Abstract:Collagen-chitosan composite film modified with grapheme oxide (GO) and 1-(3-Dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDC), termed CC-G-E film, was loaded with basic fibroblast growth factor (bFGF) as the development of an efficacious wound healing device. In this study we report a novel drug delivery system that prevents the initial burst release and loss of bioactivity of drugs in vitro and in vivo applications. The results showed that CC-G-E film possessed improved thermal stability and a higher rate of crosslinking with increased mechanical properties when the dosage of GO was between 0.03% and 0.07%. It was shown that the in vitro release of bFGF from CC-G-E film continued for more than 28d. Furthermore, the CC-G-E films demonstrated excellent in vitro biocompatibility following culture with L929 fibroblasts in terms of cell adhesion and proliferation. CC-G-E films were implanted into Sprague-Dawley rats to characterize their ability to repair full-thickness skin wounds. Results showed that the CC-G-E film accelerated the wound healing process compared with the blank control. Based on all the results, it was concluded that CC-G-E film operates as a novel drug delivery system and due to its performance in wound remodeling, has potential to be developed as a wound dressing material.
Preparation and Evaluation of Chitosan/PVA Based Hydrogel Films Loaded with Honey for Wound Healing Application

Abstract:In the present study, chitosan/polyvinyl alcohol (PVA)-based honey hydrogel films were developed for potential wound healing application. The hydrogel films were developed by a solvent-casting method and were evaluated in terms of thickness, weight variation, folding endurance, moisture content and moisture uptake. The water vapor transmission rate was found to range between 1650.50 ± 35.86 and 2698.65 ± 76.29 g/m2/day. The tensile strength and elongation at break were found to range between 4.74 ± 0.83 and 38.36 ± 5.39 N, and 30.58 ± 3.64 and 33.51 ± 2.47 mm, respectively, indicating significant mechanical properties of the films. SEM images indicated smooth surface morphology of the films. FTIR, DSC and in silico analysis were performed, which highlighted the docking energies of the protein–ligand complex and binding interactions such as hydrogen bonding, Pi–Pi bonding, and Pi–H bonding between the selected compounds and target proteins; hence, we concluded, with the three best molecules (lumichrome, galagin and chitosan), that there was wound healing potential. In vitro studies pointed toward a sustained release of honey from the films. The antimicrobial performance of the films was investigated against Staphylococcus aureus. Overall, the results signaled the potential application of chitosan/PVA based hydrogel films as wound dressings. Furthermore, in vivo experiments may be required to evaluate the clinical efficacy of honey-loaded chitosan/PVA hydrogel films in wound healing.

Abstract:Human epidermal growth factor (hEGF) has been known to have excellent wound-healing activity. However, direct application to the wound area can lead to low hEGF bioavailability due to protease enzymes or endocytosis. The use of liposomes as coatings and carriers can protect hEGF from degradation by enzymes, chemical reactions, and immune reactions. Sustained release using a matrix polymer can also keep the levels of hEGF in line with the treatment. Therefore, this study aimed to develop a film-forming spray of water-soluble chitosan (FFSWSC) containing hEGF-liposomes as a potential wound dressing. The hEGF-liposomes were prepared using the hydration film method, and the preparation of the FFSWSC was achieved by the ionic gelation method. The hydration film method produced hEGF-liposomes that were round and spread with a Z-average of 219.3 nm and encapsulation efficiency of 99.87%, whereas the film-forming solution, which provided good sprayability, had a formula containing 2% WSC and 3% propylene glycol with a viscosity, spray angle, droplet size, spray weight, and occlusion factor of 21.94 ± 0.05 mPa.s, 73.03 ± 1.28°, 54.25 ± 13.33 µm, 0.14 ± 0.00 g, and 14.57 ± 3.41%, respectively. The pH, viscosity, and particle size of the FFSWSC containing hEGF-liposomes were stable during storage for a month in a climatic chamber (40 ± 2 °C, RH 75 ± 5%). A wound healing activity test on mice revealed that hEGF-liposomes in FFSWSC accelerated wound closure significantly, with a complete wound closure on day 6. Based on the findings, we concluded that FFSWSC containing hEGF-liposomes has the potential to be used as a wound dressing.
Lupeol-loaded chitosan-Ag + nanoparticle/sericin hydrogel accelerates wound healing and effectively inhibits bacterial infection

Abstract:Lupeol, a pentacyclic triterpene, has demonstrated significant wound healing properties; however, its low water solubility has limited its clinical applicability. To overcome this limitation, we utilized Ag + -modified chitosan (CS-Ag) nanoparticles to deliver lupeol, resulting in the formation of CS-Ag-L-NPs. These nanoparticles were then encapsulated within a temperature-sensitive, self-assembled sericin hydrogel. Various analytical methods, including SEM, FTIR, XRD, HPLC, TGA assay, hemolysis and antibacterial activity tests, were employed to characterize the nanoparticles. Additionally, an infectious wound model was used to evaluate the therapeutic and antibacterial efficacy of the CS-Ag-L-NPs modified sericin hydrogel. Our results showed that the encapsulation efficiency of lupeol in CS-Ag-L-NPs reached 62.1 %, with good antibacterial activity against both gram-positive and gram-negative bacteria and a low hemolysis ratio (<5 %). The CS-Ag-L-NPs sericin gel exhibited multiple beneficial effects, including inhibiting bacterial proliferation in wound beds, promoting wound healing via accelerated re-epithelialization, reducing inflammation, and enhancing collagen fiber deposition. We conclude that the CS-Ag-L-NPs loaded sericin hydrogel has tremendous potential for development as a multifunctional therapeutic platform capable of accelerating wound healing and effectively suppressing bacterial infections in clinical settings.

Influence of gelatin on the functional characteristics and wound healing potential of chitosan/zein films loaded with ellagic acid nanoparticles

Abstract:Biopolymers based-films are an alternative to conventional bandages as the occlusion and superficial properties, important for wound healing, are supplemented with the controlled release of active molecules. Chitosan/zein/gelatin films loaded with ellagic acid (EA) nanoparticles were designed as an antimicrobial bandage platform for wound treatment. The films were characterized in terms of morphology, thickness, sorption, solubility, contact angle and water vapor permeability. The phenyl rings of EA were found to interact transiently with the polymers, what could explain the resulting controlled release. All films containing gelatin showed a burst-effect in the first hour (>29%), followed by a sustained release along the next 24 h (>78%), releasing the residual content after the next 48–168 h, according to the formulation. The release curves adjusted to the first-order model. The EA-loaded films reduced the viability of Staphylococcus aureus and Candida albicans in 2 and 3 logs, respectively and were also found to inhibit Pseudomonas aeruginosa. They presented a tensile strength between 3.6 and 10.8 MPa and an elongation at break between 18.52 and 46.34% showing good elasticity and smooth surfaces, supporting their potential use as a skin layer device. In view of the most favorable results, the film F4, with equivalent proportions of CHI (27.8%) and gelatin (24.9%), was chosen for wound healing evaluation in vivo. This film accelerated the wound contraction, reduced the number of inflammatory cells in the injured tissue and did not cause any deformation in the healing process. Accordingly, this film was found a suitable dressing device and present great potential to be used as a bandaging material in the treatment of wounds.

Abstract:A rise in the incidence of skin burns and chronic wounds present a first health care problem. Hence collagen – chitosan-based wound dressing has been seen as a way to heal wounds without leaving a scar. Collagen is the most ubiquitous protein found in marine organisms. Here, chitosan plays a vital role in improving the stability of the collagen. The Collagen-chitosan films comprised of collagen derived from skins of Catla catla which were pretreated and lyophilized and confirmed by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Thus, in this new approach collagen has been hybridized with the naturally occurring bioactive material chitosan to improve the mechanical properties with enhanced stability, and it can be effectively utilized for the wound dressing in clinical application. Physico-chemical properties and surface morphology of the films were analyzed through SEM and FTIR. Mechanical stability of collagen – chitosan film showed increased tensile strength and elongation at break. The Anti-bacterial and Anti-inflammatory activity of collagen - chitosan films were performed. Also, the in vitro biocompatibility of collagen chitosan films was confirmed by wound scratch assay on fibroblast cells. It was concluded that this present study reveals that the preparation of collagen-chitosan films from a new source of natural collagen could be suitable for the application of wound dressings.

Abstract:Crystallization of phytochromes and other photochromic proteins is hampered by the conformational changes that they undergo on exposure to light. As a canonical phytochrome, cyanobacterial Cph1 switches between two stable states upon absorption of red/far-red light. Consequently, it is mandatory to work in darkness from protein purification to crystal cryoprotection in order to ensure complete occupancy of one state or the other. With the simple and inexpensive methods that have been developed, phytochromes and other photochromic molecules can effectively be handled and crystallized, as has been demonstrated by the solution of the three-dimensional structure of the Cph1 sensory module.
A sustained‐release gel film based on chitosan for treating cutaneous candidiasis

In this study, a chitosan‐citric acid gel film was prepared using chitosan as the raw material and citric acid as the acid solvent and cross‐linking agent. In addition, to increase the antibacterial effect of the gel film, ketoconazole/β‐cyclodextrin inclusion complex was prepared by inclusion technology and added to the gel film to prepare a chitosan‐citric acid sustained‐release gel film loaded with ketoconazole/β‐cyclodextrin inclusion complex for treating cutaneous candidiasis. The physical and chemical properties of the chitosan‐citric acid gel films were investigated using infrared spectroscopy, x‐ray diffraction, water vapor transmittance test, thermal analysis, and swelling tests. Drug release, antibacterial, and toxicity tests of the chitosan‐citric acid gel film loading with ketoconazole/β‐cyclodextrin inclusion complex were carried out in vitro. Abstract:In recent decades, polysaccharide-based hydrogels have gained significant attention due to their natural biocompatibility, biodegradability, and non-toxicity. The potential for using polysaccharides to synthesize hydrogels is due to their ability to support cell proliferation, which is important for practical applications, particularly in the biomedical field. In this study, we have synthesized a chitosan-α-naphthal hydrogel film using a cost-effective one-step synthesis approach. The prepared hydrogel film exhibited high encapsulation efficiency for antibacterial drugs such as ciprofloxacin and lomefloxacin, with the ability to release the antibiotics in a controlled manner over an extended period and prevent long-term bacterial infections. Moreover, the Korsmeyer and Peppas power law, based on Fickian diffusion, was employed to model the entire complex drug release process and predict the drug release behavior. The hydrogel film also shows pH-induced swelling ability due to the presence of an imine bond in the hydrogel network, which is degradable at acidic pH. The incorporated therapeutic agents having antibacterial activity were effective against Gram-negative (Escherichia coli DH5α) and Gram-positive (Staphylococcus aureus subsp. aureus) bacterial strains. A wound dressing material should possess mechanical strength, but the prepared hydrogel film has low mechanical strength. To increase the mechanical strength, we have infused pineapple leaf fibers (PLFs) in the film network, resulting in a mechanical strength of 1.12 ± 0.89 MPa. In addition to its mechanical strength, significant cell viability against human embryonic kidney (HEK-293) cells was observed from in vitro cell culture experiments for this PLF-hydrogel film. As a result, the prepared therapeutic agent-loaded hydrogel film under study meets the requirements to be considered for use as a wound dressing material.

Abstract:Recent years have demonstrated that biopolymers can serve as effective woundhealing agents. We hereby report chitosan-oxidized cellulose (CS.OMCC) based hydrogels loaded with eugenol with distinctive antimicrobial, biocompatible, and angiogenic characteristics. CS.OMCC hydrogels in four different ratios were synthesized through Schiff base (-CN) between chitosan (CS) and oxidized microcrystalline cellulose (OMCC). The swelling and degradation study analyzed the maximum absorption strength and weight loss of 1165% and 56.8 %, respectively. Hydrogels were potent against E. coli and S. aureus . The zones of inhibition were dependent upon the chitosan and eugenol, where 30 % eugenol (Eu) loaded CS.OMCC 4:1 hydrogel revealed greater zones (17 mm). The hydrogels demonstrated favorable biocompatibility, adhesion, and migration potential of mouse fibroblast cells. Moreover, the in-vivo potential by Chorioallantoic membrane (CAM) assay displayed an increase in the number, length, and size of blood vessels, contributing superior angiogenic potential. Increased expression in the case of VEGF further supported the role of hydrogels in promoting angiogenesis. Our study presents eugenol loaded chitosan-oxidized cellulose hydrogels that presented a promising solution to target bacterial infection and promote angiogenesis, thereby promoting the wound healing process.

Abstract:Bacterial infections, excessive reactive oxygen species (ROS) accumulation and a persistent inflammatory response severely impede the wound healing process. In this study, we developed a novel multifunctional hydrogel dressing (LCPN) based on lipoic acid modified chitosan (LAMC), polypyrrole nanoparticles (PPy NPs) and nicotinamide mononucleotide (NMN) for accelerated healing of infected wounds. The synthesized LCPN hydrogel has several properties. Modification of lipoic acid significantly enhances the water solubility of chitosan making it easier to dissolve and absorb wound secretions. Interestingly, owing to the breaking and restructuring of disulfide bonds, LCPN hydrogel can be quickly bonded under UV light without relying on photoinitiators. In addition, the incorporation of PPy NPs not only enhances the electrical conductivity of LCPN hydrogel, but also confers photothermal antimicrobial capability to LCPN hydrogel. More importantly, the sustained release of NMN in LCPN hydrogel can significantly enhance cell proliferation, migration and antioxidant capacity, which is conducive to accelerated wound healing. In vitro and in vivo experiments have shown that LCPN hydrogel has excellent biocompatibility and the ability to promote wound healing. Therefore, the prepared multifunctional hydrogel is expected to be used as a novel dressing to accelerate wound healing.

Abstract:The non-toxic self-crosslinked hydrogel films designed from biocompatible materials allow for controlled drug release and have gathered remarkable attention from healthcare professionals as wound dressing materials. Thus, in the current study the chitosan (CS) film is infused with oil-in-water Pickering emulsion (PE) loaded with bioactive compound quercetin (Qu) and stabilized by dialdehyde cellulose nanocrystal-silver nanoparticles (DCNC-AgNPs). The DCNC-AgNPs play a dual role in stabilizing PE and are involved in the self-crosslinking with CS films. Also, this film could combine the advantage of the controlled release and synergistic wound-healing effect of Qu and AgNPs.

Abstract:This work proposes the development of a polymer film made up of affordable components for its use as a healthcare material. Chitosan, itaconic acid, and Randia capitata fruit extract (Mexican variation) are the unique ingredients of this biomaterial prospect. Chitosan (from crustacean chitin) is crosslinked with itaconic acid, and in situ added R. capitata fruit extract in a one-pot reaction carried out in water as the sole solvent. Structurally, the film formed is an ionically crosslinked composite characterized by IR spectroscopy and thermal analysis (DSC and TGA); cell viability was also performed in vitro using fibroblasts BALB/3T3. Dry and swollen films were analyzed to determine affinity and stability in water. This chitosan-based hydrogel is designed as a wound dressing due to the combined properties of the chitosan with R. capitata fruit extract, which has potential as bioactive material due to its properties in epithelial regeneration.

Abstract:In this work, chitosan-based films containing gelatin and chondroitin-4-sulfate (C4S) with and without ZnO particles were produced and tested in vitro to investigate their potential wound healing properties. Chitosans were produced from shrimp-head processing waste by alkaline deacetylation of chitin to obtain chitosans differing in molecular weight and degree of deacetylation (80 ± 0.5%). The film-forming solutions (chitosan, C4S and gelatin) and ZnO suspension showed no toxicity towards fibroblasts or keratinocytes. Chitosan was able to agglutinate red blood cells, and film-forming solutions induced no hemolysis. Film components were released into solution when incubated in PBS as demonstrated by protein and sugar determination. These data suggest that a stable, chitosan-based film with low toxicity and an ability to release components would be able to establish a biocompatible microenvironment for cell growth. Chitosan-based films significantly increased the percentage of wound healing (wound contraction from 65 to 86%) in skin with full-thickness excision when compared with control (51%), after 6 days. Moreover, histological analysis showed increased granulation tissue in chitosan and chitosan/gelatin/C4S/ZnO films. Chitosan-based biopolymer composites could be used for improved biomedical applications such as wound dressings, giving them enhanced properties.

Abstract:Carboxymethylated Cassia fistula gum (CCFG) and citric acid (CA) based wound healing film, (CCFG-CA) was developed using the solvent casting method. Glycerol was added as a plasticizing agent. The synthesized Carboxymethylated Cassia fistula gum cross-linked citric acid based hydrogel film (CCFG-CA) was evaluated morphologically, thermally, and structurally using FESEM, TGA, XRD and FTIR. Three essential oils (EO), rosemary ( Rosmarinus officinalis) , turmeric ( Curcuma longa) and thuja ( Thuja occidentalis L), known for antimicrobial and antioxidant activities, were loaded into the CCFG-CA film to develop essential oils loaded carboxymethylated Cassia fistula gum cross-linked citric acid based hydrogel film (CCFG-CA-EO). In vitro studies (MTT assay, disk diffusion assay, permeability tests and DPPH assay) confirm the biocompatibility, anti-oxidant and anti-microbial properties of the CCFG-CA-EO film. In vivo (wound healing studies on wistar rats and their histology) shows 99 % of wound healing and re-epithelialization in 14 days. Degradability (within 15 days), protein adsorption (12.05 μg/mL) and contact angle determination (69.43°ׄׄ ± 0.48) tests confirmed the potential of CCFG-CA-EO as an effective wound-healing material.

Novel chitosan-based hydrogels as promising wound dressing materials with advanced properties

Abstract:Herein, four different grafted chitosans were synthesized by covalent attachment of glycine, L-arginine, L-glutamic acid, or L-cysteine to the chitosan chains. All products were subsequently permethylated to obtain their corresponding quaternary ammonium salts to enhance the inherent antimicrobial properties of native chitosan. In all cases, transparent hydrogels with the following remarkable characteristics were obtained: i) high-water absorption capacity (32-44 g H2O per g of polymer), ii) viscoelastic behavior at low deformations, iii) flexibility when subjected to deformations and iv) stability over long time scales. All the permethylated derivatives successfully inhibited 100 % of the growth of S. aureus. They also exhibited higher antimicrobial activity against E. coli than native chitosan. The structure of the chemically crosslinked products was more stable under external perturbations than that of the physically crosslinked ones. Between the chemically crosslinked products, the permethylated glutamic acid-grafted chitosan exhibited a noteworthy higher water absorption capacity with respect to that modified with cysteine, which makes it the most promising material for various industrial applications, including biomedical and food industries. Regarding biomedical applications, this derivative met the required physicochemical criteria for wound dressings, which encourages the pursuit of biological studies necessary to ensure the safety of its use for this application.

Chitosan/Essential Oils Formulations for Potential Use as Wound Dressing: Physical and Antimicrobial Properties

Abstract:Film-forming emulsions and films, prepared by incorporating different concentrations of clove essential oil (CEO) and melaleuca essential oil (MEO) into chitosan (CS) were obtained and their properties were evaluated. Film-forming emulsions were characterized in terms of qualitative assessment, hydrogen potential and in vitro antibacterial activity, that was carried by the agar diffusion method, and the growth inhibition effects were tested on the Gram-positive microorganism of Staphylococcus aureus, Gram-negative microorganisms of Escherichia coli, and against isolated fungi such as Candida albicans. In order to study the impact of the incorporation of CEO and MEO into the CS matrix, the appearance and thickness of the films were evaluated. Furthermore, Fourier transform infrared spectroscopy (FTIR), contact angle measurements, a swelling test, scanning electron microscopy and a tensile test were carried out. Results showed that the film-forming emulsions had translucent aspect with cloudy milky appearance and showed antimicrobial properties. The CEO had the highest inhibition against the three strains studied. As regards the films’ properties, the coloration of the films was affected by the type and concentration of bioactive used. The chitosan/CEO films showed an intense yellowish coloration while the chitosan/MEO films presented a slightly yellowish coloration, but in general, all chitosan/EOs films presented good transparency in visible light besides flexibility, mechanical resistance when touched, smaller thicknesses than the dermis and higher wettability than chitosan films, in both distilled water and phosphate-buffered saline (PBS). The interactions between the chitosan and EOs were confirmed by. The chitosan/EOs films presented morphologies with rough appearance and with EOs droplets in varying shapes and sizes, well distributed along the surface of the films, and the tensile properties were compatible to be applied as wound dressings. These results revealed that the CEO and MEO have a good potential to be incorporated into chitosan to make films for wound-healing applications.

Abstract:The current research aims to develop and evaluate chitosan-PLGA biocomposite scaffolds in combination with quercetin liposomes to accomplish the desired impact in oral lesions where pharmacotherapeutic agent treatment through circulation could only reach the low content at the target. Optimization of quercetin-loaded liposomes was carried out using 32 factorial design. The preparation of porous scaffolds comprising produced quercetin-loaded liposomes by thin-film method was carried out in the current study using a unique strategy combining solvent casting and gas foaming procedures. The prepared scaffolds were tested for physicochemical properties, in vitro quercetin release study, ex vivo drug permeation and retention research using goat mucosa, antibacterial activity, and cell migration studies on fibroblast L929 cell lines. Improved cell growth and migration were seen in the order control < liposomes < proposed system. The proposed system has been examined for its biological and physicochemical features, and it has the potential to be utilized as an efficient therapy for oral lesions.

Abstract:As the food processing by-products, hericium erinaceus residues (HER) and pineapple peel (PP) are good sources of cellulose and chitosan that can be prepared into hydrogels for structuring a drug delivery system. Hydrogel is one new type biomaterial for drug delivery with excellent absorbent ability applied in wound dressing. In this research, one composite self-healing hydrogel with pH sensitivity for drug delivery system based on the Schiff-base reaction was fabricated. Therein aldehyde group of oxidized microcrystalline cellulose (OMCC) from PP were crosslinked with amino group of carboxymethyl chitosan (CMCS) from HER via Schiff-base reaction for structuring hydrogels. The structures of the prepared hydrogels were characterized. Meanwhile, its blood clotting activity and physical properties were investigated. The hydrogels show some favorable performances with suitable gel time (54 s of minimum), distinguish swelling rate (about 31.18 g/g), good mechanical, self-healing characteristic and well coagulation effect. The cumulative release of the rutin-loaded hydrogel OMCM-54 reached about 80 % within 6 h, suggesting the well-controlled release of rutin by crosslinking degree between the modified OMCC and CMCS based on Schiff-base reaction. The novel biomaterial based on hericium erinaceus residues and pineapple peel shows its potential use as wound dressing.
