Abstract:In this work, we prepared a novel electrochemical sensor for the detection of tramadol based on a UiO-66-NH2 metal–organic framework (UiO-66-NH2 MOF)/third-generation poly(amidoamine) dendrimer (G3-PAMAM dendrimer) nanocomposite drop-cast onto a glassy carbon electrode (GCE) surface. After the synthesis of the nanocomposite, the functionalization of the UiO-66-NH2 MOF by G3-PAMAM was confirmed by various techniques including X-ray diffraction (XRD), energy-dispersive X-ray spectroscopy (EDS), field emission-scanning electron microscopy (FE-SEM), and Fourier transform infrared (FT-IR) spectroscopy. The UiO-66-NH2 MOF/PAMAM-modified GCE exhibited commendable electrocatalytic performance toward the tramadol oxidation owing to the integration of the UiO-66-NH2 MOF with the PAMAM dendrimer. According to differential pulse voltammetry (DPV), it was possible to detect tramadol under optimized circumstances in a broad concentration range (0.5 μM–500.0 μM) and a narrow limit of detection (0.2 μM). In addition, the stability, repeatability, and reproducibility of the presented UiO-66-NH2 MOF/PAMAM/GCE sensor were also studied. The sensor also possessed an acceptable catalytic behavior for the tramadol determination in the co-existence of acetaminophen, with the separated oxidation potential of ΔE = 410 mV. Finally, the UiO-66-NH2 MOF/PAMAM-modified GCE exhibited satisfactory practical ability in pharmaceutical formulations (tramadol tablets and acetaminophen tablets).

G-quadruplex Specific Thioflavin T-based Label-Free Fluorescence Aptasensor for Rapid Detection of Tetracycline.

Abstract:A label-free fluorescence aptasensor was developed for the rapid detection of tetracycline (TET) based on G-quadruplex structure of TET aptamers and G-quadruplex specific dye Thioflavin T (ThT). The fluorescence of free ThT is essentially weak in aqueous solution, whereas it selectively identifies the G-quadruplex of aptamers to form the G-quadruplex/ThT conjugates, resulting in an enormous increase of the fluorescence intensity. However, the fluorescence intensity of G-quadruplex/ThT conjugates was drastically suppressed due to the release of free ThT from G-quadruplex/ThT conjugates after the addition of TET via specific binding with TET aptamers. The key factors affecting sensitivity and selectivity including the reaction medium, binding time of ThT to TET aptamers, incubation time between TET aptamers and TET, concentration of ThT and TET aptamers were investigated in detail. The optimal conditions were as follows: ultrapure water as reaction medium, binding time of 5min, incubation time of 1 min, 9.0 mu mol/L ThT and 0.03 mu mol/L aptamers. A good linear relationship (correlation coefficient of 0.9973) was obtained between the fluorescence quenching efficiency (F-0 - F) / F0 and the logarithm of TET concentration in the range of 0.01-1.0 mu mol/L. The limit of detection was 0.001 mu mol/L (S/N = 3). The proposed assay was applied for the detection of TET in the spiked honey and milk samples with recoveries ranging from93.5% to 106.9%. The developed label-free fluorescence aptasensor showed advantages of high specificity, low cost and short time-consuming, illustrating potential application for on-site detection of TET in foodstuffs. Abstract:Monitoring the concentration of Fludarabine phosphate, a standard chemotherapeutic drug widely used in cancer treatment, is vital for ensuring the drug's safety and effectiveness, tailoring treatments to individual needs, and consequently improving overall patient outcomes. Regarding the limitations of conventional techniques in terms of complexity, large time measurements, and a high cost, there is an urgent need to develop simple, rapid, and cost-effective devices. In this paper, we report the design of an aptasensor for the specific and selective detection of Fludarabine. Systematic evolution of ligands by exponential enrichment (SELEX) protocol was performed to select a specific aptamer for Fludarabine. Eleven rounds were carried out, and nine sequences were selected. Based on the dissociation constant (Kd), Ful-3, exhibiting the highest affinity (18.86 nM), was chosen and integrated into a simple electrochemical aptasensing platform for Fludarabine detection. Electrochemical results demonstrated good performance of the selected aptamer in detecting Fludarabine within the analytical range of 1 to 150 pg/mL and with LOD and LOQ in the order of 0.11 pg/mL (0.31 fM) and 0.39 pg/mL (1.06 fM), respectively. The developed platform also showed good selectivity against different analogous molecules and high applicability in serum-spiked samples, with recovery percentages ranging from 96.81 to 99.04%. Considering the encouraging results, this research presents an excellent alternative in terms of simplicity, stability, ease of use, reduction of sample volume, and low cost.

Abstract:Diclofenac (DCF) is among the pharmaceutical compounds of concern based on its nocuousness in environment. In this work, a novel electrochemiluminescent (ECL) immunosensor for detecting DCF was constructed using poly(etherimide)-poly(3,4-ethylene dioxythiophene): poly(styrene sulfonate) functionalized graphene oxide and CdSe@CdS quantum dots (QDs-PEI-GO/PEDOT) as bioreceptor for conjugating DCF antibody to magnify signal. It is worth noting that this is the first time GO/PEDOT has been applied to ECL sensor field. Compared with GO, GO/PEDOT exhibited a higher conductivity and more stable chemical property, indicating that the proposed immunosensor would possess stronger and more stable luminescence performance. In addition, the electrode was modified with gold nanorods (AuNRs) which increase the load capacity of DCF coating antigen through Au-N bond. Competitive immunoassay method was chosen for structuring immunosensor where polyclonal antibody (pAb) against DCF exhibited high-affinity recognition of DCF, which greatly improved the sensitively and selectivity of sensor. Consequently, the proposed immunosensor gratified in DCF detection with low detection limit (LODs) of 0.33 pg mL(-1) (S/N = 3), and displayed high stability and sensitivity, which initiated a new route for DCF determining.
Developing Aptamer-Based Colorimetric Opioid Tests

Abstract:Opioids collectively cause over 80,000 deaths in the United States annually. The ability to rapidly identify these compounds in seized drug samples on-site will be essential for curtailing trafficking and distribution. Chemical reagent-based tests are fast and simple but also notorious for giving false results due to poor specificity, whereas portable Raman spectrometers have excellent selectivity but often face interference challenges with impure drug samples. In this work, we develop on-site sensors for morphine and structurally related opioid compounds based on in vitro-selected oligonucleotide affinity reagents known as aptamers. We employ a parallel-and-serial selection strategy to isolate aptamers that recognize heroin, morphine, codeine, hydrocodone, and hydromorphone, along with a toggle-selection approach to isolate aptamers that bind oxycodone and oxymorphone. We then utilize a new high-throughput sequencing-based approach to examine aptamer growth patterns over the course of selection and a high-throughput exonuclease-based screening assay to identify optimal aptamer candidates. Finally, we use two high-performance aptamers with KD of ∼1 μM to develop colorimetric dye-displacement assays that can specifically detect opioids like heroin and oxycodone at concentrations as low as 0.5 μM with a linear range of 0-16 μM. Importantly, our assays can detect opioids in complex chemical matrices, including pharmaceutical tablets and drug mixtures; in contrast, the conventional Marquis test completely fails in this context. These aptamer-based colorimetric assays enable the naked-eye identification of specific opioids within seconds and will play an important role in combatting opioid abuse.
CuONPs/MWCNTs/carbon paste modified electrode for determination of tramadol: theoretical and experimental investigation

Abstract:A practical technique was applied to fabricate CuO nanostructures for use as the electrocatalyst. The green synthesis of cupric oxide nanoparticles (CuO NPs) via co-precipitation is described in this paper using an aqueous extract of Origanum majorana as both reductant and stabilizer, accompanied by characterization via XRD, SEM, and FTIR. The XRD pattern revealed no impurities, whereas SEM revealed low agglomerated spherical particles. CuO nanoparticles and multi wall carbon nanotubes (MWCNTs) have been used to create a modified carbon paste electrode. Voltammetric methods were used to analyze Tramadol using CuONPs/MWCNT as a working electrode. The produced nanocomposite showed high selectivity for Tramadol analysis with peak potentials of ~ 230 mV and ~ 700 mV and Excellent linear calibration curves for Tramadol ranging from 0.08 to 500.0 µM with a correlation coefficient of 0.9997 and detection limits of 0.025. Also, the CuO NPs/MWCNT/CPE sensor shows an an appreciable sensitivity of 0.0773 μA/μM to tramadol. For the first time the B3LYP/LanL2DZ, quantum method was used to compute DFT to determine nanocomposites' connected energy and bandgap energy. Eventually, CuO NPs/CNT was shown to be effective in detecting Tramadol in actual samples, with a recovery rate ranging from 96 to 104.3%.
Selection and truncation of aptamers for ultrasensitive detection of sulfamethazine using a fluorescent biosensor based on graphene oxide

Abstract:We developed a fluorescent aptamer/graphene oxide (GO)–based biosensor to detect sulfamethazine (SMZ) residues in animal-derived foods. The SMZ-bound aptamers were identified and screened with an improved GO-SELEX technique using non-immobilizing ssDNA library. After seven rounds of selection, six SMZ aptamers were sequenced and analyzed for secondary structure, and their affinity and specificity were assessed by binding assays. The truncated aptamer (SMZ1S: 5′-CGTTAGACG-3′) with a unique stem-loop structure showed the highest affinity (<i>K</i><sub>d</sub> = 24.6 nM) to SMZ and was used to develop a GO-based fluorescent aptasensor. The binding mechanism between SMZ1S and SMZ was further analyzed by molecular docking. Under optimal conditions, the fluorescent aptasensor showed low detection limits (0.35 ng/mL) and a wide dynamic linear range (from 2 to 100 ng/mL). The aptasensor was also validated against real samples spiked with SMZ, which showed a fluorescence recovery from 93.9 to 108.8% and a coefficient of variation of < 12.7%. Taken together, these results suggest that this novel aptasensor can be used to sensitively, selectively, and accurately detect SMZ residues in foods.

Abstract:The in vitro activity of piperacillin, a new semisynthetic piperazine penicillin derivative, was evaluated against 626 clinical isolates and compared with the activity of other beta-lactam antibiotics. At a concentration of 0.1 microgram/ml, piperacillin inhibited all streptococci except enterococci. Non-beta-lactamase-producing staphylococci were inhibited by 1.6 microgram or less per ml. Both beta-lactamase- and non-beta-lactamase-producing Haemophilus were inhibited by 0.1 microgram/ml. Piperacillin inhibited non-beta-lactamase-producing Escherichia coli, Salmonella, and Shigella at a concentration of 6.3 micrograms/ml, but 20% of strains of these species containing type III beta-lactamase were not inhibited by 100 micrograms/ml. Piperacillin at 25 micrograms/ml, inhibited 83% of Citrobacter, 58% of Klebsiella, 88% of Enterobacter, and 50% of indole-positive Proteus, Acinetobacter, and Providencia. At 25 micrograms/ml, piperacillin inhibited 95% of Pseudomonas aeruginosa and 78% of Bacteroides fragilis. The minimal inhibitory concentration of piperacillin against Pseudomonas was affected by increasing the inoculum size and by pH. Minimum bactericidal concentrations against Pseudomonas and Serratia often were eightfold greater than the minimum inhibitory concentrations. Piperacillin was equal in activity to ampicillin against enterococci. It was more active than carbenicillin against E. coli, Klebsiella, Enterobacter, and Bacteroides. It was the most active penicillin against Pseudomonas and inhibited many strains of Pseudomonas for which the MICs of carbenicillin were above 200 micrograms/ml. Piperacillin was hydrolyzed by many different beta-lactamases. Synergistic activity of piperacillin was demonstrated when it was combined with amikacin, gentamicin, and cefazolin against P. aeruginosa and members of the Enterobacteriaceae. No antagonism was observed when piperacillin was combined with aminoglycosides; however, antagonism was observed rarely against E. coli when piperacillin was combined with cefazolin.
Graphene Oxide-Assisted Non-Immobilized SELEX of Okdaic Acid Aptamer and the Analytical Application of Aptasensor

Abstract:Okadaic acid (OA) is a low-molecular-weight marine toxin from shellfish that causes abdominal pain, vomiting and diarrhea, i.e., diarrheic shellfish poisoning. In this study, a ssDNA aptamer that specifically binds to OA with high affinity was obtained via Systematic Evolution of Ligands by Exponential Enrichment (SELEX) assisted by graphene oxide (GO). This aptamer was then applied to fabricate a novel direct competitive enzyme-linked aptamer assay (ELAA). At the optimized conditions, this ELAA method showed a low detection limit (LOD of 0.01 ng/mL), wide linear range (from 0.025 to 10 ng/mL), good recovery rate (92.86–103.34% in OA-spiked clam samples) and repeatability (RSD of 2.28–4.53%). The proposed method can be used to detect OA in seafood products with high sensitivity and can potentially be adapted for the determination of other small molecular analytes.
Detection of theophylline using molecularly imprinted polymers based on thioglycolic acid-modified CdTe quantum dots

Abstract:Molecularly imprinted polymers (MIPs) and quantum dots (QDs) have been widely employed to fabricate highly sensitive and selective sensor. Here, we developed a fluorescence nanosensor based on thioglycolic acid-modified CdTe QDs that is coated with molecularly imprinted polymers for the specific detection of theophylline (THP). Initially, water-soluble thioglycolic acid-modified CdTe QDs were synthesized by refluxing method. Then, MIPs-coated QDs (MIPs-QDs) composite was produced by sol–gel process using THP as a template. Therefore, the selectivity of the molecular imprinting technique and advantages of QDs were combined. The prepared QDs and the MIPs-QDs were characterized using X-ray diffraction technique, transmission electron microscopy, dynamic light scattering, Fourier transform infrared spectroscopy, and fluorescence spectrometry. Finally, this sensing system was successfully used to detect THP in human plasma samples with recoveries of 90% to 108%. A very good linear relationship was observed between the decreasing in the fluorescence intensity of MIPs-QDs and increasing the THP concentration within concentration range of 0.14–3.05 µmol L−1, with a correlation coefficient of 0.9992 and detection limit of 0.07 µmol L−1.

A highly sensitive first derivative synchronous spectrofluorimetric approach for the simultaneous analysis of the anti-breast cancer co-administered drugs, letrozole and tramadol in dosage forms and human plasma at nanogram levels

Abstract:Letrozole is an anticancer medication prescribed for the management of estrogen receptor-positive breast cancer in postmenopausal women. Chronic pain is prevalent in patients receiving chemotherapy, leading to the use of adjuvant analgesics such as tramadol. This work introduces the first analytical approach for the concurrent quantification of letrozole and tramadol, two co-administered drugs, employing a rapid, highly sensitive, eco-friendly, and cost-effective first derivative synchronous spectrofluorimetric technique. The fluorescence of tramadol and letrozole was measured at wavelengths of 235.9 nm and 241.9 nm, respectively using a wavelength difference (Δλ) of 60.0 nm. The developed approach demonstrated exceptional linearity (r ˃ 0.999) within the specified concentration ranges for tramadol (10.0–1200.0 ng/mL) and letrozole (1.0–140.0 ng/mL). The results demonstrated that the proposed technique exhibits a high level of sensitivity, with detection limits of 0.569 and 0.143 ng/mL for tramadol and letrozole, respectively, indicating the good bioanalytical applicability. The within-run precisions, both intra-day and inter-day, for both analytes, were less than 0.71 % RSD. The developed approach was effectively applied to simultaneously estimate the mentioned drugs in their tablets and human plasma samples, achieving high percentage recoveries and low % RSD values. In order to assess the environmental sustainability of the developed approach, Analytical GREEnnessNNESS (AGREE) and the Green Analytical Procedure Index (GAPI) metric tools were employed. Both tools revealed that the developed approach is excellent green, suggesting its usage as an environmentally-friendly alternative for the routine assay of the investigated pharmaceuticals. The developed approach was validated according to the ICHQ2 (R1) requirements.

Abstract:Aptamers are short oligonucleotides capable of binding specifically to various targets (i.e., small molecules, proteins, and whole cells) which have been introduced in biosensors such as in the electrochemical aptamer-based (E-AB) sensing platform. E-AB sensors are comprised of a redox-reporter-modified aptamer attached to an electrode that undergoes, upon target addition, a binding-induced change in electron transfer rates. To date, E-AB sensors have faced a limitation in the translatability of aptamers into the sensing platform presumably because sequences obtained from Systematic Evolution of Ligands by Exponential Enrichment (SELEX) are typically long (> 80 nucleotides) and that obtaining structural information remains time and resource consuming. In response, we explore the utility of aptamer base truncations and in silico docking to improve their translatability into E-AB sensors. Here, we first apply this to the glucose aptamer, which we characterize in solution using NMR methods to guide design and translate truncated variants in E-AB biosensors. We further investigated applicability of the truncation and computational approaches to five other aptamer systems (vancomycin, cocaine, methotrexate, theophylline, and ochratoxin A) from which we derived functional E-AB sensors. We foresee that our strategy will increase the success rate of translating aptamers into sensing platforms to afford low-cost measurements of molecules directly in undiluted complex matrices.

In vitro isolation of small-molecule-binding aptamers with intrinsic dye-displacement functionality

Abstract:Aptamer-based sensors offer a powerful tool for molecular detection, but the practical implementation of these biosensors is hindered by costly and laborious sequence engineering and chemical modification procedures. We report a simple strategy for directly isolating signal-reporting aptamers in vitro through systematic evolution of ligands by exponential enrichment (SELEX) that transduce binding events into a detectable change of absorbance via target-induced displacement of a small-molecule dye. We first demonstrate that diethylthiatricarbocyanine (Cy7) can stack into DNA three-way junctions (TWJs) in a sequence-independent fashion, greatly altering the dye's absorbance spectrum. We then design a TWJ-containing structured library and isolate an aptamer against 3,4-methylenedioxypyrovalerone (MDPV), a synthetic cathinone that is an emerging drug of abuse. This aptamer intrinsically binds Cy7 within its TWJ domain, but MDPV efficiently displaces the dye, resulting in a change in absorbance within seconds. This assay is label-free, and detects nanomolar concentrations of MDPV. It also recognizes other synthetic cathinones, offering the potential to detect newly-emerging designer drugs, but does not detect structurally-similar non-cathinone compounds or common cutting agents. Moreover, we demonstrate that the Cy7-displacement colorimetric assay is more sensitive than a conventional strand-displacement fluorescence assay. We believe our strategy offers an effective generalized approach for the development of sensitive dye-displacement colorimetric assays for other small-molecule targets.

Abstract:In the current study, nickel oxide nanoparticles (NiO NPs) modified with dimethylglyoxime (DMG) were deposited onto the cellulose surface (Ni(DMG)2-NiO-Cell) and used as an efficient adsorbent for thin film microextraction (TFME) of tramadol (TRA). The extracted TRA was determined using a high-performance liquid chromatography-ultraviolet detector (HPLC-UV). NiO NPs were synthesized by co-precipitation method on the surface of the cellulose substrate; afterward, its surface was modified by DMG to increase the extraction capability of the thin film toward TRA. The synthesized NiO-Cell and Ni(DMG)2-NiO-Cell thin films were characterized using various techniques. The effect of modification of the NiO thin film with DMG reagent on the extraction efficiency was investigated. The crucial parameters influencing the extraction efficiency, including extraction time, desorption time, desorption solvent, pH and salt content, were investigated via a one-at-a-time approach. The figures of merit for the developed method were evaluated in urine, plasma, and deionized water under the optimized extraction and desorption condition. The limits of detection and limits of quantification were in the range of 0.1 to 1 ng mL-1 and 0.3 to 3 ng mL-1, respectively, for the studied samples. The linear dynamic ranges of the developed TFME-HPLC-UV method were 0.3-1000, 1-2500, and 3-5000 ng mL-1 for the deionized water, urine, and plasma samples, respectively. The reproducibility and repeatability of the developed method was assayed in terms of intra-day, inter-day, and inter-thin film precisions by conducting six-replicate experiments at the concentration level of 0.1 and 1 μg mL-1, which were in the range of 5.9% to 8.3%. The sufficiency and applicability of the developed TFME-HPLC-UV method was investigated by determining TRA in urine and plasma samples, and the resulting relative recoveries (RR%) were 85.9% and 91.7%, respectively.

Abstract:A first derivative synchronous spectrofluorimetric technique is proposed for the simultaneous determination of celecoxib and tramadol, which are characterized by their overlapped spectra, in their coformulated mixture. The technique was successfully applied to their dosage form and spiked human plasma with satisfactory results and validated according to International Council for Harmonization of Technical Requirements for Pharmaceuticals for Human Use (ICH). One of the most common features of many different clinical conditions is pain; hence, there is a crucial need for eliminating or reducing it to a tolerable level to retrieve physical, psychological and social functioning. A first derivative synchronous spectrofluorimetry technique is proposed for the simultaneous determination of celecoxib and tramadol HCl, a recent coformulation authorized for treating acute pain in adults. The method includes using synchronous spectrofluorimetry at ∆λ = 80 nm where tramadol HCl was determined using first derivative technique at λ = 230.2 nm, while celecoxib was determined at λ = 288.24 nm. The proposed method was successfully applied to their co‐formulated dosage forms in addition to spiked human plasma and validated in agreement with the guidelines of the International Council for Harmonization of Technical Requirements for Pharmaceuticals for Human Use (ICH). The linear ranges were found to be 0.50–5.0 and 0.15–0.50, the limits of detection to be 0.088 and 0.011 and the limits of quantification to be 0.266 and 0.032 μg/ml for celecoxib and tramadol, respectively. Statistical analysis revealed no significant difference when compared with previously reported methods as evidenced by the values of the variance ratio F‐test and Student t‐test. The proposed method was successfully applied to commercial dosage forms and spiked human samples. Moreover, the greenness of the proposed method was investigated based on the analytical eco‐scale approach, with the results showing an excellent green scale with a score of 95.

Abstract:In view of the urgent need for fluoroquinolones contamination detection in the fields of food safety, a novel aptasensor based on the fluorescence quenching property of graphene oxide (GO) and the fluorescence characteristic of cadmium selenide quantum dots (CdSe QDs) was developed for fluoroquinolones highly sensitive detection in this work. The CdSe QDs with carboxyl-rich surface were synthesized successfully and fluoresced at 525 nm under the optimal excitation light of 366 nm. Based on the hydrophobic and π-π stacking between GO and aptamer, aptamer labeled by CdSe QDs fluorescence (CdSe QDs-apt) were adsorbed by GO and the fluorescence of CdSe QDs was quenched. After the aptamer combined specifically with fluoroquinolones, greater specific force lead to the desorption of CdSe QDs-apt from GO and fluorescence recovery. Represented by Ciprofloxacin (CIP), a member of fluoroquinolones, the fluorescence emission increased with the increasing of CIP concentrations from 8 nM to 500 nM, and the detection limit was 0.42 nM. The spiked recoveries in real samples of honey and milk were 91.5–96.9 % and 90.3–95.2 %, respectively, indicating that the aptasensor was reliable. Moreover, the fluorescence responses of multiple members of fluoroquinolones were found to be consistent, denoting that the fluorescence aptasensor can be used to detect the total amount of multiple members of fluoroquinolones. These results showed that the aptasensor can be used as a promising platform for fluoroquinolones detection.

Abstract:This study focused on detecting streptomycin (STR) residues using a luminescent aptasensor encapsulated with aptamer. Utilizing MOF-74-Co with peroxidase-like activity, luminol was enclosed in its pores. The specific STR aptamer acted as a gatekeeper, ensuring excellent performance. Upon exposure to STR, the aptamers detached, releasing luminol and amplifying the luminescent signal through MOF-74-Co catalytic activity. A linear relationship between fluorescence intensity and STR concentration (50 nM ∼ 5 × 106 nM) was established, with a limit of detection of 0.065 nM. The sensor exhibited high selectivity for STR even in the presence of other aminoglycoside antibiotics. Applied to tea, egg, and honey samples, the sensor showed recovery rates of 91.38-100.2%, meeting safety standards. This MOF-based aptasensor shows promise for detecting harmful residues.

Abstract:Pholcodine, an anti-tussive medication widely used as an over-the-counter, OTC drug, has recently faced restrictions in several countries. This paper presents a sensitive electrochemical approach for pholcodine detection. The electrochemical method involved fabricating a graphene nanoplatelets electrode, incorporating polythiophene nanospheres polymer to promote electron transfer and increase the activated surface area. Characterization of the fabricated electrode was performed using transmission electron microscopy, ATR-Fourier-transform infrared spectroscopy, X-ray crystallography, X-ray photoelectron spectroscopy, and electrochemical impedance spectroscopy. The electrochemical behavior of pholcodine with the fabricated electrode was investigated using cyclic voltammetry, chronoamperometry, square wave voltammetry (SWV), and differential pulse voltammetry (DPV). The developed electrode led to a linear response for pholcodine ranging from 10 to 45 mg/L with detection limits of 1.41 and 1.51 mg/mL for SWV and DPV, respectively and quantification limits of 4.27 and 4.57 mg/L for SWV and DPV, respectively. The proposed method has accurately recovered pholcodine in spiked serum samples with a recovery percentage ranging from 1.2 to 2.9%. The optimized method is found to be accurate, precise, and robust by applying validation parameters provided by International Council for Harmonization. Two green metrics were computed to assess the method's greenness, the findings showed that the developed method is environmentally friendly with minimum sample preparation steps.
