Abstract:Nitrate accumulation is a common phenomenon in aquaculture that can lead to eutrophication of surrounding water bodies. This study used poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) as a carbon source and substrate and performed a microbial co-occurrence network ecological analysis to elucidate the denitrification processes in two packed-bed reactors with different salinities. The denitrification rate reached maximum values of 0.438 and 0.446 kg m−3 d−1 in reactor I (salinity 0 ‰) and reactor II (salinity 20 ‰), respectively. Although ammonia was formed in both systems based on dissimilation nitrate reduction to ammonia (DNRA), the concentration was very low (2.47 ± 1.99 and 2.84 ± 1.79 mg L−1); moreover, the nitrite content was average (1.01 ± 0.87 and 0.96 ± 0.86 mg L−1). These results suggested that denitrification dominated in both reactors. PHBV generally presented a stable release of DOC, although a sharp increase was observed in the start-up period of reactor II. 16S rRNA results showed that reactor I had richer microbial diversity than reactor II. Among the top ten taxa, Betaproteobacteria was the dominant class in reactor I while Gammaproteobacteria was the dominant class in reactor II. In the stable period, Thauera and Denitromonas was the most abundant genera in reactor I and reactor II, respectively. In addition, the bacterial co-occurrence network showed that reactor I had a more complex node and edge network and faster start-up time compared to reactor II; however, reactor II had a more stable nitrogen removal capacity. Higher expression of NorB and NosZ genes in reactor II indicated higher efficient denitrification in seawater system. The SEM and FTIR showed bacterial development and materials surface erosion. These findings verified the denitrification performance and niche differences between freshwater and seawater environments.

Abstract:The microalgae-based system has been applied in anaerobic digestate treatment for nutrient removal and biomass production. To optimize its performance in treating piggery digestate, here, commercial bacterial agents, including organic degrading bacteria (Cb) and nitrifying bacteria (Nb), were inoculated into the microalgae-based system dominated by Desmodesmus sp. CHX1 (D). Reactor DN (inoculated with D and Nb) and DCN (inoculated with D, and Cb to Nb at a ratio of 1:2) have better performance on NH4+-N removal, with a final efficiency at 40.26% and 39.87%, respectively, and no NO3-N or NO2-N accumulations. The final total chlorophyll concentration, an indicator of microalgal growth, reached 4.74 and 5.47 mg/L in DN and DCN, respectively, three times more than that in D. These results suggested that high NH4+-N removal was achieved by the assimilation into high microalgal biomass after the inoculation with functional bacteria. High-throughput sequencing showed that the richness of microbial community decreased but the evenness increased by inoculating functional microorganisms. Microalgae aggregating bacteria were Cellvibrio, Sphingobacterium, Flavobacterium, Comamonas, Microbacterium, Dyadobacter, and Paenibacillus. This study revealed that the inoculation with functional bacteria reconstructed the microbial community which benefited for the microalgal growth and nutrient removal, providing a promising strategy for treating highly-concentrated digestate. Characteristics and Performances of Microalgal-Bacterial Consortia in a Mixture of Raw Piggery Digestate and Anoxic Aerated Effluent

Abstract:The piggery digestate of high ammonia was mixed with the anoxic aerated effluent of high nitrate and phosphorus, to cultivate a microalgal-bacterial consortium for simultaneous pollution removal and resource recovery. The highest removal of total inorganic nitrogen was achieved at 324.77 mg/L in 40% piggery digestate mixed with 60% anoxic aerated effluent, along with the most microalgae biomass production. The crude protein and fatty acids of C14-C20 in microalgae cells were 21.80% and 69.78%, indicating that this mixing strategy could produce abundant microalgal biomass suitable for biofuel generation and animal feed. High-throughput sequencing showed that microbial diversity increased and Paenibacillus, Thiopseudomonas and Pseudomonas were the dominant species promoting microalgal growth. Overall, these results provided a new insight of mixing two types of wastewaters for cultivating microalgal-bacterial consortia, to remove contamination and recover nutrients simultaneously.
The Evolution of Bacterial Community Structure and Function in Microalgal-Bacterial Consortia with Inorganic Nitrogen Fluctuations in Piggery Digestate

Abstract:The inorganic nitrogen concentrations in full-scale anaerobic digestion plants always vary seasonally, challenging the removal stability of microalgal-bacterial consortia in treating piggery digestate. Using a continuous photobioreactor, this study revealed the evolution of bacterial community under inorganic nitrogen fluctuations with influent nitrate loading ranged from 11 to 90 mg-N L-1 and influent ammonium loading ranged from 130 to 550 mg-N L-1. The loading of gradient nitrogen concentrations demonstrated that the optimal removal rate of total inorganic nitrogen was 45.62 mg-N L-1 d-1 along with the total chlorophyll concertation of 19.27 mg L-1, when introducing 240 mg-N L-1 of ammonium and 11 mg-N L-1 of nitrate. High throughput sequencing indicated that the dominant community members for nitrogen removal were shifted from prokaryotic algae to nitrifying and denitrifying bacteria, with nitrate being changed to ammonium. With the increasing influent ammonium loading, the suspended microalgal bacterial consortia gradually changed to microalgal bacterial biofilm, which was attributed to the biofilm-producing prokaryotes Thauera Comamona and Arenimonas. Co occurrence network analyses illustrated that more functional bacterial modules were formed under ammonium fluctuation. Further correlation test identified influent ammonium loading as the important functional parameter shaping the bacterial community. Overall, this study expands our understanding of the homeostasis in microalgal-bacterial system in the cleaner production of piggery digestate.
Filamentous cyanobacteria and hydrophobic protein in extracellular polymeric substances facilitate algae—bacteria aggregation during partial nitrification

Abstract:In algae—bacteria symbiotic wastewater treatment, the excellent settling performance of algae—bacteria aggregates is critical for biomass separation and recovery. Here, the composition of extracellular polymeric substances (EPS), microbial profiles, and functional genes of algae—bacteria aggregates were investigated at different solid retention times (SRTs) (10, 20, and 40 d) during partial nitrification in photo sequencing bioreactors (PSBRs). Results showed that SRTs greatly influenced the nitrogen transformation and the formation and morphological structure of algae—bacteria aggregates. The highest nitrite accumulation, the largest particle size (~1.54 mm) and the best settling performance were observed for the algae—bacteria aggregates in the PSBR with an SRT of 10 d, where the abundant occurrence of filamentous cyanobacteria with the highest ratio of chlorophyll a /b and the lowest EPS amount with the highest protein-to-polysaccharide ratio were observed. In particular, the EPS at 10 d of SRT contained a higher amount of protein-related hydrophobic groups and a lower ratio of α-helix/(β-sheet + random coil), indicating a looser protein structure, which might facilitate the formation and stabilization of algae—bacteria aggregates. Moreover, algal-bacterial aggregation greatly depended on the composition and evolution of filamentous cyanobacteria ( unclassified _o__Oscillatoriales and Phormidium accounted for 56.29 % of the identified algae at SRT 10 d). The metagenomic analysis further revealed that functional genes related to amino acid metabolism (e.g., genes of phenylalanine, tyrosine, and tryptophan biosynthesis) were expressed at high levels within 10 d of SRT. Overall, this study demonstrates the influence of EPS structures and filamentous cyanobacteria on algae—bacteria aggregation and reveals the biological mechanisms driving photogranule structure and function.

Abstract:The purple non‐sulfur bacteria Rhodomicrobium vannielii and Rhodomicrobium udaipurense produce polyhydroxyalkanoate under various phototrophic growth conditions using N2 gas or ammonium chloride as nitrogen sources. This study adds to a growing body of literature exploring metabolically versatile microbes as biocatalysts for heterotrophic and autotrophic bioproduction. Petroleum‐based plastics levy significant environmental and economic costs that can be alleviated with sustainably sourced, biodegradable, and bio‐based polymers such as polyhydroxyalkanoates (PHAs). However, industrial‐scale production of PHAs faces barriers stemming from insufficient product yields and high costs. To address these challenges, we must look beyond the current suite of microbes for PHA production and investigate non‐model organisms with versatile metabolisms. In that vein, we assessed PHA production by the photosynthetic purple non‐sulfur bacteria (PNSB) Rhodomicrobium vannielii and Rhodomicrobium udaipurense. We show that both species accumulate PHA across photo‐heterotrophic, photo‐hydrogenotrophic, photo‐ferrotrophic, and photo‐electrotrophic growth conditions, with either ammonium chloride (NH4Cl) or dinitrogen gas (N2) as nitrogen sources. Our data indicate that nitrogen source plays a significant role in dictating PHA synthesis, with N2 fixation promoting PHA production during photoheterotrophy and photoelectrotrophy but inhibiting production during photohydrogenotrophy and photoferrotrophy. We observed the highest PHA titres (up to 44.08 mg/L, or 43.61% cell dry weight) when cells were grown photoheterotrophically on sodium butyrate with N2, while production was at its lowest during photoelectrotrophy (as low as 0.04 mg/L, or 0.16% cell dry weight). We also find that photohydrogenotrophically grown cells supplemented with NH4Cl exhibit the highest electron yields – up to 58.89% – while photoheterotrophy demonstrated the lowest (0.27%–1.39%). Finally, we highlight superior electron conversion and PHA production compared to a related PNSB, Rhodopseudomonas palustris TIE‐1. This study illustrates the value of studying non‐model organisms like Rhodomicrobium for sustainable PHA production and indicates future directions for exploring PNSB metabolisms.

Abstract:Photosynthetic bacteria (PSB) wastewater treatment is a special technology which can simultaneously remove pollutants from wastewater and produce high-value cell inclusions, but the characteristics of PSB in the heavy oil refinery wastewater treatment system are not clear. This study evaluated pollutants removal with different light intensities, and the accumulation of biomass, carotenoid, bacteriochlorophyll, and coenzyme Q10 while treating heavy oil refinery wastewater in PSB system. Furthermore, high sequencing technology and q-PCR were used to analyses microbial community structure and the quantity of Rhodopseudomonas and Pseudomonas in the systems. Results showed that 500 lx was the optimal intensity for SCOD and NH4+-N removal, the corresponding removal ratios were 62.66% and 91.54%, respectively, and the yield of coenzyme Q10 reached the highest on the 8 th day, which was 9.90 mg/g-DCW, with the highest dehydrogenase concentration reaching 71.83 mg/L. While 4000 lx was the optimal light intensity for the yield of carotenoid, bacteriochlorophyll and biomass production, the corresponding accumulations were 10.64 mg/g-DCW, 5.88 mg/g-DCW, and 550 mg/L, respectively. Furthermore, the relative abundance of Pseudomonas was the highest at 500 lx, which was 37.13%, and the gene number was 4.56 × 105 gene copies/μL, 2.34 times of that before the reaction. At 4000 lx, the relative abundance of Rhodopseudomonas was the highest, which was 58.91%, and the gene number was 9.51 × 105 gene copies/μL, 7.15 times of that before the reaction. The results proved that it was feasible for culturing Rhodopseudomonas and Pseudomonas in PSB system to treat heavy oil refinery wastewater and realized cell inclusions accumulation simultaneously. The results could provide theoretical guidance for industrial treatment of refinery wastewater.
Recovery of purple non-sulfur bacteria-mediated single-cell protein from domestic wastewater in two-stage treatment using high rate digester and raceway pond

Abstract:Wastewater resources can be used to produce microbial protein for animal feed or organic fertiliser, conserving food chain resources. This investigation hasemployed thefermented sewage to photoheterotrophically grown purple non-sulfur bacteria (PNSB) in a 2.5 m3 pilot-scaleraceway-pond with infrared light to produce proteinaceous biomass. Fermented sewage with synthetic media consisting of sodium acetate and propionic acids at a surface-to-volume (S/V) ratio of 10 m2/m3 removed 89%, 93%, and 81% of chemical oxygen demand, ammonium nitrogen, and orthophosphate, respectively; whereas respective removal in fermented sewage alone without synthetic media was 73%, 73%, and 72% during batch operation of 120 h. The biomass yield of 0.88-0.95 g CODbiomass /g CODremoved with protein content of 40.3 ± 0.3%-43.9 ± 0.2% w/w was obtained for fermented sewage with synthetic media. The results revealed enhanced possibility of scaling-up the raceway reactor to recover resources from municipal wastewater and enable simultaneous high-rate PNSB single-cell protein production.
Nutrients and biomass dynamics in photo-sequencing batch reactors treating wastewater with high nutrients loadings

Abstract:The present study investigates different strategies for the treatment of a mixture of digestate from an anaerobic digester diluted and secondary effluent from a high rate algal pond. To this aim, the performance of two photo-sequencing batch reactors (PSBRs) operated at high nutrients loading rates and different solids retention times (SRTs) were compared with a semi-continuous photobioreactor (SC). Performances were evaluated in terms of wastewater treatment, biomass composition and biopolymers accumulation during 30 days of operation. PSBRs were operated at a hydraulic retention time (HRT) of 2 days and SRTs of 10 and 5 days (PSBR2-10 and PSBR2-5, respectively), whereas the semi-continuous reactor was operated at a coupled HRT/SRT of 10 days (SC10-10). Results showed that PSBR2-5 achieved the highest removal rates in terms of TN (6.7 mg L−1 d−1), TP (0.31 mg L−1 d−1), TOC (29.32 mg L−1 d−1) and TIC (3.91 mg L−1 d−1). These results were in general 3–6 times higher than the removal rates obtained in the SC10-10 (TN 29.74 mg L−1 d−1, TP 0.96 mg L−1 d−1, TOC 29.32 mg L−1 d−1 and TIC 3.91 mg L−1 d−1). Furthermore, both PSBRs were able to produce biomass up to 0.09 g L−1 d−1, more than twofold the biomass produced by the semi-continuous reactor (0.04 g L−1 d−1), and achieved a biomass settleability of 86–92%. This study also demonstrated that the microbial composition could be controlled by the nutrients loads, since the three reactors were dominated by different species depending on the nutritional conditions. Concerning biopolymers accumulation, carbohydrates concentration achieved similar values in the three reactors (11%), whereas < 0.5% of polyhydrohybutyrates (PHB) was produced. These low values in biopolymers production could be related to the lack of microorganisms as cyanobacteria that are able to accumulate carbohydrates/PHB.

Abstract:Polyhydroxyalkanoates (PHA) are a group of biopolymers produced naturally by microorganisms with properties similar to various petroleum-based plastics. However, to date their commercial production has remained uncompetitive due to substrate, sterilization, aeration and processing costs. Purple non-sulfur bacteria (PNSB) are a group of anoxygenic photoheterotrophic bacteria that have the ability to accumulate PHA under unbalanced conditions in anaerobic environments and constant feeding with high conversion ratios. Such characteristics could potentially overcome some of the bottlenecks of conventional chemoheterotrophic PHA production. Yet these organisms have received relatively limited attention. This review explores the factors involved in the PHA accumulation process from PNSB, highlighting the differences to conventional PHA production and the areas yet to be optimized. The roles of fermentation systems, carbon substrate, feeding conditions, nutrients, pH and various aspects of light are reviewed to understand their role in PHA accumulation in PNSB.
Optimizing light sources for selective growth of purple bacteria and efficient formation of value-added products

Abstract:The slow growth and inefficient productivity of value-added products are the main obstacles to industrialization of purple non-sulfur bacteria (PNSB)-based technology for clean production from wastewater. To overcome these, a strain of PNSB was obtained through selective cultivation and purification, which was subsequently identified as Rhodobacter sphaeroides. The impacts of different light sources (i.e. halogen lamp, incandescent lamp, infrared light, and light-emitting diodes (LEDs) of white, red, blue, green and yellow) were investigated on the production of biomass, pigments and protein by Rb. sphaeroides. Results suggested that, in addition to infrared light, green and yellow LED, most light sources managed to keep the purity of Rb. sphaeroides above 96%. After 7-day cultivation, halogen lamp and infrared light yielded the highest concentrations of biomass (5.78 g SS L−1) and pigments (22.6 mg L−1), respectively. Highest protein-to-biomass ratio (82%) was observed under blue LED, while maximum protein concentration (4.43 g protein L−1) was obtained under incandescent lamp. The infrared light and incandescent lamp might account for the higher production of pigments and protein, respectively. Considering the culture screening, productivity of biomass, pigments and protein as well as energy efficiency together, incandescent lamp was the optimal light source in lab scale. It also gave rise to the highest net profit of $ 14.43 per ton of wastewater treated in a desktop extrapolation evaluating economic benefit of full-scale PNSB-based wastewater recovery process. The results jointly demonstrated that appropriate light source (i.e. incandescent lamp) would boost the microbial protein production and enhance the protein content, which is of great importance to cost control in practice.

Abstract:Hydrogen (H2) as a clean fuel holds global potential and can be produced through bio-processes. To enhance bioH2 yields, integrated systems have been proposed, combining dark fermentation (DF) of wastewater with a subsequent photofermentation (PF) stage involving purple non-sulfur (PNS) bacteria. Mixed cultures of PNS bacteria and their microbial ecology have been relatively understudied despite the known benefits of mixed cultures in industrial applications. The aim of this study was to obtain various mixed cultures of PNS bacteria under different environmental conditions during the enrichment stage. Four different mixed cultures were obtained (A, B, C, and D). However, in the H2 production phase, only Consortium A, which had been enriched with malic acid as the carbon source, exposed to 32 W m−2 of irradiance, and subjected to intermittent agitation, produced H2 with a yield of 9.37 mmol H2 g−1 COD. The consortia enriched were a hybrid of PF and DF bacteria. Especially in Consortium A, Rhodopseudomonas palustris was the dominant organism, and various DF bacteria were positively associated with H2 production, with their dominance comparable to that of PNS bacteria. Despite the reported low yields, optimizing environmental conditions for this culture could potentially enhance hydrogen production from DF effluents.

[Evolution of the microbiological parameters of cured ham].

Abstract:The main microbiological and physico-chemical parameters in dry-salted ham previously selected were determined along the elaboration process. All determinations were performed at 2 levels: surface and internal. All bacterial groups showed a similar behaviour, increasing along the first stages up to the third month of drying, then decreasing to numbers similar to the initial ones. Only the halotolerants maintained fairly high numbers during last stages. Some groups were almost absent at internal levels. The hazardous bacteria showed different behaviour: whereas the faecal streptococci were fairly abundant during the whole study, coliforms almost disappeared at the final stages. During the elaboration process at different stages of ripening they were analysed in order to know the moulds population at surface level. Aspergillus and Penicillium were the dominant genera. The latter, mainly represented by 10 species, dominated during the first stages of elaboration. Aspergillus was present throughout the whole making process. The glaucus group was clearly dominant at the final stages due to the low aw values together with the high NaCl percentages. Aspergillus halophilicus was the most abundant species identified (18%) and the most frequently isolated at the final stages.

Abstract:The global shift from traditional fish farming to aquaculture has created an aquafeed production gap. Hence, the recovery of microbial protein from organic and nutrient-rich agro-industrial wastewaters has been identified as a suitable substitute. However, such waste streams are sparse in arid climes like the Middle East. Thus, this study explores the potential of single-cell protein recovery from a novel waste stream abundant in the region–fuel synthesis process water (FSPW), via anaerobic treatment with purple non-sulfur bacteria (PNSB). The feedstock (COD = 10.3 g/L) amended with essential nutrients was inoculated with a PNSB-dominated mixed culture in replicate 1-L batch fermenters. The wastewater characteristics and microbial biomass assays were performed using standard methods. Around two-thirds of the COD was degraded within 72 h at a rate of 2100 mg L −1 d −1 , which reduced to about 710 mg L −1 d −1 by trial end. Also, total nitrogen levels (90 mg/L) were depleted within 72 h, indicating that nitrogen was a limiting nutrient. In addition, a peak biomass concentration of 1.11±0.037 g vss /L was obtained. Proximate analysis revealed that the biomass consisted of 35% protein, 32% lipid, 16% carbohydrate, 7% ash, 0.5% carotenoids, 0.6% bacteriochlorophylls, and 0.004% coenzyme Q10. Biomass protein's amino acid profile was comparable to soybean grain and meets dietary requirements for several aquatic livestock. Metal analysis of the biomass and wastewater indicated that nutritionally undesirable metals were undetected. Results show that PNSB not only efficiently degrade FSPW's organic load but also upcycles the waste to valuable feed constituents, potentially creating a regional circular economy.

Abstract:The climate crisis requires rethinking wastewater treatment to recover resources, such as nutrients and energy. In this scenario, purple phototrophic bacteria (PPB), the most versatile microorganisms on earth, are a promising alternative to transform the wastewater treatment plant concept into a biorefinery model by producing valuable protein-enriched biomass. PPB are capable of interacting with electrodes, exchanging electrons with electrically conductive materials. In this work, we have explored for mobile-bed (either stirred or fluidized) cathodes to maximize biomass production. For this purpose, stirred-electrode reactors were operated with low-reduced (3.5 e-/C) and high-reduced (5.9 e-/C) wastewater under cathodic polarization (-0.4 V and -0.8 V vs. Ag/AgCl). We observed that cathodic polarization and IR irradiation can play a key role in microbial and phenotypic selection, promoting (at -0.4 V) or minimizing (at -0.8 V) the presence of PPB. Then, we further study how cathodic polarization modulates PPB biomass production providing a fluid-like electrode as part of a so-called photo microbial electrochemical fluidized-bed reactor (photoME-FBR). Our results revealed the impact of reduction status of carbon source in wastewater to select the PPB photoheterotrophic community and how electrodes drive microbial population shifts depending on the reduction status of such carbon source.

Abstract:This work studied the formation of aggregates used for wastewater treatment in high-rate algal ponds (HRAP). For this, the establishment of microalgae-bacteria aggregates in these systems was evaluated, considering strategies for the inoculation and start-up. Two HRAP were operated in parallel, at first in batch mode and then in continuous flow. The wastewater treatment was efficient, with removal rates around 80% for COD and N-ammoniacal. Volatile suspended solids and chlorophyll for the culture grew continuously reached a concentration of 548 ± 11 mg L-1 and 7.8 mg L-1, respectively. Larger photogranules were observed when the system was placed in a continuous regime. The protein fraction of extracellular polymeric substances was identified as a determinant in photogranules formation. During the continuous regime, more than 50% of the biomass was higher than 0.2 mm, flocculation efficiency of 78 ± 6%, and the volumetric sludge index of 32 ± 5 mL g-1. The genetic sequencing showed the growth of cyanobacteria in the aggregate and the presence of microalgae from the chlorophytes and diatoms groups in the final biomass.

Abstract:A wastewater-born and settleable algal-bacterial culture, cultivated in a stirred tank photobioreactor under lab conditions, was used to remove the carbon and nutrients in municipal wastewater and accumulate biomass simultaneously. The algal-bacterial culture showed good settleable property and could totally settle down over 20 min, resulting in a reduction of total suspended solids from an initial 1.84 to 0.016 g/l. The average removal efficiencies of chemical oxygen demand, total kjeldahl nitrogen and phosphate were 98.2 ± 1.3%, 88.3 ± 1.6% and 64.8 ± 1.0% within 8 days, respectively, while the average biomass productivity was 10.9 ± 1.1 g/m(2) · d. Accumulation into biomass, identified as the main nitrogen and phosphorus removal mechanism, accounted for 44.9 ± 0.4% and 61.6 ± 0.5% of total inlet nitrogen and phosphorus, respectively. Microscopic analysis showed the main algae species in the bioreactor were filamentous blue-green algae. Furthermore, denaturing gradient gel electrophoresis and 16S rDNA gene sequencing revealed that the main bacteria present in the photobioreactor were consortia with sequences similar to those of Flavobacteria, Gammaproteobacteria, Bacteroidia and Betaproteobacteria. This study explores a better understanding of an algae-bacteria system and offers new information on further usage of biomass accumulated during treatment.

Abstract:Photosynthetic bacteria (PSB) wastewater treatment is a new method with great resource recovery potential. In order to improve the biomass and valuable resources of bacteria in the wastewater treatment of photosynthetic bacteria, the effects of different light conditions on the efficiency of Proteiniphilum acetatigenes PSB-W in the treatment of sugar wastewater and the production of bacteria and carotenoids in the wastewater were analyzed from three aspects of light source, light intensity and light period. Under the principle of priority of pollutant removal efficiency, the red LED light was selected with the light intensity of 4000 Lux, 16L (light):8D (dark) for light cycle, and the best efficiency of wastewater removal and bacterial yield were obtained when the culture time was 3d, Chemical oxygen demand (COD) removal rate was 80.9%, bacterial OD600 was 0.907 ± 0.023, carotenoid yield was 7.83 ± 0.27 mg/L, respectively. These results show that PSB-W strains can provide more biomass and valuable substances in suitable environment. It provides a theoretical basis for the realization of wastewater recycling.
