Effect of inorganic arsenic in paddy soil on the migration and transformation of selenium species in rice plants

Abstract:Selenium (Se) in paddy rice is one of the significant sources of human Se nutrition. However, the effect of arsenic (As) pollution in soil on the translocation of Se species in rice plants is unclear. In this research, a pot experiment was designed to examine the effect of the addition of 50 mg As/kg soil as arsenite or arsenate on the migration of Se species from soil to indica Minghui 63 and Luyoumingzhan. The results showed that the antagonism between inorganic As and Se was closely related to the rice cultivar and Se oxidation state in soil. Relative to the standalone selenate treatment, arsenite significantly (p < 0.05) decreased the accumulation of selenocystine, selenomethionine and selenate in the roots, stems, sheaths, leaves, brans and kernels of both cultivars by 21.4%-100.0%, 40.0%-100.0%, 41.0%-100%, 5.4%-96.3%, 11.3%-100.0% and 26.2%-39.7% respectively, except for selenocystine in the kernels of indica Minghui 63 and selenomethionine in the leaves of indica Minghui 63 and the stems of indica Luyoumingzhan. Arsenate also decreased (p < 0.05) the accumulation of selenocystine, selenomethionine and selenate in the roots, stems, brans and kernels of both cultivars by 34.9%-100.0%, 30.2%-100.0%, 11.3%-100.0% and 5.6%-39.6% respectively, except for selenate in the stems of indica Minghui 63. However, relative to the standalone selenite treatment, arsenite and arsenate decreased (p < 0.05) the accumulation of selenocystine, selenomethionine and selenite only in the roots of indica Minghui 63 by 45.5%-100.0%. Our results suggested that arsenite and arsenate had better antagonism toward Se species in selenate-added soil than that in selenite-added soil; moreover, arsenite had a higher inhibiting effect on the accumulation of Se species than arsenate.

Abstract:Arsenic (As) and selenium (Se) are two metalloids found in the environment. As it poses a significant threat to human health and plant growth due to its prevalence and toxicity, however Se is a required micronutrient for human health. In this study hydroponic experiments were performed to investigate whether endogenous Se can mitigate As toxicity in rice (Oryza sativa L.). We found that As uptake by rice roots increased by pretreatment with selenateSe(VI) or selenite Se(IV). However, co-application of arsenate As(V) or arsenite As(III) with selenate markedly reduced the uptake of As by roots. Co- or pretreatment with Se with five μM of As(V) or one μM of As(III) significantly decreased shoot As content. Conversely, Se pretreatment before the addition of five μM of As(III) or one μM of As(V) resulted in As accumulation in the shoot compared to As and Se co-application. As translocation to the shoot was lower whereas the transfer factor was higher upon the simultaneous application of Se and As compared to Se pretreatment. Se supplementation with As(III) or pretreatment increased antioxidant enzymes: Superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) increased in the root and shoot, but decreased glutathione (GSH) and malondialdehyde (MDA) contents in the shoot. Plants under As(V) treatment showed the same trend except that CAT content decreased in the root and shoot, while MDA content increased in the shoot. These results suggest that cultivating rice in the presence of Se can reduce the accumulation of toxic As in seedlings, thus ensuring the safety of this important crop for human consumption.

Selenite Foliar Application Alleviates Arsenic Uptake, Accumulation, Migration and Increases Photosynthesis of Different Upland Rice Varieties

Abstract:This study investigates how arsenic (As) uptake, accumulation, and migration responds to selenium (Se) foliar application (0–5.0 mg × kg−1). Rice varieties known to accumulate low (DOURADOAGULHA) and high (SINALOAA68) concentrations of arsenic were chosen to grow on soil with different As concentrations (20.1, 65.2, 83.9 mg × kg−1). The results showed that Se of 1.0 mg × L−1 significantly alleviated As stress on upland rice grown on the As-contaminated soil. Under light (65.2 mg × kg−1) and moderate (83.9 mg × kg−1) As concentration treatments, the biomass of upland rice was increased by 23.15% and 36.46% for DOURADOAGULHA, and 46.3% and 54.9% for SINALOAA68. However, the high Se dose (5.0 mg × kg−1) had no significant effect on biomass and heights of upland rice compared to plants where no Se was added. Se significantly decreased As contents in stems and leaves and had different effects on As transfer coefficients for the two rice varieties: when grown on soil with low and moderate As concentrations, Se could reduce the transfer coefficient from stems to leaves, but when grown on the high As soils, this was not the case. The chlorophyll content in plants grown in soil with the moderate concentration of As could be improved by 27.4%–55.3% compared with no Se treatment. Under different As stress, the Se foliar application increased the net photosynthesis, stomatal conductance, and transpiration rate, which meant that Se could enhance the photosynthesis of rice. The intercellular CO2 concentration variation implied that the stomatal or non-stomatal limitations could both occur for different rice varieties under different Se application doses. In conclusion, under moderate As stress, foliar application of Se (1.0 mg × L−1) is recommend to overcome plant damage and As accumulation.

Abstract:Although the single role of selenium (Se) or phosphorus (P) in regulating the As contamination of rice plants has been reported in some studies, the combined impacts of Se and P on the fate of As and the underlying mechanisms are poorly understood. To address this knowledge gap, the uptake, translocation, and biotransformation of As mediated by Se were investigated in rice (Oryza sativa L.) seedlings hydroponically cultured with P-normal and P-deficient conditions. The results showed Se addition stimulated the uptake of arsenite and arsenate by 15.6% and 30.7%, respectively in P-normal condition, and such effect was more profound in P-deficient condition with the value of 43.8% and 70.8%. However, regardless of Se addition, P-deficiency elevated the As uptake by 47.0%-92.1% for arsenate but had no obvious effects for arsenite. Accompanying with the As transfer factorShoot/Root reduced by 74.5%-80.2% and 71.1%-85.7%, Se addition decreased the shoot As content by 65.8%-69.7% and 59.6%-73.1%, respectively, in the arsenite- and arsenate-treated rice plants. Relative to the corresponding treatments of P-normal condition, P-deficiency reduced the As transfer factorShoot/Root by 38.9%-52.5% and thus decreasing the shoot As content by 35.2%-42.5% in the arsenite-treated plants; while the opposite impacts were observed in the arsenate-treated plants, in which the shoot As content was increased by 22.4%-83.7%. The analysis results of As species showed As(III) was dominant in both shoots (68.9%-75.1%) and roots (94.9%-97.2%), and neither Se addition nor P-deficiency had obvious impacts on the interconversion between As(III) and As(V). Our results demonstrate the regulating roles of Se in As accumulation mainly depend on P regimes and the specific rice tissues, but the effects of P-deficiency on the fate of As were influenced by the form of As added to the culture.

Abstract:We describe the clinical, histopathologic, and immunohistochemical characteristics of five examples of a distinctive subtype of neurothekeoma we term “cellular neurothekeoma” (CNT). These lesions are nondescript papules or nodules primarily involving the head and neck areas of young adults. Histopathologically, CNT are fairly well-defined proliferations involving the reticular dermis; they consist of fascicles of polygonal and spindle cells with eosinophilic or pale-staining cytoplasm and neuroid characteristics. Low-grade cytologic atypia and mitotic activity are common. All immunohistochemical markers—including S-100 protein, myelin basic protein, epithelial membrane antigen, and histiocytic antigens— have failed to show positivity in our laboratory. Separation from myxomatous variants of neurothekeoma is based on greater cellularity, less myxomatous change, and less pronounced plexiform compartmentalization by fibrous septae, which resemble perineurium. The differential diagnosis usually includes spindle and epithelioid cell (Spitz) nevus, malignant melanoma (particularly desmoplastic-neurotropic melanoma), cellular blue nevus, and fibrohistiocytic proliferations. The recognition of CNT and its differentiation from melanoma are important so that overly aggressive therapy is avoided.
Arsenic Contents, Speciation and Toxicity in Germinated Rice Alleviated by Selenium

Abstract:Rice can accumulate more organic and inorganic arsenic (iAs) than other crop plants. In this study, the localization of As in rice grains was investigated using High Performance Liquid Chromatography-Inductively Coupled Plasma Mass Spectrometry (HPLC-ICP-MS) based on 26 rice varieties collected from two provinces. In all the samples, the total As contents in polished rice were 0.03-0.37 mg/kg, with average values of 0.28 and 0.21 mg/kg for two sample sets. The results of the determination of arsenic speciation in different components of rice grain showed that in the polished and brown rice the mean value of arsenite (As(III)) was nearly twice than that of arsenate (As(V)). The regional difference was observed in both total As contents and As speciation. The reason may be that As(III) is more mobile than As(V) in a dissociated form and because of soil properties, rice varieties, and the growing environment. The proportion of iAs and the total As in rice bran was higher than that in polished rice, and this is because As tends accumulate between the husk and the endosperm. In our study, selenium could alleviate the risk of arsenic toxicity at the primary stage of rice growth. Co-exposure to As and Se in germinated rice indicated that the reduction in As accumulation in polished rice reached 73.8%, 76.8%, and 78.3% for total As, As(III), and As(V) when compared with rice treated with As alone. The addition of Se (0.3 mg/kg) along with As significantly reduced the As amount in different parts of germinated rice. Our results indicated that Se biofortification could alleviate the As accumulation and toxicity in rice crops.

Abstract:Arsenic (As), one of the major pollutants in the soil, is an important environmental concern as its consumption can cause adverse health symptoms in living organisms. Its contamination of rice grown over As-contaminated areas is a serious concern in South Asian countries. Selenium (Se) has been reported to influence various osmolytes under metal stress in plants. The present study reports the role of Se in mitigating As stress in rice by modulating osmolyte metabolism. Rice plants grown in As-amended soil (2.5–10 mg kg −1 ) in pots were treated with sodium selenate (0.5–1.0 mg Se kg −1 soil) in glass house conditions and leaf samples were collected at 60 and 90 days after sowing (DAS). As-treated rice leaves displayed a reduction in relative water content (RWC) and dry weight than control with a maximum reduction of 1.68- and 2.47-fold in RWC and 1.95- and 1.69-fold in dry weight in As10 treatment at 60 and 90 DAS, respectively. Free amino acids (1.38–2.26-fold), proline (3.88–3.93-fold), glycine betaine (GB) (1.27–1.72-fold), choline (1.67–3.1-fold), total soluble sugars (1.29–1.61-fold), and reducing sugars (1.67–2.19-fold) increased in As-treated rice leaves as compared to control at both stages. As stress increased the γ-aminobutyric acid (GABA), putrescine content, and glutamate decarboxylase activity whereas diamine oxidase and polyamine oxidase activities declined by 1.69–1.88-fold and 1.52–1.86-fold, respectively. Se alone or in combination with As improved plant growth, RWC, GB, choline, putrescine, and sugars; lowered proline and GABA; and showed a reverse trend of enzyme activities related to their metabolism than respective As treatments. As stress resulted in a higher accumulation of osmolytes to combat its stress which was further modulated by the Se application. Hence, the current investigation suggested the role of osmoprotectants in Se-induced amelioration of As toxicity in rice plants.

Abstract:AbstractA green house experiment was conducted to evaluate the efficacy of soil application of selenium (Se) in modulating metabolic changes in rice under arsenic (As) stress. Rice plants were grown over soil amended with sodium arsenate (25, 50 and 100 μM kg-1 soil) with or without sodium selenate @ 0.5 and 1 mg kg-1 soil in a complete randomized experimental design, and photosynthetic efficiency, nutrient uptake and nitrogen metabolism in rice leaves were estimated at tillering and grain filling stages. Se treatments significantly improved the toxic effects of As on plant height, leaf dry weight and grain yield. Arsenate treatment reduced uptake of Na, Mg, P, K, Ca, Mn, Fe and Zn and lowered chlorophyll, carotenoids and activities of enzymes of nitrogen metabolism (nitrate reductase, nitrite reductase, glutamine synthase and glutamate synthase) in rice leaves at both the stages in a dose-dependent fashion. Se application along with As improved photosynthesis, nutrient uptake and arsenate-induced effects on activities of enzymes of nitrogen metabolism with maximum impact shown by As50 + Se1 combination. Application of Se can modulate photosynthetic efficiency, nutrient uptake and alterations in nitrogen metabolism in rice Cv PR126 due to As stress that helped plants to adapt to excess As and resulted in improved plant growth.

Abstract:Rice (Oryza sativa L.) is the staple food for more than 50% of the world's population. Owing to its growth characteristics, rice has more than 10-fold the ability to enrich the carcinogen arsenic (As) than other crops, which seriously affects world food security. The consumption of rice is one of the primary ways for humans to intake As, and it endangers human health. Effective measures to control As pollution need to be studied and promoted. Currently, there have been many studies on reducing the accumulation of As in rice. They are generally divided into agronomic practices and biotechnological approaches, but simultaneously, the problem of using the same measures to obtain the opposite results may be due to the different species of As or soil environments. There is a lack of systematic discussion on measures to reduce As in rice based on its mechanism of action. Therefore, an in-depth understanding of the molecular mechanism of the accumulation of As in rice could result in accurate measures to reduce the content of As based on local conditions. Different species of As have different toxicity and metabolic pathways. This review comprehensively summarizes and reviews the molecular mechanisms of toxicity, absorption, transport and redistribution of different species of As in rice in recent years, and the agronomic measures to effectively reduce the accumulation of As in rice and the genetic resources that can be used to breed for rice that only accumulates low levels of As. The goal of this review is to provide theoretical support for the prevention and control of As pollution in rice, facilitate the creation of new types of germplasm aiming to develop without arsenic accumulation or within an acceptable limit to prevent the health consequences associated with heavy metal As as described here.

Regulation of oxidative stress and mineral nutrient status by selenium in arsenic treated crop plant Oryza sativa

Abstract:The present study was conducted to examine the impact of selenium (Se) on mineral nutrient status and oxidative stress in crop plant Oryza sativa treated with arsenic (As). Scanning electron microscopy (SEM) coupled with Energy dispersive x-ray spectroscopy (EDS) study revealed the morphological deformities in leaf veins along with granular deposition on the leaf surface. The EDS analysis exhibited loss of elements (S, Si, Cl, K, Ca, Fe and Cu) in As(III) treatment in rice roots as compared to untreated root. In the case of As(III) treated shoot, changes in elements content in term of percent atomic weight was K (1.17-0.90%), Cl (1.04-24.75%), Na (0.65-3.52%) and S (0.49-2.52%) when compared with untreated shoot. The result of EDS analysis showed that As limits the concentration of important mineral elements present in the rice root and shoot. Rice plant treated with Se (10µM) and sub lethal dose of As(III) (60µM) showed better growth responses in term of root, shoot length (11.4% and 10.71%, respectively), biomass (11.7%), reduced malonyldialdehyde content (35.14%) and stimulated antioxidant level indicating better As tolerance potential against As. Further, a selenium dependent significant reduction in As accumulation was also observed in root (14.24%) and shoot (23.78%) of rice plant when compared with plant treated with As alone. This study highlights the potential of Se to ameliorate the ecotoxicological risks associated with the As buildup in agricultural land.

Abstract:Selenate enhances arsenic (As) accumulation in As-hyperaccumulator , but the associated molecular mechanisms are unclear. Here, we investigated the mechanisms of selenate-induced arsenic accumulation by exposing to 50 μM arsenate (AsV) and 1.25 (Se) or 5 μM (Se) selenate in hydroponics. After 2 weeks, plant biomass, plant As and Se contents, As speciation in plant and growth media, and important genes related to As detoxification in were determined. These genes included P transporters and (AsV uptake), arsenate reductases and (AsV reduction), and arsenite (AsIII) antiporters and (AsIII translocation) in the roots, and AsIII antiporters and (AsIII sequestration) in the fronds. The results show that Se was more effective than Se in increasing As accumulation in both roots and fronds, which increased by 27 and 153% to 353 and 506 mg kg. The As speciation analyses show that selenate increased the AsIII levels in , with 124-282% more AsIII being translocated into the fronds. The qPCR analyses indicate that Se upregulated the gene expression of by 1.2-fold, and and by 1.0- to 2.5-fold in the roots, and and by 0.6- to 1.1-fold in the fronds under AsV treatment. Though arsenate enhanced gene expression of P transporters and , selenate had little effect. Our results indicate that selenate effectively increased As accumulation in , mostly by increasing reduction of AsV to AsIII in the roots, AsIII translocation from the roots to fronds, and AsIII sequestration into the vacuoles in the fronds. The results suggest that selenate may be used to enhance phytoremediation of As-contaminated soils using .
Microbiological quality of Australian beef.

Abstract:A survey of the microbiological quality of beef carcasses and boneless beef produced in Australia was conducted during the period June to November 1998. Sponge samples were collected from 1,275 carcasses, and meat samples were drilled from 990 cartons of frozen boneless beef. Carcass and boneless beef samples were respectively collected from 21 and 27 establishments that concentrated on export and from 38 and 3 establishments supplying the Australian domestic market of which 31 were very small plants slaughtering no more than 150 cattle equivalents per week. The mean log total viable counts (TVCs) were 2.42/cm2 and 2.52/g for carcasses and boneless meat, respectively. Escherichia coli was detected on 10.3% of carcasses and 5.1% of boneless beef samples and coagulase-positive staphylococci on 24.3% of carcasses and 17.5% of boneless beef. Salmonella was detected on 0.2% of carcasses and 0.1% of boneless beef and E. coli O157:H7 recovered from 0.1% of carcasses but not detected on 990 boneless beef samples. Mean log TVCs/cm2 differed significantly (P < 0.05) between establishment types. They were lower on carcasses from export establishments (2.20) compared with domestic (2.61) and very small plants (3.10). There were no significant differences in prevalence of Salmonella or E. coli O157:H7 between establishment types. Excision samples were taken from 670 carcasses to make comparisons with the first baseline study of Australian meat, carried out in 1993 to 1994. While there were differences in sampling and microbiological techniques between the two studies that require detailed consideration, there were small but significant improvements in several microbiological criteria for carcasses and boneless meat.

Abstract:Anthropogenic activities such as mining, industrialization and subsequent emission of industrial waste, and agricultural practices have led to an increase in the accumulation of metal(loid)s in agricultural soils and crops, which threatens the health of people; the risk is more pronounced for individuals whose survival depends on food sources from several contaminated regions. Selenium (Se) is an element essential for the normal functioning of the human body and is a beneficial element for plants. Se deficiency in the diet is a common issue in many countries around the world, such as China and Egypt. >40 diseases are associated with Se deficiency. In practice, Se compounds have been applied through foliar sprays or via base application of fertilizers to increase Se concentration in the edible parts of crops and to satisfy the daily Se intake. Moreover, Se at low concentrations has been used to mitigate the toxicity of many metal(loid)s. In this review, we present an overview of the latest knowledge and practices with regards to the utilization of Se to reduce the uptake/toxicity of metal(loid)s in plants. We have focused on the following issues: 1) the current status of understanding the mechanisms of detoxification and uptake restriction of metal(loid)s regulated by Se; 2) the optimal dose and speciation of Se, and stage of plant growth that is optimal for application; 3) the differences in the efficiency of different application methods of Se including seed priming, base application, and foliar spray of Se fertilizers; 4) the possibility of using Se along with other methods to reduce multiple metal(loid) accumulation in crops; and 5) potential risks when Se is used to reduce metal(loid) accumulation in crops.
Arsenic and Cadmium Bioavailability to Rice ( Oryza Sativa L.) Plant in Paddy Soil: Influence of Sulfate Application

Abstract:Arsenic (As) and cadmium (Cd) accumulate easily in rice grains that pose a non-negligible threat to human health worldwide. Sulfur fertilizer has been shown to affect the mobilization of As and Cd in paddy soil, but the effect of co-contamination by As and Cd has not been explored. This study selected three soils co-contaminated with As and Cd from Shangyu (SY), Tongling (TL) and Ma'anshan (MA). Incubation experiments and pot experiments were carried out to explore the effect of sulfate supply (100 mg kg-1) on the bioavailability of As and Cd in soil and the rice growth. The results showed that the exogenous sulfate decreased As concentrations in porewater of SY and TL by 51.1% and 29.2% through forming arsenic-sulfide minerals. The exchangeable Cd in soil also declined by 25.6% and 18.6% and transformed into Fe and Mn oxides-bound Cd. The relative abundance of Desulfotomaculum, Desulfurispora and dsr gene increased remarkably indicated that sulfate addition stimulated the activity of sulfate-reducing bacteria. In MA soil, sulfate addition immobilized Cd but had little effect on As solubility, which was speculated to be related to the high sulfate background of the soil. Further pot experiments showed that sulfate application significantly increased rice tillers, biomass, chlorophyll content in shoots, and decreased electrolyte leakage in root. Finally, sulfate significantly reduced As and Cd in SY rice shoots by 60.2% and 40.8%, respectively, while As decreased by 39.6% in TL rice shoots and Cd decreased by 23.0% in MA rice shoots. These results indicate that the application of sulfate can reduce the bioavailability of As and Cd in the soil-rice system and promote rice growth, and it is possible to reduce the accumulation of As and Cd in rice plants simultaneously.
Sulfur reduces arsenic accumulation in rice shoot by enhancing root retention and altering arsenic metabolism

Abstract:Rice can potentially pose serious health risks due to its higher arsenic (As) uptake. Sulfur (S) is not only an essential macronutrient, but it also has the ability to decrease As accumulation. In the present study, a hydroponic experiment was conducted to investigate the mechanisms underlying the effects of S on the As uptake and transport at different S (pre-)treatments and additional supply levels. It was found that additional S supply decreased As content by 20%–50% in both S-deficient and S-normal pre-treated shoots compared to the no S supply throughout the treatment; As root-to-shoot translocation factors was reduced by 7%–46% with S supply. On the one hand, additional S supply could elevate levels of thiol compounds (by 15%–280%) and increase the As percentage in soluble cytosol of roots. Additional S supply also enhanced the casparian strip development of rice roots, which could block As transfer in roots apoplast pathway. Moreover, additional S supply lead to the down-regulation of OsLsi2 expression (e.g., reduced by 71% by S at 2 mmol L −1 with the S-normal pre-treatment). Sulfur also promoted the biotransformation of As(III) in shoots into less toxic As species; reducing the As(III) proportion by 25% by 2 mmol L −1 of S under S-normal pre-treatment. These results suggest that S could play an important role in the inhibition of As transfer and the detoxification of As in rice by enhancing root retention (the vacuole sequestration), impeding transportation pathway of root apoplast, and regulating As-related gene expression. Thus, providing a basis for the potential application of S in rice production in As-contaminated paddy soil.

Abstract:Arsenic (As) exposure poses a serious threat to human health. The present study investigated the effects of organic Se on As accumulation, migration, and As bioaccessibility in As-stressed radish. The results showed that organic Se can effectively block the accumulation of As in radish, reduce As bioaccessibility, and promote the conversion of As from inorganic to organic form. The total As content decreased with increasing Se application in raw radish roots, the gastric fraction and the gastrointestinal fraction. Compared to the control (CK) group, the As bioaccessibility in the 24Se treatment of the yeast Se and malt Se groups decreased by 26% and 37%, respectively. These findings provide new comprehensive information for the application of organic Se to alleviate the toxicological effects of As and reduce the health risks of As in edible plants. In the future, it is necessary to carry out a deeper study of the interaction between Se and As through advanced analytical methods.
