Abstract:Microstructure of hydroxyapatite is known to influence cellular behavior, can be performed as a substrate for osteoblast growth, and exploited as a drug release platform. However, easy delamination and self-decomposition of hydroxyapatite caused by poor adhesion with substrates are the main problems currently. In this paper, we successfully fabricated TiO2/SrHA composite scaffolds by self-generated strontium substituted hydroxyapatite microspheres in TiO2 nanotubes. Moreover, the active compound 1-(5-bromo-2-hydroxy-methoxyphenyl)-ethanone (BHM) from Seahorse (Hippocampus kuda Bleeler) was loaded in this scaffold, and the controlled release kinetics of BHM was studied. It was found that in the first 5 hours, the release concentration and time of BHM had a good linear relationship, and the correlation coefficient reached 0.98. TiO2/SrHA/BHM composites exhibited favorable cytocompatibility at a given concentration of BHM (20 μmol/L). Compared to pure SrHA, TiO2 nanotubes and traditional TiO2/SrHA composites, the superior cytocompatibility (cell adhesion and proliferation) of MC3T3-E1 was obtained on TiO2/SrHA/BHM composites. The expression levels of osteogenic marker genes such as ALP, OPN and RunX2 are also up-regulated to varying degrees. This TiO2/SrHA composite scaffold mediated phenolic compounds BHM could be applied in bone tissue repair.
Preparation of Cu 2+ /TA/HAP composite coating with anti-bacterial and osteogenic potential on 3D-printed porous Ti alloy scaffolds for orthopedic applications

Abstract:Because of stress shielding effects, traditional titanium (Ti) alloy scaffolds have a high elastic modulus, which might promote looseness and bone disintegration surrounding the implant, increasing the likelihood of a second surgery. In contrast, 3D-printed porous Ti alloy scaffolds can reduce the scaffold weight while enhancing biocompatibility. Further, these scaffolds' porous nature allows bone tissue ingrowth as well as strong pore connectivity, which can improve nutrient absorption. Nevertheless, bare Ti alloy implants may fail because of inadequate bone integration; hence, adding a coating on the implant surface is an effective technique for improving implant stability. In this study, a composite coating comprising hydroxyapatite (HAP), chitosan (CS), tannic acid (TA) and copper ions (Cu 2+ ) (Cu 2+ /TA/HAP composite coating) was prepared on the surface of 3D printed porous Ti alloy scaffolds using electrophoretic deposition. Using the standard plate count method, Live/Dead bacteria staining assay, FITC Phalloidin and 4′,6-diamidino-2-phenylindole staining assay, and live/dead staining of cells we determined that the composite coating has better antibacterial properties and cytocompatibility as well as lower cytotoxicity. The Alkaline Phosphatase assay revealed that the coating results showed good osteogenesis potential. Overall, the composite coatings produced in this investigation give new potential for the application of Ti alloys in clinics.

Abstract:Journal of Biomaterials Applications, Ahead of Print. The 3D printing of porous titanium scaffolds reduces the elastic modulus of titanium alloys and promotes osteogenic integration. However, due to the biological inertness of titanium alloy materials, the implant-bone tissue interface is weakly bonded. A calcium silicate (CS) coating doped with polymetallic ions can impart various biological properties to titanium alloy materials. In this study, CuO and SrO binary-doped CS coatings were prepared on the surface of 3D-printed porous titanium alloy scaffolds using atmospheric plasma spraying and characterized by SEM, EDS, and XRD. Both CuO and SrO were successfully incorporated into the CS coating. The in vivo osseointegration evaluation of the composite coating-modified 3D-printed porous titanium alloy scaffolds was conducted using a rabbit bone defect model, showing that the in vivo osseointegration of 2% CuO-10% SrO-CS-modified 3D-printed porous titanium alloy was improved. The in vitro antimicrobial properties of the 2% CuO-10% SrO-CS-modified 3D-printed porous titanium alloy were evaluated through bacterial platform coating, co-culture liquid absorbance detection, and crystal violet staining experiments, demonstrating that the composite coating exhibited good antimicrobial properties. In conclusion, the composite scaffold possesses both osteointegration-promoting and antimicrobial properties, indicating a broad potential for clinical applications.

Abstract:Bone tissue engineering offers a promising alternative to stimulate the regeneration of damaged tissue, overcoming the limitations of conventional autografts and allografts. Recently, titanium alloy (Ti) implants have garnered significant attention for treating critical-sized bone defects, especially with the advancement of 3D printing technology. Although Ti alloys have impressive versatility, their lack of cellular adhesion, osteogenic and antibacterial properties are significant factors that contribute to their failure. Hence, to overcome these obstacles, this study aimed to incorporate osteoinductive and antibacterial cue-loaded hydrogels into 3D-printed Ti (3D-Ti) scaffolds. 3D-Ti scaffolds were synthesized using the direct metal laser sintering method and loaded with a gelatin (Gel) hydrogel containing strontium-doped silver nanoparticles (Sr-Ag NPs). Compared with Ag NPs, Sr-doped Ag NPs increased the expression of Runx2 mRNA, which is a key bone transcription factor. We subjected the bioactive 3D-hybrid scaffolds (3D-Ti/Gel/Sr-Ag NPs) to physicochemical and material characterization, followed by cytocompatibility and osteogenic evaluation. The microporous and macroporous topographies of the scaffolds with Sr-Ag NPs showed increased Runx2 expression and matrix mineralization, with potent antibacterial properties. Therefore, the 3D-Ti scaffolds incorporated with Sr-Ag NP-loaded Gel hydrogels favored osteoblast differentiation and antibacterial activity, indicating their potential for orthopedic applications.
Facile Fabrication of 3D-Printed Porous Ti6Al4V Scaffolds with a Sr-CaP Coating for Bone Regeneration

Abstract:To improve osseointegration caused by the stress-shielding effect and the inert nature of titanium-based alloys, in this work, we successfully constructed a strontium calcium phosphate (Sr-CaP) coating on three-dimensional (3D)-printed Ti6Al4V scaffolds to address this issue. The energy-dispersive X-ray spectroscopy (EDS) and X-ray diffraction (XRD) results indicated that the coatings with and without Sr doping mainly consisted of CaHPO4. The bonding strength of Sr doping coating met the required ISO 13 779-4-2018 standard (≥15 MPa). The in vitro results suggested that the Sr-CaP-modified Ti6Al4V scaffolds were found to effectively promote mice bone-marrow stem cell (mBMSC) adhesion, spreading, and osteogenesis. The in vivo experiments also showed that the Sr-CaP-modified Ti6Al4V scaffolds could significantly improve bone regeneration and osseointegration. More importantly, Sr-doped CaP-coated Ti6Al4V scaffolds were found to accelerate bone healing in comparison to CaP-coated Ti6Al4V scaffolds. The Sr-CaP-modified Ti6Al4V scaffolds are considered a promising strategy to develop bioactive surfaces for enhancing the osseointegration between the implant and bone tissue.

Porous surface with fusion peptides embedded in strontium titanate nanotubes elevates osteogenic and antibacterial activity of additively manufactured titanium alloy

Abstract:It is still a big challenge in orthopedics to treat infected bone defects properly using medical metals. The use of three-dimensional (3D) scaffold materials that simultaneously mimic the skeletal hierarchy and induce sustainable osteogenic and antibacterial functions are a promising solution with an increasing appeal. In this study, we first designed a bifunctional fusion peptide (HHC36-RGD, HR) by linking antimicrobial peptide (HHC36) and arginine-glycine-aspartate (RGD) peptide via 6-aminohexanoic acid. Then the 3D scaffold was fabricated by additive manufacturing, and the strontium titanate nanotube structure (3D-STN) was constructed on its surface. Finally, the HR was anchored to the 3D-STN with the aid of polydopamine (PDA, P), forming the 3D-STN-P-HR scaffold. The results showed that the scaffold exhibited an ordered 3D porous structure, and that the surface was covered by a dense HHC36-RGD layer. Expectedly, the adsorption of PDA effectively slowed down the release of HR. Moreover, the functionalized scaffold had a significant inhibitory effect on Staphylococcus aureus and Escherichia coli, and its antibacterial rate could reach more than 95%. The results of in vitro cell culture experiments demonstrated that the 3D-STN-P-HR scaffold possessed excellent cytocompatibility and could promote the transcription of osteogenic differentiation-related genes and the expression of related proteins. In conclusion, the functionally modified 3D porous titanium alloy scaffold (3D-STN-P-HR) has a balanced antibacterial and osteogenic function, which bodes well for future potential in the customized functional reconstruction of complex-shaped infected bone defects.

Abstract:Titanium was functionalized by bionic micro/nano-structure and Sr-HA coating.Functionalized titanium could promote cell adhesion and inhibit osteoclast activity.Functionalized titanium could improve the bone-implant bonding strength.As the population ages, more and more people are suffering from osteoarthritis (OA), resulting in an increasing requirement for joint implants. Surface modification to improve the topology and composition of the implants has been proved to be an effective way to improve the primary stability and long-term success rate of joint implants. In this work, a bionic micro/nano-structure accompanied with a strontium-substituted hydroxyapatite (SrHA) coating was fabricated on titanium (Ti) surface via electrochemical corrosion, ultrasonic treatment, and hydrothermal deposition methods. The in vitro study demonstrated that the bionic structure and the bioactive apatite could synergistically increase the expressions of integrin-related gene (ITG α5β1) and osteoblastic genes (Col-I and OCN), and thus promote osteoblast growth. In addition, owing to the anti-bone resorption property of Sr2+, the coating could effectively inhibit osteoclast differentiation and proliferation. In a word, the prepared samples not only promoted osteogenesis but also inhibited osteoclastogenesis. The in vivo experiment via a rabbit model found that the bionic structured surface provided the pore for new bone ingrowth, which was beneficial to the mechanical interlocking between the implant and bone. Moreover, the bionic structure and bioactive SrHA coating had a synergistic effect on promoting bone formation, osseointegration, and bone-implant bonding strength. Combined Antibacterial and Osteogenic in Situ Effects of a Bifunctional Titanium Alloy with Nanoscale Hydroxyapatite Coating

Abstract:To resolve the problems of bacterial infections and the low efficiency of osteogenesis of implanted titanium alloys in clinical dental and bone therapy, we developed a bifunctional titanium alloy (Ti) with a nano-hydroxyapatite (HA) coating (HBD+BMP/HA-Ti), which enables the sustained release of the natural antimicrobial peptide human beta-defensin 3 (HBD-3) and bone morphogenetic protein-2 (BMP-2). Due to the poriferous nano-sized structure of the HA coating with a 20-30 mu m thickness, the HBD+BMP/HA-Ti material had a high encapsulation efficiency (>74%) and exhibited synchronized slow release of HBD-3 and BMP-2. In an antibacterial test, HBD+BMP/HA-Ti prevented the growth of bacteria in an inoculated medium, and its surface remained free from viable bacteria after a continuous incubation with Gram-negative and Gram-positive strains for 7days. Furthermore, good adhesion, proliferation and osteogenic differentiation of hBMSCs in contact with HBD+BMP/HA-Ti were achieved in 7days. Therefore, the bifunctional titanium alloy HBD+BMP/HA-Ti has a great potential for eventual applications in the protection of implants against bacteria in the orthopaedic and dental clinic.
Bacterial Inhibition and Osteogenic Potentials of Sr/Zn Co-Doped Nano-Hydroxyapatite-PLGA Composite Scaffold for Bone Tissue Engineering Applications

Abstract:Bacterial infection associated with bone grafts is one of the major challenges that can lead to implant failure. Treatment of these infections is a costly endeavor; therefore, an ideal bone scaffold should merge both biocompatibility and antibacterial activity. Antibiotic-impregnated scaffolds may prevent bacterial colonization but exacerbate the global antibiotic resistance problem. Recent approaches combined scaffolds with metal ions that have antimicrobial properties. In our study, a unique strontium/zinc (Sr/Zn) co-doped nanohydroxyapatite (nHAp) and Poly (lactic-co-glycolic acid) -(PLGA) composite scaffold was fabricated using a chemical precipitation method with different ratios of Sr/Zn ions (1%, 2.5%, and 4%). The scaffolds' antibacterial activity against Staphylococcus aureus were evaluated by counting bacterial colony-forming unit (CFU) numbers after direct contact with the scaffolds. The results showed a dose-dependent reduction in CFU numbers as the Zn concentration increased, with 4% Zn showing the best antibacterial properties of all the Zn-containing scaffolds. PLGA incorporation in Sr/Zn-nHAp did not affect the Zn antibacterial activity and the 4% Sr/Zn-nHAp-PLGA scaffold showed a 99.7% bacterial growth inhibition. MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) cell viability assay showed that Sr/Zn co-doping supported osteoblast cell proliferation with no apparent cytotoxicity and the highest doping percentage in the 4% Sr/Zn-nHAp-PLGA was found to be ideal for cell growth. In conclusion, these findings demonstrate the potential for a 4% Sr/Zn-nHAp-PLGA scaffold with enhanced antibacterial activity and cytocompatibility as a suitable candidate for bone regeneration.

Impact of structural features of Sr/Fe co-doped HAp on the osteoblast proliferation and osteogenic differentiation for its application as a bone substitute

Abstract:The potential of external ions doped biomaterials has been extensively explored; however, the co-doped biomaterials remain typically unexplored for their biological properties for precise biomedical applications. The current study was aimed to explore the impact of structural features of Sr/Fe co-doped hydroxyapatite (HAp) bionanomaterial on osteoblastic proliferation and osteogenic differentiation for its application as a bone substitute. A 10 mol% isomorphous co-doping of strontium and iron with respect to calcium was carried into HAp in the solid solution. Raman spectroscopy verified the presence of major functional groups of apatite structure together with the carbonate peaks. The Sr/Fe co-doped HAp bionanomaterials showed slightly negative zeta potential (at neutral pH), versatile DLS particle size (140-205 nm), high BET surface area (186 m2/g), and narrow width pore size (13-19 nm). TG/DTA analysis showed low thermal stability of the Sr/Fe co-doped HAp groups. The nanoparticles showed an initial burst release of amoxicillin for 15 h followed by extended-release up to 81 h and demonstrated an excellent antibacterial activity by producing inhibition zones of 17.6 ± 0.3 mm and 19.5 mm ± 0.4 mm for Escherichia coli and Staphylococcus aureus. Live/dead cell staining and MTT assay confirmed the non-toxic nature of Sr/Fe co-doped HAp bionanomaterial towards MC3T3-E1 cells. Further, an improved ALP activity, increased calcium deposition, enhanced RUNX2 expression, and regulated OPN and OCN expression levels suggest in MC3T3-E1 cells demonstrate the maturation of osteoblasts. This study provides the unique advantages of the co-doping approach for the applications Sr/Fe co-doped HAp bionanomaterials as a bone substitute.
Estimating Pediatric Acute Respiratory Distress Syndrome Outcomes From Oxygenation Variables.

Abstract:654 www.ccmjournal.org April 2018 • Volume 46 • Number 4 Pediatric acute respiratory distress syndrome (ARDS) remains a significant antecedent of childhood morbidity and mortality worldwide (1, 2). Quality interventional trials are needed to identify evidence-based interventions to define best practice regarding critical care for these patients. It makes sense to identify the population that is most likely to derive benefit from the treatment under investigation: some patients may be too moribund for any intervention to be effective, whereas others may be minimally affected by the illness or injury and will survive intact regardless and may be exposed to potential adverse effects of the intervention without potential benefit (3, 4). It has previously been established that discrimination of mortality in pediatric ARDS based on oxygenation variables is improved after 24 hours compared with at ARDS onset (5–7). Ideally, discrimination of the subject population should occur early in the disease process, however, in order for the intervention to have the greatest chance of being effective (8). Accordingly, identifying patients at high risk for poor outcomes while also shortening the timeframe for ARDS screening and trial recruitment would be desirable. The article by Yehya et al (9) in this issue of Critical Care Medicine describes an observational cohort study that examined oxygenation variables (Pao 2 /Fio 2 and oxygenation index [Fio 2 × mean airway pressure × 100]/Pao 2 ) to predict pulmonary-focused outcomes (duration of mechanical ventilation, ventilator free days, probability of extubation) and global outcomes (mortality) among two cohorts of children with ARDS. Data for the derivation cohort were gleaned from a database at Children’s Hospital of Philadelphia (CHOP), whereas data for the validation cohort were obtained from a similar but independent database maintained at Children’s Hospital of Los Angeles (CHLA). It is important to stress that ventilation strategies and use of ancillary therapies were not protocolized for either of these cohorts. It is unclear if mechanical ventilator–related outcomes are reliable in the absence of ventilator weaning and extubation readiness testing (and perhaps sedation weaning) protocols, as multiple variables besides natural history of the pulmonary disease process and interventions are operative. The investigators first ascertain that no relationship exists between oxygenation variables at ARDS onset and mortality in either cohort (although discrimination improved when the cohorts were combined with added power). Some patients initially categorized with ARDS will improve quickly, perhaps with a little lung recruitment strategy; indeed, in this cohort, over half of patients only had mild ARDS or no longer met ARDS criteria by 12 hours. These patients would be poor candidates for additional experimental intervention, just adding noise to the outcome signal. Next, the investigators demonstrated a statistically significant dose-response relationship between Pediatric Acute Lung Injury Consensus Conference (PALICC) oxygenation index categories and risk of mortality at 12 and 24 hours after ARDS diagnosis (increasing mortality with worsening oxygenation, Fig. 1 in [9]). These findings provide face validity for use of oxygenation index to identify an appropriate experimental cohort with risk for the gold standard patient-centered outcome, mortality. Table 1 in (9) provides a summary of areas under the receiver operating characteristic (AUROC) curve indicating that between 6 and 24 hours, both Pao 2 /Fio 2 (Berlin criteria) and oxygenation index (PALICC criteria) predict statistically relevant risks for mortality. However, the AUROC for Pao 2 /Fio 2 and oxygenation index were only 0.57–0.62 and 0.60–0.62, respectively. For this type of analysis, AUROC of 0.70–0.80 is considered acceptable discrimination (10). With AUROC less than this, considerable lack of sensitivity and/or specificity must exist, suggesting that oxygenation variables alone do not consistently identify the appropriate population for an ARDS experimental intervention. At 12 and 24 hours, calibration for mortality was poor in the CHLA validation cohort, with r less than or equal to 0.70. The fact that mortality at CHLA was 23.1% as compared to 14.6% at CHOP with both cohorts exhibiting similar Pediatric Risk of Mortality (PRISM) scores underscores the fact that multiple variables (not quantified in this investigation) influence pediatric ARDS mortality. In fact, most children succumbing from ARDS do not die of refractory hypoxemia (11, 12); specifically, for this cohort, only 2.8% died from this cause. On the other hand, ventilator-free days, ventilator days among survivors, and probability of tracheal extubation all demonstrated improved association with the oxygenation variable severity at 12 and 24 hours, with better *See also p. 619.

Abstract:Actions of a large number of nucleic acid derivatives were studied on the aortic pressure pulse, arterial pressure, femoral artery flow, and the electrocardiogram of anesthetized dogs, and on the rate and amplitude of contraction and coronary outflow of perfused rabbit hearts. Of the compounds examined only adenosine-3-phosphoric acid, adenosine-5-phosphoric acid, and guanylic acid had significant cardiovascular activity in the doses used. Some general relationships between chemical structure and vascular activity are presented.
Development and evaluation of osteogenesis and antibacterial properties of strontium/silver-functionalized hierarchical micro/nano-titanium implants

Abstract:Promoting implant osseointegration while preventing infection is one of the major challenges in the development of dental implants. In this study, nano-needle-like structures were prepared on titanium by hot alkali treatment and then coated with uniform silver (Ag) and strontium titanate (SrTiO 3 ) through magnetron sputtering. The results of ion release showed that the SrTiO 3 outer coating reduced the release of Ag ions by 30% and 15% respectively on the 4th and 7th days, thus effectively improving the bioactivity of the Ag-modified substrates. In vitro experiments demonstrated that the Ag/SrTiO 3 -coated specimens (AH-Ti/Ag/Sr) had an excellent effect on the osteogenic differentiation of MC3T3-E1 cells and exhibited antibacterial function against Staphylococcus aureus and Escherichia coli (percentage of dead/total bacteria: ∼93% and ∼88%). Moreover, the in vivo results further confirmed that the Ag/SrTiO 3 bilayer coating significantly promoted osteogenesis around the AH-Ti/Ag/Sr implant (the new bone volume and the number of trabeculae: 1.5 and 1.4 fold of the control group) and enhanced the bonding strength between implant and surrounding bone (about 1.9 fold of the control group). All these findings indicate that AH-Ti/Ag/Sr can substantially improve osteogenesis in complex oral microenvironment, and thus has potential applications in oral implantology.

Abstract:Accelerated osseointegration is well considered as a critical facilitated factor for titanium scaffold longevity，but immensely limited by bioinert material surfaces and restricted biological responses in vivo. Here, we fabricated three-dimensional printed porous titanium scaffold modified with titanium dioxide (TiO2) nanotubes (p-TNTs), for mimicking the hierarchical trabecular bone structure and building local drug delivery system to accelerate osseointegration. The scaffold at macro and micro gradient was manufactured by selective laser melting process with pure titanium powder according to the computer-aided design (CAD), which could be further anodized to construct nano-gradient frameworks and drug-loading districts. Sequentially, 1α,25-Dihydroxyvitamin D3 (VD3) was loaded into anodized TiO2 nanotubes，thermo-sensitive hydrogel Pluronic F-127 (F-127) was coated and charged as a bio-cap for control release. Biological responses in vitro and in vivo was evaluated, showing significant osteogenesis enhancement of hierarchical structure with local drug delivery. It suggests that a promising bioactive implant system may provide a far-reaching impact on accelerating and enhancing early osseointegration.

Abstract:Biomaterial with the dual-functions of bone regeneration and antibacterial is a novel therapy for infective bone defects. Three-dimensional (3D)-printed porous titanium (pTi) benefits bone ingrowth, but its microporous structure conducive to bacteria reproduction. Herein, a multifunctional hydrogel was prepared from dynamic supramolecular assembly of sodium tetraborate (Na 2 B 4 O 7 ), polyvinyl alcohol (PVA), silver nanoparticles (AgNPs) and tetraethyl orthosilicate (TEOS), and composited with pTi as an implant system. The pTi scaffolds have ideal pore size and porosity matching with bone, while the supramolecular hydrogel endows pTi scaffolds with antibacterial and biological activity. In vitro assessments indicated the 3D composite implant was biocompatible, promoted bone marrow mesenchymal stem cells (BMSCs) proliferation and osteogenic differentiation, and inhibited bacteria, simultaneously. In vivo experiments further demonstrated that the implant showed effective antibacterial ability while promoting bone regeneration. Besides distal femur defect, the innovative scaffolds may also serve as an ideal biomaterial (e.g. dental implants) for other contaminated defects.

Abstract:As a traditional bone implant material, titanium (Ti) and its alloys have the disadvantages of lack of biological activity and susceptibility to stress shielding effect. Adipose stem cells (ADSCs) and exosomes were combined with the scaffold material in the current work to effectively create a hydroxyapatite (HA) coated porous titanium alloy scaffold that can load ADSCs and release exosomes over time. The composite made up for the drawbacks of traditional titanium alloy materials with higher mechanical characteristics and a quicker rate of osseointegration. Exosomes (Exos) are capable of promoting the development of ADSCs in porous titanium alloy scaffolds with HA coating, based on experimental findings from in vitro and in vivo research. Additionally, compared to pure Ti implants, the HA scaffolds loaded with adipose stem cell exosomes demonstrated improved bone regeneration capability and bone integration ability. It offers a theoretical foundation for the combined use of stem cell treatment and bone tissue engineering, as well as a design concept for the creation and use of novel clinical bone defect repair materials.

Abstract:Poor osseointegration and postoperative bacterial infections are the main causes of clinical failure of titanium (Ti) based implants. In this study, calcium (Ca) and strontium (Sr), which have osteogenic activity, were simultaneously introduced into titanium dioxide nanotubes (TN) in order to balance the potential cytotoxicity of GL13K (Antibacterial peptides), and a multifunctional Sr, Ca and GL13K-doped TN (SrCaTN-GL13K) coating was successfully constructed. The results showed that the SrCaTN-GL13K coating exhibited good mechanical properties, hydrophilicity, corrosion resistance and bioactivity. Sr2+, Ca2+ and GL13K could be continuously released from the coating and was pH-responsive. In addition, SrCaTN-GL13K showed good antibacterial properties against E. coli and S. aureus with antibacterial rates of approximately 70.32% and 70.78%, respectively. Cellular assays showed that RAW264.7 (immune), human umbilical vein endothelial cell (HUVEC, angiogenic) and mouse embryo osteoblast precursor cell (MC3T3-E1, osteogenic) exhibited good adhesion, survival and proliferation activities on the SrCaTN-GL13K coated surface. At both gene level and protein level, expression of anti-inflammatory markers (Interleukin-10, IL-10; mannose receptor, CD206), angiogenic markers (vascular endothelial growth factor, VEGF; platelet endothelial cell adhesion molecule-1, CD31) and osteogenic markers (alkaline phosphatase, ALP; runt-related transcription factor 2, RUNX2; human collagen type I, COL1a1; osteocalcin, OCN) were significantly upregulated in SrCaTN-GL13K coating. At the cellular and molecular level, SrCaTN-GL13K possessed the ability to induce RAW264.7 towards M2 polarization and it possessed the ability to induce HUVEC tube-formation; meanwhile, it exhibited the ability to induce MC3T3-E1 towards osteogenic differentiation. In conclusion, Ti implants exhibited improved antibacterial, anti-inflammatory, angiogenic, and osteoinductive capabilities by loading Sr, Ca, and GL13K to alter TN. In this study, a multifunctional modification of Ti surface was achieved by an economical and efficient method, which provides a new idea for the design of antimicrobial implantable devices with immunomodulatory functions in the clinic.

Abstract:We present subpicosecond time-resolved four-wave-mixing studies on GaAs/AlAs short-period superlattices having different miniband widths. Photon echoes are observed as a consequence of the inhomogeneous broadening of the excitonic resonance. In addition, the photon-echo signal exhibits periodic modulations as a function of the time delay between the excitation laser pulses. Free excitons and excitons bound to neutral acceptors are identified as the underlying three-level system leading to the observed photon-echo quantum beats. From the beat period we can directly infer the dissociation energy of the bound exciton. In accordance with theory, we find that the dissociation energy decreases with increasing miniband width, i.e., when the excitons become less confined. In addition, we find that the dephasing rate of bound excitons is about one order of magnitude less than that of free excitons and is mainly determined by energy relaxation.
Metal Ion-Dependent Tailored Antibacterial Activity and Biological Properties of Polydopamine-Coated Titanium Implants

Abstract:There is a great need to develop new approaches for preventing bacterial adhesion and promoting cell growth on the surface of Ti implants. To this end, various metal ions (Ag+, Cu2+, Sr2+ or Zn2+) were sprayed onto the surface of the PDA (polydopamine)-immobilized Ti. The antibacterial activity of metal ion coatings against Gram-negative E. coli and Gram-positive S. aureus were examined. The content of reactive oxygen species (ROS) production from the metal ions in bacteria was also determined. L929, RAW 264.7 and human mesenchymal stem cells (hMSCs) were used to analyze biological function of the coatings. The results of phase composition and microstructure showed that metal ions were successfully coated on PDA-immobilized Ti surface. Three divalent metal ion coatings effectively inhibited bacterial growth in a concentration-dependent manner. Samples of Cu, Sr and Zn ions in E. coli and S. aureus produced 3–8 times more ROS than the negative control. The presence of Cu and Ag remarkably reduced the cell function, as evidenced by poor cell viability and differentiation. In contrast, high content of Sr promoted hMSC proliferation and differentiation. It is eventually concluded that, taking antibacterial ability and osteogenic activity into account, 10% Sr-coated Ti had potential for implant applications.
