Abstract:Introduction: Epstein-Barr virus (EBV) is a highly dangerous virus that is globally prevalent and closely linked to the development of nasopharyngeal cancer (NPC). Plasma EBV DNA analysis is an effective strategy for early detection, prognostication and monitoring of treatment response of NPC. Methods: Here, we present a novel molecular diagnostic technique termed EBV-MCDA-LFB, which integrates multiple cross displacement amplification (MCDA) with nanoparticle-based lateral flow (LFB) to enable simple, rapid and specific detection of EBV. In the EBV-MCDA-LFB system, a set of 10 primers was designed for rapidly amplifying the highly conserved tandem repeat BamHI-W region of the EBV genome. Subsequently, the LFB facilitate direct assay reading, eliminating the use of extra instruments and reagents. Results: The outcomes showed that the 65°C within 40 minutes was the optimal reaction setting for the EBV-MCDA system. The sensitivity of EBV-MCDA-LFB assay reached 7 copies per reaction when using EBV recombinant plasmid, and it showed 100% specificity without any cross-reactivity with other pathogens. The feasibility of the EBV-MCDA-LFB method for EBV detection was successfully validated by 49 clinical plasma samples. The complete detection process, consisting of rapid template extraction (15 minutes), MCDA reaction (65°C for 40 minutes), and LFB result reading (2 minutes), can be finalized within a 60-minutes duration. Discussion: EBV-MCDA-LFB assay designed here is a fast, extremely sensitive and specific technique for detecting EBV in field and at the point-of-care (PoC), which is especially beneficial for countries and regions with a high prevalence of the disease and limited economic resources.

Abstract:The quality of cellular products used in biological research can directly impact the ability to obtain accurate results. Epstein–Barr virus (EBV) is a latent virus that spreads extensively worldwide, and cell lines used in experiments may carry EBV and pose an infection risk. The presence of EBV in a single cell line can contaminate other cell lines used in the same laboratory, affecting experimental results. We developed three EBV detection systems: (1) a polymerase chain reaction (PCR)-based detection system, (2) a recombinase polymerase amplification (RPA)-based detection system, and (3) a combined RPA-lateral flow assay (LFA) detection system. The minimum EBV detection limits were 1 × 103 copy numbers for the RPA-based and RPA-LFA systems and 1 × 104 copy numbers for the PCR-based system. Both the PCR and RPA detection systems were applied to 192 cell lines, and the results were consistent with those obtained by the EBV assay methods specified in the pharmaceutical industry standards of the People's Republic of China. A total of 10 EBV-positive cell lines were identified. The combined RPA-LFA system is simple to operate, allowing for rapid result visualization. This system can be implemented in laboratories and cell banks as part of a daily quality control strategy to ensure cell quality and experimental safety and may represent a potential new technique for the rapid detection of EBV in clinical samples.

Abstract:Abstract Epstein-Barr virus (EBV) is a ubiquitous gamma herpesvirus that maintains a lifelong latent association with B lymphocytes. Here, a rapid and reliable diagnosis platform for detecting EBV infection, employing loop-mediated isothermal amplification (LAMP) combined with a gold nanoparticles–based lateral flow biosensors (AuNPs-LFB) (termed LAMP Amplification Mediated AuNPs-LFB Detection, LAMAD), was developed in the current study. A set of specific LAMP primers targeting the Epstein-Barr nuclear antigen (EBNA) leader protein ( EBNA-LP ) gene was designed and synthesized. Subsequently, these templates extracted from various pathogens and whole blood samples were used to optimize and evaluate the EBV-LAMAD assay. As a result, the limit of detection (LoD) of the EBV-LAMAD assay was 45 copies/reaction. The EBV-LAMAD assay can detect all representative EBV pathogens used in the study, and of note, no cross-reactions were observed with other non-EBV organisms. Moreover, the whole workflow of the EBV-LAMAD assay can be completed within 70 min, including rapid EBV template preparation, EBV-LAMP amplification, and AuNPs-LFB-mediated detection. Taken together, the EBV-LAMAD assay targeting the EBNA-LP gene is a rapid, simplified, sensitive, reliable, and easy-to-use detection protocol that can be used as a competitive potential diagnostic/screening tool for EBV infection in clinical settings, especially in basic laboratories in resource-limited regions. Performance evaluation of magnetic bead nucleic acid extraction in real-time fluorescence quantitative PCR detection system for plasma EBV-DNA

Abstract:Objective To evaluate the application performance of magnetic bead nucleic acid extraction method in realtime fluorescence quantitative PCR detection for plasma Epstein-Barr virus (EBV) DNA.Methods Automated magnetic bead method was used to extract nucleic acid,and combined real-time fluorescence quantitative PCR assays for quantitative EBV-DNA.The detection performance of the method was evaluated byusing the third-party reference materials.In addition,100 patients with NPC were selected,and the plasma EBV-DNA was quantitatively detected by magnetic beads method and boiling method.The correlation between the two methods was analyzed.Results In the automated magnetic bead method-based real-time fluorescence quantitative PCR detection system for EBV-DNA,the repeatability precision of the low,medium and high were all ＜3/5 TEa (TEa =target ± 0.4 lg),and the intermediate precision were all ＜ 4/5 Tea.In the range of 5.4×101-5.4×105 U/mL,there had good linear relationship.The minimum detection limit was about 3.334×101 U/mL.And the concentration of Hb interference in plasma was lower than 8.667 g/L,the results of magnetic bead test were not significantly affected.The detection results between magnetic bead method and boiling method were linearly related,and the qualitative coincidence rate was 83％.Conclusion The detection of plasma EBV-DNA by automated magnetic bead nucleic acid extraction method can achieve good detection performance.
Rapid and sensitive detection of Epstein-Barr virus antibodies in nasopharyngeal carcinoma by chemiluminescence strips based on iron-porphyrin single atom nanozyme

Abstract:The correlation between Epstein-Barr virus (EBV) infection and nasopharyngeal carcinoma (NPC) risk has been extensively researched. The continual monitoring of EBV-IgAs provides a promising approach of NPC screening in its early stage. In this study, we successfully synthesized a single-atom nanozyme (SANzyme) through the application of iron-porphyrin based metal organic framework (MOF-FeP). The MOF-FeP possesses precisely-defined electronic and geometric structures that accurately mimic highly-evolved catalytic site of natural peroxidase. The peroxidase-like activity of MOF-FeP enables it to catalyze the chemiluminescence of luminol substrate. By integrating MOF-FeP into a traditional strip, we created a rapid and highly-sensitive evaluation tool for detecting EBV-IgAs. Importantly, the MOF-FeP strip enables the simultaneous detection of three EBV-IgAs, greatly improving the accuracy of EBV-associated NPC screening. The sensitivities of the MOF-FeP strip (75.56%–93.30%) surpass those of current enzyme-linked immunosorbent assay (ELISA) methods (64.44%–82.22%). This test takes only 16 min to perform as opposed to the customary 1–2 h required for standard ELISA. Additionally, the MOF-FeP strip is suitable for whole blood samples, thereby significantly simplifying the sample preparation and detection process. In conclusion, the MOF-FeP strip combines the simplicity of traditional strip with the high catalytic activity of SANzyme. Our innovative MOF-FeP strip offers a new point-of-care strategy for EBV-IgAs detection, which is expected to markedly facilitate early screening for EBV-associated diseases.

Abstract:A method that uses specific oligonucleotide probes coupled to a specific array of fluorescent microspheres in multi-analyte suspension arrays was employed for the detection of common viruses, such as Herpes virus (HSV), Human papillomavirus (HPV) and Hepatitis B virus (HBV). Sixteen species-specific probes and 9 sets of specific primers were designed based on conserved sequences of these viruses in the GenBank database. Serial symmetric PCR, asymmetric PCR and multiple PCR assays were employed to evaluate the sensitivity, specificity and reproducibility of multi-analyte suspension arrays analyzed on a Luminex-100 analyzer instrument. The symmetric PCR amplification of four types of HSV, four types of HPV and HBV genotypes of B, C and D, combined with their corresponding species-specific probes and specificities were completely concordant with the results from a comparative sequence analyses. There was no significant difference in the median fluorescence intensity (MFI) value between symmetric PCR and asymmetric PCR when the viral DNA concentration was above 10(4)copies/test. Both PCR products were negative in the multi-analyte suspension arrays with viral DNA concentrations less than 10(3)copies/test. A multi-analyte suspension array is a flexible, high-throughput, relatively simple method for rapid identification of common viruses in the clinical laboratory.

Abstract:<p>Herein, we have proposed a soft metal-phenolic capsule (sMPC)-based electrochemical immunoassay for ultrasensitive detection of Epstein-Barr virus capsid antigen IgA (EBVCA-IgA), a biomarker of nasopharyngeal carcinoma. Metal probes with large size contain a number of metal ions, which are very beneficial to signal amplification for anodic stripping voltammetry; however, these probes easily precipitate due to their heavy weight, leading to low recognition efficiency and compromised performance. In this study, we demonstrate sMPCs fabricated by metal-coordination interactions exhibit unique surface behavior compared with their solid counterparts, which significantly enhance recognition efficiency and thus improve sensitivity despite of their micrometer size. Taking advantage of the sMPCs, the involved electrochemical immunoassay shows a much-improved sensitivity with an ultralow detection limit of 0.46 fM for EBVCA-IgA and can also be used in real sample analysis. So far as we know, this is the first report on a sMPC-based electrochemical strategy. Furthermore, it clarifies the potential effect of the rigidness of probes on the performance of an involved biosensor, which is meaningful to guide the design of other functional probes. The advantages of this method, including easy to fabrication, ultrasensitivity and good selectivity, ensure a promising potential in the point-of-care diagnostics of critical diseases.</p>

Abstract:Background: Infectious mononucleosis is the common clinical manifestation of primary Epstein-Barr virus (EBV) infection in young children. We evaluated a chemiluminescent immunoassay method for the determination of serum anti-viral capsid antigen IgM antibody and its clinical value in the diagnosis of infectious mononucleosis.Methods: Concentrations of the antibody in serum samples from 187 children measured by chemiluminescent immunoassay were compared with those measured by ELISA.Results: Assessment of technologic quality (methodology) in diagnostic tests demonstrated that sensitivity of CLIA was 0.64 U/ ml and the functional sensitivity was < 0.9 U/ml. The within-assay and the between-assay imprecisions of different concentrations were all < 5%. Recoveries were all in 93-107%. The linear regression equation between expected values and measured values was y=0.0967+1.0093x, correlation coefficient was 0.9996 (p < 0.0001). The ROC curve showed that the sensitivity and specificity of the CLIA both were > 90%. The area under the curve was 0.992, which was significantly higher than that of ELISA (p < 0.05).Conclusion: The CLIA was the excellent method for EBV-VCA IgM measurement at present and can improve the clinical diagnosis of infectious mononucleosis. UV-Assisted Hyperbranched Poly(β-amino Ester) Modification of a Silica Membrane for Two-Step Microfluidic DNA Extraction from Blood

Abstract:Integrating nucleic acid extraction in amplification-based point-of-care diagnostics will be a significant feature for next-generation point-of-care virus detection devices. However, extracting DNA efficiently on a microfluidic chip poses many technological and commercialization challenges, including manual steps, multiple instruments, pretreatment processes, and the use of organic solvents (ethanol, IPA) that inhibit detection, which is not viable with routine testing such as viral load monitoring of transplant patients for post-operative care. This paper presents a microfluidic system capable of two-step DNA extraction from blood using a UV-assisted hyperbranched poly(β-amino ester) (HPAE)-modified silica membrane for cytomegalovirus (CMV) detection in a rapid and instrument-free manner without the presence of amplification inhibitors. HPAEs of varying branch ratios were synthesized, screened, and coated on a silica membrane and bonded between two layers of poly(methyl methacrylate) (PMMA) substrates. Our system could selectively extract DNA from blood with an efficiency of 94% and a lower limit viral load of 300 IU/mL in 20 min. The extracted DNA was used as the template for real-time loop-mediated isothermal amplification (LAMP)-based detection of CMV and was found to produce a fluorescent signal intensity that was comparable with commercially extracted templates. This system can be integrated easily with a nucleic acid amplification system and used for routine rapid testing of viral load in patient blood samples.

Abstract:Enzyme-linked immunosorbent assay (ELISA) provides a convenient way for the detection of viral pathogens. However, conventional ELISA performed on mirowell plates suffers from poor sensitivity, laborious coating and complicated blocking procedures. Herein, we designed a sensitive colorimetric immunoassay by taking advantages of the enrichment and isolation ability of magnetic beads (MBs) and the high loading capacity of gold nanoparticles (AuNPs) for detecting respiratory syncytial virus (RSV) as a pathogen model. RSV was selectively captured and preconcentrated from samples with antibodies functionalized MBs, followed by binding with antibodies labeled AuNPs, which carrying a large amount of alkaline phosphatase (ALP) molecules for colorimetric signal amplification by catalyzing the dephosphorylation of non-colored pNPP to generate colored product pNP. After optimizing the experimental conditions based on the principle of low nonspecific signal, low cost, and high sensitivity, the analytical sensitivity of the developed immunoassay can be improved to 0.27 pg/mL, which is over sevenfold higher than that of commercially available RSV ELISA kits (2 pg/mL). In addition, the total assay time was less than 2.5 h without any pretreatment, which is much more rapid than other reported assays. Therefore, the proposed immunoassay holds great promise for the fabrication of rapid, sensitive, and economic method for the viral pathogen detection.

Abstract:A microfluidic chip was designed and fabricated to achieve visual and quantitative sensing for the biomarker of Epstein-barr virus antibody (EBV IgG) based on TMB chromogenic unit and Raman detection unit. The biosensor was composed of polystyrene/Au nanoparticles (PS/AuNPs) microsphere arrays on a quartz substrate, where the initial SERS signal was amplified ten times than pure AuNPs through synergetic coupling between the optical field of PS whispering gallery microcavity and plasmon of AuNPs. Meanwhile, we introduced a signal probe by assembling horseradish peroxidase with high catalytic activity to TMB and Raman reporter rhodamine 6 G, in which TMB turned from colorless to blue and gradually deepened with increasing amount of EBV IgG being captured while the unique fingerprint information of rhodamine 6 G provided the accurate Raman signal for point-of-care testing of target. In sum, an integrated microchip that allows visual prediction under the assistance of TMB color as well as SERS responses with a wide dynamic linear range of 10 -1 ∼10 5 pg·mL -1 and detection limit as low as 0.045 pg·mL -1 were obtained simultaneously, this lab-on-a-chip design offers a powerful model system for the early diagnosis of various cancer markers in peripheral blood.

Abstract:Immunoagglutination assay is a promising approach for the detection of waterborne analytes like virus, cells, proteins with its advantages such as a smaller amount of reagents and easier operation. This paper presents a microfluidic agglutination assay on which all the assay processes including analyte capture, agglutination, and detection are performed. The chip integrates an on-chip pump for sample loading, a dynamic magnetic bead (MB) clump for analyte capture and agglutination, and a sheath-less flow cytometry for particle detection, sizing, and counting. The chip is tested with streptavidin-coated MBs and biotinylated bovine serum albumin as a model assay, which realizes a limit of detection (LOD) of 1 pM. Then, an antigen/antibody assay using rabbit IgG and goat anti-rabbit IgG coated MBs is tested and a LOD of 5.5 pM is achieved. At last, human ferritin in 10% fetal bovine serum is tested with Ab-functionalized MBs and the detection achieves a LOD of 8.5 pM. The whole procedure takes only 10 min in total.

Abstract:Background Bead-based immunoassays typically require researchers to perform the analysis in a laboratory setting, involving complex processing procedures, most of the time, constant potential or current instruments are used to measure the bio-impedance change. However, almost all biological materials are decorated on the electrode surface to obtain the maximum sensitivity, resulting in difficulty to clean, separate, and other applications of fixed bead-based immunoassay. Result We present an innovative unfixed bead-based immunoassay technology that utilizes electrical impedance characteristics, offering the advantages of being rapid, portable, miniaturized, and cost-effective. This technique which is known as Impedance-enhanced electrochemical immunoassay (IE-ECIA) employs antibody-coated Magnetic beads (MBs) and Dielectric beads (DBs) linked to antigens for quantitative with miniaturization electronic devices, which has been successfully applied in an immune impedance sensor for detecting the Dengue virus (DENV), which is an affordable test with a low-cost microfluidic impedance biosensor combined with immune magnetic and dielectric micro/nanobeads for antigen detection. Highlight The proposed immunoassay technology introduces a novel microfluidic platform incorporating magnetic and dielectric beads alongside with an electrochemical impedance biosensor, therefore facilitating continuous-fluid quantitative analysis. This innovative and unconventional approach leverages free-motion multiplex beads housing magnetic and dielectric bead-bound antigens within a simple linear microchannel of microfluidic chip architecture, making the high-through measurements more efficient, enhanced Electrochemical immunoassay (ECIA) signals of Bio-on-magnetic-beads (BOMB). Abstract:Herein, a spatial confined enzyme-linked immunoassay (ELIA) is developed for ultrasensitive biomarker detection using a microfluidic magnetic delivery (μMD) disc. In μMD-ELIA, magnetic immunocomplexes are delivered into microchannel and trapping/preconcentration in the microchamber of μMD disc for fluorescence detection. The microchamber offers a confined space both for effective magnetic preconcentration and accelerated concentration of enzyme products. After μMD disc optimization, the assay performances of μMD-ELIA have been evaluated by model targets of human monocyte differentiation antigen cluster of differentiation14 (CD14) and interleukin (IL)–8. The limits of detection (LOD) of μMD-ELIA for CD14 and IL-8 are 0.17 pg/mL and 0.012 pg/mL, respectively, which corresponded to an over 300-fold LOD sensitivity enhancement compared to their standard ELISA results. In human serum analysis, good accuracy is obtained on both CD14 and IL-8 when compared to the results of ELISA kits, and satisfied recoveries of 81.3–112 % with RSDs of 1.0–9.7 % for IL-8 were obtained. Moreover, the μMD-ELIA is than applied to IL-6 concentration variation monitor in rat plasma during the progression of inflammatory reaction successfully, which further confirmed that the μMD-ELIA has enormous potential in disease early diagnosis and curative effect monitoring.

Abstract:Viral infections are major causes of mortality in solid-organ and hematopoietic stem cell transplant recipients. Epstein-Barr virus (EBV) and Parvovirus B19 (B19V) are among the common viral infections after transplantation and were recommended for increased screening in relevant guidelines. Therefore, the development of rapid, specific, and cost-effective diagnostic methods for EBV and B19V is of paramount importance. We applied Fluorescence of Loop Primer Upon Self-Dequenching Loop-mediated Isothermal Amplification (FLOS-LAMP) for the first time to develop a novel multiplex assay for the detection of EBV and B19V; the fluorophore attached to the probe are self-quenched in unbound state. After binding to the dumbbell-shaped DNA target, the fluorophore is dequenched, resulting in fluorescence development. The novel multiplex FLOS-LAMP assay was optimized by testing various ratios of primer sets. This novel assay, with great specificity, did not cross-react with the common virus. For the detection of EBV and B19V, the limits of detection could reach 969 and 798 copies/μL, respectively, and the assay could be completed within 25 min. Applying this novel assay to detect 200 clinical transplant individuals indicated that the novel assay had high specificity and good sensitivity. We developed multiplex FLOS-LAMP assay for the detection of EBV and B19V, which has the potential to become an important tool for clinical transplant patient screening.
A Rapid Immunochromatographic Method Based on a Secondary Antibody-Labelled Magnetic Nanoprobe for the Detection of Hepatitis B preS2 Surface Antigen

Abstract:Hepatitis B is a globally prevalent viral infectious disease caused by the hepatitis B virus (HBV). In this study, an immunochromatographic assay (ICA) for the rapid detection of hepatitis B preS2 antigen (preS2Ag) was established. The magnetic nanoparticles (MNPs) indirectly labelled with goat anti-mouse (GAM) secondary antibody were applied as a nanoprobe for free preS2 antibody (preS2Ab) capturing and signal amplification. By employing sample pre-incubation processing as well, preS2Ag-preS2Ab was sufficiently caught by the GAM-MNPs probe in 5 min. A qualitative sensitivity of 625 ng/mL was obtained by naked-eye observation within 15–20 min. A standard curve (0–5000 ng/mL) was established, with a quantitative limit of detection (LOD) of 3.6 ng/mL, based on the stability and penetrability of the magnetic signal characteristics. The proposed method for preS2Ag was rapid (~25 min, cf. ELISA ~4 h) and had a good accuracy, which was verified using an ELISA kit (relative error < 15%). Large equipment and skilled technicians were not required. The sensitivity and specificity of the developed GAM-MNPs-ICA method were 93.3% and 90% in clinical serum samples (n = 25), respectively. A good detection consistency (84%) was observed between the developed ICA method and 2 types of commercial ELISA kits, indicating that the GAM-MNPs-ICA has a potential application in large-scale screening for and point-of-care diagnosis of hepatitis B or other infectious diseases.
Evaluation of Performance and Application of Three Nucleic Acid Extraction Methods for Quantification of Plasma Epstein-Barr Virus DNA

Abstract:Objective To evaluate and compare the analytical performances and application values of three nucleic acid extraction methods for quantification of plasma Epstein-Barr Virus ( EBV ) DNA. Methods It used silica membrane spin column , boiling and automated magnetic bead method to extract viral nucleic acid in parallel , and combined real-time fluorescence quantitative PCR assays for quantitative EBV-DNA quantification.The performances of three methods were determined and compared by using the third-party reference materials , and the clinical values were analyzed by pairing detecting 100 NPC patients and 100 healthy subjects in pair .Results The accuracy and imprecision of three methods were all in line with requirements , and the results of clinical samples were linearly correlated . But actually the reproducibility and intermediate imprecision of the magnetic bead method were smaller and stable than those of the spin column method and the boiling method ( all <3％);the limit of detection for the magnetic bead method was 3.334 ×101 IU/ml, better than that of spin column method (4.159 ×101 IU/ml) and boiling method (8.511 ×101 IU/ml);the linear range of the magnetic bead method was 5.4 ×101 -5.4 ×105 IU/ml, slightly wider than that of the boiling method (5.4 ×102 -5.4 ×105 IU/ml); the ability of anti -Hb interference ability of magnetic bead method is better than that of boiling method ;and the positive rate and the mean viral load of the NPC samples measured with the magnetic bead method were significantly higher (95％, 8.342 ×103 IU/ml) than those measured with the spin column method (84％, 4.707 ×103 IU/ml) and the boiling method (78％, 2.571 ×103 IU/ml) ( P all<0.05).Conclusion The automated magnetic bead nucleic acid extraction method offered better analytical performance and higher clinical value for EBV DNA quantification in plasma .
Magnetic-based Microfluidic Chip: A Powerful Tool for Pathogen Detection and Affinity Reagents Selection

Abstract:The global outbreak of pathogen diseases has brought a huge risk to human lives and social development. Rapid diagnosis is the key strategy to fight against pathogen diseases. Development of detection methods and discovery of related affinity reagents are important parts of pathogen diagnosis. Conventional detection methods and affinity reagents discovery have some problems including much reagent consumption and labor intensity. Magnetic-based microfluidic chip integrates the unique advantages of magnetism and microfluidic technology, improving a powerful tool for pathogen detection and their affinity reagent discovery. This review provides a summary about the summary of pathogen detection through magnetic-based microfluidic chip, which refers to the pathogen nucleic acid detection (including extraction, amplification and signal acquisition), pathogen proteins and antibodies detection. Meanwhile, affinity reagents are served as the critical tool to specially capture pathogens. New affinity reagents are discovered to further facilitate the pathogen diagnosis. Microfluidic technology has also emerged as a powerful tool for affinity reagents discovery. Thus, this review further introduced the selection progress of aptamer as next generation affinity through the magnetic-based microfluidic technology. Using this selection technology shows great potential to improve selection performance, including integration and highly efficient selection. Finally, an outlook is given on how this field will develop on the basis of ongoing pathogen challenges.
A novel and efficient method for synthesizing magnetic PS-PMMA@Fe3O4 microspheres for protein separation and detection

Abstract:Immunoassay is the most widely used detection technique in clinical testing. Compared with the traditional enzyme-linked immunoassay, the chemiluminescence immunoassay system based on carboxylated magnetic beads as the separation tool is more advantageous, which can rapidly separate proteins and achieve the purpose of quantitative detection of proteins. Separation tools in chemiluminescence immunoassay techniques are key and the focus of research. However, the domestic technology of preparing carboxylated magnetic beads is still immature, and the market is monopolized by imported products, which is not conducive to the development of domestic chemiluminescence immunoassay technology. Based on this, we propose a simple and convenient new method for the preparation of magnetic microbeads. Firstly, styrene-methyl methacrylate microspheres were polymerized by dispersion polymerization and hydrolyzed to form carboxylated microspheres, then carboxylated microspheres were introduced in the process of classical coprecipitation reaction to synthesize magnetic microbeads, and magnetic microbeads with different magnetic contents were prepared and characterized. The separation effect was then tested by a fully automated chemiluminescence immunoassay analyzer, and it was found that carboxylated magnetic beads with a magnetic content of 20% were the most effective in separating proteins, and the coefficient of variation was as low as 3.41%, with a stable and reproducible performance. The chemiluminescence immunoassay technique can separate proteins in a short period of time with a very small amount of carboxylated magnetic microbeads, which is fast and efficient and will help in the early diagnosis of diseases in healthcare facilities and may be a better point-of-care assay.

Abstract:A simple and sensitive method has been proposed to determine a trace level of α-fetoprotein (AFP), hepatocellular carcinoma biomarker, using poly(methyl methacrylate) (PMMA) microfluidic chips coupled with electrochemical detection system. The PMMA microchannels have been modified with poly(ethyleneimine) (PEI) containing abundant NH2 groups to covalently immobilize AFP monoclonal antibody. Afterward, the antigen AFP and horseradish peroxidase (HRP)-conjugated AFP antibody can sequentially bind through antigen–antibody specific interaction. Atomic force microscopy (AFM) and confocal fluorescence microscope (CFFM) were utilized to characterize the surface topography and protein immobilization after modification. Coupled with three-electrode electrochemical detection system, the immunochip can perform the detection limit of AFP down to 1pgmL−1, and achieve a detectable linear concentration range of 1–500pgmL−1 by differential pulse voltammetry (DPV). The on-chip immunoassay platform can not only provide rapid and sensitive detection for target proteins but also be resistant to non-specific adsorption of proteins, which contributes to the detection of low-level protein in real sample. Finally, AFP existing in healthy human serum was detected to demonstrate the utility of the immunochip. The result shows that the proposed approach is feasible and has the potential application in clinical analysis and diagnosis.
