Abstract:In this work, an aeration-enhanced membrane reactor (AEMR) was built to control dissolved oxygen in Schizochytrium sp. broth. The effect of culture modes, i.e. single shake and AEMR mode, on the docosahexaenoic acid (DHA) productivity of Schizochytrium sp. was investigated. Experimental results showed that the biomass production in the AEMR mode was higher than that in the single shake mode, while the final DHA productivity in single shake mode was higher than that in the AEMR mode. A two-stage mixed culture mode was proposed, in which Schizochytrium sp. was cultured in AEMR mode at a flow rate of 0.2 L min(-1) for 60 h to first increase biomass production, and then shifted to shake mode to improve DHA productivity. Compared to the single shake mode, the DHA productivity in mixed mode costed 40% less culture time and the DHA productivity at 96 h exhibited a relative increase of 60%. (C) 2013 Elsevier Ltd. Abstract:Thraustochytrids are aquatic unicellular protists organisms that represent an important reservoir of a wide range of bioactive compounds, such as essential polyunsaturated fatty acids (PUFAs) such as arachidonic acid (ARA), docosahexaenoic acid (DHA), eicosapentaenoic acid (EPA), which are involved in the regulation of the immune system. In this study, we explore the use of co-cultures of Aurantiochytrium sp. and bacteria as a biotechnological tool capable of stimulating PUFA bioaccumulation. In particular, the co-culture of lactic acid bacteria and the protist Aurantiochytrium sp. T66 induce PUFA bioaccumulation, and the lipid profile was evaluated in cultures at different inoculation times, with two different strains of lactic acid bacteria capable of producing the tryptophan dependent auxins, and one strain of Azospirillum sp., as a reference for auxin production. Our results showed that the Lentilactobacillus kefiri K6.10 strain inoculated at 72 h gives the best PUFA content (30.89 mg g-1 biomass) measured at 144 h of culture, three times higher than the control (8.87 mg g-1 biomass). Co-culture can lead to the generation of complex biomasses with higher added value for developing aquafeed supplements.

Abstract:Microalgae are the most propitious feedstock for biofuel production due to their lipid and fatty acid content. Microalgae cultivation shares many features with bioreactors, such as thermal and pH regulation, feeding procedures, and mixing to enhance heat and mass transfers. Aeration and stirring speeds are important parameters to reduce the costs of producing microalgae. In this study, three different photobioreactor types (stirred tank, airlift, bubble column) were characterized and compared for microalgae production. Hydrodynamics, mass transfer, and power consumption were determined for various aeration rates (0.9, 1.2, 1.5 L/min), and stirring speeds (100, 200 rpm), and Chlorella sorokiniana growth performance was compared under the conditions that provided the highest volumetric mass transfer and the lowest mixing time. Photo-bioreactor homogenization was good as indicated by low mixing times (< 10 s). Bubble column had the highest volumetric mass transfer due to its sparger design. Gas holdup and volumetric mass transfer coefficient were found to increase with the air flow rate and stirring speed. For stirred tank, bubble column, and airlift photobioreactors, maximum specific growth rates of C. sorokiniana were 0.053, 0.061, 0.057 h −1 , and biomass productivities were 0.064, 0.097, 0.072 gdw/L.day, respectively. Under the conditions tested, growth was limited by the volumetric mass transfer in the airlift and stirred tank and bubble column was the best option for producing microalgae. These findings pave way for more extensive use of these systems in producing microalgae and provide a basis to compare photobioreactors of different designs.

Abstract:Microalgae's potential to produce high value bioactives is contingent on the cost-effective harvesting of algal biomass. The use of CTAB (cetyl trimethylammonium bromide) assisted dispersed air flotation as a harvesting technique for Schizochytrium biomass, was optimized as a cost-effective means for recovery ( R ) and concentration factor ( CF ) using Response Surface Methodology. To the best of our knowledge, this is the first study to employ surfactant assisted dispersed air flotation (SDAF) in the recovery of a heterotrophic thraustochytrid strain. A Box-Behnken design of experiment investigating the operating parameters of CTAB amount, air flow rate and volume on Schizochytrium was employed. Initially, both responses were analyzed individually and then used to simultaneously maximize both variables. The optimized conditions of CTAB (500 mg/L), air flow of 2 L/min and volume of 600 mL resulted in a R of 91 % and CF of 19 times. Although a secondary step is necessary for further concentration, this technique utilizes 70 times less energy as compared to conventional centrifugation techniques which are used to recover Schizochytrium sp. This is significant as this technique can be easily adapted to existing bioreactors, as they are already equipped with gas spargers. In addition, the presence of surfactant carried through demonstrated an improvement in the recovery of long chain poly unsaturated fatty acid (PUFA) by 6 %, particularly in DHA and DPA which was not observed in washed biomass.

Abstract:Fermentations of Xanthomonas campestris, NRRL B-1459, were carried out in a bubble column fermentor (BCF) and in a stirred tank fermentor (STF) to allow comparison of representative variables measured during the microbial growth and the gum production.The microbial growth phase was described by a logistic rate equation where maximum cell concentration was provided by nitrogenous compounds balance. The average value of the maximum specific growth rate was higher in the bubble column (μM=0.5 h−1) than in the stirred reactor (μM=0.4 h−1).The upper values of xanthan yield (Yg-x=0.65 kg xanthan/kg glucose; YO2−x xanthan/kg oxygen) and specific production rate (qx=0.26 kg xanthan/kg biomass · h) were measured when the oxygen transfer coefficient was kept up above 80 h−1 in the STF fermentor. In the bubble column the fermentation achieved in the same culture medium lasts two times longer than in the stirred aerated tank; this was attributed to the low value of the oxygen transfer coefficient (KLa =20 h−1) at the beginning of the gum synthesis phase. The results obtained in the stirred tank were the basis to estimate the optimal biomass concentration which enables to achieve a culture in non-limiting oxygen transfer conditions.Nevertheless, the transfer characteristics were more homogeneous in the bubble column than in the stirred tank where dead stagnant zones were observed. This is of primary importance when establishing fermentation kinetics models.
Optimization of Biomass and Arachidonic Acid Production by Aureispira maritima Using Response Surface Methodology

Abstract:Statistically based experimental designs, based on the Plackett–Burman protocol, were applied to the optimization of biomass and arachidonic acid (ARA) production in Aureispira maritima shake‐flask cultures. Tryptone and culture temperature were identified to have a significant effect on biomass production, whereas ARA production was only affected significantly by the pH and agitation rate. These four factors were subsequently optimized using response surface methodology. The validity of the optimum conditions was verified by separate experiments in which biomass and ARA yield were increased 4.02‐fold (2.05 g l−1) and 3.59‐fold (21.50 mg g−1), respectively, in 3‐day fermentations. Under non‐optimized culture conditions the corresponding values were 0.51 g l−1 and 5.99 mg g−1, respectively. The results suggest that A. maritima might be a potential strain for further large scale investigations to determine whether this bacterium might be suitable for commercial production of ARA. To our knowledge, this is the first report of the statistically optimization of biomass and ARA production from the marine gliding bacterium A. maritima.

Abstract:Citric acid production is generally carried out in an aqueous medium in stirred tank reactors (STR), where the solubility of oxygen is low and the oxygen demand of microbial cultures is high. Thus, for this bioprocess, providing adequate oxygen mass transfer rate (OTR) from the gas phase into the aqueous culture medium is the main challenge of bioreactor selection and operation. In this study, citric acid production by Yarrowia lipolytica W29 from crude glycerol, in batch cultures, was performed in two non-conventional bioreactors normally associated with high mass transfer efficiency: a pressurized STR and an airlift bioreactor. Increased OTR was obtained by raising the total air pressure in the pressurized STR and by increasing the aeration rate in the airlift bioreactor. An improvement of 40% in maximum citric acid titer was obtained by raising the air pressure from 1 bar to 2 bar, whereas, in the airlift bioreactor, a 30% improvement was attained by increasing the aeration rate from 1 vvm to 1.5 vvm. Both bioreactor types can be successfully applied for the citric acid production process using alternative ways of improving OTR than increasing mechanical stirring power input, thus leading to important operating saving costs.

Abstract:Previous study has demonstrated that freshwater can be replaced with seawater for dilution of feed to algal production and wastewater treatment, but high harvest cost in suspended-growth systems is still a troublesome limitation for large-scale production. Therefore, a novel inclined algal biofilm photobioreactor (IABPBR) was constructed for algal bioproduct production and treatment of seawater-diluted anaerobically digested effluent (SA) in this study. Fluffy polyester was selected as the best carrier for the algal biofilm among ten discarded materials. With the help of phytohormones, the viability of SDEC-18 was clearly enhanced and an algal biomass productivity of 5.66 g/m(2)/d was achieved. The SDEC-18 biofilm provided removal capacities of 0.65, 0.25 and 3.31 g/m(2)/d for TN, TP and COD. Phytohormones clearly enhanced the lipid biosynthesis, with an extraordinary lipid productivity of 3.98 g/m(2)/d being achieved. Moreover, an automatic harvesting system was designed for the efficient harvesting process during large-scale production.

Abstract:Laboratory scale conventional single‐use bioreactor was used to investigate the effect of different stirrer speeds on the Arthrospira platensis (Spirulina platensis) culture. Experiments were handled in two steps. First step was the selection of the stirring speeds, which was simulated via using CFD, and the second was the long term cultivation with the selected speed. During 10 days of batches as the first step, under identical culture conditions, stirrer speed of 230 rpm gave higher results, compared to 130 and 70 rpm, with respect to dry biomass weight, absorbance value (AB) and chlorophyll‐a concentration. Volumetric productivity during the growth phase of the cultures were calculated as 0.39 ± 0.03, 0.28 ± 0.01, and 0.19 ± 0.02 g L−1 d−1, from the fast to the slower speeds. According to the results a 17 day batch was handled with 230 rpm in order to monitor the effects on the culture. The culture reached a volumetric productivity of 0.33 ± 0.04 g L−1 d−1. Statistical analysis showed the significance of the parameters related with the stirring speed.

Effect of different inocula on the granulation process, reactor performance and biodiesel production of algal-bacterial granular sludge (ABGS) under low aeration conditions

Abstract:The algal-bacterial granular sludge (ABGS) system is a prospective wastewater treatment technology, but few studies focused on the effects of different inoculum types on the establishment of the ABGS system under low aeration conditions (step-decrease superficial gas velocity from 1.4 to 0.5 cm/s). Results from this study indicated that compared with other inocula, the ABGS formed by co-inoculating aerobic granular sludge (AGS) and targeted algae ( Chlorella ) exhibited a shorter granulation period (shortened by 15 days), higher total nitrogen (89.4%) and PO 4 3- -P (95.0%) removal efficiencies, and a greater yield of fatty acid methyl esters (FAMEs) (9.04 mg/g MLSS). This was possibly attributed to that the functional bacteria (e.g. Thauera , Gemmobacter and Rhodobacter ) in the inoculated AGS facilitated the ABGS granulation. The inoculated algae promoted their effective enrichment under illumination conditions and enhanced the production of extracellular polymeric substances, thus improving the stability of ABGS. The enriched algae were attached to the outer layer of the granules, which could provide sufficient oxygen for bacterial metabolism, revealing the inherent mechanisms for the good stability of ABGS under low aeration intensity. Overall, the rapid granulation of ABGS can be achieved by inoculating optimal inocula under low aeration conditions, which is convenient and economically feasible, and motivates the application of algal-bacterial consortia.

Abstract:The cost of bio-oil refining from microalgal biomass can be significantly reduced by combining extraction and transesterification. The characterisation and optimisation of the combined steps have been carried out on strains of Stichococcus bacillaris, focusing on catalyst type and concentration, reaction time and temperature, methanol/biomass ratio, pre-mixing time and water content in the biomass. The bio-oil yield has been referenced as production of fatty acid methyl esters (FAMEs). The maximum yield (∼17%) was achieved using dried biomass with alkaline catalyst at 60°C and methanol/biomass weight ratio of 79:1. Alkaline catalyst conditions gave faster reaction rates and higher bio-oil yields than acid catalyst. Yield was also strongly affected by water content in the biomass. A mechanistic interpretation has been proposed to elucidate the effect of the different operating conditions. However, the structural characteristics of the Chlorophyta cell wall can be very different, leading to different bio-oil yields when the same protocol is applied. Therefore, the optimised protocol of direct transesterification for Stichococcus bacillaris strains was tested on other Stichococcus strains and several other Chlorophyta species characterised by a different cell wall structure. It was clearly demonstrated that different results for bio-oil yield were obtained within the same microalgal species and much more within different microalgal genera.
The effect of volatile fatty acids on the growth and lipid properties of two microalgae strains during batch heterotrophic cultivation

Abstract:To overcome the bottlenecks of waste resource utilization and energy shortage that restrict the commercial production of microalgae biodiesel, volatile fatty acids (VFAs) derived from activated sludge were used as the sole carbon source to culture oleaginous microalgae Chlorella pyrenoidosa FACHB-1216 and Scenedesmus quadricauda FACHB-1297 under the mixotrophic and heterotrophic cultivation. Four VFAs ratios (acetic acids (AA): propionic acids (PA): butyric acids (BA)) were tested to determine the effects and mechanisms of the VFAs on the two microalgae. The highest lipid content (29.54%) and lipid production (71.10 mg L−1) were achieved by S. quadricauda at the VFAs ratio of 6: 1: 3 under heterotrophic condition, with 46.27% and 67.52% removal efficiencies of total nitrogen and phosphorus, respectively. The assimilation efficiency of AA was the highest at 73.37%, followed by that of PA and BA. For C. pyrenoidosa, VFAs promoted the rapid reproduction within 2 days under the heterotrophic condition at different initial inoculation densities. At the optimal VFA ratio, algae achieved the highest biomass concentration (0.14 ± 0.02 g L−1), with a specific growth rate of 0.91 d−1 and biomass productivity of 125.17 mg L−1 d−1. The removal rates of total nitrogen and phosphorus were 47.03% and 74.40%, respectively, and the assimilation efficiency of AA was the best (61.06%). High AA assimilation efficiency under the heterotrophic condition was beneficial for the algal growth and lipid accumulation. These results simultaneously produced microalgae-based bioenergy and recycled VFAs in anaerobically digested effluent.

Abstract:Abstract In order to achieve strain improvement and reduction of fermentation medium cost, atmospheric and room temperature plasma (ARTP) mutagenesis and microbial microdroplet culture (MMC) high-throughput screening were used to obtain high-yield Aurantiochytrium sp. strain. Meanwhile, low-cost food waste liquid was added into culture medium to reduce cost. The results showed that the optimal treatment time of ARTP mutagenesis for Aurantiochytrium sp. was 25 s, and 20 mutant strains were screened out by MMC to investigate their performance. The most significant increase in DHA yield was mutant Aurantiochytrium sp. MA20, whose biomass, lipid, and DHA yield were 29.0 ± 0.24, 15.8 ± 0.11, and 9.22 ± 0.37 g/L and 5.77, 16.9, and 83.2% higher than those of original strain, respectively. Compared with the original strain, the DHA proportion in total fatty acids of Aurantiochytrium sp. MA20 increased from 37.2 to 58.3%, while the DHA proportion in biomass increased from 18.4 to 31.7%. The genetic stability for Aurantiochytrium sp. MA20 showed that the yield of biomass, lipid, and DHA of the 50th generation were 29.1 ± 0.13, 16.2 ± 0.31, and 9.47 ± 0.26 g/L, respectively, which was no significant difference between original strain, indicating that the mutant MA20 could be inherited stably. Using food waste liquid to replace 100% yeast extract and 50% sea water of the fermentation medium, the DHA yield reached 9.54 g/L, which was equivalent to that of traditional medium, while the cost of medium was reduced by 45% that achieved the recycle of food waste resource.Graphical abstract

Abstract:To increase the lipid productivity and thus to reduce the production cost of microalgal biodiesel, effects of cultivation conditions including KNO3-level, CO2 concentration and irradiance on the cell growth, chlorophyll a content and lipid accumulation of Chlorella vulgaris were systematically investigated in a membrane sparged photobioreactor. The biochemical compositions including carbohydrates, proteins and lipids were analyzed simultaneously by the FT-IR spectroscopy. The results showed that the largest biomass productivity and the highest lipid content were obtained at different cultivation conditions. The algae should be harvested at a point that optimized the biomass productivity and lipid content. When the cultivation conditions were controlled at 1.0mM KNO3, 1.0% CO2 and 60μmol photons m−2s−1 at 25°C, the highest lipid productivity obtained was 40mgL−1d−1, which was about 2.5-fold that had been reported by Illman et al. (2000). The influences of cultivation conditions on the cell growth, lipid accumulation, and other biochemical compositions of cells were further discussed and illustrated by a schematic which was also useful for other microalgal species.
Optimization of Growth and Production of Docosahexaenoic Acid by Schizochytrium limacinum BR 2 . 1 . 2 Using Plackett-Burman Design and Central Composite Design with Response Surface Methodology

Abstract:Culture conditions to maximize growth and the production of docosahexaenoic acid (DHA) by Schizochytrium limacinum BR2.1.2 were optimized using two statistical experimental designs. Three critical factors—temperature, glycerol concentration and peptone concentration—for biomass and DHA production were pre-evaluated by the Plackett-Burman design using low and high levels of the three variables. Low temperature, high glycerol and high peptone concentrations signifi cantly infl uenced both determined parameters. A central composite design and response surface plots were used to optimize the proved signifi cant factors and to generate maximum biomass and DHA production. A second order polynomial equation was generated for dried cell weight (DCW) as YDCW = -83.17811 + 7.77533X1 + 22.49818X2 32.53142X3 0.12375X1X2 + 0.73850X1X3 + 5.41750X2X3 0.20343X1 2 1.95541X2 2 9.21053X3 and that for DHA as YDHA = -21.51223 + 1.32761X1 + 5.27758X2 4.69083X3 0.01150X1X2 + 0.15400X1X3 + 0.65500X2X3 0.036655X1 2 0.45401X2 2 1.51840X3 . Five levels of each factor at -1.68, -1, 0, +1 and +1.68 were tested in 17 experiments to validate both models. The P-values were less than 0.05 suggesting that the models were acceptable at this level of signifi cance. The optimal values for temperature, glycerol and peptone concentrations were 18.9 °C, 6.1% and 0.7%, respectively, and yielded 48.31 g⋅L-1 of DCW and 5.34 g⋅L-1 of DHA. Verifi cation of the optimal conditions showed that deviations of DCW and DHA from the experimental data and predicted values were 3–5% and less than 2%, respectively, suggesting that the experimental designs used in this work were effective for growth and DHA production.

The highest biomass yield (0.46 g/g substrate ) and lipid productivity (0.239 g/g substrate ) were observed with 60 g/L of glucose, following cultivation in a bioreactor, with the DHA content to be 67.76% w/w total lipids . To reduce costs, cheaper feedstocks and simultaneous production of various value-added products for pharmaceutical or energy use should be attempted. To this end, we replaced pure glucose with organosolv-pretreated spruce hydrolysate and assessed the simultaneous production of DHA and squalene from S. limacinum SR21. After the 72 h of cultivation period in bioreactor, the maximum DHA content was observed to 66.72% w/w total lipids that was corresponded to 10.15 g/L of DHA concentration. While the highest DHA productivity was 3.38 ± 0.27 g/L/d and squalene reached a total of 933.72 ± 6.53 mg/L (16.34 ± 1.81 mg/g CDW ). In summary, we show that the co-production of DHA and squalene makes S. limacinum SR21 appropriate strain for commercial-scale production of nutraceuticals.

Abstract:Rhamnolipids are biosurfactants that tend to cause strong foaming, making microbial production in an aerated stirred tank fermenter challenging. The continuous removal of rhamnolipids from the cultivation broth via in situ liquid-liquid extraction can remedy this foam challenge, and thereby supports long-term cultivation and production. However, for efficient processing and stable phase separation, a specialized apparatus is required. In this study, the novel multiphase loop reactor, which is a modified airlift reactor with an internal loop enabling continuous in situ liquid-liquid extraction, was designed and adapted to produce rhamnolipids with a recombinant bacterium, Pseudomonas putida KT2440. The initially designed multiphase loop reactor showed a low oxygen transfer rate, unable to meet the oxygen demand of the whole-cell biocatalyst, resulting in inefficient growth and production. A re-design of the sparger via 3D printing enabled a high oxygen supply allowing rhamnolipid production at key performance indicators that matched stirred-tank reactor cultivations. Advantageously, the multiphase loop reactor allowed stable and constant phase separation and solvent removal enabling continuous cultivation in the future. Concluding, the successful use of the multiphase loop reactor for rhamnolipid synthesis is presented, highlighting its potential to become a new platform technology for intensified bioprocessing.
Enhanced Production of Microalgal Metabolites Through Aeration Coupled with Stirring

Abstract:Adequate mixing is a key factor for microalgal cultivation to achieve high biomass production, so it is essential to clarify the comparative effects of different mixing methods on microalgal productivity, which has rarely been studied previously. This work therefore aimed to investigate the effects of different mixing methods (stirring, aeration, and aeration coupled with stirring) on the growth and metabolite composition of Chlorella sorokiniana SDEC-18, a strain with potential for large-scale application. The results showed that mixing was beneficial for carbohydrate accumulation, while dual mixing (aeration coupled with stirring) promoted growth and achieved the highest dry mass and metabolite productivities (including carbohydrates, proteins, and lipids) through enhancement of light energy capture in the entire system. The stirring speed in the dual mixing approach of aeration coupled with stirring was also considered: the optimal condition was found to be 800 rpm. The maximum biomass was 3.56 g L−1, and the carbohydrate productivity was as high as 119.45 mg L−1 d−1, which was the highest metabolite productivity (higher than proteins or lipids), obtained from aeration coupled with stirring at 800 rpm. Our study suggests that aeration coupled with stirring provides a feasible strategy for microalgal production, due to the optimal availability of CO2 and light achieved through effective mixing.

Abstract:Traditional autotrophic microalgae exhibit low rates of organic carbon assimilation and conversion to useful compounds when switching to mixotrophic or heterotrophic growth. The goal of this study was to investigate the effect of inorganic carbon limitation on the efficiency of organic carbon (glycerol) assimilation and conversion to total fatty acids (TFAs) or the long-chain polyunsaturated fatty acid eicosapentaenoic acid (EPA). An oleaginous Monodus subterraneus was selected and six cultivation conditions were set, including Autotrophy-no aeration, Autotrophy-aeration, Mixotrophy-no aeration, Mixotrophy-no aeration & no Na2CO3, Mixotrophy-aeration, and Heterotrophy. The results showed M. subterraneus could utilize glycerol and grow under mixotrophic condition, while it was not occurred under heterotrophy. Superiority of mixotrophy to autotrophy on biomass productivity was more obvious under inorganic carbon limitation (no aeration or no Na2CO3) than inorganic carbon supply (aeration and existing Na2CO3 in the medium). CO2 limitation (no aeration) decreased content (of dry weight) and production (in medium) of TFAs, which was not evident in mixotrophy. CO2 limitation and inorganic carbon substrate stress largely improved the COD yield of TFAs and EPA under mixotrophic condition. TFA yield (%COD) under Mixotrophy-no aeration & no Na2CO3 was maximum (22.82%), and was almost two-fold higher than that under Mixotrophy-no aeration and nearly three-fold higher than that with Mixotrophy-aeration. EPA yield (% COD) under mixotrophy-no aeration & no Na2CO3 was maximum (6.58%). These results suggested that inorganic carbon limitation is a potentially useful method to enhance conversion of organic carbon to TFAs. Furthermore, the results suggest an application to obtain high value compounds (TFAs or EPA) combined with a high assimilation rate of waste glycerol from biodiesel and epichlorohydrin production by microalgae.
