Abstract:The objective of this study was to evaluate the preventive activity of methyl jasmonate (MeJA) alone and in combination with antagonistic yeast in suppressing green mold decay in citrus fruit, and to explore the mechanisms involved. At 100 mu mol/L, MeJA inhibited disease incidence and lesion diameter of mold decay compared with the control (P<0.05) The preventive application of Cryptococcus laurentii at 1 x 10(8) cells/mL combined with 100 mu mol/L MeJA reduced green mold incidence compared to the control and the other treatment groups (P<0.05) when tested in wounded citrus fruit inoculated with Penicillium digitatum. MeJA and C laurentii induced higher activity of polyphenol oxidase, peroxidase and catalase than control. Moreover, treatment with MeJA and C. laurentii induced a rise in the mRNA expression level of PR5 (pathogenesis-related protein family 5), which was stronger than in the single-treatment groups and the control. In addition, 100 mu mol/L MeJA improved the rapid proliferation of C. laurentii in citrus fruit wounds. This combined treatment can induce natural resistance and stimulate the proliferation of antagonistic yeast on the fruit surface. (C) 2013 Elsevier B.V. All rights reserved.
Rhodosporidium Paludigenum Induces Resistance and Defense-Related Responses Against Penicillium Digitatum in Citrus Fruit

Abstract:Induced disease resistance against plant pathogens is a promising non-fungicidal decay control strategy. In this study, a potential biocontrol yeast, Rhodosporidium paludigenum, was investigated for its induction of disease resistance against Penicillium digitatum in citrus fruit. The results showed that R. paludigenum is the most effective yeast among three selected yeasts in stimulating the resistance of citrus fruit to green mold. When R. paludigenum was applied 48-72 h before inoculation with P. digitatum, disease incidence and disease severity in citrus fruit significantly decreased. Application of R. paludigenum at concentrations of 1 x 10(8) and 1 x 10 cells mL(-1) respectively resulted in 49.6% and 52.5% reductions in the percentage of infections. Induction of resistance to P. digitatum by R. paludigenum treatment significantly enhanced the activities of defense-related enzymes, including beta-1,3-glucanase, phenylalanine ammonia-lyase, peroxidase, and polyphenoloxidase, which may be an important mechanism by which the biocontrol yeast reduces the fungal disease of citrus fruit caused by P. digitatum. Inhibiting Penicillium Expansum Infection on Pear Fruit by Cryptococcus Laurentii and Cytokinin

Abstract:The effectiveness of the cytokinin N6-benzyladenine (6-BA), alone or in combination with the biocontrol yeast Cryptococcus laurentii, in controlling blue mold on pear fruit was assessed. The application of 6-BA (500–2000μgmL−1) or C. laurentii was effective in reducing Penicillium expansum infection in pear fruit wounds, but its efficacy declined rapidly as the incubation time increased. Integrated application of C. laurentii and 6-BA at 500–2000μgmL−1, especially at the optimal concentration (1000μgmL−1), resulted in a more effective and stable inhibition of the mold rots than that of the 6-BA or C. laurentii alone. Treatments of pears with 6-BA at 1000μgmL−1, alone or with C. laurentii also led to an increase in catalase activity and an inhibition of the activities of both peroxidase and lipoxygenase as well as ethylene production. In addition, 6-BA from 20 to 2000μgmL−1 did not influence the population growth of C. laurentii in pear fruit wounds. These data suggested that a combination of 6-BA and C. laurentii could integrate the dual biological activities from 6-BA and C. laurentii and might be developed into a novel protection strategy for reduction of the blue mold rot of pear fruit.
Resistance monitoring of Penicillium digitatum from Gannan navel orange to commonly used fungicides

Abstract:The frequency and level of resistance of Penicillium digitatum population from 7 counties of Gannan,Jiangxi Province to commonly used fungicide imazalil,prochloraz,thiophanate-methyl and iminoctadine tris,as well as the resistant molecular mechanism to imazalil were studied.The results showed that the consistent resistance to imazalil and prochloraz was presented in P.digitatum.The resistant frequencies of P.digitatum to imazalil were 82% and 90% in 2011 and 2012,respectively.The average resistant factor was 51.5.The resistance molecular mechanism of P.digitatum to imazalil from Gannan belonged to IMZ-R3,which was conferred by an insertion mutation of 199 bp in the promoter region of CYP51B gene.The resistant frequencies of P.digitatum to thiophanate-methyl were 82% and 91% in 2011 and 2012,respectively.However,all strains tested in the study were sensitive to fungicide iminoctadine tris.The study provided a scientific basis for the choice of fungicides in the post-harvest disease control of citrus.
The mechanism involved in enhancing the biological control efficacy of Rhodotorula mucilaginosa with salicylic acid to postharvest green mold decay of oranges

Abstract:The main postharvest pathogen of citrus fruit is Penicillium digitatum. The present study looked at the role of Rhodotorula mucilaginosa enhanced with 0.2 mM salicylic acid (SA) on pH, lignin content accumulation and growth dynamics as a resistance mechanism against P. digitatum in orange fruit. Our findings revealed that the increase in lignin content at 20 °C storage temperature resulted in the significant decrease (P ≤ 0.05) in the lesion diameter in fruit treated with R. mucilaginosa enhanced with or without 0.2 mM SA compared to the untreated fruit (control). In addition, the pH values of the fruit treated with R. mucilaginosa enhanced with or without 0.2 mM SA were 4.43 ± 0.07 and 4.15 ± 0.11, respectively, around the infection site compared to the untreated group. Moreover, the in vivo trial showed that the addition of 0.2 mM SA to the antagonist augmented its growth, and subdued substantially the hyphae and spore germination of P. digitatum in vitro. Also, lower levels of malondialdehyde (MDA) and hydrogen peroxide (H2O2) content were observed in fruit treated with the antagonist enhanced with or without 0.2 mM SA compared to the control. The results established that pH, lignin content accumulation and growth of the yeast played a significant role in the control of green mold decay of orange fruit.

Abstract:Penicillium italicum, the cause of citrus blue mold, is a pathogenic fungus that seriously affects the postharvest quality of citrus fruit and causes serious economic loss. In this study, a eugenol nanoemulsion containing limonin, an antimicrobial component from citrus seeds, was prepared using a high-pressure microfluidizer and the antifungal activity of the nanoemulsions against P. italicum was evaluated based on the conidial germination rate, mycelial growth, and scanning electron microscopy analysis. The results showed that the minimum inhibitory concentration and the inhibition rate of limonin-loaded eugenol nanoemulsion was 160 μg/mL and 59.21%, respectively, which was more potent than that of the limonin-free eugenol emulsion. After treatment with the nanoemulsions, the integrity of the P. italicum cell membrane was disrupted, the cell morphology was abnormal, and the leakage of nucleic acid and protein was observed. In addition, the challenge test on citrus fruits revealed that the limonin-loaded eugenol emulsion inhibited citrus infection for longer periods, with an infection rate of 29.2% after 5 days. The current research shows that nanoemulsions containing limonin and eugenol have effective antifungal activity against P. italicum, and may be used as a substitute for inhibiting blue mold in citrus fruits.

Abstract:Citrus fruit were easily infected by Penicillium digitatum, and caused green mold rapidly, resulting in enormous post-harvest losses. ε-poly-l-lysine (ε-PL) was generally regarded as a safe (GRAS) substance. Besides, it was proved to have a dual effect on harming fungi and triggering fruit defense responses. Fatty acid metabolism is closely related to fruit defense response. However, little is known about how ε-PL affected fatty acid metabolism in citrus fruit. Here, we found that ε-PL increased the expression of CsFATA, CsACSL, CsFAD2, CsFAD3, CsLOX2S, and CsHPL in fatty acid metabolism, decreasing oleic acid levels and enhancing linoleic and linolenic acid levels. Additionally, ε-PL enriched the activities of LOX and HPL during the oxidative decomposition of fatty acids, and activating C9 aldehyde biosynthesis. Interestingly, ε-PL combined with (2E,4E)-nonadienal (C9 aldehyde) would improve the inhibitory effect against Penicillium digitatum. And the combined bio-fungicide significantly delayed the citrus green mold compared to single concentrations of the individual components. These results suggested that ε-PL improved citrus fruit defense responses through fatty acid-mediated defense responses. Combined bio-fungicide consisting of ε-PL and (2E,4E)-nonadienal have an excellent prospect for controlling citrus green mold.

Control of citrus blue and green molds by Actinomycin X2 and its possible antifungal mechanism

Abstract:Citrus blue and green molds caused by Penicillium digitatum, P. italicum, and P. polonicum, are the major postharvest diseases of citrus fruit. In the present study, Actinomycin X2 (Act-X2), a naturally occurring antibiotic produced by Streptomyces species, was found to show excellent antifungal effect against these three pathogens with a minimum inhibitory concentration (MIC) value of 62.5 μg/mL for them all, which was better than the positive control thiophanate-methyl. Act-X2 significantly reduced the percentage of spore germination, and highly inhibited the mycelial growth of P. italicum, P. digitatum, and P. polonicum with EC50 values being 34.34, 13.76, and 37.48 μg/mL, respectively. In addition, Act-X2 greatly decreased the intracellular protein content while increasing the reactive oxygen species (ROS) level and superoxide anion (O2-) content in the mycelia of pathogens. In vivo test indicated that Act-X2 strongly inhibited the infection of navel oranges by these three Penicillium species, with an inhibition percentage of >50% for them all at the concentration of 10 MIC. Transcriptome analysis suggested that Act-X2 might highly influence the ribosomal functions of P. polonicum, which was supported as well by the molecular docking analysis of Act-X2 with some key functional proteins and RNAs of the ribosome. Furthermore, Act-X2 significantly reduced the decay percentage and improved the firmness, color, and sugar-acid ratio of navel oranges spray-inoculated with P. polonicum during the postharvest storage at 4 °C for 60 d.

Biological and physical approaches to improve induced resistance against green mold of stored citrus fruit

Abstract:Health and environmental concerns have point out the need to improve or change several manufacturing steps in the food chain. In this context particular attention should be given to the technologies involved in fruits and vegetables production. Nearly all fresh fruit and vegetables are subjected to different periods of storage and/or shelf-life before of their consumption. This implies the need to protect the commodities from microbial spoilage. Some Citrus species (e.g. lemon and grapefruit) may be stored for several months before consumption and then post-harvest treatments are essential to contain green (Penicillium digitatum) and blue (P. italicum) moulds. Alternative approaches to chemicals usually have a lower efficacy in containing rots but fulfill the consumer's expectation. Among the alternative strategies, the improvement of host natural resistance is promising. In this regard, we report some results concerning the use of biotic (yeast) and abiotic agents as inducers of phytoalexin (i.e. scoparone and/or scopoletin) accumulation in Citrus rind and its importance in the control of fungal decay. In all experiments the inducers were applied on fruits before or 24 h after inoculation with P. digitatum and the rot severity was monitored 7 days later. The accumulation of phytoalexins was monitored according to a standard methodology by HPLC. In all experiments a positive correlation was found between increase of the phytoalexin scoparone in host tissue and reduction of decay.

Abstract:Sleep disturbances in obstructive sleep apnea are caused mainly by repetitive apneas and hypopneas. An alternative factor contributing to disordered sleep may be the obesity, which is frequently associated with sleep apnea. The sleep disturbing effect of obesity was found previously in obese nonapneic subjects. The aim of this study was to evaluate the effect of obesity on sleep quality in sleep apnea patients in particular in patients under continuous positive airway pressure (CPAP) with successfully normalized respiration. We reviewed the archive data of 18 non-obese, 18 obese and 17 severely obese age and gender matched sleep apnea patients treated with CPAP. The polysomnographic parameters from the diagnostic night, from the second night under CPAP and from the follow up night (after three months of CPAP use) were compared. Before CPAP the apnea hypopnea index was worse in obese and in severely obese group and it normalised under CPAP in all groups. The severely obese group showed more light sleep and less REM sleep before CPAP and inversely - less light and more REM sleep in the second night under CPAP than the non-obese group. In the follow up, there was no differences in sleep profile between groups. This study indicates obesity does not affect the sleep independently of respiratory disorders. Before therapy it is associated with more severe sleep apnea and indirectly with worse sleep quality.

Abstract:Green mold disease, a common citrus post-harvest disease caused by Penicillium digitatum, has an unresolved initial infection mechanism. Understanding the infection mechanism leads to the development of potential controls and preventive measures against the disease. The present study aimed to delineate the infection mechanism by investigating spore germination, changes of organic molecules and enzyme activity, and differential expression of genes in the P. digitatum infection. P. digitatum spore germination was observed by a pathology section scanner and it was found that in vivo germination was 3 h behind the in vitro germination. In addition, cell wall degrading enzymes and soluble sugar and titratable acid content during the infection process measured dynamically. The level of pectinase reached its maximum of 6067 U/g before 48 hpi, while cellulase increased rapidly after 48 hpi. The soluble sugar and organic acid content increased considerably with the progression of the infection. The transcriptomic profile of P. digitatum before and after infection was analyzed by RNA-seq. The genes related to cell wall degrading enzymes were significantly up-regulated and annotated to participate in two major carbon source synthesis pathways. The study delineated the initial infection mechanism of P. digitatum which eventually opened the gate way for the development of new control strategies in the future.
Rhodosporidium Paludigenum Induced Resistance in Ponkan Mandarin Against Penicillium Digitatum Requires Ethylene-Dependent Signaling Pathway

Abstract:This work determined if the ethylene dependent signal pathway was required for antagonist-mediated fruit defense mechanisms through investigation of disease resistance against Penicillium digitatum in Ponkan mandarin induced by 1-methylcyclopropene (1-MCP), an inhibitor of ethylene perception, and Rhodosporidium paludigenum. Blocking ethylene perception with 1-MCP resulted in an increase in ACS1, ACS2 and ACO expression, and consequently an increase in ethylene production during mechanical wounding and resistance induction. The expression of the ethylene receptors ETR1, ETR2 and ETR4 as well as ethylene response factor (ERF) were observed with similar responses to yeast and 1-MCP stimuli, with ETR3 mRNA accumulation being the most sensitive to yeast application while ERS1 was the least sensitive. When applied at concentrations greater than 500 nL L-1, 1-MCP pre-fumigation significantly reduced the fruit's natural protection and R. paludigenum induced disease resistance to Penicillium decay, indicating that ethylene perception was involved in inducting disease resistance. Moreover, expression of the defensive genes CHI, beta-1,3-glucanase, PAL and CIN up-regulated by yeast was inhibited to different degrees by the 1-MCP pre-treatment. This study provides evidence that the biocontrol yeast R. paludigenum increased disease resistance in Ponkan mandarin against P. digitatum infection due to ethylene and signaling pathway dependent mechanisms. Effects of postharvest oligochitosan treatment on anthracnose disease in citrus (Citrus sinensis L. Osbeck) fruit

Abstract:Citrus fruit (Citrus sinensis L. Osbeck) is susceptible to infection by Colletotrichum gloeosporioides during poststorage, which rapidly decreases sensory and nutritional quality of the fruit. The ability of oligochitosan treatment to control C. gloeosporioides of citrus fruit during storage was examined, and possible underlying mechanisms were discussed. Disease incidence and lesion diameter were lower in oligochitosan-treated fruits compared with their respective controls. The fruits dipped in oligochitosan showed increased contents of lignin, hydroxyproline-rich glycoprotein (HRGP), hydrogen peroxide (H2O2), ascorbate, glutathione, total phenol, and flavonoid compounds. In addition, enzymatic activities of superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), peroxidase (POD, EC 1.11.1.7), polyphenoloxidase (PPO, EC 1.14.18.1), ascorbate peroxidase (APX, EC 1.11.1.11), and β-1, 3-glucanase (GLU, EC 3.2.1.39) also increased in citrus fruit peels, all of which were correlated with the onset of induced disease resistance. These results indicated that oligochitosan treatment can induce disease resistance of citrus fruit to C.gloeosporioides Penz. Oligochitosan can be a potential alternative to conventional control methods of postharvest anthracnose in citrus fruit.

Abstract:Sodium dehydroacetate (SD) has been widely used in various fields due to its high antimicrobial activities. However, the inhibitory mechanisms of SD against fungi, especially on transcriptional level, are still unclear. In this study, the inhibitory property of SD against Penicillium digitatum (imazalil (IMZ)-sensitive P. digitatum (Pds01) and IMZ-resistant P. digitatum (Pdw03) strains) was investigated. Here, we found that SD treatment had a distinct inhibitory effect on Pdw03 strain in vitro and mitigated the disease prevalence of inoculated citrus. Morphology and microstructure observation showed that SD caused remarkable changes in the hyphae of Pds01 and Pdw03. Transcriptomic analysis displayed that SD treatment blocked the expression of genes related to the ribosome, genetic information processing and energy metabolism, while promoting the genes expression related to cell walls degradation, membrane lipid metabolism and biosynthesis of sphingolipid. Among them, the ribosomal pathway was most prominent, and key genes in this pathway (RPSA, RPS9, RPL7A and RPL5) were inhibited by SD treatment in both strains. Moreover, we found that the expression of 12 MFS (major facilitator superfamily) transporters, 6 ABC (ATP-binding cassette transporter) transporters, and 9 glutathione metabolism encoding genes related to SD detoxification were increased. The results of fourier transform infrared spectroscopy (FT-IR) fingerprints further confirmed that SD caused remarkable changes in cell membrane lipids, nucleic acids, proteins and cell wall polysaccharides in both strains. These findings indicated that the interference with ribosome, genetic information processing, cell membrane metabolism and energy metabolism might be closely involved in the antifungal mechanism of SD against P. digitatum. Our results may provide novel insights into the antifungal molecular mechanism of SD against P. digitatum and deepen our understanding of the regulatory network of SD.

Abstract:It has been reported that Yarrowia lipolytica can effectively control blue mold and green mold decay of mandarins (Citrus reticulata), and also the possible physiological mechanisms underlying this efficacy were studied in our previously research. However, the related molecular mechanisms were not clearly. In this study, the molecular mechanisms of disease-resistance in mandarins induced by Y. lipolytica were investigated. It was found that, after the treatment of Y. lipolytica, the related resistance enzyme activities of β-1,3-glucanase, superoxide dismutase, and ascorbate peroxidase in mandarins were enhanced, while the malondialdehyde content was decreased. Interestingly, based on the transcriptome analysis, it was found that Y. lipolytica can induce the up-regulation of genes related to the synthesis of resistant substances; key genes in the pathway of plant-pathogen interaction; genes related to scavenging reactive oxygen species; genes related to jasmonic acid, salicylic acid and ethylene signal transduction pathway in mandarins. Furthermore, Y. lipolytica down-regulated the expression levels of genes encoding cell wall degrading enzymes. Definitively, this unravelling of the molecular mechanisms of Y. lipolytica will contribute to consolidate its potential to serve as an effective biocontrol agent against post-harvest diseases of mandarins.

Abstract:Thirty-three patients with JRA were treated with proquazone during a 4-wk open-labeled, non-controlled trial. Increasing doses started at 400 mg/m2/d up to 800 mg/m2/d, not to exceed 960 mg/d. Oral administration was 4 times/d. One patient did not complete the study because an erythematous rash developed. Efficacy analysis showed significant decreases in the total number of active joints, the number and severity of joints with tenderness and LOM, DMS, and travel time (in all cases p less than .05). The drug appears to be similar in efficacy and safety to other NSAID that have been studied in children.
Mechanism of Cell Wall Polysaccharides Modification in Harvested ‘Shatangju’ Mandarin (Citrus reticulate Blanco) Fruit Caused by Penicillium italicum

Abstract:Modification of cell wall polysaccharide in the plant plays an important role in response to fungi infection. However, the mechanism of fungi infection on cell wall modification need further clarification. In this study, the effects of Penicillium italicum inoculation on ‘shatangju’ mandarin disease development and the potential mechanism of cell wall polysaccharides modification caused by P. italicum were investigated. Compared to the control fruit, P. italicum infection modified the cell wall polysaccharides, indicated by water-soluble pectin (WSP), acid-soluble pectin (ASP), hemicellulose and lignin contents change. P. italicum infection enhanced the activities of polygalacturonase (PG), pectin methylesterase (PME), and the expression levels of xyloglucanendotransglucosylase/hydrolase (XTH) and expansin, which might contribute to cell wall disassembly and cellular integrity damage. Additionally, higher accumulation of reactive oxygen species (ROS) via decreasing antioxidant metabolites and the activities of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX) also contributed to the cell wall polysaccharides modification. Meanwhile, the gene expression levels of hydroxyproline-rich glycoprotein (HRGP) and germin-like protein (GLP) were inhibited by pathogen infection. Altogether, these findings suggested that cell wall degradation/modification caused by non-enzymatic and enzymatic factors was an important strategy for P. italicum to infect ‘shatangju’ mandarin.

Abstract:Combined use of biocontrol agents and plant extracts can be considered a viable and promising strategy for protecting plant tissues with different synergistic mechanisms of action that improve the antimicrobial activity of the mixtures. Treatments of citrus fruits with Wickerhamomyces anomalus BS91 have been previously reported as effective measures to reduce the incidence of green mold caused by Penicillium digitatum. On the opposite, the knowledge of the antifungal activity of cultivated cardoon (Cynara cardunculus L. var. altilis DC.) leaf extract, vegetable widespread in some Mediterranean areas, is still very limited. In this study, experimental trials were conducted to evaluate the effectiveness in vitro of leaf aqueous, methanolic and ethanolic extracts of C. cardunculus against seven fungal pathogens responsible for considerable food losses in the postharvest stage. In addition, biocontrol yeast W. anomalus BS91 and the three C. cardunculus extracts were tested in vivo both as a single treatment and in mixture, against Penicillium digitatum on 'Tarocco' oranges and 'Femminello' lemons. The combination of W. anomalus BS91 and leaf ethanolic extract reduced with the highest efficacy the incidence and severity of green mold on orange and lemon fruits with respect to the control, and was more effective than treatment with antagonistic yeast or leaf extracts applied alone. Incidence and severity of citrus decay were more consistently reduced when mixtures were applied 24 h before the inoculation of the pathogen, thus suggesting the relevance of preventive treatments. The mixtures of antagonistic W. anomalus BS91 and ethanolic leaf extract were more effective in controlling green mold decay on oranges than on lemons. These results indicate that biocontrol agents and leaf extracts, used in appropriate combination, can provide a stronger protection than when used singularly. However, compatibility between microbial antagonist and antimicrobial extract should be preliminary verified.
