Abstract:Abstract Separating plasma from whole blood is an important sample processing technique required for fundamental biomedical research, medical diagnostics, and therapeutic applications. Traditional protocols for plasma isolation require multiple centrifugation steps or multiunit microfluidic processing to sequentially remove large red blood cells (RBCs) and white blood cells (WBCs), followed by the removal of small platelets. Here, we present an acoustofluidic platform capable of efficiently removing RBCs, WBCs, and platelets from whole blood in a single step. By leveraging differences in the acoustic impedances of fluids, our device generates significantly greater forces on suspended particles than conventional microfluidic approaches, enabling the removal of both large blood cells and smaller platelets in a single unit. As a result, undiluted human whole blood can be processed by our device to remove both blood cells and platelets (>90%) at low voltages (25 Vpp). The ability to successfully remove blood cells and platelets from plasma without altering the properties of the proteins and antibodies present creates numerous potential applications for our platform in biomedical research, as well as plasma-based diagnostics and therapeutics. Furthermore, the microfluidic nature of our device offers advantages such as portability, cost efficiency, and the ability to process small-volume samples.

Microfluidic Separation of Particles from Whole Blood Using Shear Induced Diffusion

Abstract:Extraction of cells of interest directly from human whole blood is in high demand. However, it is extremely challenging due to the excessive cell populations leading to non-Newtonian hemodynamics. Herein, we describe a simple microfluidic approach to take the advantage of the concentrated cells for direct isolation of larger particles spiked in whole blood, which could shift the current paradigm in cell separation from low volume fraction sample to concentrated suspension and from the Newtonian to non-Newtonian fluid. Our device fabricated in Polydimethylsiloxane (PDMS) via standard soft photolithography. With saline solution flowing in the channel center and splitting the blood sample into two side streams, the spiked larger particles rapidly migrate from the whole blood to the saline stream without external force fields. Our experimental results has suggested such intriguing particle translocation is mainly attributed to the shear induced diffusion in concentrated suspensions as well as the viscoelastic property of blood. Inertial force can also contribute to this process. The harmonic interactions among these passive force fields have successfully led to extraction, focusing and separation of 18.7 mu m -diameter particles from whole blood with a high efficiency (similar to 89%) and a superb throughput (>10(6) cells per second), which outperforms existing approaches such as spiral microchannels. Shortly, we have demonstrated a novel simple and effective separation scheme which directly works for complex bodily samples without a hassle of sample preparation. Our approach is very promising as it holds potential to separate cells from whole blood.

Abstract:Separation of blood components is required in many diagnostic applications and blood processes. In laboratories, blood is usually fractionated by manual operation involving a bulk centrifugation equipment, which significantly increases logistic burden. Blood sample processing in the field and resource-limited settings cannot be readily implemented without the use of microfluidic technology. In this study, we developed a small footprint, rapid, and passive microfluidic channel device that relied on margination and inertial focusing effects for blood component separation. No blood dilution, lysis, or labeling step was needed as to preserve sample integrity. One main innovation of this work was the insertion of fluidic restrictors at outlet ports to divert the separation interface into designated outlet channels. Thus, separation efficiency was significantly improved in comparison to previous works. We demonstrated different operation modes ranging from platelet or plasma extraction from human whole blood to platelet concentration from platelet-rich plasma through the manipulation of outlet port fluidic resistance. Using straight microfluidic channels with a high aspect ratio rectangular cross section, we demonstrated 95.4% platelet purity extracted from human whole blood. In plasma extraction, 99.9% RBC removal rate was achieved. We also demonstrated 2.6× concentration of platelet-rich plasma solution to produce platelet concentrate. The extraction efficiency and throughput rate are scalable with continuous and clog-free recirculation operation, in contrast to other blood fractionation approaches using filtration membranes or affinity-based purification methods. Our microfluidic blood separation method is highly tunable and versatile, and easy to be integrated into multi-step blood processing and advanced sample preparation workflows.
Lab-on-a-disk extraction of PBMC and metered plasma from whole blood: An advanced event-triggered valving strategy.

Abstract:In this paper, we present a centrifugal microfluidic concept employing event-triggered valving for automated extraction of metered plasma and peripheral blood mononuclear cells (PBMCs). This "lab-on-a-disk" system has been developed for retrieving different density layers from a liquid column by "overflowing" the layers sequentially using the pressure exerted by a density-gradient liquid. Defined volumes of plasma and PBMCs were efficiently forwarded into designated microfluidic chambers as a sample preparation step prior to further downstream processing. Furthermore, the extracted PBMCs were counted directly on-disk using an automated optical unit by object-based image analysis, thus eliminating the requirement for the post-processing of the extracted PBMCs. This study is a direct continuation of our previous work1 where we demonstrated combined on-disk detection of C-reactive protein and quantification of PBMCs following on-disk extraction of plasma and PBMCs from a single blood sample using a centrifugo-pneumatic valving mechanism. However, the former valving technique featured limited PBMC extraction efficiency. Here, integrating the novel concept along with event-triggered valving mechanism, we eliminated the occurrence of a specific microfluidic effect, which led us to increase PBMC extraction efficiency to 88%. This extraction method has the potential to be utilized for efficiently separating multiple density layers from a liquid sample in relevant biomedical applications.

Abstract:Rapid separation of white blood cells from whole blood sample is often required for their subsequent analyses of functions and phenotypes, and many advances have been made in this field. However, most current microfiltration-based cell separation microfluidic chips still suffer from low-throughput and membrane clogging. This paper reports on a high-throughput and clogging-free microfluidic filtration platform, which features with an integrated bidirectional micropump and commercially available polycarbonate microporous membranes. The integrated bidirectional micropump enables the fluid to flush micropores back and forth, effectively avoiding membrane clogging. The microporous membrane allows red blood cells passing through high-density pores in a cross-flow mixed with dead-end filtration mode. All the separation processes, including blood and buffer loading, separation, and sample collection, are automatically controlled for easy operation and high throughput. Both microbead mixture and undiluted whole blood sample are separated by the platform effectively. In particular, for white blood cell separation, the chip recovered 72.1% white blood cells with an over 232-fold enrichment ratio at a throughput as high as 37.5 μl/min. This high-throughput, clogging-free, and highly integrated platform holds great promise for point-of-care blood pretreatment, analysis, and diagnosis applications.

Abstract:Extraction of cells of interest directly from whole blood is in high demand, yet extraordinary challenging due to the complex hemodynamics and hemorheology of the sample. Herein, we describe a new microfluidic platform that exploits the intrinsic complex properties of blood for continuous size-selective focusing and separation of cells directly from unprocessed whole blood. The novel system only requires routinely accessible saline solution to form a sandwiched fluid configuration and to initiate a strong effect of shear-induced diffusion of cells, which is coupled with fluid inertia for effective separation. Separations of beads and cells from whole blood have been successfully demonstrated with high efficiency (89.8%) at throughput of 6.75 mL/hr (106–107 cells/s) of whole blood. Rapid isolation of circulating tumor cells (CTCs) from peripheral blood sample of hepatocarcinoma patients is also shown as a proof of principle.

Abstract:This study reports a microfluidic device for whole blood processing. The device uses the bifurcation law, cross-flow method, and hydrodynamic flow for simultaneous extraction of plasma, red blood cells, and on-chip white blood cell trapping. The results demonstrate successful plasma and red blood cell collection with a minimum dilution factor (0.76x) and low haemolysis effect. The extracted red blood cells can also be applied for blood type tests. Moreover, the device can trap up to ~1,800 white blood cells in 20 minutes. The three components can be collected simultaneously using only 6 μL of whole blood without any sample preparation processes. Based on these features, the microfluidic device enables low-cost, rapid, and efficient whole blood processing functionality that could potentially be applied for blood analysis in resource-limited environments or point-of-care settings.

Abstract:Drug selection and treatment monitoring via minimally invasive liquid biopsy using circulating tumor cells (CTCs) are expected to be realized in the near future. For clinical applications of CTCs, simple, high-throughput, single-step CTC isolation from whole blood without red blood cell (RBC) lysis and centrifugation remains a crucial challenge. In this study, we developed a novel cancer cell separation chip, "hybrid double-spiral chip", that involves the serial combination of two different Dean flow fractionation (DFF) separation modes of half and full Dean cycles, which is the hybrid DFF separation mode for ultra-high-throughput blood processing at high precision and size-resolution separation. The chip allows fast processing of 5 mL whole blood within 30 min without RBC lysis and centrifugation. RBC and white blood cell (WBC) depletion rates of over 99.9% and 99%, respectively, were achieved. The average recovery rate of spiked A549 cancer cells was 87% with as low as 200 cells in 5 mL blood. The device can achieve serial reduction in the number of cells from approximately 1010 cells of whole blood to 108 cells, and subsequently to an order of 106 cells. The developed method can be combined with measurements of all recovered cells using imaging flow cytometry. As proof of concept, CTCs were successfully enriched and enumerated from the blood of metastatic breast cancer patients (N = 10, 1-69 CTCs per 5 mL) and metastatic prostate cancer patients (N = 10, 1-39 CTCs per 5 mL). We believe that the developed method will be beneficial for automated clinical analysis of rare CTCs from whole blood.

Abstract:Human Peripheral Blood Mononuclear Cells (PBMCs) are isolated from peripheral blood and identified as any blood cell with a round nucleus that exhibits immune responses and undergoes immunophenotypic changes upon exposure to various pathophysiological stimuli. Obtaining high-recovery and clinical-grade PBMCs without decreasing cell viability and causing stress is crucial for disease diagnosis and successful immunotherapy. However, traditional manual PBMCs extraction methods rely on manual intervention with less recovery rate and reliability. In this study, we introduced a novel and efficient strategy for the fully automated extraction of PBMCs based on a Lab-on-a-Disk (LoaD) platform. The centrifugal chip used percoll as density gradient media (DGM) for separation and extraction on account of the density difference of cells in whole blood, without labeling and any additional extra cellular filtration or cell lysis steps. Above all, we proposed a high-speed triggered siphon valve, which was closed under the speed of cell sedimentation and subsequently opened by increasing speed to complete the extraction of PBMCs. It can avoid the problem that previous siphon valves rely on unstable hydrophilic surface treatment and prime under low/zero speed conditions. With valves and the clock channel integrated on the chip, users can achieve fully automated collection of PBMCs. Compared with the clinical laboratory results, the recovery rate of extracted PBMCs was 80 %. The experimental results prove that the high-speed triggered siphon valve improves the extraction efficiency of PBMCs. The robust chips, which are not only simple to manufacture and assemble but also stable and reliable to use, have great potential in biomedical and clinical applications.

Risk for maternal depression and child aggression in Early Head Start families: A test of ecological models.

Abstract:Current literature indicates that risk for maternal depression is substantial in low-income families. A large body of research also indicates that when mothers are depressed, children are at risk for a number of developmental difficulties. While mutual influence between child and parental difficulties has been noted, few studies examine risk factors for both depression and child aggression within ecological models. The present cross-site study examined the unique and additive contributions of contextual factors, including SES and family functioning, on maternal depression and child aggression in Early Head Start families. A multiethnic sample of parents and their children, between the ages of 12 and 43 months, participated in this study. Families came from five Early Head Start programs across the United States, representing both urban and rural areas. Structural equation models (SEM) demonstrate mutual links between depression and aggression, mediated at least in part by ecological factors. SEM indicated that 36.4% of the variance in child aggression is accounted for in a model linking aggressive behavior to parent depression, stress, and couple-level functioning, as well as other family interaction variables. A second model focusing on maternal depression revealed that 44.5% of the variance in maternal depression was accounted for through family factors, including couple-related support and satisfaction and parenting stress. In this second model, child aggression was indirectly linked to maternal depression. These data have important implications for programs serving at-risk families.

Abstract:Separating plasma or serum from blood is essential for precise testing. However, extracting precise plasma quantities outside the laboratory poses challenges. A recent study has introduced a capillary force-driven membrane filtration technique to accurately separate small plasma volumes. This method efficiently isolates 100–200 μL of pure human whole blood with a 48% hematocrit, resulting in 5–30 μL of plasma with less than a 10% margin of error. The entire process is completed within 20 min, offering a simple and cost-effective approach to blood separation. This study has successfully addressed the bottleneck in self-service POCT, ensuring testing accuracy. This innovative method shows promise for clinical diagnostics and point-of-care testing.

Abstract:Microfluidics can be utilized to manipulate cells and to separate the plasma from blood samples. Based on the crossflow and the centrifugation effect, a novel chip utilizing spiral microchannel with fence and cofferdam as filtration structures was fabricated by the micromachining technology. A cofferdam and a pillared fence along with the spiral microchannel was used to separate blood cells from the plasma, while white blood cells (WBCs) and red blood cells (RBCs) could also be segregated at the same time. Clogging and jamming in this filtration structure are efficiently alleviated in this device. The presented separation chip for cells and plasma isolation could be integrated into μTAS for point-of-care diagnostics in the near future.

Abstract:Sample preparation for mass spectroscopy typically involves several liquid and solid phase clean-ups, extractions, and other unit operations, which are labour-intensive and error-prone. We demonstrate a centrifugal microfluidic platform that automates the whole blood sample's preparation and clean-up by combining traditional liquid-phase and multiple solid-phase extractions for applications in mass spectroscopy (MS)-based small molecule detection. Liquid phase extraction was performed using methanol to precipitate proteins in plasma separated from a blood sample under centrifugal force. The preloaded solid phase composed of C18 beads then removed lipids with a combination of silica particles, which further cleaned up any remaining proteins. We further integrated the application of this sample prep disc with matrix-assisted laser desorption/ionization (MALDI) MS by using glancing angle deposition films, which further cleaned up the processed sample by segregating the electrolyte background from the sample salts. Additionally, hydrophilic interaction liquid chromatography (HILIC) MS was employed for detecting targeted free amino acids. Therefore, several representative ionic metabolites, including several amino acids and organic acids from blood samples, were analysed by both MALDI-MS and HILIC-MS to demonstrate the performance of this sample preparation disc. The fully automated blood sample preparation procedure only took 35 mins, with a throughput of three parallel units.

Abstract:Herein, we described an integrated microfluidic system for multi-target biochemical analysis of micro-volumes of blood samples. This system mainly consists of a microfluid handling module, an integrated microwell array chip, a temperature control module and a spectral detection module. A novel microfluidic plasma extraction approach was developed by coupling the filter membrane-based plasma separation technique with the microfluidic liquid handling technique. With this approach, quantitative extraction of trace blood with volumes as low as a drop (30 mu L) can be automatically realized. Multiple operations in multi-target biochemical analysis are achieved including micro-volume blood collecting, quantitative plasma extraction, plasma dilution, plasma distribution, transferring, biochemical reaction and absorption spectroscopy detection. This system was applied to the multi target biochemical analysis of glucose, cholesterol and total protein in blood samples and could achieve the determination of the target analytes of a 30-mu L blood sample within 11 min. The experimental results are consistent with those obtained by a commercial biochemical analyzer.

Abstract:A major hurdle for blood-based proteomic diagnostics is efficient transport of specimens from the collection site to the testing laboratory. Dried blood spots have shown utility for diagnostic applications, specifically those where red blood cell hemolysis and contamination of specimens with hemoglobin is not confounding. Conversely, applications that are sensitive to the presence of the hemoglobin subunits require blood separation, which relies on centrifugation to collect plasma/serum, and then cold-chain custody during shipping. All these factors introduce complexities and potentially increased costs. Here we report on a novel whole blood-collection device (BCD) that efficiently separates the liquid from cellular components, minimizes hemolysis in the plasma fraction, and maintains protein integrity during ambient transport. The simplicity of the design makes the device ideal for field use. Whole blood is acquired through venipuncture and applied to the device with an exact volume pipette. The BCD design was based on lateral-flow principles in which whole blood was applied to a defined area, allowing two minutes for blood absorption into the separation membrane, then closed for shipment. The diagnostic utility of the device was further demonstrated with shipments from multiple sites (n = 33) across the U.S. sent to two different centralized laboratories for analyses using liquid chromatography/mass spectrometry (LC/MS/MS) and matrix assisted laser desorption/ionization-time of flight (MALDI-ToF) commercial assays. Specimens showed high levels of result label concordance for the LC/MS/MS assay (Negative Predictive Value = 98%) and MALDI-ToF assay (100% result concordance). The overall goal of the device is to simplify specimen transport to the laboratory and produce clinical test results equivalent to established collection methods.

Low-stress Microfluidic Density-gradient Centrifugation for Blood Cell Sorting

Abstract:Density gradient centrifugation exploits density differences between different blood cells to accomplish separation of peripheral blood mononuclear cells (PBMCs) from polymorphonuclear (PNM) cells, and erythrocytes or red blood cells (RBCs). While density gradient centrifugation offers a label-free alternative avoiding the use of harsh lysis buffers for blood cell isolation, it is a time-consuming and labor-intensive process during which blood cells are subject to high-levels of centrifugal force that can artifactually activate cells. To provide a low-stress alternative to this elegant method, we miniaturized and automated this process using microfluidics to ensure continuous PBMCs isolation from whole blood while avoiding the exposure to high-levels of centrifugal stress in a simple flow-through format. Within this device, a density gradient is established by exploiting laminar flow within microfluidic channels to layer a thin stream of blood over a larger stream of Ficoll. Using this approach we demonstrate successful isolation of PBMCs from whole blood with preservation of monocytes and different lymphocyte subpopulations similar to that seen with conventional density gradient centrifugation. Evaluation of activation status of PBMCs isolated using this technique shows that our approach achieves minimal isolation process induced activation of cells in comparison to conventional lysis or density gradient centrifugation. This simple, automated microfluidic density gradient centrifugation technique can potentially serve as tool for rapid and activation-free technique for isolation of PBMCs from whole blood for point-of-care applications.

Label-free blood cells separation and enrichment from whole blood by high-throughput hydrodynamic and inertial force

Abstract:White blood cells or rare cells such as cancer cells with red blood cells in the whole blood, the concentration difference is approximately 1000 times in whole blood. To handle this extremely large cells population in microfluidic chip, we created a microdevice that takes advantage of the intrinsic features of blood flow in the microcirculation to separate leukocytes from whole blood, and solve clogging problem due to the large amount of blood cells (108-9 cell/ml) by high-throughput hydrodynamic separation in microchannel. Results also showed high separation efficiency of blood cells by inertial force in microfluidics that containing high concentration whole blood cells (4.3×106 cells/μl). We demonstrated whole blood human separation in double layer structured microfluidic devices, utilizing the principle of hydrodynamic inertial forces to separate red blood cells from whole blood and enrich leukocytes (white blood cells) extracting by the elastic properties of blood cell and the inertial force, like shear lift force in a continuous flow separation, a high separation efficiency (~92%) and high throughput flow rate (10 mL, 300 μl/s) was obtained in this microfluidic chip.

Abstract:Isolation and molecular characterization of rare cells (e.g. circulating tumor and stem cells) within biological fluids and tissues has significant potential in clinical diagnostics and personalized medicine. The present work describes an integrated platform of sample procurement, preparation, and analysis for deep proteomic profiling of rare cells in blood. Microfluidic magnetophoretic isolation of target cells spiked into 1 ml of blood at the level of 1000-2000 cells/ml, followed by focused acoustics-assisted sample preparation has been coupled with one-dimensional PLOT-LC-MS methodology. The resulting zeptomole detection sensitivity enabled identification of ∼4000 proteins with injection of the equivalent of only 100-200 cells per analysis. The characterization of rare cells in limited volumes of physiological fluids is shown by the isolation and quantitative proteomic profiling of first MCF-7 cells spiked into whole blood as a model system and then two CD133+ endothelial progenitor and hematopoietic cells in whole blood from volunteers.

Abstract:The separation of cancer cells from blood samples is one of the most crucial tasks in cancer research. However, existing methods tend to be expensive and labor intensive. Accordingly, the present study proposes a low-cost platform that uses hydrodynamic effects for the label-free separation of cancer cells from whole blood samples using a simple centrifugal microfluidic device consisting of a Y-shaped microchannel, a contraction–expansion array (CEA) microchannel, and a bifurcation region. To enhance the separation efficiency, the input branches of the Y-shaped microchannel are designed with different widths to generate a sheath flow rate greater than the sample flow rate. As the sample flows through the CEA microchannel, the cancer cells are separated from the blood cells through inertial effects and the bifurcation law. Finally, the cancer cells are collected from the low-flow-rate branch of the bifurcation region. The feasibility of the device is first demonstrated by numerical simulations. Experimental trials are then performed to separate K562 cancer cells from blood samples with various hematocrit concentrations at disk rotational speeds ranging from 1000 to 3000 rpm. The experimental results show that the cancer cells can be successfully separated from a diluted blood sample with a ratio of 1:1.2 × 10 5 K562 cells to blood cells with a high efficiency of 90% at an angular velocity of 2000 rpm.
