Abstract:Arsenic (As) and selenium (Se) are two metalloids found in the environment. As it poses a significant threat to human health and plant growth due to its prevalence and toxicity, however Se is a required micronutrient for human health. In this study hydroponic experiments were performed to investigate whether endogenous Se can mitigate As toxicity in rice (Oryza sativa L.). We found that As uptake by rice roots increased by pretreatment with selenateSe(VI) or selenite Se(IV). However, co-application of arsenate As(V) or arsenite As(III) with selenate markedly reduced the uptake of As by roots. Co- or pretreatment with Se with five μM of As(V) or one μM of As(III) significantly decreased shoot As content. Conversely, Se pretreatment before the addition of five μM of As(III) or one μM of As(V) resulted in As accumulation in the shoot compared to As and Se co-application. As translocation to the shoot was lower whereas the transfer factor was higher upon the simultaneous application of Se and As compared to Se pretreatment. Se supplementation with As(III) or pretreatment increased antioxidant enzymes: Superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) increased in the root and shoot, but decreased glutathione (GSH) and malondialdehyde (MDA) contents in the shoot. Plants under As(V) treatment showed the same trend except that CAT content decreased in the root and shoot, while MDA content increased in the shoot. These results suggest that cultivating rice in the presence of Se can reduce the accumulation of toxic As in seedlings, thus ensuring the safety of this important crop for human consumption.

Effect of inorganic arsenic in paddy soil on the migration and transformation of selenium species in rice plants

Abstract:Selenium (Se) in paddy rice is one of the significant sources of human Se nutrition. However, the effect of arsenic (As) pollution in soil on the translocation of Se species in rice plants is unclear. In this research, a pot experiment was designed to examine the effect of the addition of 50 mg As/kg soil as arsenite or arsenate on the migration of Se species from soil to indica Minghui 63 and Luyoumingzhan. The results showed that the antagonism between inorganic As and Se was closely related to the rice cultivar and Se oxidation state in soil. Relative to the standalone selenate treatment, arsenite significantly (p < 0.05) decreased the accumulation of selenocystine, selenomethionine and selenate in the roots, stems, sheaths, leaves, brans and kernels of both cultivars by 21.4%-100.0%, 40.0%-100.0%, 41.0%-100%, 5.4%-96.3%, 11.3%-100.0% and 26.2%-39.7% respectively, except for selenocystine in the kernels of indica Minghui 63 and selenomethionine in the leaves of indica Minghui 63 and the stems of indica Luyoumingzhan. Arsenate also decreased (p < 0.05) the accumulation of selenocystine, selenomethionine and selenate in the roots, stems, brans and kernels of both cultivars by 34.9%-100.0%, 30.2%-100.0%, 11.3%-100.0% and 5.6%-39.6% respectively, except for selenate in the stems of indica Minghui 63. However, relative to the standalone selenite treatment, arsenite and arsenate decreased (p < 0.05) the accumulation of selenocystine, selenomethionine and selenite only in the roots of indica Minghui 63 by 45.5%-100.0%. Our results suggested that arsenite and arsenate had better antagonism toward Se species in selenate-added soil than that in selenite-added soil; moreover, arsenite had a higher inhibiting effect on the accumulation of Se species than arsenate.
The combined impacts of selenium and phosphorus on the fate of arsenic in rice seedlings (Oryza sativa L.)

Abstract:Although the single role of selenium (Se) or phosphorus (P) in regulating the As contamination of rice plants has been reported in some studies, the combined impacts of Se and P on the fate of As and the underlying mechanisms are poorly understood. To address this knowledge gap, the uptake, translocation, and biotransformation of As mediated by Se were investigated in rice (Oryza sativa L.) seedlings hydroponically cultured with P-normal and P-deficient conditions. The results showed Se addition stimulated the uptake of arsenite and arsenate by 15.6% and 30.7%, respectively in P-normal condition, and such effect was more profound in P-deficient condition with the value of 43.8% and 70.8%. However, regardless of Se addition, P-deficiency elevated the As uptake by 47.0%-92.1% for arsenate but had no obvious effects for arsenite. Accompanying with the As transfer factorShoot/Root reduced by 74.5%-80.2% and 71.1%-85.7%, Se addition decreased the shoot As content by 65.8%-69.7% and 59.6%-73.1%, respectively, in the arsenite- and arsenate-treated rice plants. Relative to the corresponding treatments of P-normal condition, P-deficiency reduced the As transfer factorShoot/Root by 38.9%-52.5% and thus decreasing the shoot As content by 35.2%-42.5% in the arsenite-treated plants; while the opposite impacts were observed in the arsenate-treated plants, in which the shoot As content was increased by 22.4%-83.7%. The analysis results of As species showed As(III) was dominant in both shoots (68.9%-75.1%) and roots (94.9%-97.2%), and neither Se addition nor P-deficiency had obvious impacts on the interconversion between As(III) and As(V). Our results demonstrate the regulating roles of Se in As accumulation mainly depend on P regimes and the specific rice tissues, but the effects of P-deficiency on the fate of As were influenced by the form of As added to the culture.

Abstract:Rice can accumulate more organic and inorganic arsenic (iAs) than other crop plants. In this study, the localization of As in rice grains was investigated using High Performance Liquid Chromatography-Inductively Coupled Plasma Mass Spectrometry (HPLC-ICP-MS) based on 26 rice varieties collected from two provinces. In all the samples, the total As contents in polished rice were 0.03-0.37 mg/kg, with average values of 0.28 and 0.21 mg/kg for two sample sets. The results of the determination of arsenic speciation in different components of rice grain showed that in the polished and brown rice the mean value of arsenite (As(III)) was nearly twice than that of arsenate (As(V)). The regional difference was observed in both total As contents and As speciation. The reason may be that As(III) is more mobile than As(V) in a dissociated form and because of soil properties, rice varieties, and the growing environment. The proportion of iAs and the total As in rice bran was higher than that in polished rice, and this is because As tends accumulate between the husk and the endosperm. In our study, selenium could alleviate the risk of arsenic toxicity at the primary stage of rice growth. Co-exposure to As and Se in germinated rice indicated that the reduction in As accumulation in polished rice reached 73.8%, 76.8%, and 78.3% for total As, As(III), and As(V) when compared with rice treated with As alone. The addition of Se (0.3 mg/kg) along with As significantly reduced the As amount in different parts of germinated rice. Our results indicated that Se biofortification could alleviate the As accumulation and toxicity in rice crops.

Abstract:Abstract Background Selenium (Se) in soil mainly consists of selenite, selenate, and elemental Se. However, little is known about the mechanism involved in the uptake and biotransformation of elemental Se by plants. Results In this study, the uptake, translocation, subcellular distribution and biotransformation of selenium nanoparticles (SeNPs) in rice ( Oryza sativa L.), and a comparison with selenite and selenate, were investigated through hydroponic experiments. The study revealed that SeNPs could be absorbed by rice plants; and aquaporin inhibitor was responsible for a 60.4% inhibition of SeNP influx, while metabolic inhibitor was ineffective. However, the SeNPs uptake rate of rice roots was approximately 1.7 times slower than that of selenite or selenate. Under the SeNPs or selenite treatment, Se was primarily accumulated in roots rather than in shoots, whereas an opposite trend was observed with selenate treatment. Additionally, most of the absorbed Se was distributed in cell wall of the SeNPs or selenite treated-rice plants, while its proportion was the highest in soluble cytosol of the selenate treated-rice plants. The absorbed SeNPs or selenite was rapidly assimilated to organic forms, with SeMet being the most predominant species in both shoots and roots of the rice plants. However, following selenate treatment, Se(VI) remained as the most predominant species, and only a small amount of it was converted to organic forms. Conclusion Therefore, this study provides a deeper understanding of the mechanisms associated SeNPs uptake and biotransformation within plants.

Regulatory Mechanism of Copper Oxide Nanoparticles on Uptake of Different Species of Arsenic in Rice.

Abstract:Copper oxide nanoparticles (CuO NPs) are widely used as a fungicide in agriculture. The application of CuO NPs in agriculture affects the growth of rice and metal accumulation in rice. However, the mechanism of CuO NPs on arsenic (As) accumulation in rice remains unclear. In this study, a hydroponic culture was produced to investigate the mechanism of the effect of 50 and 100 mg L-1 CuO NPs on As accumulation in rice. Our results showed that CuO NPs decreased As(III/V) accumulation in the roots and shoots by adsorbing As(III/V), oxidizing of As(III) on the surface, and thickening the root cell wall. The addition of CuO NPs regulated the expression of the OsNIP1;1, OsHAC1;1, and OsHAC4 genes, which decreased As(III) transport and promoted As(V) reduction in the roots. Moreover, when CuO NPs were co-exposed to As, a negative correlation between the concentration of Cu and As in rice was also found in our study. However, CuO NPs significantly increased Cu accumulation in rice and constrained the rice growth. In conclusion, CuO NPs might be a promising way to decrease As accumulation in rice, but the negative effects such as growth inhibition should be further considered. Therefore, the application of CuO NPs in rice plants should take a more restrained approach.
Regulation of oxidative stress and mineral nutrient status by selenium in arsenic treated crop plant Oryza sativa

Abstract:The present study was conducted to examine the impact of selenium (Se) on mineral nutrient status and oxidative stress in crop plant Oryza sativa treated with arsenic (As). Scanning electron microscopy (SEM) coupled with Energy dispersive x-ray spectroscopy (EDS) study revealed the morphological deformities in leaf veins along with granular deposition on the leaf surface. The EDS analysis exhibited loss of elements (S, Si, Cl, K, Ca, Fe and Cu) in As(III) treatment in rice roots as compared to untreated root. In the case of As(III) treated shoot, changes in elements content in term of percent atomic weight was K (1.17-0.90%), Cl (1.04-24.75%), Na (0.65-3.52%) and S (0.49-2.52%) when compared with untreated shoot. The result of EDS analysis showed that As limits the concentration of important mineral elements present in the rice root and shoot. Rice plant treated with Se (10µM) and sub lethal dose of As(III) (60µM) showed better growth responses in term of root, shoot length (11.4% and 10.71%, respectively), biomass (11.7%), reduced malonyldialdehyde content (35.14%) and stimulated antioxidant level indicating better As tolerance potential against As. Further, a selenium dependent significant reduction in As accumulation was also observed in root (14.24%) and shoot (23.78%) of rice plant when compared with plant treated with As alone. This study highlights the potential of Se to ameliorate the ecotoxicological risks associated with the As buildup in agricultural land.

Abstract:Copper oxide nanoparticles (CuO NPs) are widely used as fungicides in agriculture. Arsenic (As) is a ubiquitous contaminant in paddy soil. The present study was focused on the adsorption behavior of CuO NPs with regard to As as well as the characteristics of the microbial community changes in As-contaminated soil-rice systems in response to CuO NPs. The study found that CuO NPs could be a temporary sink of As in soil; a high dose of CuO NPs promoted the release of As from crystalline iron oxide, which increased the As content in the liquid phase. The study also found that the As bioavailability changed significantly when the dose of CuO NPs was higher than 50 mg kg-1 in the soil-rice system. The addition of 100 mg kg-1 CuO NPs increased the microbial diversity and the abundance of genes involved in As cycling, decreased the abundance of Fe(III)-reducing bacteria and sulfate-reducing genes, and decreased As accumulation in grains. Treatment with 500 mg kg-1 CuO NPs increased the abundance of Fe(III)-reducing bacteria and sulfate-reducing genes, decreased Fe plaques, and increased As accumulation in rice. The adverse effects of CuO NPs on crops and associated risks need to be considered carefully.

Abstract:Arsenic (As) pollution in rice ( Oryza sativa L.), a staple food for over 3.5 billion people, is a global problem. Mixed effects of Zn, Cu, and Si amendments on plant growth and yield, including in the presence of As pollution have been reported in previous studies. To better investigate the effectiveness of these amendments on rice growth, yield, and As accumulation, we conducted a rice greenhouse experiment with 11 treatments, including control pots with and without As contamination and pots with amendments of ZnO, CuO, and SiO 2 nanoparticles (ZnO NPs, CuO NPs, and SiO 2 NPs), their ionic counterparts (ZnSO 4 , CuSO 4 , and Na 2 SiO 3 ), and bulk particles (ZnO BPs, CuO BPs, and SiO 2 BPs). Compared with the background soil, the treatment of adding As decreased rice plant height, panicle number, and grain yield by 16.5%, 50%, and 85.7%, respectively, but significantly increased the As accumulation in milled rice grains by 3.2 times. Under As contamination, the application of Zn amendments increased rice grain yield by 4.6–7.3 times; among the three Zn amendments, ZnSO 4 performed best by fully recovering grain yield to the background level and significantly reducing grain As III /total As ratio by 46.9%. Under As contamination, the application of Cu amendments increased grain yield by 3.8–5.6 times; all three Cu amendments significantly reduced grain As III /total As ratio by 20.2–65.6%. The results reveal that Zn and Cu amendments could promote rice yield and prevent As accumulation in rice grains under As contamination. Despite the observed reduction in As toxicity by the tested NPs, they do not offer more advantages over their ionic counterparts and bulk particles in promoting rice growth under As contamination. Future field research using a broader range of rice varieties, investigating various As concentrations, and encompassing diverse climate conditions will be necessary to validate our findings in achieving more extensive understanding of effective management of arsenic contaminated rice field.

Abstract:Arsenic (As), one of the major pollutants in the soil, is an important environmental concern as its consumption can cause adverse health symptoms in living organisms. Its contamination of rice grown over As-contaminated areas is a serious concern in South Asian countries. Selenium (Se) has been reported to influence various osmolytes under metal stress in plants. The present study reports the role of Se in mitigating As stress in rice by modulating osmolyte metabolism. Rice plants grown in As-amended soil (2.5–10 mg kg −1 ) in pots were treated with sodium selenate (0.5–1.0 mg Se kg −1 soil) in glass house conditions and leaf samples were collected at 60 and 90 days after sowing (DAS). As-treated rice leaves displayed a reduction in relative water content (RWC) and dry weight than control with a maximum reduction of 1.68- and 2.47-fold in RWC and 1.95- and 1.69-fold in dry weight in As10 treatment at 60 and 90 DAS, respectively. Free amino acids (1.38–2.26-fold), proline (3.88–3.93-fold), glycine betaine (GB) (1.27–1.72-fold), choline (1.67–3.1-fold), total soluble sugars (1.29–1.61-fold), and reducing sugars (1.67–2.19-fold) increased in As-treated rice leaves as compared to control at both stages. As stress increased the γ-aminobutyric acid (GABA), putrescine content, and glutamate decarboxylase activity whereas diamine oxidase and polyamine oxidase activities declined by 1.69–1.88-fold and 1.52–1.86-fold, respectively. Se alone or in combination with As improved plant growth, RWC, GB, choline, putrescine, and sugars; lowered proline and GABA; and showed a reverse trend of enzyme activities related to their metabolism than respective As treatments. As stress resulted in a higher accumulation of osmolytes to combat its stress which was further modulated by the Se application. Hence, the current investigation suggested the role of osmoprotectants in Se-induced amelioration of As toxicity in rice plants.

Abstract:This study investigates how arsenic (As) uptake, accumulation, and migration responds to selenium (Se) foliar application (0–5.0 mg × kg−1). Rice varieties known to accumulate low (DOURADOAGULHA) and high (SINALOAA68) concentrations of arsenic were chosen to grow on soil with different As concentrations (20.1, 65.2, 83.9 mg × kg−1). The results showed that Se of 1.0 mg × L−1 significantly alleviated As stress on upland rice grown on the As-contaminated soil. Under light (65.2 mg × kg−1) and moderate (83.9 mg × kg−1) As concentration treatments, the biomass of upland rice was increased by 23.15% and 36.46% for DOURADOAGULHA, and 46.3% and 54.9% for SINALOAA68. However, the high Se dose (5.0 mg × kg−1) had no significant effect on biomass and heights of upland rice compared to plants where no Se was added. Se significantly decreased As contents in stems and leaves and had different effects on As transfer coefficients for the two rice varieties: when grown on soil with low and moderate As concentrations, Se could reduce the transfer coefficient from stems to leaves, but when grown on the high As soils, this was not the case. The chlorophyll content in plants grown in soil with the moderate concentration of As could be improved by 27.4%–55.3% compared with no Se treatment. Under different As stress, the Se foliar application increased the net photosynthesis, stomatal conductance, and transpiration rate, which meant that Se could enhance the photosynthesis of rice. The intercellular CO2 concentration variation implied that the stomatal or non-stomatal limitations could both occur for different rice varieties under different Se application doses. In conclusion, under moderate As stress, foliar application of Se (1.0 mg × L−1) is recommend to overcome plant damage and As accumulation.

Abstract:Selenium (Se) is crucial for both plants and humans, with plants acting as the main source for human Se intake. In plants, moderate Se enhances growth and increases stress resistance, whereas excessive Se leads to toxicity. The physiological mechanisms by which Se influences rice seedlings' growth are poorly understood and require additional research. In order to study the effects of selenium stress on rice seedlings, plant phenotype analysis, root scanning, metal ion content determination, physiological response index determination, hormone level determination, quantitative PCR (qPCR), and other methods were used. Our findings indicated that sodium selenite had dual effects on rice seedling growth under hydroponic conditions. At low concentrations, Se treatment promotes rice seedling growth by enhancing biomass, root length, and antioxidant capacity. Conversely, high concentrations of sodium selenite impair and damage rice, as evidenced by leaf yellowing, reduced chlorophyll content, decreased biomass, and stunted growth. Elevated Se levels also significantly affect antioxidase activities and the levels of proline, malondialdehyde, metal ions, and various phytohormones and selenium metabolism, ion transport, and antioxidant genes in rice. The adverse effects of high Se concentrations may directly disrupt protein synthesis or indirectly induce oxidative stress by altering the absorption and synthesis of other compounds. This study aims to elucidate the physiological responses of rice to Se toxicity stress and lay the groundwork for the development of Se-enriched rice varieties.

Abstract:Chronic exposure to arsenic (As) in rice has raised many health and environmental problems. As reported, great variation exists among different rice genotypes in As uptake, translocation, and accumulation. Under hydroponic culture, we find that the Chinese wild rice (Oryza rufipogon; acc. 104624) takes up the most arsenic among tested genotypes. Of the cultivated rice, the indica cv. 93-11 has the lowest arsenic translocation factor value but accumulates the maximum concentration of arsenic followed by Nipponbare, Minghui 86, and Zhonghua 11. Higher level of arsenite concentration (50 μM) can induce extensive photosynthesis and root growth inhibition, and cause severe oxidative stress. Interestingly, external silicate (Si) supplementation has significantly increased the net photosynthetic rate, and promoted root elongation, as well as strongly ameliorated the oxidative stress by increasing the activities of antioxidant enzymes superoxide dismutase, ascorbate peroxidase, and peroxidase in roots and/or leaves of 93-11 seedlings. Notably, 1.873 mM concentration of Si considerably decreases the total As uptake and As content in roots, but significantly increases the As translocation from roots to shoots. In contrast, Si supplementation with 1.0 mM concentration significantly increases the total As uptake and As concentrations in roots and shoots of 93-11 seedlings after 50 μM arsenite treatment for 6 days.

Abstract:Arsenic contamination of groundwater is a major concern for its usage in crop irrigation in many regions of the world. Arsenic is absorbed by rice plants mainly from arsenic contaminated water during irrigation. It hampers growth and agricultural productivity. The aim of the study was to mitigate the toxic effects of arsenate (As-V) [25 μM, 50 μM, and 75 μM] by silicon (Si) [2 mM] and selenium (Se) [5 μM] amendments on the activity of the TCA cycle, synthesis of γ-aminobutyric acid (GABA) and polyamines (PAs) in rice (Oryza sativa L. cv. MTU-1010) seedlings and to identify which chemical was more potential to combat this threat. As(V) application decreased the activities of tested respiratory enzymes and increased the levels of organic acids (OAs) in the test seedlings. Application of Si with As(V) and Se with As(V) increased the activities of respiratory enzymes and the levels of OAs. The effects were more pronounced during Si amendments. The activities of GABA synthesizing enzymes along with accumulation of GABA were increased under As(V) stress. During joint application of Si with As(V) and Se with As(V) the activity and the level of said parameters were decreased that indicating defensive role of these chemicals to resist As(V) toxicity in rice and Si amendments showed greater potential to reduce As(V) induced damages in the test seedlings. PAs trigger tolerance mechanism against As(V) in plants. PAs such as putrescine, spermidine and spermine were synthesized more during Si and Se amendments in As(V) contaminated rice seedlings to combat the toxic effects of As(V). Si amendments substantially modulated the toxic effects caused by As(V) over Se amendments in the As(V) challenged test seedlings. Thus, in future application of Si enriched fertilizer will be beneficial to grow rice plants with normal vigor in arsenic contaminated soil.

Abstract:Arsenic (As) is a heavy toxic metalloid found in air, water and soil that adversely affects the plant growth by inducing oxidative stress in plants. Its contamination of rice is a serious problem throughout the world. Selenium (Se) is a beneficial micronutrient for plants that acts as an antioxidant at low doses and protect the plants against number of environmental stresses either by modulating the primary metabolic pathways or regulating the production of phenolic compounds. In the present investigation, effect of Se on different phenolics, enzymes related to their metabolism and antioxidative potential were studied in As stressed rice leaves. Rice plants were grown in pots containing sodium arsenate (2–10 mg As(V) kg −1 soil) and sodium selenate (0.5–1 mg Se kg −1 soil), both alone and in combination and leaf samples were analyzed for various biochemical parameters. Phenolic constituents increased in rice leaves with As(V) treatment from 2 to 5 mg kg −1 soil and leaves exposed to As(V) @ 5 mg kg −1 soil exhibited 1.7, 1.9 and 2.5 fold increase in total phenolics, o-dihydroxyphenols and flavonols, respectively at grain filling stage. Binary application of Se + As improved various phenolic constituents, FRAP, reducing power and antioxidant activities as compared to control. PAL, TAL and PPO activities increased from 1.3 to 4.6 fold in combined As + Se treatment at both the stages. Anthocyanin contents showed a decline (10.8 fold) with increasing As doses and its content improved at both the stages with maximum increase of 3.76 fold with As5+Se1 combination. Binary application of As + Se improved gallic acid, chlorogenic acid, 3-hydroxy benzoic acid and kaempferol contents than control whereas catechin and coumaric acid showed the reverse trend. Application of Se can modulate phenolic constituents in leaf and grains of rice Cv PR126 due to As stress that helped plants to adapt to excess As and resulted in improved plant growth.

Abstract:Selenium (Se) fortification is witnessed to simultaneously inhibit absorbing Cadmium (Cd) and Arsenic (As) by rice plants, but the mechanism is unclear. Here, the effects of Se on the root morphology, iron plaque (IP) content, soil Fe2+ content, radial oxygen loss (ROL), and enzyme activities of the rice plants in the soil contaminated by Cd and As were intensively investigated through the hydroponic and soil experiments. Se effectively alleviated the toxic effects of Cd and As on the plants and the dry weight, root length, and root width were increased by 203.18%, 33.41%, and 52.81%, respectively. It also elucidated that ROL was one of the key factors to elevate IP formation by Se and the specific pathways of Se enhancing ROL were identified. ROL of the plants in the experiment group treated by Se was increased 36.76%, and correspondingly IP was magnified 50.37%, compared to the groups with Cd and As. It was owing to Se significantly increased the root porosity (62.11%), facilitating O2 transport to the roots. Additionally, Se enhanced the activities of catalase (CAT) and superoxide dismutase (SOD) to promote the catalytic degradation of ROS induced by Cd and As stress. It indirectly increased O2 release in the rhizosphere, which benefit to form more robust IP serve as stronger barrier to Cd and As. The results of our study provide a novel molecular level insight for Se promoting root IP to block Cd and As uptake by the rice plants.
