Abstract:Bombyx mori silk fibroin (SF) is a promising natural biocompatible protein. However, its interaction with graphene oxide (GO) has never been studied and the resultant SF/GO matrix has not been used to direct stem cell fate. Herein, we found out that mixing SF molecules and GO nanosheets in an aqueous solution can trigger the assembly of SF nanoparticles into oriented nanofibrils due to the guidance of GO nanosheets, forming SF/GO films with unique nanotopographies and improved modulus upon the removal of the solvent. When GO mass percentage in the SF/GO films is 2 and 10%, the SF assemblies are necklace-like nanofibrils (assembled from loosely linked SF nanoparticles) and solid nanofibrils (assembled from densely linked SF nanoparticles) in the resultant films, termed SG2 and SG10, respectively. GO nanosheets guided the SF assembly into nanofibrils by triggering the structural change of SF molecules from random coils to β-sheets, as confirmed by Fourier transform infrared spectroscopy and circular dichroism measurements. Furthermore, oxidative groups in the GO nanosheets were reduced by the reducing groups in SF during the nanofibril formation according to X-ray photoelectron spectroscopy and Raman spectroscopy. The reduction of the oxidative groups in GO by SF was further verified by the good cell viability on the SF/GO films. The unique nanotopographies of the SF/GO films were found to accelerate the early cell adhesion and induce the osteogenic differentiation of human mesenchymal stem cells (MSCs) even in the absence of additional inducers in the medium. More importantly, SG10 presents a stronger capability in promoting early MSC adhesion by promoting F-actin assembly, increasing cell spreading area, and inducing the osteogenic differentiation of the MSCs by the unique SF/GO nanofibrous matrix. To the best of our knowledge, it is the first report that the SF/GO substrates can induce the osteogenic differentiation of MSCs in the absence of osteogenic differentiation medium. Therefore, SF/GO composite materials would have a potential application in the field of bone tissue engineering.
Polycaprolactone/graphene oxide/magnesium oxide as a novel composite scaffold for bone tissue engineering: Preparation and physical/biological assessment

Mechanically robust biocomposite scaffolds were fabricated by the electrospinning method for bone tissue engineering using a blend of polycaprolactone (PCL), and graphene oxide (GO) with magnesium oxide (MgO) nanoparticles. Physicochemical characteristics including morphology, tensile strength, swelling behavior, biodegradability, contact angle, cell viability, alkaline phosphatase (ALP) activity, mineralization ability, and osteogenic gene expression of the scaffolds were characterized. The addition of small amounts of GO and MgO nanoparticles significantly improved the morphological and mechanical properties of the PCL scaffold. The hydrophilicity, swelling ratio, and biodegradability of the developed composite scaffolds were improved. The PCL/GO/MgO scaffold demonstrated excellent biocompatibility and in-vitro biological performance with adipose-derived mesenchymal stem cells. Cell attachment, proliferation, ALP activity, mineral deposition, and osteogenesis-related gene expression were enhanced when compared to the pure PCL and PCL/GO scaffolds. Simultaneous incorporation of GO and MgO nanoparticles at a concentration of 2 wt% dramatically increased the differentiation of MSCs into osteoblasts. These findings may suggest that the hydrophilic properties and high protein adsorption of PCL/GO/MgO scaffold can stimulate cell proliferation, and nucleation to help improve bone mineralization. Enhanced biocompatibility of PLGA nanofibers with gelatin/nano-hydroxyapatite bone biomimetics incorporation.

Abstract:The biocompatibility of biomaterials is essentially for its application. The aim of current study was to evaluate the biocompatibility of poly(lactic-co-glycolic acid) (PLGA)/gelatin/nanohydroxyapatite (n-HA) (PGH) nanofibers systemically to provide further rationales for the application of the composite electrospun fibers as a favorable platform for bone tissue engineering. The PGH composite scaffold with diameter ranging from nano- to micrometers was fabricated by using electrospinning technique. Subsequently, we utilized confocal laser scanning microscopy (CLSM) and MTT assay to evaluate its cyto-compatibility in vitro. Besides, real-time quantitative polymerase chain reaction (qPCR) analysis and alizarin red staining (ARS) were performed to assess the osteoinductive activity. To further test in vivo, we implanted either PLGA or PGH composite scaffold in a rat subcutaneous model. The results demonstrated that PGH scaffold could better support osteoblasts adhesion, spreading, and proliferation and show better cyto-compatibility than pure PLGA scaffold. Besides, qPCR analysis and ARS showed that PGH composite scaffold exhibited higher osteoinductive activity owing to higher phenotypic expression of typical osteogenic genes and calcium deposition. The histology evaluation indicated that the incorporation of Gelatin/nanohydroxyapatite (GH) biomimetics could significantly reduce local inflammation. Our data indicated that PGH composite electrospun nanofibers possessed excellent cyto-compatibility, good osteogenic activity, as well as good performance of host tissue response, which could be versatile biocompatible scaffolds for bone tissue engineering.

Abstract:The effects of type 1 diabetes and key metabolic variables on brain structure are not well understood. Sensitive methods of assessing brain structure, such as voxel-based morphometry (VBM), have not previously been used to investigate central nervous system changes in a diabetic population. Using VBM, we compared type 1 diabetic patients aged 25-40 years with disease duration of 15-25 years and minimal diabetes complications with an age-matched, nondiabetic control group. We investigated whether lower than expected gray matter densities were present, and if so, whether they were associated with glycemic control and history of severe hypoglycemic events. In comparison with control subjects, diabetic patients showed lower density of gray matter in several brain regions. Moreover, in the patient group, higher HbA(1c) levels and severe hypoglycemic events were associated with lower density of gray matter in brain regions responsible for language processing and memory. Our study represents the first comprehensive study of gray matter density changes in type 1 diabetes and suggests that persistent hyperglycemia and acute severe hypoglycemia have an impact on brain structure.
Improved Biocompatibility and Osteogenesis of Porous Graphene Oxide/silk Fibroin Scaffold for Potential Applications in Bone Tissue Engineering

Abstract:To better mimic the inherent porous architecture of the natural bone, we fabricated graphene oxide/silk fibroin (GO/SF) scaffolds using a salting leaching method, followed by in situ reduction and reinforcement with ascorbic acid. SEM images revealed that the scaffolds exhibited an interconnectivity porous structure both on the surface and throughout the interior. We found that the pore size could be readily adjusted by varying the concentration of GO in the scaffolds. FTIR results demonstrated that the addition of GO didn't form chemical interaction between GO and SF, but it significantly increased the content of beta-sheet structure from 46.36 % to 51.12 % and improved the thermal stability of the scaffolds. Moreover, the reduced graphene oxide/silk fibroin (rGO/SF) scaffolds exhibited a superior capacity to promote the adhesion, proliferation, and mineral deposition of mouse mesenchymal stem cell (MSCs) when compared to pure SF scaffolds. Notably, the equilibrium Young's modulus of the scaffolds gradually increased from 5.5 +/- 0.5 KPa to 14.6 +/- 2.1 KPa with the increment of rGO content. Osteogenic differentiation with higher BSP (2. 84-fold) and RUNX2 (2.91-fold) expression level was also observed in 3 % rGO scaffold, suggesting a strong inclination towards bone formation. Based on these findings, the prepared rGO/SF scaffold in this study hold great potential as a viable option for bone tissue engineering.

Abstract:Novel three-dimensional porous silk fibroin/graphene oxide (SF/GO) nanocomposite scaffolds with different graphene oxide (GO) concentrations were prepared by using the freeze-drying technique. The obtained SF/GO scaffolds were characterized by thermogravimetric analysis, X-ray diffraction, scanning electron microscopy, Brunauer–Emmett–Teller isotherm and Fourier transform infrared spectroscopy techniques. The water absorption, compressive properties, porosity, degradation, biomineralization capability, cell attachment and cell viability of the composite scaffolds were studied as well. Cytocompatibility of the scaffolds was studied in vitro by employing the methylthiazoletetrazolium assay. The results showed that the presence of graphene oxide nanoparticles throughout the fibroin matrix led to an increase in water uptake and mechanical properties; at the same time, the porosity of the scaffolds was decreased. The cell adhesion results also indicated that human osteoblast cells (MG-63) could adhere to the surface of SF/GO nanocomposites and develop on them. These suggest that SF/GO nanocomposite scaffolds may be a good candidate for bone tissue engineering applications.

Abstract:Objective: The use of biocompatible scaffolds with appropriate characteristics to treat large bone defects has attracted significant attention. The main objective of the current study is to fabricate a 3D nanocomposite structure that contains green synthesized magnesium oxide nanoparticles (MgONPs) and bacterial cellulose (BC) nanofibres, as a bioscaffold for bone regeneration. Materials and methods: In this experimental study, Camellia sinensis extract was used as the green method to synthesize MgONPs. The synthesized hydrogels were evaluated for their porosity, morphology, degradation rate, mechanical features, cell attachment, and cytocompatibility. Osteogenic differentiation was assessed by alkaline phosphatase (ALP) activity, real-time reverse transcription-polymerase chain reaction (RT-PCR), and alizarin red staining. Results: MgONPs significantly increased both mechanical strength (P=0.009) and porosity (P=0.01) of the BC hydrogels. Human MG-63 osteoblast proliferation significantly increased in the MgONP-BC group compared to the pure BC group (P=0.003). Expression rates of both the ALP (P=0.001) and osteocalcin (OCN) genes were significantly enhanced in cells seeded on the MgONP-incorporated BC. MG-63 cells had significantly greater calcium deposition and ALP activity (P=0.002) on the MgONP-BC scaffold compared to the BC at day 21. Conclusion: The MgONP-BC scaffold can promote the osteogenic activity of osteoblast-like cells, which indicates its therapeutic potential for bone tissue regeneration.

Abstract:Nanocomposite hydrogels offer remarkable potential for applications in bone tissue engineering. They are synthesized through the chemical or physical crosslinking of polymers and nanomaterials, allowing for the enhancement of their behaviour by modifying the properties and compositions of the nanomaterials involved. However, their mechanical properties require further enhancement to meet the demands of bone tissue engineering. Here, we present an approach to improve the mechanical properties of nanocomposite hydrogels by incorporating polymer grafted silica nanoparticles into a double network inspired hydrogel (gSNP Gels). The gSNP Gels were synthesised via a graft polymerization process using a redox initiator. gSNP Gels were formed by grafting 2-acrylamido-2-methylpropanesulfonic acid (AMPS) as the first network gel followed by a sequential second network acrylamide (AAm) onto amine functionalized silica nanoparticles (ASNPs). We utilized glucose oxidase (GOx) to create an oxygen-free atmosphere during polymerization, resulting in higher polymer conversion compared to argon degassing. The gSNP Gels showed excellent compressive strengths of 13.9 ± 5.5 MPa, a strain of 69.6 ± 6.4%, and a water content of 63.4% ± 1.8. The synthesis technique demonstrates a promising approach to enhance the mechanical properties of hydrogels, which can have significant implications for bone tissue engineering and other soft tissue applications.

Abstract:Graphene oxide (GO) is widely used in bone tissue engineering due to its good biocompatibility and proliferation, and is often used in combination with other hydrogels, which not only reduces the cytotoxicity of GO but also improves the mechanical properties of the hydrogels. We developed injectable carboxymethyl chitosan (CMC)/hydroxyethyl cellulose (HEC)/β-tricalcium phosphate (β-TCP)/GO hydrogel via hydrogen bonding cross-linked between (CMC) and (HEC), also, calcium cross-linked by β-TCP was also involved to further improvement of mechanical properties of the hydrogel, and incorporate different concentration of GO in these hydrogel systems. The characterization of the novel hydrogel was tested by scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FT-IR). The swelling ratio and mechanical properties were investigated, the results showed that the addition of GO was able to reduce the swelling rate of hydrogels and improve their mechanical properties, with the best effect in the case of 1 mg/mL content. In vivo experimental studies showed that the hydrogel significantly promoted the osteogenic differentiation of rat bone marrow mesenchymal stem cells (rBMSCs), with the best effect at a concentration of 2 mg/mL. The results of the cellular experiments were similar. Therefore, the novel environment-friendly and non-toxic injectable CMC/HEC/β-TCP/GO hydrogel system may have potential applications in bone tissue engineering.

Abstract:Blend polymers composed of natural polymers are a ubiquitous biomaterial class due to their suitable mechanical and biological characterization. In the present study, composite scaffolds based on bacterial cellulose (BC)/silk fibroin (SF) with bioactive glass nanoparticles (BGNPs) were developed to enhance osteogenesis in human adipose derived stem cells (hASCs). The scanning electron microscopy (SEM) results of BGNPs indicated a spherical morphology and size ranging from 15 to 30 nm . The presence of BC and BGNPs reduced the pore diameter of SF scaffolds to about 210 ± 10 μm and 205 ± 10 μm, respectively, while increasing their compressive strength and compressive modulus. FTIR analyses proved the presence of BGNPs, BC and SF in the scaffolds. Flow cytometry data confirmed the surface markers for hASCs. The results also showed that BC and BGNPs addition to BC/SF scaffolds decreased degradation and swelling rate. The gene expression (Runx2, alkaline phosphatase and osteocalcin) studies signified the osteogenic potential of BGNPs in BC/SF scaffolds on hASCs. Eventually, the increased cell adhesion, viability and differentiation in the BC/SF and BC/SF/BGNPs composite scaffolds drawn from MTT, SEM, Alizarin red staining and alkaline phosphatase activity confirmed that these scaffolds promise to serve as a therapeutic candidate for bone defects.

Abstract:Tissue engineering applications in bone defect treatment have shown promising outcomes over the past decade. In this context, taking advantage of bioactive scaffolds mimicking the intrinsic composition of bone has resulted in remarkable effects on bone regeneration. In this study, Zirconium-based metal-organic framework, known as MOF 801, was synthesized and incorporated into the gelatin matrix as an osteoconductive agent to fabricate a nanocomposite bone scaffold by freeze-drying method. Physical and chemical characteristics of MOF 801 and nanocomposite scaffolds were analyzed using FE-SEM, EDS, XRD, ICP and FTIR. Besides, the biological activity of the prepared scaffolds was evaluated by MTT, alizarin red staining, crystal violet staining, and ALP assays. Inclusion of MOF 801 NPs into the scaffolds resulted in enhancement of their compressive strength up to 15±0.05 MPa. The nanocomposite samples containing MOF 801 NPs also exhibited favorable bioactive feature that was confirmed by their apatite-forming ability on their surfaces upon immersion in SBF solution. The Zr ion and fumarate release studies illustrated sustained release profiles from the gelatin matrix, leading to both antioxidant and anti-inflammatory properties. MTT assay results affirmed biocompatibility of scaffolds containing up to 5% w/w MOF 801, besides the crystal violet assay exhibited proper cell confluency and adhesion. Eventually, Alizarin red and ALP activity assays revealed that an increment of MOF 801 could induce calcium mineralization and Alkaline Phosphatase enzyme production in MG-63 cells confirming the potential of prepared scaffolds for related bone tissue engineering applications.

Abstract:GA: This study investigates 3D‐printed bone scaffolds, employing fused‐deposition modeling and graphene‐oxide (GO) coating to enhance biocompatibility. Analytical techniques confirm GO presence and effects. Computational fluid dynamics assesses shear stress, critical for cell proliferation. Mechanical tests show lower properties due to coating process. Biological responses reveal up to 220% increased viability with simultaneous GO coating and dynamic cell culture. Employing 3D printing bone scaffolds with various polymers is growing due to their biocompatibility, biodegradability, and good mechanical properties. However, their biological properties need modification to have fewer difficulties in clinical experiments. Herein, the fused‐deposition modeling technique is used to design triply‐periodic‐minimal‐surfaces polylactic‐acid scaffolds and evaluate their biological response under static and dynamic cell culture conditions. To enhance the biological response of 3D‐printed bone scaffolds, graphene‐oxide (GO) is coated on the surface of the scaffolds. Fourier‐transform infrared spectroscopy, X‐ray diffraction, and energy‐dispersion X‐ray analysis are conducted to check the GO presence and its effects. Also, computational fluid dynamics analysis is implemented to investigate the shear stress on the scaffold, which is a critical parameter for cell proliferation under dynamic cell culture conditions. Compression tests and contact‐angle measurements are performed to assess the GO effect on mechanical properties and wettability, respectively. Also, it was shown that surface‐treated scaffolds have lower mechanical properties and higher wettability than uncoated scaffolds. A perfusion bioreactor is used to study cell culture. Also, field‐emission‐scanning‐electron‐microscope and 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5 diphenyl‐tetrazolium‐bromide (MTT) assay analyses are conducted to observe cell viability and cell attachment. An increase of up to 220% in viability was achieved with GO and dynamic cell culture.

Abstract:Bioactive scaffolds fabricated from a combination of organic and inorganic biomaterials are a promising approach for addressing defects in bone tissue engineering. In the present study, a self-crosslinked nanocomposite hydrogel, composed of gelatin/aldehyde-modified xanthan (Gel-AXG) is successfully developed by varying concentrations of porous silicon nanoparticles (PSiNPs). The effect of PSiNPs incorporation on physical, mechanical, and biological performance of the nanocomposite hydrogel is evaluated. Morphological analysis reveals formation of highly porous 3D microstructures with interconnected pores in all nanocomposite hydrogels. Increased content of PSiNPs results in a lower swelling ratio, reduced porosity and pore size, which in turn impeded media penetration and slowed down the degradation process. In addition, remarkable enhancements in dynamic mechanical properties are observed in Gel-AXG-8%Si (compressive strength: 0.6223 MPa at 90 % strain and compressive modulus: 0.054 MPa), along with improved biomineralization ability via hydroxyapatite formation after immersion in simulated body fluid (SBF). This optimized nanocomposite hydrogel provides a sustained release of Si ions at safe dose levels. Furthermore, in-vitro cytocompatibility studies using MG-63 cells exhibited remarkable performance in terms of cell attachment, proliferation, and ALP activity for Gel-AXG-8%Si. These findings suggest that the prepared nanocomposite hydrogel holds promising potential as a scaffold for bone tissue engineering.

Abstract:Biomaterials that can harness the intrinsic osteogenic potential of stem cells offer a promising strategy to accelerate bone regeneration and repair. Previously, we had used methacrylated gelatin (GelMA)-based scaffolds to achieve bone formation from human mesenchymal stem cells (hMSCs). In this study, we aimed to further enhance hMSC osteogenesis by incorporating graphene oxide (GO)-based nanosheets into GelMA. In vitro results showed high viability and metabolic activities in hMSCs encapsulated in the newly developed nanocomposite. Incorporation of GO markedly increased mineralization within hMSC-laden constructs, which was further increased by replacing GO with silica-coated graphene oxide (SiGO). Mechanistic analysis revealed that the nanosheet enhanced the production, retention, and biological activity of endogenous bone morphogenetic proteins (BMPs), resulting in robust osteogenesis in the absence of exogenous osteoinductive growth factors. Specifically, the osteoinductive effect of the nanosheets was abolished by inhibiting the BMP signaling pathway with LDN-193189 treatment. The bone formation potential of the technology was further tested in vivo using a mouse subcutaneous implantation model, where hMSCs-laden GO/GelMA and SiGO/GelMA samples resulted in bone volumes 108 and 385 times larger, respectively, than the GelMA control group. Taken together, these results demonstrate the biological activity and mechanism of action of GO-based nanosheets in augmenting the osteogenic capability of hMSCs, and highlights the potential of leveraging nanomaterials such as GO and SiGO for bone tissue engineering applications.This study demonstrates the mechanism of action of graphene oxide nanosheets in augmenting bone regeneration.

Graphene Oxide Strengthens Gelatine through Non-Covalent Interactions with Its Amorphous Region

Abstract:Graphene oxide (GO) has attracted huge attention in biomedical sciences due to its outstanding properties and potential applications. In this study, we synthesized GO using our recently developed 1-pyrenebutyric acid-assisted method and assessed how the GO as a filler influences the mechanical properties of GO–gelatine nanocomposite dry films as well as the cytotoxicity of HEK-293 cells grown on the GO–gelatine substrates. We show that the addition of GO (0–2%) improves the mechanical properties of gelatine in a concentration-dependent manner. The presence of 2 wt% GO increased the tensile strength, elasticity, ductility, and toughness of the gelatine films by about 3.1-, 2.5-, 2-, and 8-fold, respectively. Cell viability, apoptosis, and necrosis analyses showed no cytotoxicity from GO. Furthermore, we performed circular dichroism, X-ray diffraction, Fourier-transform infrared spectroscopy, and X-ray photoelectron spectroscopy analyses to decipher the interactions between GO and gelatine. The results show, for the first time, that GO enhances the mechanical properties of gelatine by forming non-covalent intermolecular interactions with gelatine at its amorphous or disordered regions. We believe that our findings will provide new insight and help pave the way for potential and wide applications of GO in tissue engineering and regenerative biomedicine.

The "Yin and Yang" of Immunomodulatory Magnesium-Enriched Graphene Oxide Nanoscrolls Decorated Biomimetic Scaffolds in Promoting Bone Regeneration

Abstract:Tissue regeneration driven by immunomodulatory agents has emerged as a potential solution for repairing bone defects. However, the therapeutic benefits are compromised by disturbances in the pro- and anti-inflammatory balance. Here, using magnesium nanoparticles (MgNPs) as a template, magnesium-enriched graphene oxide nanoscrolls (MgNPs@GNSs) designed for combinational modulation of the inflammatory response are reported. First, the different effects of graphene oxide (GO) and magnesium ions (Mg2+ ) on Raw264.7 macrophage phenotype transformation are screened. The results reveal that GO activates inflammatory M1 macrophages, and that Mg2+ facilitates repolarization of M1 macrophages to the pro-healing M2 phenotype. With sustained release of Mg2+ , the MgNPs@GNS nanoplatform can orchestrate harmonious type 1 and type 2 inflammatory responses. Mg2+ decrease the internalization of GO and downregulate the nuclear factor kappa-B pathway, which is profoundly involved in the inflammatory process. A series of experiments show that the ordered inflammatory response induced by MgNPs@GNSs stimulates in vitro angiogenesis and osteogenesis through chemotactic, mitogenic, and morphogenic actions. Obvious vascularized bone regeneration is achieved in a rat cranial bone defect model via MgNPs@GNS deposited decellularized bone matrix scaffold. Therefore, the potential of using inherently therapeutic nanomedicine to modulate biomaterial-induced immune responses and thus enhance bone regeneration is demonstrated.
A New Shear-thinning Nanocomposite Hydrogel from GelMA- GO for Soft Tissue Engineering

Abstract:The use of bio ink materials for fabricating scaffolds is the best option to replace aggressive surgery. This study aims to formulate an injectable hydrogel with suitable structural, mechanical, and rheological properties for tissue engineering applications. In this respect, gelatin methacrylate (GelMA) nanocomposites with different amounts of the modified graphene oxide nanoplates (0.25, 0.5, 0.75, and 1 wt.%) were fabricated. The effect of the nanoplates on the structural, degradability, mechanical, rheological, and biological properties of the nanocomposites were evaluated. The results revealed that the functionalization of graphene oxide with dopamine (GOPD) resulted in the formation of a layer of polydopamine on the surface of the nanoplates, which developed the interaction of graphene oxide with GelMA chains. Adding GOPD to GelMA from 0 to 1 wt.% decreased the water absorption (0.3 times) and its weight loss percentage (0.2 times). The addition of 0.5 wt.% of GOPD to GelMA improved the compressive strength (7.5 times) and compressive modulus (7 times) compared to the GelMA hydrogel. Also, a similar amount of GOPD developed the thin shear properties of GelMA by three logarithmic units. Besides, the addition of the modified graphene oxide nanoplates to GelMA enhanced cell viability (93.26%) after 72 hours of cell seeding. The proper behavior of the GelMA hydrogel with 0.5% GOPD , and its stability during the 3D printing process was confirmed. Accordingly, the injectable GelMA hydrogel containing dopamine-modified graphene oxide could be a proper candidate for tissue engineering expenditures.

Abstract:Zwitterionic materials are widely applied in the biomedical field due to their excellent antimicrobial, non-cytotoxicity, and antifouling properties but have never been applied in bone tissue engineering. In this study, we synthesized a novel zwitterionic hydrogel incorporated with graphene oxide (GO) using maleic anhydride (MA) as a cross-linking agent by grafted L-cysteine (L-Cys) as the zwitterionic material on maleilated chitosan via click chemistry. The composition and each reaction procedure of the novel zwitterionic hydrogel were characterized via X-ray diffraction (XRD) and Fourier transformed infrared spectroscopy (FT-IR), while the morphology was imaged by scanning electron microscope (SEM). In vitro cell studies, CCK-8 and live/dead assay, alkaline phosphatase activity, W-B, and qRT-CR tests showed zwitterionic hydrogel incorporated with GO remarkably enhanced the osteogenic differentiation of bone mesenchymal stem cells (BMSCs); it is dose-dependent, and 2 mg/mL GO is the optimum concentration. In vivo tests also indicated the same results. Hence, these results suggested the novel zwitterionic hydrogel exhibited porous characteristics similar to natural bone tissue. In conclusion, the zwitterionic scaffold has highly biocompatible and mechanical properties. When GO was incorporated in this zwitterionic scaffold, the zwitterionic scaffold slows down the release rate and reduces the cytotoxicity of GO. Zwitterions and GO synergistically promote the proliferation and osteogenic differentiation of rBMSCs in vivo and in vitro. The optimal concentration is 2 mg/mL GO.
