Abstract:A novel nanoprobe was prepared by encapsulating carbon dots (CDs) and gold nanoclusters (AuNCs) into zeolitic imidazolate framework-8 (ZIF-8) for sensitive detecting adenosine triphosphate (ATP). Under excitation at 360 nm, the obtained CDs/ nanoprobe exhibits dual-emissions at 469 nm and 660 nm, respectively, corresponding to the fluorescence emission of CDs and the aggregation-induced emission enhancement (AIEE) of AuNCs. The framework of ZIF-8 in this probe can be degraded by ATP due to the coordination competition of ATP and 2-Methylimidazole towards zinc ion (Zn 2+ ), resulting in the release of CDs and AuNCs. The following dispersion of CDs would improve efficiencies of the fluorescence excitation and the consequent emission of CDs. On the contrary, the AIEE of AuNCs would be decreased spontaneously after the AuNCs originally restricted in ZIF-8 were allowed to escape. The intensity ratio of fluorescence at 469 nm to that at 660 nm ( I 469 / I 660 ) was conveniently employed as the response signal for representing the amount of ATP. This nanoprobe exhibits excellent sensitivity and selectivity toward ATP, with a limit of detection (LOD) of 0.061 μM. Besides, low cytotoxicity of this nanoprobe facilitates its application as a fluorescent indicator in fluorescence imaging of living cells. Encapsulating two types of fluorescent nanomaterials by a degradable ZIF-8 structure makes the ratiometric fluorescence response of the nanocomposite probe towards the target analyte that destroys the ZIF-8 structure possible, and simplifies the application of the probe.

Abstract:The accumulation of cobalt(II) and antibiotics in the environmental water system has posed a significant threat to both human health and the safety of the ecological environment. Therefore, it is of utmost importance to look for a proficient means to detect cobalt(II) and antibiotics. Herein, we introduce a luminescent Zn-MOF, constructed with a thiazolo[5,4- d ]thiazole ligand that features a chromophore group, which serves as a sensor to detect cobalt(II) and antibiotics. The Crystal structure of {[Z n 2 (OBA) 2 (4 bpyttz )]·2DMF} (1) (4bpyttz=2,5-bis(pyridine-4-yl)thiazolo[5,4- d ]thiazole, H 2 OBA=4,4′-oxybis(benzoic acid)) displayed a 6-c net three-dimensional (3D) framework structure with novel topological type denoted as {4 4 ·6 10 ·8}. Moreover, luminescence sensing studies demonstrated that 1 exhibits high selectivity and multi-responsiveness of multiple ions mainly for Co 2+ , and nitrofuran antibiotics mainly for NZF, and NFT in an aqueous medium inducing turning-off effects. The detection limits are determined to be 1.14, 0.14, and 0.072 μ M for 1 , respectively. Additionally, MOF-derived fluorescence test papers and composite films were utilized for the visual detection of Co 2+ , NZF, and NFT, thereby expanding the potential applications of the system in real-world sensing. Furthermore, the potential luminescence quenching mechanism of MOF 1 regarding Co 2+ ions and nitrofuran antibiotics could be deduced through various characterization methods.

Abstract:Out of 83 patients studied 72 were certified as dying from asthma, and 11 aged under 45 as dying from chronic bronchitis and pneumonia. Fifty-three deaths were thought to be due to asthma. There were avoidable factors associated with several of these deaths from asthma. Recent discharge from hospital (16%), non-availability of aerosol bronchodilators (45%), underuse of corticosteroids (66%), and lack of objective measurements of airflow obstruction (100%) were found in deaths outside hospital. Inadequate initial assessment including baseline spirometry and blood gases (50%), significant underusage of corticosteroids (93%) and intravenous and nebulised bronchodilators (100%), and failure to monitor treatment objectively (100%) were found in deaths in hospital. "False-positive" and "false-negative" certifications of asthma were studied, and the findings suggest that these may lead to appreciable inaccuracy in the reporting of deaths from asthma.

Abstract:It is of great significance to propose simple methods to detect DNA bases sensitively for biological analysis and medical diagnosis. Herein, a highly crystalline polyimide covalent organic framework (TAPM-COF) has been successfully synthesized via a solvothermal route using pyromellitic dianhydride (PMDA) and tris(4-aminophenyl) amine (TAPA), which possessed large specific surface area (2286 m 2 g −1 ) and excellent thermal stability. Intriguingly, the crystallinity of the TAPM-COF improved significantly with the increase of water content in the reaction medium. To verify this phenomenon, we synthesized TPPM-COF with two pores by pyromellitic dianhydride (PMDA) and N,N,N′,N'-tetrakis(4-aminophenyl)-1,4-benzenediamine (TPDA), which bonding was similar to TAPM-COF. Furthermore, the prepared TAPM–COF–0.3 was used to construct a novel and independent electrochemical biosensor on glassy carbon electrode for simultaneously determination of adenine (A) and guanine (G) without other additives. However, to further improve signal of TAPM-COF in the field of electrochemical sensors, the crystalline TAPM–COF–0.3 can be readily integrated with amino-functionalized multiwalled carbon nanotubes (NH 2 -MWCNT) to form core-shell TAPM–COF–0.3@NH 2 -MWCNT driven by a π–π stacking interaction for more sensitive electrochemical sensing toward purine bases. In comparison to TAPM-COF/GCE, the TAPM-COF@NH 2 -MWCNT/GCE exhibited more favorable linear range and lower limit of detection. The work provided a new strategy for amplifying signal of COF in the field of electrochemical sensors.

Abstract:Metal-organic frameworks (MOFs) receive more and more interest in the field of analytical chemistry for their diverse structures and multifunctionality. In this study, we have designed and fabricated nanoscale MOF-based nanoprobes for multicolor detection of DNA targets with improved sensitivity. To do so, MOF-based nanoprobes, constructed by using porous MOFs as a scaffold to load signal dyes and a DNA hairpin structure as capping shell, have been prepared. Once the target has been introduced, a competitive displacement reaction triggers the release of fluorophores from the MOFs' pores. Consequently, a significantly enhanced fluorescence signal can be observed owing to the high loading capacity of MOFs. Therefore, the stimuli-responsive nanoprobes can enable sensitive detection of DNA targets with a low detection limit of 20 fM and selective identification to discriminate single-base mismatch. Moreover, the MOFs can encapsulate different fluorophores with different DNA gatekeepers designed according to the sequence of the target DNA, resulting in more kinds of stimuli-responsive nanoprobes for multiplexed DNA analysis in the same solution. Furthermore, these smart nanoprobes reported in this paper may provide a unique MOF-based tool for detection of various targets via stimuli-responsive systems in the future to widen the applications of MOFs.

Abstract:Two kinds of Luminescent metal organic framework (LMOF) based probes were developed as fluorescent oligonucleotide indicators for ratio-metric microRNA (miRNA) sensing. One LMOF (AuNCs@ZIF-8) is ZIF-8 encapsulated with gold nanoclusters (AuNCs). Another LMOF (Zn2Ph2Da) is prepared by using 2, 2′-dithiodibenzoic acid (dtba) and 1, 10-phenanthroline (phen) as ligands, and Zn2+ as metal center. Both of them possess fluorescence emission behavior and fluorescence quenching ability toward adsorbed fluorophore labeled miRNA aptamers. When target miRNAs hybridize with aptamers, the fluorescence of labeled fluorophores would recover. Meanwhile, the fluorescence intensity of LMOFs keeps unchanged. For enhancing miRNA detection sensitivity, novel fluorophore-labeled aptamers (SYBR-dsDNA-Aps) composed of miRNA aptamers and dsDNA fragments embedded with SYBR Green I molecules were designed and optimized. The detection of miR-21 and miR-155 in the blood serum of breast cancer bearing mice or inflammatory rats prove that prepared fluorescent oligonucleotide indicators could be promising choices for ratiometric miRNA sensing.

Abstract:Enrichment and detection of circulating free nucleic acids in biological samples have gained great attention for disease diagnosis or prognostic evaluation. Nanoscale metal-organic frameworks (NMOFs) have been used for aptamer-based nucleic acid sensing. In this work, different NMOFs, including ZIF-8, MIL-88, MIL-100, MIL-101, as well as Eu-TDA and Tb-TDA [prepared by the coordination of 2,2′-thiodiacetic acid (TDA) and Eu3+ or Tb3+], were investigated in nucleic acid sensing by employing their aptamer adsorption ability and fluorescence quenching capacity for the labeled dyes. Two types of dye aptamer, FAM-labeled aptamer (FAM-Ap) and TexasRedaptamer (TexasRed-Ap) were designed, and their adsorption properties on NMOFs-were compared. It was found that the TexasRed-Ap can be well used for nucleic acid (miR-21) extraction and sensing by linking with a pH-responsive nucleotide chain (TexasRed-Ap-pH) or with an additional random chain ssDNA-1′ (TexasRed-Ap-a). After interacted with the target miR-21 in biosamples, the TexasRed-dsDNA+NMOFs composites can be collected, and the formed TexasRed-dsDNA can be released by changing pH value or addition of ssDNA-1, which is matched with ssDNA-1′. A linear relationship from 0.1 to 200 pM for miR-21 detection was obtained. The results show that the NMOFs can be used as promising platforms for nucleic acid extraction and fluorescent sensing.

Abstract:The detection of nucleic acid marker is essential for accurate and timely diagnosis and prognosis of different types of diseases. However, the sensitivity and specificity of the assay still need to be improved. Due to the highly ordered porous structure, adjustable pore size, and large specific surface area, metal/covalent-organic frameworks (MOFs/COFs) can construct multifunctional sensors, which show significant advantages of adsorption, fluorescence quenching/emission, and electrocatalysis in nucleic acid detection. This review provides an overview of the design and construction of biosensors using MOFs/COFs for nucleic acid detection. The interaction between MOFs/COFs and nucleic acids and the detection of ctDNAs, microRNAs, mRNAs, genes, and other nucleic acid molecules based on these biosensors have been comprehensively discussed. In addition, the successful cases proposed by our and other groups have been analyzed in detail. The focus is on the rational design of biosensors by taking advantage of the unique structures and optical properties of MOFs/COFs to improve the sensitivity and selectivity of nucleic acid detection. We hope that the perspectives and insights provided in this review will promote the exploration of further potential applications of MOFs and/or COFs in biomarker sensing and imaging.

Electrochemical behavior of MOF-801/MWCNT-COOH/AuNPs: A highly selective electrochemical sensor for determination of guanine and adenine

Abstract:In this work, a novel sensor based on gold nanoparticle-modified zirconium-metal-organic frameworks (MOF-801) and carboxyl-functionalized carbon nanotube (MWCNT-COOH) composite material was built successfully for electrochemical detection of guanine and adenine in DNA sample, which exhibits diverse advantages such as fast, convenience and high efficiency. Guanine and adenine were detected by the cyclic voltammetry (CV) and differential pulse voltammetry (DPV). Under the optimal conditions, MOF-801/MWCNT-COOH/ AuNPs/GCE delivered linear DPV responses to guanine in the range of 0.8 ~ 60, 60 ~ 180 μM and the adenine in the range of 0.8 ~ 60, 60 ~ 180 μM with the low detection limit of 0.09 μM for guanine and 0.08 μM for adenine, respectively. The advantages of this method include simplicity, rapidity, high sensitivity, good selectivity as well as low cost. The modified GCE was successfully applied to the analysis of guanine and adenine in DNA samples.

Abstract:Real-time monitoring of adenosine-5′-triphosphate (ATP) and investigating its ability to bind Fe3+ are of great significance for tracking of Fe3+ in cells and cell functions. However, most nanomaterials for fluorescent nanoprobes have been randomly designed, and there are no reports about band gap of nanomaterials for matching with the redox potential of Fe3+/Fe2+, which could be helpful for further sequential sensing of Fe3+ and ATP in cells. Herein, the fluorescent nanoscale covalent organic framework (NCOF) is synthesized and has −2.97 eV of the conduction band and 0.87 eV of the valence band from the theoretical calculations. Coincidentally, the energy band of NCOF fluorescence excellently meets the requirements of energy transfer for the redox potential of Fe3+/Fe2+ (0.77 V vs normal hydrogen electrode (NHE)). This allows us to easily design a NCOF-based fluorescent probe for sensing Fe3+ and ATP in cells. The experimental results demonstrated that the NCOF biosensor had significant superiority in Fe3+ and ATP detection, which had a broad linear response to Fe3+ and ATP, ranging from 0.5 to 100 μM and 0.005 to 5 μM, respectively. These results allowed to achieve ultralow detection limits of 0.061 and 0.0024 μM for the detection of Fe3+ and ATP in cells, respectively. Based on the excellent fluorescence properties and good biocompatibility of the NCOF, the probe has been further used for sensitive imaging of Fe3+ and real-time dynamic monitoring of ATP in cells. In addition, the mechanism of interaction among NCOFs, Fe3+, and ATP was first verified by theoretical calculations. These results are promising to improve the development of fluorescent nanoprobes for biosensor and bioimaging applications.

Abstract:Metal-organic frameworks (MOFs) have been successfully used as efficient quenchers for fluorescent DNA detection. However, the surface charge property of MOFs can inevitably affect their fluorescence quenching behavior. Herein, nanoscale MOFs (NMOFs), including MOF nanosheets and nanoparticles, have been employed to investigate the relationship between the fluorescence quenching and surface properties of NMOFs. We find that the positively and negatively charged NMOFs exhibited totally opposite fluorescence quenching properties toward negatively charged FAM-labeled double-stranded DNA (dsDNA). On the contrast, they show negligible influence on the sensing of positively charged TAMRA-labeled dsDNA. This study provides a new insight of the fluorescence quenching property of NMOFs and offers a new concept for construction of ratiometric fluorescence DNA biosensors.

Abstract:Due to the excellent ion-sensing potential of covalent organic frameworks (COFs), the new imine-linked conjugated COF (IC-COF) is synthesized through a water-based synthesis reaction between 1,5-diaminonaphthalene and 2,4,6-tris(4-formylphenoxy)-1,3,5-triazine to create a luminescence sensor. It is noteworthy that the green synthesized IC-COF shows excellent selectivity to phosphate ions (PO43-) with a detection limit of 0.61 μM. The recyclability performance of IC-COF is high, indicating that it can be reused without a significant reduction in performance (5.2% decline after 5 cycles). Theoretical calculations using the density functional theory are performed on the IC-COF-PO43- and IC-COF-Cu+ complexes to explore the sensing mechanism. The fluorescence quenching in the presence of PO43- ions is attributed to the difference between PO43- binding sites to the IC-COF compared to Cu+, which leads to the considerable change in the IC-COF absorption spectrum from 400 to 600 nm.

Abstract:Special recognition sites in luminescent covalent organic frameworks (LCOFs) are extremely important to improve the selectivity of detection. Herein, we report on a novel LCOF named TFBD-Py-COF with three-in-one recognition traps composed of hydrogen bonding, reversible protonation of N and O–H⋯NC chelating unit. TFBD-Py-COF is synthesized via 3,3′,5,5′-tetraformyl-4,4′biphenyldiol (TFBD) and 4,4′,4′′,4′′′-(pyrene-1,3,6,8-tetrayl) tetra aniline (PyTTA), which has a two-dimensional dual rhombic pore structure based on AA stacking with the pore diameters of 0.88 and 1.38 nm. The hydroxyl group of TFBD-Py-COF can form hydrogen bonds with imines and induce the trans conformation of the mesoimine, as evidenced by the single crystal structure of the model compound. TFBD-Py-COF exhibits good chemical and thermal stability with a high BET surface area of 750 m2 g−1. TFBD-Py-COF shows visible fluorescent color from blue to green when dispersed in different organic solvents. TFBD-Py-COF has the fluorescence chemo-sensing ability for nitro-pesticides based on electron transfer-induced fluorescence quenching mechanism, allowing a high sensitivity and low detection limit of 4.0 × 10−2 ppm for p-nitroaniline. In addition, TFBD-Py-COF shows reversible acidochromic behaviors and concomitant distinct fluorescent color change due to the protonation of the imine N. Furthermore, the fluorescence of TFBD-Py-COF is selectively quenched by Fe3+ with a Ksv of 1.1 × 104 and a detection limit of 0.2 μM, owing to the coordination of Fe3+ with the O–H⋯NC chelating unit. This study demonstrates great potential of LCOFs for multifunctional detection.

Abstract:With increasing accessibility and advancements in abdominal imaging modalities, the incidence of pancreatic neuroendocrine neoplasms has increased steadily during the past few decades. By definition, neuroendocrine neoplasms of the pancreas show neuroendocrine differentiation, but they represent a broad and heterogeneous group of neoplasms with diverse clinical and pathological characteristics. The majority of pancreatic neuroendocrine neoplasms can be classified as well‐differentiated pancreatic neuroendocrine tumours (PanNETs) or poorly differentiated pancreatic neuroendocrine carcinomas (PanNECs). While PanNETs and PanNECs are distinct entities with respect to clinical presentation, outcome and therapeutic approach, they may exhibit overlapping histopathological features. Moreover, the frequent modifications in nomenclature and prognostic grading systems over the years of not only pancreatic neuroendocrine neoplasms, but neuroendocrine neoplasms from other organ sites, has created confusion for both pathologists and clinicians as to the appropriate use of terminology and grading when evaluating these neoplasms. This review examines the current concepts and issues of nomenclature and grading of PanNETs and PanNECs. In addition, considering the morphological overlap between high‐grade (G3) PanNETs and PanNECs, we discuss an integrative and practical diagnostic approach to aid in discriminating challenging cases.

Abstract:Various sensing platforms based on molecular or nanosystems are widely exploited through molecular diversity and specific recognition. However, it is extremely challenging to develop systems with tunable sensing ability and utilize the systems as information carriers/covers for communication and safety. Herein, DNA nanosensing systems based on cobalt oxyhydroxide (CoOOH) nanosheets were constructed for tunable detection and valence distinction of metal ions, molecular crypto-steganography, and information coding. CoOOH nanosheets absorb fluorescence-labeled single-stranded DNA with different bases and lengths, resulting in fluorescence quenching. The binding priority of bases with CoOOH nanosheets was guanine (G) > cytosine (C) > adenine (A) ≈ thymine (T) and the short chain excelled long chain. Due to the differences in the interaction among CoOOH, DNA, metal ions and variability of DNA bases, various DNA−CoOOH nanosystems have significantly different selective response patterns (that is selectivity) to metal ions and tunable linear ranges to Fe3+, Hg2+, Cr3+. Interestingly, by utilizing their molecular diversity, recognition, selective patterns, DNA−CoOOH sensing systems can be served as doubly cryptographic and steganographic systems to implement information encoding, encryption, and hiding and to reversely improve the selectivity of metal ions. This study provides an idea and platform for adjustable detection and valence distinction of metal ions, and gives a set of "molecular programming languages" for designing intelligent programmable sensing and molecular information communication and safety systems.

Abstract:Copper ion (Cu2+) detection remains an important task for monitoring water quality because of its specific toxicity. Herein, a new dual-signal fluorescent probe was developed by combining zeolitic imidazolate framework-8 (ZIF-8) and lanthanide for the detection of Cu2+ for the first time. The lanthanide coordination polymer (guanosine monophosphate and Eu3+, GMP/Eu) was initially incorporated into ZIF-8 to yield ZIF-8/GMP/Eu nanomaterials with extremely weak single emission fluorescence at 618 nm. It was found that the resulted nanomaterials could display a dual emission fluorescence at 515 nm and 618 nm after the introduction of tetracycline (TC) due to the synergistic effect of aggregation-induced emission effect (AIE, TC induced by ZIF-8) and antenna effect (AE, between TC and GMP/Eu). Interestingly, in the presence of Cu2+, the AIE of TC was destroyed because of the interaction of Cu2+ with ZIF-8 and TC. The AE between TC and GMP/Eu disappeared due to the formation of complex between TC and Cu2+. A dual-signal fluorescent probe of ZIF-8/GMP/Eu/TC was thereby established for sensing Cu2+ in the range of 0.5-100 μM. Such a dual-signal response strategy that intelligently utilized the "ON"/"OFF" of AIE and AE can not only eliminate the background interference, but also ensure the improved selectivity of Cu2+ sensing. Subsequently, the dual-signal fluorimetric strategy was applied for the detection of Cu2+ in environmental water samples, indicating the potential feasibility of applications for water quality monitoring.

Abstract:Abstract Metal–organic frameworks (MOFs) with high stability and porosity have gained great attention in bioanalysis due to their potential in improving sensitivity and robustness of assays. Herein, to improve both the stability and the emission intensity of Cu nanoclusters (CuNCs), in situ entrapment strategy of CuNCs into zeolitic imidazolate framework-8 (ZIF-8) is described. Blue emissive and stable CuNCs was prepared, for the first time, using thiamine hydrochloride as capping agents, and showed strong and stable emission at 440 nm when excited at 375 nm with fluorescence quantum yields 12%. Encapsulation of CuNC into ZIF-8 showed dramatic enhancement of the fluorescence intensity up to 53% fluorescence quantum yield. Furthermore, the CuNCs@ZIF-8 possesses better stability (more than three months) due to protective and confinement effect of MOFs. Upon the addition of tetracycline to CuNCs@ZIF-8 solution, the blue emission intensity was significantly decreased. The fluorescence ratio (Fo/F) against the concentration of tetracycline exhibited a satisfactory linear relationship from 1.0 to 10.0 μM with a detection limit (LOD) of 0.30 μM. Abstract:An electrochemical DNA sensor was developed based on the electrocatalysis of porphyrinic metal-organic framework (MOF) and triple-helix molecular switch for signal transduction. The streptavidin functionalized zirconium-porphyrin MOF (PCN-222@SA) was prepared as signal nanoprobe via covalent method and demonstrated high electrocatalysis for O2 reduction. Due to the large steric effect, the designed nanoprobe was blocked for the interaction with the biotin labeled triple-helix immobilized on the surface of glassy carbon electrode. In the presence of target DNA, the assistant DNA in triple-helix will hybridize with target DNA, resulting in the disassembly of triple-helix molecular. Consequently, the end biotin away from the electrode was ''activated'' for easy access to the signal nanoprobe, PCN-222@SA, on the basis of biotin-streptavidin biorecognition. The introduction of signal nanoprobe to a sensor surface led to a significantly amplified electrocatalytic current towards oxygen reduction. Integrating with DNA recycling ampliﬁcation of Exonuclease III, the sensitivity of the biosensor was improved signiﬁcantly with detection limit of 0.29 fM. Moreover, the present method has been successfully applied to detect DNA in complex serum matrix. This porphyrinic MOF-based strategy has promising application in the determination of various analytes for signal transduction and has great potential in bioassays.
