Abstract:Individuals differ not only in the ability to make decisions, but also in the degree to which they respond adaptively to uncertainty about those decisions. We examined how optimally 124 participants used an uncertain response on near-threshold trials of a psychophysical task. All participants showed overconfidence, but women tended to be more adaptive than men in responsiveness to uncertainty. Participants who responded to uncertainty most optimally exhibited more cognitive failures, fewer attention deficit and hyperactivity disorder symptoms, greater need for closure, better attention scanning skills, but larger effects of Stroop-task incongruity compared with participants who were least optimal in responsiveness to uncertainty. These data suggest that response competition might provide a mechanism for the cognitive experience of uncertainty.
A Novel, Robust, and Portable Platform for Magnetoencephalography using Optically Pumped Magnetometers

Abstract:Magnetoencephalography (MEG) measures brain function via assessment of magnetic fields generated by neural currents. Conventional MEG uses superconducting sensors, which place significant limitations on performance, practicality, and deployment; however, the field has been revolutionised in recent years by the introduction of optically-pumped-magnetometers (OPMs). OPMs enable measurement of the MEG signal without cryogenics, and consequently the conception of ‘OPM-MEG’ systems which ostensibly allow increased sensitivity and resolution, lifespan compliance, free subject movement, and lower cost. However, OPM-MEG remains in its infancy with limitations on both sensor and system design. Here, we report a new OPM-MEG design with miniaturised and integrated electronic control, a high level of portability, and improved sensor dynamic range (arguably the biggest limitation of existing instrumentation). We show that this system produces equivalent measures when compared to an established instrument; specifically, when measuring task-induced beta-band, gamma-band and evoked neuro-electrical responses, source localisations from the two systems were highly comparable and temporal correlation was >0.7 at the individual level and >0.9 for groups. Using an electromagnetic phantom, we demonstrate improved dynamic range by running the system in background fields up to 8 nT. We show that the system is effective in gathering data during free movement (including a sitting-to-standing paradigm) and that it is compatible with simultaneous electroencephalography (EEG – the clinical standard). Finally, we demonstrate portability by moving the system between two laboratories. Overall, our new system is shown to be a significant step forward for OPM-MEG technology and offers an attractive platform for next generation functional medical imaging.

Abstract:Optically-pumped magnetometers (OPMs) are highly sensitive, compact magnetic field sensors, which offer a viable alternative to cryogenic sensors (superconducting quantum interference devices - SQUIDs) for magnetoencephalography (MEG). With the promise of a wearable system that offers lifespan compliance, enables movement during scanning, and provides higher quality data, OPMs could drive a step change in MEG instrumentation. However, this potential can only be realised if background magnetic fields are appropriately controlled, via a combination of optimised passive magnetic screening (i.e. enclosing the system in layers of high-permeability materials), and electromagnetic coils to further null the remnant magnetic field. In this work, we show that even in an OPM-optimised passive shield with extremely low (<2 nT) remnant magnetic field, head movement generates significant artefacts in MEG data that manifest as low-frequency interference. To counter this effect we introduce a magnetic field mapping technique, in which the participant moves their head to sample the background magnetic field using a wearable sensor array; resulting data are compared to a model to derive coefficients representing three uniform magnetic field components and five magnetic field gradient components inside the passive shield. We show that this technique accurately reconstructs the magnitude of known magnetic fields. Moreover, by feeding the obtained coefficients into a bi-planar electromagnetic coil system, we were able to reduce the uniform magnetic field experienced by the array from a magnitude of 1.3±0.3 nT to 0.29±0.07 nT. Most importantly, we show that this field compensation generates a five-fold reduction in motion artefact at 0‒2 Hz, in a visual steady-state evoked response experiment using 6 Hz stimulation. We suggest that this technique could be used in future OPM-MEG experiments to improve the quality of data, especially in paradigms seeking to measure low-frequency oscillations, or in experiments where head movement is encouraged.

Abstract:We describe a multichannel magnetoencephalography (MEG) system that uses optically pumped magnetometers (OPMs) to sense the magnetic fields of the human brain. The system consists of an array of 20 OPM channels conforming to the human subject's head, a person-sized magnetic shield containing the array and the human subject, a laser system to drive the OPM array, and various control and data acquisition systems. We conducted two MEG experiments: auditory evoked magnetic field and somatosensory evoked magnetic field, on three healthy male subjects, using both our OPM array and a 306-channel Elekta-Neuromag superconducting quantum interference device (SQUID) MEG system. The described OPM array measures the tangential components of the magnetic field as opposed to the radial component measured by most SQUID-based MEG systems. Herein, we compare the results of the OPM- and SQUID-based MEG systems on the auditory and somatosensory data recorded in the same individuals on both systems.

Abstract:Optically pumped magnetometers (OPMs) are compact and lightweight sensors that can measure magnetic fields generated by current flow in neuronal assemblies in the brain. Such sensors enable construction of magnetoencephalography (MEG) instrumentation, with significant advantages over conventional MEG devices including adaptability to head size, enhanced movement tolerance, lower complexity and improved data quality. However, realising the potential of OPMs depends on our ability to perform system calibration, which means finding sensor locations, orientations, and the relationship between the sensor output and magnetic field (termed sensor gain). Such calibration is complex in OPMMEG since, for example, OPM placement can change from subject to subject (unlike in conventional MEG where sensor locations or orientations are fixed). Here, we present two methods for calibration, both based on generating well-characterised magnetic fields across a sensor array. Our first device (the HALO) is a head mounted system that generates dipole like fields from a set of coils. Our second (the matrix coil (MC)) generates fields using coils embedded in the walls of a magnetically shielded room. Our results show that both methods offer an accurate means to calibrate an OPM array (e.g. sensor locations within 2 mm of the ground truth) and that the calibrations produced by the two methods agree strongly with each other. When applied to data from human MEG experiments, both methods offer improved signal to noise ratio after beamforming suggesting that they give calibration parameters closer to the ground truth than factory settings and presumed physical sensor coordinates and orientations. Both techniques are practical and easy to integrate into real world MEG applications. This advances the field significantly closer to the routine use of OPMs for MEG recording.

Abstract:Wearable magnetoencephalography based on optically pumped magnetometers (OPM-MEG) offers non-invasive and high-fidelity measurement of human brain electrophysiology. The flexibility of OPM-MEG also means it can be deployed in participants of all ages and permits scanning during movement. However, the magnetic fields generated by neuronal currents - which form the basis of the OPM-MEG signal - are much smaller than environmental fields, and this means measurements are highly sensitive to interference. Further, OPMs have a low dynamic range, and should be operated in near-zero background field. Scanners must therefore be housed in specialised magnetically shielded rooms (MSRs), formed from multiple layers of shielding material. The MSR is a critical component, and current OPM-optimised shields are large (>3 m in height), heavy (>10,000 kg) and expensive (with up to 5 layers of material). This restricts the uptake of OPM-MEG technology. Here, we show that the application of the Maxwell filtering techniques signal space separation (SSS) and its spatiotemporal extension (tSSS) to OPM-MEG data can isolate small signals of interest measured in the presence of large interference. We compare phantom recordings and MEG data from a participant performing a motor task in a state-of-the-art 5-layer MSR, to similar data collected in a lightly shielded room: application of tSSS to data recorded in the lightly shielded room allowed accurate localisation of a dipole source in the phantom and neuronal sources in the brain. Our results point to future deployment of OPM-MEG in lighter, cheaper and easier-to-site MSRs which could catalyse widespread adoption of the technology.

Abstract:Optically pumped magnetometers (OPMs) can capture brain activity but are susceptible to magnetic noise. The objective of this study was to evaluate a novel methodology used to reduce magnetic noise in OPM measurements. A portable magnetoencephalography (MEG) prototype was developed with OPMs. The OPMs were divided into primary sensors and reference sensors. For each primary sensor, a synthetic gradiometer (SG) was constructed by computing a secondary sensor that simulated noise with signals from the reference sensors. MEG data from a phantom with known source signals and six human participants were used to assess the efficacy of the SGs. Magnetic noise in the OPM data appeared predominantly in a low frequency range (<4 Hz) and varied among OPMs. The SGs significantly reduced magnetic noise (p < 0.01), enhanced the signal-to-noise ratio (SNR) (p < 0.001) and improved the accuracy of source localization (p < 0.02). The SGs precisely revealed movement-evoked magnetic fields in MEG data recorded from human participants. SGs provided an effective method to enhance SNR and improve the accuracy of source localization by suppressing noise. Software-simulated SGs may provide new opportunities regarding the use of OPM measurements in various clinical and research applications, especially those in which movement is relevant.

Abstract:Magnetoencephalography (MEG) non-invasively provides important information about human brain electrophysiology. The growing use of optically pumped magnetometers (OPM) for MEG, as opposed to fixed arrays of cryogenic sensors, has opened the door for innovation in system design and use cases. For example, cryogenic MEG systems are housed in large shielded rooms to provide sufficient space for the system dewar. Here, we investigate the performance of OPM recordings inside of a cylindrical shield with a 1×2 m footprint. The efficacy of shielding was measured in terms of field attenuation and isotropy, and the value of post hoc noise reduction algorithms was also investigated. Localization accuracy was quantified for 104 OPM sensors mounted on a fixed helmet array based on simulations and recordings from a bespoke current dipole phantom. Passive shielding attenuated direct current (DC) and power line (60 Hz) fields by approximately 60 dB, and all other frequencies by 80-90 dB. Post hoc noise reduction provided an additional 5-15 dB attenuation. Substantial field anisotropy remained in the volume encompassing the sensor array. The consistency of the anisotropy over months suggests that a field nulling solution could be readily applied. A current dipole phantom generating source activity at an appropriate magnitude for the human brain generated field fluctuations on the order of 0.5-1 pT. Phantom signals were localized with 3 mm localization accuracy and no significant bias in localization was observed, which is in line with performance for cryogenic and OPM MEG systems. This validation of the performance of a small footprint MEG system opens the door for lower cost MEG installations in terms of raw materials and facility space, as well as mobile imaging systems (e.g., truck-based). Such implementations are relevant for global adoption of MEG outside of highly resourced research and clinical institutions.

Abstract:ObjectiveCurrent commercial magnetoencephalography (MEG) systems detect neuro-magnetic signals using superconducting quantum interferometers devices (SQUIDs), which require liquid helium as cryogen and have many limitations during operation. In contrast, optical pumped magnetometers (OPMs) technology provides a promising alternative to conventional SQUID-MEG. OPMs can operate at room temperature, offering benefits such as flexible deployment and lower costs. However, the validation of OPM-MEG has primarily been conducted on small sample sizes and specific regions of interest in the brain, lacking comprehensive validation for larger sample sizes and assessment of whole-brain. ApproachWe recruited 100 participants, including healthy and neurological disorders individuals. Whole-brain OPM-MEG and SQUID-MEG data were recorded sequentially during auditory (n = 50) and visual (n = 50) stimulation experiments. By comparing the task-evoked responses of the two systems, we aimed to validate the performance of the next-generation OPM-MEG. Main resultsThe results showed that OPM-MEG enhanced the amplitude of task-related responses and exhibited similar magnetic field patterns and neural oscillatory activity as SQUID-MEG. There was no difference in the task-related latencies measured by the two systems. The signal-to-noise ratio was lower for the OPM-MEG in the auditory experiment, but did not differ in the visual experiment, suggesting that the results may be task-dependent. SignificanceThese results demonstrate that OPM-MEG, as an alternative to traditional SQUID-MEG, shows superior response amplitude and comparable performance in capturing brain dynamics. This study provides evidence for the effectiveness of OPM-MEG as a next-generation neuroimaging technique.

Abstract:Optically pumped magnetometers (OPMs) offer a new wearable means to measure magnetoencephalography (MEG) signals, with many advantages compared to conventional systems. However, OPMs are an emerging technology, thus characterizing and replicating MEG recordings is essential. Using OPM-MEG and SQUID-MEG, this study investigated evoked responses, oscillatory power, and functional connectivity during emotion processing in 20 adults, to establish replicability across the two technologies. Five participants with dental fixtures were included to assess the validity of OPM-MEG recordings in those with irremovable metal. Replicable task-related evoked responses were observed in both modalities. Similar patterns of oscillatory power to faces were observed in both systems. Increased connectivity was found in SQUID-versus OPM-MEG in an occipital and parietal anchored network. Notably, high quality OPM-MEG data were retained in participants with metallic fixtures, from whom no useable data were collected using conventional MEG.

Abstract:Multimodal neuroimaging plays an important role in neuroscience research. Integrated noninvasive neuroimaging modalities, such as magnetoencephalography (MEG), electroencephalography (EEG) and functional near-infrared spectroscopy (fNIRS), allow neural activity and related physiological processes in the brain to be precisely and comprehensively depicted, providing an effective and advanced platform to study brain function. Noncryogenic optically pumped magnetometer (OPM) MEG has high signal power due to its on-scalp sensor layout and enables more flexible configurations than traditional commercial superconducting MEG. Here, we integrate OPM-MEG with EEG and fNIRS to develop a multimodal neuroimaging system that can simultaneously measure brain electrophysiology and hemodynamics. We conducted a series of experiments to demonstrate the feasibility and robustness of our MEG-EEG-fNIRS acquisition system. The complementary neural and physiological signals simultaneously collected by our multimodal imaging system provide opportunities for a wide range of potential applications in neurovascular coupling, wearable neuroimaging, hyperscanning and brain-computer interfaces.
Modelling, simulation and identification of a mobile concrete pump

Abstract:Due to the light-weight construction of modern large-scale manipulators used, e.g., in mobile concrete pumps, the elasticity of the construction elements plays a significant role in the dynamic behaviour of the system. Therefore, current research is concerned with control strategies for active damping of elastic vibrations and trajectory planning. For this purpose, tailored mathematical models are required. Apart from the mathematical modelling, the identification of the model parameters constitutes a challenging task. This is mainly due to the large number of parameters to be identified and, considering the large scale, due to the fact that the boom movement cannot be measured by means of standard sensors. This paper presents a systematic approach for the mathematical modelling and identification of hydraulically actuated large-scale manipulators. The feasibility of the overall approach is demonstrated by means of measurement results of a mobile concrete pump.

Abstract:To address the demands in healthcare and industrial settings for spatially resolved magnetic imaging, we present a modular optically pumped magnetometer (OPM) system comprising a multi-sensor array of highly sensitive quantum magnetometers. This system is designed and built to facilitate fast prototyping and testing of new measurement schemes by enabling quick reconfiguration of the self-contained laser and sensor modules as well as allowing for the construction of various array layouts with a shared light source. The modularity of this system facilitates the development of methods for managing high-density arrays for magnetic imaging. The magnetometer sensitivity and bandwidth are first characterised in both individual channel and differential gradiometer configurations before testing in a real-world magnetoencephalography environment by measuring alpha rhythms from the brain of a human participant. We demonstrate the OPM system in a first-order axial gradiometer configuration with a magnetic field gradient sensitivity of 10 fT/cm/√Hz. Bandwidths exceeding 200 Hz were achieved for two independent modules. The system's increased temporal resolution allows for the measurement of spinal cord signals, which we demonstrate by using phantom signal trials and comparing with an existing commercial sensor.

Non-rigid-registration-based Positioning and Labelling of Triaxial OPMs on a Flexible Cap for Wearable Magnetoencephalography.

Abstract:Background: Recent advances in highly sensitive miniaturized optically pumped magnetometers (OPMs) have enabled the development of wearable magnetoencephalography (MEG) offering great flexibility in experimental setting. The OPM array for wearable MEG is typically attached to a flexible cap and exhibits a variable spatial layout across different subjects, which imposes challenges concerning the efficient positioning and labelling of OPMs.New method: A pair of reflective markers are affixed to each triaxial OPM sensor above its cable to determine its location and sensitive axes. A non-rigid registration of optically digitized marker locations with a pre-labelled template of marker locations is performed to map newly digitized markers to OPMs.Results: The positioning and labelling of 66 OPM sensors could be completed within 35 s. Across ten experiments, all OPMs were accurately labelled, and the mean test-retest errors were 0.48 mm for sensor locations and 0.20 degree for sensitive axes. By combining six OPMs' positions with their respective recordings, magnetic dipoles inside a phantom were located with a mean error of 5.5 mm, and the best fitted dipole for MEG with auditory stimuli presented was located on a subject's primary auditory cortex. Comparison with existing methods: The proposed method reduces the reliance on error-prone and laborious manual operations inherent in existing methods, therefore significantly improving the efficiency of OPM positioning and labelling on a flexible cap.Conclusion: We developed a method for the precise and rapid positioning and labelling triaxial OPMs on a flexible cap, thereby facilitating the practical implementation of wearable OPM-MEG.

Abstract:MagnetoEncephaloGraphy (MEG) provides a measure of electrical activity in the brain at a millisecond time scale. From these signals, one can non-invasively derive the dynamics of brain activity. Conventional MEG systems (SQUID-MEG) use very low temperatures to achieve the necessary sensitivity. This leads to severe experimental and economical limitations. A new generation of MEG sensors is emerging: the optically pumped magnetometers (OPM). In OPM, an atomic gas enclosed in a glass cell is traversed by a laser beam whose modulation depends on the local magnetic field. MAG4Health is developing OPMs using Helium gas (4He-OPM). They operate at room temperature with a large dynamic range and a large frequency bandwidth and output natively a 3D vectorial measure of the magnetic field. In this study, five 4He-OPMs were compared to a classical SQUID-MEG system in a group of 18 volunteers to evaluate their experimental performances. Considering that the 4He-OPMs operate at real room temperature and can be placed directly on the head, our assumption was that 4He-OPMs would provide a reliable recording of physiological magnetic brain activity. Indeed, the results showed that the 4He-OPMs showed very similar results to the classical SQUID-MEG system by taking advantage of a shorter distance to the brain, despite having a lower sensitivity.

However, MEG excels in spatial resolution, enabling it to capture even the subtlest and weakest brain signals for brain-computer interfaces (BCIs). Leveraging MEG's capabilities, specifically with optically pumped magnetometers (OPM-MEG), proves to be a promising avenue for advancing MEG-BCIs, owing to its exceptional sensitivity and portability. This study harnesses the power of high-frequency steady-state visual evoked fields (SSVEFs) to build an MEG-BCI system that is flickering-imperceptible, user-friendly, and highly accurate. Approach. We have constructed a nine-command BCI that operates on high-frequency SSVEF (58–62 Hz with a 0.5 Hz interval) stimulation. We achieved this by placing the light source inside and outside the magnetic shielding room, ensuring compliance with non-magnetic and visual stimulus presentation requirements. Five participants took part in offline experiments, during which we collected six-channel multi-dimensional MEG signals along both the vertical (Z-axis) and tangential (Y-axis) components. Our approach leveraged the ensemble task-related component analysis algorithm for SSVEF identification and system performance evaluation. Main Results. The offline average accuracy of our proposed system reached an impressive 92.98% when considering multi-dimensional conjoint analysis using data from both the Z and Y axes. Our method achieved a theoretical average information transfer rate (ITR) of 58.36 bits min −1 with a data length of 0.7 s, and the highest individual ITR reached an impressive 63.75 bits min −1 . Significance. This study marks the first exploration of high-frequency SSVEF-BCI based on OPM-MEG. These results underscore the potential and feasibility of MEG in detecting subtle brain signals, offering both theoretical insights and practical value in advancing the development and application of MEG in BCI systems.

Abstract:Magnetoencephalography (MEG) allows the non-invasive measurement of brain activity at millisecond precision combined with localization of the underlying generators. So far, MEG-systems consisted of superconducting quantum interference devices (SQUIDS), which suffer from several limitations. Recent technological advances, however, have enabled the development of novel MEG-systems based on optically pumped magnetometers (OPMs), offering several advantages over conventional SQUID-MEG systems. Considering potential improvements in the measurement of neuronal signals as well as reduced operating costs, the application of OPM-MEG systems for clinical neuroscience and diagnostic settings is highly promising. Here we provide an overview of the current state-of-the art of OPM-MEG and its unique potential for translational neuroscience. First, we discuss the technological features of OPMs and benchmark OPM-MEG against SQUID-MEG and electroencephalography (EEG), followed by a summary of pioneering studies of OPMs in healthy populations. Key applications of OPM-MEG for the investigation of psychiatric and neurological conditions are then reviewed. Specifically, we suggest novel applications of OPM-MEG for the identification of biomarkers and circuit deficits in schizophrenia, dementias, movement disorders, epilepsy, and neurodevelopmental syndromes (autism spectrum disorder and attention deficit hyperactivity disorder). Finally, we give an outlook of OPM-MEG for translational neuroscience with a focus on remaining methodological and technical challenges.

Abstract:We present a novel four-channel OPM sensor for magnetoencephalography that utilizes a two-color pump/probe scheme on a single optical axis. We characterize its performance across 18 built sensor modules. The new sensor implements several improvements over our previously developed sensor including lower vapor-cell operating temperature, improved probe-light detection optics, and reduced optical power requirements. The sensor also has new electromagnetic field coils on the sensor head which are designed using stream-function-based current optimization. We detail the coil design methodology and present experimental characterization of the coil performance. The magnetic sensitivity of the sensor is on average 12.3 fT/rt-Hz across the 18 modules while the average gradiometrically inferred sensitivity is about 6.0 fT/rt-Hz. The sensor 3-dB bandwidth is 100 Hz on average. The on-sensor coil performance is in good agreement with the simulations.
