Abstract:A quick, easy, cheap, effective, rugged, and safe method was developed for the multi-residue analysis of pesticides and antibiotics in aquaculture sediment using high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The developed method is based on ultrasonic extraction with acetonitrile and phosphate buffer, salting with sodium chloride, and cleaning with dispersive solid-phase extraction adsorbent using primary secondary amine, C18, and graphitized carbon black, followed by HPLC-MS/MS detection. We optimized different extraction methods and the ratio of the cleanup adsorbents to achieve good recoveries at three spiking levels that ranged from 60.4% to 114% with a relative standard deviation below 15% (n = 6). For all analytes, except for flufenoxuron, the limits of quantification were between 1 and 5 µg/kg (dry weight). The validated method was successfully applied to real samples collected from 20 aquacultural ponds, confirming the feasibility of the proposed method. The concentrations of the target analytes in the sediments (dry weight) were in the ranges of 2.2-35.0 µg/kg for sulfonamides, 0-409.1 µg/kg for quinolones, 0-6.56 µg/kg for macrolides, and 0-4.9 µg/kg for pesticides. Moreover, the co-occurrence of pesticides and antibiotics may potentially pose a high risk to sediment-dwelling organisms in nine out of the 20 investigated locations.
[Determination of 13 perfluorinated and polyfluoroalkyl substances in fishes by QuEChERS-ultra-high performance liquid chromatography-tandem mass spectrometry]

Abstract:Perfluorinated and polyfluoroalkyl substances (PFASs) are compounds characterized by at least one perfluorinated carbon atom in an alkyl chain linked to side-chain groups. Owing to their unique chemical properties, these compounds are widely used in industrial production and daily life. However, owing to anthropogenic activities, sewage discharge, surface runoff, and atmospheric deposition, PFASs have gradually infiltrated the environment and aquatic resources. With their gradual accumulation in environmental waters, PFASs have been detected in fishes and several fish-feeding species, suggesting that they are bioconcentrated and even amplified in aquatic organisms. PFASs exhibit high intestinal absorption efficiencies, and they bioaccumulate at higher trophic levels in the food chain. They can be bioconcentrated in the human body via food (e. g., fish) and thus threaten human health. Therefore, establishing an efficient analytical technique for use in analyzing PFASs in typical fish samples and providing technical support for the safety regulation and risk assessment of fish products is necessary. In this study, by combining solvent extraction and magnetic dispersion-solid phase extraction (d-SPE), an improved QuEChERS method with ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) was developed for the determination of 13 PFASs in fish samples. Fe3O4-TiO2 can be used as an ideal adsorbent in the removal of sample matrix interference and a separation medium for the rapid encapsulation of other solids to be isolated from the solution. Based on the matrix characteristics of the fish products and structural properties of the target PFASs, Fe3O4-TiO2 and N-propyl ethylenediamine (PSA) were employed as adsorbents in dispersive purification. The internal standard method was used in the quantitative analyses of the PFASs. To optimize the sample pretreatment conditions of analyzing PFASs, the selection of the extraction solvent and amounts of Fe3O4-TiO2 and PSA were optimized. Several PFASs contain acidic groups that are non-dissociated in acidic environments, thus favoring their entry into the organic phase. In addition, acidified acetonitrile can denature and precipitate the proteins within the sample matrix, facilitating their removal. Finally, 2% formic acid acetonitrile was used as the extraction solvent, and 20 mg Fe3O4-TiO2, 20 mg PSA and 120 mg anhydrous MgSO4 were used as purification adsorbents. Under the optimized conditions, the developed method exhibited an excellent linearity (R≥0.9973) in the range of 0.01-50 μg/L, and the limits of detection (LODs) and quantification (LOQs) ranged from 0.001-0.023 and 0.003-0.078 μg/L, respectively. The recoveries of the 13 PFASs at low, medium, and high spiked levels (0.5, 10, and 100 μg/kg) were 78.1%-118%, with the intra- and inter-day precisions of 0.2%-11.1% and 0.8%-8.7%, respectively. This method was applied in analyzing real samples, and PFASs including perfluorooctanesulfonic acid, perfluorooctanoic acid, perfluoroundecanoic acid, perfluorododecanoic acid, and perfluorotridecanoic acid, were detected in all 11 samples evaluated. This method is simple, sensitive, and suitable for use in analyzing PFASs in fish samples.

Abstract:Nitroimidazoles (NMZs) are a crucial group of antibacterial compounds from a historical perspective. In the past, they were used for treating and preventing parasitic infections in fish. Benzodiazepines (BZDs) are second-generation sedative-hypnotics. Some fish farmers or vendors use them illegally to keep aquatic products fresh during the transportation of aquatic animals. Aquatic products are one of the most common food sources of protein and can be contaminated by NMZs and BZDs, which could impact humans through the food chain. Until recently, there was limited information on the simultaneous determination of NMZs and BZDs. Thus, it is critical to accurately quantify NMZs and BZDs for risk assessment and risk monitoring of food safety. For the simultaneous determination of five nitroimidazoles and six benzodiazepines in aquatic products, a new approach based on the dispersive solid-phase extraction (dSPE) coupled with ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) was developed. First, the samples were extracted with acetonitrile containing 1% (v/v) ammonium hydroxide, and the extracts were purified using dSPE with C18 and primary secondary amine sorbents. Second, the extracts were collected and dried at 45 ℃ under nitrogen flow. Finally, the extracts were redissolved in 1 mL methanol-water (1∶9, v/v) mixture, filtered through a nylon-66 microfiltration membrane, and analyzed using UHPLC-MS/MS. The separation of compounds was conducted on a Kinetex F5 column (100 mm×3.0 mm, 2.6 μm) using gradient elution with 1% (v/v) formic acid aqueous solution and methanol as the mobile phase. The analytes were detected using MS/MS with positive electrospray ionization (ESI+) source under the multiple reaction monitoring modes. The matrix-matched external standard approach was used for quantitative analysis. The compounds of five nitroimidazoles and six benzodiazepines could be examined within 8.5 min. Under the optimal conditions, the standard curves were linear in the range of 0.5-20 μg/L, with the correlation coefficients exceeding 0.995. The limits of detection and limits of quantification were 0.2-0.5 μg/kg and 0.5-1.0 μg/kg, respectively. The average recoveries at three spiked levels in blank samples (grass carp, large yellow croaker, and prawn) ranged from 73.2% to 110.6%, with relative standard deviations of less than 15%. The developed approach is simple, sensitive, fast, and inexpensive. It can be used for determining five nitroimidazoles and six benzodiazepines in aquatic products.
A coupled method of on-line solid phase extraction with the UHPLC‒MS/MS for detection of sulfonamides antibiotics residues in aquaculture

Abstract:The use of a variety of antibiotics in fish farming raises serious concern about the development of antibiotic resistance. Sulfonamides antibiotics (SAs), which are widely used in aquaculture and generate large eco‒toxicological effects with significant mutagenicity and teratogenic consequences, are still difficult to determine in aquatic organisms. In this study, an automatic technology was developed by coupling on‒line solid phase extraction system (on‒line SPE) with ultra‒high‒performance liquid chromatography spectrometry‒mass spectrometry (UHPLC‒MS/MS). Particularly, using a single on‒line column in the process of sample pretreatment, e.g., HLB or C18, phospholipids that potentially caused the matrix effect cannot be removed form biological sample. We applied a mixed cation exchange column (Oasis® MCX) connected with a hydrophilic lipophilic balance column (Oasis® HLB) in series in on‒line SPE clean‒up to remove interferences and finally obtained a clear and stable eluant. The on‒line SPE working conditions and UHPLC‒MS/MS parameters were optimized for their sensitivity, accuracy, decision limit, and detection capability, which were further calibrated for fish, shrimp and crab. The results showed that the limits of detection and limits of quantification ranged from 1.46 to 15.5 ng/kg, and 4.90–51.6 ng/kg, respectively. Accuracy values covered 71.5%–102% at the three concentration levels (0.1, 0.5, 1.0 μg/kg) for all compounds and average repeatability (relative standard deviation, RSD%) ranged from 3.47% to 14.2%. This on‒line SPE coupled with UHPLC‒MS/MS method is a way forward for an automatic, powerful detection technology for determination of antibiotics from complex matrix.

Abstract:Amphenicol antibiotics including chloramphenicol (CAP), florfenicol (FF) and thiamphenicol (TAP) have been widely used in aquaculture and veterinary to treat various infections. Because of their high toxicity, it is important to detect these amphenicol antibiotics simultaneously. Using the monoclonal antibody against CAP (mAb(CAP)) binding CAP only and the mAb(FF) recognizing both FF and TAP, the two antibodies were covalently coupled to CNBr-activated Sepharose-4B for the simultaneous extraction of CAP, FF and TAP from samples. The bound CAP, FF and TAP were eluted from the Sepharose-4B and measured by LC-MS/MS. The extraction conditions of the two-antibodies-immobilized immunoaffinity clean-up (IAC) for CAP, FF and TAP were optimized and the maximum capacity was investigated. The extraction efficiency of IAC for CAP, FF and TAP was 1.1-2.1 times higher than that of SPE. The LC-MS/MS coupled with IAC was a powerful analytical method for the detection of CAP, FF and TAP simultaneously.
Preparation of bovine serum albumin restricted access octadecyl/phenyl-mixed-functionalized magnetic silica nanoparticles for magnetic solid phase extraction of antidepressants in aquatic products followed by UHPLC-MS/MS

Abstract:Pharmaceuticals are widespread in aquatic ecosystem, which may pose a potential threat to fish and human. In the study, a robust and reliable magnetic solid-phase extraction (MSPE) based on bovine serum albumin (BSA) restricted access octadecyl/phenyl-mixed-functionalized magnetic silica nanoparticles (BSA-C18/Ph-Fe3O4@SiO2 NPs) as sorbent was developed for the extraction of venlafaxine, paroxetine, fluoxetine, norfluoxetine, sertraline and diphenhydramine from the muscle extracts of aquatic products followed by ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) detection. The homemade sorbent showed appropriate compatibility in aqueous solution and good performance in reducing matrix interference of snakehead muscle tissue extracts with absolute matrix effect ranging in 95.4%–105.5%. The protocol was validated in analyte-free snakehead muscle with favorable recoveries ranging in 91.6%–103.6% and relative standard deviations (RSDs) less than 9.5%. Limits of detection (LODs) of the drugs were lower than 0.018 μg kg–1. Short-term drug-exposure experiments at low and high doses were conducted on snakeheads, and the measured contents of analytes were in the range 0.029–9.58 μg kg–1 with appropriate recoveries (90.0%–114.0%). The approach was extensively applied for the analysis of twelve species of market-sale aquatic products (total 37 samples), and up to 1.868 and 0.521 μg kg–1 of diphenhydramine and venlafaxine were measured, respectively. The approach shows remarkable potential in biological complex samples.

Abstract:The extraction of quinolone antibiotics (QAs) is crucial for the environment and human health. In this work, polyacrylonitrile (PAN)/covalent organic framework TpPa–1 nanofiber was prepared by an electrospinning technique and used as an adsorbent for dispersive solid-phase extraction (dSPE) of five QAs in the honey and pork. The morphology and structure of the adsorbent were characterized, and the extraction and desorption conditions for the targeted analytes were optimized. Under the optimal conditions, a sensitive method was developed by using PAN/TpPa–1 nanofiber as an adsorbent coupled with high-performance liquid chromatography (HPLC) for five QAs detection. It offered good linearity in the ranges of 0.5–200 ng·mL−1 for pefloxacin, enrofloxacin, and orbifloxacin, and of 1–200 ng·mL−1 for norfloxacin and sarafloxacin with correlation coefficients above 0.9946. The limits of detection (S/N = 3) of five QAs ranged from 0.03 to 0.133 ng·mL−1. The intra-day and inter-day relative standard deviations of the five QAs with the spiked concentration of 50 ng·mL−1 were 2.8–4.0 and 3.0–8.8, respectively. The recoveries of five QAs in the honey and pork samples were 81.6–119.7%, which proved that the proposed method has great potential for the efficient extraction and determination of QAs in complex samples.

Abstract:A method was established for the rapid determination of 28 prohibited and restricted veterinary drugs of five classes (sulfonamides, quinolones, chloramphenicols, nitrofurans metabolites and triphenylmethane) in fish and shrimp by ultra high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) coupled with simultaneous extraction and purification. After optimizing the sample pretreatment methods and chromatographic conditions, the samples were hydrolyzed with hydrochloric acid. The five kinds of residual drugs were simultaneously extracted with acetonitrile after nitrofuran metabolites derivatized with 2-nitrobenzaldehyde. The extraction solutions were dehydrated with sodium chloride and salted out. The acetonitrile layer was purified with n-hexane and concentrated to near dryness. The residues were dissolved in 1.0 mL methanol. Then, the solutions were purified by dispersive solid phase extraction with C18, N-(n-propyl)ethylenediamine (PSA) and amino bonded silica gel (NH2) sorbents. The residues were separated on a ZORBAX C18 chromatographic column (50 mm×2.1 mm, 1.8 μm). The MS data were acquired in positive and negative multiple reaction monitoring (MRM) modes, and quantitated by the isotope internal standard method. With this approach, the linear relationships of the 28 veterinary drugs were good. The linear correlation coefficients were greater than 0.99. The limits of detection and limits of quantification were in the range of 0.1-0.8 μg/kg and 0.3-2.7 μg/kg, respectively. The average recoveries at three spiked levels (1, 5, and 20 fold of the LOQs) were 70.0%-120% with relative standard deviations of 0.9%-10.0%. The method is simple, rapid, accurate and sensitive, and it is suitable for the rapid determination of various prohibited and restricted drugs in fish and shrimp.

Abstract:A proposal to modify the Cornell 12 GeV electron synchrotron facility to produce colliding beams of electrons and positrons was submitted to the National Science Foundation in May 1975. Subsequently, approval for design and prototype work was given and funding for the project is being requested by the NSF. Our goal is to complete this facility modification by October 1979. The status of the design and prototype work is described in this presentation.

Abstract:A novel method, solid-phase extraction combined with dispersive liquid-liquid microextraction (SPE-DLLME), was developed for ultra-preconcentration of 10 antibiotics in different environmental water samples prior to ultra-high performance liquid chromatography-tandem mass spectrometry detection. The optimized results were obtained as follows: after being adjusted to pH 4.0, the water sample was firstly passed through PEP-2 column at 10 mL min−1, and then methanol was used to elute the target analytes for the following steps. Dichloromethane was selected as extraction solvent, and methanol/acetonitrile (1:1, v/v) as dispersive solvent. Under optimal conditions, the calibration curves were linear in the range of 1–1000 ng mL−1 (sulfamethoxazole, cefuroxime axetil), 5–1000 ng mL−1 (tinidazole), 10–1000 ng mL−1 (chloramphenicol), 2-1000 ng mL−1 (levofloxacin oxytetracycline, doxycycline, tetracycline, and ciprofloxacin) and 1–400 ng mL−1 (sulfadiazine) with a good precision. The LOD and LOQ of the method were at very low levels, below 1.67 and 5.57 ng mL−1, respectively. The relative recoveries of the target analytes were in the range from 64.16 % to 99.80 % with relative standard deviations between 0.7 and 8.4 %. The matrix effect of this method showed a great decrease compared with solid-phase extraction and a significant value of enrichment factor (EF) compared with dispersive liquid-liquid microextraction. The developed method was successfully applied to the extraction and analysis of antibiotics in different water samples with satisfactory results.

Abstract:A simple and sensitive capillary zone electrophoretic method for the residue determination of lomefloxacin, enoxacin, sarafloxacin, enrofloxacin, sulfadiazine, sulfamethoxydiazine, and sulfadimethoxypyrimidine was developed. Accelerated solvent extraction with acetonitrile as solvent at 80 °C and 10.3 MPa was used for sample preparation. On-column preconcentration using large-volume sample stacking with polarity switching was applied for sensitivity enhancement. The standard curves for the determination of the seven residues have good linearity (r > 0.999). The method limits of quantification (LOQs) were 13.2–20 μg kg−1 for the quinolones and 8.0 μg kg−1 for the sulfonamides. Intraday and inter-day precisions (RSDs) at 100 μg kg−1 level were 1.4–4.1 and 2.3–4.7 %, respectively. The residue determination of the quinolones and sulfonamides in aquatic products was achieved. Their average recoveries for three spiked levels (50, 100, and 500 μg kg−1) were 85.4–95.7 % with the RSD of 1.1–4.7 % for shrimp sample and 84.3–94.8 % with the RSD of 1.4–4.5 % for sardine sample. The proposed method provides a rapid and simple extraction procedure, satisfactory repeatability, and reproducibility, as well as high accuracy, and can permit the routine detection of the studied drug residues in aquatic product at the content below or above the MRL.

Abstract:Rapid and accurate quantification of trace targets in complex samples is an extremely challenging issue in fast analysis field. Herein, we developed Fe 3 O 4 -carboxyl modified AuNPs-chitosan@AgNPs composite (Fe 3 O 4 –AuNCs–Cs@AgNPs) as a robust surface-enhanced Raman scattering (SERS) substrate for rapid analysis of tryptamine (TPA) and ofloxacin (OFX). The substrate possessed abundant surficial active sites of –NH 2 , –OH and –COOH groups. The substrate exhibited good SERS activity for several different model molecules with enhancement factors ( EFs ) of 1.2 × 10 8 for 4-mercaptobenzoic acid. The substrate presented good stability for detection of TPA at pH 6.0 and OFX at pH 8.0, and relative standard deviations less than 5.0% for intra-batch and 6.0% for inter-batch. Also, the substrate possessed good time-stability within 50 days. The substrate integrated advantages of efficient enrichment, fast magnetic separation, and strong localized surface plasmon resonance properties of AgNPs. With versatile merits, TPA and OFX can be enriched and separated within 10 min. SERS methods for analysis of TPA and OFX were developed with detection limits of 35.5 μg/L and 15.8 μg/L, respectively. TPA and OFX were actually found in aquatic product, and recoveries during sample analysis were 89.3%–110% for TPA and 89.3%–96.8% for OFX. The analytical process completed within 30 min via enrichment-separation-detection all-in-one, exhibiting great potential for rapid analysis of toxic biogenic monoamines and antibiotic residues in food.

Abstract:An analytical method was developed and validated for the simultaneous determination of 3 tetracyclines and 2 quinolones in pork and chicken by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) by positive ion mode under multiple reaction monitoring(MRM).The sample were extracted by ultrasonic extraction in Mcllvaine-Na2EDTA buffer(pH 4.0).The extract was cleaned up on a solid phase extraction(SPE) cartridge packed with multiwalled carbon nanotubes(MWCNTs) and using formic acid-acetonitrile(5 ∶95,by volume) mixture as eluting solvent.The drugs residues were analyzed by UPLC-MS/MS.The analysis time was only 10 min.Good linearities were obtained in the range of 5-500 μg/L.The limit of detection(LOD) was 15 μg/L for chlortetracycline(CTC) and 5 μg/L for the other four drugs.The recoveries at spiked levels of 50,100,200 μg/L ranged from 76% to 90% for 5 drugs with relative standard deviations(RSDs) less than 9%.With good stability and high sensitivity,the method could be used for the rapid determinaion of these drug residues in pork and chicken sample.
Development and evaluation of a semi-automatic single-step clean-up apparatus for rapid analysis of 18 antibiotics in fish samples

Abstract:Here we report a novel semi-automatic pre-treatment apparatus to fulfill a single-step clean-up aim for rapid quantification of 18 antibiotics in fish samples. Trimethoprim and 17 sulfonamides were extracted from fish samples by acetonitrile solution based on ultrasonic-assisted extraction. Cheap analytical filtration syringe (CAFS) clean-up column was used to fulfill single-step clean-up of the extract which was set on the developed semi-automatic clean-up apparatus. This semi-automatic clean-up apparatus is quite convenient for single-step purification by precisely controlling the flow rate of the extracts, in which up to 10 samples can be purified in parallel at one time. The time needed to clean sample was only about 2 min per 10 samples. Type of compatible purification column ranged from 1 mL (inner diameter × length, 5.6 × 55 mm) to 12 mL (inner diameter × length, 15.6 × 83 mm). The apparatus also did not need additional gas source such as nitrogen or air. After purification, the analytes were simultaneously separated and quantified by high-performance liquid chromatography/quadrupole-linear ion trap tandem mass spectrometry (HPLC-QTRAP-MS/MS). The limits of detections ranged from 0.5 to 1.0 μg kg−1, and the limits of quantifications ranged from 1.0 to 5.0 μg kg−1. All the recoveries ranged from 81.39% to 116.01%. Intra-day and inter-day relative standard deviations (RSDs) were lower than 14% for all analytes.

Abstract:Niclosamide is a mollusicide used for aquaculture on a large scale in China. However, it has high toxicity to aquatic animals and may cause damage to DNA. A modified QuEChERS (quick, easy, cheap, effective, rugged, and safe) extraction procedure with high-performance liquid chromatography–heated electrospray ionization-tandem mass spectrometry method was developed for its determination. Samples were extracted with ammoniated acetonitrile, and anhydrous magnesium sulfate was added to remove moisture and precipitate proteins. The extracts were further purified with octadecylsilane. The recoveries of niclosamide in fortified muscle tissue at concentrations of 0.5 to 20 µg/kg were between 76.40% and 95.60% with relative standard deviations less than 9.54%. The limits of detection and quantification were 0.2 µg/kg and 0.5 µg/kg, respectively. The developed method was used to study the adsorption of niclosamide in channel catfish after administration.

Abstract:Nowadays, anesthetics are widely used in fishery production processes, such as fish breeding, surgery, and fresh aquatic product transportation. Because of the widespread application of anesthetic drugs in aquatic products, there is an increasing demand for the rapid and sensitive detection of anesthetic drugs in aquatic products. The complex aquatic product matrix contains a variety of interfering substances, such as proteins, fats, and phospholipids, along with anesthetic drug residues at very low concentrations; therefore, it is necessary to adopt appropriate pretreatment methods for improving the sensitivity of detection. In this study, a dispersive solid-phase extraction (DSPE) method, combined with high-performance liquid chromatography, was established for the simultaneous detection of seven anesthetic drugs in aquatic products, viz. procaine, oxybuprocaine, tricaine, eugenol, methyl eugenol, isoeugenol, and methyl isoeugenol. For the DSPE step, pretreatment conditions, such as extraction solvent, extraction time, adsorbent amount, and DMSO dosage, were optimized. Sample pretreatment is a three-step process. First, in ultrasound-assisted extraction, 2.0 g samples were extracted using 10.0 mL 1.0% formic acid in acetonitrile under ultrasound conditions for 10 min. Then, DSPE was performed with mixed adsorbents: the solvent extracts were cleaned using 20 mg poly(styrene-glycidylmethacrylate) microspheres (PS-GMA), 50 mg primary secondary amines (PSA), and 10 mg C18, followed by separation by centrifugation. Finally, DMSO-assisted concentration was applied: the organic layer was collected and was dried at 40 ℃ in a N2 stream with 100 μL DMSO. Water was added to the residue to obtain a final volume of 1.0 mL for HPLC analysis. The seven anesthetic drugs were separated on a Welch welchrom C18 column (250 mm×4.6 mm, 5 μm) by gradient elution using methanol and 0.05% formic acid in 5 mmol/L ammonium acetate aqueous solution as mobile phases. The detection wavelengths were 235, 260, and 290 nm. Two matrix matching standard curves for fish and shrimp were applied for quantitative analysis. Under optimized conditions, the seven target anesthetics showed good linear relationships in their respective concentration ranges (R2>0.999), with the limit of detection (LOD) ranging from 0.011 to 0.043 mg/kg. In fish samples, the mean recoveries obtained at three concentration levels were between 79.7% and 109%, with relative standard deviations (RSDs) being less than 7.2%. In shrimp samples, mean recoveries were 78.0%-99.9%, with RSDs being less than 8.3%. This simple, rapid, accurate, and sensitive method can be applied to the detection of three kinds of aminobenzoic acid esters and four kinds of eugenol anesthetic drugs in aquatic products.

Abstract:The residues of sedative and anesthetic drugs in aquatic products are an important factor affecting food safety. In order to screen the residues of sedative and anesthesia drugs in aquatic products, a rapid and sensitive method was established using solid phase extraction (SPE) and high-performance liquid chromatography–tandem mass spectrometry (HPLC–MS/MS). The samples were extracted by 80% aqueous acetonitrile by an alumina column and separated by a C 18 column. The mass spectra were determined in positive ion mode. The standard curves were matrix matched and quantified by the external standard method. The results showed that the linear ranges of the drugs in aquatic products were from 0.5 to 100 μg kg −1 . The correlation coefficient ( R 2 ) was greater than 0.990 and the detection limits were between 0.001 and 0.322 μg kg −1 . The average recovery rate and relative standard deviation were between 78.64% and 112.15% and from 0.41%-8.26%. Overall, the developed method showed promising results for the simultaneous qualitative and quantitative determination of the drug residues in aquatic products.

Abstract:Nitrofurans, a kind of banned antibiotics for their serious toxic side effects, have often been found in aquatic products in the past and are still found nowadays which were a severe threat for human health. A rapid quantization of nitrofuran metabolites residues in aquatic products by ultra-high performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) method was developed in this study. The experimental operation consisted of an acid-catalyzed release of protein-bound metabolites, followed by derivatization with 2-chlorobenzaldehyde (2-CBA) and then extraction with ethyl acetate before analysis by positive electrospray ionisation (ESI + ) with multiple reaction monitoring (MRM) type. 2-CBA was employed as derivatization reagent in this study, which greatly reduced the derivatization time. The linearity range for the four analytes were 0.25–50 μg kg −1 with good determination coefficients ( R 2 > 0.9982). The limit of detection (LOD) were 0.1 μg kg −1 , 0.1 μg kg −1 , 0.3 μg kg −1 , 0.3 μg kg −1 and the limit of quantification (LOQ) were 0.3 μg kg −1 , 0.3 μg kg −1 , 0.9 μg kg −1 , 0.9 μg kg −1 for AOZ, AMOZ, SEM, and AHD, respectively. Recovery rates were ranged from 90% to 114% for fortified samples at levels of 1.0 μg kg −1 , 5.0 μg kg −1 and 10 μg kg −1 with relative standard deviation values ranging from 1.6% to 5.8%. The developed method was successfully applied in 100 aquatic products for NFs metabolites detection and the results were further confirmed by the national standard GB/T 21311-2007 method.

Abstract:The residues and abuse of antibiotics have seriously endangered ecological balance and human health; meanwhile, antibiotics determination is very difficult because of their low levels and multiple categories in complicated matrices. Appropriate sample pretreatment is usually imperative to enrich (ultra)trace antibiotics and eliminate matrix interference prior to chromatographic analysis. Dispersive liquid-liquid microextraction (DLLME) has become an ideal pretreatment technique owing to its simplicity, effectiveness, low-consumption, etc. In this work, an ultrasonic-assisted DLLME (UA-DLLME) was developed for the simultaneous extraction of seven sulfonamides (SAs) antibiotics in environmental water and seafood samples coupled with HPLC-DAD determination. Several parameters affecting UA-DLLME efficiency were systematically optimized, and consequently the SAs were separated and detected within 14.5 min. The obtained limits of detection (LODs) and limits of quantification (LOQs) ranged from 0.7–7.8 μg/L and 2.4–26.0 μg/L for three water samples (seawater, aquaculture wastewater and lake water) and two seafood samples (pomfrets and shrimps). High recoveries (80.0–116.0%) with low relative standard deviations (0.1–8.1%) were achieved for all the tested samples at three spiked levels. Notably, sulfadimethoxine was found at 24.49 μg/L in one seawater sample. The facile, robust and benign DLLME-HPLC method demonstrated promising perspectives for multiresidue analysis of antibiotics.

Abstract:Fluoroquinolones (FQs) are a class of synthetic antibiotics with a broad active spectrum towards mycoplasma and bacteria (both Gram positive and negative). The contamination of environmental water samples with FQs has been a concern due to the extensive use of them in the veterinary and pharmaceutical industries. Reliable and simple analytical methods are needed for the monitoring of these FQs. An off-line procedure of immunoaffinity extraction (IAE) followed by capillary electrophoresis-mass spectroscopy (CE-MS) has been developed for simultaneous determination of eight typical FQs in environment water. A broad-specific polyclonal antibody was home-prepared and immobilized on a Sepharose 4B stationary phase for sample affinity extraction and the enrichment factor of our columns was about 100. CE-MS was subsequently applied to FQs separation and quantification. The immunoaffinity column was evaluated in the parameters of selectivity, binding capacity, elution protocol, and recovery. Under the optimized conditions, the developed method was successfully applied to analyze FQs spiked with 2.5 nM to 30 nM of environmental water from a local lake, providing satisfactory recoveries from 72% to 112%, and LODs and LOQs ranging from 1.2 nM to 5.0 nM and 4.0 nM to 17 nM, respectively.
