Abstract:The enzyme-linked immunosorbent assay (ELISA) offers several advantages, including simple operation, high throughput, and low cost, making it an ideal immunoassay method for efficient screening of disease-related biomarkers in clinical samples. However, the traditional colorimetric ELISA has relatively low sensitivity, which promotes the continuous emergence of various novel signal amplification technologies. In this work, we fused the AFP-specific nanobody (A1) with the streptavidin-binding peptide (SBP) to develop a fusion protein (A1-SBP) as biorecognition element in a colorimetric ELISA for detecting AFP. Besides, to further improve the sensitivity of the traditional colorimetric ELISA, the streptavidin-conjugated polymerized horseradish peroxidase (SA-PolyHRP) were selected as a detection probe for signal amplification. The proposed signal enhancement strategy demonstrated a limit of detection (LOD) of 0.597 ng/mL for the SA-polyHRP-based ELISA, which is 7.67-fold lower than that of the traditional SA-HRP-based ELISA without additional steps. Furthermore, the proposed SA-polyHRP-based ELISA showed a good correlation with the detection of clinical samples using the Roche E601 chemiluminescence immunoassay analyzer. Therefore, the proposed signal enhancement strategy is an attractive approach for improving the sensitivity of immunoassay without requiring additional steps.

Abstract:A novel electrochemical immunoassay based on nanobody heptamer for ultrasensitive detection of alpha fetoprotein (AFP) is proposed. First, AFP heptamer fusion protein (A1-C4bp α) is prepared via fusing the AFP-specific nanobody (A1) with the C-terminal fragment of C4-binding protein (C4bp α). Then A1-C4bp α heptamer with higher affinity is used as the molecule recognizing probe to capture numerous the AFP, increasing the sensitivity of the immunosensor. Moreover, AuNPs decorated zeolitic imidazolate framework () with large surface area, and abundant binding sites serves an ideal nanocarrier to increase heptamer loading. Multiwalled carbon nanotube (MWCNTs) with excellent electrical conductivity was immobilized on glassy carbon electrode (GCE) to enhance electrochemical signals. The coupling between the @MWCNTs and heptamer technologies constructed an ultrasensitive and specific immunosensor for AFP detection in the range of 0.1 to 105 pg mL−1 with a lower detection limit of 0.033 pg mL−1. The as-prepared electrochemical sensing exhibited excellent performance and could be employed to determine AFP with satisfactory results in actual serum samples, suggesting that the proposed strategy for nanobody multimerization provides an alternative scheme for enhancing sensitivity in important biomarkers detection.

Abstract:The authors describe a sensitive electrochemical immunoassay for the cancer biomarker α-fetoprotein (AFP). It is based on a combination of an immunoassay with DNA-based signal amplification. Two-dimensional MnO2 nanosheets modified with gold nanoclusters (AuNC-MnO2) were synthesized through one-pot process and utilized as an electrochemical probe. Bovine serum albumin served as templating agent to guide the formation and assembly of the modified sheets. The detection antibody against AFP and a polycytosine DNA sequence (dC20) were immobilized onto the modified nanosheets. The electrochemical assay follows the usual sandwich protocol. The antibodies on the nanosheets then bind to AFP, while dC20 causes signal amplification. The reaction of the phosphate backbone of dC20 with molybdate leads to the formation of redox-active molybdophosphate which generates an electrochemical current, typically measured at 0.20 V (vs. Ag/AgCl). The method allows AFP to be determined in the 0.01 to 10 ng·mL−1 concentration range, and the detection limit is as low as 5 pg·mL−1. This strategy overcomes the drawbacks of conventional immunoassays whose sensitivity is often limited because many immunoassays are rather difficult to amplify. The method has a wide scope in that various other DNA signal amplification methods such as rolling circle amplification and hybridization chain reactions may also be applied.
Determination of alkaline phosphatase activity and of carcinoembryonic antigen by using a multicolor liquid crystal biosensor based on the controlled growth of silver nanoparticles

Abstract:An ultrasensitive liquid crystal biosensor is described for multicolor visualization of the activity of alkaline phosphatase (ALP) based on the controlled growth of silver nanoparticles. The enzymatic product is accumulated on the surface of the LC sensing film by means of silver deposition, and the birefringent signal (observed with a polarizing microscope) is strongly enhanced as a result. The presence of AuNPs also enhances the sensitivity by about 4 orders of magnitude. The bright spots in polarized optical microscopy (POM) images increase with increasing activities of ALP. The signal intensities of the spots are then calculated by using Photoshop software and by multiplying the average brightness of the spots by the pixel value. The detection limit for ALP is 1.2 nU·mL−1, which is 5–7 orders of magnitude lower than other colorimetric or fluorometric methods. The method was applied to a highly sensitive immunoassay for the carcinoembryonic antigen (CEA) by integrating immunomagnetic separation. The immunoassay was applied to the analysis of complex samples without tedious sample pretreatment, and a detection limit as low as 0.35 pg·mL−1 of CEA was achieved. The method has attractive features in that it provides an ultrasensitive multicolor visualization approach for enzymes such as ALP, but also paves the way to a new kind of immunoassay coupled to immunomagnetic separation.Graphical abstractA signal enhanced liquid crystal (LC)-based multicolor immunosensor is described that is based on immunomagnetic separation and biometallization. Abstract:Multifunctional nanolabels with both facile signal amplification and high selectivity are essential for the sensitive determination of the vital tumor marker alpha fetoprotein (AFP). Here, we prepared an immunosensing nanolabel (Ab2-PDA-AF) for convenient AFP detection by grafting the detection antibody (Ab2) and the electroactive redox probe aminoferrocene (AF) onto polydopamine (PDA) nanospheres through Michael addition and Schiff base reactions. The electrochemical signal is dually amplified by the Ab2-PDA-AF nanolabels when the nanolabels are selectively bound to the electrode. The electrochemical signal can be first amplified by oxidizing AF of the Ab2-PDA-AF on the electrode. The signal can be further amplified by the redox reaction of PDA nanospheres with the linked AF/AF+ on the nanospheres. More importantly, the integration of AF-PDA-Ab2 favors a quick electron transfer between AF and PDA nanospheres. Redox probes of AF/AF+ are bonded to the nanolabels instead of dispersing in an electrolyte solution, thus decreasing the electron-transfer resistance and increasing the current signal. The interfacial kinetic analysis demonstrates that the charge-transfer resistance is decreased by 80% by using Ab2-PDA-AF nanolabels. The sensor displays a low detection limit of ca. 0.01 pg/mL, as well as excellent selectivity, reproducibility, and stability. The linear calibration plot of the AFP immunosensor has been obtained in a range of 0.5–1000.0 pg/mL. The capability of the immunosensor has been verified by implementing the detection of AFP in human serum samples.

Abstract:highly sensitive electrochemiluminescence immunosensor was originally proposed based on lead sulfide nanocrystals (PbS NCs) to detect alpha fetoprotein (AFP). Herein, PbS NCs, as a kind of nanoenzyme, was synthesized and applied to the luminol-hydrogen peroxide (L-H2O2) system for the first time. Additionally, PbS NCs with superior electrocatalytic activity toward H2O2 can enhance the electrochemiluminescence response of luminol. In this protocol, chitosan was utilized to immobilize PbS NCs through its film-forming characteristics, and further developed an ECL immunosensor by amination reaction combined with AFP antibodies. When the modified electrode was incubated with different concentrations of AFP antigen, the electrochemiluminescence intensity decreased obviously. In addition, under optimal conditions, the sensor can sensitively and selectively detect the alpha fetoprotein ranging from 1.0x10(-5) to 800 ng/mL with the detection limit of 3.0 fg/mL. On this basis, the proposed ECL immunosensor will open up a new way to sensitively detect AFP and exhibit its application prospect in biological analysis.

Abstract:Immunosorbent assay is one of the most popular immunological screening techniques which has been widely used for the clinical diagnosis of alpha-fetoprotein (AFP). While traditional immunosorbent assay (ELISA) suffers from low detection sensitivity due to its low intensity of colorimetric signal. To improve the sensitivity of AFP detection, we developed a new and sensitive immunocolorimetric biosensor by combining Ps-Pt nanozyme with terminal deoxynucleotidyl transferase (TdT)-mediated polymerization reaction. The determination of AFP was achieved by measuring the visual color intensity produced by the catalytic oxidation reaction of the 3,3′,5,5′-tetramethylbenzidine (TMB) solution with Ps-Pt and horseradish peroxidase (HRP). Owing to the synergistic catalysis of Ps-Pt and horseradish peroxidase HRP enriched in polymerized amplification products, this biosensor exhibited a significant color change within 25 s in the presence of 10–500 pg/mL AFP. This proposed method allowed for the specific detection of AFP with a detection limit of 4.30 pg/mL and even 10 pg/mL target protein could be distinguished clearly by visual observation. Furthermore, this biosensor could be applied to analysis of AFP in the complex sample and could be easily extended to the detection of other proteins.

Abstract:Potentiometric immunoassays have been utilized for the quantitative detection of alpha-fetoprotein (AFP) in hepatocellular carcinoma, but most of them involve low sensitivity and enzyme labels, and thus are unfavorable for routine use. In this work, we report the proof-of-concept of a sensitive and powerful ion-selective potentiometric sensing method for AFP detection with an in situ amplified signal readout. This potentiometric immunoassay mainly contains a silver nanocluster-functionalized single-stranded DNA (AgNC-DNA), a short DNA primer and two antibodies. A sandwich-type immunoreaction is employed for AFP determination on an anti-AFP capture antibody-coated microplate using a biotinylated human AFP secondary antibody. Coupling with a typical biotin-avidin system, the biotinylated DNA initiator strands are conjugated on the microplate in the presence of AFP to induce the rolling cycle amplification (RCA) reaction, followed by AgNC-DNA hybridization. Upon addition of HNO3, the hybridized AgNCs are dissolved into numerous Ag(I) ions, which can be readily determined on a portable handheld silver-ion selective electrode (Ag-ISE). Under optimal conditions, the electrode potential increases with an increase in AFP concentration and exhibits a good linear range of 0.01-100 ng mL-1 at a detection limit of 7.9 pg mL-1. Moreover, the Ag-ISE-based potentiometric immune assay also shows good reproducibility, high specificity and long-term storage stability. Importantly, 18 human serum specimens containing the AFP analyte are screened using the potentiometric immunoassay, giving well-matched experimental results relative to the referenced enzyme-linked immunosorbent assay (ELISA) method.
Fe-N-C Single-Atom Nanozymes With Peroxidase-Like Activity For The Detection Of Alkaline Phosphatase

Abstract:Single-atom nanozymes have drawn wide attention in bio-sensing for their remarkable merits such as low cost, high stability, and maximum atom utilization. Herein, a colorimetric strategy based on Fe-N-C single-atom nanozymes (Fe/NC-SAs) was established for the detection of alkaline phosphatase (ALP) activity. The Fe/NC-SAs prepared by pyrolysis have excellent peroxidase-like activity and can oxidize 3,3 ',5,5 '-tetramethylbenzidine (TMB) to a blue color product in the presence of hydrogen peroxide (H2O2). When ascorbic acid (AA) is added to the system, the blue color fades, and the absorbance has a linear relationship with the concentration of AA. Alkaline phosphatase (ALP) can catalyze the hydrolysis of ascorbic acid 2-phosphate (AAP) to produce AA. Thus, a strategy based on Fe/NC-SAs for the detection of ALP activity was established, which provided a linear range of 0.1-1.5 U L-1 and a limit of detection as low as 0.05 U L-1. Besides, Fe/NC-SAs showed high stability under harsh conditions. Moreover, an Fe/NC-SA-based assay was successfully validated using human serum samples for ALP determination with satisfactory results, and has broad prospects in the field of biosensing.

Abstract:This work demonstrates an effective quenching electrochemiluminescent (ECL) immunosensor based on resonance energy transfer for the sensitive detection of alpha fetoprotein (AFP). In this strategy, graphitic carbon nitride (g-C3N4) was coupled with bismuth molybdenum oxide (Bi2MoO6) to form a g-C3N4/Bi2MoO6 nanocomposite as a novel type of ECL immunosensor. The as-synthesized amino-modified g-C3N4/Bi2MoO6 nanocomposite presents strong and stable cathodic ECL activity compared to pristine g-C3N4. One plausible reason is that the synergistic effect between the g-C3N4 and Bi2MoO6 could facilitate charge transfer process and thereby enhancing the separation efficiency of electron-hole pairs. The other functional part of the immunosensor, carboxyl CoS2 nanoboxes with a broad absorption range, was rationally designed and introduced. The evidence that the absorption spectra of carboxyl CoS2 NBs overlap with ECL spectra of g-C3N4/Bi2MoO6 nanocomposite holds accountable for exceptionally weakened ECL signal. This sandwich-type immunosensor was setup based on quenching mechanism concerning amino-modified g-C3N4/Bi2MoO6 as an ECL donor and carboxyl CoS2 NBs as an ECL accepter. The strategy was optimized to achieve a convincible and sensitive detection goal for AFP with a wide quantifiable range of 0.5 pg/mL-10 ng/mL whilst a sufficiently low detection limit of 0.04 pg/mL (S/N = 3). This immunosensor shows great potential for real sample analysis with reasonable recoveries ranging from 95.5 to 99.0 %, demonstrating its high precision for AFP determination.
An Electrochemical Biosensor for the Assay of Alpha-Fetoprotein-l3 with Practical Applications.

Abstract:The detection of alpha-fetoprotein-L3 (AFP-L3) is of great importance for hepatocellular carcinoma (HCC) diagnosis, but remains unsatisfactory owing to poor sensitivity and complex operating steps. In this work, a simple and sensitive method has been proposed for the detection of AFP-L3. Firstly, biotinylated Lens culinaris agglutinin-integrated silver nanoparticles (B-LCA-AgNPs) is fabricated. Owing to the specific interaction between Lens culinaris agglutinin and AFP-L3, AFP-L3 can be detected directly through the electrochemical signal readout of AgNPs, avoiding separated steps used in clinical practice. Furthermore, after the recognition between B-LCA-AgNPs and AFP-L3, large amount of AgNPs can be gathered at the binding site through avidin-biotin interactions, which can amplify the signal. Therefore, detection of AFP-L3 can be sensitively achieved. Moreover, compared with the other approaches, this new method has a better linear correlation (25–15,000pg/mL) and a lower detection limit (12pg/mL). Also, the new method developed in this work has been demonstrated to have good stability and reproducibility for the assay of AFP-L3 in human serum samples, so, it may hold a great application prospect in clinical diagnostics.

Abstract:Accurate detection of tumor biomarkers in blood is crucial for diagnosing and treating tumor disease. In this study, a metal enzyme-linked immunosorbent assay (MeLISA) was fabricated for the ultrasensitive and naked-eye detection of tumor biomarker alpha-fetoprotein (AFP) in clinical serum samples. Herein, novel copper metal-organic frameworks and gold platinum nanoparticles composites (Cu-MOFs@AuPtNPs) were synthesized for the first time by in-situ method, which showed an enormous specific surface area and excellent peroxidase (POx) mimicking property. Cu-MOFs@AuPtNPs linked with antibodies targeting AFP performed as signal nanoprobe to amplify the detection signal. Additionally, the specificity of MeLISA was significantly enhanced through conventional antigen-antibody reaction and efficient blocking of non-specific sites with BSA. In optimal conditions, the sandwich-type MeLISA exhibited a wide range from 0.001 to 1000 ng mL-1 (R2 = 0.997) and a low detection limit of 0.86 pg mL-1 (S/N = 3) with acceptable stability, selectivity, and reproducibility. It was noteworthy that the suggested MeLISA performed exceptionally well in detecting clinical serum samples, which were visible to the naked eye and did not require complex platforms. To sum up, the innovative MeLISA based on Cu-MOFs@AuPtNPs provides an alternative method for early cancer diagnosis, particularly in economically backward areas where simple diagnostic apparatus is extremely desirable.

Pathogenicity of Haemoproteus meleagridis (Haemosporina: Haemoproteidae) in experimentally infected domestic turkeys.

Abstract:Sporozoite-induced experimental infections of Haemoproteus meleagridis produced a moderate to severe myositis and significant effects on weight gain and growth in domestic turkey poults. Pathological effects occurred in both low- and high-dose infections (4,400 and 57,500 sporozoites, respectively). Low-dose birds weighed significantly less than controls at 3 wk postinfection (PI) when peripheral parasitemia reached a peak and had significantly shorter tarsometatarsal lengths at both 1 and 3 wk PI. High-dose birds were significantly lighter and smaller than control and low-dose birds throughout the course of the 8-wk study. Infected birds were not anemic in spite of high parasitemias that often exceeded 50% of circulating erythrocytes. The most serious pathological effects occurred prior to patency and were associated with development of megaloschizonts in skeletal muscle. Microscopic lesions in 4 high-dose birds that died between 19 and 22 days PI were characteristic of a severe, acute hemorrhagic myositis. Megaloschizonts were surrounded by a hemorrhagic inflammatory infiltrate composed of macrophages, heterophils, giant cells, and red blood cells. Muscle fibers adjacent to megaloschizonts were swollen, hyaline, and contained prominent calcium deposits. Other observations included enlargement of the spleen, deposition of pigment in macrophages of the lung and spleen, and secondary bacterial and fungal infections in the intestine and lungs. Necrotic and calcified muscle fibers and degenerating megaloschizonts were still present at 8 wk PI when the experiment ended. Our results demonstrated significant pathological changes in H. meleagridis-infected domestic turkeys that were associated primarily with preerythrocytic stages of development.
One-step and label-free detection of alpha-fetoprotein based on aggregation of gold nanorods

Abstract:In this study, a novel one-step label-free immunoassay using gold nanorods (GNRs) for sensitive detection of alpha-fetoprotein (AFP), one of the major markers for hepatocellular tumors, was successfully developed. GNRs were fabricated through seed-mediated growth procedure, and their surface was activated by complete replacement with alkanethiols for the attachment of antibodies to obtain GNR-antibody conjugates with special space conformation. Dramatic aggregation of GNRs was induced by the interaction between target proteins and capture antibodies, and confirmed by transmission electron microscopy (TEM) and Ultraviolet–visible (UV–vis) analysis. Associated wavelength red-shifts and intensity decreases in the absorption peak of GNRs were observed with increasing concentrations of target AFP. Therein the absorption intensity as sensing response showed excellent linearity with the concentration of AFP in logarithmic scale. It was illustrated that this method can be used reliably to detect AFP protein in phosphate-buffered saline (PBS) buffer at concentrations of picomolar level with a wide quantification range from 0.25 to 4.0nM. The complication of interfering proteins in the sample showed negligible effects on the detection results. Furthermore, the simple procedure, cost-effective and label-free features of this method offer advantages over common immunoassays. It is also expected that this new method would be applicable for detection of other tumor markers at low concentrations.

Although previous studies have shown that heavy drinking frequently leads to the generation of IgA antibodies against neo-antigens induced by ethanol metabolites and tissue deposition of IgAs, the associations between alcohol consumption, IgA immune responses, and kidney disease have not been examined. METHODS A total of 158 IgAGN patients (96 men, 62 women) were classified as abstainers (n = 38), moderate drinkers (n = 114), and heavy drinkers (n = 6) based on self-reported alcohol consumption. The reference population included 143 individuals (99 men, 44 women) who were either apparently healthy abstainers (n = 31), moderate drinkers (n = 43), or heavy drinkers devoid of liver disease (n = 69). The assessments included various biomarkers of alcohol consumption: carbohydrate-deficient transferrin (CDT), glutamyl transferase, gamma-CDT (combination of GGR and CDT), mean corpuscular volume (MCV), tests for liver and kidney function, serum immunoglobulin A (IgA), and specific IgA antibodies against acetaldehyde-protein adducts. RESULTS In male IgAGN patients, drinking status was significantly associated with MCV, p < 0.001; CDT, p < 0.01; and gamma -CDT, p < 0.05. In the reference population, all biomarkers and anti-adduct IgA levels were found to vary according to drinking status. In IgAGN patients, anti-adduct IgA levels were elevated in 63% of the cases but the titers did not associate with self-reported ethanol intake. CONCLUSIONS These data indicate high levels of IgA antibodies against acetaldehyde-derived antigens in IgAGN patients, which may hamper the use of the immune responses as markers of alcohol consumption among such patients. Future studies on the pathogenic and prognostic significance of anti-adduct immune responses in IgAGN patients are warranted.
Surface-enhanced Raman scatting microfluidic chip based on the identification competition strategy were used for rapid and simultaneous detection of liver cancer related proteins

Abstract:Biomarker testing plays a crucial role in the early detection of liver cancer. Herein, we developed a dual-signal amplification approach utilizing magnetic aggregation and a recognition competition strategy for the simultaneous detection of alpha-fetoprotein (AFP) and manganese superoxide dismutase (MnSOD) in serum. 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 were synthesized by functionalizing Raman signaling molecules and aptamer complementary chains onto the surface of gold nanoparticles (AuNPs). The detection complex Raman signal molecule@AuNPs@H-Fe3O4@cDNA was assembled by conjugating 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 with two nucleic acid aptamers (cDNA1 and cDNA2) modified with Fe3O4 through partial base complementary pairing. The target protein exhibited specific binding with the aptamer, leading to the competitive displacement of 4-MBA@AuNPs@H1 and DTNB@AuNPs@H2 from the Fe3O4 array surface, consequently resulting in a reduction of the Surface-Enhanced Raman Spectroscopy (SERS) signal. Through this approach, the limit of detection (LOD) for AFP and MnSOD in serum was achieved at remarkably low levels of 5.89 pg/mL and 6.23 pg/mL, respectively, with a reaction incubation period of only 5 min. Finally, the platform was utilized for the quantification of AFP and MnSOD in the serum of a nude mouse hepatocellular carcinoma model. The results obtained through SERS were consistent with those from enzyme-linked immunosorbent assay (ELISA), validating its accuracy. This methodology presents a novel approach for the swift and concurrent detection of proteins, holding significant clinical promise for the early diagnosis of hepatocellular carcinoma.
Development of active affibody aggregates induced by a self-assembling peptide for high sensitive detection of alpha-fetoprotein

Abstract:Alpha-fetoprotein (AFP) is one of the most important biomarkers associated with primary liver cancer and the development of accurate and sensitive methods for detecting the serum level of AFP is of great importance in cancer diagnosis. In this study, three self-assembling peptides (SAPs) tagged AFP specific affibody dimer (ZAFP D2)2 recombinant protein, (ZAFP D2)2-ELK16, (ZAFP D2)2-L6KD and (ZAFP D2)2-R18A were successfully produced and characterized. The (ZAFP D2)2-ELK16 affibody aggregates showed higher and more specific binding affinity to AFP compared to (ZAFP D2)2 and other two SAP-tagged (ZAFP D2)2 and in addition displayed enhanced AFP-detection sensitivity and improved stability. A sensitive two-site enzyme-linked immunosorbent assay for AFP using (ZAFP D2)2-ELK16 as the detection reagent was also established. The AFP detection limit and linear range using (ZAFP D2)2-ELK16 based two-site ELISA were 9.7 ng mL−1 and 20–600 ng mL−1, which were significantly improved compared to those of the (ZAFP D2)2 based two-site ELISA for AFP. To detect AFP in serum samples, the average recovery ranged from 95.02% to 110.41% and the relative standard deviation was in the range of 2.45%-3.97%, which meet the standard clinical testing requirements. This proposed strategy provides a high yield, easy purification and green biosynthesis method to prepare SAP-based affibody aggregates for enhanced detection sensitivity by improving the immunoassay signal amplification activity.

Abstract:BACKGROUND: Liver cancer is one of the most common cancers in the world, and it seriously threatens human life and health. Alpha-fetoprotein (AFP), as a carcinogenic glycoprotein, is an important serum marker for detecting liver cancer. Therefore, the accurate and sensitive determination of AFP is crucial for the early diagnosis and treatment of liver cancer. To this end, a label-free fluorescence aptasensor for detecting AFP based on the use of a novel organic Compound D with an aggregation-induced emission activity and aptamer-modified magnetic microparticles was constructed.RESULTS: Compound D could combine with the complementary short chain of the aptamer (CSC-Apt) of AFP to form the D/CSC-Apt complex and realize the fluorescence enhancement of Compound D. Then, magnetic particles modified by the Apt of AFP (Apt-Fe3O4) were prepared. When AFP (or nontarget substance) and D/CSC-Apt were successively added to the Apt-Fe3O4 solution, Apt-Fe3O4 selectively bound to AFP or the D/CSC-Apt complex. Magnetic separation technology showed the changes in the fluorescence intensity of the supernatant. The research results revealed a good linear relationship between the changes in the fluorescence intensity of the supernatant and concentration of AFP within the concentration range of 10-10000 pg mL-1. The proposed aptasensor could achieve high-sensitivity and high-specificity detection of AFP, and its limit of detection was 3 pg mL-1 (S/N = 3).SIGNIFICANCE AND NOVELTY: The sensor combines the advantages of high selectivity of Apt, high sensitivity of fluorescence analysis, AIE effect and good water solubility of Compound D, and rapid separation using magnetic separation technology. And it can be directly used for the detection of AFP in actual serum samples with high accuracy, whereas most of the methods reported in the literature can only detect AFP in spiked serum samples.
