Abstract:In vivo, endothelial cells are permanently subjected to dynamic cyclic stretch and adapt to it through the release of vasoactive substances. Among them, reactive oxygen species (ROS) and nitric oxide (NO) are indispensable redox molecules, the contents of which and their ratio are closely implicated with endothelial redox homeostasis. However, simultaneous and quantitative monitoring of ROS and NO release in endothelial mechanotransduction remains a great challenge. Herein, a stretchable electrochemical device is developed with a dual electrode based on gold nanotubes decorated with uniform and tiny platinum nanoparticles. This hybrid nanostructure endows the sensor with high sensitivity toward both hydrogen peroxide (H2O2) (as the most stable ROS) and NO electrooxidation. Importantly, the two species can be well discriminated by applying different potentials, which allows simultaneous monitoring of H2O2 and NO release in stretch-induced endothelial mechanotransduction by the same device. The results of quantitative analysis suggest that endothelial redox homeostasis and its alteration are strongly related to vascular biomechanical and biochemical milieus. Further investigation reveals that the interplay of ROS and NO signaling has an important role in the regulation of endothelial redox state. This work will greatly facilitate the deep understanding of the molecular mechanism of endothelial dysfunction and vascular disorder.

Abstract:Mechanotransduction is the essential process that cells convert mechanical force into biochemical responses, and electrochemical sensor stands out from existing techniques by providing quantitative and real-time information about the biochemical signals during cellular mechanotransduction. However, the intracellular biochemical response evoked by mechanical force has been poorly monitored. In this paper, we report a method to apply local stretch on single cell and simultaneously monitor the ensuing intracellular biochemical signals. Specifically, a ferromagnetic micropipette was fabricated to locally stretch a single cell labeled with Fe 3 O 4 nanoparticles under the external magnetic field, and the SiC@Pt nanowire electrode (SiC@Pt NWE) was inserted into the cell to monitor the intracellular hydrogen peroxide (H 2 O 2 ) production induced by the local stretch. As a proof of concept, this work quantitatively investigated the elevated amount of H 2 O 2 levels in single endothelial cell under different stretching amplitudes. This work puts forward a new research modality to manipulate and monitor the mechanotransduction at the single-cell level.

Abstract:Various electrochemical biosensors have been developed for direct and real-time recording of biomolecules released from living cells. However, since these traditional electrodes are commonly rigid and nonflexible, in situ monitoring of biochemical signals while cell deformation occurs remains a great challenge. Herein, we report a facile approach for the development of a stretchable and transparent electrochemical cell-sensing platform based on Au nanostructures (nano-Au) and carbon nanotube (CNT) films embedded in PDMS (nano-Au/CNTs/PDMS). The sandwich-like nanostructured network of nano-Au/CNTs endows the sensor with excellent mechanical stability and electrochemical performance. The obtained nano-Au/CNTs/PDMS electrode displays desired performance for H2O2 detection with a wide linear range (20 nM–25.8 μM) and low detection limit (8 nM). Owing to good biocompatibility and flexibility, HeLa and human umbilical vein endothelial cells can be directly cultured on the electrode and real-time monitoring of H2O2 release from cells under their stretched state was realized. Development of a nanozyme-based electrochemical catalyst for real-time biomarker sensing of superoxide and nitric oxide anions released from living cells and exogenous donors

Abstract:Developing quantitative biosensors of superoxide (O2•-) and nitric oxide (NO) anion is crucial for pathological research. As of today, the main challenge for electrochemical detection is to develop high-selectivity nano-mimetic materials to replace natural enzymes. In this study, the dendritic-like morphological structure of silver organic framework (Ag-MOF) was successfully synthesized via a solvothermal strategy. Owing to the introduction of polymeric composites results in improved electrical conductivity and catalytic activity, which promotes mass transfer and leads to faster electron efficiency. For monitoring the electrochemical signals of O2•- and NO, the Ag-MOF electrode substrate was produced by drop-coating, and composites were designed by cyclic voltammetric potential cycles. The designed electrode substrates demonstrate high sensitivity, wide linear concentrations of 1 nM-1000 μM and 1 nM-850 μM, and low detection limits of 0.27 nM and 0.34 nM (S/N = 3) against O2•- and NO. Aside from that, the sensor successfully monitored the cellular release of O2•-, and NO from HepG2 and RAW 264.7 living cells and has the potential to monitor exogenous NO release from donors of Diethylamine (DEA)-NONOate and sodium nitroprusside (SNP). Additionally, the developed system was applied to the analysis of O2•- and NO in real biological fluid samples, and the results were good satisfactory (94.10-99.57 ± 1.23%). The designed system provides a novel approach to obtaining a good electrochemical biosensor platform that is highly selective, stable, and flexible. Finally, the proposed method provides a quantitative way to follow the dynamic changes in O2•- and NO in biological systems.

Abstract:Recently, two-dimensional metal-organic framework (2D MOF) nanosheets have drawn a lot of attention on account of their various advantages, like ultrathin thickness, ultralarge specific surface area, abundant accessible active sites, favorable solution dispersion properties, and ease of designability. However, until now, it is still difficult to produce 2D MOF nanosheets in large scale, which hinders the practical applications of 2D MOF nanosheets. Here, for the first time, we introduced a novel shear exfoliation method to prepare scalable 2D MOF nanosheets by using a commercial blender. As a proof of concept, we used two kinds of layer-structured MOFs (ELM-12, Cu(bpy)(2)(OTf)(2), bpy = 4,4-bipyridine, OTf = trifluoromethanesulfonate and Zn-2(bim)(4), bim = benzimidazole) as samples to prepare MOF nanosheets. The thickness of the two kinds of MOF nanosheets obtained is 3-5 nm. Notably, the exfoliated MOF nanosheet (ELM-12) shows improved electrochemical catalytic activity compared with its bulk counterpart. Based on this, an ELM-12 nanosheet-based flexible sensor was developed for detecting superoxide anions (O-2(center dot-)) released from cancer cells. The fabricated flexible sensor displays excellent sensitivity, selectivity, flexibility, stability, and reproducibility.

Abstract:Real-time sensing of reactive oxygen species (ROS) and timely scavenging of excessive ROS in physiological environments are critically important in the diagnosis and prevention of ROS-related diseases. To solve the mismatch problem between conventional rigid ROS biosensors and biological tissues in terms of both modulus and composition, here, we present a flexible ferrocene-based hydrogel biosensor designed for oxidative stress detection and antioxidation treatment. The hydrogel was fabricated through a supramolecular assembly of ferrocene-grafted polyethylenimine (PEI-Fc), sodium alginate (SA), and polyvinyl alcohol (PVA). Multiple non-covalent interactions, including electrostatic interactions between PEI-Fc and SA, hydrophobic interactions and π–π stacking among ferrocene groups, and the PVA crystalline domain, synergistically improve the mechanical properties of the PVA/SA/PEI-Fc hydrogel. The flexible PVA/SA/PEI-Fc hydrogel biosensor exhibited a broad detection range for hydrogen peroxide (H 2 O 2 ), from 0 to 120 μM, using the differential pulse voltammetry method. Furthermore, the hydrogel demonstrated effective ROS scavenging and oxygen generation performance, desirable biocompatibility, and satisfactory antibacterial activity, making it suitable for biological interfaces. In vitro studies revealed that the PVA/SA/PEI-Fc hydrogel could monitor H 2 O 2 concentration in the proximity of inflammatory cells, and effectively scavenge ROS to protect cells from oxidative stress damage. This all-in-one multifunctional hydrogel, integrating both sensing and treatment functions, holds great promise for clinical applications in the diagnosis and management of ROS-related diseases.

Abstract:Comprehensive Summary Electrochemical sensing faces huge challenges in characterizing the transient release of biochemical molecules from deformed cells, due to the severely mechanical mismatch between rigid electrodes and soft cells. In recent years, the emergence of stretchable electrochemical sensors has made a breakthrough by complying with the deformation of living cells and simultaneous monitoring of mechanically evoked biochemical signals. This review first summarizes two fundamental strategies for the fabrication of stretchable electrodes from the points of structure and material. Next, recent progresses in construction of functionalized interface to improve the performance of stretchable electrochemical sensors are presented. Then, the application of stretchable electrochemical sensors in real‐time monitoring of biomolecules released by mechanically sensitive cells is introduced. Abstract:Diabetes is a metabolic syndrome associated with hyperglycemia, hypertension, atherosclerosis, and endothelial dysfunction. Applying the mechanical stretch on cells to simulate blood circulation while monitoring the cell glucose metabolism in a high-glucose environment is important for better comprehension of the underlying mechanisms of atherosclerosis caused by diabetes. Herein, we developed a facile strategy integrating zeolitic imidazolate framework-8-encapsulated glucose oxidase () and an gold (Au) stretchable electrode (Au SE) to construct a flexible and stretchable glucose sensor (/Au SE) for investigating the glucose metabolism mechanism of stretched endothelial cells in hyperglycemia. The encapsulation of GOx with ZIF-8 prevents the aggregation and detachment of GOx from the sensing interface and endows the biosensor with high stability. Additionally, the Au SE with inherent stretchability can act as an integrated platform for mechanical stimulation as well as for transient signal sensing during the mechanotransduction process. Moreover, this flexible and stretchable glucose sensor is successfully used for monitoring the glucose metabolism of mechanically stimulated cells in hyperglycemia, and it was found for the first time that the glucose utilization ability of cells under static conditions is higher than that in the stretched state. Abstract:Flexible biomimetic sensors have encountered a bottleneck of sensitivity and durability, as the sensors must directly work within complex body fluid with ultra-trace biomarkers. In this work, a wearable electrochemical sensor on a modified silk fibroin substrate is developed using gold nanoparticles hosted into N-doped porous carbonizated silk fibroin (AuNPs@CSF) as active materials. Taking advantage of the inherent biocompatibility and flexibility of CSF, and the high stability and enzyme-like catalytic activity of AuNPs, AuNPs@CSF-based sensor exhibits durable stability and superior sensitivity to monitor H2O2 released from cancer cell (4T1) and glucose in sweat. The detection limits for H2O2 and glucose are low to be 1.88 μM and 23 μM respectively, and the sensor can be applied in succession within 30 days at room temperature. Further, physical cross-linking of polyurethane (PU) with SF well matches with the skin tissue mechanically and provides a flexible, robust and stable electrode-tissue interface. AuNPs@CSF is applied successfully for wearable electrochemical monitoring of glucose in human sweat.The present AuNPs@CSF will possess a potential application in clinical diagnosing of H2O2- or glucose-related diseases in future.

Abstract:A novel flexible electrochemical sensor based on porous carbon nanosheets (PCNSs) nanozyme has been constructed for in situ and real-time monitoring of H 2 O 2 released by cells. The PCNSs are prepared with the integration of thermal transformation, thermal activation and sonochemical exfoliation by using zeolitic imidazolate frameworks as template. The PCNSs exhibit high electrical conductivity, electrochemical activity and peroxidase-like catalytic properties, which is beneficial to H 2 O 2 assay. With the transfer printing method, the flexible electrochemical sensor is obtained, which has excellent performances for H 2 O 2 electrochemical detecting with wide linear range from 1 μM to 20 mM and a low detection limit of 0.76 μM. Owing to the great biocompatibility, the flexible sensor guarantees the growth of living cells for 72 h and realizes in situ and real-time monitoring the release of H 2 O 2 from HeLa cells. The strategy of porous nanozyme preparation and flexible sensor construction provided a promising way for in situ and real-time assay of small molecules in the cellular microenvironment.

Abstract:Endothelial cells are sensitive to mechanical force and can convert it into biochemical signals to trigger mechano-chemo-transduction. Although conventional techniques have been used to investigate the subsequent modifications of cellular expression after mechanical stimulation, the in situ and real-time acquiring the transient biochemical information during mechanotransduction process remains an enormous challenge. In this work, we develop a flexible and multi-functional three-dimensional conductive scaffold that integrates cell growth, mechanical stimulation, and electrochemical sensing by in situ growth of enokitake-like Au nanowires on a three-dimensional porous polydimethylsiloxane substrate. The conductive scaffold possesses stable and desirable electrochemical sensing performance toward nitric oxide under mechanical deformation. The prepared e-AuNWs/CC/PDMS scaffold exhibits a good electrocatalytic ability to NO with a linear range from 2.5 nM to 13.95 μM and a detection limit of 8 nM. Owing to the excellent cellular compatibility, endothelial cells can be cultured directly on the scaffold and the real-time inducing and recording of nitric oxide secretion under physiological and pathological conditions were achieved. This work renders a reliable sensing platform for real-time monitoring cytomechanical signaling during endothelial mechanotransduction and is expected to promote other related biological investigations based on three-dimensional cell culture.

Abstract:Developing effective strategies for the flexible control of fluid is vital for microfluidic electrochemical biosensing. In this study, a gigahertz (GHz) acoustic streaming (AS) based sonoelectrochemical system was developed to realize an on-chip surface modification and sensitive hydrogen peroxide (H 2 O 2 ) detection from living cells. The flexible and controlled fluid surrounding the electrochemical chip was optimized theoretically and applied in the sonoelectrochemical deposition of Au nanoparticles (AuNPs) first. Under the steady and fast flow stimulus of AS, AuNPs could be synthesized with a smaller and evener size distribution than the normal condition, allowing AuNPs to show an excellent peroxidase-like activity. Moreover, the AS also accelerated the mass transport of target molecules and improved the catalytic rate, leading to the enhancement of H 2 O 2 detection, with an extremely low detection limit of 32 nM and a high sensitivity of 4.34 μA/ (mM·mm 2 ). Finally, this system was successfully applied in tracking H 2 O 2 release from different cell lines to distinguish the cancer cells from normal cells. This study innovatively integrated the surface modification and molecules detection process on a chip, and also proposed a simple but sensitive platform for microfluidic biosensing application.

Abstract:In-situ determination of cell-secreted biomolecules is important for elucidating its regulation of signal transduction pathways, but detecting these low-level endogenous biomolecules is difficult. Herein, using H2O2 as the detection object, we developed an ultrasensitive enzyme-free ratiometric electrochemical sensing platform in a simple and low-cost method for in-situ determination of cell-secreted H2O2, in which ferrocene (Fc) acts as an internal reference probe and nanoflower Au-Co-MOF acts a detection probe. First, flower-like CoFc-MOF was synthesized in situ on flexible carbon cloth (CC) by a one-step hydrothermal method, and then Au-CoFc-MOF/CC was obtained by electrodeposition of gold nanoparticles. The electrochemical measurement results show that the presence of H2O2 increases the reduction peak signal of Co-MOF (−0.32 V), while that of Fc (+0.4 V) is almost unchanged. By taking full advantage of the obviously separated reduction potential of Fc and Co-MOF, the well-designed sensing platform can reflect the concentration of H2O2 based on the I H2O2 – I blank/I Fc value in the 0.5 μM–10 mM range, with a detection limit as low as 32 nM, which is ∼15.6 times lower than the value obtained from the single signal method. In addition, the sensing platform also demonstrates excellent selectivity, reproducibility, stability, and biocompatibility. More significantly, this ratiometric electrochemical sensing platform based on flexible substrate shows great application potential in the in-situ determination of cell-secreted H2O2, which provides a feasible strategy for detecting other active substances released from living cells in clinical diagnosis.

Abstract:A highly stretchable and self-adhesive wearable biosensor is prepared from nanozyme-catalyzed conductive hydrogels. The hydrogels are first prepared from a nanozyme (Ta–Ag) based on polyphenol (tannic acid, Ta) chelate with metal ions (silver ion, Ag+), which possesses peroxidase (POD)-like activity. After that, a huge number of free radicals are formed by the interaction between the Ta–Ag NPs and ammonium persulfate (APS), which initiate the hydrogel polymerization via a facile “one-pot” compositing strategy without ultraviolet (UV) or thermal treatment. The homogeneous distribution of nanozymes in the interpenetrating network of sodium alginate (SA) and polyacrylamide (PAAm) realizes a dermis-mimicking structure in the flexible conductor, named Ta–Ag–SA/PAAm. It simultaneously improves the mechanical properties and conductivity. Similar to the adhesion of mussels, the nanozymes contain abundant phenolic hydroxyl groups, favoring the adhesiveness of hydrogels. Due to the capability of changing conductivity with the strain, the obtained hydrogels are able to accurately monitor different amplitudes of human activities in real time. In addition, the Ta–Ag nanozyme exhibits a good bacterial adhesion capability and thereby enhances the bactericidal efficiency of Ag. Based on the high performance of sensing and antibacterial activity, the obtained Ta–Ag–SA/PAAm hydrogels can be a promising candidate for a broad range of applications, such as adhesive epidemic biosensors and wearable medical patches.

Abstract:Myopic loss aversion is the combination of a greater sensitivity to losses than to gains and a tendency to evaluate outcomes frequently. Two implications of myopic loss aversion are tested experimentally. 1. Investors who display myopic loss aversion will be more willing to accept risks if they evaluate their investments less often. 2. If all payoffs are increased enough to eliminate losses, investors will accept more risk. In a task in which investors learn from experience, both predictions are supported. The investors who got the most frequent feedback (and thus the most information) took the least risk and earned the least money.

Abstract:Reactive oxygen species (ROS) including the superoxide anion (O 2 •− ) are typically studied in cell cultures using fluorescent dyes, which provide only discrete single-point measurements. These methods lack the capabilities for assessing O 2 •− kinetics and release in a quantitative manner over long monitoring times. Herein, we present the fabrication and application of an electrochemical biosensor that enables real-time continuous monitoring of O 2 •− release in cell cultures for extended periods (> 8 h) using an O 2 •− specific microelectrode. To achieve the sensitivity and selectivity requirements for cellular sensing, we developed a biohybrid system consisting of superoxide dismutase (SOD) and Ti 3 C 2 T x MXenes, deposited on a gold microwire electrode (AuME) as O 2 •− specific materials with catalytic amplification through the synergistic action of the enzyme and the biomimetic MXenes-based structure. The biosensor demonstrated a sensitivity of 18.35 nA/μM with a linear range from 147 to 930 nM in a cell culture medium. To demonstrate its robustness and practicality, we applied the biosensor to monitor O 2 •− levels in human leukemia monocytic THP-1 cells upon stimulation with lipopolysaccharide (LPS). Using this strategy, we successfully monitored LPS-induced O 2 •− in THP-1 cells, as well as the quenching effect induced by the ROS scavenger N-acetyl- l -cysteine (NAC). The biosensor is generally useful for exploring the role of oxidative stress and longitudinally monitoring O 2 •− release in cell cultures, enabling studies of biochemical processes and associated oxidative stress mechanisms in cellular and other biological environments.

Abstract:Research on flexible biosensors is mainly concentrated on their application in acquiring information on physical signals including temperature, pH, and so on. However, there are hardly any studies on using flexible biosensors for assaying biomolecules and biomarkers caused diseases. Herein, a free-standing flexible sensor is designed for H 2 O 2 sensing and cancer biomarker assaying, in which reduced graphene oxide and carbon nanotubes (rGO-CNT) flexible hybrid film is first obtained through self-assembly, then MnO 2 nanoflowers and Co nanospheres are anchored on rGO-CNT by electrodeposition. rGO-CNT has good flexibility and conductivity. MnO 2 uniformly distributed on rGO-CNT has excellent catalytic performance towards H 2 O 2 . Co not only improve the conductivity, but also supply a second active material for H 2 O 2 sensing. Profiting from these advantages, MnO 2 –Co/rGO-CNT sensor shows an expanded detection range, high sensitivity, low detection limit, excellent repeatability, reproducibility, stability and selectivity. Surprisingly, it can maintain 100% of the original signal after folding 100 times, indicating deformation stability. Moreover, it can real-time monitor H 2 O 2 released by HepG2 cells. The strategy of introducing flexible substrate material and transition metals as well as transition metal compounds provides new insight into the design of flexible electrodes and greatly promote the development of real-time detection of cancer biomarkers.
