Abstract:Antibiotics used in livestock production may be present in manure and slurry as the parent compound and/or metabolites.The environments may therefore be exposed to these antibiotics due to the application of animal manure to agricultural land.In order to understand the potential effects of long-term application of antibiotics-contained animal manure on agricultural soils,laboratory studies were conducted to characterize two typical antibiotic compounds(tylosin,C46H77NO17 and oxytetracycline,C22H24N2O9HCl) with regard to sorption and transport in seven agricultural soils,collected from the Changjiang and Zhujiang deltas of China.Both transfer and adsorption behaviors of the antibiotics were related with species of antibiotics and soil properties.Distribution coefficients(Kd values) determined by botch equilibrium method varied between 43L kg-1 and 3018L kg-1 for oxytetracyclin,and 2.3L kg-1 and 175L kg-1 for tylosin.Oxytetracyclin was more strongly sorbed in all the soils investigated than tylosin.The adsorption amount of antibiotic compounds on the soils was correlated positively with the contents of clay,organic matter and iron oxides.Soil column leaching experiments indicated that there were a significant difference in the transport of the two antibiotic compounds between sandy soil and clayey soil,which due to the difference in their respective sorption capabilities.Very low concentrations of the two antibiotic compounds were detected in the leachates from clayey soil column,indicating a much lower mobility of antibiotics in the clayey soil.However,significant amount of tylosin compound was recovered in the leachates from sandy soil,indicating that tylosin compound was not stable in sandy soil,and it had a high mobility in the soil.

Abstract:Veterinary antibiotics are widely used worldwide to treat and prevent infectious diseases, as well as (in countries where allowed) to promote growth and improve feeding efficiency of food-producing animals in livestock activities. Among the different antibiotic classes, tetracyclines and sulfonamides are two of the most used for veterinary proposals. Due to the fact that these compounds are poorly absorbed in the gut of animals, a significant proportion (up to ~90%) of them are excreted unchanged, thus reaching the environment mainly through the application of manures and slurries as fertilizers in agricultural fields. Once in the soil, antibiotics are subjected to a series of physicochemical and biological processes, which depend both on the antibiotic nature and soil characteristics. Adsorption/desorption to soil particles and degradation are the main processes that will affect the persistence, bioavailability, and environmental fate of these pollutants, thus determining their potential impacts and risks on human and ecological health. Taking all this into account, a literature review was conducted in order to shed light on the current knowledge about the occurrence of tetracycline and sulfonamide antibiotics in manures/slurries and agricultural soils, as well as on their fate in the environment. For that, the adsorption/desorption and the degradation (both abiotic and biotic) processes of these pollutants in soils were deeply discussed. Finally, the potential risks of deleterious effects on human and ecological health associated with the presence of these antibiotic residues were assessed. This review contributes to a deeper understanding of the lifecycle of tetracycline and sulfonamide antibiotics in the environment, thus facilitating decision-making for the application of preventive and mitigation measures to reduce its negative impacts and risks to public health.

Abstract:The Jarisch-Herxheimer reaction (JHR) observed after antibiotic treatment of relapsing fever caused by Borrelia recurrentis is associated with the systemic appearance of cytokines. The decrease of cytokine production and block of JHR was attempted by administering pentoxifylline prior to antibiotic treatment. Fifteen patients with confirmed relapsing fever were infused intravenously with pentoxifylline 90 min before intramuscular injection of penicillin; 4 patients were not treated with pentoxifylline. All patients developed JHR to varying degrees. Treatment with pentoxifylline failed to prevent fever, increase in pulse, respiration, or blood pressure, or decrease in white blood cell count. No reduction of circulating levels of tumor necrosis factor, interleukin 6, or interleukin-8 was observed with pentoxifylline treatment. Pentoxifylline did not prevent clearance of the B. recurrentis spirochetes. Thus, pentoxifylline treatment of patients with relapsing fever fails to prevent or diminish JHR or the associated cytokine release observed after appropriate antibiotic treatment.

Abstract:Antibiotics have a wide application range in human and veterinary medicines. Being designed for pharmacological stability, most antibiotics are recalcitrant to biodegradation after ingestion and can be persistent in the environment. Antibiotic residues have been detected as contaminants in various environmental compartments where they cause human and environmental threats, notably with respect to the potential emergence and proliferation of antibiotic-resistant bacteria. An important component of managing environmental risk caused by antibiotics is to understand exposure of soil and water resources to their residues. One challenge is to gain knowledge on the fate of antibiotics in the ecosystem along the soil-water continuum, and on the collateral impact of antibiotics on environmental microorganisms responsible for crucially important ecosystem functions. In this context, the ANTIBIOTOX project aims at studying the environmental fate and impact of two antibiotics of the sulfonamide class of antibiotics, sulfamethazine (SMZ), and sulfamethoxazole (SMX).

Abstract:Antibiotic residues that reach the environment via land application of livestock manure could impact structure and function of microbial communities and promote the spread of antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs). To assess whether there is a risk, we have reviewed extensive data on five veterinary antibiotics (VAs) that are commonly used in livestock farming (amoxicillin, enrofloxacin, sulfadiazine, tetracycline, trimethoprim). Predicted environmental concentrations (PECs) after the medication of pigs were derived using (i) a total residue approach and (ii) the VetCalc model to account for additional fate parameters and regional scenarios specific to Germany. Predicted no effect concentrations (PNECs) for microbial toxicity and ARB selection were derived from available concentration-response data. Except for enrofloxacin, the total residue PECs exceeded 100 µg kg-1 in soil and risk quotients indicated a high risk for soil porewater and surface water (PEC/PNEC > 1). After PEC refinement, the risk in surface water was generally low. However, in soil porewater still a high risk was indicated for sulfadiazine, tetracycline, and trimethoprim that could persist up to 100 days after the manure application. These findings suggest an urgent need for regulatory action to mitigate the risk resulting from the presence of antibiotic residues in soil.

Abstract:Veterinary antibiotics and estrogens are excreted in livestock waste before being applied to agricultural lands as fertilizer, resulting in contamination of soil and adjacent waterways. The objectives of this study were to 1) investigate the degradation kinetics of the VAs sulfamethazine and lincomycin and the estrogens estrone and 17β-estradiol in soil mesocosms, and 2) assess the effect of the phytochemical DIBOA-Glu, secreted in eastern gamagrass (Tripsacum dactyloides) roots, on antibiotic degradation due to the ability of DIBOA-Glu to facilitate hydrolysis of atrazine in solution assays. Mesocosm soil was a silt loam representing a typical claypan soil in portions of Missouri and the Central United States. Mesocosms (n = 133) were treated with a single target compound (antibiotic concentrations at 125 ng g-1 dw, estrogen concentrations at 1250 ng g-1 dw); a subset of mesocosms treated with antibiotics were also treated with DIBOA-Glu (12,500 ng g-1 dw); all mesocosms were kept at 60% water-filled pore space and incubated at 25 °C in darkness. Randomly chosen mesocosms were destructively sampled in triplicate for up to 96 days. All targeted compounds followed pseudo first-order degradation kinetics in soil. The soil half-life (t0.5) of sulfamethazine ranged between 17.8 and 30.1 d and ranged between 9.37 and 9.90 d for lincomycin. The antibiotics results showed no significant differences in degradation kinetics between treatments with or without DIBOA-Glu. For estrogens, degradation rates of estrone (t0.5 = 4.71-6.08 d) and 17β-estradiol (t0.5 = 5.59-6.03 d) were very similar; however, results showed that estrone was present as a metabolite in the 17β-estradiol treated mesocosms and vice-versa within 24 h. The antibiotics results suggest that sulfamethazine has a greater potential to persist in soil than lincomycin. The interconversion of 17β-estradiol and estrone in soil increased their overall persistence and sustained soil estrogenicity. This study demonstrates the persistence of these compounds in a typical claypan soil representing portions of the Central United States.

Abstract:Advances in the elucidation of cystic fibrosis transmembrane regulator (CFTR) function have resulted in a greater understanding of the relationship between the CF gene defect and clinical disease. The clinical phenotype is influenced by the class of mutation and possibly by other modifier genes. CFTR regulates the volume and composition of airways surface liquid, primarily by controlling chloride ion transport. However, CFTR also regulates other membrane channels and transports other molecules which may be important in mucocilary clearance and innate defence mechanisms. CFTR may also modulate the inflammatory response in respiratory epithelial cells and other inflammatory cells through a range of mechanisms. It is apparent that CFTR dysfunction results in a range of effects which may contribute to the clinical phenotype. These may contribute to the development of clinical disease at different stages of the natural history of cystic fibrosis. A greater understanding of the basic defect and its implications is likely to result in novel therapeutic approaches.

Abstract:This study addresses the growing concern about the high levels of antibiotics in water, outlining an alternative for their removal. The adsorption of four representative antibiotics from commonly used families (fluoroquinolones, β-lactams, trimethoprim, and sulfonamides) was performed over vegetal powdered activated carbon. The evolution of the adsorption was studied during 60 min for different initial antibiotic concentrations, not only individually but also simultaneously to determine competitive adsorption. Moreover, this research studied the adsorption isotherms and kinetics of the process, as well as the pH influence; FTIR of the activated carbon before and after adsorption was carried out. Trimethoprim and sulfadiazine showed more affinity for the adsorbent than amoxicillin and enrofloxacin. This trend might be attributed to their structure, capable of stablishing stronger π-π interactions with the adsorbent, which showed high affinity for the active sites of the adsorbent via FTIR. In addition, the sorption isotherms of trimethoprim followed a Langmuir type isotherm, amoxicillin followed a Freundlich type isotherm, and enrofloxacin and sulfadiazine followed both. The antibiotics followed pseudo-second-order kinetics. Sulfadiazine and amoxicillin gave better performances in acidic conditions. By contrast, the sorption of trimethoprim was favored in basic environments. Variations of pH had a negligible effect on the removal of enrofloxacin.

Abstract:The presence of pharmaceuticals in agricultural soils, like amoxicillin (AMX) and ciprofloxacin (CIP), poses a significant environmental challenge with potential implications for ecosystems and human well-being. This study explores the simultaneous adsorption of AMX and CIP on crop soils and bio-adsorbents, focusing on competitive adsorption dynamics. Tests were conducted with varying pharmaceutical concentrations in six soils and three bio-adsorbents. CIP consistently exhibited higher adsorption than AMX, particularly at higher concentrations. In the binary system, AMX's adsorption exceeded the individual system at higher concentrations, implying a synergistic effect. Bio-adsorbents, especially pine bark and oak ash, displayed superior adsorption capacities compared to soils. Some soils exhibited enhanced adsorption and retention of both antibiotics simultaneously, aligning with the cooperative adsorption model. Freundlich's adsorption model described the competitive adsorption systems well. These findings have implications for addressing antibiotic contamination in agricultural ecosystems, offering insights into complex interactions in soil environments amid rising pharmaceutical concerns.

Abstract:A laboratory experiment was performed to examine the medium-term influence of three tetracycline antibiotics (chlortetracycline, CTC; tetracycline, TC and oxytetracycline, OTC) at different concentrations in four agricultural soils with similar pH and different soil organic content. After a 42-days incubation period, three different soil enzymes (β-glucosidase, urease, and phosphomonoesterase) were estimated, as well as the phospholipid fatty acids (PLFAs). A residual effect was observed on all microbial parameters measured in the four soils affecting to the soil enzymes activity and soil microbial communities structure (PLFA pattern). A different microbial sensitivity to antibiotics was detected depending on both, soil type and the microbial property considered. Specifically, in general, no antibiotic effect or even a slight positive effect was observed for phosphomonoesterase and β-glucosidase enzyme activities, respectively, while a negative effect was detected for urease activity values, particularly at higher doses of the antibiotics in a soil with a low organic matter content. The principal component analysis performed with the PLFAs data obtained for all soil samples showed different microbial communities depending mainly on soil type, followed by the antibiotic added to the soil (CTC, TC or OTC) and, in a lesser extent, by its concentration. In general, the PLFA patterns showed similar microbial communities structure due to OTC and TC addition in comparison to the microbial communities structure of soil treated with CTC. These results could be environmentally relevant, especially as regards potential effects of antibiotics on the soil microbiome and hence on health risk assessment of these antibiotics in soils.

The fate of sulfamethoxazole in microcosms of the macrophyte Schoenoplectus californicus and its impact on microbial communities

Abstract:Sulfamethoxazole is a widely used antibiotic frequently found as an environmental pollutant. It can alter microbial communities and increase antibiotic resistance, becoming a public health risk. Constructed wetlands have the potential for removing sulfamethoxazole from polluted waters, but the role of different macrophytes in this process is not well understood. We investigated the fate of sulfamethoxazole and its effect on bacterial communities in microcosms containing Schoenoplectus californicus, an altitude-tolerant macrophyte. Within the first 10 h after introducing sulfamethoxazole (initial concentration 5 mg/L) to the microcosms, the concentration in the liquid phase significantly differed between microcosms with and without S. californicus. However, over the long term (15 and 30 days post-addition), the removal percentage (around 75%) in the liquid phase was not significantly influenced by S. californicus, indicating that sediments might be primarily responsible for removing the antibiotic. The presence of S. californicus promoted algae growth in the microcosms, and we determined that algae contributed to sulfamethoxazole removal from the liquid phase, likely through adsorption. Additionally, we characterized bacterial communities in the microcosm sediments via nanopore sequencing to identify changes following sulfamethoxazole addition. The relative abundance of Proteobacteria increased from 37-46% to 48-99% with the addition of the antibiotic. Conversely, the relative abundance of cyanobacteria decreased significantly after sulfamethoxazole was added (from 17 to 35% to less than 2%), suggesting it may serve as a biological marker for sulfamethoxazole pollution. In addition, the functional profile of the community was estimated from taxonomic diversity using PICRUST.

Abstract:Widely application of antibiotics in medicine and animal breeding had resulted in its pollution in soil environment. Antibiotic resistance is an increasing challenge for health care services worldwide. Antibiotics of both human and veterinary origins have been widely detected in various environmental matrices including surface water, groundwater, soils, and sediments. Within a relatively short period of time after the first antimicrobial drugs were introduced, bacteria began exhibiting varying degrees of resistance. The excessive use (and abuse) of antibiotics in agriculture, and in both human and veterinary medicine, has played a critical causative role in the development of antibiotic resistance. Sulfadimidine and its epimers/isomers were most frequently detected, and it enters soils by the use of animal excrements as fertilizers. In order to understand the degradation characteristics of antibiotics in agricultural soils, two soils with different nutrient levels were selected to carry out several pot soil culture experiments for studying the effect of fertilizers (organic manure, NPK, N, PK), tillage intensity (tillage, no tillage), water conditions (drought, udic, drought-udic alternation, wetness) and crop plantation (vegetables, not vegetables) on degradation of sulfadimidine in the soils. The results showed that the application of organic manure, NPK, and N fertilizers could promote the degradation of sulfadimidine in the soils, as compared with no fertilizer treatment. The effect of organic manure application on the degradation of sulfadimidine was the most obvious. However, the effect of PK fertilizer application on the degradation of sulfadimidine was not obvious. Tillage also could promote the degradation of sulfadimidine in the soils. Both udic moisture regimes and drought-udic moisture alternation were more favorable for the degradation of sulfadimidine in the soils than drought and wetness. Plantation of vegetable crop could improve the degradation of sulfadimidine in the soils. The degradation proportion of the antibiotics in the rhizosphere soil was higher than the whole soil. While the degradation proportion of the antibiotics in soil with high nutrient level was generally higher than soil with low soil nutrient level. It was thought that effects of fertilization, soil fertility, crop plantation on the degradation of sulfadimidine in the soils were related to alteration in soil microbial activity caused by those factors. Tillage can promote the degradation of antibiotics in the soils by increasing its photolysis.

Abstract:Sulfonamide antibiotics (SAs) are widely used antimicrobial agents in livestock and aquaculture, and most of them entering the animal's body will be released into the environment as prodrugs or metabolites, which ultimately affect human health through the food chain. Both acid deposition and salinization of soil may have an impact on the migration and degradation of antibiotics. Sulfamethazine (SM2), a frequently detected compound in agricultural soils, has a migration and transformation process in the environment that is closely dependent on environmental pH. Nevertheless, scarcely any studies have been conducted on the effect of soil pH changes on the environmental behavior of sulfamethazine. We analyzed the migration and degradation mechanisms of SM2 using simulation experiments and ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-Q-TOF-MS) techniques. The results showed that acidic conditions limited the vertical migration of sulfadimidine, and SM2 underwent different reaction processes under different pH conditions, including S-C bond breaking, S-N bond hydrolysis, demethylation, six-membered heterocyclic addition, methyl hydroxylation and ring opening. The study of the migration pattern and degradation mechanism of SM2 under different pH conditions can provide a solid theoretical basis for assessing the pollution risk of sulfamethazine degradation products under acid rain and saline conditions, and provide a guideline for remediation of antibiotic contamination, so as to better prevent, control and protect groundwater resources.
Reaction of human alpha2-antiplasmin and plasmin stopped-flow fluorescence kinetics.

Abstract:The interaction of human plasmin with human alpha2-antiplasmin was measured in the presence and absence of lysine-binding ligands using the corresponding active site fluorescence changes. The stopped-flow method allows for direct determination of reliable values of the second order rate constant for the fast association step of plasmin and alpha2-antiplasmin in the absence of another interacting compound, e.g. a plasmin substrate. At pH 7.4, 25 degrees C, k1 = 2.2 x 10(7) M(-1)s(-1) was obtained. Substantial reductions in k1 were seen in the presence of trans-4-(aminomethyl)cyclohexane-1-carboxylic acid at concentrations corresponding to lysine-binding site interactions at kringle 4 of plasmin; at saturation the rate constant is reduced 20-fold, whereas the effect of saturation of kringle 1 is only a 2-fold reduction. It is thus found that the interaction of alpha2-antiplasmin with the lysine-binding site of kringle 1 is of little importance compared with that of kringle 4 in regulating the inhibition reaction of plasmin with alpha2-antiplasmin. Similar results were recently obtained for the bovine plasmin-bovine alpha2-antiplasmin reaction (Christensen et al. (1995) Biochem. J. 305, 97-102).

Results showed that the antibiotic concentration in vegetables, soil and manure from 53 fixed-point monitoring sampling sites were ND-18.47, ND-1438.50 and ND-24710.00 μg kg−1, respectively. There were positive correlations between the antibiotic concentrations of vegetables and soil as well as between soil and manure. The average Amountsoil/manure values were 1.48–46.02, indicating that antibiotics built up pseudo persistent residues in soil due to repeated fertilization. The modified level-III fugacity model showed that tetracyclines and fluoroquinolones tend to remain in soil given their sorption and mobility, while sulfonamides were highly distributed in plants, especially in leaves. Norfloxacin, ofloxacin, sulfadiazine, sulfamethoxazole and sulfisoxazole were found to be risk factors in facility agriculture and should be continuously monitored during agricultural production. Most importantly, we used the inversion method to determine the recommended maximum residue limits of antibiotics in soil. This will not only allow for better control of the amount of the antibiotics in the environment, but also act as a potential method to assess the risks of pollutants without maximum residue limits in the environment.

From bacteria to fish: ecotoxicological insights into sulfamethoxazole and trimethoprim

Abstract:Sulfamethoxazole (SMX) and trimethoprim (TRIM) are two of the most used antibiotics in the last 50 years, to prevent and treat bacterial infections; however, the available literature about toxicity to non-target organisms is quite discrepant and incomplete. This study aims to assess the SMX and TRIM ecotoxicological effects in standard species: Aliivibrio fischeri (bioluminescence inhibition), Escherichia coli ATCC 25922 (growth inhibition), Lemna minor (growth inhibition and biochemical biomarkers), Daphnia magna (immobilization/mortality, life history traits, and biochemical biomarkers), and Danio rerio (survival, hatching, abnormalities, and biochemical biomarkers). The species tested showed different acute sensitivities to SMX (A. fischeri < D. magna < E. coli < L. minor) and TRIM (L. minor < A. fischeri < D. magna < E. coli). Overall, TRIM reveals less toxicity than SMX, except for E. coli (Ecotoxicological approach based on Antimicrobial Susceptibility Testing - EcoAST procedure). Both antibiotics affect individually (e.g., growth and survival) and sub-individually (e.g., antioxidant defenses) L. minor, D. magna, and D. rerio. This study allowed us to generate relevant data and fill gaps in the literature regarding the effects of SMX and TRIM in aquatic organisms. The here-obtained results can be used to (i) complete and re-evaluate the Safety Data Sheet to improve the assessment of environmental safety and management of national and international entities; (ii) clarify the environmental risks of these antibiotics in aquatic ecosystems reinforcing the inclusion in the 4th Watch List of priority substances to be monitored in whole inland waters by the Water Framework Directive; and (iii) combat the development of antimicrobial resistance, as well as supporting the definition of environmental measurements in the context of European One Health Action Plan. However, it is essential to continue studying these antibiotics to better understand their toxicity at ecologically relevant concentrations and their long-term effects under different climatic change scenarios.
Comparison of by-products as adsorbents for the removal of the antibiotics ciprofloxacin, trimethoprim and clarithromycin

Abstract:Antibiotics in the environment are considered emerging pollutants, with special relevance and concern due to the proliferation of antibiotic-resistant bacteria and genes. Therefore, finding ways to remediate antibiotics-contaminated soil and water through the use of bio-adsorbents is imperative. In this research, we investigate three by-products (hemp waste, oak ash, and mussel shell) as potential low-cost bio-adsorbents for the antibiotics Ciprofloxacin (CIP), Clarithromycin (CLA), and Trimethoprim (TRI), using batch-type and stirred flow chamber experiments to study their retention and release. The results indicate that hemp waste has higher sorption capacity for CIP and TRI (20891.8 and 2481.6 μmol L-1, respectively), while oak ash yields the highest retention for CLA (3078.4 μmol L-1). In addition, it was shown that the pH value significantly influences the sorption of these pollutants onto hemp waste. Among the three antibiotics, CLA was the most mobile, given the release experiments (903.9-1758.9 μmol kg-1), while ciprofloxacin (440.3-542.4 μmol kg-1) and trimethoprim (639.4-1652.1 μmol kg-1) are released less. Overall, the results of this research (the first of this kind including these antibiotics and sorbents simultaneously) suggest that while the individual antibiotics retention on each of the three by-products may not be entirely satisfactory, its potential combination (among them and/or with other low-cost sorbents) could significantly contribute to addressing antibiotics environmental pollution, favouring recycling and promoting a circular economy, which is a matter of global relevance.
