Abstract:BACKGROUND:Moringa oleifera Lam (MO) is native to India and is a cash crop widely cultivated in tropical and sub-tropical areas. The health improving properties of MO has been studied from a long time ago for the numerous phenolic compounds, including vitamins, flavonoids, phenolic acids, isothiocyanates, tannins and saponins, which are present in considerable amounts in the plant. A growing spectrum of therapeutic characteristics of MO leaves has been found and used in the remission or treatment of oxidative stress, liver disease, neurological disease, hyperglycemia and cancer.HYPOTHESIS:This review focused on researches applying MO or MO leaf extract as a functional food or cure against various disease and cellular injuries. We believed it would help the discovery of therapeutic application of MO and understanding of MO phytochemistry.METHODS:The data collected in this review were extracted from researches indexed in Web of Science, google scholar, PubMed, Science Direct and Scopus to find out health benefits and biological activities of MO leaves polyphenols. The studies reporting mechanistic route of phenolic compounds of MO leaves were also considered in the present study.RESULTS:It has been reported that polyphenols of MO leaf have protective characteristics against neurodegenerative disorders through reducing DNA damage, activation of AchE activity and inhibition of caspase-3 activity. It has been reported that, they protected the kidney from damage caused by melamine through suppressed the pro-inflammatory cytokine, metallopeptidase inhibitor 1 (TIMP-1), and kidney injury molecule 1 (KIM-1). Similarly, methanol extract of MO leaves has low hypoglycemic attributes and attenuate the risk of diabetes caused by alloxan by enhancing lipid metabolism and stimulating insulin release, glucose uptake, and glycogen synthesis. In addition, MO leaves are becoming the best phytomedicine to reduce hypertension, which are naturally known as angiotensin-1converting enzyme (ACE), acetylcholinesterase, arginase and phosphodiesterase 5 (PDE5) inhibitors.CONCLUSION:MO leaves extract as a health promoting food additives for human and animals due to its great protective effect against many diseases and the widely persistent environmental toxins which disrupted cellular metabolic function. More studies are required to use the phenolic compounds of MO leaves to develop and produce drugs for controlling and treatment of various diseases.
Supplementation with Moringa oleifera Lam leaf and seed flour during the pregnancy and lactation period of Wistar rats: Maternal evaluation of initial and adult neurobehavioral development of the rat progeny

Abstract:Ethnopharmacological relevance Moringa oleifera Lam. ( M. oleifera) is a tree species of Indian origin popularly known as the "tree of life". In various cultures, it is used by pregnant women to increase milk production, yet studies on its effects during pregnancy and lactation are lacking. Aim of the study To evaluate the nutraceutical aspects of flours produced from the leaves and seeds of M. oleifera , and to evaluate the effect of supplementation of pregnant Wistar rats during the gestation and lactation period, with the aim of studying the weight gain and neonatal parameters of the pregnant rats, as well as effects on the neurobehavioral development and memory in their offspring. Materials and methods The flour supplementation was conducted at a concentration of 100 mg per kg of animal body weight. For the memory tests, the Open Field Habituation test was performed and repeated after seven days. The Object Recognition test was conducted with the animal exposed to the open field in short and long familiarization sessions. The data obtained were analyzed using Kruskal-Wallis tests for non-parametric data and one-way and two-way ANOVA for parametric data. Results Flour produced from both the leaf and seed of M. oleifera was found to contain significant amounts of nutrients (protein, fibre, carbohydrates, etc.), making them suitable for supplementation. The exposure of pregnant rats to M. oleifera leaf and seed flours did not affect weight gain, did not have harmful effects on the birth of offspring, and did not result in abortions or mutations in the offspring. Regarding the supplemented group's offspring, early maturation of the senses in the offspring compared to the control group was observed in all tests were conducted; indicating that supplementation positively impacted cognitive development. Further, the offspring of the supplemented rats presented reduced locomotion and greater exploration of new objects compared to the control group offspring, indicating positive effects on learning. Conclusion This study describes for the first time the beneficial effects on pregnant Wistar rats and their offspring of maternal supplementation with flour products from the leaves and seeds of M. oleifera .

Abstract:Moringa oleifera (M. oleifera) Lam belongs to the family Moringaceae. It is an important multipurpose tree that is largely distributed globally and has been used almost in every aspect of traditional medicine for the treatment of various illnesses including cancers, diabetes mellitus, asthma, arthritis, etc. This study investigated the effects of oral acute and sub-acute administration of M. oleifera hydroethanolic leaf extract (MOHE) in ICR-mice. Its major phenolic compounds were also determined. Ten (10) female, 8-week old mice were grouped into control and treatment groups for acute toxicity study. A dose of 2000 mg/kg MOHE was given once to the treatment group via oral gavage. However, for the sub-acute toxicity study, 25 mice were grouped into groups A (control), B (125 mg/kg), C (250 mg/kg), D (500 mg/kg) and E (1000 mg/kg). MOHE was given via oral gavage to groups B, C, D and E daily for 28 days. Group A received only distilled water. The mice were sacrificed at the end of the experiments and samples were collected for evaluation. The results of the chemical profiling of MOHE revealed the presence of glucomoringin, niaziminine, quercetin and kaempferol as the major compounds. The treated mice in the acute toxicity study were slightly anaemic and showed evidence of stress leukogram. Moreover, a slight increase in creatinine, significant increases in AST and CK, hepatic degeneration and necrosis, none-obstructive sinusoidal dilatation, renal tubular necrosis, interstitial nephritis and renal interstitial oedema were observed. It is concluded that the LD 50 of MOHE is higher than 2000 mg/kg. However, oral administration of MOHE causes acute mild anaemia and moderate hepato-nephrotoxicity in ICR-mice. Its major phenolic compounds are glucomoringin, niaziminine, quercetin and kaempferol.

Abstract:Abstract: The endothelium modulates vascular tone by producing vasodilator andvasoconstrictor substances. Of these, the best characterized and potentially most important are nitric oxide (NO•) and O2−•. These small molecules exhibit opposing effects on vascular tone and chemically react with each other in a fashion that negates their individual effects and leads to the production of potentially toxic substances, such as peroxynitrite (ONOO−). These dynamic interactions may likely have important implications, altering not only tissue perfusion but also contributing to the process of atherosclerosis. The precise O2−• source within vascular tissue remains to be determined. Recent work demonstrated that in endothelial cells as well as in vascular smooth muscle cells, a membrane‐associated NAD(P)H‐dependent oxidase represents the most significant O2−• source. Interestingly, this oxidase is activated upon stimulation with angiotensin II, suggesting that under all conditions of an activated circulating and/or local renin‐angiotensin system endothelial dysfunction secondary to increased vascular O2−• production is expected.

Abstract:The authors of this brief interviewed stakeholders in states with high-ranking and low-ranking health system performance, according to The Commonwealth Fund's State Scorecard on Health System Performance. Findings suggest there are market, political, and cultural characteristics that can help or hinder health system improvement. High-performing states are more likely to have: (1) a history of continuous reform and government leadership; (2) a culture of collaboration among stakeholders; transparency of price and quality information; and (3) a congruent set of policies that focus on system improvement. Regardless of starting point, state policymakers and proponents for health system improvement can work to align incentives to change provider, health plan, purchaser, and consumer behavior; frame health in terms of economic development to gain public and political support; engage purchasers and payers to drive value and quality improvement; bring stakeholders together to develop goals and build trust; and take advantage of federal funding, incentives, and reform opportunities.

Abstract:Aim: The study was designed to investigate antioxidant, survivability, fecundity, and locomotor activity of Moringa oleifera leaf (MOL) extract in Drosophila melanogaster. Materials and Methods: The study was conducted at the Africa Centre of Excellence in phytomedicine Research and Development (ACEPRD), University of Jos, Nigeria, in August 2019 - March 2020. In the first place, in vitro analysis of the antioxidant activity of extracts of M. oleifera in various solvents (Aqueous, Methanol-Aqueous co-solvent (80 % v/v) and n-Hexane) were evaluated using DPPH (1,1-Diphenyl-2-Picrylhydrazyl) assay. Based on the in vitro result, the methanol extract with the best free radical scavenging activity was used in graded doses for conducting the in vivo studies, and the observations were recorded. Distilled water (1000 μL) was used alone in 10 g fly food (as negative control) or as a solvent to dissolve MOL extract or ascorbic acid (positive control) separately before mixing with the fly food. Statistical significance was taken at P0.05) improved movement, significantly (P<0.05) increased survivability, fecundity, and total thiol level. The activities of glutathione-S-transferase (GST) and catalase (CAT) significantly (P 0.05) decreased, while malondialdehyde (MDA) concentration decreased significantly (P<0.05) compared with controls, respectively. Conclusion: In vitro study suggested better antioxidant activity of MEMOL. In vivo study also revealed that MEMOL was relatively safe in D. melanogaster, supported by high LD50, increased survivability, fecundity, locomotor ability, antioxidant enzyme activities, total thiol level, along with a concomitant decrease in MDA content.
Anti-anaemic effects of Moringa oleifera seeds (Lam) in protein energy malnourished rats

Abstract:The effects of defatted Moringa oleifera seed (L) on haemotological indices and body weight was evaluated in rat model of Protein Energy Malnutrition (PEM). Protein Energy Malnutrition is associated with muscle degeneration and anaemia. Protein Energy Malnutrition (PEM) was induced in fifteen (15) rats by feeding them with low protein diet (3% fish meal protein) and water for twenty eight days. Five (5) rats served as positive control (Group A) received commercial feed containing 21% crude protein. The PEM rats were assigned to three groups namely B, C and D. Group B and C were fed with recovery diets ad libitum for another twenty eight days. Group B received feed containing 20% Fishmeal protein and Group C received feed containing 20% defatted M. oleifera seed. Group D were Protein-energy malnourished rats maintained on protein deficient diet (3% fish meal protein). Protein energy malnourished rats had significantly reduced (p<0.05) body weight and haematological indices with exception of the Mean Capsular Volume (MCV) and Mean Capsular Haemoglobin (MCH) which significantly (p< 0.05) increased. Phenotypical examination revealed that PEM animals had characteristic typical of catabolic states, which include, retarded growth, loss of hair, scaly tail etc. The recovery diets containing 20% fishmeal protein or 20% M. oleifera in place of 3% fishmeal caused significant (p<0.05) increase in the Haematological Indices (with exception of MCV and MCH which decreased significantly) and body weight when compared with the malnourished group D. The study support the claims that PEM can cause muscle degeneration and disturbance to haematological parameters. Furthermore, supplementation of diet with M. oleifera seeds ameliorate muscle degeneration and anaemia in PEM rats comparatively similar to 20% fish meal.

Abstract:Moringa oleifera (MO) is an excellent source of dietary antioxidant. MO is used traditionally to enhance libido and as an aphrodisiac in the treatment of sexual dysfunction. This study aimed to investigate the direct effect of aqueous leaf extract of MO on Leydig cell in vitro. Specifically, the effect of MO on viability, testosterone production, antioxidant activity and lipid peroxidation on TM3 cells were evaluated. TM3 cells seeded for 24 hr were exposed to aqueous leaf extract of MO (0, 10, 50, 100, 250, 500 and 1,000 µg/ml) for 24 hr, in the absence or presence of human chorionic gonadotropin (hCG; 6 mIU/200 µl). Cell viability remained unchanged while testosterone production significantly increased at 500 and 1,000 µg/ml of the extract under stimulatory conditions by 34 and 45% respectively. Glutathione level substantially increased at 250 µg/ml, while lipid peroxidation, catalase and superoxide dismutase activity, and total antioxidant capacity remained unchanged. Our results demonstrate the androgenic effect of MO especially at high concentrations in TM3 cells. The androgenic effect may be attributed to its antioxidant enzyme activities.

Abstract:Maternal endotoxemia disturbs the intrauterine environment, impairs nephrogenesis, and increases the risk of hypertension and kidney disease in adulthood. Here, it was investigated whether maternal treatment with the water extract of Moringa oleifera seeds (WEMoS) or the water-soluble M. oleifera seed lectin (WSMoL) prevents the oxidative stress induced by lipopolysaccharide (LPS) in pregnant rats, and the renal injury and hypertension in the adult offspring. The administration of WEMoS or WSMoL prevented the stimulatory effects of LPS on lipid peroxidation in the maternal-placenta-fetuses environment. The impact of WEMoS was linked to decreased superoxide anions production in the placenta. The effects of WSMoL were parallel to the inhibition of superoxide anion production and NADPH oxidase activity. The WSMoL also prevented increased NADPH oxidase activity in the fetal kidney. The LPS offspring presented higher systolic blood pressure (SBP) and increased lipid peroxidation, reactive oxygen species (ROS), NADPH oxidase activity, and nitrate/nitrite in the kidney; the maternal treatment with WEMoS and WSMoL prevented these changes. In conclusion, the present study demonstrates that WEMoS and WSMoL have protective effects on maternal endotoxemia, which involve antioxidant and anti-inflammatory actions that prevent the programming of hypertension.
The ameliorative impacts of Moringa oleifera leaf extract against oxidative stress and methotrexate-induced hepato-renal dysfunction

Abstract:Moringa Oleifera (MO) is a herbal plant native to South Asia known for its anti-oxidative and anti-inflammatory properties. This study explored the protective effects of MO leaf extract (MOLE) against oxidative stress and hepatic and renal injuries caused by methotrexate (MTX) therapy. Mice received a single intraperitoneal injection of 20 mg/kg body weight MTX to induce hepatic and kidney injuries. They then received 300 mg/kg body weight of MOLE orally for seven days, followed by MTX on day 7 then five more days of MOLE (12 days total). Blood, liver and kidney samples were collected from all groups and the following biochemical parameters were tested: serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), catalase, superoxide dismutase (SOD), malondialdehyde (MDA) and total proteins. Quantitative real time PCR (qRT-PCR) was used to examine Nrf2, HO-1, BAX, TIMP, XIAP, and NFkB, which are associated with apoptosis, anti-apoptosis and oxidative stress. Capase-9 and Bcl2 genes underwent immunohistochemical analysis. Pretreatment with MOLE reduced the effect of MTX on ALT, AST and total proteins, and reversed its effect on serum and tissue antioxidants. Nrf2/HO-1, apoptotic and anti-apoptotic gene expression was regulated, and Bax and TIMP were reduced; XIAP expression was increased in both the liver and kidney samples, and immunoreactivity of caspase-9 and Bcl2 was restored in the MOLE-administered experimental group. Overall, the study concluded that MOLE can inhibit the effects of hepato-renal injuries caused by MTX by regulating oxidative stress, apoptosis and anti-apoptotic genes at biochemical, molecular and cellular levels.
Reproductive outcome in female wistar rats treated with nhexane, dichloromethane and aqueous ethanol extracts of Cucurbita pepo seed

Abstract:In developing countries, healthcare challenges and expensive infertility treatments has resulted in resurgent interest in medicinal plants. This study was designed to determine if Curcubita pepo seed can enhance female fertility, by assessing the reproductive outcome in female wistar rats treated with n-hexane (nHE), dichloromethane (DCM) and aqueous ethanol (Aq. Eth) extracts of Curcubita pepo seed. Total of 48 rats randomly grouped into 12 (n=4), were treated for 21 days by oral gavage as follows: A (control) = 0.5ml 20% tween 80 (vehicle); B (positive control) = 10mg/kg clomiphene citrate, C, D & E = 142.86, 285.71 and 428.57 mg/kg nHE; F, G & H = 142.86, 285.71 and 428.57 mg/kg DCM ; and I, J & K =142.86, 285.71 and 428.57 mg/kg Aq.Eth extracts. Group L (positive control 2) = 10mg/kg clomiphene citrate for 8 days. Following treatment, the rats were paired with males for mating, designating the confirmation day as gestational day 0 (GD 0). On GD 20, the animals were laparatomised and reproductive outcome was determined by assessing foetal weight, foetal crown-rump length, litter size, number of implantation and resorption sites. Results showed all extracts had no significant (p >0.05) effect on the reproductive outcome indices. Clomiphene citrate significantly decreased reproductive outcome indices. In conclusion, Cucurbita pepo seed did not enhance the reproductive outcome of treated female rats at the doses and duration used in this study. This finding may serve as a springboard for future studies exploring the effect of C.pepo at different doses or durations.

Abstract:Moringa oleifera leaves, seeds, bark, roots, sap, and flowers are widely used in traditional medicine, and the leaves and immature seed pods are used as food products in human nutrition. Leaf extracts exhibit the greatest antioxidant activity, and various safety studies in animals involving aqueous leaf extracts indicate a high degree of safety. No adverse effects were reported in association with human studies. Five human studies using powdered whole leaf preparations of M. oleifera have been published, which have demonstrated anti-hyperglycemic (antidiabetic) and anti-dyslipidemic activities. These activities have been confirmed using extracts as well as leaf powders in animal studies. A rapidly growing number of published studies have shown that aqueous, hydroalcohol, or alcohol extracts of M. oleifera leaves possess a wide range of additional biological activities including antioxidant, tissue protective (liver, kidneys, heart, testes, and lungs), analgesic, antiulcer, antihypertensive, radioprotective, and immunomodulatory actions. A wide variety of polyphenols and phenolic acids as well as flavonoids, glucosinolates, and possibly alkaloids is believed to be responsible for the observed effects. Standardization of products is an issue. However, the results of published studies to date involving M. oleifera are very promising. Additional human studies using standardized extracts are highly desirable.

Moringa oleifera leaf ethanolic extract benefits cashmere goat semen quality via improving rumen microbiota and metabolome

Abstract:Background: Artificial insemination (AI) is an effective reproductive technique to improve the performance of cashmere goats and prevent the spread of diseases, and the quality of the semen determines the success of AI. The potential of Moringa oleifera leaf powder (MOLP) and Moringa oleifera leaf ethanolic extract (MOLE) to improve semen quality has been reported, but the underlying mechanisms remain unclear. For the purpose, 18 mature male cashmere goats were randomly assigned into three groups: the control (CON), MOLP, and MOLE groups. The CON group received distilled water orally; the MOLP group was orally treated with 200 mg/kg body weight (BW) MOLP; and the MOLE group was orally treated with 40 mg/kg BW MOLE. Results: Results showed that MOLE contained long-chain fatty acids and flavonoids. Treatment with MOLP and MOLE increased the activities of the serum catalase, superoxide dismutase, and glutathione peroxidase (P < 0.05), enhanced the total antioxidant capacity (P < 0.05), and reduced the serum malondialdehyde level (P < 0.05). At the same time, MOLE increased the contents of serum gonadotropin releasing hormone and testosterone (P < 0.05). Moreover, MOLE significantly increased sperm concentration, motility, and viability (P < 0.05). Meanwhile, MOLE raised the Chao1 index (P < 0.05) and altered the composition of the rumen microbiota; it also raised the relative abundance of Treponema (P < 0.05) and Fibrobacter (P < 0.05) and reduced the relative abundance of Prevotella (P < 0.1). Correlation analysis revealed the genus Prevotella was significantly negatively correlated with sperm concentration, as well as sperm motility and viability. Furthermore, MOLE significantly increased the rumen levels of the steroid hormones testosterone and dehydroepiandrosterone (P < 0.05), as well as the polyunsaturated fatty acids (PUFAs) alpha-Linolenic acid, gamma-Linolenic acid, docosapentaenoic acid, and 9-S-Hydroperoxylinoleicacid (P < 0.05). Conclusions: Oral MOLE supplementation can improve semen quality by increasing the antioxidant capacity and altering the rumen microbiota and metabolites of cashmere goats. Moreover, the MOLP supplementation could enhance the antioxidant capacity of cashmere goats.

Abstract:Obesity and fatty liver are relatively benign states but continued inflammatory stress and its metabolic implications turn them into one of the most devastating diseases of humankind. Generally, obesity and fatty liver precede diabetes mellitus, cardiovascular problems and malignant growths. The present research aimed to explore the efficacy of methanolic extract of ( . ) for the management of obesity and fatty liver and related inflammatory state that prime the body for devastating effects. A series of and studies were employed. Data from HPLC analysis confirmed the presence of flavonoids and phenolic acids. Rats were fed on either normal diet (ND) or high fat diet (HFD and streptozocin (STZ) in the presence or absence of . (250 mg/kg & 500 mg/kg) or metformin (70 mg/kg). Findings showed that rats received 500 mg/kg . showed a significant ( > 0.01) decrease in body weights, liver weights, and plasma glucose level. Laboratory data exhibited a significant ( < 0.05) inhibitory effect on . on pro-inflammatory mediators (IL-1B and TNF) and caused a sharp increase in anti-inflammatory cytokines levels (IL-10, IL-6 and COX-2) in all treatment groups. Histopathological analysis exhibited no structural and functional alteration in the liver and adipose tissues. Altogether, . ameliorates experimentally induced obesity accompanying fatty liver and inflammatory stress. However, further investigations are still needed to confirm the safety and efficacy of (MO) for clinical application.

Abstract:Protein-energy malnutrition (PEM) is a severe health condition affecting millions, especially in developing regions. This study investigates the potential of Moringa oleifera seeds as a low-cost protein source to address protein energy malnutrition. Moringa oleifera seeds were defatted using solvent and used in formulating diets for PEM-induced Wistar rats. The study analyzed the seeds' phytochemical constituents, proximate composition, amino acid profiles, and bioactive compounds through Gas Chromatograph-Mass Spectroscopy (GC-MS). 20 Rats were randomly assigned to four groups: A=control, B= PEM, C= malnourished rats treated with 15% soya bean based-diet and D= malnourished rats treated with 15% defatted Moringa oleifera seed-based diet. The results show that defatting increased protein content and reduced anti-nutritional factors like oxalates, saponins, and tannins, enhancing nutrient bioavailability. Defatted Moringa oleifera seed-based diets improved liver and kidney functions, lipid profiles, and protein digestibility in protein energy malnutrition-induced rats. Additionally, the seeds contained essential bioactive compounds with potential antioxidant, antimicrobial, and anti-inflammatory properties. These findings suggest that Moringa oleifera seeds could be a promising alternative protein source for combating protein energy malnutrition

Abstract:This study aimed to evaluate the safety of Moringa by comparing the effects of different gavage doses of Moringa. The general behavior, body weight, food intake, blood indexes, serum biochemical indexes, and histopathology of rats were used to determine the safety threshold and to provide a reference for the further development and use of Moringa as animal feed. 40 Sprague Dawley rats were selected and given transoral gavage for 28 consecutive days. The T1, T2 and T3 groups were observed for general behavior, body weight, and food intake. Blood and serum biochemical indices were quantified, and histopathology was performed to evaluate the effect and safety of Moringa. The results of the toxicological test showed that (1) Only T1 groups experienced diarrhea. (2) The body weight and food intake of rats in each group were normal compared with the control group. (3) The hematological and serum biochemical indices of rats in the T1 group were significantly different from those of CK but were in the normal range; (4) The results of microscopic examination of the heart, liver, spleen, lung, and kidney of rats in each group were normal, but inflammation occurred in stomach and jejunum of rats in the T1 group, but not in the ileum. The gastrointestinal tract of rats in the T2 and T3 groups were normal. (5) No abnormal death occurred in any of the treatment groups.The results of this study revealed that gavage of Moringa homogenate at a dose of 6 g/kg BW can cause diarrhea in rats. Although there is no pathological effect on weight, food intake, blood and serum biochemical indicators in rats, there are pathological textures in the gastrointestinal tissue caused by diarrhea. Therefore, the safety threshold of Moringa homogenate should be ≤ 3 g/kg BW.

Abstract:The current study aimed to investigate the hepatoprotective efficacy of Moringa oleifera (MOLE) against liver damage in male rats caused by paracetamol. Four groups of twenty-four male Wister rats, weighing between 150 and 200 gm each, were created. To cause liver damage, a dose of 2g/kg of paracetamol was given once a day for a week, and a dose of 300mg/kg B.W. of MOLE was given orally via gavage for four weeks. As an example hepatoprotective medication, silymarin was administered orally at a rate of 100 mg/kg body weight. Enzyme levels in the liver including total bilirubin, ALT, AST, and ALP were utilized to gauge the extent of recovery after hepatic damage. As antioxidant enzymes, SOD and CAT were evaluated. In male rats, paracetamol-induced liver damage was successfully avoided by silymarin and MOLE. The hepatoprotective effects of the extract were evidenced by a noteworthy reduction in the levels of blood enzymes include TB, ALT, AST, and ALP, along with a decline in tumour necrosis factor alpha (TNF-α). Additionally, there was a rise in the levels of antioxidant enzymes SOD and CAT, as well as interleukin 6 (IL-6).
Assessment of oral toxicity of Moringa oleifera Lam aqueous extract and its effect on gout induced in a murine model

Abstract:Background and aim: Although widely employed in traditional remedies globally, the safety and efficacy of Moringa oleifera remain inadequately documented through scientific research. This study evaluated the oral toxicity of M. oleifera leaf aqueous extract (MoAE) and its impact on gout-induced rats. Materials and methods: 2000 mg/kg was given in a single dose during the acute oral toxicity test, while 100 mg/kg, 250 mg/kg, and 500 mg/kg were given daily for 28 days in the repeated dose toxicity test. 100 mg/kg, 250 mg/kg, and 500 mg/kg MoAE doses were administered during the assessment of its impact on gout caused by monosodium urate. In the hyperuricemia model induced by oxonic acid, serum uric acid levels were assessed and pain response was measured through acetic acid-induced writhing. Results: In acute oral and 28-day repeated dose tests, no indications of toxicity were detected, while MoAE alleviated ankle joint swelling and reduced serum uric acid concentrations in arthritic rats, causing a significant reduction in acetic acid-induced contortions. Conclusion: No acute oral toxicity or toxicity in 28-day repeated doses was found for MoAE, while it exhibited antiarthritic, antihyperuricemic, and pain-relieving effects in the murine model.

Abstract:Moringa oleifera is a medicinal plant that has anti-inflammatory, antihypertensive, antidiabetic, tissue-protective, and antioxidant activities. Here, we evaluated the protective effect of M. oleifera leaf powder (MoLP) and 70% ethanol M. oleifera leaf extract (MoLE) on mitigating polycystic ovary syndrome (PCOS)-induced liver and kidney dysfunction via regulating oxidative stress in female albino mice (Mus musculus). The efficacy of M. oleifera was compared with metformin (standard medicine used to treat infertility in women). PCOS was induced by intramuscular injection of testosterone enanthate at 1.0 mg/100 g BW for 35 days. PCOS-induced mice were treated with MoLP (250 and 500 mg/Kg), MoLE (250 and 500 mg/kg), and metformin (250 mg/kg) orally for 14 days. Renal function test (RFT), liver function test (LFT), and oxidative stress biomarker malondialdehyde (MDA) were quantified in serum at 0, 7, and 14 days of intervention. Mice treated with M. oleifera and metformin showed a significant decrease (p < .001) in alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphate (ALP), total bilirubin, urea, creatinine, and a significant increase (p < .001) in total protein, albumin, globulin, and albumin/globulin (A/G) ratio. Oxidative stress decreased significantly (p = .00) with respect to treatments, exposure days, and their interaction in metformin and all M. oleifera-treated groups. M. oleifera leaf powder and extract reduce oxidative stress and enhance nephron-hepatic activity in PCOS-induced female albino mice.
