Abstract:Tissue engineered constructs can serve as in vitro models for research and replacement of diseased or damaged tissue. As an emerging technology, 3D bioprinting enables tissue engineering through the ability to arrange biomaterials and cells in pre-ordered structures. Hydrogels, such as alginate (Alg), can be formulated as inks for 3D bioprinting. However, Alg has limited cell affinity and lacks the functional groups needed to promote cell growth. In contrast, graphene oxide (GO) can support numerous cell types and has been purported for use in regeneration of bone, neural and cardiac tissues. Here, GO was incorporated with 2% (w/w) Alg and 3% (w/w) gelatin (Gel) to improve 3D printability for extrusion-based 3D bioprinting at room temperature (RT; 25°C) and provide a 3D cellular support platform. GO was more uniformly distributed in the ink with our developed method over a wide concentration range (0.05%-0.5%, w/w) compared to previously reported GO containing bioink. Cell support was confirmed using adipose tissue derived stem cells (ADSCs) either seeded onto 3D printed GO scaffolds or encapsulated within the GO containing ink before direct 3D printing. Added GO was shown to improve cell-affinity of bioinert biomaterials by providing more bioactive moieties on the scaffold surface. 3D cell-laden or cell-seeded constructs showed improved cell viability compared to pristine (without GO) bio-ink-based scaffolds. Our findings support the application of GO for novel bio-ink formulation, with the potential to incorporate other natural and synthetic materials such as chitosan and cellulose for advanced in situ biosensing, drug-loading and release, and with the potential for electrical stimulation of cells to further augment cell function.
3D printed biocompatible graphene oxide, attapulgite, and collagen composite scaffolds for bone regeneration

Abstract:Tissue-engineered bone material is one of the effective methods to repair bone defects, but the application is restricted in clinical because of the lack of excellent scaffolds that can induce bone regeneration as well as the difficulty in making scaffolds with personalized structures. 3D printing is an emerging technology that can fabricate bespoke 3D scaffolds with precise structure. However, it is challenging to develop the scaffold materials with excellent printability, osteogenesis ability, and mechanical strength. In this study, graphene oxide (GO), attapulgite (ATP), type I collagen (Col I) and polyvinyl alcohol were used as raw materials to prepare composite scaffolds via 3D bioprinting. The composite materials showed excellent printability. The microcosmic architecture and properties was characterized by scanning electron microscopy, Fourier transform infrared and thermal gravimetric analyzer, respectively. To verify the biocompatibility of the scaffolds, the viability, proliferation and osteogenic differentiation of Bone Marrow Stromal Cells (BMSCs) on the scaffolds were assessed by CCK-8, Live/Dead staining and Real-time PCR in vitro. The composited scaffolds were then implanted into the skull defects on rat for bone regeneration. Hematoxylin-eosin staining, Masson staining and immunohistochemistry staining were carried out in vivo to evaluate the regeneration of bone tissue. The results showed that GO/ATP/COL scaffolds have been demonstrated to possess controlled porosity, water absorption, biodegradability and good apatite-mineralization ability. The scaffold consisting of 0.5% GO/ATP/COL have excellent biocompatibility and was able to promote the growth, proliferation and osteogenic differentiation of mouse BMSCs in vitro. Furthermore, the 0.5% GO/ATP/COL scaffolds were also able to promote bone regeneration of in rat skull defects. Our results illustrated that the 3D printed GO/ATP/COL composite scaffolds have good mechanical properties, excellent cytocompatibility for enhanced mouse BMSCs adhesion, proliferation, and osteogenic differentiation. All these advantages made it potential as a promising biomaterial for osteogenic reconstruction.

Abstract:Bioprinting has emerged as a powerful manufacturing platform for tissue engineering, enabling the fabrication of 3D living structures by assembling living cells, biological molecules, and biomaterials into these structures. Among various biomaterials, hydrogels have been increasingly used in developing bioinks suitable for 3D bioprinting for diverse human body tissues and organs. In particular, hydrogel blends combining gelatin and gelatin methacryloyl (GelMA; "GG hydrogels") receive significant attention for 3D bioprinting owing to their many advantages, such as excellent biocompatibility, biodegradability, intrinsic bioactive groups, and polymer networks that combine the thermoresponsive gelation feature of gelatin and chemically crosslinkable attribute of GelMA. However, GG hydrogels have poor electroactive properties, which hinder their applications in neural tissue engineering where electrical conductivity is required. To overcome this problem, in this study, a small amount of highly electroactive graphene oxide (GO) was added in GG hydrogels to generate electroactive hydrogels for 3D bioprinting in neural tissue engineering. The incorporation of GO nanoparticles slightly improved mechanical properties and significantly increased electrical conductivity of GG hydrogels. All GO/GG composite hydrogels exhibited shear thinning behavior and sufficient viscosity and hence could be 3D printed into 3D porous scaffolds with good shape fidelity. Furthermore, bioinks combining rat bone marrow-derived mesenchymal stem cells (rBMSCs) with GO/GG composite hydrogels could be 3D bioprinted into GO/GG constructs with high cell viability. GO nanoparticles in the constructs provided ultraviolet (UV) shading effect and facilitated cell survival during UV exposure after bioprinting. The GO/GG composite hydrogels appear promising for 3D bioprinting applications in repairing damaged neural tissues.

Abstract:Journal of Bioactive and Compatible Polymers, Ahead of Print. Oral and maxillofacial tumors, trauma and infections are the main causes of jaw defects, whose clinical treatment is very complicated. With the development of biological tissue engineering, many biological materials have been widely used in various fields of stomatology, and they play a very important role in the repair and replacement of maxillofacial bone defects. In this study, we intended to prepare a graphene oxide/hyaluronic acid/chitosan (GO/HA/CS) composite hydrogel with different mass ratios of GO: 0.1% (0.1% GO/HA/CS), 0.25% (0.25% GO/HA/CS), 0.5% (0.5% GO/HA/CS), and 1% (1% GO/HA/CS), prepare it into a multilayered and stable composite scaffold through 3D-printing technology, observe the surface morphology of the composite scaffold through scanning electron microscopy (SEM), and then test its physical and chemical properties, mechanical properties, water swelling rate, in vitro degradation and other material properties. Moreover, the biological performance of the GO/HA/CS composite scaffold was studied through experiments, such as cell morphology observation, cell adhesion, cell proliferation, and live-dead cell staining. The results showed that through chemical cross-linking and 3D-printing technology, a porous (pore size: 450–580 μm) and multilayered GO/HA/CS biological scaffold could be successfully constructed, and its surface was an interconnected microporous structure, and the porosity decreased (94%−40%) gradually with the increase of GO. Meanwhile, with the change in GO concentration, some mechanical properties of the scaffold could be improved, such as water swelling rate, degradation rate, and elastic modulus. In addition, the composite scaffold with the appropriate amount of GO had almost no cytotoxicity and could promote cell growth and proliferation, especially 0.25% GO/HA/CS composite scaffold. Consequently, the 0.25% GO/HA/CS composite scaffold had excellent biological material properties and good biocompatibility with osteoblasts, which may provide a new idea for the repair of jaw defects.

Abstract:Currently, a major challenge in material engineering is to develop a cell-safe biomaterial with significant utility in processing technology such as 3D bioprinting. The main goal of this work was to optimize the composition of a new graphene oxide (GO)-based bioink containing additional extracellular matrix (ECM) with unique properties that may find application in 3D bioprinting of biomimetic scaffolds. The experimental work evaluated functional properties such as viscosity and complex modulus, printability, mechanical strength, elasticity, degradation and absorbability, as well as biological properties such as cytotoxicity and cell response after exposure to a biomaterial. The findings demonstrated that the inclusion of GO had no substantial impact on the rheological properties and printability, but it did enhance the mechanical properties. This enhancement is crucial for the advancement of 3D scaffolds that are resilient to deformation and promote their utilization in tissue engineering investigations. Furthermore, GO-based hydrogels exhibited much greater swelling, absorbability and degradation compared to non-GO-based bioink. Additionally, these biomaterials showed lower cytotoxicity. Due to its properties, it is recommended to use bioink containing GO for bioprinting functional tissue models with the vascular system, e.g., for testing drugs or hard tissue models.

Abstract:GA: This study investigates 3D‐printed bone scaffolds, employing fused‐deposition modeling and graphene‐oxide (GO) coating to enhance biocompatibility. Analytical techniques confirm GO presence and effects. Computational fluid dynamics assesses shear stress, critical for cell proliferation. Mechanical tests show lower properties due to coating process. Biological responses reveal up to 220% increased viability with simultaneous GO coating and dynamic cell culture. Employing 3D printing bone scaffolds with various polymers is growing due to their biocompatibility, biodegradability, and good mechanical properties. However, their biological properties need modification to have fewer difficulties in clinical experiments. Herein, the fused‐deposition modeling technique is used to design triply‐periodic‐minimal‐surfaces polylactic‐acid scaffolds and evaluate their biological response under static and dynamic cell culture conditions. To enhance the biological response of 3D‐printed bone scaffolds, graphene‐oxide (GO) is coated on the surface of the scaffolds. Fourier‐transform infrared spectroscopy, X‐ray diffraction, and energy‐dispersion X‐ray analysis are conducted to check the GO presence and its effects. Also, computational fluid dynamics analysis is implemented to investigate the shear stress on the scaffold, which is a critical parameter for cell proliferation under dynamic cell culture conditions. Compression tests and contact‐angle measurements are performed to assess the GO effect on mechanical properties and wettability, respectively. Also, it was shown that surface‐treated scaffolds have lower mechanical properties and higher wettability than uncoated scaffolds. A perfusion bioreactor is used to study cell culture. Also, field‐emission‐scanning‐electron‐microscope and 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5 diphenyl‐tetrazolium‐bromide (MTT) assay analyses are conducted to observe cell viability and cell attachment. An increase of up to 220% in viability was achieved with GO and dynamic cell culture.

Abstract:In this work, one-dimensional and layered parallel folding of cobalt oxalate nanostructures have been selectively prepared by a one-step, template-free, water-controlled precipitation approach by simply altering the solvents used at ambient temperature and pressure. Encouragingly, the feeding order of solutions played an extraordinary role in the synthesis of nanorods and nanowires. After calcination in air, the as-prepared cobalt oxalate nanostructures were converted to mesoporous Co(3)O(4) nanostructures while their original frame structures were well maintained. The phase composition, morphology, and structure of the as-obtained products were studied in detail. Electrochemical properties of the Co(3)O(4) electrodes were carried out using cyclic voltammetry (CV) and galvanostatic charge-discharge measurements by a three-electrode system. The electrochemical experiments revealed that the layered parallel folding structure of mesoporous Co(3)O(4) exhibited higher capacitance compared to that of the nanorods and nanowires. A maximum specific capacitance of 202.5 F g (-1) has been obtained in 2 M KOH aqueous electrolyte at a current density of 1 A g(-1) with a voltage window from 0 to 0.40 V. Furthermore, the specific capacitance decay after 1000 continuous charge-discharge cycles was negligible, revealing the excellent stability of the electrode. These characteristics indicate that the mesoporous Co(3)O(4) nanostructures are promising electrode materials for supercapacitors.
Wood-based nanocellulose and bioactive glass modified gelatin–alginate bioinks for 3D bioprinting of bone cells

Abstract:A challenge in the extrusion-based bioprinting is to find a bioink with optimal biological and physicochemical properties. The aim of this study was to evaluate the influence of wood-based cellulose nanofibrils (CNF) and bioactive glass (BaG) on the rheological properties of gelatin-alginate bioinks and the initial responses of bone cells embedded in these inks. CNF modulated the flow behavior of the hydrogels, thus improving their printability. Chemical characterization by SEM-EDX and ion release analysis confirmed the reactivity of the BaG in the hydrogels. The cytocompatibility of the hydrogels was shown to be good, as evidenced by the viability of human osteoblast-like cells (Saos-2) in cast hydrogels. For bioprinting, 4-layer structures were printed from cell-containing gels and crosslinked with CaCl2. Viability, proliferation and alkaline phosphatase activity (ALP) were monitored over 14 d. In the BaG-free gels, Saos-2 cells remained viable, but in the presence of BaG the viability and proliferation decreased in correlation with the increased viscosity. Still, there was a constant increase in the ALP activity in all the hydrogels. Further bioprinting experiments were conducted using human bone marrow-derived mesenchymal stem cells (hBMSCs), a clinically relevant cell type. Interestingly, hBMSCs tolerated the printing process better than Saos-2 cells and the ALP indicated BaG-stimulated early osteogenic commitment. The addition of CNF and BaG to gelatin-alginate bioinks holds great potential for bone tissue engineering applications.

Abstract:There is a growing demand for bone graft substitutes that mimic the extracellular matrix properties of the native bone tissue to enhance stem cell osteogenesis. Composite hydrogels containing human bone allograft particles are particularly interesting due to inherent bioactivity of the allograft tissue. Here, we report a novel photocurable composite hydrogel bioink for bone tissue engineering. Our composite bioink is formulated by incorporating human allograft bone particles in a methacrylated alginate formulation to enhance adult human mesenchymal stem cell (hMSC) osteogenesis. Detailed rheology and printability studies confirm suitability of our composite bioinks for extrusion-based 3D bioprinting technology. In vitro studies reveal high cell viability (~90%) for hMSCs up to 28 days of culture within 3D bioprinted composite scaffolds. When cultured within bioprinted composite scaffolds, hMSCs show significantly enhanced osteogenic differentiation as compared to neat scaffolds based on alkaline phosphatase activity, calcium deposition, and osteocalcin expression.

Abstract:The scaffold structures formed by the incorporation of reduced graphene oxide (RGO) nanomaterials can be studied to improve the 3D matrix properties in the applications of bone regeneration. On the other hand, RGO has delayed the incorporation into the mixtures that were determined to generate scaffolds on using 3D printing because of its poor colloidal stability of particles and weak solubility in water. Moreover, to produce RGO, the graphene oxide (GO) was treated with hydrazine hydrate solution, which was well known as a reducing agent that is highly hazardous. To outcome these drawbacks, an unique eco-friendly method was established to gather scaffolds that were 3D printed by the incorporation of RGO. This was obtained using the Gallic acid induced in situ reduction of the GO already existing on the gelatin/chitosan/tricalcium phosphate scaffolds. Moreover, scaffolds also exhibited antibacterial activity without conceding osteoblasts’ cell proliferation and viability. Furthermore, the obtained scaffolds showed good osteoblasts proliferation and viability indicating their possibility for applications in bone fracture healing.

Improved Biocompatibility and Osteogenesis of Porous Graphene Oxide/silk Fibroin Scaffold for Potential Applications in Bone Tissue Engineering

Abstract:To better mimic the inherent porous architecture of the natural bone, we fabricated graphene oxide/silk fibroin (GO/SF) scaffolds using a salting leaching method, followed by in situ reduction and reinforcement with ascorbic acid. SEM images revealed that the scaffolds exhibited an interconnectivity porous structure both on the surface and throughout the interior. We found that the pore size could be readily adjusted by varying the concentration of GO in the scaffolds. FTIR results demonstrated that the addition of GO didn't form chemical interaction between GO and SF, but it significantly increased the content of beta-sheet structure from 46.36 % to 51.12 % and improved the thermal stability of the scaffolds. Moreover, the reduced graphene oxide/silk fibroin (rGO/SF) scaffolds exhibited a superior capacity to promote the adhesion, proliferation, and mineral deposition of mouse mesenchymal stem cell (MSCs) when compared to pure SF scaffolds. Notably, the equilibrium Young's modulus of the scaffolds gradually increased from 5.5 +/- 0.5 KPa to 14.6 +/- 2.1 KPa with the increment of rGO content. Osteogenic differentiation with higher BSP (2. 84-fold) and RUNX2 (2.91-fold) expression level was also observed in 3 % rGO scaffold, suggesting a strong inclination towards bone formation. Based on these findings, the prepared rGO/SF scaffold in this study hold great potential as a viable option for bone tissue engineering.
Alendronate releasing silk fibroin 3D bioprinted scaffolds for application in bone tissue engineering: Effects of alginate concentration on printability, mechanical properties and stability

Abstract:3D bioprinting uses biomaterials combined with cells to develop living constructs. This study explores the optimization of natural polymers, including silk fibroin, gelatin, and alginate, as bioink for 3D bioprinting in bone tissue engineering. The physicochemical properties of the bioink were thoroughly examined, revealing high water uptake and reduced degradation rate due to the addition of silk fibroin. The compressive modulus increased with higher alginate concentration. Rheological analysis confirmed shear-thinning properties and viscoelasticity of the ink. Through meticulous parameter optimization, the ink achieved the highest print accuracy with 4 % w/v alginate content. The printed scaffolds exhibited both macro and micro porosity, making them suitable for bone tissue regeneration. Furthermore, the scaffolds remained stable in culture medium for 36 days. The optimal composition for the hydrogel was determined to be a blend of 5 % fibroin, 7 % gelatin, and 4 % alginate in equal ratios. The bioink demonstrated excellent biocompatibility and, when supplemented with alendronate, enhanced alkaline phosphatase activity in MG-63 osteoblast-like cells. This finding indicates the commitment of cells toward the osteoblastic phenotype. Overall, this study successfully optimized the bioink and bioprinting process for bone tissue engineering applications, highlighting its promising potential for future advancements in the field.
3D Bioprinting of shear-thinning hybrid bioinks with excellent bioactivity derived from gellan/alginate and thixotropic magnesium phosphate-based gels

Abstract:3D Bioprinting is expected to become a strong tool for regenerative medicine, but satisfactory bioinks for the printing of constructs containing living cells are lacking due to the rigorous requirement of high printability and biocompatibility, which are often contradictory. Here, we have reported the development of a novel hybrid bioink by combining rigid gellan gum (GG), flexible sodium alginate (SA), and a bioactive substance thixotropic magnesium phosphate-based gel (TMP-BG). The ratio of these components was first optimized to obtain satisfactory gelating, mechanical, rheological, and printing properties. The formulated hybrid GG-SA/TMP-BG bioink had a good printability due to the shear-thinning and its multiple cross-linking by Mg2+ and Ca2+. The tunable mechanical performance of the hybrid bioink could simulate various extracellular matrices of the different tissues and support integrity of 3D printing constructs. Moreover, the hybrid bioink induced apatite deposition during immersion in simulated body fluids, and also promoted cell proliferation in vitro. MG-63 osteosarcoma cells were dispersed in the bioink and printed into 3D constructs. The cells exhibited good cell survival due to the shear-thinning property of the bioink and the ion concentration used for cross-linking. The proliferation rate of the cells also significantly exceeded those in non-printed samples. Confocal microscopy revealed a homogeneous distribution of cells in the printed constructs, and survival for more than 7 d. In vivo animal experiments showed that the hybrid bioink without cells could induce osteochondral repair. Therefore, this hybrid bioink has good printability, biocompatibility, mechanical support, and bioactivity, which is expected to have promising applications in 3D bioprinting.
Direct Ink Writing of Graphene Oxide Reinforced 13-93B1 Bioactive Glass Scaffolds for Bone Tissue Engineering Applications

Abstract:Graphene-reinforced bioactive glass scaffolds have gained significant attention in the field of bone tissue engineering due to their unique combination of mechanical strength, bioactivity, and electrical conductivity. Additive manufacturing techniques, such as 3D printing, provide a versatile platform for fabricating these scaffolds with precise control over their architecture and composition. Consequently, in this work, we have fabricated graphene oxide (GO)-reinforced 13-93B1 bioactive glass scaffold using the direct ink writing technique. A Pluronic F-127- based ink was prepared for scaffold fabrication, and its rheological properties were assessed for shear thinning behaviour, structural support, and recovery. Further, the fabricated scaffolds were characterized using micro-computed tomography, scanning electron microscopy, and energy dispersive x-ray spectroscopy. Additionally, computational fluid dynamics simulations with Dulbecco’s modified eagle medium and blood were performed to evaluate the perfusion kinetics of the scaffolds. The inclusion of GO enhanced the compressive strength of the fabricated scaffolds by ∼225%. The morphological characterization based on micro-computed tomography showed that additively manufactured scaffolds have appropriate porosity, pore size, pore throat size, and interconnectivity. The live-dead assay results showed no cytotoxicity towards C2C12 mouse myoblast cells. Also, cell adhesion and cell viability results show better cell growth on the nanocomposite scaffolds. Overall, the fabricated scaffold is found suitable for bone tissue engineering applications.

Abstract:Hydrogels have been widely applied to the fabrication of tissue engineering scaffolds via three-dimensional (3D) bioprinting because of their extracellular matrix-like properties, capacity for living cell encapsulation, and shapeable customization depending on the defect shape. However, the current hydrogel scaffolds show limited regeneration activity, especially in the application of periodontal tissue regeneration. In this study, we attempted to develop a novel multi-component hydrogel that possesses good biological activity, can wrap living cells for 3D bioprinting and can regenerate periodontal soft and hard tissue. The multi-component hydrogel consisted of gelatin methacryloyl (GelMA), sodium alginate (SA) and bioactive glass microsphere (BGM), which was first processed into hydrogel scaffolds by cell-free 3D printing to evaluate its printability and in vitro biological performances. The cell-free 3D-printed scaffolds showed uniform porous structures and good swelling capability. The BGM-loaded scaffold exhibited good biocompatibility, enhanced osteogenic differentiation, apatite formation abilities and desired mechanical strength. The composite hydrogel was further applied as a bio-ink to load with mouse bone marrow mesenchymal stem cells (mBMSCs) and growth factors (BMP2 and PDGF) for the fabrication of a scaffold for periodontal tissue regeneration. The cell wrapped in the hydrogel still maintained good cellular vitality after 3D bioprinting and showed enhanced osteogenic differentiation and soft tissue repair capabilities in BMP2- and PDGF-loaded scaffolds. It was noted that after transplantation of the cell- and growth factor-laden scaffolds in Beagle dog periodontal defects, significant regeneration of gingival tissue, periodontal ligament, and alveolar bone was detected. Importantly, a reconstructed periodontal structure was established in the treatment group eight weeks post-transplantation of the scaffolds containing the cell and growth factors. In conclusion, we developed a bioactive composite bio-ink for the fabrication of scaffolds applicable for the reconstruction and regeneration of periodontal tissue defects.

Abstract:Bone defects as a common orthopedic disease lead to severe pains over a long period. Scaffolds are novel approaches in tissue engineering to treat bone problems and deal with their challenges. Here for the first time, freeze-drying and salt etching techniques are utilized to develop 3D porous polyurethane (PU) scaffolds containing different percentages of graphene oxide (GO; 0, 0.1, 0.3, and 0.5 wt.%) for bone tissue engineering applications. The morphologies of scaffolds, physicochemical properties, the degree of crystallinity, and hydrophilicity were evaluated by SEM, FTIR, XRD, and water contact angle assay, respectively. The porosity, degradation behavior, compressive strength, and elastic modulus of 3D porous scaffolds were also determined. To assess the scaffold bioactivity, the morphology of the deposited calcium phosphate layer on the scaffold with macro-structure was evaluated by SEM images. The viability and adhesion of MG63 osteoblast-like cells cultured on the fabricated scaffolds were examined by MTT assay and SEM images, respectively. The results show that adding GO particles not only had no effect on the interconnectivity and porosity of 3D porous macroscopic structures of neat PU but also smaller and more uniformed microscopically pores were obtained. The crystallinity, water contact angle, and weight loss of scaffolds increased as the higher GO concentrations were employed. Followed by increasing GO contents from 0 to 0.5 wt.%, the compressive strength and Young's modulus were increased by 232% and 245%, respectively. The bioactivity of scaffolds was fostered as GO concentration increased. Although, the MTT assay proved the biocompatibility of PU scaffolds containing 0.1 and 0.3 wt.% GO, the samples loaded with 0.5 GO had a negative impact on the viability of MG63 cell lines. In conclusion, the present study demonstrates a high potential of PU scaffolds loaded with 0.1 and 0.3 wt.% GO particles in bone tissue engineering applications.

Abstract:Background: Complex bone defects are challenging to treat. Autografting is the gold standard for regenerating bone defects; however, its limitations include donor-site morbidity and increased surgical complexity. Advancements in 3D bioprinting (3DBP) offer a promising alternative for viable bone grafts. In this experiment, gels composed of varying levels of gelatin methacrylate (GelMA) and hydroxyapatite (HA) and gelatin concentrations are explored. The objective was to increase the hydroxyapatite content and find the upper limit before the printability was compromised and determine its effect on the mechanical properties and cell viability. Methods: Design of Experiments (DoE) was used to design 13 hydrogel bioinks of various GelMA/HA concentrations. These bioinks were assessed in terms of their pipettability and equilibrium modulus. An optimal bioink was designed using the DoE data to produce the greatest stiffness while still being pipettable. Three bioinks, one with the DoE-designed maximal stiffness, one with the experimentally defined maximal stiffness, and a literature-based control, were then printed using a 3D bioprinter and assessed for print fidelity. The resulting hydrogels were combined with human bone-marrow-derived mesenchymal stromal cells (hMSCs) and evaluated for cell viability. Results: The DoE ANOVA analysis indicated that the augmented three-level factorial design model used was a good fit (p 0.05). There was, however, a significantly lower cell viability in the gel composed of 12.3% GelMA, 15.7% HA, and 2% gelatin compared to the other gels with a lower HA concentration (p < 0.05), showing that a higher HA content or print pressure may be cytotoxic within hydrogels. Conclusions: Extrusion-based 3DBP offers significant advantages for bone–tissue implants due to its high customizability. This study demonstrates that it is possible to create printable bone-like grafts from GelMA and HA with an increased HA content, favorable mechanical properties (145 kPa), and a greater than 80% cell viability.

Abstract:In the field of bone tissue engineering, porous hydroxyapatite (HA) bone repair scaffolds with controlled architecture and porosity are challenging to manufacture and lack sufficient osteoinductive activity, which limit its wider clinical application. This study fabricated porous graphene oxide (GO)/HA composite ceramic scaffolds using digital light processing (DLP) technology. The properties of the composite ceramic slurry and the process parameters of curing, degreasing, and sintering were initially examined to ensure molding accuracy. The research showed that a small amount of GO (0.1–0.4 wt%) could better maintain the pore structure and improve the mechanical properties of composite ceramics. Porous scaffolds with pore sizes of 300–400 μm were prepared for co-culture with rat bone marrow mesenchymal stem cells to detect biocompatibility. The results showed that all scaffolds have no cytotoxicity. Compared with HA, the GO/HA ceramic scaffold significantly promoted the cell adhesion, proliferation, and expression of osteogenesis-related genes; GO (0.1–0.2 wt%) /HA scaffolds exhibited superior alkaline phosphatase activity and more effective bone mineralization, showing enhanced osteoinductivity. The porous GO/HA composite scaffold fabricated by DLP presented a strong potential for repairing bone defects.
