on the reduction of chemical oxygen demand (COD), nitrogen compounds, and soluble phosphates. Microalgal and bacterial communities growing during the treatment process were studied. A high removal rate of soluble COD (= 91%) and nutrients (= 86%) was achieved. The average concentrations of nitrogen, phosphates, and COD in the reactor effluent were 95 mgN/L, 49 mg/L, and 735 mg O 2 /L, respectively. The overall algae-bacteria biomass productivity of 22 mg/L/d, calculated on the total suspended solids (TSS) basis, was recorded. The microbial analysis revealed the dominance of Tetradesmus obliquus followed by Microglena sp. in the first 14 weeks of the experiment. At the end of the experimental run, Chlorella sorokiniana cells appeared as a result of illumination intensity changes. The dominating bacteria belonged to Firmicutes (26.31%), Patescibacteria (17.38%), and Actinobacteriota (14.86%) and could have been responsible for the transformation of nitrogen and oxidation of organic contaminants. The research demonstrated that natural sunlight can successfully be used for efficient liquid digestate treatment with the algae-bacterial community.

Abstract:In response to the rapid expansion of the worldwide population, there is an urgent need for the development of new, more environmentally-conscious, food sources. In this context, microalgae, such as Euglena , are of interest due to their capacity for large-scale cultivation and flexible bioproduct output. Media recycling techniques have been proposed as waste- and cost-reducing measures for large-scale algal cultivation. Culture dynamics and phytohormone production during novel fermentation conditions can enhance growth and biomass production; yet, little is known about the impacts on growth regulating phytohormones such as cytokinins (CKs) and abscisic acid (ABA) under these conditions. Using Euglena gracilis, an organic culture medium was recycled at 25, 50 and 75% inclusion rates through multiple growth cycles to evaluate impacts on biomass production and phytohormone profiles. A 25% recycled inclusion rate resulted in biomass accumulation similar to the control over one growth cycle. Subsequent recycled rates and growth cycling reduced E. gracilis biomass accumulation relative to the control, even with cell counts continuing to increase throughout, highlighting that the recycled medium inclusion and the number of repeated culture cycles impacts the biomass output of E. gracilis . Phytohormone analysis indicated that E. gracilis synthesized CKs via the tRNA-degradation pathway with increased hormone levels corresponding to higher recycle rates; ABA increased in the supernatant at higher recycle rates but was not detected in any pellet fractions. This research expands upon the roles of CKs and ABA as signaling molecules and adds guidance for building a suite of conditions specific for industrial product development.

Abstract:To improve nutrients removal from real centrate wastewater and enhance the microalgae biomass production, cultivation of Chlorella vulgaris in lab and a pilot-scale photobioreactor with waste glycerol was studied. The results showed the optimal concentration of the crude glycerol was 1.0 g L-1 with the maximum biomass productivity of 460 mg L-1 d(-1) TVS, the maximum lipid content of 27%, the nutrient removal efficiency of all above 86%, due to more balanced C/N ratio. The synergistic relationship between the wastewater-borne bacteria and the microalgae had significant good influence on nutrient removal. In pilot-scale wastewater-based algae cultivation, with 1 g L-1 waste glycerol addition, the average biomass production of 16.7 g m(-2) d(-1), lipid content of 23.6%, and the removal of 2.4 g m(-2) d(-1) NH4+-N, 2.7 g m(-2) d(-1) total nitrogen, 3.0 g m(-2) d(-1) total phosphorous, and 103.0 g m(-2) d(-1) of COD were attained for 34 days semi-continuous mode.

Abstract:Natural resources are becoming increasingly scarce, and the need to control their consumption and recycle their use is growing. Water is one of the essential resources for human survival. Therefore, there has been an increasing interest in ways to save, recycle and treat water supplies. Aquaculture is one of the most polluting activities as it produces a significant wastewater volume, which needs proper treatment before being discharged into the environment or recycled. Microalgae are a potential solution for wastewater treatment. Due to their numerous advantages, the use of microalgal biomass is being studied, and, at present, there is already a market and room for profit in the sale of microalgal components in various forms, such as animal and human supplements. From a biorefinery point of view, it is important to take advantage of all the qualities and benefits that microalgae have by combining their great capacity to treat wastewater and exploit the produced biomass, analysing its composition for subsequent valorisation, for example. In this study, Chlorella vulgaris was used to treat aquaculture wastewater from a trout farm aquaculture facility, and the treatment efficiency was evaluated. To valorise the resulting biomass, its composition was also assessed. C. vulgaris successfully grew in the effluent with growth rates of 0.260 ± 0.014 d−1 and with average productivity of 32.9 ± 1.6 mg L−1 d−1. The achieved removal efficiencies were 93.5 ± 2.1% for total nitrogen, 98.0 ± 0.1% for nitrate-nitrogen and 92.7 ± 0.1% for phosphate-phosphorus. Concerning biomass composition, the lipids (15.82 ± 0.15%), carbohydrates (48.64 ± 0.83%), and pigment contents (0.99 ± 0.04% for chlorophyll a + b and 0.21 ± 0.04% for carotenoids) were similar to the values of similar studies. However, the protein content obtained (17.93 ± 1.21%) was lower than the ones mentioned in the literature.

Abstract:The main purpose of this study is to investigate the effects of operative parameters and bioprocess strategies on the photo-autotrophic cultivation of the microalgae Scenedesmus almeriensis for lutein production. S. almeriensis was cultivated in a vertical bubble column photobioreactor (VBC-PBR) in batch mode and the bioactive compounds were extracted by accelerated solvent extraction with ethanol at 67°C and 10 MPa. The cultivation with a volume fraction of CO2 in the range 0–3.0%v/v showed that the highest biomass and lutein concentrations – 3.7 g/L and 5.71 mg/g, respectively – were measured at the highest CO2 concentration and using fresh growth medium. Recycling the cultivation medium from harvested microalgae resulted in decreased biomass and lutein content. The nutrient chemical composition analysis showed the highest consumption rates for nitrogen and phosphorus, with values higher than 80%, while sulfate and chloride were less consumed.

Abstract:Four microalgae species were evaluated for their bioremediation capacity of anaerobic digestion effluent (ADE) rich in ammonium nitrogen, derived from a biogas plant. Chlorella vulgaris, Chlorella sorokiniana, Desmodesmus communis and Stichococcus sp. were examined for their nutrient assimilation efficiency, biomass production and composition through their cultivation in 3.7% v/v ADE; their performance was compared with standard cultivation media which consisted in different nitrogen sources, i.e., BG-11NO3 and BG-11ΝH4 where N-NO3 was replaced by N-NH4. The results justified ammonium as the most preferable source of nitrogen for microalgae growth. Although Stichococcus sp. outperformed the other 3 species in N-NH4 removal efficiency both in BG-11NH4 and in 3.7% ADE (reaching up to 90.79% and 69.69% respectively), it exhibited a moderate biomass production when it was cultivated in diluted ADE corresponding to 0.59 g/L, compared to 0.89 g/L recorded by C. vulgaris and 0.7 g/L by C. sorokiniana and D. communis. Phosphorus contained in the effluent and in the control media was successfully consumed by all of the species, although its removal rate was found to be affected by the type of nitrogen source used and the particular microalgae species. The use of ADE as cultivation medium resulted in a significant increase in carbohydrates content in all investigated species.

Abstract:Algae were able to grow healthy on bacteria-containing centrate wastewater in a pilot-scale bioreactor. The batch experiment indicated that the co-cultivation of algae and wastewater-borne bacteria improved the removal efficiencies of chemical oxygen demand and total phosphorus in centrate wastewater to 93.01% and 98.78%, respectively. A strain of beneficial aerobic bacteria, Acinetobacter sp., was isolated and its biochemical characteristics were explored. Synergistic cooperation was observed in the growth of algae and Acinetobacter sp. Removal efficiencies of some nutrients were improved significantly by the co-cultivation of algae and Acinetobacter sp. After treatment, residual nutrients in centrate wastewater reached the permissible discharge limit. The cooperation between algae and Acinetobacter sp. was in part attributed to the exchange of carbon dioxide and oxygen between the algae and bacteria. This synergetic relationship between algae and Acinetobacter sp. provided a promising way to treat the wastewater by improving the nutrients removal and biomass production.
Bioprospecting of microalgae for integrated biomass production and phytoremediation of unsterilized wastewater and anaerobic digestion centrate

Abstract:Eighteen microalgae, including two local isolates, were evaluated for their ability to grow and remove nutrients from unsterilized primary or secondary wastewater effluents as well as wastewater supplemented with nutrient-rich anaerobic digester centrate (ADC). Most of the tested species except several phylogenetically clustered Chlorella sorokiniana including local isolates and Scenedesmus strains were unable to grow efficiently. This may reflect the presence of certain genetic traits important for robust growth in the unsterilized wastewater. The maximum algal-specific growth rates and biomass density obtained in these bacterial-contaminated cultures were in the range of 0.8–1 day−1 and 250–350 mg L−1, respectively. ADC supplementation was especially helpful to biologically treated secondary effluent with its lower initial macronutrient and micronutrient content. As a result of algal growth, total nitrogen and orthophosphate levels were reduced by as much as 90 and 70 %, respectively. Biological assimilation was estimated to be the main mechanism of nitrogen removal in primary and secondary effluents with ammonia volatilization and bacterial nitrification-denitrification contributing for cultures supplemented with ADC. Assimilation by algae served as the principal mechanism of orthophosphate remediation in secondary wastewater cultures, while chemical precipitation appeared also to be important for orthophosphate removal in primary wastewater. Overall, cultivation of microalgae in primary and primary + 5 % ADC may be more favorable from an economical and sustainability perspective due to elimination of the costly and energy-intensive biological treatment step. These findings demonstrate that unsterilized wastewater and ADC can serve as critical nutrient sources for biomass generation and that robust microalgae can be potent players in wastewater phytoremediation.

Abstract:Microalgae-based wastewater treatment is a promising technology efficient for nutrient recycling and biomass production. Studies continuously optimize processes to reduce costs and increase productivity. However, changes in the operational conditions affect not only biomass productivity but the dynamics of the overall microbial community. This study characterizes a microalgae culture from an 80 m2 pilot-scale raceway reactor fed with untreated urban wastewater. Operational conditions such as pH, dissolved oxygen control strategies (On-off, PI, Event-based, no control), and culture height were varied to assess microbial population changes. Results demonstrate that increased culture height significantly promotes higher microalgal and bacterial diversity. pH, dissolved oxygen and culture height highly affects nitrifying bacteria activity and nitrogen accumulation. Furthermore, the system exhibited high disinfection capability with average Logarithmic Reduction Values (LRV) of 3.36 for E. coli and 2.57 for Clostridium perfringens. Finally, the fungi species detected included Chytridiomycota and Ascomycota, while purple photosynthetic bacteria were also found in significant abundance within the medium.
The Microbiome of Bioreactors Containing Mass-Cultivated Marine Diatoms for Industrial Carbon Capture and Utilization

Abstract:Marine microalgae are a promising innovation platform for carbon capture and utilization (CCU) biotechnologies to mitigate industrial greenhouse gas emissions. However, industrial-scale cultivation of algal mono-cultures is challenging and often unscalable. Non-axenic microalgae in large semi-open photobioreactors lead to the co-cultivation of diverse microbial communities. There is limited knowledge about the “bioreactor ecology” involving microalgae interacting with the microbiome and its subsequent impact on process stability and productivity. In this study, we describe the semi-continuous industrial mass cultivation of the cold-adapted marine diatom, UiT201, by investigating the prokaryotic and microeukaryotic (phytoplankton and heterotrophic protist) communities. Data were collected in two consecutive time series experiments, representing the initiation and operation of an preindustrial scale CCU photobioreactor (300,000 liters). The first experiment experienced a culture “crash” of the focal strain after 39 days, while the second culture remained stable and “healthy” for 60 days. The results highlight that this mass cultivation system represents a unique industrial marine microbial ecosystem. The succession of the prokaryotic community was primarily driven by species replacement, indicating turnover due to selective bioreactor conditions and/or biological interactions. Nonetheless, the bioreactor consistently harbors a recurring and abundant core microbiome, suggesting that the closely associated bacterial community is influenced by microalgae-specific properties and can endure a dynamic and variable environment. The observed culture collapse of coincided with changes in the core microbiome structure and different environmental growth conditions compared to the stable and “healthy” experiment. These findings imply that cohabiting microbial taxa within industrial microalgae cultivation likely play a critical role in stabilizing the conversion of industrial CO into marine biomass, and changes in community structure serve as an indicator of process stability.

Abstract:Nitrogen and phosphorus concentration in the effluent of a wastewater treatment plant can vary significantly, which could affect the growth kinetic and chemical composition of microalgae when cultivated in this medium. The aim of this work was to study the rate of growth, nutrient removal and carbon dioxide biofixation as well as biomass composition of Scenedesmus obliquus (S. obliquus) when it is cultivated in wastewater at different nitrogen and phosphorus ratio, from 1:1 to 35:1. A more homogeneous method for calculating productivities in batch reactors was proposed. The proper N:P ratio for achieving optimum batch biomass productivity ranged between 9 and 13 (263 and 322 mg L(-1) d(-1) respectively). This was also the ratio range for achieving a total N and P removal. Above and below this range (9-13) the maximum biomass concentration changed, instead of the specific growth rate.The maximum carbon dioxide biofixation rate was achieved at N:P ratio between 13 and 22 (553 and 557 mg CO2 L(-1) d(-1) respectively). Lipid and crude protein content, both depend on the aging culture, reaching the maximum lipid content (34%) at the lowest N:P (1:1) and the maximum crude protein content (34.2%) at the highest N:P (35:1).
