Abstract:Herein we report a highly specific electrochemical aptasenseor for AFB1 determination based on AFB1-controlled diffusion of redox probe (Ru(NH3)63+) through nanochannels of AFB1-specific aptamer functionalized VMSF. A high density of silanol groups on the inner surface confers VMSF with cationic permselectivity, enabling electrostatic preconcentration of Ru(NH3)63+ and producing amplified electrochemical signals. Upon the addition of AFB1, the specific interaction between the aptamer and AFB1 occurs and generates steric hindrance effect on the access of Ru(NH3)63+, finally resulting in the reduced electrochemical responses and allowing the quantitative determination of AFB1. The proposed electrochemical aptasensor shows excellent detection performance in the range of 3 pg/mL to 3 μg/mL with a low detection limit of 2.3 pg/mL for AFB1 detection. Practical analysis of AFB1 in peanut and corn samples is also accomplished with satisfactory results by our fabricated electrochemical aptasensor.

Abstract:Aflatoxin in food is a significant concern that threatens human health and life and causes a heavy burden on international trade and healthcare. In this work, we developed a new type of label-free electrochemical immunosensor for the high-sensitivity determination of aflatoxin B1 (AFB1) based on Au nanoparticles decorated urchin-like Bi 2 S 3 (Au/Bi 2 S 3 ) anchored on electrochemically reduced graphene oxide (ERGO) modified carbon fiber (CF) microelectrode (Au/Bi 2 S 3 /ERGO/CF). The morphology and characterization of the modified microelectrode were investigated using scanning electron microscopy (SEM), cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and differential pulse voltammetry (DPV). Under the optimum conditions, the label-free electrochemical immunosensor detected AFB1 within the linear range of 10 pg mL -1 to 20 ng mL -1 , with a low detection limit of 8 pg mL -1 and high sensitivity (0.48 μA ng -1 mL). The immunosensor showed high selectivity, good reproducibility, and acceptable stability. The immunosensor was applied to detecting AFB1 in cornflour samples, offering excellent reliability and accuracy compared with typical detection methods. Finally, this platform provides a superior prospect for the development of a nanohybrid immunosensor.

Effect of different calcium antagonists on transcapillary fluid shift.

Abstract:Chronic treatment with dihydropyridines, and to a lesser extent other calcium antagonists, often results in peripheral edema without fluid retention. The possibility that calcium antagonists affect extracellular fluid volume partition was tested by comparing the effects of nicardipine (Nic), diltiazem (Dil) and Vehicle (Veh) on hematocrit and plasma protein concentration in anephric anesthetized rats. In response to a 45 min infusion of Nic (0.1 or 1 micrograms/kg/min) and Dil (10 or 100 micrograms/kg/min), blood pressure decreased by 4 or 21%, and 4 or 19%, respectively, whereas hematocrit increased only with both doses of Nic (5.3 +/- 0.2 and 5.5 +/- 0.2%). Plasma protein concentration also increased significantly, although slightly, in the Nic group. The possibility of protein extravasation was then assessed using Evans blue dye as a marker. The drug-induced extravasation of dyed albumin in skeletal and cardiac muscles, but not other organs, was significantly increased following Nic, as compared to Dil or Veh. These observations suggest that two structurally different calcium antagonists used at equihypotensive doses may exert different effects on extracellular fluid partition.
Label-Free Amperometric Immunosensor Based on Versatile Carbon Nanofibers Network Coupled with Au Nanoparticles for Aflatoxin B1 Detection

Abstract:Facile detection methods for mycotoxins with high sensitivity are of great significance to prevent potential harm to humans. Herein, a label-free amperometric immunosensor based on a 3-D interconnected carbon nanofibers (CNFs) network coupled with well-dispersed Au nanoparticles (AuNPs) is proposed for the quantitative determination of aflatoxin B1 (AFB1) in wheat samples. In comparison to common carbon nanotubes (CNTs), the CNFs network derived from bacterial cellulose biomass possesses a unique hierarchically porous structure for fast electrolyte diffusion and a larger electrochemical active area, which increases the peak current of differential pulse voltammetry curves for an immunosensor. Combined with AuNPs that are incorporated into CNFs by using linear polyethyleneimine (PEI) as a soft template, the developed Au@PEI@CNFs-based immunosensor showed a good linear response to AFB1 concentrations in a wide range from 0.05 to 25 ng mL-1. The limit of detection was 0.027 ng mL-1 (S/N = 3), more than three-fold lower than that of an Au@PEI@CNTs-based sensor. The reproducibility, storage stability and selectivity of the immunosensor were proved to be satisfactory. The developed immunosensor with appropriate sensitivity and reliable accuracy can be used for the analysis of wheat samples.

Abstract:Aflatoxin B1 (AFB1) is the most toxic mycotoxin, is widely found in foods and animal feeds, and can pose a serious threat to our lives. A label-free photoelectrochemical (PEC) immunosensor was fabricated for the sensitive detection of AFB1. A Z-scheme heterojunction of gold nanoparticles (Au NPs) loaded on graphitic carbon nitride sheet and tungsten trioxide sphere composite (g-C3N4/Au/WO3) acted as the highly sensitive platform. The g-C3N4/Au/WO3 is capable, not only of immobilizing antibodies via Au NPs, but also enhancing the separation of electron-hole pairs due to its good energy band matching efficiency. The mechanism of photo-generated electron/hole transfer on g-C3N4/Au/WO3 was explored using scavengers to eliminate active components. On this basis, an electron transfer pathway for the immunosensor was deduced. The PEC immunosensor displayed a linear concentration range from 1.0 pg mL-1 to 100 ng mL-1 and a low detection limit of 0.33 pg mL-1 (S/N = 3) for AFB1. Good reproducibility, stability, and specificity provide a solid foundation for the practical application of this immunosensor.

Abstract:Aflatoxin B1 (AFB1), a hepatotoxic and carcinogenic food contaminant, is commonly found in agricultural food. Herein, Au NPs anchored ZIF-8-derived porous carbon-ZnO (Au NPs/PCZIF-8-ZnO) was firstly synthesized to act as the sensing substrate. Then, a ratiometric electrochemical (EC) and "off-on" photoelectrochemical (PEC) dual-mode paper-based aptasensor was presented for AFB1 detection based on a distance-modulation sensing strategy. The independent signal transduction mechanisms and output mode not only broaden the dynamic detection range but also provide a self-verification to assay results, improving the sensitivity and reliability. The wide detection ranges of 0.1 pg/mL-100 ng/mL (EC mode) and 0.02 pg/mL-100 ng/mL (PEC mode) were obtained using dual-mode aptasensor, with detection limits of 36.7 and 9.3 fg/mL, respectively. The fabricated aptasensor exhibited excellent selectivity, reproducibility and stability. Furthermore, it exhibited good practicability for AFB1 assays in real samples, demonstrating great potential applications for food safety evaluation.

Abstract:Aflatoxin B 1 (AFB 1 ), classified as a class I carcinogen, is a widespread mycotoxin that poses a serious threat to public health and economic development, and the food safety problems caused by AFB 1 have aroused worldwide concern. The development of accurate and sensitive methods for the detection of AFB 1 is significant for food safety monitoring. In this work, a sandwich-type photoelectrochemical (PEC) biosensor for AFB 1 detection was constructed on the basis of an aptamer–antibody structure. A good photocurrent response was obtained due to the sensitization of In 2 S 3 by Ru(bpy) 3 2+ . In addition, this sandwich-type sensor constructed by modification with the antibody, target detector, and aptamer layer by layer attenuated the migration hindering effect of photogenerated carriers caused by the double antibody structure. The aptamer and antibody synergistically recognized and captured the target analyte, resulting in more reliable PEC response signals. CdSe@CdS QDs-Apt were modified as a signal-off probe onto the sensor platform to quantitatively detect AFB 1 with a "signal-off" response, which enhanced the sensitivity of the sensor. The PEC biosensor showed a linear response range from 10 −12 to 10 −6 g mL −1 with a detection limit of 0.023 pg mL −1 , providing a feasible approach for the quantitative detection of AFB 1 in food samples.

Fabrication of a high-performance photoelectrochemical aptamer sensor based on Er-MOF nanoballs functionalized with ionic liquid and gold nanoparticles for aflatoxin B1 detection

Abstract:One of the most dangerous mycotoxins is Aflatoxin B1 (AFB1) that is commonly found in food. Thus, AFB1 has been considered as a serious food safety concern. Herein, using Er-MOF nanoballs assembled with gold nanoparticles (AuNPs) as photoactive elements, a photoelectrochemical aptasensor with high analytical performance was developed for sensing AFB1. Er-MOFs nanoballs were prepared by using ionic liquid as a ligand and Er 3+ as a metal center through a solvothermal method. Chloroauric acid was reduced in-situ onto Er-MOF nanoballs to form nanocomposites. AuNPs can effectively enhance the photocurrent of Er-MOF nanoballs attributed to their excellent conductivity and local surface plasma resonance effect. In addition, they can provide active sites for immobilizing more biorecognition elements that can further enhance the sensing performance. Through self-assembling reaction, thiol-functionalized hairpin DNA (hDNA) was immobilized onto an interface to construct a photoelectrochemical biosensing platform. As the AFB1 aptamers labeled with gold nanoparticles (AuNPs-Apt) were selectively recognized, the photocurrent response can be further amplified. Due to the high affinity of AFB1 and its aptamer (Apt), when Apt is reacted with AFB1, it causes AuNPs-Apt detaching from the PEC aptasensing interface, and thus a reduced photocurrent response is generated. Under optimized conditions, the changes of photocurrent before and after recognition of AFB1 exhibits a linear response from 0.005 to 10.0 ng mL -1 . In addition, the limit of detection obtained was 19.6 fg mL -1 . This PEC aptasensor provides relatively high selectivity, reproducibility and accuracy in the determination of AFB1 in real corn samples.

Abstract:This paper presents a novel nano-material composite membrane for detecting aflatoxin B1 (AFB1). The membrane is based on carboxyl-functionalized multi-walled carbon nanotubes (MWCNTs-COOH) @ antimony-doped tin oxide (ATO)-chitosan (CS). To prepare the immunosensor, MWCNTs-COOH were dissolved in the CS solution, but some MWCNTs-COOH formed aggregates due to the intertwining of carbon nanotubes, blocking some pores. ATO was added to the solution containing MWCNTs-COOH, and the gaps were filled by adsorbing hydroxide radicals to form a more uniform film. This greatly increased the specific surface area of the formed film, resulting in a nano-composite film that was modified on screen-printed electrodes (SPCEs). The immunosensor was then constructed by immobilizing anti-AFB1 antibodies (Ab) and bovine serum albumin (BSA) on an SPCE successively. The assembly process and effect of the immunosensor were characterized using scanning electron microscopy (SEM), differential pulse voltammetry (DPV), and cyclic voltammetry (CV). Under optimized conditions, the prepared immunosensor exhibited a low detection limit of 0.033 ng/mL with a linear range of 1 × 10−3–1 × 103 ng/mL. The immunosensor demonstrated good selectivity, reproducibility, and stability. In summary, the results suggest that the MWCNTs-COOH@ATO-CS composite membrane can be used as an effective immunosensor for detecting AFB1.

Abstract:Abstract Recent experiments with 16 male and 16 female subjects indicated that they like to look down rather steeply at an average of −29° below horizontal, s.d. 11·6°, when sitting with trunk and head upright. This angle is steeper when the visual target is at 0·50 m distance (−33°±11·3°) but flatter when the target is at 1·00 m (−24° ± 10·4°). When head and trunk are reclined to 105° behind horizontal, the overall angle of the preferred line of sight is reduced to −20° ±11·7° below horizontal, with −23° (± 12·5°) at 0·5 m and −16° (±9·6°) below horizontal at 1·0 m target distance. These preferred visual angles did not depend on the visual task. The finding that people look down much more steeply than listed in most human engineering design guides has obvious implications for the layout of workplaces where the task has a strong visual component, e.g. at computer workstations.

Abstract:A novel electrochemical aptasensor was prepared for the simultaneous determination of aflatoxin B1 (AFB1) and ochratoxin A (OTA). Composites of Au nanoparticles and polyethyleneimine-reduced graphene oxide (AuNPs/PEI-RGO) with good electrical conductivity and high specific surface area were employed as the supporting substrate, demonstrating the ability to provide more binding sites for aptamers and accelerate the electron transfer. Aptamers were immobilized on a AuNPs/PEI-RGO surface to specifically recognize AFB1 and OTA. A metal-organic framework of UiO-66-NH2 served as the signal carrier to load metal ions of Cu2+ and Pb2+, which facilitated the generation of independent current peaks and effectively improved the electrochemical signals. The prepared aptasensor exhibited sensitive current responses for AFB1 and OTA with a linear range of 0.01 to 1000 ng/mL, with detection limits of 6.2 ng/L for AFB1 and 3.7 ng/L for OTA, respectively. The aptasensor was applied to detect AFB1 and OTA in cereal samples, achieving results comparable with HPLC-MS, with recovery results from 92.5% to 104.1%. With these merits of high sensitivity and good selectivity and stability, the prepared aptasensor proved to be a powerful tool for evaluating contaminated cereals.

Abstract:Protecting cereals from contamination by mycotoxins requires rapid and sensitive detection methods. In this study, a screening-detection electrochemiluminescent (ECL) aptasensor was constructed by combining Ru(bpy) 3 2+ -doped silica nanoparticles (RuSiNPs) and anodized aluminum oxide (AAO). This aptasensor was successfully used in the ultrasensitive and highly selective detection of aflatoxin B 1 (AFB 1 ) in wheat. In the aptasensor, MXene and gold nanoparticles (AuNPs) act as the conductive material and surface plasmon resonance (SPR) source, respectively, to enhance the ECL signals of RuSiNPs. Meanwhile, due to their highly ordered porous structure, AAO pasted on the electrode surface can effectively screen out the macromolecular interferents, providing a pure environment for the aptamer (Apt) to specifically capture AFB 1 . Because the formed AFB 1 -Apt complex hinders electron transfer, the ECL intensity of the aptasensor decreases with increasing AFB 1 concentration to achieve quantitative detection. Under the optimal conditions, the linear range and detection limit of the screening-detection ECL aptasensor were 0.01−100 μg/kg and 1.5 × 10 −3 μg/kg (S/N = 3), respectively. In summary, the proposed screening-detection ECL aptasensor for the quantitative detection of AFB 1 is a promising new approach for the early detection of moldy wheat.

Abstract:We reported a highly efficient electrochemical immunosensor utilizing chitosanвҖ“graphene nanosheets (CS-GNs) nanocomposites for the detection of aflatoxin B1 (AFB1) in corn samples. The CS-GNs nanocomposites, serving as a modifying layer, provide a significant specific surface area and biocompatibility, thereby enhancing both the electron transfer rate and the efficiency of antibody immobilization. The electrochemical characterization was conducted utilizing both differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS). Moreover, the antibody concentration, pH, antibody immobilization time, and immunoreaction time, were optimized. The results showed that the current change (О”I) before and after the immunoreaction demonstrated a strong linear relationship (R2=0.990) with the AFB1 concentration, as well as good specificity and stability. The linear range extended from 0.05 to 25 ng/mL, with a detection limit of 0.021 ng/mL (S/N=3). The immunosensor exhibited a recovery rate ranging from 97.3% to 101.4% in corn samples, showing a promising performance using an efficient method, and indicating a remarkable prospect for the detection of fungal toxins in grains.

Colorimetric and ECL dual-mode aptasensor for smartphone-based onsite sensitive detection of aflatoxin B1 in combination with ZnO@MWCNTs/g-C3N4 nanosheets and CuO@CuPt nanocomposites

Abstract:The development of dual-mode strategies with superior sensitivity and accuracy have garnered increasing attention for researchers in Aflatoxin B1 (AFB1) analysis. Herein, a colorimetric-electrochemiluminescence (ECL) dual-mode biosensor was constructed for onsite and ultrasensitive determination of AFB1. The multi-wall carbon nanotubes (MWCNTs) were integrated with the ZnO metal organic frameworks (MOFs) to accelerate the electron transfer and boost the ECL intensity of g-C3N4 nanoemitters. Through the aptamer-based DNA sandwich assay, the CuO@CuPt nanocomposites were introduced onto the electrode and acted as the dual functional signal nanoprobes. Due to the good spectrum overlap between the CuO@CuPt nanoprobes and g-C3N4 nanosheets, ECL signal could be efficiently quenched. Additionally, the CuO@CuPt nanoprobes show superior catalytic properties towards the TMB and H2O2 colorimetric reactions, and an obvious color alteration from colorless to blue can be observed using the smartphone. Under optimized conditions, a sensitive and accurate dual-mode analysis of the AFB1 was accomplished with the colorimetric detection limit of 3.26 fg/mL and ECL detection limit of 0.971 fg/mL (S/N = 3). This study combines innovative nanomaterial properties of ZnO@MWCNTs, g-C3N4 and CuO@CuPt for ultrasensitive dual-mode detection, which offers new opportunities for the innovative engineering of the dual-mode sensors and demonstrates significant potential in food safety analysis.

Abstract:In this work, a new competitive immunosensor for aflatoxin B1 (AFB1) detection was developed using europium (Eu) fluorescent nanospheres and magnetic beads. Firstly, Eu nanospheres were synthesized through two steps including carboxylated polystyrene nanospheres and Eu-doped polystyrene nanospheres preparation. Then Eu nanospheres were covalently tagged to anti-AFB1 monoclonal antibody (anti-AFB1 mAb) through an EDC coupling method. Carboxylated Fe 3 O 4 magnetic beads were conjugated to AFB1-BSA through EDC/NHS crosslinking to obtain AFB1-BSA-Fe 3 O 4 . In the absence of AFB1, Eu-anti-AFB1 mAb were incubated with AFB1-BSA-Fe 3 O 4 to form Eu-anti-AFB1 mAb-AFB1-BSA-Fe 3 O 4 in PBS buffer. However, in the presence of AFB1, the competitive interaction of AFB1 and AFB1-BSA-Fe 3 O 4 to bind with Eu-anti-AFB1 mAb occurred. With the increasing concentration of AFB1, less Eu-anti-AFB1 mAb-AFB1-BSA-Fe 3 O 4 formed. So the fluorescence intensity of Eu-anti-AFB1 mAb-AFB1-BSA-Fe 3 O 4 was gradually decreased after magnetic separation. The degree of fluorescence decrease was linear with respect to the logarithm of AFB1 concentration in the range of 0.01–2 ng/mL in both buffer solution and feed samples and the detection limit was 0.003 ng/mL. What's more, the immunosensor showed excellent specificity for AFB1 without being interfered by other mycotoxins. In consideration of the excellent performance of this immunosensor, we can speculate that the proposed method could be widely used in detecting food contaminants.

Abstract:Aflatoxin B1 (AFB1) is a highly toxic mycotoxin that can contaminate foods. Hence, a surface-enhanced Raman scattering (SERS) and fluorescence dual signal-on biosensor was developed for AFB1 detection. Specifically, aminated mesoporous silica nanoparticles (MSNs) were used as nanocontainers to load the Rhodamine 6G (R6G) with fluorescence and Raman effects, aptamers and polydopamine (PDA) were sequentially assembled on the surface of the MSNs as dual-gated molecules, and MXenes and Au nanorods were used as SERS substrates. After adding AFB1 under acidic conditions, the degradation of PDA and specific bringing of the aptamer with AFB1 resulted in the release of R6G. Results showed that AFB1 could be quantified by Raman and fluorescence spectroscopic with detection limits of 0.133 pg mL −1 and 0.214 pg mL −1 , respectively. Moreover, the biosensor could be applied to the detection of AFB1 in real peanut, maize and badam samples, providing a possible alternative solution for the detection of other analytes.

Abstract:Herein, a sandwich electrochemical sensing strategy for aflatoxin b 1 (AFB 1 ) detection based on hybrid-nanoengineering was presented. First, Au nanoparticle was doped into zeolitic imidazolate framework-8 (ZIF-8) to form Au@ZIF-8 by in-situ growth method, followed by multi-walled carbon nanotubes (MWCNTs) addition to synthesize MWCNTs/Au@ZIF-8 via self-assembly. The structural "confinement effect" of ZIF-8 afforded a microenvironment for Au nanoparticles and SMCNTs in a certain spatial region, giving MWCNTs/Au@ZIF-8 excellent electrochemical property as the substrate material. In addition, Au-Pt bimetallic nanoparticle, which exhibited excellent stability and catalytic activity was loaded on the hollow cerium oxide (CeO 2 ) to form AuPt@CeO 2 nanoparticle through one-step aqueous phase reduction. Owning to its high surface-to-volume ratio, satisfied electron transfer efficiency and biocompatibility, massive toluidine blue (TB) and AFB 1 antibody (Ab) could be modified on the AuPt@CeO 2 to form AuPt@CeO 2 -Ab-TB, which acted as signal tag for the ultrasensitive assay of AFB 1 . The proposed electrochemical sensing system exhibited wide detection range (2 × 10 -5 − 20 ng/mL) and low detection limit (2.13 fg/mL), which has been successfully applied to AFB 1 determination in four real samples. The hybrid nanoengineering presented in this work is an active attempt to prepare high-performance substrate material and signal tag, which provides a new insight for the development of highly sensitive and specific electrochemical sensing systems.

Abstract:This work is aimed to develop of a new class of versatile aptasensor to specifically detect aflatoxin B1 (AFB1) using dual-channel detection method. To achieve this objective, gold nanoparticles (AuNPs) having peroxidase-like activity and capability of promoting silver deposition were used as the versatile label for both colorimetric and electrochemical techniques. First of all, aptamer (apt) modified Fe3O4@Au magnetic beads (MBs-apt) and cDNA modified AuNPs (cDNA-AuNPs) were prepared to use as capture probes and signal probes, respectively. Taking advantage of hybridization reaction between apt and cDNA, these two probes were coupled with each other to generate MBs-apt/cDNA-AuNPs bioconjugations. The high affinity between apt and AFB1 made cDNA-AuNPs detached from MBs-apt, and the released signal probes were separated and collected using an external magnetic field and used for both colorimetric and electrochemical detection channels. The dual-channel signals were directly proportional to logarithm of AFB1 concentration within the ranges of 5-200 ng mL-1 and 0.05-100 ng mL-1. The detection limit can reach as low as 35 pg mL-1 and 0.43 pg mL-1 for colorimetric and electrochemical channel, respectively. Moreover, the proposed aptasensor has been successfully applied to determine AFB1 in corn samples with satisfactory results. This dual-channel detection method can not only improve the detection precision and diversity significantly, but also can reduce the false-negative and-positive rates in food quality monitoring. We believe we have provided a general strategy with the convincing dual-readout mode which possess great promising in all of the aptamer related fields.

Abstract:A sandwich electrochemical sensor was fabricated based on multi-walled carbon nanotubes/ordered mesoporous carbon/AuNP (MWCNTs/CMK-3/AuNP) nanocomposites and porous core-shell nanoparticles Au@PdNPs to achieve rapid and sensitive detection of AFB1 in complex matrices. MWCNTs/CMK-3/AuNP nanocomposite, which was prepared by self-assembly method, served as a substrate material to increase the aptamer loading and improve the conductivity and electrocatalytic activity of the electrode for the first signal amplification. Then, Au@PdNPs, which were synthesized by one-pot aqueous phase method, were applied as nanocarriers loaded with plenty of capture probe antibody (Ab) and signal molecule toluidine blue (Tb) to form the Au@PdNPs-Ab-Tb bioconjugates for secondary signal amplification. The sensing system could still significantly improve the signal output intensity even in the presence of ultra-low concentration target compound due to the dual signal amplification of MWCNTs/CMK-3/AuNP nanocomposites and Au@PdNPs-Ab-Tb. The method exhibited high selectivity, low detection limit (9.13 fg/mL), and strong stability to differentiate AFB1 from other mycotoxins. Furthermore, the sensor has been successfully applied to the quantitative determination of AFB1 in corn, malt, and six herbs, which has potential applications in food safety, quality control, and environmental monitoring.
