Abstract:Two fluorescent covalent organic frameworks (COFs) which bear two kinds of pores with different sizes and shapes have been synthesized. The heteropore COFs exhibit spectroscopic and color changes to 2,4,6-trinitrophenol (TNP) with extremely high selectivity and sensitivity, which makes them excellent macroscopic chemosensors for the selective detection of TNP.

Abstract:A porphyrin-based two-dimensional (2D) covalent organic framework (COF) was developed by a C-4 + C-4 topological diagram. It was constructed by the condensation of zinc 5,10,15,20-tetra(4-aminophenyl)porphyrin (TAPP) and zinc 5,10,15,20-tetra(4-formylphenyl)porphyrin (TFPP) under typical solvothermal conditions, leading to the formation of a porphyrin-based TAPP-TFPP-COF with tetragonal micropores at a size of 1.8 nm. The resultant crystalline framework exhibited high crystallinity, excellent stability, and good porosity. Resulting from the specific pi-unit stacking columnar structure and excellent organic semiconducting property of porphyrins, the TAPP-TFPP-COF shows many promising applications in optoelectronics. Notably, after doping with iodine, the conductivity of this TAPP-TFPP-COF can be greatly enhanced from 1.12 x 10(-)(10) to 1.46 X 10(-7) S cm(-1). Furthermore, the nanometer-thick TAPP-TFPP-COF films were obtained using a liquid-air interface growth strategy. A spectroscopic detection device was constructed using COF thin films which displayed highly selective sensitivity toward the near infrared irradiation at 700 nm with an on-off ratio of up to 2.8 X 10(4). This value ranks as the highest among other COF-based and metal-organic-framework-based semiconducting materials under similar conditions. These results illustrated the enormous potential of 2D porphyrin COFs for future applications in optoelectronic devices and constituted an important step toward the development of new types of functional crystalline materials.

Abstract:Selective and sensitive detection of heavy metal ions is a theme of increasing scientific interest, great technological importance and extensive public concern. To realize this target, design of high-efficient and fluorescent materials as sensor is crucial yet challenging task. In this work, we describes the synthesis and functions of a new hydroxyl and imine groups functionalized covalent organic framework (COFs). The synthesized COFs-DT integrates multi-functionality such as extended π-conjugation framework, tunable functionality, regular pore structure, specific sites to interact with targets as well as robust chemical and thermal stability. COFs-DT shows strong and stable fluorescence when dispersed in isopropanol. The fluorescent property together with the unique bidentate ligand sites enables COFs-DT in the construction of a high-performance fluorescent sensor for detection of Cu2+ with high sensitivity and selectivity. Furthermore, we use a series of characterizations to investigate the fluorescence quenching mechanism and found the quenching through a photoinduced electron transfer mechanism with COFs-DT as donor and Cu2+ as acceptor. Our strategy not only highlights opportunities for utilizing COFs with integrated multiple unique nature as new type of fluorescent materials, but also expends the encapsulation scope of COFs for ion sensor in environmental field.

Cardiac Actin Capping Protein Reduction and Protein Kinase C Inhibition Maintain Myofilament Function During Cardioplegic Arrest

Abstract:Background: Heart transplantation is associated with cold, cardioplegic arrest that impairs myocardial function. Protein Kinase C (PKC) suppression of myofilaments may contribute to this dysfunction. CapZ-deficient cardiac myofilaments are unresponsive to PKC. We hypothesized that myofilaments from CapZ-deficient transgenic hearts are resistant to cardioplegic dysfunction and that PKC inhibition improves function. Methods: Heart function was assessed using a Langendorff apparatus. Myofilaments isolated from murine hearts were assessed with an actomyosin MgATPase assay and protein phosphorylation gels. PKC activation was examined by immunoblotting. Results: Wildtype hearts showed impaired function after cardioplegic arrest. CapZ-deficient transgenic mouse hearts performed significantly better after 1 h cardioplegia than wildtype hearts, but not after 4 h cardioplegic arrest. Wildtype myofilaments had depressed activation at 1 and 4 h cardioplegic arrest, as demonstrated by reduced actomyosin MgATPase activity. CapZ-deficient myofilaments showed no reduced actomyosin MgATPase activity at either time. Troponin I (TnI) phosphorylation increased by approximately 20% at 1 and 4 h in wildtype mice. Myosin binding protein C (MyBP-C),and troponin T (TnT) phosphorylation increased by less than 10% at 1 h, and tended to rise at 4 h. Myofilament protein phosphorylation was largely unchanged in CapZ-deficient hearts at 1 h, but MyBP-C tended to be dephosphorylated at 4 h cardioplegic arrest. Myofilament-associated PKC-α, -βII, -δ, and -ε increased at 1 and 4 h cardioplegia in wildtype hearts, whereas only PKC-α increased in transgenic myofilaments at 1 h. PKC inhibition abolished the cardioplegic-dependent changes in actomyosin MgATPase activity and TnI phosphorylation of wildtype myofilaments. Conclusions: We demonstrate a direct link between PKC activation and myofilament dysfunction associated with cold, cardioplegic arrest. Moreover, we show for the first time a cardioprotective benefit of decreased cardiac CapZ.

Abstract:Covalent organic frameworks (COFs) offer ordered it structures that are useful for developing light-emitting materials. However, most COFs are weak in luminescence. Here we report the conversion of less emissive COFs into light-emitting materials via a pinpoint surgery on the pore walls. Deprotonation of the N-H bond to form an anionic nitrogen species in the hydrazone linkage can eliminate the nitrogen-related fluorescence quenching pathway. The resulting COF enhances the fluorescence in a linear proportion to the progress of deprotonation, achieving a 3.8-fold improved emission. This pinpoint N-H cleavage on the pore walls can be driven only by the fluoride anion while other halogen anions, including chloride, bromide, and iodide, remain inactive, enabling the selective fluorescence switch-on sensing of the fluoride anion at a ppb level.
A pillar-layer metal-organic framework as a turn-on luminescent sensor for highly selective and sensitive detection of Zn(II) ion

Abstract:The intriguing architectures of metal-organic frameworks (MOFs), makes them an emerging class of promising materials in various application fields ranging from gas adsorption to sensing. In the present work, a three-dimensional Zn-based MOF, Zn2(BDC)2(DABCO), (H2BDC = 1,4-benzene dicarboxylic acid, DABCO = 1,4-diazabicyclo[2.2.2]octane) was prepared and its sensing behavior to metal ions was investigated by fluorescence assays. Our results show that the luminescence intensity of Zn2(BDC)2(DABCO) is strongly sensitive to Zn2+ ions. The effects of selectivity, initial concentration, the limit of detection, response time and regeneration were studied in the case of Zn2+ detection operations. The linear response to Zn2+ was obtained across the concentration range of 50–200 μM with the detection limit of 0.7 μM. The sensor also displayed excellent performance in selectivity for Zn2+ over other metal ions including Fe3+, Mn2+, Cu2+, As3+, Ni2+, Pb2+, Co2+, Al3+ and Cd2+ ions. The luminescence enhancement and recovery cycle were reiterated multiple times without much effect on the sensitivity. All the above features showed that this sensor as a sensing platform holds great potential for detection of Zn2+ with a response time of <1 min that has been rarely reported.

Abstract:Hydrogen‐bonded organic frameworks (HOFs) represent an emerging class of porous crystalline materials, have attracted growing interest in the field of fluorescence sensing area due to their distinctive luminescent characteristics, large surface area and abundant recognition sites. Herein, the porous material HOF‐BTB‐NH2 featuring one‐dimensional pore structure was successfully synthesized through hydrogen‐bonding self‐assembly using the organic building molecule 2,4,6‐Tris(4‐carboxyphenyl)aniline (BTB‐NH2) in methanol. HOF‐BTB‐NH2 showed excellent sensitivity and selectivity for metal ion Cu2+ and anion CO32‐, and the limits of detection (LOD) reached 0.23 μM and 0.33 μM, respectively. In addition, HOF‐BTB‐NH2 was employed as a biosensing material for the highly selective and sensitive detection of dopamine (DA). The synthesis of porous HOF‐BTB‐NH2 and the excellent sensing performance greatly expanded the application of hydrogen‐bonded organic framework materials in fluorescence sensing.

Abstract:The rapid development of functionalization of lanthanide metal organic frameworks (Ln-MOFs) offers the potential for biomarkers sensing. It is closely associated with biology science and clinic medicine. In this work, a porous and luminescent terbium metal-organic framework TbTATB has been designed and solvothermally synthesized. The PXRD results demonstrated the commendable stability of this sensor. Enhanced luminescence of TbTATB origin from Tb3+ was observed in the presence of Zn2+, which is a kind of biologically important ion. This turn-on pattern was utilized for sensing Zn2+. TbTATB exhibited excellent sensitivity and selectivity towards Zn2+ in aqueous solution with the detection limit of 10.5 nM. What's more, it has fairly acceptable bio-compatibility which was fully verified by cytotoxicity test. In the state of coexistence with living cells, visible increased green light of TbTATB emerged after the addition of Zn2+, while the survival state of cells was hardly disturbed. Markedly, this is the first time that a luminescent Tb-MOF can act as a sensor for sensing and imaging of Zn2+ ions simultaneously. What's more, the results also guide a new application direction of MOFs and supply a new way for the detection of other analytes in biological systems.

Abstract:The accumulation of cobalt(II) and antibiotics in the environmental water system has posed a significant threat to both human health and the safety of the ecological environment. Therefore, it is of utmost importance to look for a proficient means to detect cobalt(II) and antibiotics. Herein, we introduce a luminescent Zn-MOF, constructed with a thiazolo[5,4- d ]thiazole ligand that features a chromophore group, which serves as a sensor to detect cobalt(II) and antibiotics. The Crystal structure of {[Z n 2 (OBA) 2 (4 bpyttz )]·2DMF} (1) (4bpyttz=2,5-bis(pyridine-4-yl)thiazolo[5,4- d ]thiazole, H 2 OBA=4,4′-oxybis(benzoic acid)) displayed a 6-c net three-dimensional (3D) framework structure with novel topological type denoted as {4 4 ·6 10 ·8}. Moreover, luminescence sensing studies demonstrated that 1 exhibits high selectivity and multi-responsiveness of multiple ions mainly for Co 2+ , and nitrofuran antibiotics mainly for NZF, and NFT in an aqueous medium inducing turning-off effects. The detection limits are determined to be 1.14, 0.14, and 0.072 μ M for 1 , respectively. Additionally, MOF-derived fluorescence test papers and composite films were utilized for the visual detection of Co 2+ , NZF, and NFT, thereby expanding the potential applications of the system in real-world sensing. Furthermore, the potential luminescence quenching mechanism of MOF 1 regarding Co 2+ ions and nitrofuran antibiotics could be deduced through various characterization methods.

Abstract:For the first time, we develop a terbium (III)-functionalized covalent organic framework named 3+, through anchoring Tb3+ onto a two-dimensional imine COF (Dpy-NhBt-COF), as the selective and sensitive turn-on fluorescent switch for ochratoxin A (OTA) monitoring. Of particular significance, Tb3+ actually plays two roles during sensing process: the specific response signal, and exclusive recognition sites for OTA, while Dpy-NhBt-COF acts as the protector for Tb3+. The sensing process involves the replacement of coordinated water molecules from Tb3+ by OTA and the energy transfer from OTA to Tb3+ centers, resulting in remarkable fluorescence emergence of Tb3+. The stabilization of Tb3+ via coordination with bipyridine of Dpy-NhBt-COF not only reduces the nonselective binding of naturally occurring ligands, but also avoids the non-radiative quenching caused by solvents molecules. As a sensing platform, 3+ possesses noticeable selectivity and high sensitivity toward OTA with an ultralow detection limit of 13.5 nM and rapid response of 10 s. Taken together, our work not only demonstrates great prospect of Tb3+-functionalized COF for OTA detection, but also provides a potential way to explore other functionalized materials as promising sensors for other targets.

Abstract:Zinc ion (Zn2+) is a necessary transition metal ion in the human body and plays a vital role in biological systems. Therefore, it is meaningful for rapid, sensitive, and specific detection of Zn2+. Herein, a “switch-on” fluorescence sensing platform for the detection of Zn2+ was successfully established. In this work, we report an optical nanoprobe with amino functional groups based on porphyrin metal-organic frameworks (PCN-NH2) through a simple treatment method. The modification of the salicylaldehyde (SA) through the C=N acts as a quencher of PCN-NH2 and the ligand of Zn2+ can effectively inhibit the fluorescence emission of PCN-NH2. The complex of porphyrin-based metal-organic framework and salicylaldehyde (PCN/SA) acts as the fluorescence sensing probe. The presence of Zn2+ results in the new emission peak at 448 nm due to the formation of a complex between Zn2+ and SA. The corresponding fluorescence signal changes were used to accurately detect the concentration of Zn2+. The fluorescence sensing platform shows advantages, including rapid response, high sensitivity, and excellent specificity with a linear detection range of Zn2+ between 0.1 and 250 μM, and a low detection limit (LOD) of 0.07 μM. And the feasibility of the method was verified for the quantification of Zn2+ in cell lysate, environmental water, and plasma.Graphical abstract

Abstract:The development of luminescent metal-organic frameworks for effective sensing and monitoring of environmental pollutants is of great significance for human health and environmental protection. In this work, a novel water-stable ZnII-based luminescent coordination polymer, namely {[Zn(BBDF)(ATP)]·2DMF·3H2O}n ((BBDF = 2,7-bis(1H-benzimidazol-1-yl)-9,9-dimethyl-9H-fluorene) and H2ATP = 2-aminoterephthalic acid), was designed and obtained using the mixed-ligand method. Structural analysis indicated that 1 presents a two-fold interpenetrated two-dimensional layer structure with one dimensional (1D) channels along the a axis. Intriguingly, the uncoordinated -NH2 group was danged onto the pore walls of 1. Remarkably, compound 1 shows good aqueous stability at different pH values of 3-13 and exhibits a fluorescence turn-off sensing behavior for Hg2+, Cr2O72-, CrO42-, and antibiotics (NFZ, NFT) in aqueous solution with high selectivity and sensitivity. The limits of detection (LOD) are 0.12 μM (Hg2+), 0.17 μM (Cr2O72-), 0.21 μM (CrO42-), 0.098 μM (NFZ), and 0.14 μM (NFT). The luminescence quenching mechanism analysis by experiment and theoretical calculation revealed that the competitive absorption and the photoinduced electron transfer process are largely responsible for the sensing of the two antibiotics, while the weak interaction contributes to the selective luminescence quenching for Hg2+.

Abstract:Copper ions (Cu2+) play an essential role in various cellular functions, including respiration, nerve conduction, tissue maturation, oxidative stress defense, and iron metabolism. Covalent organic framework (COF) is a class of porous crystalline material with directed structural designability and high stability due to the combination of different monomers by covalent bond. In this study, we synthesized porphyrin-tetrathiazole COF (TT-COF(Zn)) with Zn-porphyrin and tetrathiafulvalene (TTF) as monomers and used it as photoactive materials. The strong light absorption of metalloporphyrin and the electron-rich properties of supplied TTF contribute to its photoelectrochemical performance. Additionally, the sulfur (S) in the TTF can coordinate with Cu2+. Based on these principles, we constructed a highly sensitive photoelectrochemical sensor for detecting Cu2+. The sensor exhibited a linear range from 0.5 nM to 500 nM (R2 = 0.9983) and a detection limit of 0.15 nM for Cu2+. Notably, the sensor performed well for detecting Cu2+ in water samples.

Abstract:Due to the dual functions of fluorescence detection and adsorption, fluorescent covalent organic frameworks (COFs) have attracted significant attention. However, common fluorescent COFs often exhibit unsatisfactory fluorescence properties and selectivity, coupled with poor solution dispersibility, which limit their effectiveness in detection and adsorption applications. In response, a novel post-modified fluorescent COF (named AZC-COF) was synthesized by connecting a fluorescent COF (COF-TB) with 2-azidacetic acid through a copper-catalyzed aide-alkyne cycloaddition (CuAAC) reaction. AZC-COF demonstrated excellent solution dispersibility and robust green fluorescence, boasting an absolute fluorescence quantum yield (QY) of 7.58%, which was 13.5 times higher than that of COF-TB. Furthermore, leveraging the active carboxylic acid and triazole sites, AZC-COF exhibited remarkable binding abilities for mitoxantrone (MIX) and Fe3+, enabling sensitive detection and efficient adsorption of them. In contrast, due to the absence of these functional sites, COF-TB showed poor detection and enrichment capabilities for MIX and Fe3+. The impressive detection and adsorption efficiencies of MIX and Fe3+ in environmental water, aquatic organism (fish) and plasma samples underscore the potential of AZC-COF as a detection-adsorption platform. Additionally, AZC-COF demonstrated low toxicity and hemolytic activity, alongside promising potential for cell imaging and detection of MIX and Fe3+, highlighting its considerable application prospect in biological systems.

Abstract:A novel metal-organic framework with the formula [Zn3(DDB)(DPE)]·H2O (1) (H5DDB = 3,5-di(2',4'-dicarboxylphenyl)benzoic acid and DPE = 1,2-di(4-pyridyl)ethylene) has been solvothermally synthesized by employing a rigid carboxylate ligand H5DDB to assemble with Zn(ii) ions in the presence of a flexible bis(pyridyl) linker DPE. The Zn-MOF is a 3D framework with six-nuclear clusters and possesses remarkable water stability and pH stability. Interestingly, complex 1 can sensitively and selectively sense Fe(iii), Cr(iii), Cr(vi), Mn(vii) and the pesticide 2,6-Dich-4-NA with low detection limits in aqueous solution. Moreover, complex 1 also exhibits selectivity for 2,6-Dich-4-NA detection in real samples including carrot, grape and nectarine extracts, and its detection ability is almost unchanged in the presence of the surfactant sodium dodecyl sulfate (SDS). The possible mechanisms of luminescence quenching have been explained by the weak affinity of nitrogen atoms, resonance energy transfer, and photoinduced electron transfer. To our knowledge, this is the first example of a MOF-based multiresponsive fluorescent probe for the simultaneous detection of Fe(iii), Cr(iii/vi), Mn(vii) and the pesticide 2,6-Dich-4-NA in aqueous solution.

Abstract:A new quinoline‐based COF (Covalent Organic Framework), obtained by Povarov reaction, containing 2,6‐diisopropylphenyl moieties as substituents over the heterocyclic ring is described for detecting Zn2+ in aqueous solution. The introduction of the mentioned bulky phenyl rings into the network favors an increase of the distance between the reticular sheets and their arrangement, obtaining a new material with an alternating AB type stacking. The new material exhibits good selectivity to detect Zn2+ by fluorescence emission in aqueous solutions up to a concentration of 1.2 × 10ꟷ4 M of the metal ion. In order to have a deeper insight into the interaction between the COF and the zinc cation, a thorough spectroscopical, microscopical and theoretical study is also presented and discussed in this communication. Terbium-based metal-organic frameworks through energy transfer modulation for visual logical sensing zinc and fluorine ions

Abstract:Zinc is the second most abundant trace element in the human central nervous system, which is closely related to various physiological activities in the human body. Fluoride ion is one of the most harmful elements in drinking water. Excessive intake of F- may cause dental fluorosis, renal failure, or DNA damage. Therefore, it is urgent to develop sensors with high sensitivity and selectivity for the detection of Zn2+ and F- ions at the same time. In this work, a series of mixed lanthanide metal-organic frameworks (Ln-MOFs) probes are synthesized using a simple method of in situ doping. The luminous color can be finely modulated by changing the molar ratio of Tb3+ and Eu3+ during synthesis. Benefiting from the unique energy transfer modulation mechanism, the probe has the continuous detection capability of zinc ions and fluoride ions. The detection of Zn2+ and F- in a real environment shows that the probe has a good practical application prospect. The as-designed sensor at 262 nm excitation can sequentially detect Zn2+ concentrations ranging from 10-8 to 10-3 M (LOD = 4.2 nM) and F- levels ranging from 10-5 to 10-3 M (LOD = 3.6 μM) with high selectivity. Based on different output signals, a simple Boolean logic gate device is constructed to realize intelligent visualization of Zn2+ and F- monitoring.

Abstract:Through a hydrothermal reaction of 3-(3-carboxylphenyl) isonicotinic acid (H2L) and dipyridyl with Zn(Ac)2·2H2O, two metal–organic frameworks (MOFs), [Zn(L)(bpa)0.5] n (1) and {[Zn(L)(bpp)0.5]·H2O} n (2) (bpa = 1,2-bi(4-pyridyl)ethane, bpp = 1,3-di(4-pyridyl)propane), are prepared. Complexes 1 and 2 have similar twofold interpenetrated three-dimensional (3D) layer-pillared networks. Compounds 1 and 2 exhibit outstanding fluorescence-sensing performances toward Cr2O7 2–, especially, featuring the ratiometric fluorescence against levofloxacin (LEVO) and ciprofloxacin (CIP). To the best of our knowledge, it is the first example of ratiometric detection using antibiotics. The quenching mechanism is mainly attributed to the large area of competitive absorption between the ultraviolet (UV) absorption bands of substances and the excitation peaks of complexes. Furthermore, compounds 1 and 2 could serve as efficient heterogeneous catalysts for the fixation of CO2 with various epoxides benefiting from the presence of unsaturated zinc ions as Lewis acid sites.
