Abstract:Many waste streams that are suitable substrates for mixed culture bioplastic (polyhydroxyalkanoate, PHA) production are nutrient limited and may need to be supplemented to allow sufficient growth of PHA accumulating bacteria. The scope of this study was to investigate the necessity of nutrient supplementation for the enrichment of an efficient PHA producing mixed culture. We studied the influence of different degrees of carbon and nitrogen limitation on the performance of an acetate-fed feast-famine sequencing batch reactor (SBR) employed to enrich PHA storing bacteria. The microbial reaction rates in the SBR showed a shift with a change in the limiting substrate: high acetate uptake rates were found in carbon-limited SBRs (medium C/N ratios 6-13.2 Cmol/Nmol), while nitrogen-limited SBRs (medium C/N ratios 15-24 Cmol/Nmol) were characterized by high ammonia uptake rates. Biomass in strongly nitrogen-limited SBRs had higher baseline PHA contents in the SBR, but carbon-limited SBRs resulted usually in biomass with higher maximal PHA storage capacities. The PHA storage capacity in a nitrogen-limited SBR operated at 0.5 d SRT decreased significantly over less than 5 months operation. For the microbial selection and biomass production stage of a PHA production process carbon limitation seems thus favourable and nutrient deficient wastewaters may consequently require supplementation with nutrients for the selection of a stable PHA storing biomass with a high storage capacity.

Abstract:The use of residual streams as feedstock for the production of polyhydroxyalkanoates (PHAs) is growing steadily, as it allows the valorization of waste and nutrients otherwise disposed of and the potential production of a biodegradable bioplastic. To date, the environmental and economic costs associated with this process limit its scale-up, which is why it is important to identify possible solutions and optimize the costliest steps. With this in mind, a laboratory-scale sequenced batch reactor (SBR, 5 L) was constructed to allow the selection of a mixed microbial culture able to convert volatile fatty acids (VFAs) into PHA. The reactor is fed with synthetic water containing VFAs, ammonium, phosphate, and micronutrients, typical compounds of fermented streams of certain wastes, such as cheese whey, food waste, or wastewater sludge. The biomass selected and produced by this first reactor is sent to an accumulation reactor, which is fed with a solution rich in VFAs, allowing the accumulation of PHAs. The role of aeration and its impacts on the main process parameters were analyzed. Three scenarios corresponding to different aeration rates were analyzed: 0.08, 0.16, and 0.32 vvm. The SBR was operated at an organic load rate of 600 mgCOD L−1d−1, under a dynamic feeding regime (feast–famine) and a short hydraulic retention time (HRT; 1 day). The results obtained showed that a value of 0.32 enabled better selection and better settling of the sludge. Furthermore, a potential correlation between aeration rate and VFA and NH4+ consumption rates was identified. The resulting biomass was able to accumulate up to 0.15 ± 0.02 g PHAgVSS−1.

Abstract:The production of polyhydroxyalkanoates (PHA) using industrial wastewaters as feedstocks is a current and challenging topic. This study investigated the production of biopolymers by a mixed microbial culture under different OLRs equal to 1 kgCOD m−3d−1 (Period 1), 2 kgCOD m−3d−1 (Period 2) and 3 kgCOD m−3d−1 (Period 3). The maximum PHA content was achieved in Period 2 (0.38 gPHA gTSS−1), whereas lower values were obtained in Period 1 (0.13 gPHA gTSS−1) and Period 3 (0.26 gPHA gTSS−1). Overall, the maximum PHA productivity resulted equal to 0.08 gPHA L−1 h−1 (P2), 0.05 gPHA L−1 h−1 (P1) and 0.04 gPHA L−1 h−1 (P3), respectively.The molecular weight of the PHA increased from Period 1 (250 kDa) to Period 2 (417 KDa) and Period 3 (463 KDa), although resulting in a slight decrease of crystallinity degree. Microbial community analysis revealed a reduction in bacterial diversity and a progressive shift of the microbial community with the increasing OLR. Alpha-diversity indexes based on Operational Taxonomic Units (OTUs) at 99% identity revealed higher species richness (Taxa (S) 280) and diversity (Shannon (H) 4,06) in Period 1, whereas Period 3 was characterized by reduced richness and diversity and higher dominance (Taxa (S) 133, Shannon (H) 2,40).Based on the results obtained, it was pointed out that the OLR variation determined significant effects on the process performances, as well as on the productivity and quality of the biopolymers. This means that OLR is a key control parameter to maximize the PHA production and control the physical-chemical characteristics of the polymers.

Abstract:Polyhydroxyalkanoates (PHA) production via a combination of mixed microbial culture (MMC) and waste carbon substrate has shown to be a more economical solution compared to pure culture process. Although crude glycerol has been proven as a promising feedstock, there are still limited study on the effect of operating conditions such as pH and nutrient feeding strategy on PHA production using crude glycerol. This study aims to investigate the feasibility of enrichment of PHA accumulators in mixed culture without pH control when uncoupled carbon and nitrogen feeding strategy was implemented. Enrichment was conducted in a sequencing batch reactor (SBR) with cycle duration of 24 hr and organic loading rate (OLR) of 1000 mg C/(L.d). Feast and famine strategy was successfully implemented, with feast/famine (F/F) ratio of 0.43 indicating good PHA storage response. The cyclic profile of substrate and PHA concentration has also proven the successful implementation of F/F regime. Biomass concentration of 779 mg/L was obtained by the end of week 5 with PHA content of 16.2% g PHA/g VSS. During the cycle, pH between 6 to 8 was recorded which suggests that extreme acidic or alkaline condition detrimental to PHA accumulators cultivation was not observed in this study. This study demonstrates that enrichment of PHA accumulators is feasible when pH is not controlled under uncoupled C and N feeding strategy.

Abstract:In the last years, the production of plastics at large-scale has been an issue of great concern due to their environmental and health impacts. Biobased and biodegradable plastics, such as polyhydroxyalkanoates (PHA), have emerged as a possible more ecological and sustainable alternative to those conventional plastics. PHA can be produced by mixed microbial cultures (MMC) from waste-based feedstocks. This work aimed to maximize the PHA production from fermented brewer's spent grain by selecting an efficient PHA-accumulating MMC, applying different organic loading rates (OLR, 45 or 60 Cmmol FP /(L.d)) and sludge retention times (SRT, 4 or 2 days). Additionally, the biomass reactivation capacity after storage was evaluated. The global PHA productivity depends on the operating conditions, achieving its maximum (3.55 ± 0.8 g PHA /(L.d)) at the highest OLR and SRT tested. After storage, the global PHA productivity was similar to the one achieved before the starvation period. Depending on the operating conditions, the microbiome has its own bacterial composition signature containing various PHA-producers genera (e.g., Acidovorax , Amaricoccus , Brevundimonas , Leucobacter , Leadbetterella and Thauera ) that persisted over time, albeit at different relative abundancies. A comprehensive overview of PHA production with data on system performance and microbiome is presented, which thereby can further contribute to the upscale of such valorisation processes.

Abstract:Production of polyhydroxyalkanoates (PHAs) by open mixed microbial cultures (MMCs) has been attracting increasing interest as an alternative technology to PHA production by pure cultures, due to the potential for lower costs associated with the use of open systems (eliminating the requirement for sterile conditions) and the utilisation of cheap feedstock (industrial and agricultural wastes). Such technology relies on the efficient selection of an MMC enriched in PHA-accumulating organisms. Fermented cheese whey, a protein-rich complex feedstock, has been used previously to produce PHA using the feast and famine regime for selection of PHA accumulating cultures. While this selection strategy was found efficient when operated at relatively low organic loading rate (OLR, 2g-CODL-1d-1), great instability and low selection efficiency of PHA accumulating organisms were observed when higher OLR (ca. 6g-CODL-1d-1) was applied. High organic loading is desirable as a means to enhance PHA productivity. In the present study, a new selection strategy was tested with the aim of improving selection for high OLR. It was based on uncoupling carbon and nitrogen supply and was implemented and compared with the conventional feast and famine strategy. For this, two selection reactors were fed with fermented cheese whey applying an OLR of ca. 8.5g-CODL-1 (with 3.8g-CODL-1 resulting from organic acids and ethanol), and operated in parallel under similar conditions, except for the timing of nitrogen supplementation. Whereas in the conventional strategy nitrogen and carbon substrates were added simultaneously at the beginning of the cycle, in the uncoupled substrates strategy, nitrogen addition was delayed to the end of the feast phase (i.e. after exogenous carbon was exhausted). The two different strategies selected different PHA-storing microbial communities, dominated by Corynebacterium and a Xantomonadaceae, respectively with the conventional and the new approaches. The new strategy originated a more efficient PHA-production process than the conventional one (global PHA productivity of 6.09g-PHAL-1d-1 and storage yield of 0.96 versus 2.55g-PHAL-1d-1 and 0.86, respectively). Dissociation between the feast to famine length ratio (F/F) and storage efficiency was shown to be possible with the new strategy, allowing selection of an efficient PHA-storing culture with complex feedstock under high organic loading rates.
PHA and EPS production from industrial wastewater by conventional activated sludge, membrane bioreactor and aerobic granular sludge technologies: A comprehensive comparison

Abstract:The present study has focused on the mainstream integration of polyhydroxyalkanoate (PHA) production with industrial wastewater treatment by exploiting three different technologies all operating in sequencing batch reactors (SBR): conventional activated sludge (AS-SBR), membrane bioreactor (AS-MBR) and aerobic granular sludge (AGS). A full aerobic feast/famine strategy was adopted to obtain enrichment of biomass with PHA-storing bacteria. All the systems were operated at different organic loading (OLR) rate equal to 1-2-3 kgCOD/m<sup>3</sup>∙d in three respective experimental periods. The AS-MBR showed the better and stable carbon removal performance, whereas the effluent quality of the AS-SBR and AGS deteriorated at high OLR. Biomass enrichment with PHA-storing bacteria was successfully obtained in all the systems. The AS-MBR improved the PHA productivity with increasing OLR (max 35% w/w), whereas the AS-SBR reduced the PHA content (max 20% w/w) above an OLR threshold of 2 kgCOD/m<sup>3</sup>∙d. In contrast, in the AGS the increase of OLR resulted in a significant decrease in PHA productivity (max 14% w/w) and a concomitant increase of extracellular polymers (EPS) production (max 75% w/w). Results demonstrated that organic carbon was mainly driven towards the intracellular storage pathway in the AS-SBR (max yield 51%) and MBR (max yield 61%), whereas additional stressors in AGS (e.g., hydraulic selection pressure, shear forces) induced bacteria to channel the COD into extracellular storage compounds (max yield 50%) necessary to maintain the granule's structure. The results of the present study indicated that full-aerobic feast/famine strategy was more suitable for flocculent sludge-based technologies, although biofilm-like systems could open new scenarios for other biopolymers recovery (e.g., EPS). Moreover, the AS-MBR resulted the most suitable technology for the integration of PHA production in a mainstream industrial wastewater treatment plant, considering the greater process stability and the potential reclamation of the treated wastewater.

Abstract:In this study, the enrichment sequencing batch biofilm reactor (SBBR) and conventional enrichment reactor (SBR) were established, and a new strategy was developed to achieve higher biomass yield and polyhydroxyalkanoate (PHA) production from food waste than those achieved through the conventional method. After the 10th day of an expanded cultivation period, the respective active biomass concentrations in SBR and SBBR were 5.16 and 13.92 g/L, indicating a significant improvement in the biomass yield of mixed microbial culture in the SBBR. The PHA accumulation test revealed that maximum PHA concentration in SBR and SBBR reached 6.60 and 12.70 g/L, respectively, after 80 days of enrichment, indicating that compared to SBR, the SBBR could more effectively improve the PHA accumulation capacity. Meganema, Thauera, and Paracoccus were the dominant PHA-accumulating bacteria in the SBR and SBBR. Additionally, the volumetric productivity of the SBBR was 3.22 g COD/L/d, twice that of the SBR. This study provides new strategies and insights for improving PHA production and reducing the costs of PHA production and reactor investment.

Abstract:The response of a mixed-microbial-culture (MMC) biomass for PHA accumulation was evaluated over a range of relative nitrogen (N) and phosphorus (P) availabilities with respect to the supply of either complex (fermented whey permeate - FWP) or simpler (acetic acid) organic feedstocks. Fed-batch feed-on-demand PHA accumulation experiments were conducted where the feed N/COD and P/COD ratios were varied ranging from conditions of nutrient starvation to excess. A feast-famine enrichment (activated sludge) biomass, produced in a pilot-scale aerobic sequencing batch reactor on FWP and with a long history of stable PHA accumulation performance, was used for all the experiments as reference material. FWP with N/COD ratios of (2, 5, 15, 70 mg/g all with P/COD = 8 mg/g) as well as simulated FWP with nutrient starvation (N/COD = P/COD = 0) conditions were applied. For the acetic acid accumulations, nutrient starvation as well as N/COD variations (2.5, 5, 50 mg/g all with P/COD = 9 mg/g) and P/COD variations (0.5, 2, 9, 15 mg/g all with N/COD = 10 mg/g) were evaluated. An optimal range of combined N and P limitation with N/COD from 2 to 15 mg/g and P/COD from 0.5 to 3 mg/g was considered to offer consistent improvement of productivity over the case of nutrient starvation. Productivity increased due to active biomass growth of the PHA storing biomass without observed risk for a growth response overtaking PHA storage activity. PHA production with respect to the initial active biomass was significantly higher even in cases of excess nutrient additions when compared to the cases of nutrient starvation. The 24-h PHA productivities were enhanced as much as 4-fold from a base value of 1.35 g-PHA per gram initial active biomass with respect nutrient starvation feedstock. With or without nutrient loading the biomass consistently accumulated similar and significant PHA (nominally 60% g-PHA/g-VSS). Based on results from replicate experiments some variability in the extant biomass maximum PHA content was attributed to interpreted differences in the biomass initial physiological state and not due to changes in feedstock nutrient loading. We found that the accumulation process production rates for mixed cultures can be sustained long after the maximum PHA content of the biomass was reached. Within the specific context of the applied fed-batch feed-on-demand methods, active biomass growth was interpreted to have been largely restricted to the PHA-storing phenotypic fraction of the biomass. This study suggests practical prospects for mixed culture PHA production using a wide range of volatile fatty acid (VFA) rich feedstocks. Such VFA sources derived from residual industrial or municipal organic wastes often naturally contain associated nutrients ranging in levels from limitation to excess.
Optimization of an enriched mixed culture to increase PHA accumulation using industrial saline complex wastewater as a substrate

Abstract:Polyhydroxyalkanoates (PHA) appear as good candidates to substitute conventional petroleum-based plastics since they have similar properties but with the advantage of being biodegradable. Wastewater streams with high organic content are feasible substrates for PHA production resulting in an opportunity for waste recovery. One of the main challenges is the optimization of the selection of microorganisms with high PHA storage capacity. This microbial selection is performed in sequencing batch reactors (SBR) operated under an aerobic feast/famine (F/F) regime. In the present study, a settling stage was added at the end of the feast phase of the enrichment cycle of a SBR fed with pre-acidified cooked mussel processing wastewater (containing up to 12 g NaCl/L). Settling and subsequent supernatant discharge favoured the wash-out of non-accumulating microorganisms as well as the removal of substances that enhanced their undesired development (proteins and carbohydrates). Microbial analysis performed by fluorescence in situ hybridization (FISH) technique showed shifts in the microbial community; the presence of genus Paracoccus increased whereas genera Comamonas decreased. Moreover, the process efficiency was improved with the increase of the PHA production yield (YPHA) and the maximum PHA storage capacity (max. PHA) from 0.48 to 0.72 CmmolPHA/CmmolVFA and from 40 to 60 wt%, respectively. The polymer composition also changed, its HB:HV ratio varied from 83:17 to 70:30. Results obtained in the present study showed that settling after the feast phase promoted the removal of carbon sources that did not contribute to PHA production and the washout of non-storing bacteria, which favoured the culture enrichment.

Abstract:Volatile fatty acid (VFA) rich streams from fermentation of organic residuals and wastewater are suitable feedstocks for mixed microbial culture (MMC) Polyhydroxyalkanoate (PHA) production. However, many such streams have low total VFA concentration (1–10 gCOD/L). PHA accumulation requires a flow-through bioprocess if the VFAs are not concentrated. A flow through bioprocess must balance goals of productivity (highest possible influent flow rates) with goals of substrate utilization efficiency (lowest possible effluent VFA concentration). Towards these goals, dynamics of upshift and downshift respiration kinetics for laboratory and pilot scale MMCs were evaluated. Monod kinetics described a hysteresis between the upshift and downshift responses. Substrate concentrations necessary to stimulate a given substrate uptake rate were significantly higher than the concentrations necessary to sustain the attained substrate uptake rate. A benefit of this hysteresis was explored in Monte Carlo based PHA accumulation bioprocess numerical simulations. Simulations illustrated for a potential to establish continuous flow-through PHA production bioprocesses even at a low (1 gCOD/L) influent total VFA concentration. Process biomass recirculation into an engineered higher substrate concentration mixing zone, due to the constant influent substrate flow, enabled to drive the process to maximal possible PHA production rates without sacrificing substrate utilization efficiency.
Improving simultaneous N, P, and C removal and microbial population dynamics in an anaerobic–aerobic–anoxic SBR (A-O-A-SBR) treating municipal wastewater by altering organic loading rate (OLR)

Abstract:To assess the impacts of the influent organic loading rate (OLR) on the performance of an improved sequencing batch reactor (A-O-A-SBR system), four OLRs (1.35, 2.03, 2.70, and 3.38 g-COD/L/d) were used for enhancing simultaneous N, P, and C removals in the system. Results showed that high simultaneous N, P, and C removal efficiencies of the A-O-A-SBR were achieved with an OLR of 2.03 g-COD/L/d. The corresponding TN, TP, and COD removal efficiencies were 98.10%, 87.01%, and 93.43%, respectively. Dominant phyla, including Proteobacteria, Planctomycetes, Candidatus Saccharibacteria, Bacteroidetes, and critical genus Thiothrix, and essential classes such as Gammaproteobacteria, Betaproteobacteria, Sphingobacteriia, contributed to the excellent A-O-A-SBR performance. Functional genes forecasts demonstrated that the metabolic potential on pollutants degradation metabolism improved with the proper OLR condition.

Abstract:In this paper, a study was conducted on the effect of polyhydroxyalkanoates (PHA) and glycogen transformations on biologic nitrogen and phosphorus removal in low dissolved oxygen (DO) systems. Two laboratory-scale sequencing batch reactors (SBR1 and SBR2) were operating with anaerobic/aerobic (low DO, 0.15–0.45 mg·L −1 ) configurations, which cultured a propionic to acetic acid ratio (molar carbon ratio) of 1.0 and 2.0, respectively. Fewer poly-3-hydroxybutyrate (PHB), total PHA, and glycogen transformations were observed with the increase of propionic/acetic acid, along with more poly-3-hydroxyvalerate (PHV) and poly-3-hydroxy-2-methyvalerate (PH2MV) shifts. The total nitrogen (TN) removal efficiency was 68% and 82% in SBR1 and SBR2, respectively. In the two SBRs, the soluble ortho-phosphate (SOP) removal efficiency was 94% and 99%, and the average sludge polyphosphate (poly-P) content (g·g-MLVSS −1 ) was 8.3% and 10.2%, respectively. Thus, the propionic to acetic acid ratio of the influent greatly influenced the PHA form and quantity, glycogen transformation, and poly-P contained in activated sludge and further determined TN and SOP removal efficiency. Moreover, significant correlations between the SOP removal rate and the (PHV + PH2MV)/PHA ratio were observed ( R 2 >0.99). Accordingly, PHA and glycogen transformations should be taken into account as key components for optimizing anaerobic/aerobic (low DO) biologic nitrogen and phosphorus removal systems.
Enhanced polyhydroxyalkanoate (PHA) production from the organic fraction of municipal solid waste by using mixed microbial culture

Abstract:BackgroundIn Europe, almost 87.6 million tonnes of food waste are produced. Despite the high biological value of food waste, traditional management solutions do not consider it as a precious resource. Many studies have reported the use of food waste for the production of high added value molecules. Polyhydroxyalkanoates (PHAs) represent a class of interesting bio-polyesters accumulated by different bacterial cells, and has been proposed for production from the organic fraction of municipal solid waste (OFMSW). Nevertheless, until now, no attention has been paid to the entire biological process leading to the transformation of food waste to organic acids (OA) and then to PHA, getting high PHA yield per food waste unit. In particular, the acid-generating process needs to be optimized, maximizing OA production from OFMSW. To do so, a pilot-scale Anaerobic Percolation Biocell Reactor (100 L in volume) was used to produce an OA-rich percolate from OFMSW which was used subsequently to produce PHA.ResultsThe optimized acidogenic process resulted in an OA production of 151 g kg−1 from fresh OFMSW. The subsequent optimization of PHA production from OA gave a PHA production, on average, of 223 ± 28 g kg−1 total OA fed. Total mass balance indicated, for the best case studied, a PHA production per OFMSW weight unit of 33.22 ± 4.2 g kg−1 from fresh OFMSW, corresponding to 114.4 ± 14.5 g kg−1 of total solids from OFMSW. PHA composition revealed a hydroxybutyrate/hydroxyvalerate (%) ratio of 53/47 and Mw of 8∙105 kDa with a low polydispersity index, i.e. 1.4.ConclusionsThis work showed how by optimizing acidic fermentation it could be possible to get a large amount of OA from OFMSW to be then transformed into PHA. This step is important as it greatly affects the total final PHA yield. Data obtained in this work can be useful as the starting point for considering the economic feasibility of PHA production from OFMSW by using mixed culture.

Abstract:Polyhydroxyalkanoates (PHAs) are important and completely biodegradable alternatives to regular plastics, and they can be produced by activated sludge systems during wastewater treatment. Wastewaters with high organic content are being used for PHA production, which is an important resource recovery option. In this context, the effect of sludge retention time and different carbon sources, such as acetate, peptone-mixture and industrial wastewater (containing acetic acid (AA), lactic acid (LA) and propionic acid (PA)), on PHA storage was investigated. Oxygen utilisation rate (OUR) profiles were generated in respirometric tests and were evaluated by activated sludge modelling. Results showed that high storage (AA: 70%; LA: 49%; PA: 60% and industrial wastewater: 52%) was achievable in the feast phase even when the biomass was fed with a high organic acid content substrate to which it is not acclimated.

Effects of increasing organic loading rate on performance and microbial community shift of an up-flow anaerobic sludge blanket reactor treating diluted pharmaceutical wastewater

Abstract:The performance of an up-flow anaerobic sludge blanket (UASB) reactor was investigated in the treatment of diluted pharmaceutical fermentation wastewater for a continuous operation of 140 days. The dynamics and compositions of the microbial community were monitored using polymerase chain reaction (PCR)-restriction fragment length polymorphism (PCR-RFLP) analysis. Increase of the organic loading rate (OLR) from 2.7 kg COD/m(3) d to 7.2 COD/m(3) d led to an increase in the COD removal efficiency from 83% to 91%. The dominant bacteria shifted from Proteobacteria (23.8%), Chloroflexi (14.5%) and Firmicutes (4.0%) to Firmicutes (48.4%), Bacteroidetes (9.5%) and Proteobacteria (5.4%). For archeaon, the dominant groups changed from Thermoplasmata (24.4%), Thermoprotei (18.0%) and Methanobacteria (30.8%) to Thermoplasmata (70.4%) and Methanomicrobia (16.8%). Firmicutes, Bacteroidetes, Thermoplasmata and Methanobacteria could outcompete other species and dominated in the reactor under higher OLR. The results indicated that, to some extent, microbial community shift could reflect the performance of the reactor and a significant community shift corresponded to a considerable process event.

Abstract:Vascular angiotensin-converting enzyme (ACE) activity is increased in a variety of disorders representing both early and late stages of atherosclerosis. The factors governing the induction of vascular ACE are poorly understood. We hypothesized that vascular ACE activity might be increased by hypertension-induced vessel distension. Hypertension was induced in rats by suprarenal coarctation of the aorta. Analyses were performed 5 days and 4 weeks post-operation. ACE activity and ACE mRNA level were increased in thoracic aortae from coarctation hypertensive rats that had been exposed to elevated blood pressure, whereas they remained at normal level in abdominal aortae from those rats that had been exposed to normal blood pressure. The degree of aortic ACE induction correlated well with the degree of the trans-stenotic blood pressure gradient. An increase in ACE transcript level was also observed in carotid arteries from coarctation hypertensive rats that had been exposed to elevated blood pressure. In contrast, ACE activity and ACE mRNA expression were not altered in tissues that did not contain any large arteries from coarctation hypertensive rats, although these tissues had been exposed to elevated blood pressure. These results demonstrate an induction of ACE in large arteries that had been exposed to elevated blood pressure, and they imply that the induction of vascular ACE is due to hypertension-induced vessel distension.
Poly-hydroxyalkanoates production from mixed microbial cultures: effect of dissolved oxygen and pH.

Abstract:The accumulation potential of poly-hydroxyalkanoates (PHAs) produced from an acclimated mixed microbial culture (MMC) under different dissolve oxygen (DO) levels and pH was investigated, using volatile fatty acids (VFAs) as carbon source. Specifically, the PHAs production potential of the MMC was evaluated in batch mode in a 5L bioreactor under controlled high aeration (DO level set at 25% saturation, DO25) and low aeration (DO level set at 5% saturation, DO5). The effect of the controlled DO level on the PHAs yield expressed in % PHAs/DCW (w/w) and also the monomeric composition was evaluated in comparison to the results obtained during constant aeration at 2L/min/L rector i.e. with highly varying DO (DOv). The optimal yield of 79.5±0.14% PHAs/DCW was achieved for DO25, while in the cases of DO5 results were similar to those from DOv, i.e. 60 % PHAs/DCW and 65 % PHAs/DCW, respectively . In all cases, P(3HB-co-3HV) was produced whereas the assimilation of propionate to HV in the polymers did not seem to be correlated to the DO changes, with HV content being above 5% in all cases. The subsequent simultaneous advantageous for the process, indicating that control of DO and not of pH proved to be even more advantageous for the process enhancing the assimilation rates as well, indicating that the production of PHAs from MMCs may be quite efficient if the cultivation conditions are properly controlled.

Abstract:Optimizing anoxic biological phosphorus removal in the anaerobic-anoxic sequencing batch reactor (A2 SBR) was observed to depend greatly on three parameters: the amount of added nitrate, the sludge retention time (SRT) and the phosphorus/carbon feeding ratio (P/C, wt/wt). The concentration of 120 mg N-NO3 l-1 in the anoxic medium corresponding to 800 mg COD l-1 acetic acid and 60 mg P-PO4 l-1 in the synthetic wastewater, the SRT of 15 days and the P/C feeding ratio of 20/100 were determined as optimal for complete phosphorus removal in the A2 SBR. The acetate uptake, the phosphorus release and the phosphorus removal increased with the P/C feeding ratio and the phosphorus sludge content (Ps). The polyphosphate-accumulating organisms (PAO) were dominant under operation at all P/C feeding ratios tested except 2/100. At a P/C feeding ratio of 20/100, PAO could accumulate a high content of polyphosphate compared with other P/C ratios. In contrast, the P/C ratio of 2/100 caused a decrease in the polyphosphate content in PAO, the deterioration of enhanced biological phosphorus removal (EBPR) activity, and the dominance of glycogen-accumulating organisms (GAO). This study was completed by microscopic observations which revealed three morphological types of PAO. This is the first time that an oval form of PAO could be observed in the A2 SBR.

Abstract:An innovative approach has been recently proposed in order to link polyhydroxyalkanoates (PHA) production with sludge minimization in municipal wastewater treatment, where (1) a sequencing batch reactor (SBR) is used for the simultaneous municipal wastewater treatment and the selection/enrichment of biomass with storage ability and (2) the acidogenic fermentation of the primary sludge is used to produce a stream rich in volatile fatty acids (VFAs) as the carbon source for the following PHA accumulation stage. The reliability of the proposed process has been evaluated at lab scale by using substrate synthetic mixtures for both stages, simulating a low-strength municipal wastewater and the effluent from primary sludge fermentation, respectively. Six SBR runs were performed under the same operating conditions, each time starting from a new activated sludge inoculum. In every SBR run, despite the low VFA content (10 % chemical oxygen demand, COD basis) of the substrate synthetic mixture, a stable feast–famine regime was established, ensuring the necessary selection/enrichment of the sludge and soluble COD removal to 89 %. A good process reproducibility was observed, as also confirmed by denaturing gradient gel electrophoresis (DGGE) analysis of the microbial community, which showed that a high similarity after SBR steady-state had been reached. The main variation factors of the storage properties among different runs were uncontrolled changes of settling properties which in turn caused variations of both sludge retention time and specific organic loading rate. In the following accumulation batch tests, the selected/enriched consortium was able to accumulate PHA with good rate (63 mg CODPHA g CODXa−1 h−1) and yield (0.23 CODPHA CODΔS−1) in spite that the feeding solution was different from the acclimation one. Even though the PHA production performance still requires optimization, the proposed process has a good potential especially if coupled to minimization of both primary sludge (by its use as the VFA source for the PHA accumulation, via previous fermentation) and excess secondary sludge (by its use as the biomass source for the PHA accumulation).
