Abstract:An in situ transesterification approach was demonstrated for converting lipid-rich wet algae (Nannochloropsis sauna) into fatty acid ethyl esters (FAEE) under microwave-mediated supercritical ethanol conditions, while preserving the nutrients and other valuable components in the algae. This single-step process can simultaneously and effectively extract the lipids from wet algae and transesterify them into crude biodiesel. Experimental runs were designed to optimize the process parameters and to evaluate their effects on algal biodiesel yield. The algal biomass characterization and algal biodiesel analysis were carried out by using various analytical instruments such as FTIR, SEM-EDS, TLC, GC-MS and transmission electron microscopy (TEM). The thermogravimetric analysis (TGA) under nitrogen and oxygen environments was also performed to examine the thermal and oxidative stability of ethyl esters produced from wet algae. This simple in situ transesterification process using a green solvent and catalyst-free approach can be a potentially efficient route for algal biodiesel production. (C) 2013 Elsevier Ltd. Abstract:To valorize oleaginous Schizochytrium sp. biomass, a novel sequential process was addressed and developed to simultaneously produce high value DHA-enriched glycerides and low value bulky biodiesels. In this process, Schizochytrium sp. biomass was first treated by alkaline protease; then the resulting biomass was directly subjected to enzymatic ethanolysis catalyzed by liquid enzymes from C. antarctica (CALA), T. lanuginosus (TL), R. miehei (RM) and A. oryzae (PLA). For all four liquid lipases, the alkaline protease treatment on Schizochytrium sp. biomass significantly enhanced total lipid recovery, biodiesel yield and yield of DHA-enriched glycerides. Under optimized conditions, CALA and TL achieved the highest biodiesel yields 76.28-76.66 wt% based on microalgae TFAs while PLA had the lowest (55.25 wt%). Moreover, CALA attained the highest DHA (77.93 wt%) and LC PUFAs (93.08 wt%) contents in glyceride fraction, suggesting a superior performance to concentrate DHA and LC PUFAs in glycerides. C-13 NMR analysis illustrated that non-regiospecificity and unfavorability towards LC PUFAs of CALA made it possible to transesterify saturated fatty acids located in all 3 positions of microalgal oil; thus LC PUFAs were highly enriched in glycerides. Overall, the sequential process using enzymes and ethanol as green media was successfully established for the co-production of high-value microalgal bioproducts rich in LC PUFAs and sustainable biodiesels in a comprehensive manner.

Abstract:Biochemical pathways to generate renewable and high value-added diols and dicarboxylic acids have attracted increasing interest. However, the diols/dicarboxylic acids usually exist in the form of low-concentration aqueous solution after the bioprocesses, causing the issues of low efficiency and high energy consumption in the separation and recovery of these highly hydrophilic bioproducts. Here, we developed an aqueous two-phase system (ATPS) consisting of ionic liquid (IL) and inorganic salt for efficient extraction of the bio-based diols and dicarboxylic acids. The optimal salt, IL, and extractant to feed volume ratio were K4P2O7, [Bmim][C12H23O2], and 1:4, respectively. The selected IL showed large capacity and good reusability, with the highest dehydration rate reaching up to 97.6%. More importantly, the IL-based ATPS exhibited effectiveness and high efficiency in the separation of various low-concentration diols and dicarboxylic acids, with the recovery exceeding 91.9%. To explore the extraction mechanism, the structural and interactions analyses were conducted by DFT calculations. The strength of hydrogen bond interactions formed between diols/dicarboxylic acids and ILs and those between salts and water largely determines the extraction performance. Moreover, three descriptors that can well represent the H-bond basicity and polarizability/dipolarity of ILs were established to facilitate the IL evaluation and screening.
Short alkyl-imidazolium ionic liquids enhanced in-situ transesterification of microalgae

Abstract:Ionic liquids (ILs) catalyzed in-situ transesterification (IST) of microalgae is a promising strategy to improve biodiesel yield through enhancing cellulose solubility and catalyzing transesterification. Effect of anions of ILs on biodiesel has been widely investigated previously. However, how do cations of ILs affect IST of microalgae is still unclear. On this basis, ILs composed of alkyl-imidazolium cations with different length, 1-ethyl-3-methylimidazolium tetrafluoroborate ([Emim][BF 4 ]), 1-butyl-3-methylimidazolium tetrafluoroborate ([Bmim][BF 4 ]), and 1-hexyl-3-methylimidazolium tetrafluoroborate ([Hmim][BF 4 ]) catalyzed IST of microalgae are experimentally and theoretically investigated. The results indicate that increasing alkyl chains of cations weakens interactions of ILs-cellulose, and reduces acidities of ILs, subsequently leads to decrease of cellulose extraction yield and biodiesel yield. Although cellulose extraction yield and biodiesel yield are affected by reaction temperature, dosage of methanol, and MILs: Algae, however, H-bonding interactions of ILs-cellulose and acidities of ILs are still dominate parameters affecting cellulose extraction yield and biodiesel yield, respectively, leading to their unchanged sort under variable reaction conditions as following: [Emim][BF 4 ] > [Bmim][BF 4 ] > [Hmim][BF 4 ]. This work reveals that short alkyl chain of cation is favorable to both cellulose extraction and biodiesel yield, providing theoretical guides for screen and design of benign ILs to further improve biodiesel of in-situ transesterification of microalgae.

Abstract:Given the recent extensive synthesis and application of ionic liquids (ILs), finding a sensitive and visual indicator to provide a fast-initial risk assessment of IL use has become a pressing issue. In this study, we verified that the phototaxis of Chlamydomonas reinhardtii is a valid indicator of the environmental risk associated with chiral ILs L-(+)- and D-(−)-1-butyl-3-methylimidazolium lactate (BMIM L). Briefly, C. reinhardtii was exposed to a 4000-lx side light source for varying lengths of time. Following the allotted exposure time, the algae aggregation was photographed, and then quantitative analysis was conducted using Image-J software to obtain the corresponding relationship between IL stimulation and C. reinhardtii phototaxis. The gray areas from each treatment were measured and the percentage was calculated. After 16 h of side lighting, for control, the percentage of gray areas was − 22%, while for L-(+)- and D-(−)- BMIM L were 17% and 33%, respectively. Then, after 8 h of darkness, where D-(−)-BMIM L and the control showed the positive phototaxis, but the L-(+)-BMIM L-treated group showed virtually no change. This phenomenon is consistent with excessive production of reactive oxygen species (ROS). Moreover, atomic force microscope (AFM) results indicated distinct aggregation between D-(−)- and L-(+)-BMIM L, which caused changes in cell permeability that induced a change in ROS transfer. Furthermore, relationship between phototaxis and changes in cell ultrastructure and photosynthetic efficiency was also investigated. This work demonstrates the potential of phototaxis to serve as a sensitive, convenient, and cost effective qualitative assessment of ILs’ toxic impact, with the understanding that quantitative evaluation requires further improvement.

Abstract:Carotenoids are high added-value products primarily known for their intense coloration and high antioxidant activity. They can be extracted from a variety of natural sources, such as plants, animals, microalgae, yeasts, and bacteria. Gordonia alkanivorans strain 1B is a bacterium recognized as a hyper-pigment producer. However, due to its adaptations to its natural habitat, hydrocarbon-contaminated soils, strain 1B is resistant to different organic solvents, making carotenoid extraction through conventional methods more laborious and inefficient. Ionic liquids (ILs) have been abundantly shown to increase carotenoid extraction in plants, microalgae, and yeast; however, there is limited information regarding bacterial carotenoid extraction, especially for the Gordonia genus. Therefore, the main goal of this study was to evaluate the potential of ILs to mediate bacterial carotenoid extraction and develop a method to achieve higher yields with fewer pre-processing steps. In this context, an initial screening was performed with biomass of strain 1B and nineteen different ILs in various conditions, revealing that tributyl(ethyl)phosphonium diethyl phosphate (IL#18), combined with ethyl acetate (EAc) as a co-solvent, presented the highest level of carotenoid extraction. Afterward, to better understand the process and optimize the extraction results, two experimental designs were performed, varying the amounts of IL#18 and EAc used. These allowed the establishment of 50 μL of IL#18 with 1125 μL of EAc, for 400 μL of biomass (cell suspension with about 36 g/L), as the ideal conditions to achieve maximal carotenoid extraction. Compared to the conventional extraction method using DMSO, this novel procedure eliminates the need for biomass drying, reduces extraction temperatures from 50 °C to 22 ± 2 °C, and increases carotenoid extraction by 264%, allowing a near-complete recovery of carotenoids contained in the biomass. These results highlight the great potential of ILs for bacterial carotenoid extraction, increasing the process efficiency, while potentially reducing energy consumption, related costs, and emissions.

Abstract:The co-production of lipids and high value-added products from microalgae is a key milestone for practical, microalgae-derived food and biofuel production. However, the use of ethanol as a solvent for lipid extraction requires a drying pretreatment to avoid wastewater contamination and large energy consumption during solvent recovery. Previous studies on lipid production have reported liquefied dimethyl ether (DME) as an alternative solvent that can extract lipids from high water content microalgae with low energy consumption by eliminating the traditional drying step. In this study, liquefied DME is used as a solvent to extract fucoxanthin, antioxidants, and lipids from wet diatom Chaetoceros simplex var. calcitrans . DME extracted 9.2 mg/g-dry microalgae of fucoxanthin from wet sample; this is slightly less than the 11.9 mg/g-dry microalgae extracted from dry sample by ethanol. The total phenolic content and antioxidant capacity of the liquefied DME extracts were only slightly lower than those of the ethanol extract. Using DME, all water and 22.7 wt% of lipid, and 5.6 wt% of atty acid were extracted. Lipids extracted with liquefied DME have a higher C/N, an indicator of suitability for conversion to biofuel, than lipids obtained by ethanol extraction. The microalga contains high levels of unsaturated fatty acids. Liquefied DME extraction is sustainable method because liquefied DME can extract most of the lipids and functional components without drying the diatom. DME avoids risks associated with ethanol.

Abstract:AIE based nanomaterials are progressively gaining momentum owing to their evolvement into an interdisciplinary field ranging from biomass and biomolecule yield to image‐guided photodynamic therapy. This study focuses on a novel strategy to enhance growth, lipid accumulation, and in vivo fluorescence visualisation in green microalgae Chlamydomonas reinhardtii using AIE nanoparticles to quantify radical changes. The absorption of AIE photosensitiser, TTMN was recorded from 420 to 570 nm with a peak at 500 nm, and the emission ranged from 550 to 800 nm with a peak at 650 nm. As a ROS molecule, H2O2 generation of TTMN in C. reinhardtii cells was detected with AIE nanoprobes TPE‐BO. H2O2 accumulation increased with the increase of TTMN concentrations. The maximum growth was observed at 10 μM TTMN‐exposed C. reinhardtii cells. Significant lipid accumulation was found in both 10 and 15 μM TTMN‐treated cells. For lipid visualisation, an AIE nanoprobe, 2‐DPAN was used, and superior fluorescence was determined and compared with the traditional BODIPY dye. Cytotoxicity analysis of 10 μM TTMN on the HaCat cell line with 86.2% cell viability revealed its high biocompatibility on living cells. This AIE‐based nanotechnology provides a novel approach for microalgae‐derived sustainable biomass and eco‐friendly biofuel production.

Interplay between interfacial behaviour, cell structure and shear enables biphasic lipid extraction from whole diatom cells (Navicula sp.)

Abstract:Hypothesis: Bacillariophyceae (i.e., diatoms) are an important class of algae with potential use in the production of proteins and lipids including long-chain ω-3 polyunsaturated fatty acids. Biphasic extraction of microalgae lipids using water-immiscible solvents such as hexane, can avoid the excessive energy required to distil solvents from water, but generally requires energy-intensive rupture of the cells. The unique cell structure and surface chemistry of diatoms compared to other microalgae species might allow biphasic lipid extraction without prior cell rupture. Experiments: The kinetics of biphasic lipid extraction from intact Navicula sp. cells was investigated during low-shear and high-shear mixing, and with prior or simultaneous application of ultrasound (20 kHz at 0.57 W/mL). Dynamic interfacial tension measurements and electron microscopic analysis were used to investigate lipid extraction in relation to interfacial behaviour and cell structure. Results: High yields (>80%) of intracellular lipids were extracted from intact cells over the course of hours upon low-shear contacting with hexane. The cells associated with and stabilised the hexane-water interface, allowing hexane to infiltrate pores in the frustule component of the cell walls and access the intracellular lipids. It was shown that mucilaginous extracellular polymeric substances (EPS) bound to the cell walls acted as a barrier to solvent penetration into the cells. This EPS could be removed by prior ultrasonication. Biphasic extraction was greatly accelerated by shear applied by rotor-stator mixing or ultrasound. High-shear could remove mucilaginous EPS from the cell surfaces to facilitate direct contact of the cell surface with hexane and produced smaller emulsion droplets with increased surface area. The combination of high-shear in the presence of hexane resulted in the in-situ rupture of the cells, which greatly accelerated lipid extraction and allowed high yields of neutral lipid (>95%) to be recovered from freshly harvested cells within less than 5 min. The study demonstrated the ability of shear to enable simultaneous cell rupture and lipid extraction from a diatom alga based on its cell structure and interfacial behaviour.

Abstract:Thraustochytrium sp. (T18) is a marine heterokont that exhibits a fast growth rate and accumulates a high proportion of polyunsaturated fatty acids (PUFAs), particularly docosahexaenoic acid (DHA), as essential compounds for health. The extraction of DHA-rich oil from marine microorganisms is challenging due to the difficulty in the substantial cost of lyophilization, the incompatibility of solvents with wet biomass, and the large volumes of organic solvents required by traditional extraction techniques. To address these concerns, ionic liquids (ILs) have been used to improve the oil yield from a wet oleaginous microorganism under mild conditions. In this study, a novel hydrophobic IL, 1-decyl-3-methyl-imidazolium bis(2,4,4-trimethylpentyl)phosphinate ([C10mim][( i C8)2PO2]) was first applied as pretreatment to aid in the extraction of DHA-rich oil from T18 biomass in the aqueous phase. A central composite design and response surface methodology were used to study the effect of different extraction variables on the oil yield (temperature, pH, and the mass ratio of IL to dried T18). The extraction conditions were numerically optimized to maximize the oil yield [69.4 ± 0.5% (w/w)] within the experimental range (the temperature of 76 °C, pH of 5.0, and mass ratio of 4.2). In addition, the extraction efficiency was up to 97.4 ± 0.4% of the total lipid content. The extracted oil contained 44.7 ± 0.2% DHA under 1 h of the pretreatment process. In this work, the proposed approach was found to be efficient and sustainable for the water-compatible recovery of a DHA-rich oil recovery process.

Optimizing light regimes for neutral lipid accumulation in Dunaliella salina MCC 43: a study on physiological status and carbon allocation

Abstract:Dunaliella salina is a favourable source of high lipid feedstock for biofuel and medicinal chemicals. Low biomass output from microalgae is a significant barrier to industrial-scale commercialisation. The current study aimed to determine how photosynthetic efficiency, carbon fixation, macromolecular synthesis, accumulation of neutral lipids, and antioxidative defence (ROS scavenging enzyme activities) of D. salina cells were affected by different light intensities (LI) (50, 100, 200, and 400 μmol m −2 s −1 ). The cells when exposed to strong light (400 μmol m −2 s −1 ) led to reduction in chlorophyll a but the carotenoid content increased by 19% in comparison to the control (LI 100). The amount of carbohydrate changed significantly under high light and in spite of stress inflicted on the cells by high irradiation, a considerable increase in activity of carbonic anhydrase and fixation rate of CO 2 were recorded, thus, preserving the biomass content. The high light exposed biomass when subjected to nitrogen-deficient medium led to increase in lipid content (59.92% of the dry cell weight). However, neutral lipid made up 78.26% of the total lipid while other lipids like phospholipid and glycolipid content decreased, showing that the lipid was redistributed in these cells under nitrogen deprivation, making the organism more appropriate for biodiesel/jet fuel use. Although D. salina cells had a relatively longer generation time (3.5 d) than other microalgal cells, an economic analysis concluded that the amount of carotenoid they produced and the quality of their lipids made them more suited for commercialization.

Abstract:In this research, waste lime sludge (LS) nanoparticles procured from integrated pulp and paper mill were used as the flocculent material to harvest the marine microalgae Tetraselmis indica (T. indica). LS contains 80–85% of calcium carbonate in the form of calcite with rhombohedral and scalenohedral structures along with other minerals such as Mg, K, Fe, and Na. XRD results showed that lime sludge particles have an averaged crystallite size of ~ 39 nm. The biomass harvesting efficiency of T. indica was observed at different doses of flocculent (0–160 mg L−1), temperature (30, 35, 40, 45, and 50 °C) and mixing rate (100–300 rpm), respectively. Maximum biomass harvesting of 86% was observed at 50 °C with 140 mg L−1 flocculent dose, pH 7, and 150 rpm in 90 min. It was also observed that LS has no effect on biodiesel profile of T. indica. Scanning electron microscopy (SEM) images revealed agglomeration of microalgal cells and deposition of calcium carbonate on its surface upon treatment with LS at 50 ºC. Experimental data were found to be in good agreement with pseudo-second-order kinetics model. This study indicated that waste lime sludge is a potential material for high harvesting efficiency in low settling time and low-cost harvesting of microalgae and it makes the production of biodiesel cost-effective.

Abstract:Microalgae are a kind of promising raw material for bioenergy production and carbon sequestration. To enhance cell growth and production efficiency, great efforts were made for exploring the appropriate intensity and quality of light to improve the synthesis of the value-added products. The aggregation-induced emission (AIE) material (Astrazon Brilliant Red 4G), active for converting blue-green light to red light (630–680 nm), was for the first time applied in the research to achieve high fatty acid yield with considerable carbon–neutral ability. After 10 days-incubation, it revealed that AIE light-converting films boosted the biomass of microalgae Scenedesmus obliquus , Phaeodactylum tricornutum , and Chlamydomonas reinhardtii by 13.00 %, 28.00 %, and 26.00 %, and the total lipid contents increased by 34.60 %, 17.74 %, and 22.63 %, with the carbon sequestration capacity raised by 17.18 %, 38.89 %, and 35.71 %. Furthermore, AIE light-converting films promoted the accumulation of unsaturated fatty acids (UFAs) in all three microalgae. Our study demonstrated the potential application of AIE light-converting films for cultivating microalgae to capture high carbon dioxide for photosynthesize carbon–neutral energy and high-valued active compounds.

Abstract:Liquid byproducts and organic wastes generated from dairy processing units contribute as the largest source of industrial food wastewater. Though bacteria-mediated treatment strategies are largely implemented, a more effective and innovative management system is needed of the hour. Thus, the current study involves the cultivation of centric diatoms, Chaetoceros gracilis , and Thalassiosira weissflogii in simulated dairy wastewater (SDWW) formulated using varying amounts of milk powder with artificial seawater f/ 2 media (ASW). The results revealed that cell density and biomass productivity were highest in the 2.5% SDWW treatment cultures of both the strains, the maximum being in C. gracilis (7.5 × 10 6 cells mL − 1 ; 21.1 mg L −1 day −1 ). Conversely, the total carotenoid, chrysolaminarin, and phenol content were negatively impacted by SDWW. However, a considerable enhancement in the total lipid content was reported in the 2.5% SDWW culture of both species. Furthermore, the fatty acid profiling revealed that though the total polyunsaturated fatty acid (PUFA) content was highest in the control setups, the total mono polyunsaturated fatty acid (MUFA) content was higher in the 5% SDWW setups (30.66% in C. gracilis and 33.21% in T. weissflogii ). In addition to it, in the cultures utilizing energy from external carbon sources provided by SDWW, the biodiesel produced was also enhanced owing to the heightened cetane number. Thus, the current study evidently highlights the organic carbon acquisition potential of marine diatoms with the scope of providing sustainable biorefinery.

Abstract:Efficient algal harvesting, cell pretreatment and lipid extraction are the major steps challenging the algal biofuel industrialization. To develop sustainable solutions for economically viable algal biofuels, our research aims at devising innovative reactive electrochemical membrane (REM) filtration systems for simultaneous algal harvesting and pretreatment for lipid extraction. The results in this work particularly demonstrated the use of the Ti4O7-based REM in algal pretreatment and the positive impacts on lipid extraction. After REM treatment, algal cells exhibited significant disruption in morphology and photosynthetic activity due to the anodic oxidation. Cell lysis was evidenced by the changes of fluorescent patterns of dissolved organic matter (DOM) in the treated algal suspension. The lipid extraction efficiency increased from 15.2 +/- 0.6 g-lipid g-algae(-1) for untreated algae to 23.4 +/- 0.7 g-lipid g-algae(-1) for treated algae (p < 0.05), which highlights the potential to couple algal harvesting with cell pretreatment in an integrated REM filtration process. Abstract:Abstract Background Microalgae are well-established feedstocks for applications ranging from biofuels to valuable pigments and therapeutic proteins. However, the low biomass productivity using commercially available growth mediums is a roadblock for its mass production. This work describes a strategy to boost algal biomass productivity by using an effective CO 2 supplement. Results In the present study, a novel nanoemulsion-based media has been tested for the growth of freshwater microalgae strain Chlorella pyrenoidosa . Two different nanoemulsion-based media were developed using 1% silicone oil nanoemulsion (1% SE) and 1% paraffin oil nanoemulsion (1% PE) supplemented in Blue-green 11 media (BG11). After 12 days of cultivation, biomass yield was found highest in 1% PE followed by 1% SE and control, i.e., 3.20, 2.75, and 1.03 g L −1 , respectively. The chlorophyll-a synthesis was improved by 76% in 1% SE and 53% in 1% PE compared with control. The respective microalgal cell numbers for 1% PE, 1% SE and control measured using the cell counter were 3.00 × 10 6 , 2.40 × 10 6 , and 1.34 × 10 6 cells mL −1 . The effective CO 2 absorption tendency of the emulsion was highlighted as the key mechanism for enhanced algal growth and biomass production. On the biochemical characterization of the produced biomass, it was found that the nanoemulsion-cultivated C. pyrenoidosa had increased lipid (1% PE = 26.80%, 1% SE = 23.60%) and carbohydrates (1% PE = 17.20%, 1% SE = 18.90%) content compared to the control (lipid = 18.05%, carbohydrates = 13.60%). Conclusions This study describes a novel nanoemulsion which potentially acts as an effective CO 2 supplement for microalgal growth media thereby increasing the growth of microalgal cells. Further, nanoemulsion-cultivated microalgal biomass depicts an increase in lipid and carbohydrate content. The approach provides high microalgal biomass productivity without altering morphological characteristics like cell shape and size as revealed by field emission scanning electron microscope (FESEM) images.
