Abstract:Tangential flow filtration (TFF) through a 30 kDa nominal molecular weight cut-off (MWCO) ultrafiltration membrane is widely employed to concentrate purified monoclonal antibodies (mAbs) to levels required for their formulation into injectable biologics. While TFF has been used to remove casein from milk for cheese production for over 35 years, and in pharmaceutical manufacture of biotherapeutic proteins for 20 years, the rapid decline in filtration rate (i.e., flux) at high protein concentrations is a limitation that still needs to be addressed. This is particularly important for mAbs, many of which are 140-160 kDa immunoglobulin G (IgG) type proteins recovered at concentrations of 200 mg/mL or higher. This work reports the direct measurement of local transmembrane pressure drops and off-line confocal imaging of protein accumulation in stagnant regions on the surface of a 30 kDa regenerated cellulose membrane in a flat-sheet configuration widely used in manufacture of biotherapeutic proteins. These first-of-a-kind measurements using 150 kDa bovine IgG show that while axial pressure decreases by 58 psi across a process membrane cassette, the decrease in transmembrane pressure drop is constant at about 1.2 psi/cm along the 20.7 cm length of the membrane. Confocal laser scanning microscopy of the membrane surface at the completion of runs where retentate protein concentration exceeds 200 mg/mL, shows a 50 μm thick protein layer is uniformly deposited. The localized measurements made possible by the modified membrane system confirm the role of protein deposition on limiting ultrafiltration rate and indicate possible targets for improving membrane performance.
Tunable membrane-less dielectrophoretic microfiltration by crossing interdigitated electrodes

Abstract:Filtration is a crucial step in the analysis of living microparticles. In particular, the selective microfiltration of phytoplankton by size and shape remains an open problem, even though these criteria are essential for their gender and/or species identification. However, microfiltration devices necessitate physical membranes which complicate their fabrication, reduce the sample flow rate and can cause unwanted particle clogging. Recent advances in microfabrication such as electrode High Precision Capillary Printing allow to rapidly build electrode patterns over wide areas. In this study, we introduce a new concept of membrane-less dielectrophoretic (DEP) microfiltration suitable for large scale microfabrication processes. The proposed design involves two pairs of interdigitated electrodes at the top and the bottom of a microfluidic channel. We use finite-element calculations to analyse how the DEP force field throughout the channel, as well as the resulting trajectories of particles depend on the geometry of the system, on the physical properties of the particles and suspending medium and on the imposed voltage and flow rates. We numerically show that in the negative DEP regime, particles are focused in the channel mid-planes and that virtual pillars array leads either to their trapping at specific stagnation points, or to their focusing along specific lines, depending on their dielectrophoretic mobility. Simulations allow to understand how particles can be captured and to quantify the particle filtration conditions by introducing a critical dielectrophoretic mobility. We further illustrate the principle of membrane-less dielectrophoretic microfiltration using the proposed setup, by considering the separation of a binary mixture of polystyrene particles with different diameters, and validate it experimentally.

Automated Membrane Characterization: In-situ Monitoring of the Permeate and Retentate Solutions using a 3D Printed Permeate Probe Device

Abstract:Self-driving laboratories and automated experiments can accelerate the design workflow and decrease errors associated with experiments that characterize membrane transport properties. Within this study, we use 3D printing to design a custom stirred cell that incorporates inline conductivity probes in the retentate and permeate streams. The probes provide a complete trajectory of the salt concentrations as they evolve over the course of an experiment. Here, automated diafiltration experiments are used to characterize the performance of commercial NF90 and NF270 polyamide membranes over a predetermined range of KCl concentrations from 1-100 mM. The measurements obtained by the inline conductivity probes are validated using offline post-experiment analyses. Compared to traditional filtration experiments, the probes decrease the amount of time required for an experimentalist to characterize membrane materials by more than 50× and increase the amount of information generated by 100×. Device design principles to address the physical constraints associated with making conductivity measurements in confined volumes are proposed. Overall, the device developed within this study provides a foundation to establish high-throughput, automated membrane characterization techniques.

A mini-module with built-in spacers for high-throughput ultrafiltration

Abstract:Ultrafiltration membrane modules suffer from a permeate flow decrease arising during filtration and caused by concentration polarization and fouling in, e.g., fermentation broth purification. Such performance losses are frequently mitigated by manipulating the hydrodynamic conditions at the membrane-fluid interface using, e.g., mesh spacers acting as static mixers. This additional element increases manufacturing complexity while improving mass transport in general, yet accepting their known disadvantages such as less transport in dead zones. However, the shape of such spacers is limited to the design of commercially available spacer geometries. Here, we present a methodology to design an industrially relevant mini-module with an optimized built-in 3D spacer structure in a flat-sheet ultrafiltration membrane module to eliminate the spacer as a separate part. Therefore, the built-in structures have been conceptually implemented through an in-silico design in compliance with the specifications for an injection molding process. Ten built-in structures were investigated in a digital twin of the mini-module by 3D-CFD simulations to select two options, which were then compared to the empty feed channel regarding mass transfer. Subsequently, the simulated flux increase was experimentally verified during bovine serum albumin (BSA) filtration. The new built-in sinusoidal corrugation outperforms conventional mesh spacer inlays by up to 30% higher permeation rates. The origin of these improvements is correlated to the flow characteristics inside the mini-module as visualized online and in-situ by low-field and high-field magnetic resonance imaging velocimetry (flow-MRI) during pure water permeation.

Abstract:Membrane filtration processes are best known for their application in the water, oil, and gas sectors, but also in food production they play an eminent role. Filtration processes are known to suffer from a decrease in efficiency in time due to e.g., particle deposition, also known as fouling and pore blocking. Although these processes are not very well understood at a small scale, smart engineering approaches have been used to keep membrane processes running. Microfluidic devices have been increasingly applied to study membrane filtration processes and accommodate observation and understanding of the filtration process at different scales, from nanometer to millimeter and more. In combination with microscopes and high-speed imaging, microfluidic devices allow real time observation of filtration processes. In this review we will give a general introduction on microfluidic devices used to study membrane filtration behavior, followed by a discussion of how microfluidic devices can be used to understand current challenges. We will then discuss how increased knowledge on fundamental aspects of membrane filtration can help optimize existing processes, before wrapping up with an outlook on future prospects on the use of microfluidics within the field of membrane separation.

Accelerated Development of AAV Purification Process Using a High-Throughput and Automated Crossflow System

Abstract:Adeno-associated viruses (AAV) are currently predominant viral transfer tools for gene therapy, and efforts are being made to design faster and more efficient methods and technologies for their manufacturing. The early selection of high-performing filters is essential for developing an ultrafiltration and diafiltration (UF/DF) process, especially when feed material is scarce, and timelines are short. However, few methods and technologies exist to enable process optimization with multiple variations in a single run. In this study, we explored the potential of Ambr® Crossflow for high-throughput, automated screening of different membrane materials, pore sizes and different process conditions for the UF/DF step of AAV8. The best overall performance was obtained with a 100 kDa PES flat sheet cassette. The UF/DF process was further transferred to a larger scale to the Sartoflow® Smart Tangential Flow Filtration (TFF) system using a 100 kDa PES Sartocon® Slice 200 cassette and compared to a 100 kDa PES hollow fiber. Virus recovery, permeate flux and total protein removal values of the flat sheet cassette were similar to those achieved in small-scale devices, and higher than those of the hollow fiber, thus demonstrating similar performance at a larger process scale. The high-throughput, automated method described herein allowed to screen multiple materials and process parameters of a UF/DF process in a time- and resource-efficient way, making it a useful tool to accelerate early-stage downstream process development of AAV.

Microfluidic Buffer Exchange for Interference-free Micro/Nanoparticle Cell Engineering

Abstract:Engineering cells with active-ingredient-loaded micro/nanoparticles (NPs) is becoming an increasingly popular method to enhance native therapeutic properties, enable bio imaging and control cell phenotype. A critical yet inadequately addressed issue is the significant number of particles that remain unbound after cell labeling which cannot be readily removed by conventional centrifugation. This leads to an increase in bio imaging background noise and can impart transformative effects onto neighboring non-target cells. In this protocol, we present an inertial microfluidics-based buffer exchange strategy termed as Dean Flow Fractionation (DFF) to efficiently separate labeled cells from free NPs in a high throughput manner. The developed spiral microdevice facilitates continuous collection (>90% cell recovery) of purified cells (THP-1 and MSCs) suspended in new buffer solution, while achieving >95% depletion of unbound fluorescent dye or dye-loaded NPs (silica or PLGA). This single-step, size-based cell purification strategy enables high cell processing throughput (10(6) cells/min) and is highly useful for large-volume cell purification of micro/nanoparticle engineered cells to achieve interference-free clinical application.
Flow and filtration imaging of single use sterile membrane filters

Abstract:Sterile filters incorporating endotoxin adsorption function emerged recently to pretreat dialysate liquids fed to a hemodialysis filtration process. Their application significantly enhances the survival rate during dialysis treatment as they function as steril filters as well as an endotoxin adsorber. Little is known about the fluid flow distribution in such single use membrane modules. We report a detailed analysis of the local 3D flow field distribution in such membrane modules using magnetic resonance flow imaging. Next to pure water filtration representing the application case of endotoxin adsorption from an already pure liquid, we also used the module as a filtration device rejecting for instance colloidal silica. Such experiments performed in-situ allow the quantification of cake layer development and its concomitant redistribution of the flow field. Particularly novel is the quantification of the time evolution of local permeate flux distribution. These detailed insights of this study encourage the use of flow-MRI when designing and applying new membrane module configurations.
Filtration of Particulates and Emulsions with a Pleated, Thin Channel, Cross-Flow Module

Abstract:The pleated thin channel crossflow module represents a novel flat sheet membrane configuration combining the modular advantages of hollow fiber design with the hydrodynamics of plate and frame units containing turbulence promoting elements. It consists of a circular array of parallel pleated thin channels, each of whose inner wall is a membrane supported by a porous backing resting on a central product drain tube. The outer flexible wall is separated from the membrane by a turbulence promoting spacer and serves to create the flow channel, without fulfilling a direct filtrative purpose. The design allows operation at elevated particulate levels and is highly suited to microfiltration of stray matter and bacteria from aqueous solutions to be injected into well heads for enhanced oil recovery. Data are presented for 0.45 and 0.2 μm pore size membrane modules containing 3.3 ft2 of membrane area and operating on dispersions of standard particulates of known size distribution (80 μm to 0.2 μm). Effects of filtration mode (i.e., constant pressure or constant permeate rate), and crossflow velocity were examined at particulate concentrations from 250 to 800 ppm. For seperation involving paraffinic oil/water emulsions, modules are either fabricated with ultrafiltration (UF) membrane, or a dynamically formed membrane is created on the microporous support. The latter approach was used to purify waste textile scouring washwater at 60°, thereby recovering both water and energy. Waste scouring water, containing ±1200 ppm of emulsified total organic carbon (TOC), 500 ppm of freon extractable hydrocarbon, and a turbidity of 66 nephalometric turbidity units (NTU) was ultrafiltered at 50 to 60°C with a dynamically formed silic a membrane at an average pressure of 17 psi and 1.6 gpm crossflow in a unit containing 1 ft 2 of 0.2 pm Acropor support. The product flow rate was 35 to 50 gpd and contained 400 ppm TOC with 125 ppm freon extractable hydrocarbon and a turbidity of 0.5 NTU. Yarn samples scoured with recovered scouring water showed no difference in their dyeability.

Abstract:Protein crystallization has the potential to provide nearly pure protein products in a single processing step. Most previous studies of protein crystallization have used centrifugation for dewatering and washing the crystallized solids, but this approach would be difficult to adopt in continuous bioprocessing operations. This study provides the first demonstration that crystallized proteins can be successfully filtered using hollow fiber tangential flow filtration (TFF) modules. Experiments were performed with human serum albumin (HSA) crystallized by the addition of trivalent CeCl 3 , both with and without polyethylene glycol (PEG). Greater than 85 % protein yield was achieved using 4.5 mM CeCl 3 in the presence of 3 wt% PEG with the crystallization occurring in less than 20 s, which is ideal for continuous processing. The addition of PEG had minimal effect on the crystallization yield, but it did lead to the formation of larger and denser crystals and a corresponding increase in the critical flux, which is the maximum value of the flux that causes minimal increase in the transmembrane pressure (i.e., minimal fouling). Filtration experiments performed at optimal conditions were successfully run for 24 h with the transmembrane pressure remaining below 5 kPa, demonstrating the feasibility of using continuous crystallization with tangential flow filtration for the purification of high value protein products.

Abstract:In this paper, we investigate the physical mechanisms underlying one of the most efficient filtration devices: the kidney. Building on a minimal model of the Henle loop - the central part of the kidney filtration - we investigate theoretically the detailed out-of-equilibrium fluxes in this separation process in order to obtain absolute theoretical bounds for its efficiency in terms of separation ability and energy consumption. We demonstrate that this separation process operates at a remarkably small energy cost as compared to traditional sieving processes while working at much smaller pressures. This unique energetic efficiency originates in the double-loop geometry of the nephron, which operates as an active osmotic exchanger. The principles for an artificial-kidney-inspired filtration device could be readily mimicked based on existing soft technologies to build compact and low-energy artificial dialytic devices. Such a "kidney on a chip" also points to new avenues for advanced water recycling, targeting, in particular, sea-water pretreatment for decontamination and hardness reduction.

Abstract:Microfluidics has been the key component for many applications, including biomedical devices, chemical processors, microactuators, and even wearable devices. This technology relies on soft lithography fabrication which requires cleanroom facilities. Although popular, this method is expensive and labor-intensive. Furthermore, current conventional microfluidic chips precludes reconfiguration, making reiterations in design very time-consuming and costly. To address these intrinsic drawbacks of microfabrication, we present an alternative solution for the rapid prototyping of microfluidic elements such as microtubes, valves, and pumps. In addition, we demonstrate how microtubes with channels of various lengths and cross-sections can be attached modularly into 2D and 3D microfluidic systems for functional applications. We introduce a facile method of fabricating elastomeric microtubes as the basic building blocks for microfluidic devices. These microtubes are transparent, biocompatible, highly deformable, and customizable to various sizes and cross-sectional geometries. By configuring the microtubes into deterministic geometry, we enable rapid, low-cost formation of microfluidic assemblies without compromising their precision and functionality. We demonstrate configurable 2D and 3D microfluidic systems for applications in different domains. These include microparticle sorting, microdroplet generation, biocatalytic micromotor, triboelectric sensor, and even wearable sensing. Our approach, termed soft tubular microfluidics, provides a simple, cheaper, and faster solution for users lacking proficiency and access to cleanroom facilities to design and rapidly construct microfluidic devices for their various applications and needs.

Zweifach-Fung Microfluidic Device for Efficient Microparticle Separation: Cost-Effective Fabrication Using CO2 Laser-Ablated PMMA

Abstract:Microfluidic separators play a pivotal role in the biomedical and chemical industries by enabling precise fluid manipulations. Traditional fabrication of these devices typically requires costly cleanroom facilities, which limits their broader application. This study introduces a novel microfluidic device that leverages the passive Zweifach-Fung principle to overcome these financial barriers. Through Lagrangian computational simulations, we optimized an eleven-channel Zweifach-Fung configuration that achieved a perfect 100% recall rate for particles following a specified normal distribution. Experimental evaluations determined 2 mL/h as the optimal total flow rate (TFR), under which the device showcased exceptional performance enhancements in precision and recall for micrometer-sized particles, achieving an overall accuracy of 94% ± 3%. Fabricated using a cost-effective, non-cleanroom method, this approach represents a significant shift from conventional practices, dramatically reducing production costs while maintaining high operational efficacy. The cost of each chip is less than USD 0.90 cents and the manufacturing process takes only 15 min. The development of this device not only makes microfluidic technology more accessible but also sets a new standard for future advancements in the field.

Abstract:Nanofiltration has been attracting great attention in alleviating the global water crisis because of its high efficiency, mild operation, and strong adaptability. Over decades, it remains a challenge to break the upper limit of performance and establish the formation-structure-property relationship for nanofiltration membranes. This feature article summarizes our recent progress in the preparation of high-performance thin-film composite (TFC) nanofiltration membranes, focusing on the mussel-inspired deposition method and the optimized interfacial polymerization (IP). By accelerating the oxidation of polydopamine and equilibrating the rate of aggregation and deposition processes, the mussel-inspired deposition method realizes the rapid and uniform formation of selective coatings or nanofilms. Diverse deposition systems endow the selective layer with rich chemical structures and easy post-functionalization, highlighting its potential in water treatment. As for optimizing the conventional IP, the rapid polycondensation of amine and acid chloride groups is slowed down to enable the controllability of IP at the water-organic interface. The homogeneity and integrity of the TFC membranes are improved by constructing a uniform reaction platform and introducing a viscous medium to control the amine diffusion, which facilitates the water permeability and promotes the separation efficiency. We have proposed a series of practical strategies for improving TFC membranes and might provide more inspiration for other nanofiltration techniques.
Establishment of a Perfusion Process with Antibody-Producing CHO Cells Using a 3D-Printed Microfluidic Spiral Separator with Web-Based Flow Control

Abstract:Monoclonal antibodies are increasingly dominating the market for human therapeutic and diagnostic agents. For this reason, continuous methods-such as perfusion processes-are being explored and optimized in an ongoing effort to increase product yields. Unfortunately, many established cell retention devices-such as tangential flow filtration-rely on membranes that are prone to clogging, fouling, and undesirable product retention at high cell densities. To circumvent these problems, in this work, we have developed a 3D-printed microfluidic spiral separator for cell retention, which can readily be adapted and replaced according to process conditions (i.e., a plug-and-play system) due to the fast and flexible 3D printing technique. In addition, this system was also expanded to include automatic flushing, web-based control, and notification via a cellphone application. This set-up constitutes a proof of concept that was successful at inducing a stable process operation at a viable cell concentration of 10-17 × 106 cells/mL in a hybrid mode (with alternating cell retention and cell bleed phases) while significantly reducing both shear stress and channel blockage. In addition to increasing efficiency to nearly 100%, this microfluidic device also improved production conditions by successfully separating dead cells and cell debris and increasing cell viability within the bioreactor.
High throughput clogging free microfluidic particle filter by femtosecond laser micromachining

Abstract:In recent decades, driven by the needs of industry and medicine, researchers have been investigating how to remove carefully from the main flow microscopic particles or clusters of them. Among all the approaches proposed, crossflow filtration is one of the most attractive as it provides a non‐destructive, label‐free and in‐flow sorting method. In general, the separation performance shows capture and separation efficiencies ranging from 70% up to 100%. However, the maximum flow rate achievable (μL/min) is still orders of magnitude away from those suitable for clinical or industrial applications mainly due to the low stiffness of the materials typically used. In this work, we propose an innovative hydrodynamic‐crossflow hybrid filter geometry, buried in a fused silica substrate by means of the femtosecond laser irradiation followed by chemical etching technique. The material high stiffness combined with the accuracy of our manufacturing technique allows the 3D fabrication of non‐deformable channels with higher aspect ratio posts, while keeping the overall device dimensions compact. The filter performance has been validated through experiments with both Newtonian (water‐based solution of microbeads) and non‐Newtonian fluids (blood), achieving separation efficiencies of up to 94% and large particles recovery rates of 100%, even at very high flow rates (mL/h).

Abstract:A new contorted monomer featuring a 2,2'‐biphenyldiamine (BIPDA) motif used as the building block to generate ultra‐thin microporous nanofilms via IP, enabling rapid and precise molecular nanofiltration. The oil‐phase cross‐linker diacyl chloride TPC results in selective cross‐linking and a more ordered film structure compare to triacyl chloride TMC. In addition, it can enable effective recycling of organometallic homogeneous catalysts. A key challenge in efficient molecular separation is fabricating large‐scale, highly selective polymeric membranes with precise pore control at the molecular scale. Herein, a new contorted monomer 6,6′‐dihydroxy‐2,2′‐biphenyldiamine (DHBIPDA) is introduced as a building block to generate cross‐linked, ultra‐thin microporous nanofilms (sub‐10 nm) via interfacial polymerization, enabling rapid, and precise molecular nanofiltration. Using diacyl chloride (TPC) as the cross‐linker instead of trimesoyl chloride (TMC) significantly reduces the pore sizes within the membranes and achieves a narrower pore distribution due to a semi‐crystalline structure. The film structures are confirmed using comprehensive characterization techniques including wide‐angle X‐ray scattering (WAXS), X‐ray diffraction (XRD), positron annihilation lifetime spectroscopy (PALS), CO2 adsorption analysis, and molecular‐scale simulation. The DHBIPDA/TPC and DHBIPDA/TMC membranes achieve methanol permeance values of up to 16.4 and 15.1 LMH bar−1 coupled with molecular weight cutoffs (MWCOs) as low as 283 and 306 Da, respectively. The DHBIPDA/TPC membrane demonstrates both higher permeance and higher selectivity compared to its relatively disordered counterpart DHBIPDA/TMC, consistent with characterization data. The DHBIPDA‐derived membrane efficiently separates dye mixtures with similar molecular weights and enables effective recycling of organometallic homogeneous catalysts, suggesting its potential for industrial applications.

Abstract:Few studies have reported on the detail of the mass production process of nanofiltration (NF) membranes and modules due to technical confidentiality. This work presents a procedure of fabrication of NF membranes and modules on the pilot scale. Thin film composite (TFC) NF flat-sheet membranes of 300 mm width were continuously fabricated via custom-designed equipment. The fabrication parameters and their effects on NF performance were systematically investigated. The optimized conditions parameters were achieved for excellent NF modules' performance. The modules show over 75 L m(-2) h(-1) flux at 6 bar and over 95% MgSO4 rejection. This study may provide insightful guidelines for the scale-up of NF membranes for water treatment and other applications. (C) 2020 Institution of Chemical Engineers. Published by Elsevier B.V. A Ceramic Microfiltration Tube Membrane Dispersion Extractor

Abstract:Although the phase-free membrane extraction process has shown many advantages over the traditional extraction, the process is still at the experimental stage. On the other hand, in a new extractor a microfiltration membrane is used as a dispersion medium. The mass-transfer performance of the new extractor was tested with 30% TBP (in kerosene)nitric acid-H2O as an experimental system. The overall mass-transfer coefficient and the equipment efficiency were calculated with the concentrations of the inlet and outlet. The extractor was designed and constructed with two special inner elements for improving the performance further. The effects of the transmembrane pressure, the continuous-phase flow rate, and the geometric parameters of the inner elements on the mass-transfer performance, as well as on the flux of the dispersion phase, are discussed. The experimental results showed that very higher efficiency was reached. The inner elements could improve the mass-transfer performance greatly by changing the two-phase contact status. The results suggested that the mass-transfer process could be completed quickly while the drop size was in the range of micrometers. In addition, the flux of the dispersed phase was mainly influenced by the transmembrane pressure, not by the equipment structures, inner elements, and the continuous-phase flow rate. The new extractor can be operated with very higher efficiency and higher flux, and the efficiency can be predicted with a cubic polynomial. (C) 2004 American Institute of Chemical Engineers.
