Abstract:Herein we report a highly specific electrochemical aptasenseor for AFB1 determination based on AFB1-controlled diffusion of redox probe (Ru(NH3)63+) through nanochannels of AFB1-specific aptamer functionalized VMSF. A high density of silanol groups on the inner surface confers VMSF with cationic permselectivity, enabling electrostatic preconcentration of Ru(NH3)63+ and producing amplified electrochemical signals. Upon the addition of AFB1, the specific interaction between the aptamer and AFB1 occurs and generates steric hindrance effect on the access of Ru(NH3)63+, finally resulting in the reduced electrochemical responses and allowing the quantitative determination of AFB1. The proposed electrochemical aptasensor shows excellent detection performance in the range of 3 pg/mL to 3 μg/mL with a low detection limit of 2.3 pg/mL for AFB1 detection. Practical analysis of AFB1 in peanut and corn samples is also accomplished with satisfactory results by our fabricated electrochemical aptasensor.

Abstract:Aflatoxin B1 (AFB1) is the most toxic mycotoxin, is widely found in foods and animal feeds, and can pose a serious threat to our lives. A label-free photoelectrochemical (PEC) immunosensor was fabricated for the sensitive detection of AFB1. A Z-scheme heterojunction of gold nanoparticles (Au NPs) loaded on graphitic carbon nitride sheet and tungsten trioxide sphere composite (g-C3N4/Au/WO3) acted as the highly sensitive platform. The g-C3N4/Au/WO3 is capable, not only of immobilizing antibodies via Au NPs, but also enhancing the separation of electron-hole pairs due to its good energy band matching efficiency. The mechanism of photo-generated electron/hole transfer on g-C3N4/Au/WO3 was explored using scavengers to eliminate active components. On this basis, an electron transfer pathway for the immunosensor was deduced. The PEC immunosensor displayed a linear concentration range from 1.0 pg mL-1 to 100 ng mL-1 and a low detection limit of 0.33 pg mL-1 (S/N = 3) for AFB1. Good reproducibility, stability, and specificity provide a solid foundation for the practical application of this immunosensor.

Electrochemical/photoelectrochemical dual-mode aptasensor for sensitive aflatoxin B1 assay based on distance-modulation strategy using Au NPs/PCZIF-8-ZnO as sensing substrate

Abstract:Aflatoxin B1 (AFB1), a hepatotoxic and carcinogenic food contaminant, is commonly found in agricultural food. Herein, Au NPs anchored ZIF-8-derived porous carbon-ZnO (Au NPs/PCZIF-8-ZnO) was firstly synthesized to act as the sensing substrate. Then, a ratiometric electrochemical (EC) and "off-on" photoelectrochemical (PEC) dual-mode paper-based aptasensor was presented for AFB1 detection based on a distance-modulation sensing strategy. The independent signal transduction mechanisms and output mode not only broaden the dynamic detection range but also provide a self-verification to assay results, improving the sensitivity and reliability. The wide detection ranges of 0.1 pg/mL-100 ng/mL (EC mode) and 0.02 pg/mL-100 ng/mL (PEC mode) were obtained using dual-mode aptasensor, with detection limits of 36.7 and 9.3 fg/mL, respectively. The fabricated aptasensor exhibited excellent selectivity, reproducibility and stability. Furthermore, it exhibited good practicability for AFB1 assays in real samples, demonstrating great potential applications for food safety evaluation.

Abstract:Aflatoxin B 1 (AFB 1 ), classified as a class I carcinogen, is a widespread mycotoxin that poses a serious threat to public health and economic development, and the food safety problems caused by AFB 1 have aroused worldwide concern. The development of accurate and sensitive methods for the detection of AFB 1 is significant for food safety monitoring. In this work, a sandwich-type photoelectrochemical (PEC) biosensor for AFB 1 detection was constructed on the basis of an aptamer–antibody structure. A good photocurrent response was obtained due to the sensitization of In 2 S 3 by Ru(bpy) 3 2+ . In addition, this sandwich-type sensor constructed by modification with the antibody, target detector, and aptamer layer by layer attenuated the migration hindering effect of photogenerated carriers caused by the double antibody structure. The aptamer and antibody synergistically recognized and captured the target analyte, resulting in more reliable PEC response signals. CdSe@CdS QDs-Apt were modified as a signal-off probe onto the sensor platform to quantitatively detect AFB 1 with a "signal-off" response, which enhanced the sensitivity of the sensor. The PEC biosensor showed a linear response range from 10 −12 to 10 −6 g mL −1 with a detection limit of 0.023 pg mL −1 , providing a feasible approach for the quantitative detection of AFB 1 in food samples.

Abstract:To enhance the performance of photoelectrochemical (PEC) sensors, heterojunction construction is a workable technique. However, the interface defects inside the structure are often ignored. In this work, an innovative photoelectrochemical (PEC) sensor based on a "turn-off" PEC model was constructed using the synergistic effects of in situ growing heterojunction and local surface plasmon resonance (LSPR) to detect aflatoxin B1 (AFB1) instantly. In situ heterojunctions were constructed based on bismuth-based perovskite, and the internal electric field spontaneously established during the growth process can effectively compensate for the interfacial defects and promote the separation and transfer of e-/h+ pairs. Moreover, the gold nanoparticles (Au NPs) LSPR effect can improve PEC performance by enhancing the light trapping ability of the material. The PEC sensor's detecting area for AFB1 analysis is 10.0 fg/ml-100 ng/mL, and the detection limit is 4.34 fg/mL. Good stability, reproducibility, and selectivity underscore the sensor's suitability for meeting the requirements of food quality monitoring.

Abstract:Designing a photoelectrochemical (PEC) biosensor with preponderant sensitivity and anti-interference is a challenge for detecting small molecules in real samples with complex matrices. To this end, the Bi2O3/CuBi2O4 was synthesized in one step to enhance visible light's absorption ability, transferring the interfacial carrier's efficiency, a high-active Z-scheme heterojunction, and a photocathode biosensor was proposed. For the first time, we used the density functional theory to verify a Z-scheme transfer pathway of photogenerated electrons in Bi2O3/CuBi2O4 and the energy band structure of Bi2O3 and CuBi2O4, respectively. Bi2O3/CuBi2O4-based PEC biosensor was developed for competive immunoassay of small molecular, aflatoxin B1 (AFB1) as an example, resulting in a low detection limit of 297.4 fg/mL and a linear range of 1.4 pg/mL-280 ng/mL in urine, water, peanut, and wheat samples. Using spiked experiments, the satisfied repeatability, reproducibility, stability, and specificity of the Bi2O3/CuBi2O4-based PEC biosensor indicated a promise for application in health care, the environment, and food.

Abstract:As one of the most toxic chemical substances, aflatoxin B1 (AFB1) has a strong carcinogenic effect even at a trace level in human and animal, which severely threatens human health and even causes cancers. Therefore, ultrasensitive detection of AFB1 is of significant importance. For such analysis, dual II-scheme sheet-like Bi2S3/Bi2O3/Ag2S heterostructures were prepared by the in-situ growth method, which exhibited high separation efficiency for the electron-hole (e--h+) pairs, prominent stability, and high photoactivity. Moreover, the dendritic nanorod-like Au@Pd@Pt (Au@Pd@Pt DNRs) nanozyme was homely synthesized, whose peroxidase-like activity was scrupulously investigated by catalytical oxidation of diaminobenzidine (DAB) in the presence of H2O2. Integration by the aptasensing strategy, a photoelectrochemical (PEC) "signal-on" aptasensor was prepared, which exhibited a broader linear range of 0.5 pg mL-1-100 ng mL-1 with a lower limit of detection (LOD = 0.09 pg mL-1, S/N = 3). This work provides a feasible strategy to develop advanced PEC biosensors for actual analysis of environmental pollutants.

Abstract:Chronic treatment with dihydropyridines, and to a lesser extent other calcium antagonists, often results in peripheral edema without fluid retention. The possibility that calcium antagonists affect extracellular fluid volume partition was tested by comparing the effects of nicardipine (Nic), diltiazem (Dil) and Vehicle (Veh) on hematocrit and plasma protein concentration in anephric anesthetized rats. In response to a 45 min infusion of Nic (0.1 or 1 micrograms/kg/min) and Dil (10 or 100 micrograms/kg/min), blood pressure decreased by 4 or 21%, and 4 or 19%, respectively, whereas hematocrit increased only with both doses of Nic (5.3 +/- 0.2 and 5.5 +/- 0.2%). Plasma protein concentration also increased significantly, although slightly, in the Nic group. The possibility of protein extravasation was then assessed using Evans blue dye as a marker. The drug-induced extravasation of dyed albumin in skeletal and cardiac muscles, but not other organs, was significantly increased following Nic, as compared to Dil or Veh. These observations suggest that two structurally different calcium antagonists used at equihypotensive doses may exert different effects on extracellular fluid partition.
Construction of a dual-functional CuO/BiOCl heterojunction for high-efficiently photoelectrochemical biosensing and photoelectrocatalytic degradation of aflatoxin B1

Abstract:Aflatoxin B1 (AFB1) exposure often causes serious food safety problems and illnesses in humans and animals even at extremely low content. Therefore, ultrasensitive detection and effective degradation of AFB1 is vitally significant. Photoelectrochemical (PEC) approach has been widely applied in sensing and catalysis fields. For achieving robust PEC performance, exploring highly photoactive semiconductor materials is critical. Herein, we constructed a novel and dual-functional copper oxide/bismuth oxychloride (CuO/BiOCl) composite by in-situ growth of CuO on BiOCl surface. The photo-absorption region of CuO/BiOCl was efficaciously broadened from UV to visible range, making for enhanced light harvest. Meanwhile, the p-n heterostructure in CuO/BiOCl clarified that the formed built-in internal electric field could accelerate band-band transfer of carriers. Driven by this, PEC response of CuO/BiOCl was greatly boosted comparing with that of pure CuO or BiOCl. Further combing with the specific aptamer, a favorable CuO/BiOCl-based PEC biosensor was fabricated for AFB1 detection with ultra-sensitivity (detection limit of 0.07 pg mL−1) and satisfactory recoveries (96.4% ∼ 105.7%) in real maize samples. Subsequently, under light irradiation and suitable bias voltage, a degradation rate of ∼81.3% was facilely attained for 5.0 µg mL−1 AFB1, indicating excellent photoelectrocatalytic activity of CuO/BiOCl material. The catalytic mechanism and the main product of AFB1 degradation were analyzed. Taken together, the heterostructured CuO/BiOCl-based PEC assay provides a potential way for monitoring and controlling the AFB1 contamination in the food security areas.

Abstract:This paper presents a novel nano-material composite membrane for detecting aflatoxin B1 (AFB1). The membrane is based on carboxyl-functionalized multi-walled carbon nanotubes (MWCNTs-COOH) @ antimony-doped tin oxide (ATO)-chitosan (CS). To prepare the immunosensor, MWCNTs-COOH were dissolved in the CS solution, but some MWCNTs-COOH formed aggregates due to the intertwining of carbon nanotubes, blocking some pores. ATO was added to the solution containing MWCNTs-COOH, and the gaps were filled by adsorbing hydroxide radicals to form a more uniform film. This greatly increased the specific surface area of the formed film, resulting in a nano-composite film that was modified on screen-printed electrodes (SPCEs). The immunosensor was then constructed by immobilizing anti-AFB1 antibodies (Ab) and bovine serum albumin (BSA) on an SPCE successively. The assembly process and effect of the immunosensor were characterized using scanning electron microscopy (SEM), differential pulse voltammetry (DPV), and cyclic voltammetry (CV). Under optimized conditions, the prepared immunosensor exhibited a low detection limit of 0.033 ng/mL with a linear range of 1 × 10−3–1 × 103 ng/mL. The immunosensor demonstrated good selectivity, reproducibility, and stability. In summary, the results suggest that the MWCNTs-COOH@ATO-CS composite membrane can be used as an effective immunosensor for detecting AFB1.

Fabrication of a high-performance photoelectrochemical aptamer sensor based on Er-MOF nanoballs functionalized with ionic liquid and gold nanoparticles for aflatoxin B1 detection

Abstract:One of the most dangerous mycotoxins is Aflatoxin B1 (AFB1) that is commonly found in food. Thus, AFB1 has been considered as a serious food safety concern. Herein, using Er-MOF nanoballs assembled with gold nanoparticles (AuNPs) as photoactive elements, a photoelectrochemical aptasensor with high analytical performance was developed for sensing AFB1. Er-MOFs nanoballs were prepared by using ionic liquid as a ligand and Er 3+ as a metal center through a solvothermal method. Chloroauric acid was reduced in-situ onto Er-MOF nanoballs to form nanocomposites. AuNPs can effectively enhance the photocurrent of Er-MOF nanoballs attributed to their excellent conductivity and local surface plasma resonance effect. In addition, they can provide active sites for immobilizing more biorecognition elements that can further enhance the sensing performance. Through self-assembling reaction, thiol-functionalized hairpin DNA (hDNA) was immobilized onto an interface to construct a photoelectrochemical biosensing platform. As the AFB1 aptamers labeled with gold nanoparticles (AuNPs-Apt) were selectively recognized, the photocurrent response can be further amplified. Due to the high affinity of AFB1 and its aptamer (Apt), when Apt is reacted with AFB1, it causes AuNPs-Apt detaching from the PEC aptasensing interface, and thus a reduced photocurrent response is generated. Under optimized conditions, the changes of photocurrent before and after recognition of AFB1 exhibits a linear response from 0.005 to 10.0 ng mL -1 . In addition, the limit of detection obtained was 19.6 fg mL -1 . This PEC aptasensor provides relatively high selectivity, reproducibility and accuracy in the determination of AFB1 in real corn samples.

Abstract:Aflatoxin in food is a significant concern that threatens human health and life and causes a heavy burden on international trade and healthcare. In this work, we developed a new type of label-free electrochemical immunosensor for the high-sensitivity determination of aflatoxin B1 (AFB1) based on Au nanoparticles decorated urchin-like Bi 2 S 3 (Au/Bi 2 S 3 ) anchored on electrochemically reduced graphene oxide (ERGO) modified carbon fiber (CF) microelectrode (Au/Bi 2 S 3 /ERGO/CF). The morphology and characterization of the modified microelectrode were investigated using scanning electron microscopy (SEM), cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and differential pulse voltammetry (DPV). Under the optimum conditions, the label-free electrochemical immunosensor detected AFB1 within the linear range of 10 pg mL -1 to 20 ng mL -1 , with a low detection limit of 8 pg mL -1 and high sensitivity (0.48 μA ng -1 mL). The immunosensor showed high selectivity, good reproducibility, and acceptable stability. The immunosensor was applied to detecting AFB1 in cornflour samples, offering excellent reliability and accuracy compared with typical detection methods. Finally, this platform provides a superior prospect for the development of a nanohybrid immunosensor.

An enhanced photoelectrochemical sensor for aflatoxin B1 detection based on organic-inorganic heterojunction nanomaterial: poly(5-formylindole)/NiO

Abstract:A new strategy for enhancing the photoelectric activity of poly(5-formylindole) (P5FIn) was developed by introducing the inorganic semiconductor material (NiO) to form organic-inorganic heterojunctions. P5FIn/NiO heterojunctions were firstly prepared by combining hydrothermal synthesis and electrochemical polymerization. Due to the synergistic effect between P5FIn and NiO, the photoelectrochemical (PEC) performance of this heterojunction was significantly enhanced compared to pure P5FIn and NiO. The reason for the enhanced PEC performance is mainly attributed to the increased visible light utilization and the bandgap matching effect of the P5FIn/NiO heterojunctions. Based on the prepared P5FIn/NiO heterojunctions, a novel PEC sensor for aflatoxin B1 (AFB1) detection was also constructed with a wide linear range of 0.005-50 ng mL-1 and a limit of detection (LOD) of 0.0015 ng mL-1. Moreover, this constructed PEC sensor also had good stability, reproducibility, selectivity, and satisfactory actual sample detection ability. This strategy may inspire more design and application of high-performance photoelectric active material based on inorganic semiconductor and organic conducting polymer heterojunctions. Abstract:Herein, we fabricated a label-free ECL immunosensor for aflatoxin B 1 (AFB 1 ) detection. In this system, a small organic aggregation-induced electrochemiluminescence luminophore, 2,5-di-tetraphenylethylene-ylthiazolo [5,4-d] thiazole, was designed, named TPETTZ. Polyaniline-wrapped TiO 2 nanoparticles (PANI/TiO 2 NPs) complex was synthesized through one-step in situ oxidation polymerization of aniline, and performed excellent electrical conductivity and abundant amino groups. As an ECL accelerator, TiO 2 nanoparticles (TiO 2 NPs) promoted the oxidation of tri- n -propylamine (TPA) to generate more TPA • ; in addition, it also acted as a donor to improve the ECL intensity of TPETTZ (acceptor) through electrochemiluminescence resonance energy transfer (ECL-RET). Encouraged by the above, under the existence of TPA, TPETTZ displayed a strong and continuously stable ECL anode signal due to the introduction of PANI/TiO 2 NPs. Therefore, the immunosensor was constructed for AFB 1 detection based on the quenching effect of target on the ECL signal, and a linearly decreasing ECL signal was obtained as the increasement of AFB 1 in the range of 75 fg/mL to 100 ng/mL, with a lower detection limit of 27.5 fg/mL. Moreover, the as-prepared sensing platform performed a satisfactory anti-interference, stability, and reproducibility, and appeared a good accuracy in walnut sample analysis, presenting a promising application in the future.

The development of portable, low-cost, and easy to operate analytical tools is essential for AFB1 detection, especially in resource-limited regions. Herein, a dual-mode microfluidic paper-based analytical device (μPAD) with electrochemical and colorimetric readout was constructed for AFB1 detection using Au NPs anchored Ni-Co layered double hydroxides nanocages (Au/Ni-Co LDH NCs) as a signal amplifier. In this design, tetrahedral DNA nanostructures (TDNs) were employed as a scaffold to anchor “bottom-up” aptamers (Apt) for improving capture efficiency of AFB1. Au/Ni-Co LDH NCs with excellent peroxidase (POD)-like activity were bound on the TDNs through hybridization of Apt and complementary DNA (cDNA), a significant electrochemical signal of H2O2 reduction was generated. Meanwhile, the·OH from H2O2 decomposition catalyzed the oxidation of 3,3′,5,5′-tetramethylbenzidine and resulted in a colorimetric signal. The binding of target AFB1 to Apt triggered the dissociation of Apt/cDNA, causing the release of the signal amplifier from the sensing interface and signal decrease. Consequently, a detection range of 0.2 pg/mL-100 ng/mL for electrochemical and 50 pg/mL-100 ng/mL for colorimetric were achieved for AFB1, holding promising application in the fields of food safety.

An "off-on" electrochemical luminescence biosensor with aggregation-induced emission for ultrasensitive detection of aflatoxin B1

Abstract:This work describes a novel "off-on" biosensor for super-sensitive and selective detection of aflatoxin B1 (AFB1). In the biosensor, electrochemiluminescence (ECL) and aggregation-induced emission (AIE) work together to enhance ECL performance by a dibenzo-[b,d]thiophene sulfone-based conjugated microporous polymer, and hairpin DNA1 (H1) and ferrocene (Fc)-apt act as the switches for the "off-on" mode. In the absence of target AFB1, the ECL signal is inhibited because of the steric-hindrance effect, and the biosensor remains in the "off" state. In the presence of AFB1, ECL signal recovery is observed caused by the specific recognition between the AFB1 and Fc-apt, i.e., the biosensor changes to the "on" state. Under optimal experimental conditions, AFB1 can be quantified from 1 pg·mL −1 to 1 μg·mL −1 with an ultralow detection limit of 0.46 pg·mL −1 by the "off-on" biosensor. The biosensor exhibited good practicability for AFB1 assay in corn samples, thus providing a simple and sensitive method for monitoring food safety.

Abstract:Aflatoxin B1 (AFB1) is one of the most harmful mycotoxins to human and environmental health. In this paper, a novel electrochemical AFB1 immunosensor using attapulgite composite N -phenylglycine as a conductive material is developed. The strong π–π and electrostatic interactions between the AFB1 antibody and the nanomaterial are investigated. The developed electrochemical AFB1 immunosensor exhibits high sensitivity, with a detection limit of 0.011 ng mL −1 and a quantitation limit of 0.035 ng mL −1 (S/N = 3). The recoveries for three different concentrations of AFB1 range from 97.5% to 104.6%, with a relative standard deviation of stability less than 2.0%. ATP-PNPG possesses more oxygen defects and adsorbed oxygen than ATP-NPG. These properties enhance its electronegativity and make it more conducive to detecting low concentrations of AFB1 through electrochemical sensing technology. Finally, the electrochemical AFB1 immunosensor demonstrates good selectivity, stability, and reusability for detecting AFB1 in corn samples, exhibiting a high recovery rate.

Abstract:Facile detection methods for mycotoxins with high sensitivity are of great significance to prevent potential harm to humans. Herein, a label-free amperometric immunosensor based on a 3-D interconnected carbon nanofibers (CNFs) network coupled with well-dispersed Au nanoparticles (AuNPs) is proposed for the quantitative determination of aflatoxin B1 (AFB1) in wheat samples. In comparison to common carbon nanotubes (CNTs), the CNFs network derived from bacterial cellulose biomass possesses a unique hierarchically porous structure for fast electrolyte diffusion and a larger electrochemical active area, which increases the peak current of differential pulse voltammetry curves for an immunosensor. Combined with AuNPs that are incorporated into CNFs by using linear polyethyleneimine (PEI) as a soft template, the developed Au@PEI@CNFs-based immunosensor showed a good linear response to AFB1 concentrations in a wide range from 0.05 to 25 ng mL-1. The limit of detection was 0.027 ng mL-1 (S/N = 3), more than three-fold lower than that of an Au@PEI@CNTs-based sensor. The reproducibility, storage stability and selectivity of the immunosensor were proved to be satisfactory. The developed immunosensor with appropriate sensitivity and reliable accuracy can be used for the analysis of wheat samples.

Abstract:We reported a highly efficient electrochemical immunosensor utilizing chitosanвҖ“graphene nanosheets (CS-GNs) nanocomposites for the detection of aflatoxin B1 (AFB1) in corn samples. The CS-GNs nanocomposites, serving as a modifying layer, provide a significant specific surface area and biocompatibility, thereby enhancing both the electron transfer rate and the efficiency of antibody immobilization. The electrochemical characterization was conducted utilizing both differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS). Moreover, the antibody concentration, pH, antibody immobilization time, and immunoreaction time, were optimized. The results showed that the current change (О”I) before and after the immunoreaction demonstrated a strong linear relationship (R2=0.990) with the AFB1 concentration, as well as good specificity and stability. The linear range extended from 0.05 to 25 ng/mL, with a detection limit of 0.021 ng/mL (S/N=3). The immunosensor exhibited a recovery rate ranging from 97.3% to 101.4% in corn samples, showing a promising performance using an efficient method, and indicating a remarkable prospect for the detection of fungal toxins in grains.
