Abstract:Existing studies have shown the benefit of laser surface texturing (LST) in promoting the cytocompatibility of traditional metallic biomaterials. Researchers have also reported the potential of bulk metallic glasses (BMGs) as an alternative class of material for biomedical applications. However, investigations specifically focussed on studying the cytocompatibility of BMG surfaces processed with LST are still lacking. The present work demon-strated the feasibility of nanosecond LST as a method to modify the cytocompatibility of a Zr-based BMG material known as Vitreloy 105. Two different types of laser-induced surface patterns, namely grooves and dimples, were considered. Their respective influence on the resulting cell viability, attachment and morphology was studied and compared against the cytocompatibility of the original BMG surface. It was found that MG63 osteoblast-like cells on the groove-textured surface exhibited higher viability and better adhesion compared to those on the original and dimple-textured surfaces. Possible underlying mechanisms associated with LST, which can affect the in vitro cytocompatibility of Vitreloy 105 were discussed based on the induced changes to surface chemistry, wettability and roughness. It is suggested that the higher surface roughness, increased presence of metallic oxides and enhanced hydrophilicity of the groove-textured sample were the main contributors to its improved cytocompatibility.

Abstract:Abstract This study explores the enhancement of biocompatible titanium‐based implants through surface functionalization for improved bone healing. Specifically, a near‐beta type Ti‐13Nb‐13Zr alloy is 3D printed using laser powder bed fusion and subsequently textured using nanosecond (ns) and picosecond (ps) direct laser interference patterning (DLIP) to create single‐scale and multi‐scale surface textures. On these textures, the cell behavior, morphology, metabolic activity and osteogenic differentiation potential of human bone marrow stromal cells are assessed using fluorescence microscopy and MTS assays. Moreover, tissue non‐specific alkaline phosphatase activity served as an early osteoblast production marker. Compared to untextured specimens, both types of textures exhibited higher metabolic activity and cell proliferation. Single‐scale ns‐DLIP textures encouraged cell extensions anchored in groove regions, while ps‐DLIP textures with hierarchical structures promoted cell extensions attaching to nanostructures on sidewalls. The groove width and nanotopographies in groove areas facilitated cell spreading. Surface topography, roughness, and surface chemistry (surface energy, wettability) influenced cell adhesion, proliferation, and differentiation. A comprehensive evaluation of DLIP‐generated surface textures, including their topography and chemical states, complements the factors affecting in vitro cell behavior. Overall, this research demonstrates the potential of surface‐functionalized 3Dprinted titanium for a novel generation of biocompatible implants.

Abstract:This work aims to provide a topographical and biological characterization of Additively Manufactured (AMed) Ti6Al4V ELI surfaces subjected to different surface finishing techniques. Unlike traditional biomedical sandblasting methods, femtosecond laser texturing generates new surface roughness and morphologies, which show promise for enhancing the differentiation and proliferation of human bone cells. This work lays the groundwork for optimizing the surfaces of AMed medical devices. Among Additive Manufacturing (AM) technologies, Laser Powder Bed Fusion (LPBF) has made a great contribution to optimizing the production of customized implant materials. However, the design of the ideal surface topography, capable of exerting the best biological effect without drawbacks, is still a subject of study. The aim of the present study is to topographically and biologically characterize AM‐produced Ti6Al4V ELI (Extra Low Interstitial) samples by comparing different surface finishing. Vertically and horizontally samples are realized by LPBF with four surface finishing conditions (as‐built, corundum‐sandblasted, zirconia‐sandblasted, femtosecond laser textured). Bioactivity in vitro tests are performed with human osteoblasts evaluating morphology, metabolic activity, and differentiation capabilities in direct contact with surfaces. Scanning electron microscope and profilometry analysis are used to evaluate surface morphology and samples' roughness with and without cells. All tested surfaces show good biocompatibility. The influence of material surface features is evident in the early evaluation, with the most promising results of morphological study for laser texturing. Deposition orientations seem to influence metabolic activities, with XZ orientation more effective than XY. Current data provide the first positive feedback on the biocompatibility of laser texturing finishing, still poorly described in the literature, and support the future clinical development of devices produced with a combination of LPBF and different finishing treatments.

Impact of Sample Heterogeneity on Methylation Analysis

Abstract:The recent emergence of high-throughput arrays for methylation analysis has made the influence of tumor content on the interpretation of methylation levels increasingly pertinent. However, to what degree does tumor content have an influence, and what degree of tumor content makes a specimen acceptable for accurate analysis remains unclear. Taking a systematic approach, we analyzed 98 unselected formalin-fixed and paraffin-embedded gastric tumors and matched normal tissue samples using the Illumina GoldenGate methylation assay. Unsupervised hierarchical clustering showed 2 separate clusters with a significant difference in average tumor content levels. The probes identified to be significantly differentially methylated between the tumors and normals also differed according to the tumor content of the samples included, with the sensitivity of identifying the “top” candidate probes significantly reduced when including samples below 70% tumor content. We also tested whether the removal of the probes featuring single nucleotide polymorphisms and/or DNA repetitive elements, reportedly present in GoldenGate arrays, would significantly affect the study's findings, and found little change in the results with their omission. Our findings suggest that tumor content significantly influences the interpretation of methylation levels and candidate gene identification, and that 70% tumor content may be a suitable threshold for selecting samples for methylation studies.

Abstract:In a double-blind, randomized, crossover study performed in atopic subjects, the inhibitory effects of single doses of 10 mg cetirizine and 10 mg loratadine on histamine- and grass pollen-induced skin reactions were evaluated 4 h after drug intake. Cetirizine significantly inhibited histamine- and antigen-induced skin reactions, as well as the accumulation of eosinophils measured 24 h after antigen challenge. Loratadine, however, did not significantly inhibit the skin reactions induced by histamine and grass pollen, nor eosinophil accumulation.
Magnesium sulphate infusion suppresses the cardiac release of noradrenaline during a handgrip stress test.

Abstract:BACKGROUND Magnesium has several important cardiovascular effects, but its effect on cardiac sympathetic efferent neuron activity has not been clarified. OBJECTIVES To examine the effect of magnesium sulphate infusion on cardiac sympathetic efferent postganglionic neuronal liberation of noradrenaline. PATIENTS AND METHODS Twenty-two patients who underwent cardiac catheterization were randomly allocated to the control group or the magnesium group. Plasma noradrenaline and adrenaline concentrations in the aorta and the coronary sinus were measured. Noradrenaline or adrenaline release from the heart was calculated by dividing the difference in noradrenaline or adrenaline concentration between the aorta and the coronary sinus by that of the aorta. After baseline blood sampling, the control patients and the patients in the magnesium group received intravenous infusion of saline or magnesium sulphate (10 mmol). All patients were then subjected to 3 min of handgrip exercise stress test to augment sympathetic efferent neuronal activity, and the blood sampling was repeated. RESULTS Although blood pressure was increased by the handgrip stress test, there were no differences in heart rate and blood pressure between the two groups, both at baseline and during the handgrip stress test. The plasma noradrenaline and adrenaline concentrations and noradrenaline or adrenaline release from the heart did not differ between the two groups in the baseline condition. However, the handgrip stress increased plasma noradrenaline concentrations and the cardiac noradrenaline release was increased in the control group, whereas the cardiac noradrenaline release was not increased by the handgrip stress in the magnesium group (P<0.02). CONCLUSIONS These data indicate that magnesium sulphate infusion suppresses the release of catecholamines by the heart, which is an indirect index of sympathetic efferent neuronal activity.
Ultrafast Laser Processing of Nanostructured Patterns for the Control of Cell Adhesion and Migration on Titanium Alloy

Abstract:Femtosecond laser texturing is a promising surface functionalization technology to improve the integration and durability of dental and orthopedic implants. Four different surface topographies were obtained on titanium-6aluminum-4vanadium plates by varying laser processing parameters and strategies: surfaces presenting nanostructures such as laser-induced periodic surface structures (LIPSS) and 'spikes', associated or not with more complex multiscale geometries combining micro-pits, nanostructures and stretches of polished areas. After sterilization by heat treatment, LIPSS and spikes were characterized to be highly hydrophobic, whereas the original polished surfaces remained hydrophilic. Human mesenchymal stem cells (hMSCs) grown on simple nanostructured surfaces were found to spread less with an increased motility (velocity, acceleration, tortuosity), while on the complex surfaces, hMSCs decreased their migration when approaching the micro-pits and preferentially positioned their nucleus inside them. Moreover, focal adhesions of hMSCs were notably located on polished zones rather than on neighboring nanostructured areas where the protein adsorption was lower. All these observations indicated that hMSCs were spatially controlled and mechanically strained by the laser-induced topographies. The nanoscale structures influence surface wettability and protein adsorption and thus influence focal adhesions formation and finally induce shape-based mechanical constraints on cells, known to promote osteogenic differentiation.

Abstract:Surface design plays a critical role in determining the integration of dental implants with bone tissue. Femtosecond laser‐texturing has emerged as a breakthrough technology offering excellent uniformity and reproducibility in implant surface features. However, when compared to state‐of‐the‐art sandblasted and acid‐etched surfaces, laser‐textured surface designs typically underperform in terms of osseointegration. This study investigated the capacity of a bio‐inspired femtosecond laser‐textured surface design to enhance osseointegration compared to state‐of‐the‐art sandblasted & acid‐etched surfaces. Laser‐texturing facilitates the production of an organized trabeculae‐like microarchitecture with superimposed nano‐scale laser‐induced periodic surface structures on both 2D and 3D samples of titanium‐zirconium‐alloy. Following a boiling treatment to modify the surface chemistry, improving wettability to a contact angle of 10°, laser‐textured surfaces enhance fibrin network formation when in contact with human whole blood, comparable to state‐of‐the‐art surfaces. In vitro experiments demonstrate that laser‐textured surfaces significantly outperform state‐of‐the‐art surfaces with a 2.5‐fold higher level of mineralization by bone progenitor cells after 28 days of culture. Furthermore, in vivo evaluations reveal superior biomechanical integration of laser‐textured surfaces after 28 days of implantation. Notably, during abiological pull‐out tests, laser‐textured surfaces exhibit comparable performance, suggesting that the observed enhanced osseointegration is primarily driven by the biological response to the surface. Abstract:Implant surface topography plays an important role in determining cell responses, including attachment, proliferation, migration, and differentiation. With rapid developments in micro-and nano-fabrication methods, precisely controlled patterns can be generated on implant surfaces. Osteon-mimetic structures are of great interest as new surface designs on bone implants for creating optimal conditions for bone tissue growth. In this work, we used a laser ablation technique to create a biomimetic relief on Ti-6Al-4 V supports. The osteon-mimetic reliefs exhibited patterns of concentric lamellae-like rings of different width and periodicity. A distinctive feature of these reliefs is the presence of a highly porous nanocoating of titanium oxides, formed as a result of reverse deposition from the vapor-gas phase during ablation. Our investigation of the response of human mesenchymal stem cells revealed that the osteon-mimetic microrelief topography significantly influenced their adhesion, proliferation, and polarization. The presence of the nanocoating further impacted the number and localization of their focal adhesions. The outcomes of this work demonstrate an innovative direction towards the design of osteon-mimetic topographies on implantable materials. This work also holds the potential to identify optimal combinations of microrelief and nanocoating parameters to elicit specific cellular responses.

Abstract:The material and surface morphology of bone implants can influence cellular activity and proliferation properties. The preparation of biomimetic microtextures on the hydroxyapatite (HA) bio-coated surface can enhance the biological properties of the implant. In this study, a nanosecond laser was used to prepare microtextures on the surface of HA coatings, and the effect of laser parameters on the ablation morphology was investigated by micromorphological observation and response surface methods. The ablation morphology was found to be better when the laser repetition frequency was 20 kHZ, the scanning speed was 300 mm/s and the number of scans was 7. Microtextures imitating fish scales and crocodile skin of different sizes were prepared on the surface of HA coatings using this laser parameter. The biological properties of the microtextures were investigated using CCK-8 tests and cell adhesion tests and the nanosecond laser-prepared mimetic microtextures were found to promote cell proliferation. A 0.1 mm radius of the mock fish scale microtexture had optimal cellular activity, where the tentacles of the cells could fully extend to the surface of the microstructure, which facilitated cell adhesion and proliferation.

Abstract:This article explores the enhancement of material surface properties of Ti6Al4V, potentially applicable to dental implants, through ultra-short pulse laser systems. This study investigates potential connections between surface wettability and biocompatibility, addressing the challenge of improving variability in material properties with specific laser treatment. Several designed microstructures were manufactured using a picosecond laser system. After that, the wettability of these structures was measured using the sessile drop method. The basic behavior and growth activity of biological cells (MG-63 cell line) on treated surfaces were also analyzed. While the conducted tests did not conclusively establish correlations between wettability and biocompatibility, the results indicated that laser treatment of Ti6Al4V could effectively enlarge the active surface to better biological cell colonization and adhesion and provide a focused moving orientation. This outcome suggests the potential application of laser treatment in producing special dental implants to mitigate the issues during and following implantation.

Abstract:By means of a silver staining method, Nucleolar Organizer Region-associated proteins (NORs) have been evaluated on paraffin sections of a series of 58 ultrasound-guided liver biopsy specimens. These included 12 normal livers, 12 cirrhotic livers, 12 cases of chronic hepatitis and 22 cases of hepatocellular carcinoma. A significant difference (P less than 0.001) was found between the mean AgNOR scores of the normal and pathological biopsies, and between the non-neoplastic and the carcinomatous lesions. The authors suggest that AgNOR counts, in combination with conventional histocytological criteria, may be a useful method in the diagnosis of liver diseases.
Femtosecond laser upscaling strategy and biological validation for dental screws with improved osteogenic performance

Abstract:Osseointegration is one of the key conditions for long term successful dental implantation. To this end, titanium alloys undergo plethora of surface treatments able to sustain osteogenic differentiation. For these surface treatments, femtosecond laser (FSL) can generate precise and reproducible surface patterns on titanium, avoiding thermal damage and chemical pollution. We recently identify that laser-induced periodic surface structure (LIPSS) with radial orientation, generated on model (flat) titanium surface, has a high osteogenic potential. However, nano-texturing is time consuming. In the present study, we aimed to reduce the texturing time of radial LIPSS, as well as processing of large commercially available dental screws by ways of laser beam engineering. Our objectives were to maintain at least osteogenic properties demonstrated on model surfaces by adjusting laser beam diameters and to demonstrate maintenance of performance with a dental screw texturing process not exceeding 1 minute.We first textured model surfaces with radial LIPSS by laser beams of different diameters, with surface impacts of 24$\mu$m, 80$\mu$m or 180$\mu$m, named as R24, R80 or R180 respectively. Osteogenic performance of human mesenchymal stem cells (hMSCs) were compared; seeded on polished control surfaces and textured surfaces and subjected to osteogenic evaluation by cell/matrix imaging, qRT-PCR and mineral deposition quantification. All textured surfaces showed greater osteogenic potential than the control surfaces, with significantly higher efficacy on R180 surface. Therefore, R180 pattern with large beam impacts was chosen for texturing on a dental screw and its osteogenic activity was compared to that of a non-textured screw. Interestingly, R180 required only 40 seconds to be textured on whole screws, on which it preserved a high osteogenic potential. Thus, by using FSL technology, we have improved the osteogenic potential of a topographic pattern while optimizing and scaling up its processing time on a medical device.

Enhancing Bactericidal Properties of Ti6Al4V Surfaces through Micro and Nano Hierarchical Laser Texturing

Abstract:Orthopedic and dental implants made from Ti6Al4V are widely used due to their excellent mechanical properties and biocompatibility. However, the long-term performance of these implants can be compromised by bacterial infections. This study explores the development of hierarchically textured surfaces with enhanced bactericidal properties to address such challenges. Hierarchical surface structures were developed by combining microscale features produced by a microsecond laser and superimposed submicron features produced using a femtosecond laser. Microscale patterns were produced by the pulsed laser surface melting process, whereas submicrometer laser-induced periodic surface structures were created on top of them by femtosecond laser processing. Escherichia coli bacterial cells were cultured on the textured surface. After 24 h, a staining analysis was performed using SYTO9 and PI dyes to investigate the samples with a confocal microscope for live dead assays. Results showed bacterial colony formation onto the microscale surface textures with live bacterial cells, whereas the hierarchical surface textures display segregated and physically damaged bacterial cell attachments on surfaces. The hierarchical surface textures showed ∼98% dead bacterial cells due to the combined effect of its multiscale surface features and oxide formation during the laser processing steps. The efficacy of hierarchical surface textures in enhancing the antibacterial behavior of Ti6Al4V implants is evident from the conducted research. Such laser-based surface treatments can find potential applications in different industrial sectors.

Abstract:Abstract Current research on surface modifications has yielded advanced implant biomaterials. Various implant surface modifications have been shown to be promising in improving bone target cell response, but more comprehensive studies whether certain implant surface modifications can directly target cell behavioural features such as morphogenesis and proliferation are needed. Here, we studied the response of primary alveolar bone cells on various implant surface modifications in terms of osteoblast morphology and proliferation in vitro. Analyses of surface modifications led to surface-related test parameters including the topographical parameters micro-roughness, texture aspect and surface enlargement as well as the physicochemical parameter surface wettability. We compared osteoblast morphology and proliferation towards the above-mentioned parameters and found that texture aspect and surface enlargement but not surface roughness or wettability exhibited significant impact on osteoblast morphology and proliferation. Detailed analysis revealed osteoblast proliferation as a function of cell morphology, substantiated by an osteoblast size- and morphology-dependent increase in mitotic activity. These findings show that implant surface topography controls cell behavioural morphology and subsequently cell proliferation, thereby opening the road for cell instructive biomaterials.

Novel hydrophilic nanostructured microtexture on direct metal laser sintered Ti-6Al-4V surfaces enhances osteoblast response in vitro and osseointegration in a rabbit model

Abstract:The purpose of this study was to compare the biological effects in vivo of hierarchical surface roughness on laser sintered titanium-aluminum-vanadium (Ti-6Al-4V) implants to those of conventionally machined implants on osteoblast response in vitro and osseointegration. Laser sintered disks were fabricated to have micro-/nano-roughness and wettability. Control disks were computer numerical control (CNC) milled and then polished to be smooth (CNC-M). Laser sintered disks were polished smooth (LST-M), grit blasted (LST-B), or blasted and acid etched (LST-BE). LST-BE implants or implants manufactured by CNC milling and grit blasted (CNC-B) were implanted in the femurs of male New Zealand white rabbits. Most osteoblast differentiation markers and local factors were enhanced on rough LST-B and LST-BE surfaces in comparison to smooth CNC-M or LST-M surfaces for MG63 and normal human osteoblast cells. To determine if LST-BE implants were osteogenic in vivo, we compared them to implant surfaces used clinically. LST-BE implants had a unique surface with combined micro-/nano-roughness and higher wettability than conventional CNC-B implants. Histomorphometric analysis demonstrated a significant improvement in cortical bone-implant contact of LST-BE implants compared to CNC-B implants after 3 and 6 weeks. However, mechanical testing revealed no differences between implant pullout forces at those time points. LST surfaces enhanced osteoblast differentiation and production of local factors in vitro and improved the osseointegration process in vivo. © 2016 Wiley Periodicals, Inc. Agreement among four homocysteine assays and results in patients with coronary atherosclerosis and controls.

Abstract:BACKGROUND Hyperhomocysteinemia has been associated with coronary atherosclerosis in many, but not all, prospective and retrospective studies. Some on these inconsistencies may be attributed to methodological variabilities. METHODS In the present study, three newly commercially available assays and one in-house HPLC assay for total homocysteine (tHcy) were utilized in 99 subjects with angiographically documented atherosclerosis and in 91 community controls matched by age, gender, and smoking history. The in-house assay, a modified Fortin and Genest HPLC method, was compared with the Bio-Rad HPLC assay, the Abbott IMx((R)) fluorescence polarization immunoassay, and a Bio-Rad enzyme-linked immunoassay (EIA) microtiter method. RESULTS Correlation coefficient values between the in-house HPLC assay and the Bio-Rad HPLC, the Abbott IMx, and the Bio-Rad EIA assays were 0.95, 0.96 and 0.90, respectively. Although tHcy concentrations were higher in cases compared with controls by all four methods, the difference reached statistical significance only with the in-house HPLC procedure (median, 13.5 +/- 6.7 micromol/L in cases vs 10.9 +/- 4.8 micromol/L in controls; P <0. 01, adjusting for covariates), where it was an independent predictor of case or control status, along with hypertension, total cholesterol, and triglycerides. The tHcy distributions in cases and controls demonstrated significant overlap. The number of atherosclerotic major coronary vessels was associated with significantly higher tHcy (P <0.01 for trend) in all four methods. CONCLUSIONS The three commercial assays for tHcy differed in analytical and clinical performance. Analytically, the Abbott IMx method showed the best comparability with the in-house assay, but clinically, the three commercial methods were similar and did not distinguish cases from controls.

Abstract:The cellular infiltrate in mycosis fungoides lymphoma was evaluated by a quantitative morphologic technique prior to, during, and after therapy with various antilymphoma agents. Prior to therapy plaques of disease differed from tumors in having fewer total cells and fewer atypical cells per unit field. Both types of lesions responded very well to each treatment modality employed with clinical regression occurring in 30 of 32 areas. After therapy there was a marked reduction in the total number of cells and of atypical cells. The number of lymphocytes remained constant. These changes are discussed with reference to their significance to the course of the disease.

Abstract:A mismatch in bone and implant elastic modulus can lead to aseptic loosening and ultimately implant failure. Selective elemental composition of titanium (Ti) alloys coupled with surface treatment can be used to improve osseointegration and reduce bacterial adhesion. The biocompatibility and antibacterial properties of Ti-35Nb-7Zr-6Ta (TNZT) using fibre laser surface treatment were assessed in this work, due to its excellent material properties (low Young’s modulus and non-toxicity) and the promising attributes of fibre laser treatment (very fast, non-contact, clean and only causes changes in surface without altering the bulk composition/microstructure). The TNZT surfaces in this study were treated in a high speed regime, specifically 100 and 200 mm/s, (or 6 and 12 m/min). Surface roughness and topography (WLI and SEM), chemical composition (SEM-EDX), microstructure (XRD) and chemistry (XPS) were investigated. The biocompatibility of the laser treated surfaces was evaluated using mesenchymal stem cells (MSCs) cultured in vitro at various time points to assess cell attachment (6, 24 and 48 h), proliferation (3, 7 and 14 days) and differentiation (7, 14 and 21 days). Antibacterial performance was also evaluated using Staphylococcus aureus (S. aureus) and Live/Dead staining. Sample groups included untreated base metal (BM), laser treated at 100 mm/s (LT100) and 200 mm/s (LT200). The results demonstrated that laser surface treatment creates a rougher (Ra value of BM is 199 nm, LT100 is 256 nm and LT200 is 232 nm), spiky surface (Rsk > 0 and Rku > 3) with homogenous elemental distribution and decreasing peak-to-peak distance between ripples (0.63 to 0.315 μm) as the scanning speed increases (p < 0.05), generating a surface with distinct micron and nano scale features. The improvement in cell spreading, formation of bone-like nodules (only seen on the laser treated samples) and subsequent four-fold reduction in bacterial attachment (p < 0.001) can be attributed to the features created through fibre laser treatment, making it an excellent choice for load bearing implant applications. Last but not least, the presence of TiN in the outermost surface oxide might also account for the improved biocompatibility and antibacterial performances of TNZT.
