Abstract:The improper management of wound exudate around the diabetic wound bed is one of the major factors leading to delay diabetic wound healing. However, the limited water absorption of single-layer conventional dressings unavoidably maintains excessive exudate in the diabetic wound bed. Herein, a novel diabetic wound dressing is reported for achieving rapid wound healing by electrospinning hydrophobic nanofibers on a hydrophilic modified non-woven fabric. The double-layer structure of the self-pumping dressing possesses unidirectional fluid transportation to divert excessive exudate away from diabetic wounds, ultimately promoting wound healing. The hydrophilic modified non-woven fabric provides water absorption and moisture retention to absorb excess exudate passing through the hydrophobic nanofiber, thereby preventing the wound bed from rewetting. Moreover, the formation of depressions on the hydrophobic nanofiber surfaces serves as water channels to further drain wound exudate from the wound bed. In vivo wound studies confirm that the self-pumping dressing exhibits excellent unidirectional wound exudate delivery and wound moisturization. Thus, diabetic wound healing shows a low inflammatory response and excellent wound healing quality. This self-pumping dressing exhibits clinical significance to design and fabricate a next-generation dressing for diabetic wound healing.

Abstract:The treatment of diabetic wounds remains a formidable clinical challenge worldwide. Because of persistent inflammatory reaction, excessive oxidative stress, cell dysfunction, poor blood microcirculation and other microvascular complications, diabetic wounds often fall into inflammatory circulation and are difficult to heal, making patients confront the risk of amputation. In this study, silver complex nanoparticles with Resina Draconis extract and Rhodiola rosea L. extract were loaded in situ onto thermoplastic polyurethane nanofibers to develop a multifunctional electrospun nanofiber wound dressing with excellent mechanical properties, superior water absorption and breathability, good coagulation promoting activity, strong antibacterial performance and antioxidant properties. This wound dressing could effectively enhance the migration and proliferation of fibroblasts, reduce the increased thickness of regenerated epidermis caused by diabetes, and the high expression and high lipid peroxidation levels of IL-1 β, IL-6, TNF α, iNOS and MMP-9, and raise the low expression of VEGF, which shows great potential to accelerate the wound healing of diabetic mouse models. The wound healing rate reached about 87.92%, close to the non-diabetic group. Our findings suggest a breakthrough in diabetic wound care, offering a viable solution to a long-standing medical shackle.

Abstract:Chronic wound healing remains a substantial clinical challenge. Current treatments are often either prohibitively expensive or insufficient in meeting the various requirements needed for effective diabetic wound healing. A 4D printing multifunctional hydrogel dressing is reported here, which aligns perfectly with wounds owning various complex shapes and depths, promoting both wound closure and tissue regeneration. The hydrogel is prepared via digital light process (DLP) 3D printing of the mixture containing N‐isopropylacrylamide (NIPAm), curcumin‐loaded Pluronic F127 micelles (Cur‐PF127), and poly(ethylene glycol) diacrylate‐dopamine (PEGDA575‐Do), a degradable crosslinker. The use of PEGDA575‐Do ensures tissue adhesion and degradability, and cur‐PF127 serves as an antibacterial agent. Moreover, the thermo‐responsive mainchains (i.e., polymerized NIPAm) enables the activation of wound contraction by body temperature. The features of the prepared hydrogel, including robust tissue adhesion, temperature‐responsive contraction, effective hemostasis, spectral antibacterial, biocompatibility, biodegradability, and inflammation regulation, contribute to accelerating diabetic wound healing in MRSA‐infected full‐thickness skin defect diabetic rat models and liver injury mouse models, highlighting the potential of this customizable, mechanobiological, and inflammation‐regulatory dressing to expedite wound healing in various clinical settings. Abstract:Diabetic foot ulcers (DFUs) are one of the most serious complications of diabetes, often leading to necrosis and amputation. DFU is caused by the intricate diabetic microenvironment, including ischemia, hypoxia, hyperinflammation, reduced angiogenesis, and persistent infection. Traditional wound dressings made of single or mixed materials often struggle to meet all the requirements for effective diabetic wound healing. In contrast, multilayer dressings comprising more than single layers have the potential to address these challenges by combining their diverse chemical and physical properties. In this study, we developed a bilayer hydrogel comprising a GelMA-ALG-nano-ZnO protective film and a COL1-PRP regenerative hydrogel for facilitating diabetic wound healing. We demonstrated the protective properties against bacterial infection of the protective film, while highlighting the regenerative potential of the COL1-PRP hydrogel in promoting fibroblast and MUVEC migration, extracellular matrix secretion and deposition, and angiogenesis. Importantly, the bilayer hydrogel exhibited superior efficacy in promoting full-thickness wound healing in a diabetic rat model compared to its single-layer hydrogel counterparts. This multi-layer approach offers a promising strategy for addressing the complexities of diabetic foot treatment and improving clinical outcomes.

Abstract:The proficient handling of diabetic wounds, a rising issue coinciding with the global escalation of diabetes cases, poses significant clinical difficulties. A range of biofunctional dressings have been engineered and produced to expedite the healing process of diabetic wounds. This study proposes a multifunctional hydrogel dressing for diabetic wound healing, which is composed of Polyvinyl Alcohol (PVA) and N1-(4-boronobenzyl)-N3-(4-boronophenyl)-N1, N1, N3, N3-teramethylpropane-1, 3-diaminium (TSPBA), and a dual-drug loaded Gelatin methacryloyl (GM) microgel. The GM microgel is loaded with sodium fusidate (SF) and nanoliposomes (LP) that contain metformin hydrochloride (MH). Notably, adhesive and self-healing properties the hydrogel enhance their therapeutic potential and ease of application. In vitro assessments indicate that SF-infused hydrogel can eliminate more than 98% of bacteria within 24 h and maintain a sustained release over 15 days. Additionally, MH incorporated within the hydrogel has demonstrated effective glucose level regulation for a duration exceeding 15 days. The hydrogel demonstrates a sustained ability to neutralize ROS throughout the entire healing process, predominantly by electron donation and sequestration. This multifunctional hydrogel dressing, which integrated biological functions of efficient bactericidal activity against both MSSA and MRSA strains, blood glucose modulation, and control of active oxygen levels, has successfully promoted the healing of diabetic wounds in rats in 14 days. The hydrogel dressing exhibited significant effectiveness in facilitating the healing process of diabetic wounds, highlighting its considerable promise for clinical translation.
Disposal of small quantities of radioactive materials in local landfills: problems and solutions.

Abstract:in modifying a Los Angeles waste disposal policy. As a result, nuclear medicine diagnostic patient waste was es sentially excluded from detection at Los Angeles landfills. In response to the Detroit area landfill problem, mem bers of the nuclear medicine community chose to work directly with the landfill owner and operators to achieve a detection level acceptable to both parties. The landfill owner allowed two certified medical physi cists to examine the monitoring equipment on-site. The landfill monitor (Bicron Landfill Monitor, Bicron Corp, Newbury, OH) consisted of two sodium iodide (lâ€•x1â€•) detectors mounted in lead sleeves on opposite sides of the entrance to the landfill property. Each was positioned to sample the trash container at a height of 7 ft above the roadbed. A total distance of 13 ft separated the detectors; however, the location of the detector on the driver's side permitted the trash container to come as close as 2 ft to the detector. Exempt quantities of â€ ̃33Ba (2.9 @sCi) and @Â°Co (1 .0 MCi) were measured in air between the two detectors. When either source was positioned within 2 ft of either detector, the alarm was triggered. During a return visit, the medical physicists examined the trigger level for the alarm. Background levels, moni tored with no sources of radioactive material present, ranged from 370 to 510 cpm. The threshold for detection set by the landfill owner was less than twice background (600 cpm). A meeting of all concerned parties was convened. The landfill operators had reviewed the company's procedure for landfill monitoring and found that the recommended distance between each detector and the side of the trash container was 4â€”5ft. The landfill management agreed to increase the distance between the trash container and detector on the driver's side to five feet. The level ofdetection selected by the landfill owner (i.e., 600 cpm) was a more difficult problem to resolve. Most states, including Michigan, do not have regulations for landfill disposal of small quantities of radioactive mate rials. Texas allows disposal of short-lived radionuclides in landfills (1). Abstract:A diabetic foot ulcer is a common high-risk complication in diabetic patients, but there is still no universal dressing for clinical treatment. In this study, a novel dual-functional sulfated galactofucan polysaccharide/poly(vinyl alcohol) hydrogel (DPH20) is developed during freeze-thaw cycles. Experimental results indicated that DPH20 had a high specific surface area, a dense porous structure, and a good swelling property, which could effectively adsorb the exudates and keep the wound moist. Furthermore, DPH20 exhibited remarkably recruited macrophage capability and accelerated the inflammation stage by improving the expression of the mRNA of , and in macrophages. DPH20 could promote cell migration and growth factor release to accelerate tube formation under hyperglycemic conditions in cell models of L929s and HUEVCs, respectively. Significantly, DPH20 accelerates the reconstruction of the full-thickness skin wound by accelerating the recruitment of macrophages, promoting angiogenesis, and releasing the growth factor in the diabetic mouse model. Collectively, DPH20 is a promising multifunctional dressing to reshape the damaged tissue environment and accelerate wound healing. This study provides an efficient strategy to repair and regenerate diabetic skin ulcers.
Enhancement of diabetic wound healing using a core-shell nanofiber platform with sequential antibacterial, angiogenic, and collagen deposition activities

Abstract:Infected diabetic wound (DW) healing entails a series of temporally and spatially regulated stages including antibacterial responses, revascularization, and collagen deposition, underscoring the need for the controlled sequential delivery of appropriate compounds when treating these wounds. Here, we designed a multifunctional core–shell CTS@PLCL/DWJM@Cu nanofiber loaded with three bioactive agents, including chitosan (CTS), copper (Cu), and decellularized Wharton’s jelly matrix (DWJM). We used this platform to facilitate regulated antibacterial, angiogenic, and collagen deposition activity in the context of DW healing. These CTS@PLCL/DWJM@Cu nanofibers exhibited satisfactory mechanical properties, excellent absorption, acceptable biodegradability, a high level of biocompatibility, elevated adhesive rates, and the ability to sequentially mediate controlled sequential CTS and Cu release. Further in vitro analyses revealed that the initial release of the CTS shell was sufficient to exert antibacterial activity, after which the sustained release of the Cu core promoted cellular migration, neovascularization, and enhanced angiogenesis-related gene expression. In vivo, DWJM further accelerated the deposition of collagen. Together, the sequential antibacterial, angiogenic, and collagen deposition activities synergistically enhanced DW healing. We thus posit that the core–shell CTS@PLCL/DWJM@Cu nanofiber holds great promise for use as a dressing material capable of promoting the appropriate stages of DW healing in a regulated, sequential manner.

Abstract:Bacterial infections and long-term inflammation cause serious secondary damage to chronic diabetic wounds and hinder the wound healing processes. Currently, multifunctional hydrogels have shown promising effects in chronic wound repair. However, traditional hydrogels only keep the wound moist and protect it from bacterial infection, and cannot provide mechanical force to contract the wound edges to achieve facilitated wound closure. Here, an asymmetric composite dressing was created by combining biaxially oriented nanofibers and hydrogel, inspired by the double-layer structure of the traditional Chinese medicinal plaster patch, for managing chronic wounds. Specifically, electrospun Poly-(lactic acid-co-trimethylene carbonate) (PLATMC) nanofibers and methacrylate gelatin (GelMa) hydrogel loaded with Epinecidin-1@chitosan (Epi-1@CS) nanoparticles are assembled as the temperature-responsive self-contracting nanofiber/hydrogel (TSNH) composite dressing. The substrate layer of PLATMC nanofibers combines topological morphology with material properties to drive wound closure through temperature-triggered contraction force. The functional layer of GelMa hydrogel is loaded with Epi-1@CS nanoparticles that combine satisfactory cytocompatibility, and antioxidant, anti-inflammatory, and antibacterial properties. Strikingly, in vivo , the TSNH dressing could regulate the diabetic wound microenvironment, thereby promoting collagen deposition, facilitating angiogenesis, and reducing the inflammatory response, which promotes the rapid healing of chronic wounds. This study highlights the potential of synergizing mechanical and biochemical signals in enhancing chronic wound treatment. Overall, this TSNH composite dressing is provided as a reliable approach to solving the long-standing problem of chronically infected wound healing.

Abstract:The intricate microenvironment of diabetic wounds characterized by hyperglycemia, intense oxidative stress, persistent bacterial infection and complex pH fluctuations hinders the healing process. Herein, an injectable multifunctional hydrogel (QPTx) was developed, which exhibited excellent mechanical performance and triple responsiveness to pH, temperature, and glucose due to dynamic covalent cross-linking involving dynamic Schiff base bonds and phenylboronate esters with phenylboronic-modified quaternized chitosan (QCS-PBA), polydopamine coated tunicate cellulose crystals (PDAn@TCNCs) and polyvinyl alcohol (PVA). Furthermore, the hydrogels can incorporate insulin (INS) drugs to adapt to the complex and variable wound environment in diabetic patients for on-demand drug release that promote diabetic wound healing. Based on various excellent properties of the colloidal materials, the hydrogels were evaluated for self-healing, rheological and mechanical properties, in vitro insulin response to pH/temperature/glucose release, antibacterial, antioxidant, tissue adhesion, coagulation, hemostasis in vivo and in vitro, and biocompatibility and biodegradability. By introducing PDAn@TCNCs particles, the hydrogel has photothermal antibacterial activity, enhanced adhesion and oxidation resistance. We further demonstrated that these hydrogel dressings significantly improved the healing process compared to commercial dressings (Tegaderm™) in full-layer skin defect models. All indicated that the glucose-responsive QPTx hydrogel platform has great potential for treating diabetic wounds.

Abstract:Smart hydrogel dressings capable of simultaneously highly effective antimicrobial, anti-inflammatory, and promoting re-epithelialization and angiogenesis are urgently needed for the management of diabetic wounds. Herein, a H 2 S-releasing multifunctional hydrogel was developed by utilizing the dynamic Schiff base reaction between carboxymethyl chitosan (CMC) and polyhexamethylene guanidine (PHMG)-modified aldehyde F108 (PFC). Diallyl trisulfide (DATS) was encapsulated into the PFC nanoparticles. Apart from possessing the essential properties necessary for an idealized hydrogel dressing, such as excellent injectability, tissue adhesion, self-healing, and stimulus–response degradation, the &CMC also utilized the synergistic effect of the PHMG and DATS to provide an efficient antimicrobial effect; the H 2 S was slowly released from the DATS under the action of GSH and exerted excellent anti-inflammatory effects, by inhibiting the expression of p-ERK and p-STAT3 in activated macrophages, and promoting macrophage polarization to the M2 phenotype. Strikingly, following the completion of the efficient antimicrobial and anti-inflammatory effects, the continuously generated H 2 S further significantly accelerated the proliferation, migration, and vascularization of endothelial cells by extending the activation of the p-p38 and p-ERK1/2. Owing to superior performances, &CMC significantly promoted the healing of diabetic wounds induced by streptozotocin with good biocompatibility. This study demonstrates that &CMC is a versatile hydrogel dressing with great potential for the management of diabetic wounds.

Abstract:Diabetic wound healing poses a substantial challenge owing to bacterial infections, insufficient angiogenesis, and excessive exudates. Currently, most of the clinical dressings used for diabetic wounds are still conventional dressings such as gauze. In this study, a three-layer Janus dressing was developed via continuous electrostatic spinning. The top-layer was composed of polylactic acid-glycolic acid and hydroxyapatite doped with silver ions and silicate. The hydrophobic top-layer prevented the adhesion of foreign bacteria. The mid-layer was composed of polyethylene glycol, polylactic acid-glycolic acid and hydroxyapatite doped with silver ions and silicate facilitated exudate absorption and bioactive ion release. The modified sub-layer containing polylactic acid-glycolic acid, hydroxyapatite doped with silver ions and silicate and sodium alginate microspheres enabled both the transport of wound exudate from the wound bed to dressing and the backflow of bioactive silver ions and silicate to the wound bed, thereby reducing infection and stimulating angiogenesis. Through in vivo and in vivo experiments, the Janus dressing showed to have antimicrobial, angiogenic, and exudate-control properties that accelerate healing in diabetic wounds. As a novel dressing, the multifunctional, self-pumping Janus wound dressing with bi-directional biofluidic transport offers a new approach to diabetic wound healing.
Astragaloside/PVP/PLA nanofiber functional dressing prepared by coaxial electrostatic spinning technology for promoting diabetic wound healing

Abstract:Diabetic wound is often a chronic wound with complex healing process and long healing time. It is evident that conventional dressings are inadequate in promoting the healing of diabetic wounds. In this study, a novel nanofiber dressing, produced through coaxial electrostatic spinning technology, was created to facilitate the healing of diabetic chronic wounds. This dressing was composed of Astragaloside (AS), polyvinylpyrrolidone (PVP) and polylactic acid (PLA). The morphology of the nanofiber dressing was characterized by scanning electron microscopy (SEM). Confirmation of the core–shell structure was done with the help of transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD). A pharmacodynamic study was conducted both in vivo and in vitro. It was shown that the nanofibers were approximately 604.83 ± 127.96 nm and with a core–shell structure. Additionally, in vitro drug release experiments revealed that the coaxial AS/PVP/PLA nanofiber functional dressings had a first rapid and then slow release property. Antioxidant activity experiment showed that the free radical scavenging rate of Astragaloside-loaded nanofibers reached 61 %. Mechanical tests also indicated that the nanofibers had an elastic modulus of 38.37 ± 1.81 MPa. Coaxial AS/PVP/PLA nanofiber functional dressings had good antimicrobial activity, and its diameter of the circle of inhibition against Staphylococcus aureus was 17.17 ± 2.04 mm, and its diameter of the circle of inhibition against Escherichia coli was 6.80 ± 0.78 mm. In addition, the findings of in vitro cellular tests showed that coaxial AS/PVP/PLA nanofiber functional dressings could promote the multiplication, adhesion and the migration ability of L929 fibroblasts with better biocompatibility. Finally, the wound healing rate of diabetic rats in the coaxial AS/PVP/PLA nanofiber functional dressing group was 96.54 ± 0.29 % on day 14. Consequently, the coaxial AS/PVP/PLA nanofiber functional dressings prepared by coaxial electrostatic spinning technique had satisfactory morphology, met the requirements of performance, and had the potential to be developed as a dressing for improving diabetic chronic wound healing.

We show that the beat amplitude at short times, in the absence of collisions, is well described by previously developed line strength theory for (1+1) laser induced fluorescence. The subsequent pressure dependent decay of the beat amplitude is used to extract depolarization rate constants and estimates of collisional depolarization cross sections. Depolarization accompanies both inelastic collisions, giving rise to rotational energy transfer, and elastic collisions, which change m(j) but conserve j. Previous experimental studies, as well as classical theory, suggest that elastic scattering contributes around 20% to the observed total depolarization rate at low j. Simulation of the experimental beat amplitudes, using theoretical calculations presented in the preceding paper, reveals that depolarization of OH(A) by Ar has a rate constant comparable to, if not larger than, that for energy transfer. This is consistent with a significant tilting or realignment of j(') away from j on collision. The experimental data are used to provide a detailed test of quantum mechanical and quasiclassical trajectory scattering calculations performed on a recently developed ab initio potential energy surface of Kłos et al. [J. Chem. Phys. 129, 054301 (2008)]. The calculations and simulations account well for the observed cross sections at high N, but underestimate the experimental results by between 10% and 20% at low N, possibly due to remaining inaccuracies in the potential energy surface or perhaps to limitations in the dynamical approximations made, particularly the freezing of the OH(A) bond.

Abstract:Smart hydrogel dressings capable of simultaneously highly effective antimicrobial, anti-inflammatory, and promoting re-epithelialization and angiogenesis are urgently needed for the management of diabetic wounds. Herein, a H2S-releasing multifunctional hydrogel was developed by utilizing the dynamic Schiff base reaction between carboxymethyl chitosan (CMC) and polyhexamethylene guanidine (PHMG)-modified aldehyde F108 (PFC). Diallyl trisulfide (DATS) was encapsulated into the PFC nanoparticles. Apart from possessing the essential properties necessary for an idealized hydrogel dressing, such as excellent injectability, tissue adhesion, self-healing, and stimulus–response degradation, the DATS@PFC&CMC also utilized the synergistic effect of the PHMG and DATS to provide an efficient antimicrobial effect; the H2S was slowly released from the DATS under the action of GSH and exerted excellent anti-inflammatory effects, by inhibiting the expression of p-ERK and p-STAT3 in activated macrophages, and promoting macrophage polarization to the M2 phenotype. Strikingly, following the completion of the efficient antimicrobial and anti-inflammatory effects, the continuously generated H2S further significantly accelerated the proliferation, migration, and vascularization of endothelial cells by extending the activation of the p-p38 and p-ERK1/2. Owing to superior performances, DATS@PFC&CMC significantly promoted the healing of diabetic wounds induced by streptozotocin with good biocompatibility. This study demonstrates that DATS@PFC&CMC is a versatile hydrogel dressing with great potential for the management of diabetic wounds.

Abstract:Multifunctional dressings are attracting increased interest in the area of tissue engineering and regenerative medicine. Wound healing is a dynamic and complex process that requires multidisciplinary management. However, dressings that exhibit both anti‐infective and tissue regenerative properties are still limited. Herein, a hydrogel‐based multifunctional dressing weaved by drug‐loaded magnetothermally responsive [Fe3O4@SiO2‐interpenetrating polymer network (IPN)] microfibers and human umbilical cord mesenchymal stem cell‐laden microfibers is developed. Magnetothermally responsive microfibers, as smart constructs, can release antibiotics on‐demand to prevent infection, due to the magnetothermal heating effect of the embedded magnetic nanoparticles and the thermoresponsive property of the IPN hydrogels. Furthermore, stem cell‐laden microfibers markedly promote cell proliferation and tissue remodeling, with improved re‐epithelization, granulation tissue formation, and collagen deposition at the wound site. Hydrogel‐based microfibers have the additional ability of absorbing exudate, providing a moist environment, and retaining the therapeutic cells. Therefore, functional microfibers and textiles significantly accelerate the healing process. An advanced dressing for wound healing, incorporating controlled drug delivery and stem cell‐based therapy is developed. Furthermore, the results of the present study reveal the potential of multifunctional dressings in tissue engineering and regenerative medicine. A novel multifunctional dressing composed of magnetothermally responsive microfibers and stem cell‐laden microfibers is developed for versatile and high‐quality wound healing. The thermal‐responsive microfibers facilitate on‐demand release of antibiotics. The stem cell‐laden microfibers improve re‐epithelization, granulation tissue formation, and collagen deposition at the wound site. As a result, these two microfibers synergistically enhance wound healing.

Abstract:Diabetic severe wounds are difficult to heal and regenerate, and are extremely challenging to manage in clinical practice with common wound dressings. There is a need to develop multifunctional dressings that meet the requirements for efficient treatment of diabetic wounds. Therefore, the nanocomposite hydrogel with oxidized chondroitin sulfate (OCS), carboxymethyl chitosan (CMC), and phenol red-modified ε-poly- l -lysine (EPL-PR) as the backbones, encapsulated with chondroitin sulfate-modified selenium nanoparticles () and infinite coordination polymer nanomedicine (ICPs) composed of procyanidins, berberine hydrochloride and Fe(III), was designed and fabricated. The prepared nanocomposite hydrogel system allows for rapid in situ gelation and complete coverage of irregular wounds. In vivo and in vitro investigations demonstrate that the nanocomposite hydrogel shows excellent tissue adhesion, antibacterial activities, biocompatibility and free radical scavenging properties. The rapid hemostasis of the nanocomposite hydrogel is confirmed through hemorrhage models of liver and caudal amputation. All the advantages enable the nanocomposite hydrogel to effectively induce skin tissue remodeling, promote vascular and hair follicle regeneration, remove free radicals from wounds, and accelerate wound healing. The nanocomposite hydrogel can also successfully monitor the pH range of 5–10, thus enabling real-time visual monitoring of the wound healing phase, facilitating wound management in clinical settings. This multifunctional wound dressing opens up ideas for the treatment and management of diabetic full-thickness wounds.

Abstract:Excessive exudate secreted from diabetic wounds often results in skin overhydration, severe infections, and secondary damage upon dressing changes. However, conventional wound dressings are difficult to synchronously realize the non-maceration of wound sites and rapid exudate transport due to their random porous structure. Herein, a self-pumping Janus hydrogel with aligned channels (JHA) composed of hydrophilic poly (ethylene glycol) diacrylate (PEGDA) hydrogel layer and hydrophobic polyurethane (PU)/graphene oxide (GO)/polytetrafluoroethylene (PTFE) layer is designed to rapidly export exudate and accelerate diabetic wound healing. In the design, the ice-templating process endows the hydrophilic hydrogel layer with superior liquid transport ability and mechanical strength due to the formation of aligned channel structure. The hydrophobic layer with controlled thickness functions as an effective barrier to prevent exudate from wetting the skin surface. Experiments in diabetic rat model show that JHA can significantly promote re-epithelialization and collagen deposition, shorten the inflammation phase, and accelerate wound healing. This unique JHA dressing may have great potential for real-life usage in clinical patients.

Abstract:Self-pumping dressings (SPDs) have been developed as a new type of effective material for the drainage of excessive wound exudates based on the structure of asymmetric wettability. However, current SPDs are easy to lose their asymmetric wettability due to the weak interfacial mechanical stability between the hydrophobic and hydrophilic layers. Herein, we report an integrated self-pumping organohydrogel dressing with aligned microchannels (SPD-AM), prepared by an ice-templating-assisted wetting-enabled-transfer (WET) polymerization strategy, that can accelerate the healing process of diabetic wounds. The WET polymerization strategy enables strong interfacial mechanical stability between the hydrophobic organogel and hydrophilic hydrogel layers. The aligned microchannels greatly improve the draining capability of SPDs. Taking a diabetic rat model as an example, the SPD-AM can significantly reduce the bacterial colonization with low inflammatory responses, enhance dermal remodeling by about 47.31%, and shorten wound closure time by about 1/5 compared with other dressings, ultimately accelerating diabetic wound healing. This study is valuable for developing next-generation SPDs with stable mechanical performance for clinical applications.

Abstract:Hydrogels are an important category of polymeric materials with physicochemical features such as moisturizability and biocompatibility that are ideal for developing dressings for diabetic wounds. However, conventional non-ionic hydrogel materials generally exhibit poor mechanical properties and poor adhesion, which compromise their ability to self-sustain in mechanically dynamic wound microenvironments. In this research, we developed a hybrid hydrogel as a highly biocompatible adhesive wound dressing that met the mechanical requirements of the skin to promote chronic wound healing in diabetic mouse models. A 7.5% (w/v) hydrogel corresponded to a Young’s modulus of 6.3 kPa. In vitro cell-based and subcutaneous implantation experiments in mice demonstrated the excellent biocompatibility and optimal biodegradability of hydrogel dressings. In a diabetic mouse splint wound model for evaluating wound healing in vivo, the hydrogel dressing showed robust adhesion to the wound and efficiently accommodated mechanical deformations around the wound, resulting in significantly improved healing rates of chronic diabetic wounds. Thus, our work illustrates a newly alternative strategy for the simple and efficacious treatment of chronic wounds in the context of diabetes care.
