Abstract:[Objective] The soil microbial communities and N2O emissions were compared between soil samples from a vegetable greenhouse and outside farmland. [Methods] Denaturing gradient gel electrophoresis (DGGE) and quantitative PCR were used to investigate the difference of soil microbial 836 微生物学通报 Microbiol. China 2015, Vol.42, No.5 http://journals.im.ac.cn/wswxtbcn communities and functional gene abundance, respectively. A robotized incubation system was employed to analyze the gaseous products of denitrification and calculate the ratio of N2O/(N2+N2O+NO). [Results] Significant differences were observed in soil bacterial communities between the two soils. The total number of bacteria in the greenhouse soil was higher than that of farmland. However, relative abundance of functional genes (nirS and nosZ) of the two types of soil was no significant difference. There were lower N2O accumulation and N2O/(N2+N2O+NO) ratio in the farmland soil compared to the greenhouse soil. In addition, the patterns of gas emissions in the two soils were significantly different in the early stage of anaerobic incubation. DNRA might lead to the increase of ammonium nitrogen content after the anaerobic incubation. [Conclusion] The higher strength of fertilization in greenhouse soil resulted in significant changes in soil bacterial communities and higher active soil microbial mass, N2O accumulation and N2O/(N2+N2O+NO) ratio. The soils from a greenhouse and a farmland, of which the greenhouse soil came from, have distinct nitrate respiration property. DNRA may be responsible for certain amounts of N2O emissions in both soils.

Warmer and Drier Conditions Alter the Nitrifier and Denitrifier Communities and Reduce N2O Emissions in Fertilized Vegetable Soils

Abstract:Nitrous oxide (N2O) is a potent greenhouse gas and is mainly produced from agricultural soils especially vegetable soils with large N fertilizer input. How future projected climate change may impact on the N2O emissions and the related key (de)nitrifier communities in such ecosystem is poorly understood. The aim of this field study was to determine the interactive effects of a simulated warmer and drier climate on (de) nitrifier communities and N2O emissions in a vegetable soil. A warmer (+3.3 degrees C) and drier climate (-14.4% soil moisture content) was created with greenhouses with or without urea N fertilizer application. The variation of microbial population abundance and community structure of Ammonia-oxidizing archaea (AOA), bacteria (AOB) and denitrifiers (nirK/S, nosZ) were determined using Real time-PCR and sequencing. The results showed a strong interactive effect of simulated climate change with N fertilizer applications, whereby the impacts of warmer and drier conditions on the microbial communities and N2O emissions were more evident when N fertilizer was applied. The simulated warmer and drier conditions in the greenhouses significantly decreased N2O emissions largely due to the drier soil conditions. The abundance and community structure of AOB showed more rapid responses than AOA under the simulated climate conditions when N fertilizer was applied. Changes of AOB community structure were significantly correlated with soil moisture content and NH4+-N concentration. The simulated climate change did not affect the nirS gene abundance, but significantly increased nirK gene abundance, and significantly decreased nosZ gene abundance with urea application. N2O emissions were positively correlated with the bacterial amoA abundance and with the ratio of nirK/nosZ gene abundance. Therefore, bacterial amoA, nirK- and nosZ-type denitrifiers are the dominant microbial communities which were affected by the simulated climate conditions and are thus critically important for N cycling in vegetable soils under a changing climate. Abstract:A greenhouse experiment was conducted with a specially designed apparatus consisting of an upper and lower chamber where the treatment with rice was carried out (treatment 1). The apparatus also had a single chamber where treatment 2, without rice plants, was carried out. The scope of this study was to elucidate the influence of rice plant growth on gaseous N losses as N 2 and N 2 O produced by nitrification-denitrification in a flooded soil fertilized with (NH 4 ) 2 SO 4 (with 56.50 atom% 15 N). Gas samples were withdrawn weekly and analyzed for (N 2 + N 2 O)- 15 N losses by mass spectrometer and for N 2 O by gas chromatograph. The gaseous (N 2 + N 2 O)- 15 N losses of the treatment with rice plants were significantly ( P =0.01) higher than those of the treatment without rice plants, as were the amounts of N 2 O emitted. Rice plants facilitate the efflux of N 2 and N 2 O from soil to atmosphere, as about half of the total gaseous 15 N loss as N 2 and N 2 O was found in the upper chamber. The proportion of N 2 O- 15 N to (N 2 + N 2 O)- 15 N in the upper chamber was 10.56%, much higher than that of the lower chamber in treatment 1 and the headspace of treatment 2.

Abstract:Denitrification is vital to nitrogen removal and N 2 O release in ecosystems; in this regard, paddy soils exhibit strong denitrifying ability. However, the underlying mechanism of N 2 O emission from denitrification in paddy soils is yet to be elucidated. In this study, the potential N 2 O emission rate, enzymatic activity for N 2 O production and reduction, gene abundance, and community composition during denitrification were investigated using the 15 N isotope tracer technique combined with slurry incubation, enzymatic activity detection, quantitative polymerase chain reaction (qPCR), and metagenomic sequencing. Results of incubation experiments showed that the average potential N 2 O emission rates were 0.51 ± 0.20 μmol⋅N⋅kg –1 ⋅h –1 , which constituted 2.16 ± 0.85% of the denitrification end-products. The enzymatic activity for N 2 O production was 2.77–8.94 times than that for N 2 O reduction, indicating an imbalance between N 2 O production and reduction. The gene abundance ratio of nir to nos Z from qPCR results further supported the imbalance. Results of metagenomic analysis showed that, although Proteobacteria was the common phylum for denitrification genes, other dominant community compositions varied for different denitrification genes. Gammaproteobacteria and other phyla containing the nor B gene without nos Z genes, including Actinobacteria, Planctomycetes, Desulfobacterota, Cyanobacteria, Acidobacteria, Bacteroidetes, and Myxococcus, may contribute to N 2 O emission from paddy soils. Our results suggest that denitrification is highly modular, with different microbial communities collaborating to complete the denitrification process, thus resulting in an emission estimation of 13.67 ± 5.44 g N 2 O⋅m –2 ⋅yr –1 in surface paddy soils.

Distinct Denitrifying Phenotypes of Predominant Bacteria Modulate Nitrous Oxide Metabolism in Two Typical Cropland Soils

Abstract:Denitrifying nitrous oxide (N2O) emissions in agroecosystems result from variations in microbial composition and soil properties. However, the microbial mechanisms of differential N2O emissions in agricultural soils are less understood. In this study, microcosm experiments using two main types of Chinese cropland soil were conducted with different supplements of nitrate and glucose to simulate the varying nitrogen and carbon conditions. The results show that N2O accumulation in black soil (BF) was significantly higher than that in fluvo-aquic soil (FF) independent of nitrogen and carbon. The abundance of most denitrifying genes was significantly higher in FF, but the ratios of genes responsible for N2O production (nirS and nirK) to the gene responsible for N2O reduction (nosZ) did not significantly differ between the two soils. However, the soils showed obvious discrepancies in denitrifying bacterial communities, with a higher abundance of N2O-generating bacteria in BF and a higher abundance of N2O-reducing bacteria in FF. High accumulation of N2O was verified by the bacterial isolates of Rhodanobacter predominated in BF due to a lack of N2O reduction capacity. The dominance of Castellaniella and others in FF led to a rapid reduction in N2O and thus less N2O accumulation, as demonstrated when the corresponding isolate was inoculated into the studied soils. Therefore, the different phenotypes of N2O metabolism of the distinct denitrifiers predominantly colonized the two soils, causing differing N2O accumulation. This knowledge would help to develop a strategy for mitigating N2O emissions in agricultural soils by regulating the phenotypes of N2O metabolism.
Characteristics of soil N2O emission and N2O-producing microbial communities in paddy fields under elevated CO2 concentrations

Abstract:The effects of elevated carbon dioxide (CO2) concentration (e[CO2]) on nitrous oxide (N2O) emissions from paddy fields and the microbial processes involved in N2O emissions have recently received much attention. Ammonia-oxidizing microorganisms and denitrifying bacteria dominate the production of N2O in paddy soils. To better understand the dynamics of N2O production under e[CO2], a field experiment was conducted after five years of CO2 fumigation based on three treatments: CK (ambient atmospheric CO2), T1 (CK + increase of 40 ppm per year until 200 ppm), and T2 (CK + 200 ppm). N2O fluxes, soil physicochemical properties, and N2O production potential were quantified during the rice-growth period. The functional gene abundance and community composition of ammonia-oxidizing archaea (AOA) and bacteria (AOB) were analyzed using quantitative polymerase chain reaction (qPCR) and those of ammonia-denitrifying bacteria (nirS- and nirK-type) were analyzed using Illumina MiSeq sequencing. N2O emissions decreased by 173% and 41% under the two e[CO2] treatments during grain filling and milk ripening, respectively (P < 0.05). N2O emissions increased by 279% and 172% in the T2 treatment compared with T1 during the tillering and milk-ripening stages, respectively (P < 0.05). Furthermore, the N2O production potential was significantly higher in the CK treatment than in T1 and T2 during the elongation stage. The N2O production potential and abundance of AOA amoA genes in T1 treatment were significantly lower than those in CK treatment during the high N2O emission phase caused by mid-season drainage (P < 0.05). Although nirK- and nirS-type denitrifying bacteria community structure and diversity did not respond significantly (P > 0.05) to e[CO2], the abundance of nirK-type denitrifying bacteria significantly affected the N2O flux (P < 0.05). Linear regression analysis showed that the N2O production potential, AOA amoA gene abundance, and nirK gene abundance explained 47.2% of the variation in N2O emissions. In addition, soil nitrogen (N) significantly affected the nirK- and nirS-type denitrifier communities. Overall, our results revealed that e[CO2] suppressed N2O emissions, which was closely associated with the abundance of AOA amoA and nirK genes (P < 0.05).

Effects of Straw Amendment in Combination with Synthetic N Fertilizer Addition on N2O, N2, and Their Stoichiometric Ratios in Three Different Agro-Ecosystems

Abstract:Nitrogen (N) fertilizer and crop residue amendments are important agricultural practices that could increase soil health, fertility, and crop yield. Such practices may also change soil denitrification processes where contradictory observations have been reported on soil N2O emissions with fewer studies on N2 emissions due to its large atmospheric background concentrations limiting its soil-borne measurement. This study aims to investigate N2O production and reduction of N2 emissions under a conducive denitrifying environment (like anaerobic microsites, 80% WFPS, available N and C) after rice straw amendment and KNO3 application to three different soil types (fluvo-aquic, black, and paddy soils). In this regard, three treatments for three different soil types were set consisting of (a) a non-amended treatment (control), (b) a KNO3 treatment (KNO3, 20 mM KNO3), and (c) a straw plus KNO3 treatment (2.5 g rice straw kg−1 dry soil and 20 mM KNO3), which were incubated under 80% WFPS. Moreover, direct N2O and N2 fluxes were measured over 17 days in the current incubation experiment with a robotized incubation system using a helium atmosphere. Results showed that rice straw amendment combined with N fertilizer increased both N2O and N2 fluxes compared with control or KNO3 treatments in all three soil types. Overall, compared with the black and paddy soils, the N2O and N2 fluxes were higher in the fluvo-aquic soil, with a maximum of 234.2 ± 6.3 and 590.1 ± 27.3 g N ha−1 from F_SK treatment, respectively, during the incubation period. The general trends in three soil types of both N2O and N2 emissions were control < KNO3 < rice straw plus KNO3 treatments. Straw amendment in combination with KNO3 can stimulate a high denitrification rate (less N2O and higher N2), whereas their effect on stoichiometric ratios of N2O/(N2O + N2) highly depends on soil nitrate concentration, oxygen level, soil moisture content, and labile C. The current study underscores that the rice straw amendment in combination with N fertilizer can trigger denitrification with less increment on soil N2O but higher N2 emissions under conditions favoring denitrification.

Abstract:Most studies on soil N2O emissions have focused either on the quantifying of agricultural N2O fluxes or on the effect of environmental factors on N2O emissions. However, very limited information is available on how land-use will affect N2O production, and nitrifiers involved in N2O emissions in agricultural soil ecosystems. Therefore, this study aimed at evaluating the relative importance of nitrification and denitrification to N2O emissions from different land-use soils and identifying the potential underlying microbial mechanisms. A (15)N-tracing experiment was conducted under controlled laboratory conditions on four agricultural soils collected from different land-use. We measured N2O fluxes, nitrate ([Formula: see text]), and ammonium ([Formula: see text]) concentration and (15)N2O, (15)[Formula: see text], and (15)[Formula: see text] enrichment during the incubation. Quantitative PCR was used to quantify ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB). Our results showed that nitrification was the main contributor to N2O production in soils from sugarcane, dairy pasture and cereal cropping systems, while denitrification played a major role in N2O production in the vegetable soil under the experimental conditions. Nitrification contributed to 96.7% of the N2O emissions in sugarcane soil followed by 71.3% in the cereal cropping soil and 70.9% in the dairy pasture soil, while only around 20.0% of N2O was produced from nitrification in vegetable soil. The proportion of nitrified nitrogen as N2O (PN2O-value) varied across different soils, with the highest PN2O-value (0.26‰) found in the cereal cropping soil, which was around 10 times higher than that in other three systems. AOA were the abundant ammonia oxidizers, and were significantly correlated to N2O emitted from nitrification in the sugarcane soil, while AOB were significantly correlated with N2O emitted from nitrification in the cereal cropping soil. Our findings suggested that soil type and land-use might have strongly affected the relative contribution of nitrification and denitrification to N2O production from agricultural soils.

High accumulation of N 2 O was verified by the isolates of Rhodanobacter , which is predominant in BF due to its truncated denitrifying genes and lack of N 2 O reduction capacity. The dominance of complete denitrifiers such as Castellaniella in FF led to a rapid reduction in N 2 O and reduced N 2 O accumulation, as demonstrated when its corresponding isolate was inoculated into both studied soils. Therefore, the different phenotypes of N 2 O metabolism of the distinct denitrifiers maintained in the two soils caused their differing N 2 O accumulation. This knowledge could guide the regulation of the denitrifying bacterial community and the phenotypes of N 2 O metabolism in agricultural soils to reduce N 2 O emissions.
Investigating drivers of N2 loss and N2O reducers in paddy soils across China

Abstract:Denitrification plays a pivotal role in nitrogen (N) cycling in rice paddies, significantly impacting N loss and greenhouse gas emissions. Accurate quantification of net N2 emissions from paddy fields is therefore essential for improving fertilizer N use efficiency. However, challenges in directly measuring gaseous N2 hinder our understanding of microbially-mediated N loss in paddy soils at large scales. In this study, we investigated net N2 loss and its influencing factors in 45 paddy soils across China using membrane inlet mass spectrometry and N2/Ar technique, complemented by microbial community analysis via metagenomics. Potential N2 loss rates varied from 0.41 to 3.58 nmol N g-1 h-1, with no significant regional differences. However, soils from rice-upland rotation (1.72 ± 0.64 nmol N g-1 h-1) and mono-rice cropping systems (1.41 ± 0.53 nmol N g-1 h-1) exhibited higher N2 loss rates compared to double-rice cropping soils (1.13 ± 0.62 nmol N g-1 h-1). Our results revealed a unimodal relationship between soil N2 loss rates and soil pH, with N2O reducers and soil properties primarily regulating regional variations in N2 loss. Significant ecological differentiation was observed within both nosZ Clade I and Clade II, with soil pH emerging as the key factor shaping their community composition. Specifically, in rice-upland rotations, soil moisture and pH significantly influenced nosZ Clade I, while in double-rice cropping systems, soil texture and pH were the main factors affecting nosZ Clade II, thereby driving N2 loss. These findings enhance our understanding of N2 loss dynamics in paddy soil ecosystems, underscoring the critical role of N2O reducers on microbial-derived N2 loss and highlighting the importance of developing strategies to mitigate N2O emissions by balancing N2 loss through the manipulation of N2O-reducing and N2O-producing microbes.
Effects of fertilizer application schemes and soil environmental factors on nitrous oxide emission fluxes in a rice-wheat cropping system, east China

Abstract:Nitrous oxide (N2O) is a potent greenhouse gas (GHG) with agricultural soils representing its largest anthropogenic source. However, the mechanisms involved in the N2O emission and factors affecting N2O emission fluxes in response to various nitrogenous fertilizer applications remain uncertain. We conducted a four-year (2012-2015) field experiment to assess how fertilization scheme impacts N2O emissions from a rice-wheat cropping system in eastern China. The fertilizer treatments included Control (CK), Conventional fertilizer (CF), CF with shallow-irrigation (CF+SI), CF with deep-irrigation system (CF+DI), Optimized fertilizer (OF), OF with Urease inhibitor (OF+UI), OF with conservation tillage (OF+CT) and Slow-release fertilizer (SRF). N2O emissions were measured by a closed static chamber method. N2O emission fluxes ranged from 0.61 μg m-2 h-1 to 1707 μg m-2 h-1, indicating a significant impact of nitrogen fertilizer and cropping type on N2O emissions. The highest crop yields for wheat (3515-3667 kg ha-1) and rice (8633-8990 kg ha-1) were observed under the SRF and OF+UI treatments with significant reduction in N2O emissions by 16.94-21.20% and 5.55-7.93%, respectively. Our findings suggest that the SRF and OF+UI treatments can be effective in achieving maximum crop yield and lowering N2O emissions for the rice-wheat cropping system in eastern China.

Abstract:Nitrous oxide (N2O) is an important greenhouse gas and a catalyst of stratospheric ozone decay. Agricultural soils are the source of 75% of anthropogenic N2O emissions globally. Recently, significant attention has been directed at examining effects of conservation tillage on carbon sequestration in agricultural systems. However, limited knowledge is available regarding how these practices impact N2O emissions, especially for organic vegetable production systems. In this context, a three-year study was conducted in a well-drained sandy loam field transitioning to organic vegetable production in the Mid-Atlantic coastal plain of USA to investigate impacts of conservation tillage [strip till (ST) and no-till (NT)] and conventional tillage (CT) [with black plastic mulch (CT-BP) and bare-ground (CT-BG)] on N2O emissions. Each year, a winter cover crop mixture (forage radish: Raphanus sativus var. longipinnatus, crimson clover: Trifolium incarnatum L., and rye: Secale cereale L.) was grown and flail-mowed in the spring. Nearly 80% of annual N2O-nitrogen (N) emissions occurred during the vegetable growing season for all treatments. Annual N2O-N emissions were greater in CT-BP than in ST and NT, and greater in CT-BG than in NT, but not different between CT-BG and CT-BP, ST and NT, or CT-BG and ST. Conventional tillage promoted N mineralization and plastic mulch increased soil temperature, which contributed to greater N2O-N fluxes. Though water filled porosity in NT was higher and correlated well with N2O-N fluxes, annual N2O-N emissions were lowest in NT suggesting a lack of substrates for nitrification and denitrification processes. Crop yield was lowest in NT in Year 1 and CT-BP in Year 3 but yield-scaled N2O-N emissions were consistently greatest in CT-BP and lowest in NT each year. Our results suggest that for coarse-textured soils in the coastal plain with winter cover crops, conservation tillage practices may reduce N2O emissions in organic vegetable production systems.

Abstract:Numerous interrelated factors (e.g., the labile C, soil NO3− concentration, and soil moisture content) are involved in controlling the microbial sources of N2O and the product stoichiometry of denitrification; however, the interactions among different factors are still poorly understood. Here, a fully robotized continuous flow soil incubation system (allowing simultaneous measurements of N2 and N2O fluxes) was employed to investigate the interactive effects of a 51-day duration of moist spell, straw amendment, and the NO3− level on the rate and product stoichiometry (N2O/(N2O + N2) ratio) of denitrification in heavily N loaded arable soils (i.e., paddy, vegetable, and orchard soils). The rewetting-induced N2O emissions mainly originated from bacterial denitrification in all soil types, with a clear shift to fungal denitrification (plus contingent nitrification) over time. The vegetable and orchard soils showed a higher share of bacterial N2O (62–70%) than that in the paddy soils (50–54%), which may be attributed to more labile-C driven bacterial activity induced by the greater manure and crop residue input therein. Interestingly, the inhibitory effect of high soil NO3− on N2O reduction in these soils was offset by a 51-day-long moist spell, regardless of the amendment of straw. To our knowledge, our study is the first to show that the inhibitory effect of high residual NO3− on N2O reduction is suppressed by a moist spell with a certain duration in heavily N loaded arable soils, suggesting that the water regime history should be considered when optimizing the N fertilizer application timing to mitigate soil N2O emissions.

Abstract:Emissions of the greenhouse gas nitrous oxide (N2O) from soil are sensitive to many factors including temperature, nutrient status, pH and bulk-density. Interactions among these are complex, particularly in agricultural systems where fertilizer-addition has interactive influences across many soil properties. Under laboratory conditions, the temperature sensitivity of N2O emissions, as measured by Q(10) values (fractional change in rate with a 10 degrees C increase in temperature), was higher in soils receiving long-term fertilizer addition compared with control. Different fertilization regimes significantly influenced the emission pathways. Application of manure increased the proportion of potential N2O derived from denitrification although the incubation condition used in the present study might not be favorable for anaerobic denitrification. The abundance of archaeal amoA gene copies increased under all fertilizer treatments, but that of bacterial amoA only increased under mineral (NPK) fertilization. Meanwhile, abundance of nirS, nirK and nosZ only increased under OM and MNPK fertilization. T-RFLP analyses showed that both ammonia oxidizing and denitrifier community structures were altered by fertilization, but only the nirS community structure was sensitive to temperature change. Furthermore, a strong correlation was observed between nirS gene abundance and potential N2O emissions. Relationships between AOA, nirK gene abundances and potential N2O emission were significant but relatively weak. PLS path model revealed that besides direct effect, potential N2O emission was also indirectly influenced by temperature through mediation of NH4+ concentration and nirS-type denitrifier. Our work suggests that warming-induced elevation of potential N2O emission could be strengthened by long-term application of fertilizers, especially organic manure, via shifting community abundance and structure of nirS-type denitrifier. Contributions of Ammonia-Oxidizing Archaea and Bacteria to Nitrous Oxide Production in Intensive Greenhouse Vegetable Fields

Abstract:With excessive nitrogen (N) input, high nitrous oxide (N2O) emissions are frequently observed in greenhouse vegetable fields. We hypothesized that the underlying production mechanisms can be derived across a wide selection of vegetable fields in the middle and lower reaches of the Yangtze River. Thus, we investigated the emission characteristics and relative contributions of ammonia-oxidizing archaea (AOA) and bacteria (AOB) and other microbial processes to the N2O production from five long-term greenhouse vegetable fields through an incubation experiment with combined inhibition methods. The results showed that the ammonia oxidation process is the dominant contributor to N2O production at all five sites, accounting for 88–97% of the total N2O emissions. Regardless of acidic, neutral, or alkaline soil, AOA-driven N2O emission rates were consistently higher than AOB-driven N2O emission rates. Both AOA-driven and AOB-driven N2O emissions exhibited positive correlations with soil pH, with significant increases in soil N2O production associated with high pH levels. Therefore, general production mechanisms were derived, such that more attention should be paid to AOA-driven N2O emissions and to vegetable soils with a relatively high pH in the middle and lower reaches of the Yangtze River.

Abstract:Acetylene (C2H2) is often employed to assess soil total denitrification (N2O + N2) due to its ease of implementation. However, this technique underestimates soil denitrification in soils with low nutrient contents, particularly those supporting grain yields. To our knowledge, there are limited studies that have specifically investigated the impact of C2H2 on nutrient-rich vegetable soils, especially concerning the emissions of N2 and N2O and the nitrogenous gas product ratio (i.e., N2O/(N2O + N2)). In this study, we conducted both anoxic and oxic incubations at various C2H2 concentrations (0%, 0.01%, and 10%, v/v) and utilized a robotized sampling and analysis system to quantify soil N2, N2O, and CO2 emissions. Our findings revealed that the cumulative N2O production in soil treated with 10%C2H2 was significantly lower than that in soil treated with 0.01%C2H2 and soil without C2H2. Contrarily, high concentrations of C2H2 (10%, v/v) led to increased N2 production. Similar trends were observed under oxic conditions, where 10%C2H2 concentration did not enhance N2O production but markedly increased N2 and CO2 emissions. Moreover, the N2O/(N2O + N2) product ratio was notably higher in soils treated with 0%C2H2 compared to the 10%C2H2 treatment under anoxic conditions. These findings indicate that high concentrations of acetylene could facilitate the reduction of N2O to N2 and lead to underestimated soil total denitrification in vegetable soil, regardless of anoxic or oxic conditions. This discovery underscores the drawbacks when employing high concentrations of acetylene to evaluate actual total denitrification in intensive greenhouse vegetable soils, highlighting the necessity for further investigation into alternative methodologies.

Abstract:Conversion of forestland to intensively managed agricultural land occurs worldwide and can increase soil nitrous oxide (N2O) emissions by altering the transformation processes of nitrogen (N) cycling related microbes and environmental conditions. However, little research has been conducted to assess the relationships between nitrifying and denitrifying functional genes and enzyme activities, the altered soil environment and N2O emissions under forest conversion in subtropical China. Here, we investigated the long-term (two decades) effect of converting natural forests to intensively managed tea (Camellia sinensis L.) plantations on soil potential N2O emissions, inorganic N concentrations, functional gene abundances of nitrifying and denitrifying bacteria, as well as nitrifying and denitrifying enzyme activities in subtropical China. The conversion significantly increased soil potential N2O emissions, which were regulated directly by increased denitrifying enzyme activity (52 %) and nirS + nirK gene abundance (38 %) as shown by structural equation modeling, and indirectly by AOB-amoA gene abundance and inorganic N concentration. Our results indicate that converting natural forests to tea plantations directly increases soil inorganic N concentration, resulting in increases in the abundance of soil nitrifying and denitrifying microorganisms and the associated N2O emissions. These findings are crucial for disentangling the factors that directly and indirectly affect soil potential N2O emissions respond to the conversion of forest to tea plantation.

Abstract:Plastic-shed vegetable cultivation is characterized by high nitrogen (N) application rates, frequent irrigation and multiple cropping, probably leading to different soil N transformation rates compared to open-field cultivation. However, how continuous cultivation of vegetables under plastic sheds affects soil N transformation rates and associated nitrous oxide (N2O) emissions remains unclear. Here, a 15N-tracing experiment in combination with the qPCR technique was used to investigate the effects of conversion from conventional open-field cultivation to plastic-shed vegetable cultivation, as well as the age of this shift, on soil gross N transformation rates, N2O emission pathways and associated microbial abundances. Five typical fields, including a wheat–maize rotation system field and four adjacent plastic-shed vegetable fields under cultivation for 2, 5, 12 and 20 years, were selected. Soil gross N mineralization rates increased significantly after two years after conversion from a wheat–maize rotation system to a plastic-shed vegetable system, and then decreased with an increase in plantation age, which could be attributed to decreased soil total phosphorus availability. Long-term vegetable plantation under plastic-shed decreased soil gross autotrophic nitrification rates significantly by decreasing ammonia-oxidizing bacteria abundance probably due to decreasing soil pH. Such decreased rates of autotrophic nitrification further led to decreased N2O emissions under plastic-shed cultivation. Under long-term plastic-shed cultivation (>2 years), negligible gross nitrate-N (NO3−-N) immobilization rates were observed due to the preferential use of ammonium-N by soil microbes, which was probably responsible for rapid NO3−-N accumulation in soils. Overall, these results could be of help to elucidate the mechanisms of soil N2O emission and NO3−-N accumulation in plastic-shed vegetable fields.
