Abstract:The synthesis of a new kind of amphiphilic poly(vinyl-co-ester)s via simple one-pot radical ring-opening polymerization (RROP) is reported. A series of biodegradable, biocompatible, and cross-linkable copolymers P(MDO-co-PEGMA-co-CMA) with different compositions were synthesized by copolymerization of 2-methylene-1,3-dioxepane (MDO), poly(ethylene glycol) methyl ether methacrylate (PEGMA) and 7-(2-methacryloyloxyethoxy)-4-methylcoumarin methacrylate (CMA). All the copolymers were well characterized by H-1 NMR, C-13 NMR, and gel permeation chromatography (GPC). The hydrolytic degradation and enzymatic degradation of the copolymers were investigated. The amphiphilic P(MDO-co-PEGMA-co-CMA) can self-assemble into micelles in aqueous solution. Polymeric micelles with different sizes were obtained through tuning the compositions of the copolymers, i.e., the hydrophilic/hydrophobic ratio. The size and morphology of the micelles were measured by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Because of the coumarin side groups in the micellar core, the micelles can be cross-linked upon exposure to long-wavelength UV light (lambda = 365 nm). These micelles also showed excellent biocompatibility by the in vitro cytotoxicity experiments. Furthermore, the biodegradable micelles were applied for the delivery of an anticancer drug doxorubicin (DOX). The cell viability of DOX-loading micelles compared to free DOX was discussed. The effects of cross-linking and the copolymer composition on the drug release behavior were well studied. The results indicate that this new kind of amphiphilic poly(vinyl-co-ester) s could serve as promising nanocarriers for the delivery of anticancer drugs.

Abstract:Two asymmetrical, amphiphilic perylene derivatives, N -Decyl-perylene-3,4:9,10-tetracaboxylic-3,4-di(methoxyethoxyethyl)ester-9,10-imide ( D1E2 ) and N -(1-Decylundecyl)-perylene-3,4:9,10-tetracaboxylic-3,4-di(methoxyethoxyethyl)ester-9,10-imide ( D2E2 ), have been synthesized and characterized. These compounds contain one long hydrophobic chain on one end and two hydrophilic ethoxy chains on the other end. Self-assembly of these molecules in a variety of solvents has been demonstrated. Scanning electron microscopy images showed that these compounds self-assembled to various nanostructures in different solvents. The most well-defined structure was flexible nanoribbons obtained from D1E2 precipitation in methanol. The UV–vis absorption and fluorescence spectra of these compounds in solution and solid form are also reported. The self-assembled nanostructures have potential applications in optoelectronics.

Abstract:The crystallization-driven self-assembly of polymers based on semicrystalline poly(epsilon-caprolactone) cores is currently an area of high interest on account of their well-known biocompatibility and biodegradability, yet a comprehensive understanding of coil-crystalline-coil type triblock copolymer assembly behavior with respect to this core chemistry is yet to be realized. Herein, we demonstrate the simple preparation of well-defined tuneable 1D and 2D structures based on poly(epsilon-caprolactone) (PCL) triblock copolymers of different block ratios synthesized by ring-opening polymerization (ROP) and reversible addition-fragmentation chain transfer (RAFT) polymerization. In this report, the assembly of PCL-based amphiphiles in various solvents was investigated to tune the morphology and size of the assemblies with well-defined 2D platelets and long cylinders produced when using long soluble coronal blocks or under good solvent conditions. By contrast, truncated short fibers were obtained for less soluble PCL-containing block copolymers or under poor solubility conditions for the core block as a consequence of the increasing amount of nuclei formed in the crystallization process. Furthermore, the length of PCL-based 1D nanostructures could be controlled by tuning self-assembly conditions where the micelles' lengths varied from 93 to 1200 nm with narrow dispersities. This easy assembly methodology greatly simplifies the lengthy procedure required to prepare biodegradable 1D and 2D nanostructures from PCL with tuneable sizes, which demonstrate great potential as drug-delivery vehicles in the realm of biomedicine.
Flower- and Grass-like Self-Assemblies of an Oleanane-Type Triterpenoid Erythrodiol: Application in the Removal of Toxic Dye from Water

Abstract:Erythrodiol (3β-olean-12-ene-3, 28-diol) (C30H50O2) 1 is a nanosized oleanane-type fused 6-6-6-6-6 pentacyclic triterpeneoid extractable from the dried leaves of olive (Olea europia). One step reduction of oleanolic acid extracted from Lantana camara also yields the same compound. The triterpenoid has one secondary −OH group attached at C3 of the "A" ring and one primary −OH group at C28 present at the junction of the "D" and "E" rings. Here, we report the spontaneous self-assembly of erythrodiol in different neat organic liquids and aqueous-organic liquid mixtures. The nanosized dihydroxy triterpenoid having an oleanane-type lipophilic rigid skeleton self-assembled in liquids, yielding nanosized fibrils, microsized flowers, and grass-like architectures via formation of densely assembled fibrils and petals or 2D sheets. The microstructures of the self-assemblies have been characterized by different techniques like optical microscopy, electron microscopy, atomic force microscopy, FTIR, and wide angle X-ray diffraction studies. The porous self-assemblies having a large surface area obtained from 1 were capable of adsorbing toxic fluorophores like rhodamine-B, rhodamine-6G, methylene blue, and crystal violet (CV). Moreover, removal of the aforementioned toxic pigments has also been demonstrated from their aqueous solutions by using UV–visible spectrophotometry and epifluorescence microscopy.This article has not yet been cited by other publications.

Self-assembly of naturally occurring stigmasterol in liquids yielding a fibrillar network and gel

Abstract:Stigmasterol, a naturally occurring 6-6-6-5 monohydroxy phytosterol, was extracted from the leaves of Indian medicinal plant Roscoea purpurea, commonly known as Kakoli. In continuation of our studies on the self-assembly properties of naturally occurring terpenoids, herein, we report the first self-assembly properties of this phytosterol in different organic liquids. The molecule self-assembled in organic liquids yielding supramolecular gels in most of the liquids studied via the formation of fibers and belt-like architechtures of nano-to micrometer diameter. Characterization of the self-assemblies carried out by using scanning electron microscopy, transmission electron microscopy, atomic force microscopy, optical microscopy, FTIR and X-ray diffraction studies indicated fibrillar network and belt-like structures. A model for the self-assembly of stigmasterol has been proposed based on molecular modeling studies, X-ray diffraction data and FTIR studies. Rheology studies indicated that the gels were of high mechanical strength. Fluorophores such as rhodamine B, carboxy fluorescein including the anticancer drug doxorubicin could be loaded in the gels. Moreover, release of the loaded fluorophores including the drug has also been demonstrated from the gel phase into aqueous medium.

Abstract:A series of P(DL)LA-PEG(1k)-P(DL)LA tri-block co-polymers with various compositions, i.e., containing 2-10 lactoyl units, were prepared via ring opening polymerisation of d,l-lactide in the presence of poly (ethylene glycol) (PEG) (M-n = 1000 gmol(-1)) as the initiator and stannous 2-ethylhexanoate as the catalyst at different feed ratios. P(DL)LA-PEG(1k)-P(DL)LA co-polymers were then functionalised with acrylate groups using acryloyl chloride under various reaction conditions. The diacrylated P(DL)LA-PEG(1k)-P(DL)LA (diacryl-P(DL)LA-PEG(1k)-P(DL)LA) were further polymerised to synthesize soluble hyperbranched polymers by either homo-polymerisation or co-polymerisation with poly(ethylene glycol) methyl ether methylacrylate (PEGMEMA) via free radical polymerisation. The polymer samples obtained were characterised by H-1 NMR (proton Nuclear Magnetic Resonance), FTIR (Fourier Transform Infra-red spectroscopy), and GPC (Gel Permeation Chromatography). Moreover, the diacryl-P(DL)LA-PEG(1k)-P(DL)LA macromers were used for the preparation of biodegradable crosslinked hydrogels through the Michael addition reaction and radical photo-polymerisation with or without poly(ethylene glycol) methyl ether methylacrylate (PEGMEMA, M-n = 475 gmol(-1)) as the co-monomer. It was found that fine tuning of the diacryl-P(DL)LA-PEG(1k)-P(DL)LA constituents and its combination with co-monomers resulted in hydrogels with tailored swelling properties. It is envisioned that soluble hyperbranched polymers and crosslinked hydrogels prepared from diacryl-P(DL)LA-PEG(1k)-P(DL)LA macromers can have promising applications in the fields of nano-medicines and regenerative medicines.

Abstract:Poly(1,3-dioxolane) (PDO) is widely used as a pH- and temperature-sensitive water-soluble polymer applied in functional hydrogels and nanoparticles. In this contribution, we report a series of highly efficient rare-earth triflate catalysts for a Bronsted-acid-free ring-opening polymerization of DO. The repeat unit of PDO comprises perfectly alternating hydrophilic ethylene glycol and hydrophobic methyl aldehyde units leading to hydrogen-bonding-controlled self-assembly in aqueous solution. Spherical aggregates are observed in transmission electron microscopy images whose sizes can be tuned by concentration and temperature, further confirming that the hydrate is derived from "blocked" water molecules through hydrogen bonding. The size and morphology of the aggregates can be modulated not only by the annealing process owing to the realignment of semicrystalline regions but also by metal salts generating rodlike, wormlike, or spherical structures. In this case, PDO is able to act as both a surfactant and as a reductant on account of the acidolysis of acetal moieties, which form a core-shell Cu-decorated hybrid material in situ in one step, thus providing a promising future template for the generation of metal nanoparticles.

Abstract:Erucic acid, a nano-sized long-chain ω-9-cis-mono-unsaturated 22C fatty acid, is isolable from various plants, especially from the seeds of Brassicaceae (e.g., rape seed or mustard seed). Isolation of erucic acid has recently been reported by us from the pseudobulb of Crepidium acuminatum. Herein, we report the first self-assembly of erucic acid in different aqueous binary liquids yielding vesicles. Several microscopic techniques such as scanning electron microscopy, high-resolution transmission electron microscopy, atomic force microscopy, optical microscopy, and X-ray diffraction studies were carried out to characterize the vesicular morphology. Encapsulation of various fluorophores as guests inside the self-assemblies of erucic acid was also demonstrated. The examples of the guests include cationic fluorophores rhodamine B, anionic fluorophore carboxyfluorescein, and cationic fluorescence active anticancer drug doxorubicin. Release of the entrapped fluorophores was demonstrated by treating with a vesicle-rupturing agent Triton X-100. On treatment with a composite of self-assembled erucic acid and curcumin, antibacterial activity was observed for both Gram-positive as well as Gram-negative bacteria. Therefore the vesicular self-assemblies generated from the renewable nano-sized erucic acid are useful for drug delivery applications as well as antibacterial activities with poorly water-soluble drugs.

Abstract:Self‐assembled hydrogels, fabricated through diverse non‐covalent interactions, have been extensively studied in regenerative medicines. Inspired from bioactive functional motifs of ECM protein, short peptide sequences have shown remarkable abilities to replicate the intrinsic features of the natural extracellular milieu. In this direction, we have fabricated two short hydrophobic bioactive sequences derived from the laminin protein i.e., IKVAV and YIGSR. Based on the substantial hydrophobicity of these peptides, we selected a co‐solvent approach as a suitable gelation technique that included different concentrations of DMSO as an organic phase along with an aqueous solution containing 0.1% TFA. These hydrophobic laminin‐based bioactive peptides which had limited solubility in aqueous physiological environment showed significantly enhanced solubility with higher DMSO content in water. The enhanced solubility resulted in extensive intermolecular interactions that led to the formation of hydrogels with a higher‐order entangled network along with improved mechanical properties. Interestingly, by simply modulating DMSO content, highly tunable gels were accessed in the same gelator domain that displayed differential physicochemical properties. Further, the cellular studies substantiated the potential of these laminin‐derived hydrogels in enhancing cell‐matrix interactions, thereby reinforcing their applications in tissue engineering.

Abstract:Formation of nanofibers and nanovesicles in the self-assembled state of small amphiphilic molecules has applications in versatile fields such as tissue engineering, controlled delivery of drug molecules, etc. This paper demonstrated the self-aggregation behavior of three synthesized 6-acylamino nicotinic acid amphiphiles named 6-octanoylamino-nicotinic acid (OANA), 6-decanoylamino-nicotinic acid (DANA) and 6-dodecanoylamino-nicotinic acid (DDANA) in water and basic aqueous solution. The result showed that the amphiphiles successfully self-organize into vesicles and twisted ribbons in water. FT-IR study revealed existence of mixtures of handedness in the fibrous structures. CD spectroscopy and TEM study elucidated formation of chiral structures through aggregation. Results showed that DDANA forms thermoreversible hydrogel in aqueous solutions of NaOH and Na2CO3, whereas other two amphiphiles form hydrogel only in the presence of NaOH. Morphological investigation revealed that the hydrogel is formed due to self-assembly of fibrils of micron length. The elastic fibrillar networks are quite stable to external forces. Existence of bilayer columnar square packing arrangement in the self-assembled state was recognized by XRD measurement. Spectroscopy and theoretical studies established that hydrogen bonding interactions are responsible to self-assemble the amphiphilic molecules. The amphiphiles are efficient phase selective gelators of mineral oils in water–mineral oil mixtures and excellent remover of rhodamine 6G, eosin Y and rose bengal from water. The amphiphiles successfully create reproducible fiber mat applicable in tissue engineering field.

Abstract:Generally, an esterase lipase enzyme can hydrolyze specific substrates called esters in an aqueous solution. Herein, we investigate how a G‐quadruplex self‐assembly affects the hydrolysis equilibrium in reverse. The biocatalyst, lipase, activates the individual building‐blocks through fuel consumption, causing them to undergo a higher degree of self‐organization into nanofibers within spheres. We have synthesized five peptide‐lipid‐conjugated guanine base functionalized molecules to explore how the equilibrium can be shifted through reverse hydrolysis. Among these, NAC5 self‐assembled into a G‐quadruplex structure which has been confirmed by various spectroscopic techniques. The wide‐angle powder XRD, ThT dye binding assay and circular dichroism study is carried out to support the presence of the G‐quadruplex structure. The biocatalytic formation of nanofibers enclosed spheres is analyzed using CLSM, FE‐SEM and HR‐TEM experiments. Additionally, we assess the biocompatibility of the enzyme fueled dissipative self‐assembled fibers enclosed spheres, as they have potential applications as a biomaterial in protocells. MTT assay is performed to check the cytotoxicity of G‐quadruplex hydrogel, using HEK 293 and McCoy cell lines for viability assessment. Finally, the utility of the novel NAC5 hydrogel as a wound repairing biomaterial is demonstrated by cell migration experiment in a scratch assay.

Abstract:Although a few architectures have been fabricated by the self-assembly of natural triterpenoids, the precise control of shape and size is rarely studied. Herein, a methyl oleanolate-bearing amphiphile, 1-[2-(methyl oleanolate)-2-oxoethyl]pyridinium bromide (MOP), has been designed and its assembly behavior was investigated. It was found that the morphologies of MOP assemblies ranged from nanoparticles to rigid microrods and flexible nanofibers in chloroform/p-xylene and methanol/water, respectively. During the assembly process, the systematical variational solvophobic/solvophilic effect resulted in the formation of spherical nanoparticles with opposite dipoles and converse bilayer structures. Moreover, such opposite molecular orientations lead to the inversion of supramolecular chirality and distinct mechanical properties. The driving forces and packing patterns of MOP in each solvent system were clearly demonstrated by the combination of NMR, UV-vis, Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), theoretical computation, and contact angle experiments, which revealed the roles of triterpenoids and pyridinium cations in the assembly process. This work provides a facile strategy to control the supramolecular structures in triterpenoid-based assemblies by adjusting the solvent polarity and composition.

Abstract:Using supramolecular self-assembled nanocomposite materials made from protein and polysaccharide components is becoming more popular because of their unique properties, such as biodegradability, hierarchical structures, and tunable multifunctionality. However, the fabrication of these materials in a reproducible way remains a challenge. This study presents a new evaporation-induced self-assembly method producing layered hydrogel membranes (LHMs) using tropocollagen grafted by partially deacetylated chitin nanocrystals (CO-g-ChNCs). ChNCs help stabilize tropocollagen's helical conformation and fibrillar structure by forming a hierarchical microstructure through chemical and physical interactions. The LHMs show improved mechanical properties, cytocompatibility, and the ability to control drug release using octenidine dihydrochloride (OCT) as a drug model. Because of the high synergetic performance between CO and ChNCs, the modulus, strength, and toughness increased significantly compared to native CO. The biocompatibility of LHM was tested using the normal human dermal fibroblast (NHDF) and the human osteosarcoma cell line (Saos-2). Cytocompatibility and cell adhesion improved with the introduction of ChNCs. The extracted ChNCs are used as a reinforcing nanofiller to enhance the performance properties of tropocollagen hydrogel membranes and provide new insights into the design of novel LHMs that could be used for various medical applications, such as control of drug release in the skin and bone tissue regeneration.

Abstract:In this work, a series of amphiphilic hemicellulose-based grafting polymers are synthesized by homogeneous esterification with various hydrophobic chain fatty acids. With the flexible chain conformation of hemicelluloses, the hemicellulose-graft-fatty acid is self-assembled into hard sphere micelles in aqueous solution through hydrophobic interactions. The resultant micelles show apparent hydrodynamic radius (Rh) varying in the range of 34–57 nm and radius of gyration (Rg) varying from 30 to 44 nm. Moreover, the Rh, Rg, zeta potential and critical micelle concentration (CMC) of the micelles gradually decreases with the increasing hydrophobic chain length. The aggregation number of micelles can be tuned by varying alkyl chain lengths. Taking curcumin (Cur) as a model drug, the hemicellulose micelles were investigated as drug carriers, which exhibit a chain length-controlled drug release behavior. Taken together, structure and property tunable hemicellulose-based micelles were obtained, and showed potential of application in pharmaceutical, food, and cosmetics fields.

Abstract:C 3 -symmetric star-shaped aromatic compounds are known to possess unique characteristics which facilitate their industrial and biomedical applications. Herein, we report the design, synthesis, self-assembly and drug/dye delivery capabilities of C 3 -symmetric, hexa-substituted benzene-based amphiphiles. The synthesis of the hexa-substituted C 3 -symmetric core involves C -acetylation of phloroglucinol to yield the corresponding tri-acetyl derivative. This was further subjected to O -propargylation, followed by the carbonyl reduction of acetyl groups to yield the central core. Various hydrophilic (mPEG) and lipophilic units were then incorporated into this core via click and esterification reactions, respectively, to produce a new type of star shaped amphiphiles. So the obtained amphiphilic architectures have a tendency to aggregate in an aqueous medium forming nanosized assemblies with an inner hydrophobic core, allowing the substituents to control the tension-active properties. The critical aggregation concentration of the amphiphiles was evaluated by fluorescence measurement using the dye Nile red as a fluorescent probe. The hydrodynamic diameter of self-assembled aggregates in aqueous solution was studied by dynamic light scattering, while the actual size and morphology were determined by cryo-transmission electron microscopy (cryo-TEM) analysis. The physicochemical properties of the amphiphiles suggested their suitability for exploring their drug delivery applications. In this endeavor, the amphiphiles were utilized for the encapsulation of model hydrophobic entities and studying their subsequent release from their hydrophobic core in a controlled manner. The transport potential of the synthesised amphiphiles was explored for transdermal drug delivery. Furthermore, cytotoxicity studies were conducted using MCF7 and HeLa cells, which indicated that the nanocarriers had no toxic effect on the cells.

Abstract:Lignin, the most abundant source of renewable aromatic compounds on the planet, is attracting more scholarly attention due to its possibility of replacing petroleum-based chemicals and products. However, it remains underutilized because of the heterogeneity of its multi-level structure that prevents homogenization and standardization of derived products. The key to solving these problems lies in finding a general preparation method to achieve the integrated utilization of lignin molecules at all levels. The assembly-mediated granulation methods provide a significant means for the integrated value-added utilization of lignin, and for biomass productization applications, it is significant to understand the molecular mechanisms of lignin nano-colloids (LNCs) formation thus accurately guiding their functionalization. Therefore, a thorough understanding of the assembly morphology and behavior of lignin in different solutions towards colloids is of great scientific importance. In this minireview, we focus on the assembly behavior of lignin in different solvents, specifically in mono-solvent and multi-solvent, and in particular, we review various methods for preparing lignin composite colloids and concentrate on the applications in therapy, cosmetics and emulsification, which are important for guiding the preparation and efficient utilization of LNCs.

Abstract:Oil-filled microcapsules of kraft lignin were synthesized by first creating an oil in water emulsion followed by a high-intensity, ultrasound-assisted cross-linking of lignin at the water/oil interface. The rationale behind our approach is based on promoting documented lignin hydrophobic interactions within the oil phase, followed by locking the resulting spherical microsystems by covalent cross-linking using a high intensity ultrasound treatment. As further evidence in support of our rationale, confocal and optical microscopies demonstrated the uniformly spherical morphology of the created lignin microparticles. The detailed elucidation of the cross-linking processes was carried out using gel permeation chromatography (GPC) and quantitative (31)P NMR analyses. The ability of lignin microcapsules to incorporate and release Coumarin-6 was evaluated in detail. In vitro studies and confocal laser scanning microscopy analysis were carried out to assess the internalization of capsules into Chinese hamster ovary (CHO) cells. This part of our work demonstrated that the lignin microcapsules are not cytotoxic and readily incorporated in the CHO cells.

Abstract:A novel series of anionic fluorine-containing amphiphilic graft copolymers, poly(hexafluorobutyl methacrylate-co-sodium 4-vinylbenzenesulfonate)-g-poly(ethylene glycol) methyl ether methacrylate [poly(HFMA-co-NaSS)-g-PEG], were prepared via the free radical copolymerization. The micellization behavior of the copolymers was studied by critical micelle concentration (CMC), dynamic light scattering (DLS) and transmission electron microscopy (TEM). The results showed that poly(HFMA-co-NaSS)-g-PEG copolymers could self-assemble into core/shell structure micelles in aqueous solution. Afterward, the micelles stability was studied by zeta potential measurement, which demonstrated that the copolymer micelles had good stability in biological milieu. The interaction between the copolymer micelles and BSA was characterized by fluorescence spectroscopy, and the morphology of the micelles/BSA complexes was observed by TEM. The results demonstrated that the adsorption could be easily achieved between the copolymer micelles and BSA. Furthermore, the cytotoxicity was evaluated via MTT viability assay, indicating that the copolymer had low toxicity. Our study suggests that poly(HFMA-co-NaSS)-g-PEG micelles have a potential application as the protein drug carrier.

Abstract:A convenient and simple route to multifunctional surface coatings via the alternating covalent layer-by-layer (LBL) assembly of p-nitrophenyloxycarbonyl group-terminated hyperbranched polyether (HBPO-NO2) and polyethylenimine (PEI) is described. The in situ chemical reaction between HBPO-NO2 and PEI onto aminolyzed substrates was rapid and mild. Results from ellipsometry measurements, contact angle measurements, and ATR-FTIR spectra confirmed the successful LBL assembly of the building blocks, and the surface reactivity of the multilayer films with HBPO-NO2 as the outmost. layer. was, demonstrated by the Immobilization of an amine-functionalized fluorophore. Furthermore, a biomimetic surface was achieved by surface functionalization of the multilayer films with extracellular matrix protein collagen to promote the adhesion and growth of cells. The studies on the drug loading and in vitro release behaviors of the multilayer films demonstrated their application potentials in local delivery of hydrophilic and hydrophobic therapeutic agents.
