Abstract:STOFFENMANAGER (R) and the Advanced REACH Tool (ART) are recommended tools by the European Chemical Agency for regulatory chemical safety assessment. The models are widely used and accepted within the scientific community. STOFFENMANAGER (R) alone has more than 37 000 users globally and more than 310 000 risk assessment have been carried out by 2020. Regardless of their widespread use, this is the first study evaluating the theoretical backgrounds of each model. STOFFENMANAGER (R) and ART are based on a modified multiplicative model where an exposure base level (mg m(-3)) is replaced with a dimensionless intrinsic emission score and the exposure modifying factors are replaced with multipliers that are mainly based on subjective categories that are selected by using exposure taxonomy. The intrinsic emission is a unit of concentration to the substance emission potential that represents the concentration generated in a standardized task without local ventilation. Further information or scientific justification for this selection is not provided. The multipliers have mainly discrete values given in natural logarithm steps (..., 0.3, 1, 3, ...) that are allocated by expert judgements. The multipliers scientific reasoning or link to physical quantities is not reported. The models calculate a subjective exposure score, which is then translated to an exposure level (mg m(-3)) by using a calibration factor. The calibration factor is assigned by comparing the measured personal exposure levels with the exposure score that is calculated for the respective exposure scenarios. A mixed effect regression model was used to calculate correlation factors for four exposure group [e.g. dusts, vapors, mists (low-volatiles), and solid object/abrasion] by using similar to 1000 measurements for STOFFENMANAGER (R) and 3000 measurements for ART. The measurement data for calibration are collected from different exposure groups. For example, for dusts the calibration data were pooled from exposure measurements sampled from pharmacies, bakeries, construction industry, and so on, which violates the empirical model basic principles. The calibration databases are not publicly available and thus their quality or subjective selections cannot be evaluated. STOFFENMANAGER (R) and ART can be classified as subjective categorization tools providing qualitative values as their outputs. By definition, STOFFENMANAGER (R) and ART cannot be classified as mechanistic models or empirical models. This modeling algorithm does not reflect the physical concept originally presented for the STOFFENMANAGER (R) and ART. A literature review showed that the models have been validated only at the 'operational analysis' level that describes the model usability. This review revealed that the accuracy of STOFFENMANAGER (R) is in the range of 100 000 and for ART 100. Calibration and validation studies have shown that typical log-transformed predicted exposure concentration and measured exposure levels often exhibit weak Pearson's correlations (r is <0.6) for both STOFFENMANAGER (R) and ART. Based on these limitations and performance departure from regulatory criteria for risk assessment models, it is recommended that STOFFENMANAGER (R) and ART regulatory acceptance for chemical safety decision making should be explicitly qualified as to their current deficiencies.

Abstract:This paper describes a Bayesian model for the assessment of inhalation exposures in an occupational setting; the methodology underpins a freely available web-based application for exposure assessment, the Advanced REACH Tool (ART). The ART is a higher tier exposure tool that combines disparate sources of information within a Bayesian statistical framework. The information is obtained from expert knowledge expressed in a calibrated mechanistic model of exposure assessment, data on inter- and intra-individual variability in exposures from the literature, and context-specific exposure measurements. The ART provides central estimates and credible intervals for different percentiles of the exposure distribution, for full-shift and long-term average exposures. The ART can produce exposure estimates in the absence of measurements, but the precision of the estimates improves as more data become available. The methodology presented in this paper is able to utilize partially analogous data, a novel approach designed to make efficient use of a sparsely populated measurement database although some additional research is still required before practical implementation. The methodology is demonstrated using two worked examples: an exposure to copper pyrithione in the spraying of antifouling paints and an exposure to ethyl acetate in shoe repair.
Calibration of the Dermal Advanced REACH Tool (dART) Mechanistic Model

Abstract:The dermal Advanced REACH Tool (dART) is a Tier 2 exposure modelling tool currently in development for estimating dermal exposure to the hands (mg min-1) for non-volatile liquid and solids-in-liquid products. The dART builds upon the existing ART framework and describes three mass transport processes [deposition (Dhands), direct emission and direct contact (Ehands), and contact transfer (Thands)] that may each contribute to dermal exposure. The mechanistic model that underpins the dART and its applicability domain has already been described in previous work. This paper describes the process of calibrating the mechanistic model such that the dimensionless score that results from encoding contextual information about a task into the determinants of the dART can be converted into a prediction of exposure (mg min-1). Furthermore, as a consequence of calibration, the uncertainty in a dART prediction may be quantified via a confidence interval. Thirty-six experimental studies were identified that satisfied the conditions of: (i) high-quality contextual information that was sufficient to confidently code the dART mechanistic model determinants; (ii) reliable exposure measurement data sets were available. From these studies, 40 exposure scenarios were subsequently developed. A non-linear log-normal mixed-effect model was fitted to the data set of Dhands, Ehands, and Thands scores and corresponding measurement data. The dART model was shown to be consistent with activities covering a broad range of tasks [spray applications, activities involving open liquid surfaces (e.g. dipping, mixing), handling of contaminated objects, spreading of liquid products, and transfer of products (e.g. pouring of liquid)]. Exposures resulting from a particular task were each dominated by one or two of the identified mass transport processes. As a consequence of calibration, an estimate of the uncertainty associated with a mechanistic model estimate is available. A 90% multiplicative interval is approximately a factor of six. This represents poorer overall precision than the (inhalation) ART model for dusts and vapours, although better than the ART model for mists. Considering the complexity of the conceptual model compared with the ART, the wide variety of exposure scenarios considered with differing dominant routes, and the particular challenges that result from the consideration of measurement data both above and beneath a protective glove, the precision of the calibrated dART mechanistic model is reasonable for well-documented exposure scenarios coded by experts. However, as the inputs to the model are based upon user judgement, in practical use, the reliability of predictions will be dependent upon both the competence of users and the quality of contextual information available on an exposure scenario.

Abstract:This article describes the development of a mechanistic model for underpinning the dermal Advanced REACH Tool (dART), an extension of the existing ART model and its software platform. It was developed for hand exposure to low volatile liquids (vapour pressure ≤ 10 Pa at 20°C) including solids-in-liquid products. The model is based on an existing conceptual dermal source-receptor model that has been integrated into the ART framework. A structured taxonomy of workplace activities referred to as activity classes are adopted from ART. Three key processes involved in mass transport associated with dermal exposure are applied, i.e. deposition, direct emission and contact, and transfer. For deposition, the model adopts all the relevant modifying factors (MFs) applied in ART. In terms of direct emission and contact (e.g. splashes) and transfer (e.g. hand-surface contacts), the model defines independent principal MFs, i.e. substance-related factors, activity-related factors, localized- and dispersion control and exposed surface area of the hands. To address event-based exposures as much as possible, the model includes crucial events during an activity (e.g. hand immersions) and translates objective information on tools and equipment (manual or automated) to probable events (e.g. splashes) and worker behaviours (e.g. surface contacts). Based on an extensive review of peer-reviewed literature and unpublished field studies, multipliers were assigned to each determinant and provide an approximated (dimensionless) numerical value. In the absence of (sufficient) evidence, multipliers were assigned to determinants based on assumptions made during discussions by experts in the consortium. A worked example is presented to illustrate the calculation of hand exposure for a specific scenario. The dART model is not yet implemented in the ART software platform, and a robust validation of the model is necessary to determine its predictive ability. With advancing knowledge on dermal exposure and its determinants, this model will require periodic updates and refinements, in addition to further expansion of the applicability domain of the model.
'Stoffenmanager', a web-based control banding tool using an exposure process model

Abstract:In the scope of a Dutch programme to reinforce the working conditions policy on hazardous substances, an internet-based tool was developed to help small- and medium-sized companies to handle hazardous substances with more care. The heart of this tool, called the Stoffenmanager, is a risk banding scheme. It combines a hazard banding scheme similar to that of COSHH Essentials and an exposure banding scheme based on an exposure model originally presented by Cherrie et al. (1996) and further developed by Cherrie and Schneider (1999). The exposure model has been modified to allow non-expert users to understand and use the model. Exposure scores are calculated based on categorization of determinants of emission, transmission and immission. These exposure scores are assigned to exposure bands. The comparison of exposure bands and hazard bands leads to a risk band or priority band. Following the evaluation of the priority of tasks done with products, generic exposure control measures can be evaluated for their possibility to lower the risks. Relevant control measures can be put into an action plan and into workplace instruction cards. The tool has several other functionalities regarding registration and storage of products. The exposure model in the Stoffenmanager leads to exposure scores. These have been compared with measured exposure levels. The exposure scores correlated well with measured exposure levels. The development of the Stoffenmanager has facilitated a whole range of further developments of useful tools for small- and medium-sized enterprises.

Abstract:Exposure assessment is a crucial component of environmental health research, providing essential information on the potential risks associated with various chemicals. A systematic scoping review was conducted to acquire an overview of accessible human exposure assessment methods and computational tools to support and ultimately improve risk assessment. The systematic scoping review was performed in Sysrev, a web platform that introduces machine learning techniques into the review process aiming for increased accuracy and efficiency. Included publications were restricted to a publication date after the year 2000, where exposure methods were properly described. Exposure assessments methods were found to be used for a broad range of environmental chemicals including pesticides, metals, persistent chemicals, volatile organic compounds, and other chemical classes. Our results show that after the year 2000, for all the types of exposure routes, probabilistic analysis, and computational methods to calculate human exposure have increased. Sixty-three mathematical models and toolboxes were identified that have been developed in Europe, North America, and globally. However, only twelve occur frequently and their usefulness were associated with exposure route, chemical classes and input parameters used to estimate exposure. The outcome of the combined associations can function as a basis and/or guide for decision making for the selection of most appropriate method and tool to be used for environmental chemical human exposure assessments in Ontology-driven and artificial intelligence-based repeated dose toxicity testing of chemicals for next generation risk assessment (ONTOX) project and elsewhere. Finally, the choice of input parameters used in each mathematical model and toolbox shown by our analysis can contribute to the harmonization process of the exposure models and tools increasing the prospect for comparison between studies and consistency in the regulatory process in the future.

Abstract:Abstract Within Occupational Risk Prevention, the assessment of risks due to exposure to biological agents has received little attention compared to risks such as those arising from chemicals. This was the case until the impact of the COVID-19 pandemic highlighted the importance of health effects related to exposure to biological agents in the work environment. However, the tools for hazard inventory, risk assessment, and implementation of control measures for biological agents are scarce. There are many reasons that make it challenging for OHS professionals to assess the exposure to biological agents such as, little knowledge on the biological agents potentially present in the different economic activities, their effects on health, the lack of standardized methods for quantitative sampling or the lack of reference values available for most biological agents. Different qualitative risk assessment tools for biological agents have been developed over the last decades. Some examples are the Simplified Evaluation of the INSST (Spain), the Biogaval-Neo of the INVASSAT (Spain), the Bioaerosol Tool of the IRSST (Canada) or the RIE Method of the NKAL (The Netherlands). These tools differ from each other in terms of their scope and the parameters used to determine risk and were compared in order to better understand their limitations and applicability. After the comparison was carried out some additional specific parameters have been proposed as essential for the development of this new model. The proposed improvements could be implemented in the development of a new qualitative biological risk assessment model in the Stoffenmanager® tool.

Exposure assessment during paint spraying and drying using PTR-ToF-MS

Abstract:Spraying is a common way to distribute occupational products, but it puts worker's health at risk by exposing them to potentially harmful particles and gases. The objective of this study is to use time-resolved measurements to gain an understanding of spray applications at the process level and to compare them to predictions of exposure models. We used proton transfer reaction time-of-flight mass spectrometry (PTR-ToF-MS) at 1-s time resolution to monitor the gas phase concentration of the solvents acetone, ethanol, butyl acetate, xylene and 1-methoxy-2-propy acetate during outdoor spraying and indoor drying of metal plate under various conditions of outdoor air supply. We found that during spraying, gas-phase exposure was dominated by the more volatile solvents acetone and ethanol, which exhibited strong concentration variations due to the outdoor winds. During drying, exposure strongly depended on the strength of ventilation. Under conditions with high supply of outdoor air, our measurements show a near-exponential decay of the solvent concentrations during drying. Conversely, under conditions without outdoor air supply, the drying process required hours, during which the less volatile solvents passed through a concentration maximum in the gas phase, so that the exposure during drying exceeded the exposure during spraying. The concentrations measured during spraying were then compared for each of the substances individually with the predictions of the exposure models ECETOC TRA, Stoffenmanager, and ART using TREXMO. For these conditions, ECETOC TRA and Stoffenmanager predicted exposures in the measured concentration range, albeit not conservative for all solvents and each application. In contrast, ART largely overestimated the exposure for the more volatile solvents acetone and ethanol and slightly underestimated exposure to 1M2PA for one spraying. ECETOC TRA and ART do not have options to predict exposure during drying. Stoffenmanager has the option to predict drying together with spraying, but not to predict the drying phase independently. Our study demonstrates the importance of considering both the spray cloud and solvent evaporation during the drying process. To improve workplace safety, there is a critical need for enhanced exposure models and comprehensive datasets for calibration and validation covering a broader range of exposure situations.

Abstract:Various qualitative exposure assessment models based on different underlying assumptions requiring distinct inputs and providing diverse outputs are used by occupational hygienists and risk managers to evaluate the magnitude of occupational exposures. Although a wide variety of exposure assessment models are available, most models have not been validated. This study compared the inhalation risk factor of a qualitative exposure assessment model with quantitative exposure data collected at a manufacturing facility for more than 9 years for 12 worker groups involving 24 chemical agents. A Spearman's rho correlation found no significant correlation between the model's risk factor and the maximum measured exposure (rs=0.119, p=0.496). A Fisher exact test found that the maximum measured exposure was independent of the model's inhalation risk factor (chi2=0.203, p=0.653). The model accurately classified measured exposures in 18 out of 35 cases (51%), 53% of the measured exposures classified as acceptable were correctly classified by the model (sensitivity), whereas 33% of the measured exposures classified as uncertain/unacceptable were correctly classified by the model (specificity). There was a 67% probability that the model would result in a false low classification and a 47% probability that the model would result in a false high classification. Although the model was simple to apply, the overall predictive ability for the low level exposures seen in the study were poor and inconsistent among worker groups compared.

Abstract:A critical review of the current state of knowledge of chemical emissions from indoor sources, partitioning among indoor compartments, and the ensuing indoor exposure leads to a proposal for a modular mechanistic framework for predicting human exposure to semivolatile organic compounds (SVOCs). Mechanistically consistent source emission categories include solid, soft, frequent contact, applied, sprayed, and high temperature sources. Environmental compartments are the gas phase, airborne particles, settled dust, indoor surfaces, and clothing. Identified research needs are the development of dynamic emission models for several of the source emission categories and of estimation strategies for critical model parameters. The modular structure of the framework facilitates subsequent inclusion of new knowledge, other chemical classes of indoor pollutants, and additional mechanistic processes relevant to human exposure indoors. The framework may serve as the foundation for developing an open-source community model to better support collaborative research and improve access for application by stakeholders. Combining exposure estimates derived using this framework with toxicity data for different end points and toxicokinetic mechanisms will accelerate chemical risk prioritization, advance effective chemical management decisions, and protect public health.

Abstract:Objective: The differences in the methodologies of various occupational health risk assessment (OHRA) models have not been extensively reported. We aimed to understand the qualitative and quantitative differences between common OHRA models in typical industries. Methods : The Environmental Protection Agency (EPA), Australian, Romanian, Singaporean, International Council on Mining and Metals (ICMM), and the Control of Substances Hazardous to Health (COSHH) models were evaluated, and a theoretical framework was established for a comparative study. Results: Qualitative comparisons showed that each OHRA model had its own strengths and limitations, and exhibited a diverse distribution at different levels for each evaluation indicator. The Singaporean, COSHH, and EPA models had a much higher comprehensive advantage than the other models for all indicators. Quantitative comparisons demonstrated that these three models also had a stronger ability to distinguish the difference in risk ratios between different industries. The Singaporean model had the strongest correlation with the other models. Conclusion : Each model possessed its own strengths and limitations depending on its unique methodological principles. Combining the EPA, Singaporean, and COSHH models might be advantageous for developing an OHRA strategy. More studies comparing multiple models in key industries are required. (C) Article author(s).

Abstract:Errors in air pollution exposure assessment are often considered as a major limitation in epidemiological studies. However, it is difficult to obtain accurate personal level exposure on cohort populations due to the often prohibitive expense. Personal exposure estimation models are used in lieu of direct personal exposure measures but still suffer from issues of availability and accuracy. We aim to establish a personal PM2.5 exposure assessment model for a cohort population and assess its performance by applying our model on cohort subjects. We analyzed data from representative sites selected from the subclinical outcomes of polluted air in China (SCOPA-China) cohort study and established a random forest model for estimating daily PM2.5 personal exposure. We also applied the model among subjects recruited in the project mentioned above within the same area and study period to estimate the reliability of the model. The established model showed a good fit with an R2 of 0.81. The model application results showed similar patterns with empirically measured data. Our pilot study provided a validated and feasible modeling approach for assessing daily personal PM2.5 exposure for large cohort populations. Abstract:In the majority of occupational settings within China, the concentrations of benzene are observed to fall markedly below the demarcated detection thresholds. Employing traditional risk assessment models, the presence of exceptionally low airborne benzene exposure concentrations may infuse heightened degrees of uncertainty. Consequently, the necessity arises to investigate risk assessment methodologies more apt for the prevalent exposure environment among employees. In the present study, a pharmacokinetic model premised on urinary benzene metabolites (S-PMA and t, t-MA) was employed to ascertain a more precise daily airborne benzene exposure concentration per individual. This value was integrated into the linear multistage model as the 'internal exposure concentration'. In conjunction with the U.S National Environmental Protection Agency's (EPA) inhalation risk assessment model predicated on the external exposure concentration, the Singapore Ministry of Manpower's (MOM) model, and the linear multistage (LMS) model, the carcinogenic and non-carcinogenic effects of benzene were evaluated for 1781 benzene-exposed employees across 76 enterprises in Jiangsu Province. Findings suggest that in the linear multilevel model assessment, the cancer risk levels based on t, t-MA and S-PMA were higher in the printing and recording media reproduction industry, automobile manufacturing industry, general equipment manufacturing industry and the furniture manufacturing industry (median 2.842 × 10 −4 , 2.819 × 10 −4 , 2.809 × 10 −4 , and 2.678 × 10 −4 ), which align more consistently with the actual benzene exposure circumstances of each industry's study participants, with overall risk levels calculated by the linear multistage model exceeding those of the EPA inhalation risk assessment model and the MOM model. This implies that the linear multistage model of internal exposure, based on the reciprocal of benzene biomarkers S-PMA and t, t-MA for airborne benzene exposure, presents enhanced sensitivity and suitability for the current occupational health risk assessment of workers. Without doubt, biomarker-based benzene exposure risk assessment emerges as the optimal choice.

Abstract:The field of exposure assessment has matured significantly over the past 10-15 years. Dozens of studies have measured the concentrations of numerous chemicals in many media to which humans are exposed. Others have catalogued the various exposure pathways and identified typical values which can be used in the exposure calculations for the general population such as amount of water or soil ingested per day or the percent of a chemical than can pass through the skin. In addition, studies of the duration of exposure for many tasks (e.g. showering, jogging, working in the office) have been conducted which allow for more general descriptions of the likely range of exposures. All of this information, as well as the development of new and better models (e.g. air dispersion or groundwater models), allow for better estimates of exposure. In addition to identifying better exposure factors, and better mathematical models for predicting the aerial distribution of chemicals, the conduct of simulation studies and dose-reconstruction studies can offer extraordinary opportunities for filling in data gaps regarding historical exposures which are critical to improving the power of epidemiology studies. The use of probabilistic techniques such as Monte Carlo analysis and Bayesian statistics have revolutionized the practice of exposure assessment and has greatly enhanced the quality of the risk characterization. Lastly, the field of epidemiology is about to undergo a sea change with respect to the exposure component because each year better environmental and exposure models, statistical techniques and new biological monitoring techniques are being introduced. This paper reviews these techniques and discusses where additional research is likely to pay a significant dividend. Exposure assessment techniques are now available which can significantly improve the quality of epidemiology and health risk assessment studies and vastly improve their usefulness. As more quantitative exposure components can now be incorporated into these studies, they can be better used to identify safe levels of exposure using customary risk assessment methodologies. Examples are drawn from both environmental and occupational studies illustrating how these techniques have been used to better understand exposure to specific chemicals. Some thoughts are also presented on what lessons have been learned about conducting exposure assessment for health risk assessments and epidemiological studies.

Abstract:The paper contains the main lines of the modeling of a prototype for both medical and economical evaluation of radiotherapy centers using innovative therapies. We consider uncertainties according to patients and hospital complexes participating in the study. The corresponding part of the tool is called recruitment model. It evaluates a theoretical number of patients interested in treatments of one center, using spatialization into a context of competition. Moreover we developed a scheduling model, described briefly here and two shortly presented economical models in charge of the evaluation of the average price per treatment and the comparison between different radiotherapy strategies. We present preliminary results of the recruitment model.

Abstract:Experimentation demonstrates long-range surface plasmon polariton waveguides as a useful biosensor to selectively detect gram negative or gram positive bacteria in human urine having a low concentration of constituents. The biosensor can detect bacteria at concentrations of 10(5) CFU/ml, the internationally recommended threshold for diagnostic of urinary tract infection. Using a negative control urine solution of bacterial concentration 1000☓ higher than the targeted bacteria, we obtain a ratio of 5.4 for the positive to negative signals.

Abstract:Over the past several decades, reducing, refining, and replacing animal testing (three R’s) has been a prominent goal in chemical toxicology.1 The STopTox (Systemic and Topical chemical Toxicity) platform was developed for this objective as an innovative in-silico alternative to conventional animal testing for acute systemic and topical toxicity testing.2 STopTox utilizes quantitative structure-activity relationship (QSAR) models to predict the toxicity of chemicals, providing a comprehensive, accessible, and user-friendly tool for hazard identification.2 STopTox models were rigorously validated during its initial development using extensive publicly available data sets, ensuring compliance with the Organisation for Economic Co-operation and Development (OECD) principles. These models boasted high internal accuracy and substantial external predictive power.2,3 Despite these promising results, continued validation with novel compounds is integral in establishing the robustness and reliability needed for STopTox to be used as a substitute for in vivo animal testing. In this research letter, we aim to evaluate the predictive performance of STopTox using independent data sets across the six major endpoints of acute toxicity: acute oral, dermal, and inhalation systemic toxicity, as well as skin sensitization, skin irritation/corrosion, and eye irritation/corrosion through external validation. The outcomes of this validation underscore the potential of STopTox to reliably predict toxicity, thereby supporting STopTox as a reliable regulatory decision-making tool that contributes to reducing animal testing in toxicological assessments.

Evaluating the alignment of alternative assessment methods with REACH Article 57

Abstract:The assessment of chemical alternatives for hazardous substances is an important prerequisite for avoiding regrettable substitution, and several methods have been developed in the past to perform such an Assessment-of-Alternatives. We investigate whether GreenScreen®, Cradle to Cradle®, Multi Criteria Decision Analysis (MCDA), Cost Benefit Analysis, the Pollution Prevention Options Analysis System, the U.S.EPA Safer Choice Standard and Criteria, and the GHS column model 2020 from IFA use similar criteria for the evaluation of substances as Article 57 of REACH and how suitable these methods are for assessing per- and polyfluoroalkyl substances. MCDA and GreenScreen® were identified as the most promising methods and were analyzed in detail using two different datasets. The results of the assessments show that none of the investigated alternative assessment methods use the same hazard assessment criteria as described in Article 57 of REACH. It was also not possible to parameterize MCDA (using the multi-attribute utility theory approach) without redundant variables so that the results align with an assessment according to Article 57 of REACH. There is therefore an urgent need for a modified/new method that can be used in the future to assess organic substances that are used within the European Economic Area.

Abstract:Over the past few years there have been several advances in distributed systems, and more recently multi-core processors. Consequently, a natural need for concurrent and parallel programming languages arises. In this paper, we compare some aspects of two concurrency models, Actors and Agents, using benchmarks to evaluate: (i) the communication performance on a concurrency-heavy scenario; (ii) the performance in a scenario with the presence of bottleneck and synchronization problems; and (iii) the reactivity and fairness of the models. We chose Jason, 2APL, and GOAL as the agent-oriented programming languages and Erlang, Akka, and ActorFoundry as the actor-oriented programming languages. Overall, Erlang displayed the best performance of all languages used in this comparison, followed by ActorFoundry, Akka, Jason, 2APL, and GOAL, in this particular order.
