Abstract:Using mushroom to enhance meaty flavor is a common culinary practice; however, nutrition and taste studies associated with amino acid changes during mushroom cooking are limited, and mushroom aroma's enhancement of meaty flavor has not been verified. This study aimed to 1) investigate free amino acid profile changes in raw, sautéed, and roasted portobello and shiitake, and 2) investigate if certain volatiles from mushroom culinary preparation could increase meaty flavor. A total of 23 amino acids, including all 9 essential, were quantified using gas chromatography-mass spectrometry and high-performance liquid chromatography. Both cooking methods caused significant (p ≤ 0.05) free amino acid loss, while mushroom variety significantly impacted most amino acid contents. The following 8 solutions were served to 82 participants: a blank (basic taste ingredients), a control (blank with meaty flavor), and three aroma blocks (control with mushroom-like, roasted, and grilled flavors) at two intensity levels. The control solution showed significantly higher umami taste than the blank. Compared to the control and blank, the grilled aroma significantly increased meaty flavor intensity and acceptance, overall acceptance, and umami taste; while the roasted and mushroom-like aroma blocks did not show meaty flavor enhancement. Meaty flavor was mainly driven by salty, umami, and sweet tastes as well as thickness mouthfeel. This study demonstrates how culinary application impacts free amino acid content in mushroom and how meaty flavor can be enhanced by aroma compounds derived from non-animal sources.

Abstract:The aim of the study was to evaluate the effect of marinating (3 or 6 days) in kefir (KE), yogurt (YO) and buttermilk (BM) and sous-vide cooking (SV) at 60 or 80 °C on changes in the protein profile of pork in relation to its sensory quality. In the marinated raw meat, an increased share of some fractions of myofibrillar and cytoskeletal proteins and calpains were found. The greatest degradation of proteins, regardless of time, was caused by marinating in YO and KE and cooking SV at 80 °C. The lowest processing losses were in samples marinated in KE and YO and cooked SV at 60 °C, with marinating time having no significant effect. The odor, flavor, tenderness and juiciness of meat marinated in BM was better than in KE and YO. Meat marinated and cooked SV at 60 °C was rated better by the panelists. Changes in proteins significantly affect the formation of meat texture, tenderness and juiciness, which confirms the correlations. This is also reflected in the sensory evaluation. During the process of marinating and cooking meat, protein degradation should be taken into account, which can be a good tool for shaping the sensory quality of cooked pork.

Abstract:In this study, edible mushroom and soybean protein isolate (SPI) were used to prepare a fibrous meat analogue using thermos-extrusion and the extruded mushroom-based meat analogue as meat replacer was further developed with different formulations in fabricating sausage analogues. The effect of water content (35%, 70% and 100%), three types of edible mushroom (Lentinus edodes, Pleurotus ostreatus, Coprinus comatus and a mixture of equal proportions) and their amounts (from 15% to 100%) on the physicochemical and structural profiles were studied. The results showed that the extruded mushroom-based meat analogue prepared from Coprinus comatus (15% addition) and SPI with a water content of 35% exhibited close textural profiles to real beef. Furthermore, a texture profile analysis (TPA) combined with a principal component analysis (PCA) was conducted to compare and assess the textural traits of the sausage analogues with similar commercial products. The characterization and comparison of the flavor profile of post-processing mushroom-based meat sausage analogues (MMSA) were performed using headspace-phase microextraction (HS-SPME), coupled with gas chromatography-mass spectrometry (GC-MS). A total of 64 volatile compounds were identified, and the content in dried-processing treatment was significantly higher than for steamed-processing, which indicated that the natural fermentation process contributed to the increase in aroma substances in the non-animal sourced sausage. This study developed a feasible method to fabricate a meat replacement and to create high added-value products, which offer an opportunity for developing non-animal products with satisfactory sensory properties and flavor profiles.

Abstract:This study evaluated the physicochemical characteristics and storage stability (at 0, 3, and 7 days) of hybrid beef patties with different amount of shiitake mushrooms (Lentinus edodes) added. Shiitake mushrooms contain healthy ingredients such as ergosterol and β-glucan. Four proportions of shiitake mushrooms were added to beef patties (T1, 20%, T2, 40%, T3, 60%, T4, 80%) as a substitute for beef and compared with a control group (CON 0%). Chemical composition, water holding capacity (WHC), cooking loss, pH, color, texture profile analysis, and sensory properties of the products were compared on day 0. As a storage stability experiment, volatile basic nitrogen (VBN), 2-thiobarbituric acid reactive substances (TBARS), and total microbial count were compared (0, 3, and 7 days). The results revealed that replacement with shiitake improved the WHC and cooking loss of patties but had a negative effect on sensory properties and storage stability. These results indicate that shiitake mushrooms can be added along with beef to produce hybrid patties; however, the usage amount must be considered.

Abstract:The influence of mycelium of oyster mushrooms addition on the physical and flavor properties of a fried plant-based beef analogue (PBBA) was evaluated. The key odor compounds of the PBBA were 2-methyl-3-furanthiol, furfuryl mercaptan, (E),(E)-2,4-decadienal, and hexanal. The content of volatiles increased with the increase in mycelium content when the mycelium content was <8%. E-nose analysis showed that nitrogen oxides and organic sulfides were the key compounds that contribute to the flavor of samples, and these varied with mycelium content. The umami taste of the PBBA increased slowly with the mycelium content; e-tongue and sensory evaluation showed that the mycelium content <8% may inhibit bitterness and astringency. In addition, mycelium can increase the springiness, brightness, and whiteness of the PBBA and reduce the redness and beany flavor. Overall, the addition of a moderate amount of mycelium can enhance the meat-like and fat-like aroma of PBBA, reduce some undesirable odor and taste, and made it closer to meat. These findings demonstrate the potential of mycelium as a raw material for PBBAs.

Abstract:Sausages are loved by people for their unique texture, satisfying chewiness, and pleasant flavor. However, in the production of sausages, red meat and a large amount of fat are mainly used, and long-term consumption will increase the risk of diseases such as obesity, heart disease, hypertension, and cancer. Our previous studies have shown that the intake of red meat and fat can be reduced through the replacement of lean meat and fat in sausages by Lentinula edodes and Pleaurotus eryngii mushrooms, but this will lead to the deterioration of the gel of sausage products and seriously affect the sensory quality of sausages. In this study, the response surface method was used to optimize the amount of balsa fish skin gelatin, soy protein isolate, and starch added to, and the proportion of Lentinula edodes mushrooms replacing lean meat in, the new sausage, with Pleaurotus eryngii mushrooms replacing fat. The results show that Lentinula edodes mushrooms replaced 36.1% of the lean meat, and the addition of 0.96% balsa fish skin gelatin, 10.61% starch, and 9.94% soy protein isolate resulted in the highest sensory score and the sensory quality being the closest to that of traditional sausages. Compared with the control group, this novel sausage exhibits characteristics such as lower fat and saturated fatty acid content, reduced energy levels, and higher levels of amino acids (aspartic acid, glutamic acid, cysteine, methionine, and proline) and polyunsaturated fatty acids. The total phenolic content of the novel sausage is 12.52 times higher than that of the control. In comparison with the control group, the novel sausage demonstrates a 65.58% increase in DPPH radical scavenging activity and a 3.88-fold improvement in ABTS+ radical scavenging activity. These findings highlight the outstanding antioxidant performance of the novel sausage. This study provides new ideas for improving the sensory quality of new sausages, promoting the healthy development of the sausage industry, and promoting the high-value utilization of edible mushrooms.

Abstract:This study investigates vacuum impregnation technology, a non-thermal approach to treat local beef cubes (2 cm × 2 cm × 2 cm) with three distinct marinades: distilled water (CONTROL), 10% (v/v) pineapple juice (PINE), and 10% (v/v) papaya juice (PAPAYA), both containing plant proteases (bromelain and papain). The primary objective was to evaluate vacuum impregnation (VI) and traditional marination (TM) techniques and assess their impact on beef’s physicochemical attributes, shelf-life stability, and its plant protease activity. Shelf-life tests over 5 days revealed that all beef samples’ pH reached its lowest on day 4, subsequently increasing on day 5. Throughout storage, colour parameters (L*, a*, b*, and ∆E) of treated samples consistently decreased. VI-PINE exhibited the lowest water holding capacity (WHC) over 5 days. Texture profile analysis (TPA) indicated that VI samples' hardness significantly reduced in VI-PINE and VI-PAPAYA compared to TM samples, attributed to coupled plant proteases. VI-PINE showed the highest marinade uptake improvement (21.6%); while having the lowest WHC (15.52%), and all samples' pH ranged in 5.15–6.12. The colour parameters of treated samples increased compared to raw untreated samples. Compared to TM, VI-PINE (1115 g) and VI-PAPAYA (1333 g) significantly reduced beef hardness, with VI-marinated beef also exhibiting higher tyrosine release compared to TM techniques. The application of VI on beef cubes with plant protease-containing marinades provides practical insights for the food industry, offering potential avenues to enhance beef quality, improve shelf life, and optimize flavour incorporation.

Abstract:A DIA-based quantitative proteomic strategy was used to investigate the effects of different cooking procedures (steaming and boiling) on pork meat quality. Results showed that steamed meats had higher redness, cohesion, springiness, but lower lightness, yellowness, shear force, hardness, chewiness and cooking loss than boiled meats. In total of 1608 proteins were identified and 103 proteins exhibited significant difference (fold change 1.5, P < 0.05). These DAPs mainly involved in protein structure, metabolic enzyme, protein turnover and oxidation stress. ALDOC, PVALB, PPP1R14C, AMPD1, CRYAB and SOD1 were validated as potential indicators of color variations in cooked meat. CFL1, COL1A1, COL3A1, RTN4, NRAP, NT5C3A, and SOD1 might be potential biomarker for texture changes of cooked meats. Moreover, these validated proteins exhibited significant (P < 0.05) correlation with cooking loss and could be serve as candidate predictors for cooking loss changes of meats in different cooking procedures.
Evaluation of low-temperature ultrasonic marination of pork meat at various frequencies on physicochemical properties, myoglobin levels, and volatile compounds

Abstract:This study aims to evaluate the pork meat quality after ultrasonic brining at different frequencies, thereby providing a more comprehensive understanding of the effects of ultrasound marination on meat. The texture profile analysis showed that ultrasonic curing at various frequencies significantly improved the textural properties of samples, especially at 26.8 kHz, resulting in a reduction of tenderness, hardness, and chewiness values by 44%, 43%, and 44%, respectively. The cooking loss of samples marinated by ultrasound decreased from 27% without ultrasonic treatment to 22%, indicating a significant improvement in water-holding capacity, while the changes in pH had only a subtle impact on pork quality. Meanwhile, the color of pork became more rosy hue due to decreased L⁎ values and increased a⁎ values, which was mainly attributed to an elevated proportion of oxymyoglobin and reduced metmyoglobin content. Additionally, ultrasonic marination did not exert a negative impact on the oxidation of pork protein and lipids. After roasting, samples marinated by ultrasound exhibited a significantly higher abundance of volatile flavor compounds compared to static marinated meat (with an increase of 16 flavor substances) and fresh pork (with an increase of 24 flavor substances), demonstrating the efficacy of ultrasonic marination in enhancing the overall flavor and taste profile of pork. Consequently, the application of ultrasonic technology holds great potential for the "home kitchen type" rapid marination.

Abstract:This study aimed to determine the effects of fermented mixed feed (FMF) supplementation (0%, 5% and 10%) on the intestinal microbial community and metabolism, and the compositions of volatile flavor compounds and inosine monophosphate (IMP) contents in the longissimus thoracis. In this study, 144 finishing pigs (Duroc x Berkshire x Jiaxing Black) were randomly allocated to 3 groups with 4 replicate pens per group and 12 pigs per pen. The experiment lasted 38 days after 4 days of acclimation. The 16S rRNA gene sequences and an untargeted metabolomics analysis showed FMF altered the profiles of microbes and metabolites in the colon. Heracles flash GC e-nose analysis showed that 10% FMF (treatment 3) had a greater influence on the compositions of volatile flavor compounds than 5% FMF (treatment 2). Compared to 0% FMF (treatment 1), the contents of total alde-hydes, (E,E)-2,4-nonadienal, dodecanal, nonanal and 2-decenal were significantly increased by treatment 3, and treatment 3 increased IMP concentrations and gene expressions related to its synthesis. Correlations analysis showed significantly different microbes and metabolites had strong correlations with the contents of IMP and volatile flavor compounds. In conclusion, treatment 3 regulated intestinal microbial community and metabolism, that in turn altered the compositions of volatile compounds, which contributed to improving pork flavor and umami.

Abstract:This study was designed to explore the effects of marbling on meat quality characteristics and intramuscular connective tissue of beef longissimus muscle. Chemical determinations, histological and mechanical measurements were performed on the raw and cooked meat at d 4 postmortem. The results showed that crude fat, collagen, fiber diameter and maximum transition temperature of intramuscular connective tissue increased (p<0.05) with the increase of marbling score. Abstract:A liquid chromatographic/tandem mass spectrometric method using pneumatically assisted electrospray ionisation (LC/ESI-MS/MS) was developed for the determination of gangliosides GD3 and GM3 in milk and infant formulae. The gangliosides were extracted in a chloroform/methanol/water environment and cleaned up by solid-phase extraction (SPE) on an end-capped C8 sorbent. The gangliosides were detected in negative ion mode after separation on a reversed-phase (RP) C5 analytical column. From the different ganglioside molecular species, product ions at m/z 290 corresponding to an N-acetylneuraminic acid fragment were produced in the collision cell and used in selected reaction monitoring. A standard addition technique was applied for quantification. The relative repeatability standard deviations were less than 5% for GD3 (level 10 mg/L) and 14% for GM3 (level 0.1-0.2 mg/L).

Abstract:Meat alternatives are in great demand in current situation to address the increasing human health and animal welfare issues. This study focuses on microbial protein meats, emphasizing the importance of sourcing structurally similar fungal strains and optimizing processing conditions. A new strain, Neurospora intermedia, was identified and evaluated for availability. The impact of pH (3, 5, and 7) and soluble starch (SS) addition (0, 3, and 6%) on the textural and water retention properties of composite mycoprotein gel meat (CMGM) developed by using N. intermedia was investigated. Zeta potential, dynamic rheology, SEM, and FTIR techniques were employed to elucidate the mechanism. CMGM at pH 7 exhibited the sparsest internal structure but demonstrated better elasticity and recovery properties due to strong electrostatic repulsion. Lowering pH enhanced material interaction, improving gel intensity and water-holding capacity after steaming. A 6% SS addition significantly enhanced CMGM's hardness and reduced internal porosity, which further enhanced the water retention, but also reduced fracture elongation. Thus a 3% SS addition was found to be moderate. The mycelium and gel in the system exhibited complementary characteristics. SPI and SS were dispersed under the guidance of the mycelial fiber framework before steaming. The materials were fully crosslinked after steaming and the mycelial fibers were fixed in the gel material, which conferred support to the gel material. As a result, the modulation of excipients can achieve CMGM with different properties—a systematic approach to producing high-quality microbial protein meat.

Abstract:Leisure dried tofu (LD-tofu) was prepared using two different marinating processes: the repeated heating method (RHM) and the vacuum pulse method (VPM). The quality characteristics and bacterial community succession of LD-tofu and the marinade were evaluated. The results showed that the nutrients in LD-tofu were easily dissolved into the marinade during the marinating process, while the protein and moisture content of RHM LD-tofu changed most dramatically. With the increase in marinade recycling times, the springiness, chewiness and hardness of VPM LD-tofu increased significantly. The total viable count (TVC) of the VPM LD-tofu decreased from the initial value of 4.41 lg cfu/g to 2.51-2.67 lg cfu/g as a result of the marinating process, which had a significant inhibitory effect. Additionally, 26, 167 and 356 communities in the LD-tofu and marinade were detected at the phylum, family and genus levels, respectively. Pearson correlation analysis showed that Pseudomonadaceae, Thermaceae and Lactobacillaceae were closely related to the quality characteristics of LD-tofu, whereas Caulobacteriaceae, Bacillaceae and Enterobacteriae were closely related to the marinade. The present work provides a theoretical basis for the screening of functional strains and quality control in LD-tofu and marinade.

Abstract:Modern Western medical individualism has had a significant impact on health care in China. This essay demonstrates the ways in which such Western-style individualism has been explicitly endorsed in China's 2010 directive: The Basic Norms of the Documentation of the Medical Record. The Norms require that the patient himself, rather than a member of his family, sign each informed consent form. This change in clinical practice indicates a shift toward medical individualism in Chinese healthcare legislation. Such individualism, however, is incompatible with the character of Chinese familism that is deeply rooted in the Chinese ethical tradition. It also contradicts family-based patterns of health care in China. Moreover, the requirement for individual informed consent is incompatible with numerous medical regulations promulgated in the past two decades. This essay argues that while Chinese medical legislation should learn from relevant Western ideas, it should not simply copy such practices by importing medical individualism into Chinese health care. Chinese healthcare policy is properly based on Chinese medical familist resources.
Influence of mycelial integrity damaged by ultrasonic treatment on product textural properties and in vitro digestibility

Abstract:This study investigated the effects of ultrasonic treatment with different intensities (200, 400, and 600 W) and durations (15, 30, and 45 min) on the mycelial integrity, product quality, and in-vitro digestibility of mycoprotein. Results showed that the mycelium was damaged to varying degrees under different ultrasonic parameters. The mycelium showed obvious damage when the time of ultrasonic treatment lasted more than 30 min or the power exceeded 400 W. As a result, the particle size, hardness and chewiness of mycoprotein products were decreased significantly. Compared with the control group (CK), ultrasonic treatment increased the proportion of immobile water and changed the secondary structure of the fungal intracellular protein, which may be the reason for improving in-vitro digestibility of mycoprotein products. This result demonstrated that, by damaging the mycelial integrity and changing the protein secondary structure, ultrasound treatment could decrease the texture characteristics, but improve the water-holding capacity and in-vitro digestibility of products. This study would provide a potential application for the minced meat products by mycoprotein treated with ultrasound.
