Abstract:Label-free vibrational imaging by stimulated Raman scattering (SRS) provides unprecedented insight into real-time chemical distributions. Specifically, SRS in the fingerprint region (400–1800 cm −1 ) can resolve multiple chemicals in a complex bio-environment. However, due to the intrinsic weak Raman cross-sections and the lack of ultrafast spectral acquisition schemes with high spectral fidelity, SRS in the fingerprint region is not viable for studying living cells or large-scale tissue samples. Here, we report a fingerprint spectroscopic SRS platform that acquires a distortion-free SRS spectrum at 10 cm −1 spectral resolution within 20 µs using a polygon scanner. Meanwhile, we significantly improve the signal-to-noise ratio by employing a spatial-spectral residual learning network, reaching a level comparable to that with 100 times integration. Collectively, our system enables high-speed vibrational spectroscopic imaging of multiple biomolecules in samples ranging from a single live microbe to a tissue slice.
Resonant TERS of a Single-Molecule Kondo System

Abstract:Single-molecule tip-enhanced Raman spectroscopy (TERS) under ultra-high vacuum (UHV) and cryogenic conditions enables exploration of the relations between the adsorption geometry, electronic state, and vibrational fingerprints of individual molecules. TERS capability of reflecting spin states in open-shell molecular configurations is yet unexplored. Here we use the tip of a scanning probe microscope to lift a perylene-3,4,9,10-tetracarboxylic dianhydride (PTCDA) molecule from a metal surface to bring it into an open-shell spin one-half anionic state. We reveal a correlation between the appearance of a Kondo resonance in the differential conductance spectroscopy and concurrent characteristic changes captured by the TERS measurements. Through a detailed investigation of various adsorbed and tip-contacted PTCDA scenarios, we infer that the Raman scattering on the suspended PTCDA is resonant with a higher excited state. Theoretical simulation of the vibrational spectra enables a precise assignment of the individual TERS peaks to high-symmetry Ag modes, including the fingerprints of the observed spin state. These findings highlight the potential of TERS in capturing complex interactions between charge, spin, and photophysical properties in nanoscale molecular systems, and suggest a pathway for designing spin-optical devices using organic molecules.

Tip-Enhanced Raman Scattering on Both Sides of the Schrödinger Equation

Abstract:Historically, molecular spectroscopists have focused their attention to the right-hand side of the Schrödinger equation. Our major goal had and still has to do with determining a (bio)molecular system's Hamiltonian operator. From a theoretical spectroscopist's perspective, this entails varying the parameters of a model Hamiltonian until the predicted observables agree with their experimental analogues. In this context, less emphasis has been put on the left-hand side of the equation, where the interplay between a system and its immediate local environment is described. The latter is particularly meaningful and informative in modern applications of optical microscopy and spectroscopy that take advantage of surface plasmons to enhance molecular scattering cross-sections and to increase the attainable spatial resolution that is classically limited by diffraction. Indeed, the manipulation of light near the apex of a metallic nanotip has enabled single molecule detection, identification, and imaging. The distinct advantages of the so-called tip-enhanced optical nanospectroscopy/nanoimaging approaches are self-evident: ultrahigh spatial resolution (nanometer or better) and ultimate sensitivity (down to yoctomolar) are both attainable, all while retaining the ability to chemically fingerprint one molecule at a time (e.g., through Raman scattering). An equally interesting aspect of the same approach stems from using the properties of a single molecule to characterize the local environment in which it resides. This concept of single molecule spectroscopy on the left-hand side of the Schrödinger equation is certainly not novel and has been discussed in pioneering single molecule studies that ultimately led to a Nobel prize in chemistry. That said, local environment mapping through ultrasensitive optical spectroscopy acquires a unique flavor when executed using tip-enhanced Raman scattering (TERS). This is the subject of this Account.In a series of recent reports, our group utilized TERS to characterize different properties of nanolocalized and enhanced optical fields. The platforms that were used to this end consist of chemically functionalized plasmonic nanostructures and nanoparticles imaged using visible-light-irradiated gold- or silver-coated probes of an atomic force microscope. Through a detailed analysis of the recorded spectral nanoimages, we found that molecular Raman spectra may be used to track the magnitudes, resonances, spatiotemporal gradients, and even vector components of optical fields with nanometer spatial resolution under ambient conditions. On the other side of the equation, understanding how spatially varying optical fields modulate molecular nano-Raman spectra is of utmost importance to emerging areas of nanophotonics. For instance, tracking plasmon-enhanced chemical transformations via TERS necessitates a deeper fundamental understanding of the optical signatures of molecular reorientation and multipolar Raman scattering, both of which may be driven by local optical field gradients that are operative in TERS. We illustrate these concepts and introduce the readers to the generally less appreciated and equally exciting world of TERS on the left-hand side of the Schrödinger equation.

Abstract:Structure, dynamics, and coupling involving single-molecules determine function in catalytic, electronic or biological systems. While vibrational spectroscopy provides insight into molecular structure, rapid fluctuations blur the molecular trajectory even in single-molecule spectroscopy, analogous to spatial averaging in measuring large ensembles. To gain insight into intramolecular coupling, substrate coupling, and dynamic processes, we use tip-enhanced Raman spectroscopy (TERS) at variable and cryogenic temperatures, to slow and control the motion of a single molecule. We resolve intrinsic line widths of individual normal modes, allowing detailed and quantitative investigation of the vibrational modes. From temperature dependent line narrowing and splitting, we quantify ultrafast vibrational dephasing, intramolecular coupling, and conformational heterogeneity. Through statistical correlation analysis of fluctuations of individual modes, we observe rotational motion and spectral fluctuations of the molecule. This work demonstrates single-molecule vibrational spectroscopy beyond chemical identification, opening the possibility for a complete picture of molecular motion ranging from femtoseconds to minutes.
A full quantum mechanical approach assessing the chemical and electromagnetic effect in TERS

Abstract:Tip-enhanced Raman spectroscopy (TERS) is a valuable method for surface analysis with nanometer to angstrom-scale resolution, however, the accurate simulation of particular TERS signals remains a computational challenge. We present a unique approach to this challenge by combining the two main contributors to plasmon-enhanced Raman spectroscopy and to the high resolution in TERS in particular, the electromagnetic and the chemical effect, into one quantum mechanical simulation. The electromagnetic effect describes the sample's interaction with the strong, highly localized and inhomogeneous electric fields associated with the plasmonic tip, and is typically the thematic focus for most mechanistic studies. On the other hand, the chemical effect covers the different responses to the extremely close-range and highly position-sensitive chemical interaction between the apex tip atom(s) and the sample, and, as we could show in previous works, plays an often underestimated role. Starting from a (time-dependent) density functional theory description of the chemical model system, comprised of a tin(II) phthalocyanine (SnPc) sample molecule and a single silver atom as tip, we introduce the electromagnetic effect through a series of point charges that recreate the electric field in the vicinity of the plasmonic Ag nanoparticle. By scanning the tip over the molecule along a 3D grid, we can investigate the system's Raman response on each position for non-resonant and resonant illumination. Simulating both effects on their own already hints at the achievable signal enhancement and resolution, but the combination of both creates even stronger evidence that TERS is capable of resolving sub-molecular features.

Abstract:Tip-enhanced Raman scattering (TERS) has recently attracted remarkable attention as a novel nano-spectroscopy technique. TERS, which provides site-specific information, can be performed on any material surface regardless of morphology. Moreover, it can be applied in various environments, such as ambient air, ultrahigh vacuum (UHV), solutions, and electrochemical environments. This review reports on one hand progress of TERS for the last two decades, and on the other hand, its challenges in very recent years. Part of the progress of TERS starts with the prehistory and history of TERS, and then, the characteristics and advantages of TERS are described. Significant emphasis is put on the development of TERS instrumentation and equipment such as ultrahigh vacuum TERS, liquid TERS, electrochemical-TERS, and tip-preparations. Applications of TERS, particularly those with nanocarbons, biological materials, and surface and interface analysis, are mentioned in some detail. In the part on challenges, we focus on the very recent advances in TERS; progress in spatial resolution to the angstrom scale is the hottest topic. Recent TERS studies performed under UHV, for example chemical imaging at the angstrom scale and Raman detection of bond breaking and making of a chemisorbed up-standing single molecules at single-bond level, are reviewed. Of course, there is no clear border between the two parts. In the last part the perspective of TERS is discussed.

Abstract:Confocal and surface-enhanced Raman spectroscopy (SERS) are powerful techniques for molecular characterization; however, they suffer from the drawback of diffraction-limited spatial resolution. Tip-enhanced Raman spectroscopy (TERS) overcomes this limitation and provides chemical information at length scales in the tens of nanometers. In contrast to alternative approaches to nanoscale chemical analysis, TERS is label free, is non-destructive, and can be performed in both air and liquid environments, allowing its use in a diverse range of applications. Atomic force microscopy (AFM)-based TERS is especially versatile, as it can be applied to a broad range of samples on various substrates. Despite its advantages, widespread uptake of this technique for nanoscale chemical imaging has been inhibited by various experimental challenges, such as limited lifetime, and the low stability and yield of TERS probes. This protocol details procedures that will enable researchers to reliably perform TERS imaging using a transmission-mode AFM-TERS configuration on both biological and non-biological samples. The procedure consists of four stages: (i) preparation of plasmonically active TERS probes; (ii) alignment of the TERS system; (iii) experimental procedures for nanoscale imaging using TERS; and (iv) TERS data processing. We provide procedures and example data for a range of different sample types, including polymer thin films, self-assembled monolayers (SAMs) of organic molecules, photocatalyst surfaces, small molecules within biological cells, single-layer graphene and single-walled carbon nanotubes in both air and water. With this protocol, TERS probes can be prepared within ~23 h, and each subsequent TERS experimental procedure requires 3–5 h.

Abstract:Raman spectroscopy is a powerful technique in chemical information characterization. However, this spectral method is subject to two obstacles in nano-material detection. One is diffraction limited spatial resolution, and the other is its inherent small Raman cross section and weak signaling. To resolve these problems, a new approach has been developed, denoted as tip-enhanced Raman spectroscopy (TERS). TERS is capable of high-resolution and high-sensitivity detection and demonstrated to be a promising spectroscopic and micro-topographic method to characterize nano-materials and nanostructures. In this paper, the principle and experimental system of TERS are discussed. The latest application of TERS in molecule detection, biological specimen identification, nanao-material characterization, and semi-conductor material determination with some specific experimental examples are presented.
Polarized tip-enhanced Raman spectroscopy at liquid He temperature in ultrahigh vacuum using an off-axis parabolic mirror

Abstract:Tip-enhanced Raman spectroscopy (TERS) combines inelastic light scattering well below the diffraction limit down to the nanometer range and scanning probe microscopy and, possibly, spectroscopy. In this way, topographic and spectroscopic as well as single- and two-particle information may simultaneously be collected. While single molecules can now be studied successfully, bulk solids are still not meaningfully accessible. It is the purpose of the work presented here to outline approaches toward this objective. We describe a home-built, liquid helium cooled, ultrahigh vacuum tip-enhanced Raman spectroscopy system (LHe-UHV-TERS). The setup is based on a scanning tunneling microscope and, as an innovation, an off-axis parabolic mirror having a high numerical aperture of approximately $0.85$ and a large working distance. The system is equipped with a fast load-lock chamber, a chamber for the \textit{in situ} preparation of tips, substrates, and samples, and a TERS chamber. Base pressure and temperature in the TERS chamber were approximately $3\times 10^{-11}$~mbar and 15~K, respectively. Polarization dependent tip-enhanced Raman spectra of the vibration modes of carbon nanotubes were successfully acquired at cryogenic temperature. Enhancement factors in the range of $10^7$ were observed. The new features described here including very low pressure and temperature and the external access to the light polarizations, thus the selection rules, may pave the way towards the investigation of bulk and surface materials.

Subnanometer-resolved Chemical Imaging Via Multivariate Analysis of Tip-Enhanced Raman Maps

Abstract:Tip-enhanced Raman spectroscopy (TERS) is a powerful surface analysis technique that can provide subnanometer-resolved images of nanostructures with site-specific chemical fingerprints. However, due to the limitation of weak Raman signals and the resultant difficulty in achieving TERS imaging with good signal-to-noise ratios (SNRs), the conventional single-peak analysis is unsuitable for distinguishing complex molecular architectures at the subnanometer scale. Here we demonstrate that the combination of subnanometer-resolved TERS imaging and advanced multivariate analysis can provide an unbiased panoramic view of the chemical identity and spatial distribution of different molecules on surfaces, yielding high-quality chemical images despite limited SNRs in individual pixel-level spectra. This methodology allows us to exploit the full power of TERS imaging and unambiguously distinguish between adjacent molecules with a resolution of ~0.4 nm, as well as to resolve submolecular features and the differences in molecular adsorption configurations. Our results provide a promising methodology that promotes TERS imaging as a routine analytical technique for the analysis of complex nanostructures on surfaces. Advanced multivariate analysis can dramatically improve the chemical sensing capabilities of tip-enhanced Raman spectroscopy (TERS). A powerful surface analysis technique, TERS is hampered by inherently weak Raman signals, which have not permitted complex molecular architectures to be distinguished at the subnanometer scale. By employing advanced multivariate analysis, Song Jiang and co-workers from the University of Science and Technology of China used TERS to unambiguously identify adjacent molecules on a surface. The scheme’s spatial resolution of just 0.4 nanometers opens up the possibility of gathering information on a submolecular scale, such as mapping local vibrations and structural units within a single molecule. Experiments with a mixture of ZnTPP and H2TBPP molecules adsorbed on the edge of a silver step confirmed the advantages of the multivariate approach over conventional single-peak spectral analysis.

Abstract:Recent developments in tip-enhanced Raman spectroscopy (TERS) have demonstrated its capabilities in lateral resolution down to Ångström scale and chemical identification of adjacent molecules at the submolecular level. However, most studies are showcased only on the planar molecules lying flat on surfaces, and the resolving ability of TERS in the vertical direction still remains elusive, especially regarding its ability to resolve vertical heteromolecular structures. In this work, we artificially construct a bilayer-molecular system with one molecule at the tip apex and the other on the substrate to quantitatively investigate the vertical resolving ability of the TERS technique. The vertical detection depth is first measured using a pristine Ag tip by tracking the spectral evolution upon tip retraction away from a C60 molecule on the substrate until the TERS signals become practically vanished, which yields a detection depth around 500 pm. Consistently, when the tip is decorated by a C60 molecule at the tip apex, no TERS signals from the CuPc molecule on the substrate can be observed due to the large size of the decorated C60 molecules of ∼700 pm in diameter. However, when the Ag-tip apex is decorated with a single flat-lying CuPc molecule with a thickness of ∼280 pm, the TERS signals of the C60 molecule on the substrate can be detected clearly, demonstrating the capability of TERS to resolve vertical heteromolecular structures. Furthermore, the presence of the molecule at the tip apex is found to influence the local plasmonic field and extend the vertical detection depth up to ∼580 pm due to the hot-spot transfer effect. Our results not only provide a quantitative study on the vertical resolving ability of the TERS technique but also are instructive for potential applications in resolving subsurface chemical constituents and stereochemical structures.

Single-molecule time-resolved spectroscopy in a tunable STM nanocavity

Abstract:The spontaneous fluorescence rates of single-molecule emitters are typically on the order of nanoseconds. However coupling them with plasmonic nanostructures can substantially increase their fluorescence yields. The confinement between the tip and sample of a scanning tunneling microscope creates a tunable nanocavity, an ideal platform for exploring the yields and excitation decay rates of single-molecule emitters depending on the coupling strength to the nanocavity. With this setup we estimate the excitation lifetimes from the direct time-resolved measurements of the fluorescence decays of phthalocyanine adsorbates, decoupled from the metal substrates by ultrathin NaCl layers. It is found that nanosecond-range lifetimes prevail for the emitters away from the nanocavity, whereas for the tip approached to a molecule, we find a substantial effect of the nanocavity coupling, which reduces the lifetimes to a few picoseconds. An analysis is performed to investigate the crossover between the far-field and tip-enhanced photoluminescence regimes. This approach overcomes the drawbacks associated with the estimation of lifetimes for single molecules from their respective emission linewidths.

Abstract:Ultrafast studies1,2 of excitations on the nanometre scale are essential for guiding applications in nanotechnology. Efforts to integrate femtosecond lasers with scanning tunnelling microscopes (STMs)3 have yielded a number of ultrafast STM techniques4,5,6,7,8,9,10,11,12,13,14, but the basic ability to directly modulate the STM junction bias while maintaining nanometre spatial resolution has been limited to ∼10 ps (refs 7,8) and has required specialized probe or sample structures. Here, without any modification to the STM design, we modulate the STM junction bias by coupling terahertz pulses to the scanning probe tip of an STM and demonstrate terahertz-pulse-induced tunnelling in an STM. The terahertz STM (THz-STM) provides simultaneous subpicosecond (<500 fs) time resolution and nanometre (2 nm) imaging resolution under ambient laboratory conditions, and can directly image ultrafast carrier capture into a single InAs nanodot. The THz-STM accesses an ultrafast tunnelling regime that opens the door to subpicosecond scanning probe microscopy of materials with atomic resolution.

Abstract:Materials and their interfaces are the core for the development of a large variety of fields, including catalysis, energy storage and conversion. In this case, tip-enhanced Raman spectroscopy (TERS), which combines scanning probe microscopy with plasmon-enhanced Raman spectroscopy, is a powerful technique that can simultaneously obtain the morphological information and chemical fingerprint of target samples at nanometer spatial resolution. It is an ideal tool for the nanoscale chemical characterization of materials and interfaces, correlating their structures with chemical performances. In this review, we begin with a brief introduction to the nanoscale characterization of materials and interfaces, followed by a detailed discussion on the recent theoretical understanding and technical improvements of TERS, including the origin of enhancement, TERS instruments, TERS tips and the application of algorithms in TERS. Subsequently, we list the key experimental issues that need to be addressed to conduct successful TERS measurements. Next, we focus on the recent progress of TERS in the study of various materials, especially the novel low-dimensional materials, and the progresses of TERS in studying different interfaces, including both solid-gas and solid-liquid interfaces. Finally, we provide an outlook on the future developments of TERS in the study of materials and interfaces.
Tip-Enhanced Raman Spectroscopy Based on Plasmonic Lens Excitation and Experimental Detection

Abstract:A tip-enhanced Raman spectroscopy (TERS) based on plasmonic lens (PL) excitation is proposed in this work. A PL expected to realize a strong longitudinal electric field focus is designed. The focusing performance of the PL is calculated via finite-difference time-domain (FDTD) simulation and experimentally detected by a scattering-type scanning near-field optical microscope. The PL is introduced to a TERS system as a focusing device. Experimental results with carbon nanotube samples indicate that the Raman scatting signal is significantly enhanced. It proves experimentally that the combination of a PL focused excitation field with a metallic tip in a TERS system is a promising method.

Abstract:The development of time-resolved scanning tunnelling microscopy (STM), in particular, attempts to combine STM with ultrafast laser technology, is reviewed with emphasis on observed physical quantities and spatiotemporal resolution. Ultrashort optical pulse technology has allowed us to observe transient phenomena in the femtosecond range, which, however, has the drawback of a relatively low spatial resolution due to the electromagnetic wavelength used. In contrast, STM and its related techniques, although the time resolution is limited by the circuit bandwidth (∼100 kHz), enable us to observe structures at the atomic level in real space. Our purpose has been to combine these two techniques to achieve a new technology that satisfies the requirements for exploring the ultrafast transient dynamics of the local quantum functions in organized small structures, which will advance the pursuit of future nanoscale scientific research in terms of the ultimate temporal and spatial resolutions.

Abstract:After over 15 years of development, tip-enhanced Raman spectroscopy (TERS) is now facing a very important stage in its history. TERS offers high detection sensitivity down to single molecules and a high spatial resolution down to sub-nanometers, which make it an unprecedented nanoscale analytical technique offering molecular fingerprint information. The tip is the core element in TERS, as it is the only source through which to support the enhancement effect and provide the high spatial resolution. However, TERS suffers and will continue to suffer from the limited availability of TERS tips with a high enhancement, good stability, and high reproducibility. This review focuses on the tip-related issues in TERS. We first discuss the parameters that influence the enhancement and spatial resolution of TERS and the possibility to optimize the performance of a TERS system via an in-depth understanding of the enhancement mechanism. We then analyze the methods that have been developed for producing TERS tips, including vacuum-based deposition, electrochemical etching, electrodeposition, electroless deposition, and microfabrication, with discussion on the advantages and weaknesses of some important methods. We also tackle the issue of lifetime and protection protocols of TERS tips which are very important for the stability of a tip. Last, some fundamental problems and challenges are proposed, which should be addressed before this promising nanoscale characterization tool can exert its full potential.Graphical Abstractᅟ
Are charged tips driving TERS-resolution? A full quantum chemical approach

Abstract:Experimental evidence suggests an extremely high, possibly even sub-molecular, spatial resolution of tip-enhanced Raman spectroscopy. While the underlying mechanism is currently still under discussion, two main contributions are considered: The involved plasmonic particles are able to highly confine light to small spatial regions in the near-field, i.e. the electromagnetic effect, and the chemical effect due to altered molecular properties of the sample in close proximity to the plasmonic tip. Significant theoretical effort is put into the modelling of the electromagnetic contribution by various groups. In contrast, we previously introduced a computational protocol that allows for investigation of the local chemical effect -- including non-resonant, resonant and charge transfer contributions -- in a plasmonic hybrid system by mapping the sample molecule with a metallic tip model at the (time-dependent) density functional level of theory. In the present contribution, we evaluate the impact of static charges localized on the tip's frontmost atom, possibly induced by the tip geometry in the vicinity of the apex, on the TERS signal and the lateral resolution. To this aim, an immobilized molecule, i.e. tin(II) phthalocyanine (SnPc), is mapped by the plasmonic tip modeled by a single positively vs. negatively charged silver atom. The performed quantum chemical simulations reveal a pronounced enhancement of the Raman intensity under non-resonant and resonant conditions with respect to the uncharged reference system, while the contribution of charge transfer phenomena as well as of locally excited states of SnPc is highly dependent on the tip's charge.
