Abstract:Electrowetting on dielectric-based digital microfluidic platforms (EWOD-DMF) have a potential to impact point-of-care diagnostics. Conventionally, EWOD-DMF platforms are manufactured in cleanrooms by expert technicians using costly and time consuming micro-nanofabrication processes such as optical lithography, depositions and etching. However, such high-end microfabrication facilities are extremely challenging to establish in resource-poor and low-income countries, due to their high capital investment and operating costs. This makes the fabrication of EWOD-DMF platforms extremely challenging in low-income countries, where such platforms are most needed for many applications such as point-of-care testing applications. To address this challenge, we present a low-cost and simple fabrication procedure for EWOD-DMF electrode arrays, which can be performed anywhere with a commercial office inkjet printer without the need of expensive cleanroom facilities. We demonstrate the utility of our platform to move and mix droplets of different reagents and physiologically conductive buffers, thereby showing its capability to potentially perform a variety of biochemical assays. By combining our low-cost, inkjet-printed EWOD-DMF platform with smartphone imaging technology and a compact control system for droplet manipulation, we also demonstrate a portable and hand-held device which can be programmed to potentially perform a variety of biochemical assays.
Ultrathin-layer chromatography on SiO(2), Al(2)O(3), TiO(2), and ZrO(2) nanostructured thin films

Abstract:We explored four different inorganic oxides and determined their merits in miniaturized planar chromatography. Despite progression of chromatographic techniques over several decades, such alternatives to traditional planar silica gel stationary phases have not been fully evaluated. Glancing angle deposition(GLAD) provided an excellent platform for engineering nanostructured thin films in these materials for ultrathin-layer chromatography (UTLC). Separations of carotenoids and synthetic food dyes were used to investigate the attributes of SiO(2), Al(2)O(3), TiO(2), and ZrO(2)GLAD UTLC media. These anisotropic high surface area thin films possessed similar channel-like features but different chromatographic properties.TiO(2)and ZrO(2)media were especially interesting since analyte retention could be modified through sim-ple oxidation heat treatments and UV irradiation. Generally, oxidation reduced analyte retention while UV exposure increased retention. Changes in retention factor as large as ΔhRF∼ 40 (for Acid Red 14 on titanium oxide) were achieved. Food dye mixtures were applied using consumer inkjet printers as per the Office Chromatography concept and separation performance was quantified using advanced video instrumentation designed for miniaturized plates. Enhanced time-resolved UTLC methods were used to calculate figures of merit from recorded dye separation videos. Small theoretical plate heights (<4 μm)and low limits of detection (<2 ng per zone for the food dye tartrazine) were measured. The combination of engineered GLAD UTLC plates, inkjet application of analyte spots, time-resolved UTLC, and custom analysis algorithms enabled some of the best performance achieved on GLAD UTLC layers. Separations on the inorganic oxide thin films were also successfully hyphenated with electrospray ionization mass spectrometry for the first time. This investigation demonstrates the utility of alternative inorganic oxide GLADUTLC media and probes avenues of expanding the capabilities of miniaturized planar chromatography.

Abstract:<p>This investigation was carried out with the aim of stressing that technologically-advanced systems can be prototyped and produced by using 3D printing and inexpensive electronic modules. In particular, open-source technologies are opening new exciting possibilities also in the area of chemical analysis, thus making possible the assembly of customized analytical devices by using widely available and low cost materials. Here, two different field-portable optical analytical instruments, one operating in the transmittance mode and the other in reflectance mode, were built up by combining 3D printed parts with common hardware components, such as Arduino boards, LEDs and photoresistors. They were employed to analyze the antioxidant capacity of several tea infusions by profiting from the Folin-Ciocalteu assay. A performance comparable with that displayed by research-grade spectrophotometers indicates that these easy-to-operate and low-cost devices can provide accurate and precise results, thus demonstrating that highly-sensitive optical instrumentations, useful not only in research and education environments but also in resource-limited settings, can be readily constructed by a DIY (Do-It-Yourself) approach.</p>

Abstract:Electronic system control of analytical instrumentation remains a critical aspect of modern measurement science. Within the field of liquid chromatography (LC), this is especially relevant for automation, module operation, detection, data acquisition, and data analysis. Increasingly, home‐built analytical tools used for liquid‐phase separations rely upon open‐source microcontrollers and single‐board computers to aid in simplifying these operations. In this review, we detail literature reported within the past 5 years in which these types of devices were used to advance various aspects of the LC research field, including sample preparation, instrument control, and data collection.

OpenCell: A Low-cost, Open-Source, 3-in-1 device for DNA Extraction

Abstract:High-cost DNA extraction procedures pose significant challenges for budget-constrained laboratories. To address this, we introduce OpenCell, an economical, open-source, 3-in-1 laboratory device that combines the functionalities of a bead homogenizer, a microcentrifuge, and a vortex mixer. OpenCell utilizes modular attachments that magnetically connect to a central rotating brushless motor. This motor couples to an epicyclic gearing mechanism, enabling efficient bead homogenization, vortex mixing, and centrifugation within one compact unit. OpenCell’s design incorporates multiple redundant safety features, ensuring both the device’s and operator’s safety. Additional features such as RPM measurement, programmable timers, battery operation, and optional speed control make OpenCell a reliable and reproducible laboratory instrument. In our study, OpenCell successfully isolated DNA from (spinach), with an average yield of 2.3 µg and an A260/A280 ratio of 1.77, demonstrating its effectiveness for downstream applications such as Polymerase Chain Reaction (PCR) amplification. With its compact size (20 cm x 28 cm x 6.7 cm) and lightweight design (0.8 kg), comparable to the size and weight of a laptop, OpenCell is portable, making it an attractive component of a ‘lab-in-a-backpack’ for resource-constrained environments in low-and-middle-income countries and synthetic biology in remote field stations. Leveraging the accessibility of 3D printing and off-the-shelf components, OpenCell can be manufactured and assembled at a low unit cost of less than $50, providing an affordable alternative to expensive laboratory equipment costing over $4000. OpenCell aims to overcome the barriers to entry in synthetic biology research and contribute to the growing collection of frugal and open hardware.

Abstract:Lab-On-Chips (LOCs) offer several advantages over traditional approaches for studying biological models and evaluating drug efficacy. Despite their small size, they present highly complex environments where significant chemical gradients are often confined to specific regions, either through intentional manipulation by researchers or generated by the cellular model itself. To address these challenges and gain deeper insights into biological mechanisms, integrating biosensors in the LOC becomes an appealing solution. In this research, we present a cost-effective, high-resolution stereolithographic technique to create small, widespread optical biosensors directly integrated into the lab-on-chip. To extract information from these sensors, we designed a custom multispectral light source, capable of capturing the optical signature of the field of view, coupled with a dedicated machine-learning algorithm. Applying this approach, we successfully measured the pH in Human Embryonic Stem Cell (hESC) 3D cell cultures, a unique cell type that lacks a pH indicator in its medium. By analyzing local temporal pH trends across various experiments and different areas within the same experiments, we advanced our comprehension of cell-scale metabolic mechanisms and their impact on tissue/organ behavior.

Abstract:Automation and digitalization solutions in the field of small molecule synthesis face new challenges for chemical reaction analysis, especially in the field of high-performance liquid chromatography (HPLC). Chromatographic data remains locked in vendors' hardware and software components, limiting their potential in automated workflows and data science applications. In this work, we present an open-source Python project called MOCCA for the analysis of HPLC-DAD (photodiode array detector) raw data. MOCCA provides a comprehensive set of data analysis features, including an automated peak deconvolution routine of known signals, even if overlapped with signals of unexpected impurities or side products. We highlight the broad applicability of MOCCA in four studies: (i) a simulation study to validate MOCCA's data analysis features; (ii) a reaction kinetics study on a Knoevenagel condensation reaction demonstrating MOCCA's peak deconvolution feature; (iii) a closed-loop optimization study for the alkylation of 2-pyridone without human control during data analysis; (iv) a well plate screening of categorical reaction parameters for a novel palladium-catalyzed cyanation of aryl halides employing <i>O</i>-protected cyanohydrins. By publishing MOCCA as a Python package with this work, we envision an open-source community project for chromatographic data analysis with the potential of further advancing its scope and capabilities.

Abstract:This paper explores an application of 3D printing technology on the food industry. Since its inception in the 1980s, 3D printing has experienced a huge rise in popularity. This study uses cost-effective, flexible, and sustainable components that enable specific features of certain gas chromatography. This study aims to optimize the process of gas detection using a 3D printed separation column and the MiCS-6814 sensor. The principle of the entire device is based on the idea of utilizing a simple capillary chromatographic column manufactured by 3D printing for the separation of samples into components prior to their measurement using inexpensive chemiresistive sensors. An optimization of a system with a 3D printed PLA block containing a capillary, a mixing chamber, and a measuring chamber with a MiCS-6814 sensor was performed. The optimization distributed the sensor output signal in the time domain so that it was possible to distinguish the peak for the two most common alcohols, ethanol and methanol. The paper further describes some optimization types and their possibilities.

Abstract:In both batch and continuous-flow reactor technology, reproducibility can be challenging for photochemical processes due to setup variability. One major contributor to this issue is the lack of standardized reactor solutions, particularly in academic laboratories where cost is often a prohibitive factor to purchase commercially-available reactor technology. However, advancements in 3D printing technologies and the availability of high-intensity light sources present an opportunity to develop cost-effective laboratory equipment. In this work, we present a diverse set of open-source reactor designs aimed at democratizing photochemistry while reducing the barrier of expensive technology. We introduce three new reactor designs: the UFO reactor for batch reactions, the Uflow reactor for seamless transition to flow processes, and the Fidget reactor for scale-up. After detailing the design principles and rationale behind these configurations, we characterize and evaluate their performance through simulations and experiments. These designs offer a standardized and affordable point of entry for researchers interested in exploring batch and flow photochemistry.

Abstract:The development of a portable device created by 3D printing for colorimetric and fluorometric measurements is an efficient tool for analytical applications in situ or in the laboratory presenting a wide field of applications in the environmental and food field. This device uses a light-emitting diode (LED) as radiation source and a webcam as a detector. Digital images obtained by the interaction between the radiation source and the sample were analyzed using a programming language developed in Matlab (Mathworks Inc., Natick, MA, USA), which builds the calibration curves in real-time using the RGB colour model. In addition, the entire system is connected to a notebook which serves as an LED and detector power supply without the need for any additional power source. The proposed device was used for the determination in situ of norfloxacin, allura red, and quinine in water and beverages samples, respectively. For the validation of the developed system, the results obtained were compared with a conventional spectrophotometer and spectrofluorometer respectively with a t-test at a 95% confidence level, which provides satisfactory precision and accuracy values.

Abstract:Additive manufacturing (3D printing) is a disruptive technology that is changing production systems globally. In addition, microfluidic devices are increasingly being used for chemical analysis and continuous production of chemicals. Printing of materials such as polymers and metals is already a reality, but additive manufacturing of glass for microfluidic systems has received minor attention. We characterize microfluidic devices (channel cross-section dimensions down to a scale of 100 μm) that have been produced by additive manufacturing of molten soda-lime glass in tens of minutes and report their mass spectrometric and Raman spectroscopic analysis examples. The functionality of a microfluidic glass microreactor is shown with online mass spectrometric analysis of linezolid synthesis. Additionally, the performance of a direct infusion device is demonstrated by mass spectrometric analysis of drugs. Finally, the excellent optical quality of the glass structures is demonstrated with in-line Raman spectroscopic measurements. Our results promise a bright future for additively manufactured glass microdevices in diverse fields of science.

Abstract:3D-printed laboratory devices can enable ambitious research purposes even at a low-budget level. To follow this trend, here we describe the construction, calibration, and usage of the FINDUS (Fully Integrable Noncommercial Dispensing Utility System). We report the successful 3D printing and assembly of a liquid-handling workstation for less than $400. Using this setup, we achieve reliable and flexible liquid-dispensing automation with relative pipetting errors of less than 0.3%. We show our system is well suited for several showcase applications from both the biology and chemistry fields. In support of the open-source spirit, we make all 3D models, assembly instructions, and source code available for free download, rebuild, and modification.

Abstract:The possibility to print electronics by means of office tools has remarkedly increased the possibility to design affordable and robust point-of-care/need devices. However, conductive inks suffer from low electrochemical and rheological performances limiting their applicability in biosensors. Herein, a fast CO2 laser approach to activate printed carbon inks towards direct enzymatic bioelectrocatalysis (3rd generation) is proposed and exploited to build biosensors for D-fructose analysis in biological fluids. The CO2 laser treatment was compared with two lab-grade printed transducers fabricated with solvent (SB) and water (WB) based carbon inks. The use of the laser revealed significant morpho-chemical variations on the printed inks and was investigated towards enzymatic direct catalysis, using Fructose dehydrogenase (FDH) integrated into entirely lab-produced biosensors. The laser-driven activation of the inks unveils the inks' direct electron transfer (DET) ability between FDH and the electrode surface. Sub-micromolar limits of detection (SB-ink LOD = 0.47 μM; WB-ink LOD = 0.24 μM) and good linear ranges (SB-ink: 5-100 μM; WB-ink: 1-50 μM) were obtained, together with high selectivity due to use of the enzyme and the low applied overpotential (0.15 V vs. pseudo-Ag/AgCl). The laser-activated biosensors were successfully used for D-fructose determination in complex synthetic and real biological fluids (recoveries: 93-112%; RSD ≤8.0%, n = 3); in addition, the biosensor ability for continuous measurement (1.5h) was also demonstrated simulating physiological D-fructose fluctuations in cerebrospinal fluid.

Abstract:The widespread availability and diversity of open-source microcontrollers paired with off-the-shelf electronics and 3D printed technology has led to the creation of a wide range of low-cost scientific instruments, including microscopes, spectrometers, sensors, data loggers, and other tools that can be used for research, education, and experimentation. These devices can be used to explore a wide range of scientific topics, from biology and chemistry to physics and engineering. In this study, we designed and built a multifunction fluorescent open source in vivo/in vitro imaging system (openIVIS) system that integrates a Raspberry Pi with commercial cameras and LEDs with 3D printed structures combined with an acrylic housing. Our openIVIS provides three excitation wavelengths of 460 nm, 520 nm, and 630 nm integrated with Python control software to enable fluorescent measurements across the full visible light spectrum. To demonstrate the potential applications of our system, we tested its performance against a diverse set of experiments including laboratory assays (measuring fluorescent dyes, using optical nanosensors, and DNA gel electrophoresis) to potentially fieldable applications (plant and mineral imaging). We also tested the potential use for a high school biology environment by imaging small animals and tracking their development over the course of ten days. Our system demonstrated its ability to measure a wide dynamic range fluorescent response from millimolar to picomolar concentrations in the same sample while measuring responses across visible wavelengths. These results demonstrate the power and flexibility of open-source hardware and software and how it can be integrated with customizable manufacturing to create low-cost scientific instruments with a wide range of applications. Our study provides a promising model for the development of low-cost instruments that can be used in both research and education.

Abstract:The miniaturization of gas chromatography (GC) systems has made it possible to utilize the analytical technique in various on-site applications to rapidly analyze complex gas samples. Various types of miniaturized sensors have been developed for micro-gas chromatography (µGC). However, the integration of an appropriate detector in µGC systems still faces a significant challenge. We present a solution to the problem through integration of µGC with photonic crystal slab (PCS) sensors using transfer printing technology. This integration offers an opportunity to utilize the advantages of optical sensors, such as high sensitivity and rapid response time, and at the same time, compensate for the lack of detection specificity from which label-free optical sensors suffer. We transfer printed a 2D defect free PCS on a borofloat glass, bonded it to a silicon microfluidic gas cell or directly to a microfabricated GC column, and then coated it with a gas responsive polymer. Realtime spectral shift in Fano resonance of the PCS sensor was used to quantitatively detect analytes over a mass range of three orders. The integrated µGC-PCS system was used to demonstrate separation and detection of a complex mixture of 10 chemicals. Fast separation and detection (4 min) and a low detection limit (ng) was demonstrated.
Development of an automated platform for monitoring microfluidic reactors through multi-reactor integration and online (chip-)LC/MS-detection

Abstract:This work presents a novel microfluidic screening setup with real-time analytics for investigating reactions with immobilised biocatalysts. The setup combines microreactor technology, multi-reactor integration, and online (chip-)LC/MS analysis in a sequential automated workflow. We utilized in-house manufactured fused-silica glass chips as reusable packed-bed microreactors interconnected as individual tube reactors. The potential of this setup was showcased by conducting and optimising a biocatalytic aromatic bromination reaction as the first proof of concept using immobilised vanadium-dependent haloperoxidase from Curvularia inaequalis (CiVHPO). The fusion of a HaloTagTM to CiVHPO was used for efficient and mild covalent linkage of the enzyme onto chloroalkane-functionalized particles. Then, the biotransformation was continuously monitored with automated LC/MS data acquisition in a data-rich manner. By further developing the automation principle, it was possible to sequentially screen multiple different connected packed-bed microreactors for reaction optimization while using only miniature amounts of reactants and biocatalyst. Finally, we present a fast and modular chipHPLC solution for online analysis to reduce the overall solvent consumption by over 80%. We established a modern microfluidic platform for real-time reaction monitoring and evaluation of biocatalytic reactions through automation of the reactant feed integration, flexible microreactor selection, and online LC/MS analysis.

Abstract:Recently, the unique capability of three-dimensional (3D) printing strategy for patterning micro-sized features with complex 3D architectures in a single process has caught an ever increasing interest within the domain of lab-on-a-chip devices. In this review, the latest advances in the constucting development of 3D microfluidic devices with 3D printing technologies in analytical application were summed up. To begin with, the attractive properties of 3D printing technologies were described. Then, the principles of three mainly printing technologies were introduced in detail. The stimulating progress related to applications of 3D printed lab-on-a-chip devices was subsequently dealt with. Finally, the challenges and opportunities in regard to numerous aspects of 3D printing microfluidic system were discussed. It is believed that higher resolution, more user-frinendly operation endows 3D printed lab-on-a-chip devices with great potential academic and commercial value in on-site environmental monitoring, food safety, and other point-of-care assay. Laboratory Ozonolysis Using an Integrated Batch-DIY Flow System for Renewable Material Production

Abstract:Flow chemistry offers a solution for replacing batch methods in chemical preparation where intermediates or products may pose toxicity or instability hazards. Ozonolysis offers an ideal opportunity for flow chemistry solutions, but multiple barriers to entry exist for use of these methods, including equipment cost and performance optimization. To address these challenges, we developed a programmable DIY syringe pump system to use for a continuous flow multireactor process using 3D-printed parts, off-the-shelf stepper motors, and an Arduino microcontroller. Reaction kinetics of ozonide formation informed the use of an integrated batch-flow approach, where ozone addition to an olefin was timed to coincide with fluid movement of a single-syringe pump, followed by downstream Pinnick oxidation and reductive quench in flow. The system was demonstrated by continuous preparation of azelaic acid from ozonolysis of palmitoleic acid, a process limited to low production volumes via batch chemistry. High total production of azelaic acid with 80% yield was obtained from an algae oil sourced unsaturated fatty acid: a product with important applications in medicine, cosmetics, and polymers. This low-cost, scalable approach offers the potential for rapid prototyping and distributed chemical production.
