Abstract:This study investigated the effect of electron beam irradiation (EBI) on the lipid stability of oat and barley during long-term storage. Results showed that the initial free fatty acid content in oat was higher than that in barley. This may mean that lipid hydrolysis started under the function of lipase when oat and barley were milled into flours. Both storage and EBI factors influenced lipid-degrading enzyme activity and promoted lipid oxidation in oat and barley. However, it seemed that storage had higher impacts because the DPPH scavenging activity decreased greatly, and the contents of both malondialdehyde and volatile lipid oxidation products increased in all samples. Thus, the antioxidant capacity and level of lipid oxidation after EBI treatment should be considered when producing oat and barley foods. Overall, this study shows the high potential of EBI for use as a non-thermal technique in stabilising the storage quality of oat and barley.

Abstract:The effect of γ irradiation on the cooked rice texture and amylopectin structure is studied with three varieties(Zhefu 504.Zhefu 802 and Zaogeng T2)subjected to 4kGy,8kGy and 12kGy irradiation.The results indicate that irradiation has significant effect on the texture of the cooked rice;the hardness,cohesiveness,gumminess and chewiness reduce significantly with the increase of the irradiation dose.Irradiation also affects the chain length distribution of the amylopectin,the side-chains of amylopectin of Zhefu 504 with the 12≥DP,Zhefu 802 with 24≥DP≥13 and Zaogeng T2 with 24≥DP≥13and 36≥DP≥25 are broken.
Electron beam irradiation induced aggregation, structural and functional changes of soybean 11S globulin

Abstract:This study investigated the effects of different irradiation doses of an electron beam (e-beam) (0, 2, 4, 6, 8, and 10 kGy) on the structure, emulsification, foaming, and rheological and gel properties of soybean 11S globulin. The irradiation treatment at 4 and 6 kGy significantly increased the solubility, surface hydrophobicity, disulfide bonding, and ζ-potential of 11S globulin, decreased the particle size of the protein solution, and effectively improved the emulsifying activity and foaming stability of the protein solution. Moreover, irradiation induced moderate cross-linking and aggregation of the proteins, thereby increasing the apparent viscosity and shear stress of the protein solution. In addition, the low-field NMR and microstructure analysis results revealed that protein gels formed a dense and homogeneous three-dimensional mesh structure after irradiation (6 kGy), along with increased content of bound water (T2b) and water not readily flowable (T21) and a decrease content of free water (T22). Overall, our results confirmed that e-beam irradiation could significantly improve the physicochemical properties of soybean 11S globulin. Our study thus provides a new technical means for the application of electron beam irradiation technology toward protein modification and broadens the high-value utilization of soybean 11S globulin in the food processing industry.
Effects of combined fermentation and ultrasonic treatments on the structure and properties of Okara soluble dietary fiber

Abstract:In this study, okara dietary fiber samples were prepared through a combination of solid-state fermentation using Aspergillus oryzae and ultrasonication. Subsequent infrared and UV–visible spectral analyses revealed that such treatment resulted in an increase in the oligosaccharide content of these samples through the enhanced cleavage of hydrogen bonds. Scanning electron microscopy and X-ray evaluation further confirmed the changes in the structural makeup of these okara SDF (soluble dietary fiber) samples relative to control samples. Following combination treatment, the soluble dietary fiber levels in these samples were three times higher than in control samples at 18.06 g/100 g. This treatment also led to a significant increase in the water holding, oil holding, and swelling capacities of the prepared fiber samples, whereas the capacity for water retention was slightly reduced. Ultrasonication alone failed to impact SDF oil holding capacity, nor did it impact cholesterol absorption at a pH of 2 used to simulate the stomach microenvironment. Together, these data thus offer a valuable reference for further work aimed at extracting soluble dietary fiber from okara.
Electron Beam Irradiation Alters the Physicochemical Properties of Chickpea Proteins and the Peptidomic Profile of Its Digest

Abstract:Irradiation can be used for the preservation of chickpea protein as it can destroy microorganisms, bacteria, virus, or insects that might be present. However, irradiation may provoke oxidative stress, and therefore modify the functionality and nutritional value of chickpea protein. In order to study the effects of irradiation on the physicochemical properties and digestion behaviour of chickpea protein, chickpea protein concentrate (CPC) was treated with electron beam irradiation (EBI) at doses of 5, 10, 15, and 20 kGy. After irradiation, protein solubility first increased at 10 kGy and 15 kGy, and then decreased at the higher dose of 20 kGy. This was supported by SDS-PAGE, where the intensity of major protein bands first increased and then decreased. Increased doses of EBI generally led to greater oxidative modification of proteins in CPC, indicated by reduced sulfhydryls and increased carbonyls. In addition, the protein structure was modified by EBI as shown by Fourier transform infrared spectroscopy analysis, where α-helix generally decreased, and β-sheet increased. Although the protein digestibility was not significantly affected by EBI, the peptidomic analysis of the digests revealed significant differences among CPC irradiated with varying doses. A total of 337 peptides were identified from CPC irradiated with 0 kGy, 10 kGy, and 20 kGy, with 18 overlapping peptides and 60, 29, and 40 peptides specific to the groups of 0, 10, and 20 kGy respectively. Theoretical calculation showed that the distribution of peptide length, hydrophobicity, net charge, and C-terminal residues were affected by irradiation. The 2, 2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity showed a marginal decrease with an increasing dose of irradiation. In conclusion, EBI led to oxidative modification and structural changes in chickpea protein, which subsequently affected the physicochemical properties of peptides obtained from in-vitro digestion of CPC, despite similar digestibility.

Abstract:In this study, Chinese yam (Dioscorea opposita Thunb.) flour was irradiated at doses of 1.79, 6.06, 10.07 and 14.97 kGy of the electron beam, to analyze the effects of electron beam irradiated (EBI) on the solubility, transmittance, swelling, paste properties, and the microstructure of starch in yam flour. Results showed that EBI degraded the starch component of yam flour, and presented an increase in solubility from 15.59 to 30.05%, transmittance from 8.11 to 11.63%, and a decrease in swelling from 9.59 to 7.26 g/g. Also, we observed that there was a significant decrease in the paste properties of yam flour with the elevation of EBI. The scanning electron micrographs and Fourier transforms infrared spectra further illustrated the effect of EBI on the molecular structure, but not the surface structure of starch in yam flour. C-type crystalline structure in yam starch was also observed in both control and treatment, while irradiated yam starch had a lower crystallinity than that of unirradiated treatments. Yam starch exhibited a decrease trend in their thermal property values with irradiation treatment. This study concludes that the physical properties of yam flour instructed by EBI may be used in a wide application in the food and pharmaceutical industrie.

Abstract:Summary Okara is the by‐product of soybean during processing, which is rich in dietary fiber. In this study, the effects of steam explosion (SE) treatment on the physicochemical properties and biological activities of soluble dietary fiber (SDF) from okara was investigated. Results showed that SE increased the water‐holding capacity, oil‐holding capacity, swelling capacity, water solubility, foaming ability, foaming stability, emulsifying activity and emulsion stability of SDF. The structure of okara‐derived SDF was changed after SE treatment, and the surface was poriferous, rough and collapsed. SDF from okara modified by SE exhibited significantly higher glucose adsorption capacity, cholesterol adsorption capacity and antioxidant activity manifested by the increase in DPPH and ABTS‐radical scavenging activity as well as ferric ion reducing antioxidant power. In vitro colonic fermentation and 16S rDNA sequencing showed that SE treatment up‐regulated the short‐chain fatty acids (SCFAs) concentrations and increased diversity of gut microbiota. Moreover, SE treatment increased the relative abundance of specific beneficial bacteria including Lactobacillus, whereas decreased the relative abundance of Firmicutes, Enterococcus, Escherichia‐Shigella and Proteus. In conclusion, these results highlight the potential of SE in improving the physicochemical and functional properties of okara‐derived SDF and promoting its future application.

Abstract:The objective of this study was to investigate the effect of particle size and microstructure on the physical and functional properties of soybean insoluble dietary fiber (IDF) in aqueous solution. Different IDF suspensions were obtained by ball milling (BM) and high-pressure homogenization (HPH) treatments. The results showed that the BM treatment significantly reduced the particle size distribution of the IDF, and the additional HPH treatment (BM-HPH) caused the IDF to become more porous, resulting in the formation of a honeycomb structure. Samples with a honeycomb network structure had higher hydration capacity and solubility. The water holding capacity, swelling capacity and solubility of I8 reached 18g/g, 72ml/g and 35%, respectively. I8 had the highest emulsifying stability and suspension stability. These results of the present study suggested that the reduction in particle size of IDF cannot fully account for its functional properties, of which the honeycomb structure played a more significant role. From the results of this study, it was clear that the functional properties of IDF depend on its small particle size and honeycomb structure. The present study indicated that a combination of BM and HPH treatments provides a new perspective formodifying and utilizing dietary fiber.

Abstract:The endogenous components of okara can affect the extraction and modification of insoluble dietary fiber (IDF). This study was intended to investigate the effect of extraction and purification of IDFs from okara affected by the endogenous components and composite enzyme Viscozyme® L. After that, the physicochemical, functional, and structural characteristics of IDFs were analyzed. The results showed that with the purification of degreasing, deproteinizing and destarching, Viscozyme® L can accurately act on the cell wall of okara, leading to the increase in specific surface area of IDF and the formation of honeycomb-like structure. In accordance with the X-ray diffraction and Fourier-transform infrared spectrum investigations, relative crystallinity decreased and certain cellulose components were reallocated and changed into soluble dietary fiber. Moreover, the modified IDF showed improved water-holding capacity (10.73-11.59 g/g), oil-holding capacity (5.37-6.60 g/g), and swelling capacity (8.99-12.37 mL/g), leading to improved adsorption capacities of glucose (maximum: 39.60 mg/g), cholesterol (14.54-33.56 mg/g at pH 7; 10.94-19.37 mg/g at pH 2), and cholate and better cation-exchange capacity. The outcomes demonstrated the potential for the acquired IDFs, particularly the high-purity modified-IDFs T3 and T4 , to be exploited as functional fiber components in the food sector. PRACTICAL APPLICATION: This study advances the understanding of the internal mechanism of modified insoluble dietary fiber (IDF) with different purities in okara. The obtained high-purity modified IDF can be employed as a functional fiber raw material in the manufacture of functional foods, and can also be utilized to assist in the treatment of some illnesses due to its well-established in vivo function.

Abstract:In this study, we compared the effects of three methods on the structural, physicochemical, and functional properties of insoluble dietary fiber (IDF) in okara: cavitation jet plus ultrasonic treatment (UC), cavitation jet plus alkaline hydrogen peroxide treatment (HC), and cavitation jet plus fermentation with Bacillus subtilis (FC). Compared to UC-IDF, the two types of co-modified IDF showed significant differences in particle size and specific surface area. Structural analyses showed that HC-IDF had a porous structure and complex morphological structure, with a significant increase in the absorption peak area at around 3420 cm−1 and 1051 cm−1, and a significant decrease in thermal stability and crystallinity. Of the three treatments, HC most effectively enhanced the physicochemical and adsorption properties of IDF, including the emulsion stability (84.49% ± 3.5%), water-holding capacity (16.04 ± 0.05 g/g), glucose absorption capacity (53.17 ± 3.44 g/g), cholesterol absorption capacity (32.87 ± 0.89 mg/g), and nitrite absorption capacity (211.7 ± 0.17 μg/g). These results demonstrate the potential of HC-modified okara IDF as a valuable functional food ingredient. Therefore, the cavitation jet combined with alkaline hydrogen peroxide method has a good potential for application in the study of dietary fiber modification.

Structural characterization and functional properties of egg white protein treated by electron beam irradiation

Abstract:This study investigated the functional properties and structural characteristics of egg white protein (EWP) powder treated by electron beam irradiation (EBI) at the doses of 5, 10, 20, 30, 50, 70 and 100 kGy. In comparison with the untreated sample (0 kGy), the EBI-induced EWP showed generally better solubility and higher antioxidant capacity by ABTS• + radical scavenging assay. Furthermore, the emulsifying activity index of EBI-induced EWP increased with increased EBI dose, reaching the maximum value of 15.8 ± 0.4 m 2 /g at 100 kGy. The emulsifying stability index and ϛ-potential of EBI-induced EWP showed fluctuating tendencies. These functional property changes of protein were attributed to the conformational changes caused by EBI. SDS-PAGE patterns confirmed that high-dose EBI would result in protein depolymerization to low-mass molecules at 11 kDa, contributing to the increased solubility. Circular dichroism data demonstrated that EBI-induced EWP exhibited partial denaturation and a flexible structure, which was responsible for the improved emulsifying properties. Fluorescence spectra revealed that high EBI doses led to an increase in the polar environment around tryptophan, which could enhance the solubility and antioxidant activity of EWP. In addition, high EBI doses decreased the content of free and total sulphydryl groups, and changed the intermolecular forces of protein, including ionic bonds, hydrogen bonds, hydrophobic interactions, and disulfide bonds. These findings can provide solid support for practical applications of EBI technology in EWP food processing.

Abstract:Electron beam irradiation (EBI) is an environmentally friendly physical modification technology. In this study, pea starch nanocrystals (SNC) were prepared by EBI-assisted pretreatment, and investigated the effects of EBI on the multiscale structure and physicochemical properties of SNC. EBI-assisted pretreatment didn't change the particle morphology, crystalline type and FT-IR spectra of SNC. However, EBI-SNC's relative crystallinity and short-range orderliness index (R 1047/1022 ) significantly increased with increasing irradiation dose (5 KGy-20 KGy). In addition, EBI-assisted pretreatment caused the long chains of SNC's amylopectin to break into short chains. Moreover, EBI-assisted treatment significantly reduced the mean size, molecular weight, apparent amylose content, swelling power and SDS + RS content of SNC, while increasing the solubility, zeta potential and RDS content. Furthermore, the flow properties of the EBI-SNC samples were increased. The results show that EBI effectively changed the structural and functional properties of SNC, and the excellent functional properties are expected to broaden the application range of SNC.

Abstract:Moisture content is an important factor that affects rice storage. Rice with high moisture (HM) content has superior taste but is difficult to store. In this study, low-dose electron beam irradiation (EBI) was used to study water distribution in newly harvested HM (15.03%) rice and dried rice (11.97%) via low-field nuclear magnetic resonance (LF-NMR). The gelatinization, texture and rheological properties of rice and the thermal and digestion properties of rice starch were determined. Results showed that low-dose EBI could change water distribution in rice mainly by affecting free water under low-moisture (LM) conditions and free water and bound water under HM conditions. HM rice showed smooth changes in gelatinization and rheological properties and softened textural properties. The swelling power and solubility index indicated that irradiation promoted the depolymerization of starch chains. Overall, low-dose EBI had little effect on the properties of rice. HM rice showed superior quality and taste, whereas LM rice exhibited superior nutritional quality. This work attempted to optimize the outcome of the EBI treatment of rice for storage purposes by analyzing its effects. It demonstrated that low-dose EBI was more effective and environmentally friendly than other techniques.

Abstract:Okara, as a by-product of soybean processing, is rich in insoluble dietary fiber (IDF), which is a carbohydrate polymer with various insoluble polysaccharides. Nowadays, the extraction of IDF with excellent functional properties has become a research hotspot. In this work, we further proposed an alternating alkali/ultrasound method for the efficient extraction of IDF. The sequential treatments of alkali (A-ISF), alkali-ultrasonic (AU-ISF), ultrasonic-alkali (UA-ISF), ultrasonic-alkali-ultrasonic (UAU-ISF) and alkali-ultrasonic-alkali (AUA-ISF) were applied to extract insoluble soybean fiber (ISF). FTIR and XRD results proved the typical structure of ISFs, and TGA results demonstrated the improved thermal stability of UAU-ISF and AUA-ISF. Chemical composition measurement showed that UAU-ISF and AUA-ISF exhibited higher cellulose content (>83 %). SEM results revealed that ultrasonic treatment led to a decomposition of okara matrix and significant porous structure in ISFs with an amplified collapse effect, resulting in an increase of the pore size of ISFs, and strengthening the properties of UAU-ISF and AUA-ISF in higher water (>15 g/g)/oil (>12 g/g) holding capacities, cholesterol binding capacity (>36 mg/g), and cation exchange capacity (>0.3 mmol/g), thus providing new insights for the preparation of ISF with high functional properties that are beneficial for human intestinal health.

Abstract:<p>The effects of mixed fermentation by lactic acid bacteria and <em>Neurospora crassa,</em> and microwave treatment on the composition, structure, and functional properties of okara DF were studied. The results of SEM and EDX showed that the honeycomb structure was more obvious and the content of elemental components was changed after fermentation and microwave treatment. The result of TGA-DSC showed that the thermal stability of the sample improved following microwave treatment. The FT-IR results showed that fermentation and microwave treatment have an effect on the functional groups of cellulose and hemicellulose consistent with the composition of the samples, and the crystal structure of samples was slightly damaged after modification. After mixed fermentation and microwave treatment, the adsorption effect of cholesterol and sodium cholate was not obviously changed. However, the α-Amylase activity inhibition ratio of fermented DF increased from 9.55% to 15.04% and 20.92% respectively. The glucose dialysis retardation of fermented DF doubled at 30 min. Overall, both methods improve the availability of DF in okara. Modified DF from soybean residue can be used as a functional ingredient in diverse food products.</p>

Therefore, electron beam (e-beam) irradiation pretreatment was carried out to improve the deacetylation efficiency of KGM, and the physicochemical and gel properties of KGM were investigated. The results show that e-beam irradiation and deacetylation decrease the water absorption, solubility, transparency, molecular weight, and viscosity of KGM. Conversely, the moisture content, thermal stability, and water-binding capacity increase. FTIR and X-ray diffraction analysis revealed no significant changes in the chemical and crystalline structure of KGM before and after modification. However, modification weakens the intermolecular interaction of KGM hydrosols, which affects their rheology. Furthermore, deacetylation improves the mechanical properties and water retention capacity of KGM gels. Overall, the e-beam irradiation pretreatment provides a method to increase the efficiency of KGM deacetylation and improve the physical and chemical properties of KGM, thus expanding its potential applications in the food and chemical industries, among others.

Abstract:In this study, granular cold-water swelling (GCWS) starch was prepared from chestnut starch by ethanol-alkali method, after which it was further modified by electron beam irradiation (EBI) technique to investigate the effect of EBI on GCWS chestnut starch. It was shown that the alcohol-alkali treatment disrupted the starch double helix structure and the starch crystalline form had been changed from "C" to "V" type. On this basis, EBI continued to act on the disrupted starch chains and further cleaved the long chains into short chains, which significantly improved the solubility of starch to 90.08 % in cold water at a 24 kGy irradiation dose. Therefore, this study can broaden the application scope of starch and provide new ideas for GCWS starch applications in food and water-soluble pharmaceutical industries.
