Abstract:Hepatitis C virus (HCV) is a common blood-borne infection that can lead to long-term illnesses such as hepatocellular cancer and liver cirrhosis. Early diagnosis is crucial for effective management, as no vaccine is available for preventing HCV infection. However, the high cost and complexity of current molecular diagnostic tools hinder efforts to achieve early diagnosis and prevent transmission, particularly in resource-limited settings. We developed a novel electrochemical biosensor for point-of-care testing (POCT) of HCV RNA. The sensor utilizes a strand displacement method, where the target RNA displaces a gold nanoparticle-labeled reporter probe (AuRP) from a pre-hybridized duplex with a magnetic nanoparticle (MNP)-labeled capture probe. The amount of displaced AuRP, detected using differential pulse anodic stripping voltammetry (DPASV), is directly proportional to the target RNA concentration. The biosensor exhibited excellent analytical performance, with a detection limit of 4 fM for synthetic targets and 43 ng/μL for RT-PCR products. Importantly, it successfully detected HCV RNA directly in clinical plasma samples without the need for RNA extraction or amplification. The sensor was used to analyze 30 RNA samples from HCV-positive patients, 20 cDNA samples from viral RNA, 30 HCV-positive plasma samples, and 22 HCV-negative plasma samples. The sensor results show good concordance with the RT-PCR results, demonstrating the sensor's potential for detecting HCV in clinical samples.

Abstract:Precise identification of hepatocellular carcinoma (HCC) in early stages remains both a challenge and an opportunity for prevention and diagnosis of early liver cancer. Fast-detecting techniques like electrochemical sensing might provide more insight than current clinical methods owing to its high sensitivity, rapid detection, and convenient operation. Here, a single-chain variable fragment (scFv) was designed by retaining the antigen binding active sites of a recombinant humanized monoclonal antibody and modifying with histidine tags, and consequently used for the construction of cell sensor for HCC detection. The customized scFv showed specific binding signature on the membrane of HCC cell and positive correlation with cell concentration using fluorescence confocal microscopy and flow cytometry, respectively. A higher binding affinity of scFv over the original monoclonal antibody was also displayed. After the immobilization onto the screen-printed electrode through the high affinity between histidine and nickel-based nanomaterial, the constructed cell sensor exhibited a wide detection range of 102 to 107 cells ml−1 and a low detection limit of 2 cells ml−1 with a short incubation time of 5 min. The high selectivity in the liver tissue extract fluid derived from the design strategy was also displayed in the mouse samples. The customized scFv and related cell sensor demonstrated great potential for fast HCC analysis in early-stage cancer diagnosis.
Ultrasensitive electrochemical detection of hepatitis C virus core antigen using terminal deoxynucleotidyl transferase amplification coupled with DNA nanowires

Abstract:Early diagnosis of hepatitis C virus (HCV) infection is essential to prevent disease from spreading and progression. Herein, a novel electrochemical biosensor was developed for ultrasensitive detection of HCV core antigen (HCVcAg) based on terminal deoxynucleotidyl transferase (TdT) amplification and DNA nanowires (DNW). After sandwich-type antibody-antigen recognition, the antibody-conjugated DNA was pulled to the electrode surface and further extended into a long DNA sequence by robust TdT reaction. Then, large numbers of methylene blue–loaded DNW (MB@DNW) as signal labels are linked to the extended DNA sequence. This results in an amplified electrochemical signal for HCVcAg determination, typically measured at around −0.25 V (Ag/AgCl). Under the optimum conditions, the proposed biosensor achieved a wide linear range for HCVcAg from 0.1 to 312.5 pg/mL with a low limit of detection of 32 fg/mL. The good practicality of the biosensor was demonstrated by recovery experiment (recoveries from 98 to 104% with RSD of 2.5–4.4%) and comparison with enzyme-linked immunosorbent assay (ELISA). Given the highlighted performance, the biosensor is expected to act as a reliable sensing tool for HCVcAg determination in clinics.

Abstract:Hepatitis E virus (HEV) is one of the leading causes of acute viral hepatitis worldwide. In this work, a pulse-triggered ultrasensitive electrochemical sensor was fabricated using graphene quantum dots and gold-embedded polyaniline nanowires, prepared via an interfacial polymerization and then self-assembly approach. Introducing an external electrical pulse during the virus accumulation step increases the sensitivity towards HEV due to the expanded surface of the virus particle as well as the antibody-conjugated polyaniline chain length, compared to other conventional electrochemical sensors. The sensor was applied to various HEV genotypes, including G1, G3, G7 and ferret HEV obtained from cell culture supernatant and in a series of fecal specimen samples collected from G7 HEV-infected monkey. The sensitivity is similar to that detected by real-time quantitative reverse transcription-polymerase chain (RT-qPCR). These results suggests that the proposed sensor can pave the way for the development of robust, high-performance sensing methodologies for HEV detection.

Abstract:Herein, we have demonstrated highly sensitive real-time biospecific detection of a protein marker of hepatitis C—the core antigen of hepatitis C virus (HCVcoreAg)—using a nanowire (NW) biosensor. The primary element of the NW-biosensor is a chip with p-type conductance, bearing silicon-on-insulator (SOI) nanowire structures on its surface. The nanowire structures are fabricated by gas-plasma treatment and electron beam lithography. The detection specificity was provided by sensitization of the sensor surface with aptamers against HCVcoreAg. The influence of buffer pH on the sensor response signal was studied. The effect of reverse polarity of the biosensor response signal with change from the acidic buffer pH to the neutral one was found. The lowest detectable HCVcoreAg concentration was determined to be 2.0 × 10−15 M in both acidic (pH 5.1) and neutral (pH 7.4) buffer solution. The proposed aptamer-sensitized sensor was also successfully applied to detect HCVcoreAg in serum samples of hepatitis C patients.

Abstract:Pt nanoparticles deposited on single-walled carbon nanotubes (PtSWCNTs), synthesized via the deposition precipitation (DP) method, were introduced as a substrate for immobilizing antibodies on an electrode surface and then enhancing the electrochemical sensitivity. A PtSWCNT-modified paper-based screen-printed graphene electrode was successfully developed to diagnose hepatitis C virus (HCV) infection. The hepatitis C virus core antigen (HCV-cAg) level was determined by differential pulse voltammetry (DPV) using [Fe(CN)6]3-/4- as a redox solution. In the presence of HCV-cAg, the DPV current response decreased with increasing HCV-cAg concentration. Under the optimal conditions, the change in current response provides a good linear correlation with the logarithm of HCV-cAg concentration in the range 0.05 to 1000 pg mL-1 (RSD < 5%), and the limit of detection was 0.015 pg mL-1 (or 0.71 fmol L-1). Furthermore, the proposed immunosensor has been utilized to quantify HCV-cAg in human serum samples with reliable results compared with standard immunoassays (% relative error < 10%). This sensor offers a simple, sensitive, selective, disposable, and inexpensive means for determination of HCV-cAg in human serum samples. The paper-based label-free immunosensor is versatile and feasible for clinical diagnosis.
Dual-label vertical flow-based electrochemical immunosensor for rapid and simultaneous detection of hepatitis B surface and e virus antigens

Abstract:The detection of hepatitis B virus (HBV)-associated biomarkers, especially hepatitis B surface (HBsAg) and hepatitis B e (HBeAg) antigens, is of critical significance for the early diagnosis and monitoring of disease progression. However, the detection of these biomarkers has seldom been reported. Herein, we first present a dual-label electrochemical vertical flow-based immunosensor (DL-eVFIS) for the simultaneous detection of HBsAg and HBeAg. DL-eVFIS was engineered to integrate multiple time-sequenced detection procedures into a single device, thus considerably shortening the overall assaying time to 6 min. To enable simultaneous determination, thionine (Th) and ferrocenecarboxylic acid (Fc) were employed as signal tags, as they demonstrate nonoverlapping specific reduction peaks. The appearance of target antigens resulted in a substantial increase in the electrochemical signal response of the redox tags. While requiring only a miniscule sample volume (5 μL), superior performance was achieved in terms of their limits of detection (LODs) at 1.66 pg/mL for HBsAg and 5.32 pg/mL for HBeAg. DL-eVFIS further demonstrates excellent specificity and selectivity in clinical sample analysis compared with the conventional method. This study contributes to next-generation point-of-care (POC) testing devices for HBV diagnosis and a wide range of biosensing applications.

A Cell-Based Electrochemical Biosensor for the Detection of Infectious Hepatitis A Virus

Abstract:Hepatitis A virus (HAV), a major cause of acute liver infections, is transmitted through the fecal–oral route and close contact with infected individuals. Current HAV standardized methods rely on the detection of virus antigen or RNA, which do not differentiate between infectious and non-infectious HAV. The objective of this study was to develop a prototype cell-based electrochemical biosensor for detection of infectious HAV. A cell culture-adapted HAV strain (HM175/18f) and its permissive cells (FRhK-4), along with gold nanoparticle-modified screen-printed electrodes, were used to develop the biosensor. Electrochemical impedance spectroscopy was used to quantify the electrical impedance signal. Nyquist plots showed successful fabrication of the cell-based biosensor. The optimum period of HAV incubation with the biosensor was 6 h. A significant linear relationship (R2 = 0.98) was found between the signal and a 6-log range of HAV titers, with a limit of detection of ~5 TCID50/mL (tissue culture infectious dose). The biosensor did not detect non-target viruses such as feline calicivirus and human coronavirus 229E. The biosensor was stable for 3 to 7 days at an abusive temperature (37 °C), retaining ~90 to 60% of the original signal, respectively. In conclusion, this prototype cell-based biosensor is capable of rapidly detecting low levels of infectious HAV.

Abstract:A burgeoning diversified closed bipolar electrochemiluminescent (d-BPE-ECL) based on photothermal amplification biosensor via the thermophysical and photochemical properties of niobium carbide Mxene quantum dots (Nb2C MQDs) has been proposed. The device consists of three components: separated intermediate recognition, cathodic catalytic hydrogen evolution reaction (HER) and anodic ECL response channel. Wherein, the recognition compartment was innovatively designed as a temperature-sensitive conductivity modulated interface, and the introduction of photothermal material PDA@Nb2C MQDs through target mediated rolling circle amplification strategy increases the interface temperature under near-infrared light radiation, thereby enhancing the BPE current and leading to the amplification of the anode ECL signal of Nb2C MQDs. In addition, MoS2@Ni-Cu-P features excellent electrocatalytic activity, which can promote HER and thus accelerate electron transfer, further amplifying the ECL signal. Therefore, a highly sensitive d-BPE-ECL biosensor for hepatitis C virus envelope protein E2 detection with a linear range from 10-4 to 10 ng/mL and detection limit of 3.3 × 10-5 ng/mL was obtained. This work is expected to provide a new direction for exploring BPE multiple signal amplification strategy and broaden the application of BPE-ECL in bioassays.

Abstract:Highly sensitive simultaneous detection of dual-target is one of the main research directions of electrochemiluminescence (ECL), but the traditional three-electrode system makes it difficult to realize this aim. This work designed a multifunctional spatial-potential resolved ECL biosensor for dual-channel simultaneous detection of hepatitis A virus (HAV) and hepatitis C virus (HCV) DNA. Self-luminescent dual-ligand metal organic framework (D-MOF) and cerium MOF (Ce-MOF) were modified on two parts of the ITO electrode to achieve dualchannel simultaneous detection. One ligand of D-MOF acted as the luminophore and the other as the co-reactant, so D-MOF could emit excellent positive potential ECL signal without requiring external co-reactants. In channel A, the Ag nanoparticles (Ag NPs) quenching probes were introduced to D-MOF through hybridization of HAV with "Y" type DNA. Positive potential signal of D-MOF was decreased for HAV DNA detection based on resonance energy transfer (RET). In channel B, the CdSe quantum dots (QDs) were introduced to Ce-MOF through hybridization of HCV with "Y" type DNA. Due to the excellent ECL performance of CdSe QDs, negative potential signal was increased for HCV DNA detection. In this work, an ingenious spatial-potential resolved ECL biosensor was developed for simultaneous detection of dual-target by simple sealing film isolation, which opened a new strategy for dual-target detection in general ECL assays with three-electrode system.
Bioreceptors' immobilization by hydrogen bonding interactions and differential pulse voltammetry for completely label-free electrochemical biosensors

Abstract:Label-free electrochemical biosensors show great potential for the development of point-of-care devices (POCDs) for environmental and clinical applications. These sensors operate with shorter analysis times and are more economic than the labelled ones. Here, four completely label-free biosensors without electron transfer mediators were developed for hepatitis B virus (HBV) detection. The approach consisted in (i) the modification of gold surfaces with cysteamine (CT) or cysteine (CS) linkers, (ii) the subsequent antibody (Ab) immobilization, either directly by hydrogen bonding (HB) interactions or by covalent bonds (CB) using additional reagents, and (iii) measuring the biosensor response by electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV). The electrode surfaces at each stage of the modification process were characterised by X-ray photo-electron spectroscopy (XPS) and atomic force microscopy (AFM). The combination of Ab immobilization by HB with the DPV analysis displayed improved repeatability, lower interference to serum matrix and similar limits of detection and quantification than the traditional biosensors that immobilize the Ab via CB and use EIS as readout technique. The Ab immobilization by HB is shown as a simple, efficient and low-cost alternative to CB ones, while DPV was faster and showed better performance than EIS. The CT-HB biosensor displayed the lowest limits of detection and quantification of 0.14 and 0.46 ng/mL, respectively, a 0.46-12.5 ng/mL linear analytical range, and 100% of recovery for 1/10 human serum media during HBV surface antigen detection by DPV. Even, it preserved the initial sensing capability after 7 days of its fabrication.

Abstract:The ultrasensitive detection of hepatitis C virus (HCV) nucleic acid is crucial for the early diagnosis of hepatitis C. In this study, by combining Ag@Au core/shell nanoparticle (Ag@AuNP)-based surface-enhanced Raman scattering (SERS) tag with hybridization chain reaction (HCR), a novel SERS-sensing method was developed for the ultrasensitive detection of HCV nucleic acid. This SERS-sensing system comprised two different SERS tags, which were constructed by modifying Ag@AuNP with a Raman reporter molecule of 4-ethynylbezaldehyde, two different hairpin-structured HCR sequences (H1 or H2), and a detection plate prepared by immobilizing a capture DNA sequence onto the Ag@AuNP layer surface of the detection wells. When the target nucleic acid was present, the two SERS tags were captured on the surface of the Ag@AuNP-coated detection well to generate many "hot spots" through HCR, forming a strong SERS signal and realizing the ultrasensitive detection of the target HCV nucleic acid. The limit of detection of the SERS-sensing method for HCV nucleic acid was 0.47 fM, and the linear range was from 1 to 10 5 fM.

Abstract:The hepatitis C virus (HCV), a major causative agent of chronic liver disease, is responsible for the incidence of hepatitis infections worldwide. In line with the World Health Organization's goal of reducing global new infections by 90% and viral hepatitis-related mortality rates by 65% by 2030, we proposed a miniaturized indium tin oxide-based extended gate field-effect transistor biosensor for rapid and specific detection of different HCV genotypes within 20 min. The biosensor combined loop-mediated isothermal amplification (LAMP) and clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 12 (CRISPR/Cas12a)-induced cleavage. The immobilized single-stranded DNA (ssDNA) served as a signal transmitter and underwent CRISPR/Cas12a trans-cleavage, which was triggered by specific nucleic acid pairing with complementary guide RNAs. This resulted in a significant change in the surface potential, which indicated the presence of HCV-viral RNA. The sensor demonstrated high sensitivity, with a detection limit as low as 1 genomic copy/reaction and a 10-fold increase in robustness for low concentration tests. The clinical results are consistent with those of the polymerase chain reaction. This research offered a rapid, sensitive, and digital diagnostic method for HCV viral infections, particularly suited for resource-limited settings, contributing to the elimination of hepatitis.

Abstract:As the common hepatitis viruses, hepatitis B virus (HBV) and hepatitis C virus (HCV) are important causes of liver diseases, which greatly threaten human health and public hygiene. Furthermore, due to the similar transmission routes of HBV and HCV, it is highly susceptible to suffer from the cross-infection of HBV and HCV when humans are infected by hepatitis viruses. So it is urgent to develop strategies with high sensitivity to distinguish HBV/HCV and detect their overlapping infection. Herein, a new biosensor named "hepatitis virus indicator" for simultaneous detection of HBV and HCV based on the automatic particle enumeration was developed by the combination of Exo III assisted signal amplification strategy and particle counting technology with a dark-field microscopy. The limit of detections (LOD) were as low as 0.194 pM for HBV and 0.169 pM for HCV, respectively. In addition, the proposed method was capable of simultaneously detecting HBV and HCV based on the colorful light scattering from plasmonic nanoparticles. The proposed assay was simple, sensitive and selective for the simultaneous detection of HBV and HCV, which provided a new idea for the early diagnosis of diseases, such as hepatitis A virus (HAV), hepatitis D virus (HDV) and hepatitis E virus (HEV).

Abstract:Hepatitis B Surface Antigen (HBsAg) is usually recognized as an earliest diagnostic markers for Hepatitis B Virus (HBV) infections. The precise and sensitive determination of HBsAg is crucial for the diagnostic of HBV infection. In this paper, a colorimetric immunosensor has been successfully designed and fabricated based on conjugating magnetic separation and enzymatic catalysis, for point-of-care detection of HBsAg. The HBsAg was simultaneously captured by the biotinylated HBsAg primary antibody (biotin-Ab1) labeled immunomagnetic nanoparticles and detection antibody (Ab2). In conjugation with Ab2, horseradish peroxidase (HRP) labeled rabbit anti-goat secondary antibody was linked with chromogenic agents to induce the colorimetric signal. As a result, a good linear relation was achieved between the concentration of HBsAg and OD450 nm (absorbance values at 450 nm) in the 0.1–20 ng mL−1 ranges, demonstrating a great specificity to HBsAg with a limit of detection of 0.012 ng mL−1. Furthermore, considering its succeed in real human serum sample test, this immunosensor could make contribute to potential applications in rapid diagnosis, effective prevention of epidemic and infectious diseases.

Establishment and methodological evaluation of a chemiluminescence assay for detection of anti‐envelope protein (E1, E2) antibodies in the serum of hepatitis C virus‐infected patients

Abstract:Abstract Background To establish a chemiluminescence method for detecting anti‐E1 and anti‐E2 antibodies in the serum of patients with hepatitis C virus (HCV) infection. Methods The microplate was coated with recombinant envelope proteins E1 and E2 by indirect method, respectively, and the kits for detecting anti‐E1 and anti‐E2 antibodies were prepared. The methodological indexes were evaluated. Results The methodological indexes of the kits were as follows: precision test (the variation coefficient of anti‐E1 antibody 6.71%–8.95% for within run and 9.91%–12.16% for between run, the variation coefficient of anti‐E2 antibody 6.06%–8.44% for within run and 10.77%–13.98% for between run, respectively). The blank limit and detection limit were 1.18 RLIR and 3.16 RLIR for the anti‐E1 antibody, and 1.26 RLIR and 3.32 RLIR for the anti‐E2 antibody, respectively. The correlation coefficients ( r ) of anti‐E1 and anti‐E2 were 0.9963 and 0.9828, the analysis and measurement ranges (AMR) were 1.66–41.28 RLIR and 1.55–19.46 RLIR, and the average recovery was 96.4% and 93.7%, respectively. The rheumatoid factor and other positive serum samples had no interference or cross‐reaction to the test, and the kits were stable within 15 months. The positive rates of anti‐E1 and anti‐E2 antibodies in 45 patients with HCV infection were 35.6% (16/45) and 44.4% (20/45), respectively. Conclusions The kits for detecting anti‐E1 and anti‐E2 meet the requirements of methodology, and can be used in screening diagnosis, disease monitoring, prognosis evaluation, disease mechanism, and epidemiological studies of HCV infection. The HCV envelope proteins E1 and E2 have an immune response in HCV‐infected patients.

Abstract:Currently, PCR is the gold standard for the detection of hepatitis C virus (HCV). However, the PCR technique is complicated and time-consuming, which prevents its application and, clinical point-of-care testing (POCT). Herein, we report a POCT method with simplicity, high sensitivity and specificity, which consists of a catalytic hairpin assembly (CHA) signal amplification system coupled with a lateral flow immunochromatographic (LFIA) test strip for the detection of HCV. Two ingeniously designed hairpin probes were hybridized to form the H1-H2 duplex in the presence of the target DNA. The catalytic hairpin assembly which was characterized of isothermal and enzyme-free, was accomplished within 40 min and the reaction was then applied to a LFIA test strip. Only the H1-H2 duplex labeled with both digoxin and biotin could be captured by the test strip, and the fluorescence value was determined. In addition, we evaluated the application potential for the detection of clinical samples. The reported method demonstrated high sensitivity with a detectable minimum concentration at 1 fM and showed a good linear range from 10 nM to 10pM, and high specificity for various mismatched sequences. The results demonstrated that clinically positive samples could be successfully detected. In conclusion, the reported method is simple, rapid, and free of large-scale equipment. POCT is expected to be useful for HCV detection in clinic.

Abstract:We study almost F -preenvelopes in the category of rings, for a significative class F of commutative rings. We completely identify those rings which have an almost F -preenvelope when F is the class of fields, semisimple rings, integer domains and local rings. We show that rings with Krull dimension zero have (almost) V-preenvelopes, where V is the class of von Neumann regular rings. key words. Abstract:The authors describe an electrochemical immunoassay for the core antigen of hepatitis C virus (HCV). The method is based on the use of a screen-printed carbon electrode (SPCE) that was modified with a Nafion@TiO2 nanocomposite and loaded with secondary antibody (Ab2) to entrap Celestine Blue (CB). The material has architecture of the type CB/Ab2/Nafion@TiO2. A nanocomposite consisting of graphene, ionic liquid and fullerene was deposited on the SPCE first, and rhodium nanoparticles (RhNPs) were then deposited on the surface of modified electrode in order to immobilize primary antibody (Ab1). The antigen and CB/Ab2/Nafion@TiO2 were conjugated one by one to form a sandwich-type immunocomplex. The signal was obtained by differential pulse voltammetry whose intensity is related to the concentration of the antigen. The assay, if operated at a working voltage of typically 0.35 V (vs. Ag/AgCl) has a response that is linear in the 0.1 to 250 pg⋅mL−1 HCV core antigen concentration range, and the limit of detection is as low as 25 fg⋅mL−1. The assay was applied to the determination of the HCV core antigen in spiked human serum samples. In our perception, the method represents a promising platform for the detection of various antigens if appropriate antibodies are available.Graphical abstractAn electrochemical immunoassay for the core antigen of Hepatitis C virus was studied.
