Abstract:The ability of soil microorganisms to uphold a variety of ecosystem services is of increasing interest, but our understanding of the linkage between cropland ecosystem services and the soil microbiome under long-term different fertilizer regimes is still limited. Here, we measured 15 response variables commonly used as indicators of cropland ecosystem services, and employed a shotgun metagenomic sequencing technology to determine microbiome taxonomic composition and functional composition in a sandy loam soil amended with organic and inorganic fertilizers for 28 years. Long-term application of fertilizers had overall positive effects on the variables that indicate soil C and nutrient status, organic material decomposition, nutrient transformation, and plant productivity, but the strength of these effects differed between organic and inorganic fertilization. In long term, organic fertilizers significantly increased the abundance of Sphingomonas and Acidobacteria as well as genes involved in C and N metabolism and degradation of aromatic organic compounds. The observed differences in the indicators of overall biological activity, litter decomposition, nutrient transformation, cropland fertility and productivity were associated with changes in taxonomic composition and functional composition of the soil microbiome. Network analysis revealed that soil organic C, total N, L-leucine aminopeptidase, phosphatase, microbial biomass C and N, and the multifunctionality index showed strong positive associations with Acidobacterium, Lysobacter and Nitrososphaera. These microbes are implicated in the decomposition of organic material, soil C and N transformation, and plant growth promotion. The present study deepens our understanding of the relationship between the soil microbiome and agro-ecosystem functioning, and provides support for the use of organic fertilization strategy to enhance multiple ecosystem services in croplands.
Partial Replacement of Inorganic Phosphorus (P) by Organic Manure Reshapes Phosphate Mobilizing Bacterial Community and Promotes P Bioavailability in a Paddy Soil.

Abstract:The optimization of more sustainable fertilization practice to relieve phosphorus (P) resource scarcity and increase P fertilizer utilization, a better understanding of the regulatory roles of microbes in P mobilization is urgently required to reduce P input. The genes phoD and pqqC are responsible for regulating organic and inorganic P mobilization, respectively. Using high-throughput sequencing, the corresponding bacterial communities harbored by these genes were determined. We conducted a 4-year rice-rice-crop rotation to investigate the responses of phoD- and pqqC-harboring bacterial communities to the partial replacement of inorganic P fertilizer by organic manure with reduced P input. The results showed that a combination of organic and inorganic fertilization maintained high rice yield, and also produced a more complex and stable phosphate mobilizing bacterial community, which contributed to phosphatase activities more than their gene abundances in the model analysis. Compared with the conventional mineral fertilization, organic-inorganic fertilization with the reduced P input slightly increased pqqC gene abundance while significantly enhanced the abundance of phoD-harboring bacteria, especially the genera Bradyrhizobium and Methylobacterium known as potential organic P mineralizers which can maintain high rice production. Moreover, the increased pH was the most impactful factor for the phoD- and pqqC-harboring bacterial communities, by promoting microbial P turnover and greatly increasing bioavailable P pools (H2O-Pi and NaHCO3-Pi, NaOH-Pi) in this P-deficient paddy soil. Hence, our study demonstrated that the partial replacement of mineral P with organic manure could reshape the inorganic phosphate solubilizing and alkaline-phosphomonoesterase encoding bacterial communities towards more resilient and effective to the high P utilization and productivity over intense cultivation, providing insights into the potential of soil microbes in the efficient management of agricultural P fertilization.
The microbial cycling of phosphorus on long-term fertilized soil: Insights from phosphate oxygen isotope ratios

Abstract:Microorganisms mobilize phosphorus (P) from soil and make it available for plants. However, the role of microbial activity in soil P dynamics especially among different P pools is poorly understood largely due to methodological limitations. In this study, we analyzed the oxygen isotope ratios in phosphate (δ18OP) of sequentially extracted inorganic P (Pi) pools (H2O-Pi, NaHCO3-Pi, NaOH-Pi, and HCl-Pi) in a long-term agricultural research field in Henan, China with different fertilization histories and coupled with soil enzyme activity and P-cycling bacteria gene abundance studies. Results showed the dominant enzymes were alkaline phosphatase (APase) and phosphodiesterase (PDE), and the functional genes for P-cycling were bpp, cphy, phoX and pqqC. After long-term P fertilization, the δ18OP values of H2O-Pi, NaHCO3-Pi and NaOH-Pi pools approached to or achieved equilibrium, suggesting that the externally applied P was actively mobilized and cycled by soil microorganisms and speciated into different P pools. Based on the extent of isotope excursion among different Pi pools, the equilibrium fractionation oriented the source signature in the NaHCO3-Pi and NaOH-Pi pools, and HCl-Pi pool could be derived from P fertilizer but through multistep reactions, and mixed with the HCl-Pi from rock weathering product, which constituted HCl-Pi pool in the non-P treatments. Overall, the long-term P fertilization especially the balanced fertilization with nitrogen (N), P and potassium (K) was found to be beneficial for extensive utilization of soil P with abundant biological uptake and cell-internal Pi cycling. Variation partitioning analysis (VPA) indicated that the expression of functional genes may be stimulated to mobilize soil P under specific P pools distribution in this alkaline environment. Overall, our findings provide new insights to understand the roles of microbial activities in soil P biogeochemistry that are useful for agricultural P sustainability.
A Meta-analysis of the Effects of Organic and Inorganic Fertilizers on the Soil Microbial Community

Abstract:In order to investigate the general tendency of soil microbial community responses to fertilizers,a meta-analysis approach was used to synthesise observations on the effects of inorganic and organic fertilizer addi-tion(N: nitrogen; P: phosphorus; NP: nitrogen and phosphorus; PK: phosphorus and potassium; NPK: nitrogen,phosphorus and potassium; OF: organic fertilizer; OF+NPK: organic fertilizer plus NPK)on soil microbial communi-ties.Among the various studies,PK,NPK,OF and OF+NPK addition increased total phospholipid fatty acid(PLFA)by 52.0％,19.5％,334.3％and 58.3％,respectively; while NP,OF and OF+NPK addition increased fungi by 5.6％,21.0％and 8.2％,respectively.NP,NPK and OF addition increased bacteria by 6.4％,9.8％and 13.3％,respectively;while NP and NPK addition increased actinomycetes by 7.0％and 14.8％,respectively.Addition of ammonium ni-trate rather than urea decreased gram-negative bacteria(G-).N addition increased total PLFA、bacteria and ac-tinomycetes in croplands,but decreased fungi and bacteria in forests,and the F/B ratio in grasslands.NPK addition increased total PLFA in forests but not in croplands.The N addition rate was positively correlated with the effects of N addition on gram-positive bacteria(G+)and G-.Therefore,different fertilizers appear to have different effects on the soil microbial community.Organic fertilizers can have a greater positive effect on the soil microbial community than inorganic fertilizers.The effects of fertilizers on the soil microbial community varied with ecosystem types.The effect of N addition on the soil microbial community was related to both the forms of nitrogen that were added and the nitrogen addition rate.
Soil Microbial Composition and phoD Gene Abundance Are Sensitive to Phosphorus Level in a Long-Term Wheat-Maize Crop System

Abstract:Microbes associated with phosphorus (P) cycling are intrinsic to soil P transformation and availability for plant use but are also influenced by the application of P fertilizer. Nevertheless, the variability in soil P in the field means that integrative analyses of soil P cycling, microbial composition, and microbial functional genes related to P cycling remain very challenging. In the present study in the North China Plain, we subjected the bacterial and fungal communities to amplicon sequencing analysis and characterized the alkaline phosphatase gene ( phoD) encoding bacterial alkaline phosphatase in a long-term field experiment (10 years) with six mineral P fertilization rates up to 200 kg P ha –1 . Long-term P fertilization increased soil available P, inorganic P, and total P, while soil organic P increased until the applied P rate reached 25 kg ha –1 and then decreased. The fungal alpha-diversity decreased as P rate increased, while there were no significant effects on bacterial alpha-diversity. Community compositions of bacteria and fungi were significantly affected by P rates at order and family levels. The number of keystone taxa decreased from 10 to 3 OTUs under increasing P rates from 0 to 200 kg ha –1 . The gene copy numbers of the biomarker of the alkaline phosphatase phoD was higher at moderate P rates (25 and 50 kg ha –1 ) than at low (0 and 12.5 kg ha –1 ) and high (100 and 200 kg ha –1 ) rates of P fertilization, and was positively correlated with soil organic P concentration. One of the keystone taxa named BacOTU3771 belonging to Xanthomonadales was positively correlated with potential functional genes encoding enzymes such as glycerophosphoryl diester phosphodiesterase, acid phosphatase and negatively correlated with guinoprotein glucose dehydrogenase. Altogether, the results show the systematic effect of P gradient fertilization on P forms, the microbial community structure, keystone taxa, and functional genes associated with P cycling and highlight the potential of moderate rates of P fertilization to maintain microbial community composition, specific taxa, and levels of functional genes to achieve and sustain soil health.

Abstract:The excessive application of phosphorus (P) fertilizer is becoming a major agricultural problem, which reduces the utilization rate of the P fertilizer and degrades soil quality. The following five P fertilizer treatments were investigated to know how they affect soil properties, enzyme activity, bacterial and fungal community structure. 1) no P fertilizer (P0); 2) farmers' traditional P fertilization scheme (FP); 3) 30% reduction in P fertilizer application (P1, microbial blended fertilizer as base fertilizer); 4) 30% reduction in P fertilizer application (P2, diammonium phosphate as starting fertilizer); 5) 30% reduction in P fertilizer application (P3, microbial inoculum seed dressing). The P fertilizer reduction combined with microbial fertilizer significantly increased soil organic matter (SOM), total phosphorus (TP), available phosphorus (AP) available potassium (AK) contents, and acid phosphatase activity (ACP), however, soil urease activity was significantly reduced. Moreover, the P fertilizer reduction combined with microbial fertilizer significantly increased the relative abundance of a potential beneficial genus (i.e., Bacillus , Pseudomonas , Penicillium , and Acremonium ) and potentially pathogenic genus (i.e., Fusarium , Gibberella , and Drechslera ). The structural equation model (SEM) revealed that different P fertilizer reduction systems had significant indirect effects on bacterial and fungal community structures. The results suggested that the P fertilizer reduction combined with microbial fertilizer systems regulated the pathogenic and beneficial genus which created a microbial community that is favorable for maize growth. Moreover, the findings highlighted the importance of soil properties in determining the soil bacterial and fungal community structure.

Abstract:Soil fungi play an important role in agricultural ecosystems, yet the understanding of the responses of fungi community and function to different fertilization is limited. Here we report the responses of fungal communities and functions to 34 years of application of inorganic fertilizer (NPK), rice straw combined with inorganic fertilizer (NPKS), and no fertilizer (CK). The fungal communities were identified by Illumina high-throughput sequencing and the functional groups were inferred with FUNGuild. Three predominant Phyla of Ascomycota, Basidiomycota, Zygomycota were identified in three treatments. A significantly lower proportion of Ascomycota (47%) but higher relative abundance of Basidiomycota (28%) were observed in the NPKS treatment compared with the CK (71% and 14%, respectively). A low proportion of Basidiomycota (10%), and no significant changes in Ascomycota (74%) were found in the NPK treatment in comparison to the CK (14% and 71%, respectively). The relative abundance of Zygomycota in the NPKS (8%) and NPK (9%) treatments were significantly higher than in the CK (5%). Diversity analysis showed that indexes including Shannon, Simpson, Chao 1 and ACE all significantly declined in NPK but increased in NPKS compared with CK. FUNGuild revealed that symbiotrophic fungi in the NPKS treatment (8%) were significantly higher than in the NPK (3%) and CK (3%) treatments, and dominated by the genera Redeckera and Tricholoma, the species Trichoderma atroviride and the order Scleroderma. However, a higher proportion of pathotrophic fungi in the NPK treatment (19%) were detected than in the NPKS (11%) and CK (10%) treatments, and dominated by the species Schizangiella serpentis. Redundancy analysis (RDA) showed that fungal community structure and function were more strongly related to moisture, EC and porosity in the soil. We suggest that the sole application of inorganic fertilizer results in great changes in fungal community composition and the hazard of excess growth of pathogenic fungi, whereas combined organic fertilizer and rice straw is beneficial to maintain a healthy ecological environment and the diversity of fungal communities in paddy soil.

Abstract:Agricultural fertilization may change processes of elemental biogeochemical cycles and alter the ecological function. Ecoenzymatic stoichiometric feature plays a critical role in global soil carbon (C) metabolism, driving element cycles, and mediating atmospheric composition in response to agricultural nutrient management. Despite the importance on crop growth, the role of phosphorous (P) in compliance with eco-stoichiometry on soil C and nitrogen (N) sequestration in the paddy field remains poorly understood in the context of climate change. Here, we collected soil samples from a field experiment after 6 years of chemical P application at a gradient of 0 (P-0), 30 (P-30), 60 (P-60), and 90 (P-90) kg ha(-1) in order to evaluate the role of P on stoichiometric properties in terms of soil chemical, microbial biomass, and eco-enzyme activities as well as greenhouse gas (GHG: CO2, N2O and CH4) emissions. Continuous P input increased soil total organic C and N by 1.3-9.2% and 3%-13%, respectively. P input induced C and N limitations as indicated by the decreased ratio of C:P and N:P in the soil and microbial biomass. A synergistic mechanism among the ecoenzymatic stoichiometry, which regulated the ecological function of microbial C and N acquisition and were stoichiometrically related to P input, stimulated soil C and N sequestration in the paddy field. The lower emissions of N2O and CH4 under the higher P application (P-60 and P-90) in July and the insignificant difference in N2O emission in August compared to P-30; however, continuous P input enhanced CO2 fluxes for both samplings. There is a technical conflict for simultaneously regulating three types of GHGs in terms of the eco-stoichiometry mechanism under P fertilization. Thus, it is recommended that the P input in paddy fields not exceed 60 kg ha(-1) may maximize soil C sequestration, minimize P export, and guarantee grain yields.
Long-term high-P fertilizer input shifts soil P cycle genes and microorganism communities in dryland wheat production systems

Abstract:Phosphorus (P) fertilizer input influences crop yield, soil P cycling, and phosphate-solubilizing microorganism (PSM) communities, which play a key role in obtaining soil available phosphorus. However, the responses of soil P cycle genes and PSM communities to long-term high-P fertilizer application are less known in the Chinese Loess Plateau. In this study, metagenomics was used to explore the shifts in the abundance and compositions of soil P cycle genes and PSM communities at a long-term (14 years) trial site with four P fertilization rates. Long-term high-P (200 kg P2O5 ha−1 year−1) inputs resulted in a decrease in the abundance of the P-starvation response genes (phoR and phoP), the high-affinity phosphate-specific transporter (pst), the inorganic P-solubilization, and organic P-mineralization genes (gcd, pqq, and phoD). Long-term P inputs increased the abundance of phosphate-solubilizing bacteria but decreased the abundance of phosphate-solubilizing fungi and weakened the relationship of relevant key soil P genes. Soil total P, inorganic P, available P (AP), dissolved organic C contents, and alkaline phosphatase (ALP) activity were significantly related to soil P cycle genes and PSM community structure. In conclusion, the changes in soil P cycle genes and microbial P solubilization capacity under long-term P input conditions were mainly regulated by soil AP content and ALP activity, while long-term high-P inputs increased soil mineral P immobilization by altering the expression of soil P cycle genes and the PSM functional profiles. These consequences provide a new view for the study of soil P cycles in agricultural ecosystems with P inputs.

Abstract:PurposeVariation in soil microbial metabolism remains highly uncertain in predicting soil carbon (C) sequestration, and is particularly and poorly understood in agroecosystem with high soil phosphorus (P) variability.Materials and methodsThis study quantified metabolic limitation of microbes and their association with carbon use efficiency (CUE) via extracellular enzymatic stoichiometry and biogeochemical equilibrium models in field experiment employing five inorganic P gradients (0, 75, 150, 225, and 300 kg P ha−1) in farmland used to grow peas.Results and discussionResults showed P fertilization significantly increased soil Olsen-P and NO3−-N contents, and enzyme activities (β-1,4-glucosidase and β-D-cellobiosidase) were significantly affected by P fertilization. It indicated that P fertilization significantly decreased microbial P limitation due to the increase of soil available P. Interestingly, P application also significantly decreased microbial nitrogen (N) limitation, a phenomenon primarily attributable to increasing NO3−-N content via increasing biological N fixation within the pea field. Furthermore, P fertilization increased microbial CUE because the reduction in microbial N and P limitation leads to higher C allocation to microbial growth. Partial least squares path modeling (PLS-PM) further revealed that the reduction of microbial metabolic limitation is conducive to soil C sequestration.ConclusionsOur study revealed that P application in agroecosystem can alleviate not only microbial P limitation but also N limitation, which further reduces soil C loss via increasing microbial CUE. This study provides important insight into better understanding the mechanisms whereby fertilization mediates soil C cycling driven by microbial metabolism in agricultural ecosystems.

Contrasting fertilization response of soil phosphorus forms and functional bacteria in two newly reclaimed vegetable soils

Abstract:Fertilization is a pervasive approach to agricultural production enhancing vegetable nutrients such as phosphorus (P) absorption. However, unreasonable fertilization strategies result in high levels of residual P in vegetable planting systems. To better understand the mechanisms of soil phosphorus dynamics responding to inorganic/organic fertilization, we conducted a 3-year field experiment in two newly reclaimed vegetable fields in southern China. The results revealed that soil Olsen-P in CF (mineral fertilization) and OF (Combined application of organic and inorganic fertilizers) increased by approximately 210.6 % and 183.6 %, respectively, while stable P proportion decreased by approximately 9.2 % and 18.1 %, respectively, compared with CK. Combined application of organic and inorganic fertilizer increased the proportion of moderately labile P (NaOH-P) by 1-6 % in comparison with chemical fertilizer and facilitated the conversion from diester-P to monoester-P, indicating that applying pig manure enhanced the potential soil P bioavailability. Besides, organic-inorganic fertilization shaped a bacterial community with more connectivity and stability and changed keystone taxa related to the P transformation of the network. Phenylobacterium, Solirubrobacter, and Modestobacter were regarded as core genera for mobilizing soil phosphorus. However, residual P content in newly reclaimed soils under fertilization, especially for chemical fertilizer, remained non-negligible and may cause potential environmental risks. The partial least squares path modeling results demonstrated that fertilization management had both direct and indirect positive effects on P fraction through the improvement of soil nutrients e.g. total N and soil organic carbon, and bacterial community, while soil properties mainly determined the variation of soil P species. Our results provide comprehensive insights into the current status of legacy P forms and the vital role of fertilizer, key soil properties and bacteria in P dynamics in newly reclaimed vegetable field.

Abstract:Phosphorus (P) is an essential macronutrient crucial for both plant growth and crop production, playing a pivotal role in agriculture since the early 20th century. The symbiotic relationship between AMF and plants serves as a classic illustration. These fungi play a regulatory role in the growth and development of plants, especially in facilitating the absorption of P and carbon molecules by plants. While there has been a growing body of research on the community assembly of arbuscular mycorrhizal fungi (AMF) in recent decades, our knowledge of the processes governing the coexistence of these AMF communities influenced by P in agroecosystems remains limited. To investigate the impact of various P fertilizers on AMF communities in temperate agroecosystems, this study was conducted using soils sourced from wheat–maize rotation farmland at Henan Agricultural University Yuanyang Base. With the Illumina MiSeq high-throughput sequencing technique, we systematically examined the taxonomic composition of soil AMF at the Yuanyang Base of Henan Agricultural University in a wheat–maize rotation agricultural field. Our primary objective was to unravel the mechanisms behind AMF community assembly and stability under varying P gradient fertilization conditions. Nonmetric Multidimensional Scaling (NMDS) analysis revealed significant differences among AMF communities in field soil subjected to various treatments (p < 0.05). A torus translations test demonstrated positive associations with the three treatments in 36 out of the 51 examined AMF operational taxonomic units (OTUs), making up 70.59% (p < 0.05) of the results. Furthermore, 37.84% (14/37) of the OTUs displayed preferences for the low P concentration treatment, while 34.29% (12/35) and 32.26% (10/31) favored medium and high P concentrations, respectively. An analysis of the Normalized Stochasticity Ratio (NST) and Checkerboard Score (C-score) indicated that in temperate agroecosystems, deterministic processes predominantly governed AMF in all treatment groups, with high P conditions exerting a stronger influence than low or medium P conditions. This study underscores the profound impact of long-term P fertilizer application on AMF community structures within temperate agricultural systems employing wheat–maize rotation. Additionally, it highlights the dominant role played by deterministic processes in shaping the assembly of AMF communities in these temperate agricultural systems that use P fertilizers. These findings emphasize the need for balanced nutrient management, particularly concerning P, to ensure the stability of AMF communities.

Abstract:Biogeochemical cycling of phosphorus in the agro-ecosystem is mediated by soil microbes. These microbes regulate the availability of phosphorus in the soil. Little is known about the response of functional traits of phosphorus cycling microbes in soil fertilized with compost manure (derived from domestic waste and plant materials) or inorganic nitrogen fertilizers at high and low doses. We used a metagenomics investigation study to understand the changes in the abundance and distribution of microbial phosphorus cycling genes in agricultural farmlands receiving inorganic fertilizers (120 kg N/ha, 60 kg N/ha) or compost manure (8 tons/ha, 4 tons/ha), and in comparison with the control. Soil fertilization with high level of compost (Cp8) or low level of inorganic nitrogen (N1) fertilizer have nearly similar effects on the rhizosphere of maize plants in promoting the abundance of genes involved in phosphorus cycle. Genes such as ppk involved in polyphosphate formation and pstSABC (for phosphate transportation) are highly enriched in these treatments. These genes facilitate phosphorus immobilization. At a high dose of inorganic fertilizer application or low compost manure treatment, the phosphorus cycling genes were repressed and the abundance decreased. The bacterial families Bacillaceae and Carnobacteriaceae were very abundant in the high inorganic fertilizer (N2) treated soil, while Pseudonocardiaceae, Clostridiaceae, Cytophagaceae, Micromonosporaceae, Thermomonosporaceae, Nocardiopsaceae, Sphaerobacteraceae, Thermoactinomycetaceae, Planococcaceae, Intrasporangiaceae, Opitutaceae, Acidimicrobiaceae, Frankiaceae were most abundant in Cp8. Pyrenophora, Talaromyces, and Trichophyton fungi were observed to be dominant in Cp8 and Methanosarcina, Methanobrevibacter, Methanoculleus, and Methanosphaera archaea have the highest percentage occurrence in Cp8. Moreover, N2 treatment, Cenarchaeum, Candidatus Nitrososphaera, and Nitrosopumilus were most abundant among fertilized soils. Our findings have brought to light the basis for the manipulation of rhizosphere microbial communities and their genes to improve availability of phosphorus as well as phosphorus cycle regulation in agro-ecosystems.

Abstract:The influence of long-term chemical fertilization on soil microbial communities has been one of the frontier topics of agricultural and environmental sciences and is critical for linking soil microbial flora with soil functions. In this study, 16S rRNA gene pyrosequencing and a functional gene array, geochip 4.0, were used to investigate the shifts in microbial composition and functional gene structure in paddy soils with different fertilization treatments over a 22-year period. These included a control without fertilizers; chemical nitrogen fertilizer (N); N and phosphate (NP); N and potassium (NK); and N, P and K (NPK). Based on 16S rRNA gene data, both species evenness and key genera were affected by P fertilization. Functional gene array-based analysis revealed that long-term fertilization significantly changed the overall microbial functional structures. Chemical fertilization significantly increased the diversity and abundance of most genes involved in C, N, P and S cycling, especially for the treatments NK and NPK. Significant correlations were found among functional gene structure and abundance, related soil enzymatic activities and rice yield, suggesting that a fertilizer-induced shift in the microbial community may accelerate the nutrient turnover in soil, which in turn influenced rice growth. The effect of N fertilization on soil microbial functional genes was mitigated by the addition of P fertilizer in this P-limited paddy soil, suggesting that balanced chemical fertilization is beneficial to the soil microbial community and its functions.
Responses of Crop and Soil Phosphorus Fractions to Long-Term Fertilization Regimes in a Loess Soil in Northwest China

Abstract:Contrasting fertilization modifies soil phosphorus (P) transformation and bioavailability, which impact crop P uptake and P migration in the soil profile. A long-term (25-year) fertilizer experiment was employed to investigate crop yield, P uptake and changes in sequentially extracted P fractions in the soil profile, and their relationships on a calcareous soil derived from loess material under a winter wheat and summer maize double-cropping system. The experiment involved seven nutrient management treatments: control (CK, no nutrient input), N, NK, NP, and NPK, representing various combinations of synthetic nitrogen (N), phosphate (P), and potassium (K) applications, as well as combinations of NPK fertilizers with either crop residues (SNPK, where S refers to maize stalk or wheat straw) or manure (MNPK, where M refers to dairy manure). Wheat and maize yields were significantly higher with P input fertilizer relative to the P-omitted treatments. Long-term application of P-containing fertilizers markedly raised the contents of inorganic (Pi) and organic (Po) P fractions at 0–20 cm depth compared with the P-omitted treatments. Moreover, both Pi and Po fractions were markedly higher under MNPK than under NPK and SNPK treatments. For achieving high yield for wheat and maize, the critical contents of labile P were 54 and 63 mg kg−1, and those of moderately labile P were 48 and 49 mg kg−1, respectively, defined by the linear plateau model. In addition, the change points of labile P and moderately labile P were 99 and 70 mg kg−1, above which CaCl2-P content significantly increased. Moreover, long-term P input significantly accumulated different P fractions in the deeper soil layers up to 100 cm, with large portions of organic P being a composite of labile and moderately labile P, especially in MNPK treatment. Our results suggest that excessive P supply with organic manure resulted in massive P accumulation in the topsoil and promoted soil P fraction transformation and availability in the deep soil layers, especially in an organic P form that has often been neglected.

Abstract:Microorganisms regulate the responses of terrestrial ecosystems to anthropogenic nutrient inputs. The escalation of anthropogenic activities has resulted in a rise in the primary terrestrial constraining elements, namely nitrogen (N) and phosphorus (P). Nevertheless, the specific mechanisms governing the influence of soil microbial community structure and ecological processes in ecologically vulnerable and delicate semi-arid loess agroecosystems remain inadequately understood. Therefore, we explored the effects of different N and P additions on soil microbial community structure and its associated ecological processes in the farmland of Chinese Loess Plateau based on a 36-a long-term experiment. Nine fertilization treatments with complete interactions of high, medium, and low N and P gradients were set up. Soil physical and chemical properties, along with the microbial community structure were measured in this study. Additionally, relevant ecological processes such as microbial biomass, respiration, N mineralization, and enzyme activity were quantified. To elucidate the relationships between these variables, we examined correlation-mediated processes using statistical techniques, including redundancy analysis (RDA) and structural equation modeling (SEM). The results showed that the addition of N alone had a detrimental effect on soil microbial biomass, mineralized N accumulation, and β-1,4-glucosidase activity. Conversely, the addition of P exhibited an opposing effect, leading to positive influences on these soil parameters. The interactive addition of N and P significantly changed the microbial community structure, increasing microbial activity (microbial biomass and soil respiration), but decreasing the accumulation of mineralized N. Among them, N 24 P 12 treatment showed the greatest increase in the soil nutrient content and respiration. N 12 P 12 treatment increased the overall enzyme activity and total phospholipid fatty acid (PLFA) content by 70.93%. N and P nutrient contents of the soil dominate the microbial community structure and the corresponding changes in hydrolytic enzymes. Soil microbial biomass, respiration, and overall enzyme activity are driven by mineralized N. Our study provides a theoretical basis for exploring energy conversion processes of soil microbial community and environmental sustainability under long-term N and P additions in semi-arid loess areas.

Abstract:Abstract. Understanding soil phosphorus (P) transformation and turnover under various fertilization managements is important for evaluating sustainable P fertility and potential bioavailability in agriculture managements. Thus, long-term fertilization experiments (∼ 38 years) with the application of different inorganic and organic fertilizers in paddy red soils were conducted to determine the effect of different fertilizer applications on P pool accumulation and microbial communities, especially for phosphate-solubilizing microorganisms (PSMs). Long-term inorganic P (IP) fertilization increased the concentrations of total P (TP) (∼ 479 mg kg−1), available P (AP) (∼ 417 mg kg−1) and inorganic P (∼ 18 mg kg−1), but manure fertilization accelerated the accumulation of organic P, especially for orthophosphate monoesters (e.g., myo-IHP, ∼ 12 mg kg−1). Long-term mineral fertilization decreased bacterial richness, evenness and complexation of bacterial networks. In contrast, long-term manure fertilization and rhizosphere accumulated more amounts of total carbon, total nitrogen, and organic carbon, as well as regulated the soil pH, thus improving the separation of bacterial communities. Furthermore, PSM compositions were greatly influenced by fertilization managements and rhizosphere. For example, inorganic P fertilization increased the abundance of Thiobacillus (i.e., the most abundant phosphate-solubilizing bacteria (PSB) in this study) and shifted the community structure of PSB. Correspondingly, the concentrations of inorganic and total P were the key factors for the variation of the PSB community structure. These findings are beneficial for understanding the variation of inorganic and organic P pools and the microbial community, especially for PSMs under long-term inorganic and/or organic fertilization.

Abstract:The intricate decomposition pathways within soil micro-food webs are vital for cycling soil organic carbon and nutrients, influencing the quality, productivity, and sustainability of soil systems. However, the impact of diverse phosphorus addition on these organic decomposition pathways still needs to be explored. In an 8-year experiment, phosphorus (P) fertilizer was added at varying levels (0 kg ha −1 , CK; 60 kg ha −1 , P60; 120 kg ha −1 , P120; and 180 kg ha −1 , P180), to investigate the response of the soil micro-food web. The results revealed a significant effect of phosphorus addition on soil microorganisms and nematodes, with P60 exerting a greater influence than other treatments. At P60, the Shannon index of nematodes and fungi surpassed other treatments, indicating higher diversity, while the Shannon index of bacteria was lower. The Chao1 index of bacteria and fungi at P60 was higher, contrasting with the lower index for nematodes. Metabolic footprints of bacterivores and omnivores–predators (BFMF and OPMF) were higher at P60, while metabolic footprints of fungivores and plant parasites (FFMF and PPMF) were lower, signifying altered energy flow. Functional metabolic footprints and energy flow analysis unveiled a stable soil micro-food web structure at P60, with enhanced energy conversion efficiency. Network analysis illustrated positive correlations between fungi, fungivorous nematodes (FF), and omnivorous-predatory nematodes (OP) at P60, while P120 and P180 showed positive correlations among bacteria, bacterivorous nematodes (BF), and OP. Path analysis underscored the higher contribution rate of BF-C, FF-C, and OP-C to soil organic carbon at P60 compared with P120 and P180. These findings suggest that nutrient interactions between fungi and nematodes regulate soil micro-food web decomposition under low phosphorus concentrations. In contrast, interactions between bacteria and nematodes dominate at high phosphorus concentrations. The study indicates that adding phosphorus has nuanced bottom-up effects, intricately shaping the structure and activity of the pathways and underscoring the need for a comprehensive understanding of nutrient dynamics in soil ecosystems.
