Abstract:Pure Titanium (Titanium grade 2) and Ti6Al4V (Titanium grade 5) alloys are commonly used biocompatible materials for medical bone implants and dental protheses. As some implants have to be removed after a certain amount of time after the implantation, the osteo repellent surface is one of the most important property for the removable bone implants such as bone screws and plates. On contrary, for the dental protheses the property of the most importance is osseointegration. Femtosecond laser treatment causes ablation and produces LIPSS ("laser induced periodic surface structures") which can consist of cones on macro scale and ripples on nano scale which showed structures and roughness similar to that of a bone. Therefore, surface modifications by electrochemical anodization and femtosecond laser could be seen as promising methods for improving both osteo-repellence and osteointegration properties of the bone implants and dental protheses. The aim of this work is electrochemical and surface characterization of the anodized Titanium and Ti6Al4V alloy treated by the femtosecond laser. Firstly, Titanium and Ti6Al4V samples were anodized potentiostaticaly up to 100V in 25% phosphoric acid solution for 120s. Following the potentiostatic anodization, samples were structured with femtosecond laser to produce micro cones and nano ripples on top of the cones. As a final step, laser treated spots were potentiodynamicaly anodized using a SDCM (Scanning Droplet Cell Microscopy) setup. Influence of different potentials, scan rates and electrolytes were studied and optimized, consequently. Morphology and topography were examined by the means of SEM (Scanning Electron Microscopy) and AFM (Atomic Force Microscopy), while crystallography was determined by the XRD (X-Ray Diffraction). Furthermore, the composition of the oxide layer was determined by the XPS (X-ray photoelectron spectroscopy). Oxide forming factor (k ox ), a relative permittivity ε r , and ECSA (Electrochemical surface area) were obtained from the cyclic voltammograms (CV) and electrochemical impedance spectroscopy (EIS) experimental data. Electrochemical impedance spectroscopy data was modeled and compared between not preanodized, preanodized and anodized and not anodized after femtosecond laser structuring. Finally, bioassessment with osteoblast was done to examine the growth of the osteoblast cells on the modified surface structures. Value of the oxide forming factor (k ox ) for the pure Titanium and Titanium grade 5 were successfully calculated and compared with a Scanning Electron Microscopy. Calculated values of the relative permittivity ε r are in agreement with the literature values. Finally, obtained results indicate that anodized and subsequently femtosecond laser treated samples could be used as medical bone and dental implants.
Ultrafast Laser Processing of Nanostructured Patterns for the Control of Cell Adhesion and Migration on Titanium Alloy

Abstract:Femtosecond laser texturing is a promising surface functionalization technology to improve the integration and durability of dental and orthopedic implants. Four different surface topographies were obtained on titanium-6aluminum-4vanadium plates by varying laser processing parameters and strategies: surfaces presenting nanostructures such as laser-induced periodic surface structures (LIPSS) and 'spikes', associated or not with more complex multiscale geometries combining micro-pits, nanostructures and stretches of polished areas. After sterilization by heat treatment, LIPSS and spikes were characterized to be highly hydrophobic, whereas the original polished surfaces remained hydrophilic. Human mesenchymal stem cells (hMSCs) grown on simple nanostructured surfaces were found to spread less with an increased motility (velocity, acceleration, tortuosity), while on the complex surfaces, hMSCs decreased their migration when approaching the micro-pits and preferentially positioned their nucleus inside them. Moreover, focal adhesions of hMSCs were notably located on polished zones rather than on neighboring nanostructured areas where the protein adsorption was lower. All these observations indicated that hMSCs were spatially controlled and mechanically strained by the laser-induced topographies. The nanoscale structures influence surface wettability and protein adsorption and thus influence focal adhesions formation and finally induce shape-based mechanical constraints on cells, known to promote osteogenic differentiation.
Impact of Sample Heterogeneity on Methylation Analysis

Abstract:The recent emergence of high-throughput arrays for methylation analysis has made the influence of tumor content on the interpretation of methylation levels increasingly pertinent. However, to what degree does tumor content have an influence, and what degree of tumor content makes a specimen acceptable for accurate analysis remains unclear. Taking a systematic approach, we analyzed 98 unselected formalin-fixed and paraffin-embedded gastric tumors and matched normal tissue samples using the Illumina GoldenGate methylation assay. Unsupervised hierarchical clustering showed 2 separate clusters with a significant difference in average tumor content levels. The probes identified to be significantly differentially methylated between the tumors and normals also differed according to the tumor content of the samples included, with the sensitivity of identifying the “top” candidate probes significantly reduced when including samples below 70% tumor content. We also tested whether the removal of the probes featuring single nucleotide polymorphisms and/or DNA repetitive elements, reportedly present in GoldenGate arrays, would significantly affect the study's findings, and found little change in the results with their omission. Our findings suggest that tumor content significantly influences the interpretation of methylation levels and candidate gene identification, and that 70% tumor content may be a suitable threshold for selecting samples for methylation studies.

Abstract:In this study, a femtosecond laser with a wavelength of 800 nm was used to modify the surface of a titanium alloy bone scaffold created via selective laser melting (SLM). The outcomes demonstrated that the surface morphology of the bone scaffold after femtosecond laser treatment was micro-nano morphology. The hydrophobic structure of the scaffold was changed into a super-hydrophilic structure, improving the surface roughness, which was highly helpful for osteoblast adhesion and differentiation. The femtosecond laser surface treatment in vitro samples produced a thick layer of hydroxyapatite (HAP) with improved surface bioactivity. The effectiveness of osseointegration and interstitial growth of the specimens treated with the femtosecond laser surface were found to be better when bone scaffolds were implanted into the epiphysis of the tibia of rabbits. As a result, femtosecond laser therapy dramatically enhanced the surface activity of bone scaffolds and their capacity to integrate with the surrounding bone tissues, serving as a trustworthy benchmark for future biological scaffold research.

Laser-Assisted Nanotexturing and Silver Immobilization on Titanium Implant Surfaces to Enhance Bone Cell Mineralization and Antimicrobial Properties

Abstract:Despite the great advancement and wide use of titanium (Ti) and Ti-based alloys in different orthopedic implants, device-related infections remain the major complication in modern orthopedic and trauma surgery. Most of these infections are often caused by both poor antibacterial and osteoinductive properties of the implant surface. Here, we have demonstrated a facile two-step laser nanotexturing and immobilization of silver onto the titanium implants to improve both cellular integration and antibacterial properties of Ti surfaces. The required threshold laser processing power for effective nanotexturing and osseointegration was systematically determined by the level of osteoblast cells mineralized on the laser nanotextured Ti (LN–Ti) surfaces using a neodymium-doped yttrium aluminum garnet laser (Nd:YAG, wavelength of 1.06 μm). Laser processing powers above 24 W resulted in the formation of hierarchical nanoporous structures (average pore 190 nm) on the Ti surface with a 2.5-fold increase in osseointegration as compared to the pristine Ti surface. Immobilization of silver nanoparticles onto the LN–Ti surface was conducted by dip coating in an aqueous silver ionic solution and subsequently converted to silver nanoparticles (AgNPs) by using a low power laser-assisted photocatalytic reduction process. Structural and surface morphology analysis via XRD and SEM revealed a uniform distribution of Ag and the formation of an AgTi-alloy interface on the Ti surface. The antibacterial efficacy of the LN–Ti with laser immobilized silver (LN–Ti/LI–Ag) was tested against both Gram-positive (Staphylococcus aureus) and Gram-negative (Escherichia coli) bacteria. The LN–Ti/LI–Ag surface was observed to have efficient and stable antimicrobial properties for over 6 days. In addition, it was found that the LN–Ti/LI–Ag maintained a cytocompatibility and bone cell mineralization property similar to the LN–Ti surface. Abstract:In this work Ti6Al4V alloy is surface modified by femtosecond laser ablation. The microstructure image obtained by secondary electron microscopy reveals a combination of micrometer spikes or cones superimposed by nanoripples (laser‐induced periodic surface structures). To make the surface hydrophilic anodization was performed resulting in further smoothness of microstructure and a final thickness of 35±4 nm is estimated for oxide produced after anodization at 10 V (scan rate = 0.1 V s−1) vs. standard hydrogen electrode. The obtained electrochemical active surface area (ECSA) is approximately 8 times larger as compared to flat mirror polished Ti6Al4V surface. Combined chemical analysis by Pourbaix diagram and X‐ray photoelectron spectroscopy (XPS) analyses reveal that titanium and aluminium are passivating into TiO2 and Al2O3 but the dissolution of aluminium in the form of solvated ion is inevitable. Finally, cell seeding experiments on anodized and laser treated titanium alloy samples showed that the growth of murine fibroblast cells is significantly suppressed due to unique surface texture of the laser treated and anodized titanium alloy sample.This article is protected by copyright. Abstract:This paper presents a rigorous method for calculating nonreciprocal propagation characteristics of double-layered inset dielectric guide containing a magnetized ferrite. The main features of this method are the formulation of integral equations by using Fourier transforms and the derivation of the solution by means of Galerkin's method. Numerical results for the propagation constants of the forward and backward waves of the dominant mode are presented for various values of the relative permittivity of the dielectric layer. It is found that by using large relative permittivity of the dielectric layer, this IDG structure offers very high nonreciprocity in propagation constants for the realization of efficient nonreciprocal phase shifters.
Impact of Laser Structuring on Medical-Grade Titanium: Surface Characterization and In Vitro Evaluation of Osteoblast Attachment

Abstract:Improved implant osteointegration offers meaningful potential for orthopedic, spinal, and dental implants. In this study, a laser treatment was used for the structuring of a titanium alloy (Ti6Al4V) surface combined with a titanium dioxide coating, whereby a porous surface was created. The objective was to characterize the pore structure shape, treatment-related metallographic changes, cytocompatibility, and attachment of osteoblast-like cells (MG-63). The treatment generated specific bottleneck pore shapes, offering the potential for the interlocking of osteoblasts within undercuts in the implant surface. The pore dimensions were a bottleneck diameter of 27 μm (SD: 4 μm), an inner pore width of 78 μm (SD: 6 μm), and a pore depth of 129 μm (SD: 8 μm). The introduced energy of the laser changed the metallic structure of the alloy within the heat-affected region (approximately 66 μm) without any indication of a micro cracking formation. The phase of the alloy (microcrystalline alpha + beta) was changed to a martensite alpha phase in the surface region and an alpha + beta phase in the transition region between the pores. The MG-63 cells adhered to the structured titanium surface within 30 min and grew with numerous filopodia over and into the pores over the following days. Cell viability was improved on the structured surface compared to pure titanium, indicating good cytocompatibility. In particular, the demonstrated affinity of MG-63 cells to grow into the pores offers the potential to provide significantly improved implant fixation in further in vivo studies.

Abstract:This article explores the enhancement of material surface properties of Ti6Al4V, potentially applicable to dental implants, through ultra-short pulse laser systems. This study investigates potential connections between surface wettability and biocompatibility, addressing the challenge of improving variability in material properties with specific laser treatment. Several designed microstructures were manufactured using a picosecond laser system. After that, the wettability of these structures was measured using the sessile drop method. The basic behavior and growth activity of biological cells (MG-63 cell line) on treated surfaces were also analyzed. While the conducted tests did not conclusively establish correlations between wettability and biocompatibility, the results indicated that laser treatment of Ti6Al4V could effectively enlarge the active surface to better biological cell colonization and adhesion and provide a focused moving orientation. This outcome suggests the potential application of laser treatment in producing special dental implants to mitigate the issues during and following implantation.

Abstract:Abstract This study explores the enhancement of biocompatible titanium‐based implants through surface functionalization for improved bone healing. Specifically, a near‐beta type Ti‐13Nb‐13Zr alloy is 3D printed using laser powder bed fusion and subsequently textured using nanosecond (ns) and picosecond (ps) direct laser interference patterning (DLIP) to create single‐scale and multi‐scale surface textures. On these textures, the cell behavior, morphology, metabolic activity and osteogenic differentiation potential of human bone marrow stromal cells are assessed using fluorescence microscopy and MTS assays. Moreover, tissue non‐specific alkaline phosphatase activity served as an early osteoblast production marker. Compared to untextured specimens, both types of textures exhibited higher metabolic activity and cell proliferation. Single‐scale ns‐DLIP textures encouraged cell extensions anchored in groove regions, while ps‐DLIP textures with hierarchical structures promoted cell extensions attaching to nanostructures on sidewalls. The groove width and nanotopographies in groove areas facilitated cell spreading. Surface topography, roughness, and surface chemistry (surface energy, wettability) influenced cell adhesion, proliferation, and differentiation. A comprehensive evaluation of DLIP‐generated surface textures, including their topography and chemical states, complements the factors affecting in vitro cell behavior. Overall, this research demonstrates the potential of surface‐functionalized 3Dprinted titanium for a novel generation of biocompatible implants.

Femtosecond laser upscaling strategy and biological validation for dental screws with improved osteogenic performance

Abstract:Osseointegration is one of the key conditions for long term successful dental implantation. To this end, titanium alloys undergo plethora of surface treatments able to sustain osteogenic differentiation. For these surface treatments, femtosecond laser (FSL) can generate precise and reproducible surface patterns on titanium, avoiding thermal damage and chemical pollution. We recently identify that laser-induced periodic surface structure (LIPSS) with radial orientation, generated on model (flat) titanium surface, has a high osteogenic potential. However, nano-texturing is time consuming. In the present study, we aimed to reduce the texturing time of radial LIPSS, as well as processing of large commercially available dental screws by ways of laser beam engineering. Our objectives were to maintain at least osteogenic properties demonstrated on model surfaces by adjusting laser beam diameters and to demonstrate maintenance of performance with a dental screw texturing process not exceeding 1 minute.We first textured model surfaces with radial LIPSS by laser beams of different diameters, with surface impacts of 24$\mu$m, 80$\mu$m or 180$\mu$m, named as R24, R80 or R180 respectively. Osteogenic performance of human mesenchymal stem cells (hMSCs) were compared; seeded on polished control surfaces and textured surfaces and subjected to osteogenic evaluation by cell/matrix imaging, qRT-PCR and mineral deposition quantification. All textured surfaces showed greater osteogenic potential than the control surfaces, with significantly higher efficacy on R180 surface. Therefore, R180 pattern with large beam impacts was chosen for texturing on a dental screw and its osteogenic activity was compared to that of a non-textured screw. Interestingly, R180 required only 40 seconds to be textured on whole screws, on which it preserved a high osteogenic potential. Thus, by using FSL technology, we have improved the osteogenic potential of a topographic pattern while optimizing and scaling up its processing time on a medical device.

Abstract:A mismatch in bone and implant elastic modulus can lead to aseptic loosening and ultimately implant failure. Selective elemental composition of titanium (Ti) alloys coupled with surface treatment can be used to improve osseointegration and reduce bacterial adhesion. The biocompatibility and antibacterial properties of Ti-35Nb-7Zr-6Ta (TNZT) using fibre laser surface treatment were assessed in this work, due to its excellent material properties (low Young’s modulus and non-toxicity) and the promising attributes of fibre laser treatment (very fast, non-contact, clean and only causes changes in surface without altering the bulk composition/microstructure). The TNZT surfaces in this study were treated in a high speed regime, specifically 100 and 200 mm/s, (or 6 and 12 m/min). Surface roughness and topography (WLI and SEM), chemical composition (SEM-EDX), microstructure (XRD) and chemistry (XPS) were investigated. The biocompatibility of the laser treated surfaces was evaluated using mesenchymal stem cells (MSCs) cultured in vitro at various time points to assess cell attachment (6, 24 and 48 h), proliferation (3, 7 and 14 days) and differentiation (7, 14 and 21 days). Antibacterial performance was also evaluated using Staphylococcus aureus (S. aureus) and Live/Dead staining. Sample groups included untreated base metal (BM), laser treated at 100 mm/s (LT100) and 200 mm/s (LT200). The results demonstrated that laser surface treatment creates a rougher (Ra value of BM is 199 nm, LT100 is 256 nm and LT200 is 232 nm), spiky surface (Rsk > 0 and Rku > 3) with homogenous elemental distribution and decreasing peak-to-peak distance between ripples (0.63 to 0.315 μm) as the scanning speed increases (p < 0.05), generating a surface with distinct micron and nano scale features. The improvement in cell spreading, formation of bone-like nodules (only seen on the laser treated samples) and subsequent four-fold reduction in bacterial attachment (p < 0.001) can be attributed to the features created through fibre laser treatment, making it an excellent choice for load bearing implant applications. Last but not least, the presence of TiN in the outermost surface oxide might also account for the improved biocompatibility and antibacterial performances of TNZT.

Abstract:Implant surface topography plays an important role in determining cell responses, including attachment, proliferation, migration, and differentiation. With rapid developments in micro-and nano-fabrication methods, precisely controlled patterns can be generated on implant surfaces. Osteon-mimetic structures are of great interest as new surface designs on bone implants for creating optimal conditions for bone tissue growth. In this work, we used a laser ablation technique to create a biomimetic relief on Ti-6Al-4 V supports. The osteon-mimetic reliefs exhibited patterns of concentric lamellae-like rings of different width and periodicity. A distinctive feature of these reliefs is the presence of a highly porous nanocoating of titanium oxides, formed as a result of reverse deposition from the vapor-gas phase during ablation. Our investigation of the response of human mesenchymal stem cells revealed that the osteon-mimetic microrelief topography significantly influenced their adhesion, proliferation, and polarization. The presence of the nanocoating further impacted the number and localization of their focal adhesions. The outcomes of this work demonstrate an innovative direction towards the design of osteon-mimetic topographies on implantable materials. This work also holds the potential to identify optimal combinations of microrelief and nanocoating parameters to elicit specific cellular responses.

Abstract:The cellular infiltrate in mycosis fungoides lymphoma was evaluated by a quantitative morphologic technique prior to, during, and after therapy with various antilymphoma agents. Prior to therapy plaques of disease differed from tumors in having fewer total cells and fewer atypical cells per unit field. Both types of lesions responded very well to each treatment modality employed with clinical regression occurring in 30 of 32 areas. After therapy there was a marked reduction in the total number of cells and of atypical cells. The number of lymphocytes remained constant. These changes are discussed with reference to their significance to the course of the disease.
Building Orientation and Post Processing of Ti6Al4V Produced by Laser Powder Bed Fusion Process

Abstract:Laser powder bed fusion, particularly the selective laser melting (SLM), is an additive manufacturing (AM) technology used to produce near-net-shaped engineering components for biomedical applications, especially in orthopaedics. Ti6Al4V is commonly used for producing orthopaedic implants using SLM because it has excellent mechanical qualities, a high level of biocompatibility, and corrosion resistance. However, the main problems associated with this process are the result of its surface properties: it has to be able to promote cell attachment but, at the same time, avoid bacteria colonization. Surface modification is used as a post-processing technique to provide items the unique qualities that can improve their functionality and performance in particular working conditions. The goal of this work was to produce and analyse Ti6Al4V samples fabricated by SLM with different building directions in relation to the building plate (0° and 45°) and post-processed by anodization and passivation. The results demonstrate how the production and post processes had an impact on osteoblast attachment, mineralization, and osseointegration over an extended period of time. Though the anodization treatment result was cytotoxic, the biocompatibility of as-built specimens and specimens after passivation treatment was confirmed. In addition, it was discovered that effective post-processing increases the mineralization of these types of 3D-printed surfaces.

Abstract:In a double-blind, randomized, crossover study performed in atopic subjects, the inhibitory effects of single doses of 10 mg cetirizine and 10 mg loratadine on histamine- and grass pollen-induced skin reactions were evaluated 4 h after drug intake. Cetirizine significantly inhibited histamine- and antigen-induced skin reactions, as well as the accumulation of eosinophils measured 24 h after antigen challenge. Loratadine, however, did not significantly inhibit the skin reactions induced by histamine and grass pollen, nor eosinophil accumulation.
Titanium Surfaces with a Laser-Produced Microchannel Structure Enhance Pre-Osteoblast Proliferation, Maturation, and Extracellular Mineralization In Vitro

Abstract:The clinical success of dental titanium implants is profoundly linked to implant stability and osseointegration, which comprises pre-osteoblast proliferation, osteogenic differentiation, and extracellular mineralization. Because of the bio-inert nature of titanium, surface processing using subtractive or additive methods enhances osseointegration ability but limits the benefit due to accompanying surface contamination. By contrast, laser processing methods increase the roughness of the implant surface without contamination. However, the effects of laser-mediated distinct surface structures on the osteointegration level of osteoblasts are controversial. The role of a titanium surface with a laser-mediated microchannel structure in pre-osteoblast maturation remains unclear. This study aimed to elucidate the effect of laser-produced microchannels on pre-osteoblast maturation. Pre-osteoblast human embryonic palatal mesenchymal cells were seeded on a titanium plate treated with grinding (G), sandblasting with large grit and acid etching (SLA), or laser irradiation (L) for 3–18 days. The proliferation and morphology of pre-osteoblasts were evaluated using a Trypan Blue dye exclusion test and fluorescence microscopy. The mRNA expression, protein expression, and protein secretion of osteogenic differentiation markers in pre-osteoblasts were evaluated using reverse transcriptase quantitative polymerase chain reaction, a Western blot assay, and a multiplex assay, respectively. The extracellular calcium precipitation of pre-osteoblast was measured using Alizarin red S staining. Compared to G- and SLA-treated titanium surfaces, the laser-produced microchannel surfaces enhanced pre-osteoblast proliferation, the expression/secretion of osteogenic differentiation markers, and extracellular calcium precipitation. Laser-treated titanium implants may enhance the pre-osteoblast maturation process and provide extra benefits in clinical application.

Abstract:Surface design plays a critical role in determining the integration of dental implants with bone tissue. Femtosecond laser‐texturing has emerged as a breakthrough technology offering excellent uniformity and reproducibility in implant surface features. However, when compared to state‐of‐the‐art sandblasted and acid‐etched surfaces, laser‐textured surface designs typically underperform in terms of osseointegration. This study investigated the capacity of a bio‐inspired femtosecond laser‐textured surface design to enhance osseointegration compared to state‐of‐the‐art sandblasted & acid‐etched surfaces. Laser‐texturing facilitates the production of an organized trabeculae‐like microarchitecture with superimposed nano‐scale laser‐induced periodic surface structures on both 2D and 3D samples of titanium‐zirconium‐alloy. Following a boiling treatment to modify the surface chemistry, improving wettability to a contact angle of 10°, laser‐textured surfaces enhance fibrin network formation when in contact with human whole blood, comparable to state‐of‐the‐art surfaces. In vitro experiments demonstrate that laser‐textured surfaces significantly outperform state‐of‐the‐art surfaces with a 2.5‐fold higher level of mineralization by bone progenitor cells after 28 days of culture. Furthermore, in vivo evaluations reveal superior biomechanical integration of laser‐textured surfaces after 28 days of implantation. Notably, during abiological pull‐out tests, laser‐textured surfaces exhibit comparable performance, suggesting that the observed enhanced osseointegration is primarily driven by the biological response to the surface. Abstract:Bone resorption and inadequate osseointegration are considered the main problems of titanium implants. In this investigation, the texture and surface roughness of porous titanium samples obtained by the space holder technique were modified with a femtosecond Yb-doped fiber laser. Different percentages of porosity (30, 40, 50, and 60 vol.%) and particle range size (100-200 and 355-500 μm) were compared with fully-dense samples obtained by conventional powder metallurgy. After femtosecond laser treatment the formation of a rough surface with micro-columns and micro-holes occurred for all the studied substrates. The surface was covered by ripples over the micro-metric structures. This work evaluates both the influence of the macro-pores inherent to the spacer particles, as well as the micro-columns and the texture generated with the laser, on the wettability of the surface, the cell behavior (adhesion and proliferation of osteoblasts), micro-hardness (instrumented micro-indentation test, P-h curves) and scratch resistance. The titanium sample with 30 vol.% and a pore range size of 100-200 μm was the best candidate for the replacement of small damaged cortical bone tissues, based on its better biomechanical (stiffness and yield strength) and biofunctional balance (bone in-growth and in vitro osseointegration).
