Abstract:This manuscript reports a systematic study of encapsulation of fish oil into soybean microparticles by emulsification and subsequent spray drying in order to protect it from lipid autoxidation and/or facilitate its handling for incorporation into healthy food products. In particular, the effect of the formulation and the emulsification technique on the physicochemical properties of the resulting emulsions and encapsulates was evaluated. Two emulsifying processes and four protein:oil ratios were analyzed. Soy proteins managed to encapsulate the fish oil, masking its characteristic odor and its oily texture into matrix type microcapsules, as was observed by confocal microscopy. The emulsification process was determinant in the efficiency of drying and encapsulation as well as in the protection exerted on the oil oxidative stability. Although both, emulsifying and drying processes caused certain initial oil oxidation (verified by peroxide and TBA indexes), some of the studied systems showed a good perspective of oxidative stability over time (studied by accelerated rancimat test).

Abstract:The objectives of this study were to investigate the properties and stability of microcapsules containing fish oil co-encapsulated with phytosterol ester and limonene, prepared by spray-drying and freeze-drying methods. Whey protein isolate and soluble corn fiber were used as wall materials in the encapsulation process. The properties of microcapsules, including structure, glass transition, volatile/non-volatiles retention, microencapsulation efficiency, oxidation stability, color measurement, and sensory profiles, were evaluated after drying and during a seven-day accelerated storage trial. The finding reveals that drying methods have an effect on the retention of volatile fraction and the physical structure of the wall matrix consisted of WPI and SCF, consequently influencing the storage stability of the powders. Significantly higher retention of volatile fraction (p < 0.05) and lower surface oil were found in the spray-dried samples, resulting in the higher microencapsulation efficiency. However, samples dehydrated by both methods have good redispersion properties, showing no statistical significance (p > 0.05). The oxidation of the encapsulated oils was comparable for both spray- and freeze-dried samples during the seven-day accelerated storage trial but the loss of limonene flavor was significantly higher in the freeze-dried samples (p < 0.05). Sensory evaluation indicated that the addition of limonene could mask the unpleasant fishy odor in the co-encapsulated microcapsules. Overall, freeze drying did not produce powders with superior properties and did not show better protection towards the core materials than spray drying.

Abstract:Various microencapsulation techniques can result in significant differences in the properties of dried microcapsules. Microencapsulation is an effective approach to improve fish oil properties, including oxidisability and unpleasant flavour. In this study, β-carotene, lutein, zeaxanthin, and fish oil were co-encapsulated by microfluidic-jet spray drying (MFJSD), two-fluid nozzle spray drying (SD), and freeze-drying (FD), respectively. The aim of the current study is to understand the effect of different drying techniques on microcapsule properties. Whey protein isolate (WPI) and octenylsuccinic anhydride (OSA) modified starch were used as wall matrices in this study for encapsulating carotenoids and fish oil due to their strong emulsifying properties. Results showed the MFJSD microcapsules presented uniform particle size and regular morphological characteristics, while the SD and FD microcapsules presented a large distribution of particle size and irregular morphological characteristics. Compared to the SD and FD microcapsules, the MFJSD microcapsules possessed higher microencapsulation efficiency (94.0-95.1%), higher tapped density (0.373-0.652 g/cm3), and higher flowability (the Carr index of 16.0-30.0%). After a 4-week storage, the SD microcapsules showed the lower retention of carotenoids, as well as ω-3 LC-PUFAs than the FD and MFJSD microcapsules. After in vitro digestion trial, the differences in the digestion behaviours of the microcapsules mainly resulted from the different wall materials, but independent of drying methods. This study has provided an alternative way of delivering visual-beneficial compounds via a novel drying method, which is fundamentally essential in both areas of microencapsulation application and functional food development.

Abstract:The aim of this study was to encapsulate fish oil by emulsification and spray drying using soy protein and cellulose nanocrystals matrices as nanocomposite wall material, and to evaluate the effect of CNC concentration on the microcapsules structural and physicochemical properties of the microcapsules and on the stability of fish oil against lipid oxidation. All emulsions could be dehydrated by spray drying to produce matrix-type microcapsules with high encapsulation efficiencies of fish oil. The microcapsules masked the fish oil odor and didn't have an oily appearance to the touch. The addition of CNCs up to 4% in soy protein microcapsules caused an increase in the solids yield and improved the protection of fish oil against oxidation, showing low peroxide levels and thiobarbituric acid reactive substance indices. The microcapsules exhibited low water activity values, suggesting their stability against lipid oxidation and microbial growth during storage. These microcapsules could be used as vehicles for omega-3 fatty acids in the formulation of functional foods.

Abstract:BACKGROUND Incorporating fish oils into commonly consumed foods is considered an imminent technique for increasing the daily amount of omega‐3 fatty acids intake. However, the great vulnerability of fish oil towards oxidative deterioration cause shelf‐stability reduction. Microencapsulation by spray drying with different combinations of biopolymers and other parameters may provide a solution by preventing further oxidation of fish oil and permitting its delivery to food items. This study emphasised the influence of developed biopolymer materials (maltodextrin, maltodextrin + gum arabic, maltodextrin + starch, maltodextrin + whey protein isolates and maltodextrin + sodium caseinate) for emulsion formulation, inlet air temperature (160, 170, 180, 190 and 200 °C) and emulsion ratios (15, 20, 25, 30 and 35 %) towards the physicochemical of powdered Menhaden fish oil by spray drying technique. RESULTS Microencapsulated Menhaden fish oil was evaluated for its moisture content, size distribution, microcapsule efficiency, peroxide value, free fatty acid, acid value, and morphological structure. It was observed that the coating materials of maltodextrin + whey protein isolates give the lowest moisture content and peroxide value of 4.410% and 4.031 mEq/kg, respectively. The morphological images showed a smooth surface with no cracks and minimal shrinking, indicating lower permeability to gases and effectively protecting the oil against deterioration. CONCLUSION The spray drying procedure using maltodextrin + whey protein isolates at inlet air temperature 190 °C and emulsion ratios of 30% successfully produce fish oil microcapsules with better properties. Abstract:The impact of the encapsulation technology on the oxidative stability of fish-oil-loaded capsules was investigated. The capsules (ca. 13 wt% oil load) were produced via monoaxial or coaxial electrospraying and spray-drying using low molecular weight carbohydrates as encapsulating agents (e.g., glucose syrup or maltodextrin). The use of spray-drying technology resulted in larger capsules with higher encapsulation efficiency (EE > 84%), whilst the use of electrospraying produced encapsulates in the sub-micron scale with poorer retention properties (EE < 72%). The coaxially electrosprayed capsules had the lowest EE values (EE = 53–59%), resulting in the lowest oxidative stability, although the lipid oxidation was significantly reduced by increasing the content of pullulan in the shell solution. The emulsion-based encapsulates (spray-dried and monoaxially electrosprayed capsules) presented high oxidative stability during storage, as confirmed by the low concentration of selected volatiles (e.g., (E,E)-2,4-heptadienal). Nonetheless, the monoaxially electrosprayed capsules were the most oxidized after production due to the emulsification process and the longer processing time.
Optimization of the production of double-shell microparticles containing fish oil

Abstract:Fish oil incorporation into food products is a challenge because long-chain fatty acids are susceptible to oxidation. Microencapsulation is an alternative for protecting and delivering fish oil besides masking undesirable flavours. This work aimed to produce spray-chilled microparticles using spray-dried microparticles loaded with fish oil as the core material and evaluate the effects of core concentration and lipid wall material composition on the apparent viscosity of the feeding material (suspension), microparticle mean diameter (D50), moisture content and eicosapentaenoic acid and docosahexaenoic acid losses. Double-shell microparticles containing fish oil were successfully obtained. Higher core concentrations resulted in higher feeding material viscosities and microparticles with higher D50 values and higher moisture content, but suitable for food applications. Less eicosapentaenoic acid and docosahexaenoic acid loss was achieved with lipid matrixes containing palm fat/vegetable fat ratios of up to 40/60 or a ratio of 50/50 when associated with a low concentration of core material. The remaining eicosapentaenoic acid and docosahexaenoic acid content observed in the final double-shell microparticles and its good oxidative stability can be considered sufficient for the successful application of these microparticles in foods. These findings may contribute to expanding the use of microencapsulated fish oil.

Abstract:The aim of the present study was to investigate the effect of wall materials composition on physicochemical characteristics of fish oil microcapsules produced by spray drying (180 °C). Four different combination of coating materials (fish gelatin, chitosan, combination of gelatin and chitosan, and a mixture of microbial transglutaminase (MTGase) with maltodextrin) were applied to two different fish oils to produce 40 % solid emulsions. Scanning electron microscopy and extraction of surface and encapsulated oils revealed that fish gelatin provided the highest preserving effect on the covering fish oil. Meantime, addition of MTGase to gelatin could also increase this ability and reveled less surface oil than chitosan treatment (2.63 and 2.80 % versus 4.66 and 5.23 %, respectively; P < 0.05). Mixture of gelatin and maltodextrin with MTGase as wall material led to the highest encapsulation efficiency, being selected as the best microencapsulation condition; meantime, application of chitosan with maltodextrin provided the worse encapsulation efficiency (P < 0.05). All indices of powders (encapsulation efficiency, surface morphology, and particle size) showed that powders prepared from gelatin and gelatin with MTGase increased the encapsulation efficiency and would increase the stability of microcapsule powders.

Abstract:Edible oils with physiological functions, such as fish oil and krill oil, which contain polyunsaturated fatty acids (PUFAs), are highly unstable to heat and oxygen. One of the methods for improving the shelf life of these oils is microencapsulation of the oils through spray drying. This mini-review describes the effects of the surface–oil ratio, oil location within the microcapsules, and oil-droplet diameter on the oxidative stability of functional oils in microcapsules prepared through spray drying. The surface–oil ratio, s , can be expressed through the ratio of the average oil-droplet diameter to the average powder diameter, E , by s = 1 − (1 − 2 E ) 3 . Additionally, the effects of oil-droplet size and storage temperature on the stability of the encapsulated oil were examined using squalene as a model functional oil. The activation energy E a of the squalene degradation rate constant of the Avrami equation can be correlated with the average oil-droplet diameter d e . The release behavior of the microencapsulated flavors is also discussed because spray drying is also used for the microencapsulation of flavors. The flavor release rate constant of the Avrami equation is related to the oxygen diffusion coefficient of the wall material.

Abstract:The objective of this study was to evaluate the potential of double wall material combinations, using legume protein (soy protein isolate and pea protein isolate) in combination with wheat dextrin soluble fiber or trehalose, as alternative materials for microencapsulation of flaxseed oil by spray drying. The obtained preparations, with oil content of 35%, were fine and difficult flowing powders, regardless of their composition. The 1% addition of silica to the powders significantly reduced their cohesiveness and improved their flowability. The efficiency of microencapsulation, calculated based on oil fat content, ranged from 62 to 98% and was higher in the powders with trehalose and in the powders containing soy protein. Effective protection against oxidation of microencapsulated oil was achieved in the protein-trehalose matrix, especially in the case of the vacuum-packed powders with pea protein during storage at refrigeration temperature. Replacing trehalose with soluble fiber enabled formation of powders less susceptible to caking under conditions of increased humidity, but it resulted in decreased microencapsulation efficiency. The combination of pea protein/carbohydrate resulted in the formation of microcapsules with porous structure, especially in the system with soluble fiber. With time, the structure of the primary emulsions and those reconstituted from powders containing pea protein changed from liquid to greasy and paste-like.

Abstract:Sepsis and septic shock are the major causes of morbidity and mortality in critically ill patients. During the onset of sepsis, a massive inflammatory reaction involving chemical mediators such as cytokines and chemokines and inflammatory cells such as the polymorphonuclear neutrophil and macrophage takes place. In addition to this systemic inflammatory process, sepsis and septic shocks cause a profound decrease in the peripheral vasomotor tone leading to a great decrease in the peripheral resistance. This event is central to derangement of hemodynamic and perfusion parameters. Nitric oxide (NO) is produced by several cell types and has been implicated in a wide range of physiological and pathological processes, with both detrimental and beneficial effects. There is a wealth of data implicating NO as a key player in all cardiac, vascular, renal and pulmonary derangements of sepsis and septic shock. Clinical assays trying to improve sepsis by inhibiting NO formation by NO synthases have met with failure, probably due to the lack of selectivity of inhibitors towards NOS isoforms. Notwithstanding the search for selective inhibitors, a better understanding of the NO molecular effector mechanisms may provide new opportunities for therapy development. Some of these NO effector mechanisms are discussed, including guanylate cyclase, nitrosothiols, potassium channels, reactive oxygen species and gene expression in the context of sepsis. Thus, more research on the relationship between NO and sepsis is clearly needed and warranted and may provide new therapeutic targets to treat sepsis and septic shock.
Impact of chitosan and oregano extract on the physicochemical properties of microencapsulated fish oil stored at different temperature

Abstract:Fish oil is an excellent source of omega-3 fatty acids and is easily susceptible to oxidation. Microencapsulation is a commonly employed technique to protect fish oil against oxidation. In the present study, the potential of chitosan in combination with bovine gelatin and maltodextrin as wall material for microencapsulation of fish oil by spray drying was evaluated. To improve the oxidative stability of the fish oil microencapsulates, oregano (Origanum vulgare L) extract was added at 0.50 g/100 g of emulsion. The spray-dried powder showed a moisture content of 2.8 – 3.2 g/100 g of spray-dried powder. The powder contained spherical microparticles with different sizes as indicated by scanning electron microscope images. Encapsulation efficiency of microencapsulates ranged between 68.94% and 81.88%. Differential scanning calorimetry and Fourier-transformed infrared spectroscopy analysis of microencapsulates revealed the possible structural stabilization of core and wall material. The oxidative stability of fish oil microencapsulates were monitored under three different temperature (60°C, 28 ± 2°C, and 4°C). Incorporation of oregano extract minimized the generation of secondary and tertiary oxidation products as indicated by lower peroxide value and thiobarbituric acid reactive substance values compared to control. Overall, the results suggested that combination of chitosan along with bovine gelatin and maltodextrin as wall material improved the surface morphology of the microparticle and encapsulation efficiency, whereas incorporation of oregano extract in fish oil before spray drying enhanced the oxidative stability during storage.

Abstract:Evaluate the stability of microparticles loaded with fish oil produced by spray drying, spray chilling and by the combination of these techniques (double-shell) and use the microparticles for food application.Samples were stored for 180 days at 6 °C and 24 °C (75% RH). Performed investigations included encapsulation efficiency, moisture content, aw, size (laser scattering), colour (L*, a*, b*), polyunsaturated fatty acids (PUFAs) (GC), thermal behaviour (DSC) and crystalline structure (XRD).Double-shell microparticles containing 26 wt% core material, 22.74 ± 0.02 µm (D0.5) and 2.05 ± 0.03 span index, 1.262 ± 0.026 wt% moisture content and 0.240 ± 0.001 of aw had PUFAs retention higher than 90 wt% during storage at 6 °C without changes in crystalline structure (β'-type crystals) and melting temperature (54 °C). The sensory evaluation suggested low fish oil release in oral phase digestion.Double-shell microparticles were effective to protect and deliver PUFAs.

Abstract:Fish oil, rich in unsaturated fatty acids, is susceptible to oxidation. However, employing multiple emulsification technologies with polymers and antioxidants can effectively stabilise it, resulting in a stable fish oil powder. Summary Polyunsaturated fatty acids (PUFAs), which are abundant in fish oil, easily oxidise. Therefore, stabilisation is necessary, preferably through encapsulation and the addition of antioxidants. This research looked on multiple emulsification for PUFA‐rich fish oil by utilising a wall material (whey protein and chitosan) and an antioxidant (betalain) combination. Spray‐dried, microparticles filled with PUFA were successfully created (80% yield) and characterised. It was determined that the emulsion droplet size was 0.917 μm. Good oxidative stability was observed after 4 weeks of storage, as evidenced by a moderate (4%) drop in EPA and DHA content and a low peroxide value (PV) of 4.8 ± 0.23 meq/kg oil in microparticles. Microencapsulation of PUFA‐rich fish oil was successfully accomplished using multiple emulsion technology, as evidenced by encapsulation efficiency values of 87.73 ± 0.21% and loading efficiency values of 67 ± 0.11%. These results suggest that structurally stabilised healthful lipids could be added to foods using these innovative emulsions, which provide greater stability and versatility for a wide variety of uses.

Abstract:In the spray drying process, variation of outlet temperatures significantly influence the microencapsulation efficiency of fish oil and chia oil. Oil sprayed at 55 °C showed maximum encapsulation efficiency and minimum peroxide value. FTIR spectra of the capsule proved the presence of PUFA in all encapsulated samples. The spectra of microcapsules and unencapsulated samples followed the same trend, implying no interaction between the wall and core. Size of particles were found to be 5.04 and 3.89 µm for chia oil and fish oil powders, respectively; the surface showed blowholes indicating a shell structure with smooth surface. Fatty acid profile of samples indicated a considerable retention of essential fatty acids in both oil powder samples. Maximum release in gastrointestinal conditions was observed, indicating that the capsules were more stable under acidic conditions and that maximum oil would be available for absorption in the intestine.

Abstract:Fish oil is one of the most important sources of omega 3 polyunsaturated fatty acids (PUFAs), especially eicosapentaenoic acid and docosahexaenoic acid which are the most important PUFAs with several health benefits. However, PUFAs are prone to oxidation and have a poor water solubility which limits the use of fish oils into food formulations. Encapsulation techniques can be applied to overcome these challenges. There is a large number of published micro/nanoencapsulation papers, where each of them contains a limited number of wall materials, feed formulation, encapsulation technique, and storage conditions. Therefore, without systematic evaluation of the data extracted from available studies, the design of functional foods containing fish oil would not be very successful. The objective of this systematic review is a meta-analysis of published researches on the nano/microencapsulation of fish oil. A comprehensive literature search was performed between 1 October and 31 December 2019 with encapsulation, fish oil, and oxidative stability keywords. Overall, 39 qualified articles were selected for the statistical analysis. Based on the technique used for encapsulation, the fish oil-loaded carriers were classified into four main groups: (a) spray-dried particles; (b) freeze-dried particles; (c) electrospun fibers and electrosprayed capsules; and (d) other carriers prepared by supercritical antisolvent, gelation, liposomes, spray–freeze drying, and transglutaminase catalyzed cross-linking. The three most frequent methods applied for fish oil encapsulation were spray drying (42.86%), freeze drying (21.43%), and electrohydrodynamic (19.04%) methods, respectively. Averagely, the best encapsulation efficiency was obtained for electrohydrodynamic processes. Also, the combination of polysaccharide–protein based wall materials provided the best performance in terms of fish oil encapsulation efficiency.

Optimized spray-dried conditions' impact on fatty acid profiles and estimation of in vitro digestion of spray-dried chia/fish oil microcapsules

Abstract:Long-chain polyunsaturated fatty acids (LCPUFA) are of interest due to their potential health properties and have a significant role in reducing the risk of various chronic diseases in humans. It is commonly used as a supplement. However, lipid oxidation is an important negative factor caused by environmental, processing, and limited water solubility of LCPUFA, making them difficult to incorporate into food products. The objective of this research work was to prevent oxidation, extend shelf life, enhance the stability of fatty acids, and to achieve controlled release by preparing spray-dried powder (SDM). For spray-drying, aqueous emulsion blends were formulated using a 1:1 ratio of chia seed oil (CSO) and fish oil (FO) and using a laboratory-scale spray-dryer with varying conditions: inlet air temperature (IAT, 125-185 °C), wall material (WM, 5-25%), pump speed (PS, 3-7 mL/min), and needle speed (NS, 3-11 s). The maximum alpha-linolenic acid (ALA) content was 33 ± 1%. The highest values of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in the microcapsules were 8.4 ± 0.4 and 13 ± 1%, respectively. Fourier transform infrared and X-Ray diffraction analysis results indicated that SDM was successfully formulated with Gum Arabic and maltodextrin (MD). The blending without encapsulation of CSO and FO was digested more efficiently and resulted in more oil being released with simulated gastric fluid (SGF), simulated intestinal fluid (SIF), and SGF + SIF conditions without heating. No significant changes were observed for saturated, monounsaturated, and LCPUFA, whether exposed or not to gastrointestinal conditions. However, compared to the release of SDM, it can be useful for designing delivery systems for the controlled release of essential fatty acids.

Abstract:Background: Omega-3 fatty acids are essential fatty acids that the body cannot create itself; thus, they must be obtained from functional foods. Therefore, the food processing industries are becoming more interested in the production of omega-3 enriched food products, as consumers and healthcare organizations are increasingly demanding functional foods with minimal fatty acid loss and higher oxidative stability. Moreover, the stability of long-chain, polyunsaturated fatty acids in functional foods is a major challenge for the food processing industries. Therefore, the spray drying method was used to prepare spray-dried microcapsules (SDMs) with the minimum loss of, and more stable, fatty acids. Methods: In this study, emulsion blends of chia seed oil (CSO, 50%) and fish oil (FO, 50%) were spray-dried using varied operating conditions, including the inlet air temperature (IAT) (125, 140, 155, 170, and 185 °C), wall material (WM) (5, 10, 15, 20, and 25%), pump speed (PS) (3, 4, 5, 6, and 7 mL/min), and needle speed (NS) (3, 5, 7, 9, and 11 S). Results: The maximum loss of ALA in the spray-dried microcapsules (SDMs) was observed (9.90 ± 0.40%) at 170 °C, and the minimum loss was 4.18 ± 0.20% in run order 9. A similar trend was observed in the maximum retention loss of EPA and DHA (9.71 ± 0.39% and 9.77 ± 0.39%) at a high temperature of 170 °C, while the minimum losses of EPA and DHA were observed in run order 9. Furthermore, the maximum peroxide value (PV) of the SDMs was observed at a lower temperature of 140 °C (1.45 ± 0.19 meq O2/kg), and the minimum PV was 1.33 ± 0.16 meq O2/kg. Conclusions: Overall, based on the results, we concluded that the oxidative stability of the SDMs was improved and that it can be used as a fortifying agent in the processing of many food products.

Abstract:In this study, we prepared fish oil microcapsules by spray-drying using mesquite gum and chitosan as wall materials, and the encapsulation efficiency, microstructure, and lipid hydroperoxide concentration were evaluated. First, stable mesquite gum (216 nm and −22.3 mV) and mesquite gum/chitosan emulsions (333 nm and +47.9 mV) were produced. The fish oil emulsions were spray-dried, and the mesquite gum microcapsules displayed an encapsulation efficiency of 67.18% while mesquite gum/chitosan microcapsules exhibited an efficiency of 92.92%. The lipid hydroperoxide content of the fish oil microcapsules depended on the composition of the wall material, storage time, and storage temperature.Graphical

Abstract:In this study, ω-3 medium- and long-chain triacylglycerols (MLCTs) microcapsules with excellent performance were obtained using soy protein as the wall component to address the oxidation-related problems of MLCTs. Additionally, the effect of soy, whey, or pea proteins on microcapsules in terms of the changes in their structure and physicochemical properties was investigated. The results showed that the small particle size, low PDI (polydispersity index) and zeta potential, fast adsorption rate, and low interfacial tension of these protein-based samples fabricated through the O/W template method were conducive to maintaining the integrity of microcapsules during spray-drying. The microcapsules, characterized by a spherical shape, exhibited superior encapsulation efficiency of 94.56%, surpassing the findings of previous investigations. Overall, these microcapsules exhibited long-term storage stability and low controllable release rates, which could be utilized as carriers for liposoluble actives.
