Abstract:The biopharmaceutical market is driving the revolution from traditional batch processes to continuous manufacturing for higher productivity and lower costs. In this work, a batch mAb downstream process has been converted into an integrated continuous process with the combination of multiple techniques. For process intensification, two batch mode unit operations (protein A capture chromatography, ultrafiltration/diafiltration) were converted into continuous ones; for continuity, surge tanks were used between adjacent steps, and level signals were used to trigger process start or stop, forming a holistic continuous process. For process automation, manual operations (e.g., pH and conductivity adjustment) were changed into automatic operation and load mass was controlled with process analytical technology (PAT). A model-based simulation was applied to estimate the loading conditions for the continuous capture process, resulting in 21% resin capacity utilization and 28% productivity improvement as compared to the batch process. Automatic load mass control of cation exchange chromatography (CEX) was achieved through a customized in-line protein quantity monitoring system, with a difference of less than 1.3% as compared to off-line analysis. Total process time was shortened from 4 days (batch process) to less than 24 hours using the continuous downstream process with the overall productivity of 23.8 g mAb per day for the bench-scale system. Comparable yield and quality data were obtained in three test runs, indicating a successful conversion from a batch process to a continuous process. The insight of this work could be a reference to other similar situations.
Analysis of Mixing Efficiency in Continuous Polymer Processing Equipment

Abstract:[Continuous mixing equipment is widely used in polymer processing. Typical continuous mixers include single screw and twin screw extruders. The flow field in these mixing devices is difficult to analyze due to the complex geometry. In this work, a fluid dynamics analysis package-FIDAP, using the finite element method, was implemented to simulate the three dimensional, isothermal flow patterns in the region of conveying elements and shearing discs of a Leistritz intermeshing counterrotating twin screw extruder model LSM30.34, a 2{dollar}{bsol}sp{lcub}{bsol}prime{bsol}prime{rcub}{dollar} Welding Engineers tangential counterrotating twin screw extruder and the MCT section of a Multi-Cut Transfermix extrusion system. The rheology of the fluid was described by a power-law model. The problem of time dependent flow boundaries was solved by selecting a number of sequential geometries to represent a complete mixing cycle. The dispersive mixing in the region of conveying elements and shearing discs of the intermeshing counterrotating twin screw extruder was characterized in terms of shear stresses generated in the flow field and a parameter {dollar}{bsol}beta{dollar} quantifying the elongational flow components. The influence of screw rotational speed and axial pressure difference on these flow characteristics was analyzed. Comparisons of the flow characteristics in the regio n of conveying elements and in the shearing discs, and between corotating and counterrotating modes in the shearing discs were also presented. A framework was developed to evaluate distributive mixing efficiency in the intermeshing and tangential counterrotating twin screw extruders and in the Multi-Cut Transfermix. The dynamics of distributive mixing was studied numerically by means of tracking the evolution of particles originally gathered as clusters. The extent or goodness of mixing was characterized in terms of length stretch, pairwise correlation function and volume fraction of islands. The length stretch reflects the capability of the mixer to spread particles away from their neighbors initially present in the same cluster. The pairwise correlation function characterizes the global distribution of particles in the mixing region. The volume fraction of islands quantifies the regions void of minor component and provides a more local mixing analysis. This framework can then be used to differentiate among various operating conditions or various designs of the same type of equipment. Abstract:Continuous manufacturing (CM) is a promising strategy to achieve various benefits in the context of quality, flexibility, safety and cost in pharmaceutical production. One of the main technical challenges of CM is that the process needs to handle transient conditions such as the start-up phase before state of control operation is reached, which can potentially cause out-of-specification (OOS) material. In this context, the presented paper aims to demonstrate that suitable process control strategies during start-up of a continuous granulation and drying operation can limit or even avoid OOS material production and hence can ensure that the provided benefits of CM are not compromised by poor production yields. In detail, heat-up of the drying chamber prior the start of production can lead to thermal energy being stored inside of the stainless-steel housing, acting as an energy buffer that is known to cause over-dried granules in the first few minutes of the drying process. To compensate this issue, an automatic ramping procedure of dryer rotation speed (and hence drying time) was introduced into the plant’s process control system, which counteracts the excessive drying capacity during start-up. As a result, dry granules exiting the dryer complied with the targeted intermediate critical quality attribute loss-on-drying (LOD) from the very beginning of production.

Residence time and mixing capacity of a rotary tablet press feed frame

Abstract:Purpose: Most rotary tablet presses contain a feed frame to provide a continuous powder flow and to feed powder into the dies. The wide residence time distribution (RTD) of these feed frames is problematic, because it negatively affects material traceability in continuous manufacturing. In a rotary tablet press, different machine settings influence the RTD, which is characterized by the mean and the width of the distribution. This study focused on the effects of the rotational speed of the feed frame paddles and the rotary tablet press throughput on the RTD. Methods: An in-line UV/Vis measurement method was developed for determining the RTD in the feed frame. A model based on a plug flow and a continuous stirred tank reactor was adapted to model the experimentally determined RTDs. Finally, the mixing capacity of a feed frame was evaluated and correlated with a model parameter of the RTD. Results: Overall, the developed UV/Vis measurement method was suitable and could be used to obtain process information regarding content uniformity in real time. The experimentally-determined RTDs were described well by fitting an inverse mixing and a transport time. In addition, a correlation between the location and the shape of measured RTDs and tablet press throughput was found. In contrast, rotational feed frame paddle speed did not affect the RTDs. Split-feeding experiments indicated the mixing capacity of the rotary tablet press feed frame. Conclusion: The inverse mixing time can be used as an initial indicator for estimating the mixing capacity.

Abstract:The manufacturing industry is challenged by increasing costs of scarce resources, rapid change, and tight competition in global markets, and the need for efficient and sustainable process design and operating strategies. Addressing these challenges has been the focus of process intensification (PI). Polymerization processes face the additional challenge of meeting tight product specifications, which are dictated by the demands of high quality and advanced materials. Meeting these specifications requires the incorporation of complex molecular weight distribution (MWD) models based on population balances of all the polymer chains. Addressing these challenges through process intensification requires the development and application of efficient, robust, and powerful optimization strategies and modeling frameworks, leading to an essential strategy for process intensification of polymerization systems. This survey presents the development and application of novel optimization strategies for model development, process synthesis and design, and the operation of polymer processes. The effectiveness of these optimization-based approaches will be demonstrated on a slurry process for high density polyethylene (HDPE), where significant advances in process intensification have been realized. Future directions are given, with the aim of benefiting both research and application in this field.
Influence of refilling on dosing accuracy of loss-in-weight powder feeders in continuous manufacturing

Abstract:Continuous manufacturing is a novel concept to produce high quality drug products. The manufacture of solid dosage forms starts with the feeding of materials, which is consider a critical step because it could have a high influence on the quality of the final product. Therefore, this unit operation is done by the so-called loss-in-weight feeders developed by various equipment manufacturers to ensure accurate feed rates that allows obtaining products in compliance with the specifications. Nonetheless, as the feed hopper of a feeder is of limited capacity, it must be refilled on regular basis to maintain continuous manufacturing. During refilling, dosing must be done in volumetric mode which can have an impact on dosing accuracy. The present work studies the impact of the refill and different factors such as the feed rate, the refill system and the filling level at which the refill starts on the accuracy of the dose. Four performance indicators were used to evaluate this impact: the refilling time, the time out of the limits, the maximum or minimum feed rate during the refilling and the total deviation from the set point. The refiller type and the filling level at which the refill starts play an important role and should be selected considering to the material's characteristics to be fed. As a general rule, a faster and shorter refill is always preferable, however, depending on the refiller type and the material, a positive or negative deviation from the feeding dose can be observed.

Abstract:We consider the optimal reactor network synthesis of a polymerization process with detailed molecular weight distributions (MWDs). Based on an industrial high‐density polyethylene (HDPE) slurry process model including an embedded MWD, a fully connected process superstructure of continuous stirred tank reactors (CSTRs) is established through the introduction of splitters. Using this generalized superstructure as a basis, two nonlinear programming (NLP) problem formulations, which simultaneously maximize the monomer conversion and minimize the deviation between the calculated and target MWDs, are developed by applying multiobjective optimization (MO) methods. Different optimal flowsheet configurations are generated by systematically manipulating a set of continuous decision variables. Several case studies that consider different specifications on MWD are conducted to illustrate the effectiveness and efficiency of the proposed synthesis approach. Numerical results show that the optimal flowsheet configurations overcome the limitations of conventional reactor network structures and help to increase reactor productivity at the desired product quality. © 2015 American Institute of Chemical Engineers AIChE J, 62: 131–145, 2016
Reducing Side Product by Enhancing Mass‐transfer Rate

Abstract:Fast mass transfer rate and short reaction time are required to reduce side product of the premixing reaction, in the SNIA Viscosa process, as a liquid-liquid competitive consecutive system. In order to obtain high selectivity, a microstructure device, named a membrane dispersion mini-reactor, was designed and tested at several operation conditions. The mass transfer rate of cyclohexanecarboxylic acid (CCA) was significantly increased by membrane dispersion in this mini-reactor compared with a traditional batch reactor and a membrane free one, just with a residence time less than 80 ms. The results showed that the nominal yield of side product was lower than 10% in the reaction products and about 5% lower than the traditional stirring method. Additionally, a theoretical model was developed to eliminate the mass transfer flux in this mini-reactor, and the calculation results fit well with the experimental data. Abstract:Continuous manufacturing of pharmaceuticals offers several benefits, such as increased production efficiency, enhanced product quality control, and lower environmental footprint. To fully exploit these benefits, standard operation mode (production processes with no or minimal disturbance mitigation measures) should be supported by adopting novel quality-by-control (QbC) methodologies. The paper at hand is the first part of a study focused on developing QbC algorithms for optimizing twin-screw wet granulation in the industrial manufacturing line ConsiGma TM -25, specifically addressing granule composition. This work relieson previously established process-analytical-technology (PAT) equipment for real-time monitoring of the granule composition, i.e., the active pharmaceutical ingredient (API) and liquid content in wet granules. The developed control platform integrates model-based process control algorithms that aim to keep the API- and liquid content at target values through real-time adjustments of the process parameters. Furthermore, the platform integrates a digital operator assistant, which aims to detect and classify granulation disturbances and provides messages and instructions for the plant operator. The present manuscript systematically outlines all design steps from the development phase in the simulation environment to the final real system application and validation. The control platform's performance is demonstrated through selected test scenarios on the ConsiGma TM -25 manufacturing line. The obtained results indicate improved disturbance robustness and an increase in intermediate/final product quality (compared to conventional operating modes): The process control algorithms successfully maintained the API- and liquid content at target values despite process disturbances. Furthermore, realistic disturbances (feeder, pump, and material) were accurately detected and classified by the digital assistant algorithm. The information was provided through a user interface, offering real-time support for plant personnel.

Abstract:The pharmaceutical industry is looking for new and innovative ways of manufacturing to improve product quality and reduce process complexity. In manufacturing oral solid dosage products, blending is a crucial step in ensuring the homogeneity of active pharmaceutical ingredients (APIs) in the final product. Currently, batch and continuous blending are the two commonly used modes for blending in the industry. However, these methods have limitations in terms of blending time, manual intervention, and flexibility in handling multiple ingredients. To address these limitations, this study aims to explore the feasibility and benefits of using a semicontinuous blending mode in the pharmaceutical industry. A case study is conducted using a binary blend of microcrystalline cellulose and acetaminophen to compare the performance of the semicontinuous mode of blending with the batch and continuous blending modes. The results show that the semicontinuous blending setup can produce blends with good blend uniformity and homogeneity and that the output can be used for both batch and continuous downstream operations. The effect of variation in the three most important process parameters, impeller rotation per minute, blending time, and fill level on the blend uniformity, is also investigated. The semicontinuous blending mode had a higher line rate of 12.5 kg/hour than a similarly sized batch blender at 3.6 kg/hour and less than that of a continuous blender. The benefits of the new blending mode include reduced blending time, minimal manual intervention, flexibility in blending multiple ingredients, easier scale-up, and a smaller footprint. Overall, this study highlights the relative advantages of using this new semicontinuous blending mode in pharmaceutical manufacturing and its potential as a good alternative to the existing blending modes. The semicontinuous mode is well placed between the batch blending and continuous blending mode, with many benefits over the former mode and performance comparable to the latter continuous mode.

Abstract:Wet granulation is a size enlargement process in which fine powders are agglomerated into larger granules via the presence of a liquid binder. Among various types and modes of granulation processes, twin-screw granulation is a popular continuous wet granulation technique. The typical residence time that powders spend in a twin-screw granulator are in the order of seconds, hence, the rigorous mixing of solid and liquid particles across particle sizes and equipment geometry becomes even more important. This study aims to improve the mechanistic understanding of the effect of process and screw parameters on the mixing dynamics inside a twin-screw granulator using dispersion coefficient as a key metric. The dispersion coefficient quantifies the dispersion and is directly proportional to the extent of mixing occurring inside the granulator. An analysis of the experimentally obtained dispersion coefficient is performed to understand the effect of key process and design parameters such as feed rate, screw speed, number of kneading elements and, stagger angle. A semi-mechanistic model that incorporates these parameters was developed to estimate and predict the dispersion coefficient. The model accurately predicts the dispersion coefficient values and the goodness-of-fit values (R2) for the test set and full data set were found to be equal to 0.920 and 0.932 respectively. This model was further tested by predicting the complete RTD curves using the predicted dispersion coefficients. The average goodness-of-fit value (R2) for the prediction of RTD curves for all the experimental runs was calculated to be equal to 0.698.

Abstract:Compounding is an important step in processing base polymers and is used to incorporate various additives into a polymer. For this purpose, different screw elements are used for dispersive and distributive mixing on a co-rotating twin-screw extruder. Optimising the screw configuration requires precise knowledge of the screw elements' mixing properties, which have not been thoroughly investigated. This study analyses the mixing behaviour of individual screw elements regarding dispersive and distributive mixing using 3D CFD flow simulations with subsequent particle tracking. For distributive mixing, the particle distribution behind the screw elements in the XY plane is analysed and the mixing index MQ, which relates the standard deviation and the mean value of the triangular areas between the particles, is calculated. For dispersive mixing, the maximum shear stress on the particle path and the integral of the shear stress over the residence time of each individual particle are determined. The results show that screw element geometry and rotation speed have a significant influence on dispersive and distributive mixing. In addition, better dispersive mixing is achievable with highly viscous materials. These findings enable the optimisation of the mixing zone of a co-rotating twin-screw extruder for the efficient mixing of mineral fillers.

Abstract:In this work, a high-fidelity digital twin was developed to support the design and testing of control strategies for drug product manufacturing via direct compression. The high-fidelity digital twin platform was based on typical pharmaceutical equipment, materials, and direct compression continuous processes. The paper describes in detail the material characterization, the Discrete Element Method (DEM) model and the DEM model parameter calibration approach and provides a comparison of the system's response to the experimental results for stepwise changes in the API concentration at the mixer inlet. A calibration method for a cohesive DEM contact model parameter estimation was introduced. To assure a correct prediction for a wide range of processes, the calibration approach contained four characterization experiments using different stress states and different measurement principles, namely the bulk density test, compression with elastic recovery, the shear cell, and the rotating drum. To demonstrate the sensitivity of the DEM contact parameters to the process response, two powder characterization data sets with different powder flowability were applied. The results showed that the calibration method could differentiate between the different material batches of the same blend and that small-scale material characterization tests could be used to predict the residence time distribution in a continuous manufacturing process.

Abstract:Continuous Direct Compaction (CDC) has emerged as a promising route towards producing solid dosage forms while reducing material, development time and energy consumption. Understanding the response of powder processing unit operations, especially blenders, is crucial. There is a substantial body of work around how lubrication via batch blender operation affects tablet critical quality attributes such as hardness and tensile strength. But, aside from being batch operations, the design of these blenders is such that they operate with low-shear, low-intensity mixing at Froude number values significantly below 0.4 (Froude number Fr being the dimensionless ratio of inertial to gravitational forces). The present work explores the performance of a mini-blender which has a fundamentally different mode of operation (static vessel with rotating blades around a mixing shaft as opposed to rotating vessel with no mixing shaft). This difference allows a substantially wider operating range in terms of speed and shear (and Fr values). The present work evaluates how its performance compares to other blenders studied in the literature. Tablet compaction data from blends produced at various intensities and regimes of mixing in the mini-blender follow a common trajectory. Model equations from literature are suitably modified by inclusion of the Froude number Fr , but only for situations where the Froude number was sufficiently high (1 < Fr ). The results suggest that although a similar lubrication extent plateau is eventually reached it is the intensity of mixing ( i.e. captured using the Froude number as a surrogate) which is important for the lubrication dynamics in the mini-blender, next to the number of revolutions. The degree of fill or headspace, on the other hand, is only crucial to the performance of common batch blenders. Testing using alternative formulations shows the same common trend across mixing intensities, suggesting the validity of the approach to capture lubrication dynamics for this system.

Abstract:Toward further implementation of continuous tablet manufacturing, one key issue is the time needed for start-up operation because it could lead to lower product yield and reduced economic performance. The behavior of the start-up operation is not well understood; moreover, the definition of the start-up time is still unclear. This work investigates the effects of process parameters on the start-up operation in continuous wet granulation, which is a critical unit operation in solid drug manufacturing. The profiles of torque and granule size distribution were monitored and measured for the first hour of operation, including the start-up phase. We analyzed the impact of process parameters based on design of experiments and performed an economic assessment to see the effects of the start-up operation. The torque profiles indicated that liquid-to-solid ratio and screw speed would affect the start-up operation, whereas different start-up behavior resulted in different granule size. Depending on the indicator used to define the start-up operation, the economic optimal point was significantly different. The results of this study stress that the start-up time differs according to the process parameters and used definition, e.g., indicators and criteria. This aspect should be considered for the further study and regulation of continuous manufacturing.

Additive manufacturing of prototype elements with process interfaces for continuously operating manufacturing lines

Abstract:Rapid prototyping based on in silico design and 3D printing enables fast customization of complex geometries to multiple needs. This study utilizes, additive manufacturing for rapid prototyping of elements for continuously operating mixing geometries including interfaces with process analytical technology (PAT) tools, to show that 3D printing can be used for prototyping of both parts of production line and PAT interfacing solution. An additional setup was designed for measuring the dynamic calibration samples for a semi-quantitative near infrared (NIR) spectroscopic model. The powder was filled in a small calibration chamber and in-line NIR spectra of calibration samples were collected from moving material while mimicking the powder flow dynamics in a typical continuous mixer. This dynamic powder mixing system was compared with a static powder calibration model where the NIR probe was placed at different positions on a static sample. Principal component analysis (PCA) revealed that the 3D printed device with dynamic measurement of the NIR spectra had more potential for quantitative analysis. With the prototype continuous mixer, two differently placed process interfaces for NIR spectroscopic monitoring of the powder mixing were evaluated. With this approach, the importance of positioning the process analytical tools to assess the blend uniformity could be demonstrated. It was also observed that with the longer mixing geometry, a better mixing result was achieved due to a larger hold up volume and increased residence time.
Development and Use of a Residence Time Distribution (RTD) Model Control Strategy for a Continuous Manufacturing Drug Product Pharmaceutical Process

Abstract:Residence-time-distribution (RTD)-based models are key to understanding the mixing dynamics of continuous manufacturing systems. Such models can allow for material traceability throughout the process and can provide the ability for removal of non-conforming material from the finished product. These models have been implemented in continuous pharmaceutical manufacturing mainly for monitoring purposes, not as an integral part of the control strategy and in-process specifications. This paper discusses the steps taken to develop an RTD model design space and how the model was statistically incorporated into the product’s control strategy. To develop the model, experiments were conducted at a range of blender impeller speeds and total system mass flow rates. RTD parameters were optimized for each condition tested using a tank-in-series-type model with a delay. Using the experimental RTD parameters, an equation was derived relating the mean residence time to the operating conditions (i.e., blender impeller speed and mass flow rate). The RTD parameters were used in combination with real-time upstream process data to predict downstream API concentration, where these predictions allowed validation across the entire operating range of the process by comparison to measured tablet assay. The standard in-process control limits for the product were statistically tightened using the validation acceptance criteria. Ultimately, this model and strategy were accepted by regulatory authorities.

Abstract:In continuous powder handling processes, precise and consistent feeding is crucial for ensuring the quality of the final product. The intermixing effect caused by agitators, which alters the powder's bulk density, flow rate, and flow patterns, plays a significant role in this process, yet it is often overlooked. This study combines discrete element method (DEM) modeling and experiments using a commercial-scale feeder to propose a Digital Twin (DT) framework. The DEM model accurately captures key flow features, such as bypass trajectories, stagnant zones, and preferential flow patterns, while providing quantitative predictions for the feed factor and zones prone to material accumulation. Scenario analysis is performed to identify the most favorable operating ranges of the screw-agitator ratio and screw speed, considering the cohesive properties of the powder. The study demonstrates that powders with poor flow characteristics require tighter operational constraints, as the screw-agitator ratio is susceptible to variations in mass feed rate. This contribution highlights the importance of selecting an appropriate screw-agitator ratio instead of maintaining a fixed value. Properly choosing this ratio helps determine an optimal operation window, which aims to achieve a minimum agitation level needed to induce unhindered flow and reduce variability in the mass flow rate.

Abstract:Abstract This paper proposes a feed rate control strategy for a novel volumetric micro-feeder, which can accomplish low-dose feeding of pharmaceutical raw materials with significantly different powder properties. The developed feed-forward control strategy enables a constant feed rate with a minimum deviation from the set-point, even for materials that are typically difficult to accurately feed (e.g., due to high cohesion or low density) using conventional continuous feeders. Density variations observed during the feeding process were characterized via a displacement feed factor profile for each powder. The characterized effective displacement density profile was applied in the micro-feeder system to proactively control the feed rate by manipulating the powder displacement rate (i.e., computing the feed rate from the powder displacement rate). Based on the displacement feed factor profile, the feed rate can be predicted during the feeding process and at any feed rate set-point. Three pharmaceutically relevant materials were used for the micro-feeder evaluation: di-calcium phosphate (large-particle system, high density), croscarmellose sodium (small-particle system, medium density), and barium sulfate (very small-particle <10 μm, high density). A significant improvement in the feeding performance was achieved for all investigated materials. The feed rate deviation from the set-point and its relative standard deviation were minimal compared to operations without the control strategy.
