Abstract:The objective of this study was to evaluate the preventive activity of methyl jasmonate (MeJA) alone and in combination with antagonistic yeast in suppressing green mold decay in citrus fruit, and to explore the mechanisms involved. At 100 mu mol/L, MeJA inhibited disease incidence and lesion diameter of mold decay compared with the control (P<0.05) The preventive application of Cryptococcus laurentii at 1 x 10(8) cells/mL combined with 100 mu mol/L MeJA reduced green mold incidence compared to the control and the other treatment groups (P<0.05) when tested in wounded citrus fruit inoculated with Penicillium digitatum. MeJA and C laurentii induced higher activity of polyphenol oxidase, peroxidase and catalase than control. Moreover, treatment with MeJA and C. laurentii induced a rise in the mRNA expression level of PR5 (pathogenesis-related protein family 5), which was stronger than in the single-treatment groups and the control. In addition, 100 mu mol/L MeJA improved the rapid proliferation of C. laurentii in citrus fruit wounds. This combined treatment can induce natural resistance and stimulate the proliferation of antagonistic yeast on the fruit surface. (C) 2013 Elsevier B.V. All rights reserved.
Rhodosporidium Paludigenum Induces Resistance and Defense-Related Responses Against Penicillium Digitatum in Citrus Fruit

Abstract:Induced disease resistance against plant pathogens is a promising non-fungicidal decay control strategy. In this study, a potential biocontrol yeast, Rhodosporidium paludigenum, was investigated for its induction of disease resistance against Penicillium digitatum in citrus fruit. The results showed that R. paludigenum is the most effective yeast among three selected yeasts in stimulating the resistance of citrus fruit to green mold. When R. paludigenum was applied 48-72 h before inoculation with P. digitatum, disease incidence and disease severity in citrus fruit significantly decreased. Application of R. paludigenum at concentrations of 1 x 10(8) and 1 x 10 cells mL(-1) respectively resulted in 49.6% and 52.5% reductions in the percentage of infections. Induction of resistance to P. digitatum by R. paludigenum treatment significantly enhanced the activities of defense-related enzymes, including beta-1,3-glucanase, phenylalanine ammonia-lyase, peroxidase, and polyphenoloxidase, which may be an important mechanism by which the biocontrol yeast reduces the fungal disease of citrus fruit caused by P. digitatum. Inhibiting Penicillium Expansum Infection on Pear Fruit by Cryptococcus Laurentii and Cytokinin

Abstract:The effectiveness of the cytokinin N6-benzyladenine (6-BA), alone or in combination with the biocontrol yeast Cryptococcus laurentii, in controlling blue mold on pear fruit was assessed. The application of 6-BA (500–2000μgmL−1) or C. laurentii was effective in reducing Penicillium expansum infection in pear fruit wounds, but its efficacy declined rapidly as the incubation time increased. Integrated application of C. laurentii and 6-BA at 500–2000μgmL−1, especially at the optimal concentration (1000μgmL−1), resulted in a more effective and stable inhibition of the mold rots than that of the 6-BA or C. laurentii alone. Treatments of pears with 6-BA at 1000μgmL−1, alone or with C. laurentii also led to an increase in catalase activity and an inhibition of the activities of both peroxidase and lipoxygenase as well as ethylene production. In addition, 6-BA from 20 to 2000μgmL−1 did not influence the population growth of C. laurentii in pear fruit wounds. These data suggested that a combination of 6-BA and C. laurentii could integrate the dual biological activities from 6-BA and C. laurentii and might be developed into a novel protection strategy for reduction of the blue mold rot of pear fruit.
Effect of Epiphytic Bacteria from Citrus against Green Mold Post-Harvest Diseases of Citrus

Abstract:This study investigates the potential of epiphytic bacteria isolated from citrus fruits to control green mold post-harvest disease caused by Penicillium digitatum in Thompson Navel sweet oranges in the north of Iran. Twenty-three epiphytic bacteria were isolated and screened in vitro against P. digitatum, and 13 isolates with antagonistic activity were selected for further studies. Isolates B15 (Bacillus sp.), P17 (Pseudomonas sp.), and S13 (Streptomyces sp.) exhibited the most effective inhibitory effects on P. digitatum in vitro and in vivo. Among these, B15 showed the highest percentage of mycelium growth reduction and was further identified as B. cereus by 16S rRNA sequence analysis. Metabolic analysis of Bacillus sp. isolate B15 extract revealed the presence of the inhibiting antifungal substance Iturin A. The result showed that the application of epiphytic B. cereus decreased the percentage of green mold post-harvest diseases in orange fruit. This indicates their potential as an environmentally friendly alternative to chemical post-harvest control of orange diseases caused by P. digitatum.

Abstract:Managing post-harvest decays in citrus fruit without relying on conventional pesticides presents a significant challenge in modern Plant Pathology. This study aimed to evaluate the efficacy of the biological control agent strain O in controlling green mold caused by throughout various stages of the post-harvest supply chain. Using a series of experiments, different scenarios of infections in clementine tangerine, orange, and lemon fruit were examined, with treatments applied before, during or after infection. The study simulated typical conditions of the citrus supply chain, including picking, processing in packinghouses, and transportation, as well as cold storage and shelf-life phases. Results indicated that exhibited significant efficacy in reducing green mold symptoms, even at shelf-life temperatures, making it a practical alternative to conventional fungicides. Additionally, the study provided insights into the molecular mechanisms underlying the defensive response of citrus fruit to treatment, with up-regulation of defense-related genes observed across different fruit types. Overall, this study underscores the potential of as a sustainable and effective solution for managing post-harvest decays in citrus fruit within the complexities of the supply chain.

Control of citrus blue and green molds by Actinomycin X2 and its possible antifungal mechanism

Abstract:Citrus blue and green molds caused by Penicillium digitatum, P. italicum, and P. polonicum, are the major postharvest diseases of citrus fruit. In the present study, Actinomycin X2 (Act-X2), a naturally occurring antibiotic produced by Streptomyces species, was found to show excellent antifungal effect against these three pathogens with a minimum inhibitory concentration (MIC) value of 62.5 μg/mL for them all, which was better than the positive control thiophanate-methyl. Act-X2 significantly reduced the percentage of spore germination, and highly inhibited the mycelial growth of P. italicum, P. digitatum, and P. polonicum with EC50 values being 34.34, 13.76, and 37.48 μg/mL, respectively. In addition, Act-X2 greatly decreased the intracellular protein content while increasing the reactive oxygen species (ROS) level and superoxide anion (O2-) content in the mycelia of pathogens. In vivo test indicated that Act-X2 strongly inhibited the infection of navel oranges by these three Penicillium species, with an inhibition percentage of >50% for them all at the concentration of 10 MIC. Transcriptome analysis suggested that Act-X2 might highly influence the ribosomal functions of P. polonicum, which was supported as well by the molecular docking analysis of Act-X2 with some key functional proteins and RNAs of the ribosome. Furthermore, Act-X2 significantly reduced the decay percentage and improved the firmness, color, and sugar-acid ratio of navel oranges spray-inoculated with P. polonicum during the postharvest storage at 4 °C for 60 d.

Abstract:Citrus (Citrus spp.) species produce a variety of fruits that are popular worldwide. Citrus fruits, however, are susceptible to postharvest decays caused by various pathogenic fungi, including Penicillium digitatum, Penicillium italicum, Geotrichum citri-aurantii, Aspergillus niger, and Aspergillus flavus. Decays resulting from infections by these pathogens cause a significant reduction in citrus quality and marketable yield. Biological control of postharvest decay utilizing antagonistic bacteria and fungi has been explored as a promising alternative to synthetic fungicides. In the present article, the isolation of antagonists utilized to manage postharvest decays in citrus is reviewed, and the mechanism of action including recent molecular and genomic studies is discussed as well. Several recently-postulated mechanisms of action, such as biofilm formation and an oxidative burst of reactive oxygen species have been highlighted. Improvements in biocontrol efficacy of antagonists through the use of a combination of microbial antagonists and additives are also reviewed. Biological control utilizing bacterial and yeast antagonists is a critical component of an integrated management approach for the sustainable development of the citrus industry. Further research will be needed, however, to explore and utilize beneficial microbial consortia and novel approaches like CRISPR/Cas technology for management of postharvest decays.

Biological and physical approaches to improve induced resistance against green mold of stored citrus fruit

Abstract:Health and environmental concerns have point out the need to improve or change several manufacturing steps in the food chain. In this context particular attention should be given to the technologies involved in fruits and vegetables production. Nearly all fresh fruit and vegetables are subjected to different periods of storage and/or shelf-life before of their consumption. This implies the need to protect the commodities from microbial spoilage. Some Citrus species (e.g. lemon and grapefruit) may be stored for several months before consumption and then post-harvest treatments are essential to contain green (Penicillium digitatum) and blue (P. italicum) moulds. Alternative approaches to chemicals usually have a lower efficacy in containing rots but fulfill the consumer's expectation. Among the alternative strategies, the improvement of host natural resistance is promising. In this regard, we report some results concerning the use of biotic (yeast) and abiotic agents as inducers of phytoalexin (i.e. scoparone and/or scopoletin) accumulation in Citrus rind and its importance in the control of fungal decay. In all experiments the inducers were applied on fruits before or 24 h after inoculation with P. digitatum and the rot severity was monitored 7 days later. The accumulation of phytoalexins was monitored according to a standard methodology by HPLC. In all experiments a positive correlation was found between increase of the phytoalexin scoparone in host tissue and reduction of decay.
Control of postharvest blue mold of Nanfeng mandarin by application of strain YS-1 Paenibacillus brasilensis

Abstract:In order to study its commercial value, antagonistic spectrum and storage application of YS-1 Paenibacillus brasilensis were investigated in this paper. YS-1 P. brasilensis showed obvious antifungal activity to 5 different fruit pathogens, which was of broad antagonistic spectrum. Effect and application of YS-1 P. brasilensis fermentation liquid on Nanfeng mandarin at different storage temperatures were also investigated with the puncture inoculation method. Results showed that lesion diameter and disease incidence at 25 °C were higher than those at 5 °C after end of the storage, and there was significant difference between them. P. brasilensis fermentation liquid was effective for control of Penicillium italicum on Nanfeng mandarin stored at 5 °C for 25 d or 25 °C for 20 d. Preharvest treatment combined with postharvest treatment significantly reduced the decay rate of Nanfeng mandarin by 5.8% more than the control, particularly in the 1st 2 mo of storage. Fruits treated with P. brasilensis fermentation liquid in preharvest and postharvest period tended to have higher total sugar content, titratable acidity, ascorbic acid (AsA) content, and soluble solids content than those in the control group, and there was significant difference between the 2 groups. A delay was observed in the drop in AsA content. In this article, strain YS-1 is reported for the 1st time as a biocontrol agent against blue mold of Nanfeng mandarin. The research will provide an application reference for preservation of citrus.
Developing Penicillium digitatum Management Strategies on Post-Harvest Citrus Fruits with Metabolic Components and Colonization of Bacillus subtilis L1-21

Abstract:Citrus is among the most important plants in the fruit industry severely infected with pathogens. Citrus green mold caused by Penicillium digitatum is one of the most devastating diseases during post-harvest stages of citrus fruit. In this study, a potential endophyte Bacillus subtilis L1-21, isolated from healthy citrus plants, was assessed for its biocontrol activity against the pathogen P. digitatum. Based on an in vitro crosstalk assay, we suggested that B. subtilis L1-21 inhibits the pathogen with an inhibition zone of 3.51 ± 0.08 cm. Biocontrol efficacy was highest for the fermented culture filtrate of B. subtilis L1-21. Additionally, using GC-MS analysis, 13 compounds were detected in the extract of this endophyte. The culture filtrate in Landy medium could enlarge and deform pathogen spores and prevent them from developing into normal mycelium. Accordingly, the Landy culture filtrate of B. subtilis L1-21 was stable in the temperature range of 4–90 °C and pH of 3–11. Further, MALDI-TOF-MS for B. subtilis L1-21 detected surfactin, fengycin, bacillaene and bacilysin as potential antifungal compounds. GFP-tagged B. subtilis L1-21 easily colonized in citrus fruit peel and pulp, suggesting its role in eliminating the fungal pathogen. Altogether, it is highly expected that the production of antifungal compounds, and the colonization potential of B. subtilis L1-21 are required against the post-harvest P. digitatum pathogen on citrus fruit.

Abstract:The main postharvest pathogen of citrus fruit is Penicillium digitatum. The present study looked at the role of Rhodotorula mucilaginosa enhanced with 0.2 mM salicylic acid (SA) on pH, lignin content accumulation and growth dynamics as a resistance mechanism against P. digitatum in orange fruit. Our findings revealed that the increase in lignin content at 20 °C storage temperature resulted in the significant decrease (P ≤ 0.05) in the lesion diameter in fruit treated with R. mucilaginosa enhanced with or without 0.2 mM SA compared to the untreated fruit (control). In addition, the pH values of the fruit treated with R. mucilaginosa enhanced with or without 0.2 mM SA were 4.43 ± 0.07 and 4.15 ± 0.11, respectively, around the infection site compared to the untreated group. Moreover, the in vivo trial showed that the addition of 0.2 mM SA to the antagonist augmented its growth, and subdued substantially the hyphae and spore germination of P. digitatum in vitro. Also, lower levels of malondialdehyde (MDA) and hydrogen peroxide (H2O2) content were observed in fruit treated with the antagonist enhanced with or without 0.2 mM SA compared to the control. The results established that pH, lignin content accumulation and growth of the yeast played a significant role in the control of green mold decay of orange fruit.

Abstract:The objective of this study was to analyze the impact of the marine yeast Rhodosporidium paludigenum and sodium bicarbonate (SBC) treatments applied separately or in combination to control green mold of two cultivars of citrus fruit ( Citrus reticulata Blanco cv. Subcompressa and C. reticulata Blanco cv. Ponkan) caused by Penicillium digitatum . In the assay of C. reticulata Blanco cv. Subcompressa, R. paludigenum at 10 7 cells/mL significantly ( P < 0.05) reduced green mold incidence compared to the control, and the combination of 10 7 cells/mL R. paludigenum and 5 % SBC was as effective as fungicide (500 ppm Fungaflor). In the assay of C. reticulata Blanco cv. Ponkan, the treatment with R. paludigenum at 10 8 cells/mL and 2 % SBC were equally effective and significantly ( P < 0.05) reduced the decay incidence. Moreover, the combination of the two treatments completely eliminated the decay incidence, matching the effectiveness of fungicide. This work demonstrated that proper combination of R. paludigenum and SBC can be an alternative to synthetic fungicides for the control of postharvest green mold decay of citrus.

Abstract:Aureobasidin A (AbA), an antifungal cyclic depsipeptide antibiotic produced by Aureobasidium pullulans R 106, has previously been shown to be effective against a wide range of fungi and protozoa. Here we report the inhibitory effects of AbA on spore germination, germ tuber elongation and hyphal growth of five pathogenic fungi including Penicillium digitatum, P. italicum, P. expansum, Botrytis cinerea and Monilinia fructicola, which are major pathogens causing postharvest diseases of a variety of fruits. AbA inhibited five pathogenic fungi by reducing conidial germination rates, delaying conidial germination initiation, restricting elongation of germ tuber and mycelium, as well as inducing abnormal alternations of morphology of genii tubes and hyphae of these fungi. The sensitivity of these fungi to AbA was pathogen species-dependent. P digitatum was the most sensitive and M.fructicola the least. Importantly, AbA at 50 mu g/ml was effective in controlling the citrus green mold and in reducing the strawberry gray mold incidence and severity, caused by P. digitatum and B. cinerea, respectively, after artificial inoculation. AbA and/or its analogs, therefore, hold promise as relatively safe and promising fungicide candidates to control postharvest decays of fruits, because AbA targets the inositol phosphorylceramide (IPC) synthase, an enzyme essential for fungi but absent from mammals. Abstract:Green mold ( Penicillium digitatum ) and blue mold ( Penicillium italicum ) are among the most economically impactful post-harvest diseases of citrus fruit worldwide. Post-harvest citrus diseases are largely controlled with synthetic fungicides such as pyrimethanil, imazalil, fludioxonil, and thiabendazole. Due to their toxic effects, prolonged and excessive application of these fungicides is gradually restricted in favor of safe and more eco-friendly alternatives. This review comprehensively describes alternative methods for the control of P. digitatum and P. italicum : (a) antagonistic micro-organisms, (b) plant extracts and essential oils, (c) biofungicides, (d) chitosan and chitosan-based citrus coatings, (e) heat treatments, (f) ionizing and non-ionizing irradiations, (g) food additives, and (h) synthetic elicitors. Integrating multiple approaches such as the application of biocontrol agents with food additives or heat treatments have overcome some drawbacks to single treatments. In addition, integrating treatment approaches could produce an additive or synergistic effect on controlling both molds for a satisfactory level of disease reduction in post-harvest citrus. Further research is warranted on plant resistance and fruit-pathogen interactions to develop safer strategies for the sustainable control of P. digitatum and P. italicum in citrus.

Exploring the Bioprotective Potential of Halophilic Bacteria against Major Postharvest Fungal Pathogens of Citrus Fruit Penicillium digitatum and Penicillium italicum

Abstract:Citrus fruits are vulnerable to green mold (caused by Penicillium digitatum) and blue mold (caused by Penicillium italicum) during storage, posing significant challenges to the industry. Therefore, biological control utilizing antagonistic bacteria has emerged as a dependable strategy for managing postharvest diseases. In this study, halophilic bacterial isolates were carefully selected from diverse saline ecosystems, including the Dead Sea, the Agadir Sea, the Rabat Sea, saline soil, and water of the Amassine Oued in Taounate, based on rigorous in vitro and in vivo antagonism bioassays. Out of 21 bacteria from different saline environments, 10 were chosen for further characterization based on the 16S rDNA gene. Notably, the EAM1 isolate demonstrated exceptional inhibitory effects, reaching a 90% inhibition rate against P. digitatum, while the ER2 isolate closely followed with an 89% inhibition rate against P. italicum. Furthermore, in bacterial supernatant experiments, six bacterial isolates effectively curbed the growth of P. digitatum, and three demonstrated efficacy against P. italicum development. In an in vivo trial spanning ten days of incubation, three highly effective isolates against P. digitatum displayed zero severity, and two of these isolates also demonstrated zero severity against P. italicum. Interestingly, a comparison of bacterial filtrates revealed that all isolates exhibited a severity level of over 50% against the pathogen causing green rot (P. digitatum), while the severity was lower than 50% for the supernatants of the two isolates used against P. italicum. In conclusion, this study highlights the promising role of halophilic bacteria, specifically Bacillus amyloliquefaciens EAM1 and B. amyloliquefaciens ER2, in controlling postharvest fruit pathogens. The findings shed light on the potential of utilizing these bioprotective agents to address the challenges posed by green and blue citrus molds, providing valuable insights for the citrus industry.

Abstract:Citrus fruits are subjected to a diversity of postharvest diseases caused by various pathogens during picking, packing, storage and transportation. Green and blue molds, caused by Penicillium digitatum and Penicillium italicum, respectively, are two major postharvest citrus diseases and cause significant economic losses during the commercialization phase. Currently, the control of postharvest citrus diseases relies mainly on the use of synthetic fungicides, which usually result in the resistance against fungal attack, environment pollution and health hazards. In recent years, much attention has been given to the preservation of citrus by naturally isolated edible plant extracts, medicinal plant extracts, Citrus extracts and volatiles, et al. Scientists worldwide devote their time and energy to discover the high effect, low toxicity, safety and inexpensive plant-derived fungicides. The current review will highlight plant-derived fungicides and chemical constituents that aim to inhibit P. digitatum and P. italicum in vitro and in vivo. Coatings enriched with plant extracts could be good alternative methods for Citrus fruits preservation. Problems and prospects of the research and development of plant-derived natural fungicides will also be discussed in this article.
Pantoea vagans strain BWL1 controls blue mold in mandarin fruit by inhibiting ergosterol biosynthesis in Penicillium expansum

Abstract:Penicillium expansum is a major postharvest fungal pathogen of a variety of fruits. Recently, Pen. expansum has been reported to infect citrus fruit; however, the management of Pen. expansum on citrus has not been studied. Pantoea vagans strain BWL1 was isolated from the surface of mandarin fruit. Here, its inhibitory effect on postharvest blue mold on mandarin fruit was evaluated. Pan. vagans inhibited conidial germination and mycelial growth of Pen. expansum. Cell-free fermentation extract of Pan. vagans inhibited the expression of ergC, a gene involved in ergosterol biosynthesis, and decreased the ergosterol content in Pen. expansum, resulting in plasma membrane disruption in the fungal pathogen. In vivo experiments showed that the development of blue mold caused by Pen. expansum on citrus fruit was significantly decreased by treatment with Pan. vagans strain BWL1. These results suggest that Pan. vagans can produce metabolites that exhibit antifungal activity toward Pen. expansum by inhibiting the biosynthesis of ergosterol. The application of Pan. vagans strain BWL1 could be effective for the postharvest management of blue mold in citrus fruit.

Combined application of antagonistic Wickerhamomyces anomalus BS91 strain and Cynara cardunculus L. leaf extracts for the control of postharvest decay of citrus fruit

Abstract:Combined use of biocontrol agents and plant extracts can be considered a viable and promising strategy for protecting plant tissues with different synergistic mechanisms of action that improve the antimicrobial activity of the mixtures. Treatments of citrus fruits with Wickerhamomyces anomalus BS91 have been previously reported as effective measures to reduce the incidence of green mold caused by Penicillium digitatum. On the opposite, the knowledge of the antifungal activity of cultivated cardoon (Cynara cardunculus L. var. altilis DC.) leaf extract, vegetable widespread in some Mediterranean areas, is still very limited. In this study, experimental trials were conducted to evaluate the effectiveness in vitro of leaf aqueous, methanolic and ethanolic extracts of C. cardunculus against seven fungal pathogens responsible for considerable food losses in the postharvest stage. In addition, biocontrol yeast W. anomalus BS91 and the three C. cardunculus extracts were tested in vivo both as a single treatment and in mixture, against Penicillium digitatum on 'Tarocco' oranges and 'Femminello' lemons. The combination of W. anomalus BS91 and leaf ethanolic extract reduced with the highest efficacy the incidence and severity of green mold on orange and lemon fruits with respect to the control, and was more effective than treatment with antagonistic yeast or leaf extracts applied alone. Incidence and severity of citrus decay were more consistently reduced when mixtures were applied 24 h before the inoculation of the pathogen, thus suggesting the relevance of preventive treatments. The mixtures of antagonistic W. anomalus BS91 and ethanolic leaf extract were more effective in controlling green mold decay on oranges than on lemons. These results indicate that biocontrol agents and leaf extracts, used in appropriate combination, can provide a stronger protection than when used singularly. However, compatibility between microbial antagonist and antimicrobial extract should be preliminary verified.

Abstract:Cryptococcus laurentii (Kufferath) Skinner was evaluated for its activity in reducing postharvest blue mold decay of oranges caused by Penicillium italicum in vitro and in vivo . The results showed that washed cell suspensions of yeast provided control of blue mold decay better than yeast in culture broth. Autoclaved cell culture and cell-free culture filtrate failed to provide protection against the pathogen. The concentrations of antagonist had significant effects on biocontrol effectiveness. When the washed yeast cell suspension reached the concentration of 1 × 10 9 CFU/ml, challenged with pathogen spore suspension at 1 × 10 4 spores/ml, the blue mold decay was completely inhibited during 5 days of incubation at 20 °C. No complete control was obtained when oranges were stored at 4 °C for 30 days, but the decay was distinctly prevented. Efficacy of C. laurentii was maintained when applied simultaneously or prior to inoculation with P. italicum . Efficacy was reduced when C. laurentii was applied after inoculation. In drop-inoculated wounds of oranges, the populations of C. laurentii increased by approximately 50-fold during the first 24 h at 20 °C. The maximum yeast populations, approximately 250-fold over the initial populations, were reached 15 days after inoculation at 4 °C.
