Abstract:There is an increasing interest in biodegradable materials, such as magnesium, for orthopedic implants. This is driven by their potential to address challenges like stress shielding and the need for secondary removal surgery. In this study, biodegradable magnesium alloys were produced using the Vacuum Induction Casting technique. The impact of micro-alloying Zn and Ca in Mg-xZn-0.2Ca (x= 0.1, 0.2, 0.3, and 0.4 wt%) alloys on corrosion resistance, cytocompatibility, and early-stage inflammatory response was investigated. XRD and SEM-EDS analysis confirmed the presence of Ca2Mg6Zn3 secondary phases in all alloys. The Mg-0.3Zn-0.2Ca alloy exhibited the lowest corrosion rate and an elastic modulus of 33.7 GPa, resembling that of natural bone. Electrochemical measurements indicated a correlation between grain size and secondary phase volume fraction in explaining corrosion behaviour. In vitro degradation in simulated body fluid (SBF) for 21 days showed hydroxyapatite formation on alloy surfaces, aligning with electrochemical studies. In vitro cytotoxicity tests demonstrated the cytocompatibility of all alloys, with Mg-0.3Zn-0.2Ca having the highest cell viability over a 6-day cell culture. Investigation into the inflammatory response with RAW-Blue macrophages revealed the anti-inflammatory properties of Mg-0.3Zn-0.2Ca alloys. Micro-alloying with 0.3 wt% Zn and 0.2 wt% Ca enhanced mechanical properties, corrosion resistance, cytocompatibility, and immunomodulatory properties. This positions the Mg-0.3Zn-0.2Ca alloy as a promising biodegradable implant for bone fixation applications.

In Vitro and in Vivo Studies on Biomedical Magnesium Low-Alloying with Elements Gadolinium and Zinc for Orthopedic Implant Applications

Abstract:Ternary magnesium alloys with low combined addition of elements gadolinium and zinc were developed in the present work, with their microstructures, mechanical properties, in vitro degradation behaviors, and cytotoxicity being systematically studied. Furthermore, the Mg-1.8Zn-0.2Gd alloy, with the best in vitro performance, was implanted into Sprague Dawley rats to examine its in vivo degradation performance for up to 6 months. It was found that Mg-1.8Zn-0.2Gd, composed of a single α-Mg phase, owned excellent strength and toughness that were comparable to the CE marked MAGNEZIX, the mischmetal added Mg alloy. Owing to the uniform single-phased microstructure, the degradation rate of this alloy was around 0.12 mm/y measured by electrochemical testing, which was comparable to high purity magnesium. Moreover, the Mg-1.8Zn-0.2Gd alloy exhibited no cytotoxicity to L929, MG63, and VSMC cells. In vivo degradation characterized by micro-computed tomography revealed that the Mg-1.8Zn-0.2Gd implant could maintain structural integrity in the first 2 months, and serious degradation could be observed after 6 months. A remarkable 100% survival rate of experimental animals was observed with no negative effects on bone tissues. The implant and the surrounding bone were well integrated within 2 months, implying good biocompatibility and osteoconductivity of the experimental alloy. On the basis of the above findings, the feasibility of Mg-Zn-Gd alloys for use as orthopedic implants was systematically discussed. This study provides a new strategy for development of high-performance Mg-rare earth (RE)-based alloys with superior mechanical properties and corrosion resistance while effectively avoiding the possible standing toxic effect of RE elements.

Abstract:In this study, a specific Mg–Zn–RE alloy membrane with 6 wt.% zinc and 2.7 wt.% rare earth elements (Y, Gd, La and Ce) was prepared to investigate implant degradation, transport mechanism and guide bone regeneration in vivo. The Mg-membrane microstructure and precipitates were characterized by the scanning electron microscopy (SEM) and the transmission electron microscopy (TEM). The Mg-membrane degradation process and effect on osteogenesis were investigated in a critical-sized rat calvarial defect model via micro-CT examination and hard tissue slicing after 2-, 5- and 8-week implants. Then, the distribution of elements in organs after 1-, 2- and 4-weeks implantation was examined to explore their transportation routes. Results showed that two types of precipitates had formed in the Mg–membrane after a 10-h heat treatment at 175 °C: γ-phase MgZn precipitation with dissolved La, Ce and Gd, and W-phase Mg3(Y, Gd)2Zn3 precipitation rich in Y and Gd. In the degradation process of the Mg-membrane, the Mg matrix degraded first, and the rare earth-rich precipitation particles were transferred to a more stable phosphate compound. The element release rate was dependent on the precipitate type and composition. Rare earth elements may be transported mainly through the lymph system. The defects were repaired rapidly by the membranes. The Mg-membrane used in the present study showed excellent biocompatibility and enhanced bone formation in the vicinity of the implants.
Hyperproinsulinaemia in elderly subjects: evidence for age-related pancreatic β-cell dysfunction

Abstract:SUMMARY. We investigated whether pancreatic pathogenesis of glucose intolerance in the elderly by comparing plasma glucose, immunoreactive insulin, C-peptide and proinsulin concentrations during a 100-g oral glucose load in six healthy older male volunteers [aged 69 (65·9-74·9) years; median (95% confidence limits)] and seven young male controls [aged 24·0 (21·9-26·3) years]. Fasting and integrated concentrations of glucose and C-peptide were similar in the elderly and young. Although fasting, insulin and proinsulin levels were similar, integrated insulin (P=0·05) and proinsulin (P< 0·05) concentrations were higher in the elderly than in controls. Insulin resistance, measured as homeostasis model assessment, was greater (P< 0·05) in the elderly than in controls. Elderly men had greater molar ratios of integrated insulin:C-peptide (P< 0·05) and proinsulin:Cpeptide (P< 0·01) but their respective fasting molar ratios were similar when compared with controls. Pancreatic β-cell secretion in older subjects, as assessed by C-peptide concentrations, was inappropriately low in the presence of insulin resistance. Their post-prandial 'hyperinsulinaemia' is probably due to a combination of hyperproinsulinaemia and reduced metabolic clearance of insulin. Older subjects had disproportionately high proinsulin to C-peptide levels, suggesting pancreatic β-cell dysfunction. These results are consistent with the notion that decreased insulin sensitivity and pancreatic β-cell dysfunction may predispose the elderly to glucose intolerance.
In Vitro and in Vivo Investigation on Biodegradable Mg-Li-Ca Alloys for Bone Implant Application

Abstract:Magnesium alloys show promise for application in orthopedic implants, owing to their biodegradability and biocompatibility. In the present study, ternary Mg-(3.5, 6.5 wt%) Li-(0.2, 0.5, 1.0 wt%) Ca alloys were developed. Their mechanical strength, corrosion behavior and cytocompatibility were studied. These alloys showed improved mechanical strength than pure Mg and exhibited suitable corrosion resistance. Furthermore, Mg-3.5Li-0.5Ca alloys with the best in vitro performance were implanted intramedullary into the femurs of mice for 2 and 8 weeks. In vivo results revealed a significant increase in cortical bone thickness around the Mg-3.5Li-0.5Ca alloy rods, without causing any adverse effects. Western blotting and immunofluorescence staining of β-catenin illustrated that Mg-3.5Li-0.5Ca alloy extracts induced osteogenic differentiation of human bone marrow-derived mesenchymal stem cells (hBMMSCs) through the canonical Wnt/β-catenin pathway. Our studies demonstrate that Mg-3.5Li-0.5Ca alloys hold much promise as candidates for the facilitation of bone implant application.
Corrosion and bone healing of Mg-Y-Zn-Zr-Ca alloy implants: Comparative in vivo study in a non-immobilized rat femoral fracture model

Abstract:Biodegradable magnesium (Mg) alloys exhibit improved mechanical properties compared to degradable polymers while degrading in vivo circumventing the complications of permanent metals, obviating the need for surgical removal. This study investigated the safety and efficacy of Mg-Y-Zn-Zr-Ca (WZ42) alloy compared to non-degradable Ti6Al4V over a 14-week follow-up implanted as pins to fix a full osteotomy in rat femurs and as wires wrapped around the outside of the femurs as a cerclage. We used a fully load bearing model allowing implants to intentionally experience realistic loads without immobilization. To assess systemic toxicity, blood cell count and serum biochemical tests were performed. Livers and kidneys were harvested to observe any accumulation of alloying elements. Hard and soft tissues adjacent to the fracture site were also histologically examined. Degradation behavior and bone morphology were determined using micro-computed tomography scans. Corrosion occurred gradually, with degradation seen after two weeks of implantation with points of high stress observed near the fracture site ultimately resulting in WZ42 alloy pin fracture. At 14 weeks however, normal bone healing was observed in femurs fixed with the WZ42 alloy confirmed by the presence of osteoid, osteoblast activity, and new bone formation. Blood testing exhibited no significant changes arising from the WZ42 alloy compared to the two control groups. No recognizable differences in the morphology and more importantly, no accumulation of Mg, Zn, and Ca in the kidney and liver of rats were observed. These load bearing model results collectively taken, thus demonstrate the feasibility for use of the Mg-Y-Zn-Zr-Ca alloy for long bone fracture fixation applications.

Abstract:Individual studies in patient-oriented research, whether described as "comparative effectiveness" or using other terms, are based on underlying methodological designs. A simple taxonomy of study designs includes randomized controlled trials on the one hand, and observational studies (such as case series, cohort studies, and case-control studies) on the other. A rigid hierarchy of these design types is a fairly recent phenomenon, promoted as a tenet of "evidence-based medicine," with randomized controlled trials receiving gold-standard status in terms of producing valid results. Although randomized trials have many strengths, and contribute substantially to the evidence base in clinical care, making presumptions about the quality of a study based solely on category of research design is unscientific. Both the limitations of randomized trials as well as the strengths of observational studies tend to be overlooked when a priori assumptions are made. This essay presents an argument in support of a more balanced approach to evaluating evidence, and discusses representative examples from the general medical as well as pulmonary and critical care literature. The simultaneous consideration of validity (whether results are correct "internally") and generalizability (how well results apply to "external" populations) is warranted in assessing whether a study's results are accurate for patients likely to receive the intervention-examining the intersection of clinical and methodological issues in what can be called a medicine-based evidence approach. Examination of cause-effect associations in patient-oriented research should recognize both the strengths and limitations of randomized trials as well as observational studies.
Microalloying Design of Biodegradable Mg-2Zn-0.05Ca Promises Improved Bone-Implant Applications

Abstract:Mg and its alloys have been comprehensively studied and show huge potential for clinical orthopedic applications. However, balancing the mechanical strength and corrosion resistance of alloys is still a challenge. In light of this, micro-level contents of Zn and Ca were added to pure Mg to fabricate a Mg-2Zn-0.05Ca microalloy to expectedly enhance the mechanical strength and concurrently improve the corrosion resistance. The characteristics of the rolled Mg-2Zn-0.05Ca microalloy were explored using optical microscopy, X-ray diffraction, and tensile tests. The corrosion behavior and mechanical strength loss were explored using electrochemical and immersion tests. The effects of the microalloy extract on the proliferation, adhesion, and osteogenic differentiation of MC3T3-E1 cells were systematically studied. Moreover, implantations were done in femoral condyles of rabbits to study the degradation properties, osteogenic effect, mechanical strength loss, and biosafety of the microalloy. The ultimate tensile strength and yield strength of the rolled microalloy were found to be significantly elevated to 257 ± 2.74 and 237.6 ± 8.29 MPa, respectively. The microalloy showed a stable and gradual strength loss during degradation, both in vivo and in vitro. Concurrently, the microalloy exhibited improved corrosion resistance ability and especially, in vivo, the rolled microalloy exhibited a comparable degradation rate to that of rolled pure Mg within the initial 12 weeks of implantation. Additionally, the microalloy promoted osteogenesis, both in vitro and in vivo, and no short- and long-term toxicities of the microalloy were observed in rabbits. This study suggested that the rolled Mg-2Zn-0.05Ca microalloy effectively balanced the mechanical strength and corrosion resistance and showed potential application as bone implants.
In vivo degradation and bone reaction of long-term fixation with a magnesium alloy made by twin-roll casting in a rat femur model

Abstract:Background: The effect of casting parameters on the microstructure and corrosion resistance of Mg alloys is still limited, especially in clinical animal experiments. Objective: We prepared a new magnesium rare earth alloy (Mg-Re, where Re is Ce or La) by vertical two-roll casting and Mg-A by further rolling. The microstructure characteristics, degradation behavior, and bone reaction of the two alloys were studied. Method: Ti, Mg-Re, and Mg-A alloy plates were implanted in a rat femur model, and their degradation behavior was observed 48 weeks later. Results: In vivo experiments showed no significant changes around the femur in the Ti group, excluding external factors that may cause bone remodeling and lead to new bone formation. Mg-A induces more new bone formation than Mg-Re, which meets the necessary conditions to prevent pathological fracture. The specimen staining and sectioning showed that the liver and heart of rats implanted with magnesium alloys had no pathological changes and the cell structure was normal, similar to that of rats without a magnesium alloy. Conclusion: Mg-A alloy has good healing potential as a biodegradable implant material.

Abstract:Magnesium (Mg) alloys have been widely used in bone fixation, bone repair, and cardiovascular stents as biodegradable bone‐implant materials. However, their clinical application is limited due to their fast corrosion rate and poor mechanical stability. Here we report the development of Mg‐2Zn‐0.5Ca‐0.5Sr (MZCS) and Mg‐2Zn‐0.5Ca‐0.5Zr (MZCZ) alloys with improved mechanical properties, corrosion resistance, cytocompatibility, osteogenesis performance, and antibacterial capability for biodegradable bone‐implant applications. The hot‐extruded (HE) MZCZ sample exhibited the highest ultimate tensile strength of 255.8±2.4 MPa and the highest yield strength of 208.4±2.8 MPa among all alloy samples and an elongation of 15.7±0.5% due to the recrystallization and grain‐refining effect of Zr. The HE MZCS sample showed the highest corrosion resistance among all samples, with the lowest corrosion current density of 0.2±0.1 μA/cm2 and lowest corrosion rate of 4±2 μm/y obtained from electrochemical testing, and a degradation rate of 368 μm/y and hydrogen (H2) evolution rate of 0.83±0.03 mL/cm2/d obtained from immersion testing for 21 d in Hanks' solution. The MZCZ sample showed the highest cell viability in relation to MC3T3‐E1 cells among all alloy extracts, indicating good cytocompatibility except at 25% concentration. Insight the long-term biodegradable Mg-RE-Sr alloy for orthopaedics implant via friction stir processing

Abstract:Magnesium alloys, noted for their substantial mechanical strength and exceptional biocompatibility, are increasingly being considered for use in biodegradable implants. However, their rapid degradation and significant hydrogen release have limited their applications in orthopaedics. In this study, a novel Mg-RE-Sr alloy was created by friction stir processing to modify its microstructure and enhance its degradation performance. Through microstructural characterization, the friction stir processing effectively refined the grains, accelerated the re-dissolution of precipitates, and ensured a uniform distribution of these phases. The processed alloy demonstrated improved comprehensive properties, with an in vitro corrosion rate of approximately 0.4 mm/y and increases in ultimate tensile strength and elongation by 37 % and 166 %, respectively. Notably, in vivo experiments involving a rat subcutaneous implantation model revealed a slower degradation rate of 0.09 mm/y and a uniform degradation process, basically achieving the requirements for ideal performance in orthopaedic applications. The superior degradation characteristics were attributed to the synergistic effect of attenuated galvanic corrosion and the formation of a dense Y(OH)3/Y2O3 film induced by an exceptional microstructure with a highly solid-soluted matrix and uniformly refined precipitates. Meanwhile, the alloys exhibited excellent biocompatibility and did not cause undesirable inflammation or produce toxic degradation products. These improvements in biocompatibility and degradation characteristics indicate great promise for the use of this friction stir processed alloy in osteosynthesis systems in the clinical setting.
The Development of Binary Mg-Ca Alloys for Use As Biodegradable Materials Within Bone

Abstract:Binary Mg–Ca alloys with various Ca contents were fabricated under different working conditions. X-ray diffraction (XRD) analysis and optical microscopy observations showed that Mg–xCa (x=1–3 wt%) alloys were composed of two phases, α(Mg) and Mg2Ca. The results of tensile tests and in vitro corrosion tests indicated that the mechanical properties could be adjusted by controlling the Ca content and processing treatment. The yield strength (YS), ultimate tensile strength (UTS) and elongation decreased with increasing Ca content. The UTS and elongation of as-cast Mg–1Ca alloy (71.38±3.01MPa and 1.87±0.14%) were largely improved after hot rolling (166.7±3.01MPa and 3±0.78%) and hot extrusion (239.63±7.21MPa and 10.63±0.64%). The in vitro corrosion test in simulated body fluid (SBF) indicated that the microstructure and working history of Mg–xCa alloys strongly affected their corrosion behaviors. An increasing content of Mg2Ca phase led to a higher corrosion rate whereas hot rolling and hot extrusion could reduce it. The cytotoxicity evaluation using L-929 cells revealed that Mg–1Ca alloy did not induce toxicity to cells, and the viability of cells for Mg–1Ca alloy extraction medium was better than that of control. Moreover, Mg–1Ca alloy pins, with commercial pure Ti pins as control, were implanted into the left and right rabbit femoral shafts, respectively, and observed for 1, 2 and 3 months. High activity of osteoblast and osteocytes were observed around the Mg–1Ca alloy pins as shown by hematoxylin and eosin stained tissue sections. Radiographic examination revealed that the Mg–1Ca alloy pins gradually degraded in vivo within 90 days and the newly formed bone was clearly seen at month 3. Both the in vitro and in vivo corrosion suggested that a mixture of Mg(OH)2 and hydroxyapatite formed on the surface of Mg–1Ca alloy with the extension of immersion/implantation time. In addition, no significant difference (p>0.05) of serum magnesium was detected at different degradation stages. All these results revealed that Mg–1Ca alloy had the acceptable biocompatibility as a new kind of biodegradable implant material. Based on the above results, a solid alloy/liquid solution interface model was also proposed to interpret the biocorrosion process and the associated hydroxyapatite mineralization.

Abstract:Magnesium alloys have shown prospective applications as a new biodegradable metal within bone. To garantee the longterm biocompatibility, a Mg-Zn-Ca alloy, composing of essential elements for human, was prepared and its feasibility for orthopedic applications was investigated. The in vitro and in vivo corrosion of Mg-Zn-Ca alloy as well as the biocompatibility were studied. The in vitro corrosion tests in five kinds of physiological solutions showed that the corrosion rates and corrosion morphologies of the alloy were strongly influenced by the solution used. The addition of serum in Hank’s and MEM significantly slowed down the corrosion rate and improved the corrosion uniformity of the alloy. The corrosion rate decreased with increasing serum concentration. The alloy showed the slowest corrosion rate as well as homogeneous corrosion morphology in MEM+10%FBS. Both the indirect and direct cell experiments indicated good cytocompatibility of the extruded Mg-Zn-Ca alloy. In vivo , we observed a gradual degradation process from the surface of extruded Mg-Zn-Ca alloy and only 40% in volume of implant was left after 4 weeks implantation in medullary cavities of mice. The micro-CT and histological analyses revealed its good biocompatibility with peri-implant new bone formation and increasing cortical bone thickness with increasing implantation period. This study showed that the extruded Mg-Zn-Ca alloy provided sufficient biocompatibility for orthopedic application, though the in vivo corrosion rate should be further reduced for clinical use.
Biocompatibility of bioabsorbable Mg–Ca alloys with rare earth elements addition

Abstract:The objectives were to investigate the mechanical strength and biocompatibility of Mg2Ca2Gd and Mg1Ca2Nd (wt%) alloys developed for biomedical application as implantable bioabsorbable devices. Samples were implanted in New-Zealand rabbits tibia for 3, 6 and 8 weeks and compatibility analysis involved whole blood test, biochemistry, histopathology, histology, and radiographs. Refinement in grains were observed in Mg2Ca2Gd alloy; and Mg5Gd, Mg41Nd5, α-Mg and Mg2Ca phases were identified. Polarization curves revealed easier oxidation of Mg2Ca2Gd alloy, smaller values of corrosion rate and a higher polarization resistance of Mg1Ca2Nd. Adequate compatibility of both alloys was identified with pre-osteoblast stem cells. Red and white cells stayed compatible with reference ranges. Enzymes from liver and kidneys stayed at regular values and samples from kidneys and liver tissues presented similar organization to control animals. Histological displays from implantation sites disclosed well-structured tissues with evidences of bone cells activities compatible with the new bone tissues observed. Radiographs from tibias did not revealed relevant gas pockets. Mg2Ca2Gd alloy demonstrated faster degradation. Adequate biocompatibility was observed in Mg–Ca alloys with RE addition, being potential candidates for development of metallic implantable bioabsorbable devices.
In vivo biodegradation and biological properties of a Mg-Zn-Ca amorphous alloy for bone defect repair

Abstract:Amorphous magnesium alloys, characterised by their unique disordered structure, exhibit exceptional mechanical properties and superior corrosion resistance. This study introduced a novel biodegradable Mg-Zn-Ca amorphous alloy. Immersion and electrochemical tests revealed uniform degradation in a simulated body fluid environment. The degradation products predominantly contained zinc and oxygen. After 28 days of immersion, the alloy's structure largely remained intact, and it exhibited a minimal weight loss rate of 11.64 ± 1.85%, indicating its excellent corrosion resistance. In addition, the homogeneous and rapid degradation of the Mg-Zn-Ca amorphous alloy in rats (the residual volume at 12 weeks was 14.94 ± 5.05%, at least 2–3 times of the corrosion rate of immersion in vitro), thus increasing the bone volume/total volume and trabecular number. The new bone and the residual material surface showed good bone integration. Blood and organ tissue tests confirmed the in vivo biological safety. Therefore, Mg-Zn-Ca amorphous alloys have excellent potential as bone-repair materials, especially in load-bearing areas.

Development of rare-earth oxide reinforced magnesium nanocomposites for orthopaedic applications: A mechanical/immersion/biocompatibility perspective

Abstract:Magnesium-Zinc based nanocomposites containing cerium oxide nanoparticles were developed in the present work. A systematic study on their microstructure, mechanical properties, in vitro degradation behaviour, and cytotoxicity are presented. It was found that the developed nanocomposites exhibited excellent strength and toughness that are superior to the commercially available magnesium alloys. From corrosion perspective, nanocomposites exhibited reduced pH increase compared to pure Mg with Mg-0.5Zn/0.5CeO2 showing the least corrosion rate. Moreover, the developed nanocomposites exhibited no cytotoxicity to MC3T3-E1 pre-osteoblast cells. Based on the above findings, the feasibility of Mg-Zn/CeO2 nanocomposites for use as orthopaedic implants is systematically discussed. This study provides an insight into the development of new high-performance Mg alloy-rare earth oxide (REO)-based nanocomposites with superior mechanical properties and corrosion resistance while effectively avoiding the possible standing toxic effect of RE elements.
Chemiluminescent Analysis of Plasma Antioxidant Capacity in Uremic Patients Undergoing Hemodialysis

Abstract:Objective. Hemodialysis is a common therapeutic strategy for patients with end stage renal failure. During the hemodialytic process, the neutrophils are activated (neutrophil burst) due to the hemoincompatibility induced by hemodialysis. As a result, the activated neutrophils release reactive oxygen species (ROS), such as hydrogen peroxide, singlet oxygen, and hypochlorite, into the bloodstream and cause oxidative damage. Methods. This study investigated the antioxidant alteration of plasma in uremic patients undergoing hemodialysis by chemiluminescent analysis. The antioxidant capacities of plasma in scavenging hydrogen peroxide, singlet oxygen, and hypochlorite were investigated in this experiment. In addition, investigation of the ferric-reducing ability of plasma (FRAP) would be covered in this study as well. Results. This study found that after hemodialysis, the antioxidant capacities of plasma in scavenging hydrogen peroxide, singlet oxygen, and hypochlorite decreases 7.9%, 18.8%, and 18.9%, respectively. Moreover, the FRAP is reduced by 56%. We speculate that the loss of dialyzable solutes (such as uremic solutes and antioxidants with small molecular weight) in plasma resulted in its decrease in antioxidant capacity. Conclusion. We therefore suggest that the supplement of antioxidants with small molecular weight is capable of regaining antioxidant defense in plasma and preventing oxidative damage induced by hemodialysis.
Microstructure, Mechanical Properties, in Vitro Degradation and Cytotoxicity Evaluations of Mg–1.5Y–1.2Zn–0.44Zr Alloys for Biodegradable Metallic Implants

Abstract:Mg-1.5Y-1.2Zn-0.44Zr alloys were newly developed as degradable metallic biomaterials. A comprehensive investigation of the microstructure, mechanical properties, in vitro degradation assessments and in vitro cytotoxicity evaluations of the as-cast state, as-heat treated state and as-extruded state alloys was done. The microstructure observations show that the Mg-1.5Y-1.2Zn-0.44Zr alloys are mainly composed of the matrix α-Mg phases and the Mg12ZnY secondary phases (LPS structure). The hot extrusion method significantly refined the grains and eliminated the defects of both as-cast and heat treated alloys and thereby contributed to the better mechanical properties and biodegradation resistance. The values of tensile strength and tensile yield strength of the alloy in the as-extruded condition are about 236 and 178 MPa respectively, with an excellent elongation of 28%. Meanwhile, the value of compressive strength is about 471 MPa and the value of bending strength is about 501 MPa. The superior bending strength further demonstrates the excellent ductility of the hot extruded alloys. The results of immersion tests and electrochemical measurements in the SBF indicate that a protective film precipitated on the alloy's surface with the extension of degradation. The protective film contains Mg(OH)2 and hydroxyapatite (HA) which can reinforce osteoblast activity and promote good biocompatibility. No significant cytotoxicity towards L-929 cells was detected and the immersion extracts of alloy samples could enhance the cell proliferation with time in the cytotoxicity evaluations, implying that the Mg-1.5Y-1.2Zn-0.44Zr alloys have the potential to be used for biomedical applications.
In Vitro and in Vivo Studies of Mg-30Sc Alloys with Different Phase Structure for Potential Usage Within Bone.

Abstract:Proper alloying magnesium with element scandium (Sc) could transform its microstructure from alpha phase with hexagonal closed-packed (hcp) structure into beta phase with body-cubic centered (bcc) structure. In the present work, the Mg-30 wt% Sc alloy with single alpha phase, dual phases (alpha + beta) or beta phase microstructure were developed by altering the heat-treatment routines and their suitability for usage within bone was comprehensively investigated. The beta phased Mg-30 wt% Sc alloy showed the best mechanical performance with ultimate compressive strength of 603 +/- 39 MPa and compressive strain of 31 +/- 3%. In vitro degradation test showed that element scandium could effectively incorporate into the surface corrosion product layer, form a double-layered structure, and further protect the alloy matrix. No cytotoxic effect was observed for both single alpha phased and beta phased Mg-30 wt% Sc alloys on MC3T3 cell line. Moreover, the beta phased Mg-30 wt%Sc alloy displayed acceptable corrosion resistance in vivo (0.06 mm y(-1)) and maintained mechanical integrity up to 24 weeks. The degradation process did not significantly influence the hematology indexes of inflammation, hepatic or renal functions. The bone-implant contact ratio of 75 +/- 10% after 24 weeks implied satisfactory integration between beta phased Mg-30 wt%Sc alloy and the surrounding bone. These findings indicate a potential usage of the bcc-structured Mg-Sc alloy within bone and might provide a new strategy for future biomedical magnesium alloy design. Statement of Significance Scandium is the only rare earth element that can transform the matrix of magnesium alloy into bcc structure, and Mg-30 wt%Sc alloy had been recently reported to exhibit shape memory effect. The aim of the present work is to study the feasibility of Mg-30 wt%Sc alloy with different constitutional phases (single alpha phase, single beta phase or dual phases (alpha+beta) as biodegradable orthopedic implant by in vitro and in vivo testings. Our findings showed that beta phased Mg-30 wt%Sc alloy which is of bcc structure exhibited improved strength and superior in vivo degradation performance (0.06 mm y(-1)). No cytotoxicity and systematic toxicity were shown for beta phased Mg-30 wt%Sc alloy on MC3T3 cell model and rat organisms. Moreover, good osseointegration, limited hydrogen gas release and maintained mechanical integrity were observed after 24 weeks' implantation into the rat femur bone. (C) 2019 Acta Materialia Inc. Abstract:The development direction of bioresorbable fixing structures is currently very relevant because it corresponds to the priority areas in worldwide biotechnology development. Magnesium (Mg)-based alloys are gaining high levels of attention due to their promising potential use as the basis for fixating structures. These alloys can be an alternative to non-degradable metal implants in orthopedics, maxillofacial surgery, neurosurgery, and veterinary medicine. In our study, we formulated a Mg-2Zn-2Ga alloy, prepared pins, and analyzed their biodegradation level based on SEM (scanning electron microscopy) and EDX (energy-dispersive X-ray analysis) after carrying out an experimental study on rats. We assessed the resorption parameters 1, 3, and 6 months after surgery. In general, the biodegradation process was characterized by the systematic development of newly formed bone tissue. Our results showed that Mg-2Zn-2Ga magnesium alloys are suitable for clinical applications.
