Abstract:A study was conducted to determine the adsorption/desorption of butachlor, myclobutanil and chlorpyrifos on five soils using a batch equilibration technique and to study the relationship between bioavailability to Allolobophora caliginosa and the adsorption/desorption of these three pesticides. The results showed that the adsorption/desorption processes of the tested compounds were mainly controlled by soil organic matter content (OM) and octanol/water-partitioning coefficient (Kow), and that the bioavailability of the pesticides was dependent on characteristics of pesticides, properties of soils, and uptake routes of earthworms. Bioconcentration of butachlor and myclobutanil was negatively correlated with Freundlich adsorption constant Kaf and Kdf. However, only a slightly positive correlation between bioconcentration and Kaf and Kdf was observed for chlorpyrifos due to its high affinity onto soil.
Adsorption and leaching of β-blockers in fluvo-aquic and black soil: Behavior characteristic and enantiomer selectivity

Abstract:β-blockers are widely used chiral pharmaceuticals to treat hypertension and cardiovascular diseases, which are ubiquitously detected in the water-soil environment. However, little is known about their biogeochemical behaviors and enantiomer selectivity during soil migration and transformation. In this study, the adsorption and leaching behaviors of β-blockers in fluvo-aquic soil and black soil were investigated. The adsorption of β-blockers was fit well by the Freundlich adsorption isotherm (R2 > 0.913) and the adsorption affinity of β-blockers decreased in the following order: propranolol (logarithm of Freundlich adsorption coefficient log Kf = 1.46-2.55) > atenolol (log Kf = 0.53-1.04) > sotalol (log Kf = 0.32-1.01). An increase in ionic strength and dissolved organic matter (DOM) inhibited their soil adsorption. Ionic change is the main driving force for adsorption. Besides, hydrophobic partitioning and hydrogen bonding played key roles in the adsorption of propranolol and atenolol, respectively. The leaching behaviors of β-blockers are related to their hydrophobicity. An increase in ionic strength enhanced the migration of β-blockers to deeper soil layers, and the presence of DOM accelerated the migration of sotalol and propranolol. The migration potential of β-blockers in black soil is lower than that in fluvo-aquic soil, which could be ascribed to the higher organic matter content and strong ion exchange ability of black soil. Further, more significant enantiomer selectivity of β-blockers was found in black soil (e.g. enantiomer fraction of atenolol = 0.61) than in fluvo-aquic soil (e.g. enantiomer fraction of atenolol = 0.53) during the leaching process. The microbial activity might influence the enantiomer selectivity of studied β-blockers during soil leaching.
Adsorption and Transport Characteristics of Two Exterior-Source Antibiotics in Some Agricultural Soils

Abstract:Antibiotics used in livestock production may be present in manure and slurry as the parent compound and/or metabolites.The environments may therefore be exposed to these antibiotics due to the application of animal manure to agricultural land.In order to understand the potential effects of long-term application of antibiotics-contained animal manure on agricultural soils,laboratory studies were conducted to characterize two typical antibiotic compounds(tylosin,C46H77NO17 and oxytetracycline,C22H24N2O9HCl) with regard to sorption and transport in seven agricultural soils,collected from the Changjiang and Zhujiang deltas of China.Both transfer and adsorption behaviors of the antibiotics were related with species of antibiotics and soil properties.Distribution coefficients(Kd values) determined by botch equilibrium method varied between 43L kg-1 and 3018L kg-1 for oxytetracyclin,and 2.3L kg-1 and 175L kg-1 for tylosin.Oxytetracyclin was more strongly sorbed in all the soils investigated than tylosin.The adsorption amount of antibiotic compounds on the soils was correlated positively with the contents of clay,organic matter and iron oxides.Soil column leaching experiments indicated that there were a significant difference in the transport of the two antibiotic compounds between sandy soil and clayey soil,which due to the difference in their respective sorption capabilities.Very low concentrations of the two antibiotic compounds were detected in the leachates from clayey soil column,indicating a much lower mobility of antibiotics in the clayey soil.However,significant amount of tylosin compound was recovered in the leachates from sandy soil,indicating that tylosin compound was not stable in sandy soil,and it had a high mobility in the soil.
Fate of beta blockers in aquatic-sediment systems: sorption and biotransformation

Abstract:The fate of beta blockers (atenolol, acebutolol, bisoprolol, celiprolol, metoprolol, nadolol, pindolol, propranolol, and sotalol) was studied in surface water-sediment systems. A new analytical method was developed to determine the beta blockers in sediments by LC-ESI-tandem MS detection. The relative recoveries in sediments ranged from 89 +/- 7% (acebutolol) to 102 +/- 3% (nadolol) using deuterated surrogate standards. Beta blockers were present with concentrations up to 86 ng/g (bisoprolol) in the sediments of small German streams containing an elevated percentage of treated wastewater. Biotransformation studies and sorption isotherms of the beta blockers were performed with two natural river sediments ("Burgen", "Dausenau") differing in organic carbon content and particle size distribution. Biotransformation of beta blockers in the surface water-sediment systems exhibited a low to high persistence with 90% disappearance (DT(90)) ranging from 0.4-10 d (pindolol, atenolol) to >100 d (sotalol, propranolol or celiprolol). For sorption studies neither NaN(3) addition nor autoclavation led to a complete mass balance of the beta blockers, probably due to biotransformation. Isotherms at 6 h (apparent equilibrium, measuring aqueous and sediment phase) fitted by the Freundlich equation show that sorption of all beta blockers to the Burgen sediment were linear or close to it (i.e., n-values between 0.93 and 1.13), while in the Dausenau sediment the sorptions were slightly non linear (i.e., n-values 0.77-0.91). In river water the sorbed fraction is negligible in comparison to the dissolved fraction. Nevertheless, beta blockers can be detected with concentrations up to 86 ng/g (bisoprolol) in sediments of small streams containing more than 50% treated wastewater.

Abstract:The presence of pharmaceuticals in agricultural soils, like amoxicillin (AMX) and ciprofloxacin (CIP), poses a significant environmental challenge with potential implications for ecosystems and human well-being. This study explores the simultaneous adsorption of AMX and CIP on crop soils and bio-adsorbents, focusing on competitive adsorption dynamics. Tests were conducted with varying pharmaceutical concentrations in six soils and three bio-adsorbents. CIP consistently exhibited higher adsorption than AMX, particularly at higher concentrations. In the binary system, AMX's adsorption exceeded the individual system at higher concentrations, implying a synergistic effect. Bio-adsorbents, especially pine bark and oak ash, displayed superior adsorption capacities compared to soils. Some soils exhibited enhanced adsorption and retention of both antibiotics simultaneously, aligning with the cooperative adsorption model. Freundlich's adsorption model described the competitive adsorption systems well. These findings have implications for addressing antibiotic contamination in agricultural ecosystems, offering insights into complex interactions in soil environments amid rising pharmaceutical concerns.

Abstract:The environmental risks of wide application and occurrence of antibiotics have attracted great concern but their environmental behaviors are still unclear. The fast development of nanotechnology also arise environmental concerns, one of which is that the discharge of nanomaterials into the environment may alter the environmental behavior and risks of various contaminants. This study conducted batch adsorption experiments of sulfamethoxazole (SMX), a widely studied antibiotic, on various carbon nanotubes (CNTs) and sediment particles. The SMX adsorption data on other natural adsorbents were also collected from literature. The surface area of CNTs is an important parameter controlling their adsorption to SMX. Other properties, including diameters and surface functional groups, were also observed to be important in regulating SMX adsorption. Inorganic mineral particles have comparable adsorption coefficients with other natural sorbents including soils, sediments and sludges, suggesting the contribution of inorganic fractions to SMX adsorption could not be ignored and the normalization of adsorption coefficient by organic carbon content should be avoided. The adsorption coefficients of SMX on CNTs were generally two orders of magnitude higher than other natural sorbents. The presence of CNTs in the environment, especially in waste water and solid waste of water treatment plant, could greatly alter SMX behavior as well as its risks.

Abstract:The widely utilization of pharmaceutical and personal care products (PPCPs) in the pharmaceutical therapies and agricultural husbandry has led to the world pollution. Thus, it is of great interest to study the mobility and fate of PPCPs in the environment. The aim of this study was to investigate the influence of heavy metal Pb(II), anionic surfactant sodium dodecylbenzenesulfonate (SDBS), nonionic surfactant Triton X-100 (TX-100) on the adsorption of representative PPCPs (tetracycline (TC) and oxytetracycline (OTC) on two types of adsorbents. Results showed that the adsorption isotherms of TC and OTC on two sediments were in good comparison with Freundlich equation. However compared to the sediment B without organic matter (OM), the sediment A with higher OM exhibited the higher adsorption affinity to TC and OTC. The presence of Pb (II) caused a significant increase on the TC and OTC adsorption to sediment A due to the formation of ternary complex between OM ligand groups and tetracycline ligand groups. However the influence of Pb (II) on TC and OTC adsorption on sediment B was minimal. The presence of SDBS increased TC and OTC adsorption on two sediments, which could be due to the significantly increasing OM content of two sediments via the SDBS adsorption. In contrast, the nonionic surfactant TX-100 caused a slight reduction on TC and OTC adsorption on two sediments even at high concentration.

Abstract:The fate of contaminants in soils and sediments is a function of a diverse number of biogeochemical and hydrologic processes, including sorption and bioavailability. Sorption of organic contaminants may play an important role in limiting contaminant bioavailability and thereby decreasing rates of in situ biodegradation. Sorption of toxic metals can also influence microbial processes through the influence of metal sorption on the toxicity of the metal. While there have been many macroscopic studies which have quantified the influence of sorption an bioavailability, a relatively limited number of studies have characterized the specific mechanism(s) controlling organic or inorganic interactions with soils and sediments and the subsequent influence of sorption on contaminant bioavailability We have coupled NMR spectroscopy, macroscopic sorption experiments, and microbial activity and the kinetics of substrate biodegradation in microcosms to explore the relationship between sorption and bioavailability of PAHs and cadmium in soils and sediments. We are using these results to explain field-scale contaminant bioavailability based upon the complexity of spatial patterns of mineral surface chemistry Spatial patterns of mineral surface chemistry within unconsolidated aquifer sediments are likely due to the importance of fluvial processes that sorted minerals during deposition and the hydrologic and biogeochemical processes that control inorganic and organic coatings on these minerals.

Abstract:Nowadays anticancer drugs (ADs), like other pharmaceuticals, are recognized as new emerging pollutants, meaning that they are not commonly monitored in the environment; however, they have great potential to enter the environment and cause adverse effects there. The current scientific literature highlights the problem of their presence in the aquatic environment by publishing more and more results on their analytics and ecotoxicological evaluation. In order to properly assess the risk associated with the presence of ADs in the environment, it is also necessary to investigate the processes that are important in understanding the environmental fate of these compounds. However, the state of knowledge on mobility of ADs in the environment is still very limited. Therefore, the main aim of our study was to investigate the sorption potential of two anticancer drugs, 5-fluorouracil (5-FU) and methotrexate (MTX), onto different soils. Special attention was paid to the determination of the influence of pH and ionic strength as well as presence of co-contaminants (cadmium (Cd2+) and another pharmaceutical—metoprolol (MET)) on the sorption of 5-FU and MTX onto soil. The obtained distribution coefficient values (Kd) ranged from 2.52 to 6.36 L·kg−1 and from 6.79 to 12.94 L·kg−1 for 5-FU and MTX, respectively. Investigated compounds may be classified as slightly or low mobile in the soil matrix (depending on soil). 5-FU may be recognized as more mobile in comparison to MET. It was proved that presence of other soil contaminants may strongly influence their mobility in soil structures. The investigated co-contaminant (MET) caused around 25-fold increased sorption of 5-FU, whereas diminished sorption of MTX. Moreover, the influence of environmental conditions such as pH and ionic strength on their sorption has been clearly demonstrated.

Abstract:A natural black soil (BS) was treated to obtain three individual soils referred to as removed organic matter (ROM), removed metal oxide (ROX) and humic acid (HA), and the sorption behaviors of tetracycline (TC) and heavy metals (Cu2+ and Cd2+) on BS and three treated soils were investigated to evaluate the role and contribution of different soil components (organic matter, clay and metal oxide). The three treated soils all showed stronger sorption capacities toward TC than BS, and the sorption amount of ROM, ROX, and HA for TC was greater than that of BS by 1.2 times, 2.3 times, and 3.3 times at an initial TC concentration of 25 mg L-1. Differently, the sorption capacity of BS for Cu2+ and Cd2+ was stronger than that of ROM. The multiple linear regression analysis suggested that soil organic matter made the greatest contribution toward the sorption of TC, whereas, metal oxide was the key component influencing the sorption of Cu2+ and Cd2+. The presence of Cu2+ enhanced the TC sorption of BS, ROM and ROX, but had a suppression effect on HA because of the competition of Cu2+. The presence of Cd2+ did not exert obvious effects on TC sorption of BS, but exhibited a suppression effect on TC sorption of ROM, ROX and HA, which was likely to relate to the surface potentials of the soils. The results in this study are expected to give an insight into the role of different soil components in the sorption and co-sorption of TC and heavy metal through a mathematical model and to reveal the sorption mechanism.
The fate of sulfamethoxazole in microcosms of the macrophyte Schoenoplectus californicus and its impact on microbial communities

Abstract:Sulfamethoxazole is a widely used antibiotic frequently found as an environmental pollutant. It can alter microbial communities and increase antibiotic resistance, becoming a public health risk. Constructed wetlands have the potential for removing sulfamethoxazole from polluted waters, but the role of different macrophytes in this process is not well understood. We investigated the fate of sulfamethoxazole and its effect on bacterial communities in microcosms containing Schoenoplectus californicus, an altitude-tolerant macrophyte. Within the first 10 h after introducing sulfamethoxazole (initial concentration 5 mg/L) to the microcosms, the concentration in the liquid phase significantly differed between microcosms with and without S. californicus. However, over the long term (15 and 30 days post-addition), the removal percentage (around 75%) in the liquid phase was not significantly influenced by S. californicus, indicating that sediments might be primarily responsible for removing the antibiotic. The presence of S. californicus promoted algae growth in the microcosms, and we determined that algae contributed to sulfamethoxazole removal from the liquid phase, likely through adsorption. Additionally, we characterized bacterial communities in the microcosm sediments via nanopore sequencing to identify changes following sulfamethoxazole addition. The relative abundance of Proteobacteria increased from 37-46% to 48-99% with the addition of the antibiotic. Conversely, the relative abundance of cyanobacteria decreased significantly after sulfamethoxazole was added (from 17 to 35% to less than 2%), suggesting it may serve as a biological marker for sulfamethoxazole pollution. In addition, the functional profile of the community was estimated from taxonomic diversity using PICRUST.

Abstract:Multiple antibiotics that are used in veterinary medicine coexist in soils, but their interaction and the effects on adsorption and desorption in soils have not been extensively studied. In this study, using batch experiments, we evaluated the adsorption and desorption of sulfadiazine (SDZ), tetracycline (TC), and norfloxacin (NFX) using four different soil aggregate size fractions and discovered that: (1) TC had the highest adsorption (76-98 %) and the lowest desorption in each tested system, whereas SDZ showed opposite adsorption and desorption ability, (2) the highest adsorption and the lowest desorption of all three tested antibiotics were observed with soil macroaggregates (250-2000 µm) in all the cases; in contrast, opposite adsorption and desorption ability were observed for soil clay (<53 µm), and (3) adsorption of each antibiotic was in the following order: single system (71-89 %) > binary system (56-84 %) > ternary system (50-78 %); however, desorption were in the reverse order. The Freundlich equation fitting and Brunauer-Emmett-Teller (BET) analysis further demonstrated that the adsorption competition between the tested antibiotics depended mainly on the specific surface area of each soil aggregate size fractions and its chemical properties. In conclusion, soil macroaggregates play a key role in the retention of antibiotics in soils, and the coexistence of multiple antibiotics greatly increases leaching risk.

Abstract:The long-term groundwater contamination risks posed by steroidal estrogens (SEs) in animal-manured agricultural soils are closely associated with the soil organic matter (SOM) content and composition. In this study, the bioavailability of estrone (E1) and 17β-estradiol (17β-E2) under different sorption mechanism in humic acids (HA1 and HA2) and humin (HM) extracted with sequential alkaline-extraction technique (SAET) were examined. These SOMs extracted by SAET showed various properties and sorption characteristics for SEs. The alkyl carbon and condensed SOM increased during SAET, but aromatic carbon decreased and the same trend for polarity. Quick sorption was the major SEs sorption mechanism on HA1 and HA2, which contributed more than 69%; whilst slow sorption rate was about 50% in soil and HM. The logKoc values were proportional to the TOC of SOM according to Freundlich fitting, and the sorption capacity of sorbent for E1 and 17β-E2 was related to the logKow values, indicating that the main mechanism controlling the SEs sorption was hydrophobic interaction. The larger micropore volume of HM and soil was more conducive to the micropore filling of SEs. Meanwhile, the specific sorption of SEs on condensed domain of SOM was the main reason for the strong desorption hysteresis and slow sorption in HM and soil. The SEs degradation rate was positively correlated with the contribution rate of quick adsorption and negatively correlated with the contribution rate of slow adsorption, indicating that the bioavailability of SEs sorbed by hydrophobic interaction was higher than that of micropore filling or specific sorption, which was also the reason for the low bioavailability of SEs in HM and soil. This work confirms the regulation of on-site SOM compositions and their properties on SEs sorption and bioavailability. Characterization of these details is crucial for the improved prediction of long-term risks to groundwater.

Abstract:Microplastics (MPs) and sulfamethoxazole (SMX) are emerging contaminants that are ubiquitous in the soil environment. In this study, we investigated MPs polymer type and soil environmental factor effects on SMX adsorption behavior in the soil system. Our results showed that MPs dosage affected the soil particles' SMX adsorption rate and capacity (Qe). Adding 1 % polystyrene (PS) increased the SMX adsorption rate significantly. The value of K1, which represented the adsorption rate, increased from 0.569 h-1 to 1.019 h-1. However, the addition of MPs reduced the soil's SMX equilibrium adsorption capacity slightly. Moreover, increasing salinity strength enhanced SMX adsorption capacity by MPs significantly. However, increasing calcium ions concentration decreased SMX adsorption in the MPs amended soil due to multivalent cationic bridging and competitive adsorption mechanisms. In addition, we observed that fulvic acid addition inhibited SMX adsorption. This study suggests that the addition of MPs reduced the adsorption of SMX in the soil slightly due to dilution effect. Meanwhile, changes in environmental factors also affected the adsorption behavior of SMX in soil amended with MPs.

Abstract:Pharmaceuticals are known for their great effects and applications in the treatment and suppression of various diseases in human and veterinary medicine. The development and modernization of science and technologies have led to a constant increase in the production and consumption of various classes of pharmaceuticals, so they pose a threat to the environment, which can be subjected to the sorption process on the solid phase. The efficiency of sorption is determined by various parameters, of which the physicochemical properties of the compound and the sorbent are very important. One of these parameters that determine pharmaceutical mobility in soil or sediment is the soil−water partition coefficient normalized to organic carbon (Koc), whose determination was the purpose of this study. The influence of organic matter, suspended in an aqueous solution of pharmaceutical (more precisely: cefdinir, memantine, and praziquantel), was studied for five different types of soil and sediment samples from Croatia. The linear, Freundlich, and Dubinin−Raduskevich sorption isotherms were used to determine specific constants such as the partition coefficient Kd, which directly describes the strength of sorbate and sorbent binding. The linear model proved to be the best with the highest correlation coefficients, R2 > 0.99. For all three pharmaceuticals, a positive correlation between sorption affinity described by Kd and Koc and the amount of organic matter was demonstrated.

Abstract:Supported lipid bilayers (SLBs) have contributed invaluable information about the physiochemical properties of cell membranes, but their compositional simplicity often limits the level of knowledge that can be gained about the structure and function of transmembrane proteins in their native environment. Herein, we demonstrate a generic protocol for producing polymer-supported lipid bilayers on glass surfaces that contain essentially all naturally occurring cell-membrane components of a cell line while still retaining transmembrane protein mobility and activity. This was achieved by merging vesicles made from synthetic lipids (PEGylated lipids and POPC lipids) with native cell-membrane vesicles to generate hybrid vesicles which readily rupture into a continuous polymer-supported lipid bilayer. To investigate the properties of these complex hybrid SLBs and particularly the behavior of their integral membrane-proteins, we used total internal reflection fluorescence imaging to study a transmembrane protease, β-secretase 1 (BACE1), whose ectoplasmic and cytoplasmic domains could both be specifically targeted with fluorescent reporters. By selectively probing the two different orientations of BACE1 in the resulting hybrid SLBs, the role of the PEG-cushion on transmembrane protein lateral mobility was investigated. The results reveal the necessity of having the PEGylated lipids present during vesicle adsorption to prevent immobilization of transmembrane proteins with protruding domains. The proteolytic activity of BACE1 was unadulterated by the sonication process used to merge the synthetic and native membrane vesicles; importantly it was also conserved in the SLB. The presented strategy could thus serve both fundamental studies of membrane biophysics and the production of surface-based bioanalytical sensor platforms.
Single and mixture toxicity of four pharmaceuticals of environmental concern to aquatic organisms, including a behavioral assessment

Abstract:Pharmaceuticals are frequently detected in aquatic environments as mixtures and can cause toxic effects to non-target organisms. We aimed to evaluate the single and mixture effects of the pharmaceuticals metformin, bisoprolol, ranitidine and sotalol using Daphnia similis and Danio rerio. In addition, we aimed to test the predictive accuracy of the mathematical models concentration addition and independent action and to evaluate the nature of the possible toxicological interactions among these pharmaceuticals using the combination index-isobologram model. The acute toxicity of these four pharmaceuticals individually and of their binary mixtures were evaluated using the D. similis tests. Developmental and behavioral effects induced by the pharmaceuticals in quaternary mixtures were evaluated using D. rerio embryos. We observed that most of the binary mixture effects were in the zone between the effects predicted by the concentration addition and the independent action model. The combination index-isobologram model showed to be adequate to describe the nature of possible interactions occurring between the combined pharmaceuticals. Developmental and behavioral acute adverse effects seem not to be induced by the joint action of the quaternary mixture of the evaluated pharmaceuticals on D. rerio embryos, at the concentrations at which they are usually found in surface fresh waters. However, from the results obtained with D. similis, we can conclude that assessing the ecological risk based on the effects of individual pharmaceuticals can underestimate the risk level posed by these environmental contaminants.

Abstract:We studied the role of vascular collagen on the responsiveness of isolated pulmonary arteries to vasoactive agonists. Dose-response curves to prostaglandin F2 alpha, (PGF2 alpha), angiotensin II (ANG II), and norepinephrine (NE) and responses to a depolarizing concentration of KCl were obtained on rings of pulmonary artery from normal rats, chronically hypoxic (10% O2 for 10 days) rats, and chronically hypoxic rats treated with cis-4-hydroxy-L-proline (cHyp), an agent that blocks collagen accumulation. Treatment with this agent prevented the rise in pulmonary blood pressure normally seen during hypoxia. Collagen content was twice normal in hypoxic vessels and was significantly reduced in vessels from cHyp-treated hypoxic rats. There was no change in reactivity to PGF2 alpha, but reactivity to ANG II, NE, and KCl was decreased in vessels from hypoxic rats. cHyp treatment completely restored reactivity to NE and KCl and partially restored reactivity to ANG II. The changes in contractility could be a response to the lower blood pressure. Another explanation is that collagen deposited in hypertensive pulmonary arteries may impair reactivity to some constrictor agonists.

Abstract:Purified human granulocyte elastase is inactivated by serum owing to com-plexation with α1-antitrypsin and α2-macroglobulin. The molar combining ratios are 1:1 for elastase/α1-antitrypsin and 2:1 for elastase/α2-macroglobulin. The affinity of granulocyte elastase is approximately the same for both protease inhibitors. Considering the molar concentration of the two inhibitors in normal human plasma, α1-antitrypsin thus is the dominating elastase inhibitor.
