Abstract:Structural evolution, digestibility and inhibition on starch digestion of rice glutelin fibril aggregates (RGFAs) as affected by incubation were investigated. Thioflavin T fluorescence intensity of the RGFAs, incubated for 4-day, reached the maximum values, which ranged from 845.00 ± 23.52 to 873.67 ± 50.30. Transmission electron microscopy (TEM) observed that the samples heated for 2 h (2 h fibril) were self-assembled from small glutelin aggregates and a few protofibrils into mature fibrils, the samples heated for 4–10 h (4–10 h fibril) were elongated into long, branched fibrils, and the longer fibrils of 15 h fibril sample dissociated into short fibrils after 4-day of incubation. Compared to rice glutelin, the RGFAs showed thermal stability and resistance to proteolysis. The fluorescence retention rate of 6 h fibril, after incubation for 4-day, was 8.62 ± 0.61 % after in vitro stomach and pancreas digestion, which was the highest among all of the samples. The RGFAs incubated for 1-day displayed much better inhibition effects on starch digestion. This was the first study to clarify the relationship between incubation and physicochemical/functional properties of protein fibrils, which could help understand the preservation of food protein fibrils and their application.

Abstract:Plant-derived proteins like rice protein are considered an ideal source for yielding amyloid fibrils due to sustainability and affordability. However, the inherent low solubility of rice protein often restricts its self-assembly capability under common fibrillization protocols. Hence, this study comprehensively investigated the effect of modification on the self-assembly behavior, protein structure and foaming properties of rice protein and its components. The modification facilitated the transformation of alpha-helix to beta-sheet, promoting fibrils formation. All modified proteins, except rice protein, exhibited significantly increased zeta potential values. AFM results revealed that the modified rice protein and globulin fibrils exhibited entangled aggregation behavior, while the glutelin fibril was notably extended, with several micrometers in length. Additionally, modification promoted the aggregation of albumin and prolamin particles to form short and straight fibrils. Amino acid analysis indicated that the content difference of amino acids with different characteristics and the change of aromatic amino acid interaction caused by modification may lead to different self-assembly behavior of the same protein. Furthermore, the foaming capability and stability of most protein fibrils were significantly improved by the modification. These findings provide valuable insights into the regulation of plant-derived protein fibrillation through modification.

Abstract:This work studied effects of different amounts of rice glutelin (RG) on physicochemical and structural properties of extruded rice starch (ERS) and explored the underlying mechanism of interaction between rice starch and RG upon extrusion processing. The results showed that the addition of RG altered the pasting properties, improved the viscoelastic, and increased the water mobility of ERS. The weight loss of ERS decreased from 71.40 % to 62.61 %, while the degradation temperature increased from 290.48 °C to 296.25 °C as the RG content increased from 0 % to 12 %. The complex index of extruded starch-glutelin complexes significantly elevated from 10.40 % to 35.81 % when RG content increased from 6 % to 12 %. Fourier-transform infrared spectra confirmed that RG interacted with starch via Maillard reactions, and the binding strength between RG and starch was enhanced at a higher RG content. Furthermore, results of rheological property and chemical interactions demonstrated that hydrogen bonding, hydrophobic, and electrostatic interaction were formed between RG and starch during extrusion. In summary, the obtained results of this study can further enrich the theory of starch-protein interactions and show the possibility of RG applied in the extruded starchy foods.

Inhibition mechanism of rice glutelin on extruded starch digestion: From the structural properties of starch and enzyme activity

Abstract:To increase the anti-digestion ability of extruded rice starch (ERS), the influence of rice glutelin (RG) on digestive and structural characteristics of ERS were investigated. The resistant starch content increased from 4.49 % to 18.08 % as the RG content increased, while the digestion rate and digestion velocity constant were reduced by the incorporation of RG. Morphological observations showed that ERS was adhered and encapsulated by RG, and the specific area of starch granules were decreased after the addition of RG. The results of XRD and FTIR suggested that the long-range and short-range orders of ERS were improved due to the complexation with RG. The thickness of crystalline of ERS was increased while its amorphous region thickness was reduced by the supplementation with RG. The 1H NMR and 13C NMR data revealed that the branching degree and double helix content of ERS was increased by 46.24 % and 52.67 % when RG content reached to 12 %. Additionally, the addition of RG altered the molecular weight and chain length distribution of ERS. The α-amylase activity and glucoamylase activity was inhibited by RG. These results could provide a valuable basis for the application of RG in extruded rice starchy foods with lower glycemic index.
Structural Characterization and Binding Interaction of Rice Glutelin Fibrils Complexing with Curcumin

Abstract:The rice glutelin (RG), the separated retentate (RGFs) and filtrate (FGFs) fractions from total glutelin fibrils (TGFs) at pH 3.5 were used as carrier for curcumin in this test. The solubility and antioxidant activities of curcumin were improved after binding with protein and fibrils. Compared to other complexes, the RGFscurcumin complex exhibited a highest curcumin solubility (48.05%) and a superior sustained release property, probably owing to the stable hydrogen bond between the surface groups of fibrils and hydroxyl groups of polyphenols. In addition, thermodynamic parameters revealed that the RG/TGFs/RGFs-curcumin complexes were stabilized by hydrogen bonds and van der Waals forces, whereas FGFs interacted with curcumin through specific electrostatic interaction. Besides, after interacting with curcumin, the fibrils gathered into coarsened and agglutinated fibrillar aggregates, relating to the increment of a -helix and beta -sheet structure. These results suggested that RGFs could be a good alternative for curcumin delivery in food industry.

Abstract:The influence of the degree of hydrolysis (DH) on the structure, solubility, rheology, and emulsifying properties of rice glutelin was investigated. Structural properties were characterized using analysis of molecular weight distribution, surface hydrophobicity, intrinsic fluorescence, and Circular Dichroism (CD) spectra. Protein hydrolysis changed molecular weight, increased flexibility, altered surface hydrophobicity, and increased solubility. Oil-in-water emulsions were prepared from rice glutelin with different DH (0.5%, 2%, and 6%) and their stability to storage, pH, salt, and thermal processing was assessed. The storage, pH, salt, and temperature stability of the emulsions increased with decreasing hydrolysis. Emulsions prepared with 2% DH rice glutelin were stable over a range of environmental conditions: pH 7–9; NaCl <100 mM (pH 7); temperatures < 90 °C (pH 7, 0 mM NaCl) studied. These results will facilitate the formulation and production of natural emulsion-based products using rice glutelin as an emulsifier.

Abstract:Reconstituted rice produced by extrusion has been attracted attention due to nutritional fortification and convenient production. Nevertheless, how to achieve desirable qualities and physicochemical properties of reconstituted rice nearly to natural rice by regulating extrusion process parameters is difficult. Herein, rice starch/glutelin mixture as raw material of reconstituted rice was extruded at varying extrusion conditions. Specific mechanical energy (SME) and sectional expansion index (SEI) dropped with rise in density (R2 = 0.9117 and 0.8207). Solubility was enhanced with increase in product temperature (R2 = 0.9085), color darkened and shifted to reddish and yellowish as extrusion temperature increased (R2 = 0.8577). These trends were well fitted by sigmoid models. Furthermore, SME enhanced hydrophobic and electrostatic interactions between rice starch and glutelin and caused the reduction in crystallinity and thermal stability, promoting the formation of a bi-continuous matrix of protein aggregates with rice starch. The obtained results can be applied to guide the production of reconstituted rice with desirable qualities.

Abstract:Metal ions have been considered as an important factor on fibrils assembly. Herein, a comprehensive analysis of specific ions effect on fibril formation and structural changes was investigated. The addition of ions (except Zn2+) accelerated the aggregation kinetics of rice glutelin fibrils (RGFs) from 0.93 to 1.28-2.19 h(-1). In addition, the fibrillization rate followed the order of NH4+ > Li+ > Na+ > K+ > Cu2+ > Mg2+ > Ca2+ > Zn2+. The highest yield and length of fibrils were observed with Ca2+, probably due to the ionic bridging effect and hydrated capacity of Ca2+. However, Cu2+ reduced the fibrils yield, which was attributable to the fact that Cu2+ disrupted beta-sheet structure and inhibited the transition of monomer to fibrils. The polymorphism of fibrils was observed with different salts, and the light metals presented a superior effect on fibrils formation than heavy metals. Overall, this work will provide a further information into how to tune the structure of RGFs using various ions.
Effects of enzyme hydrolysis-assisted fibrillation treatment on the solubility, emulsifying properties and antioxidant activity of rice protein

Abstract:This work aimed to explore the changes of rice protein (RP) in solubility, emulsifying properties, and antioxidant activity after the enzyme hydrolysis-assisted fibrillation dual modification. Results showed that enzyme hydrolysis by papain and fibrillation treatments significantly affected the secondary and tertiary structures of RP. The modified proteins, including RP hydrolysate (RPH), RP nanofibrils (RPN), and RPH nanofibrils (RPHN), demonstrated enhanced solubility and antioxidant activity compared to RP, with RPHN exhibiting the superior performance. The emulsifying capacity of RPH, RPN, and RPHN increased by 9.55 %, 22.86 %, and 26.57 %, respectively, compared to that of RP. Furthermore, RPHN displayed the highest emulsion stability index. Nanoemulsion stabilized by RPHN showed enhanced centrifugal, storage, and oxidative stabilities. Neither RPHN nor RPN exhibited cytotoxicity to human cell lines, and could provide nutrients for cells. Overall, the functional properties and antioxidant activity of RP were significantly improved by enzyme hydrolysis-assisted fibrillation dual modification. This study may provide reference for the development and utilization of nanofibrils from plant proteins.

Abstract:The objective of this study was to improve the physical stability of emulsions containing oil droplets stabilized by hydrolyzed rice glutelin (HRG) by coating them with anionic polysaccharides. Polysaccharide type and concentration had a strong influence on the properties of the coated droplets. Smaller particles with better pH-stability could be formed from xanthan gum or pectin than from alginate or gum arabic. The impact of environmental stresses on the stability of oil droplets coated with HRG-xanthan gum or HRG-pectin complexes was tested by measuring the impact of salt addition (0–500 mM NaCl, pH 3.5) and thermal processing (30–90 °C 30 min, 0 mM NaCl, pH 3.5). The emulsions containing xanthan gum were stable at all NaCl concentrations, whereas those containing pectin were only stable at low salt levels. Emulsions containing xanthan gum or pectin were stable to thermal processing. The improved stability of the emulsions containing the polysaccharides can be attributed to an increase in electrostatic and steric repulsion between the droplets. These results are useful for expanding the application of rice glutelin as a functional ingredient in the food industry.

Abstract:Rice gluten, as the hydrophobic protein, exhibits restricted application value in hydrophilic food, which may be enhanced through interaction with soybean 11S globulin, characterized by favorable functional properties. This study aims at revealing their interaction mechanism via multi-spectroscopy and molecular dynamics simulation. The formation and structural change of rice glutelin-soybean 11S globulin complexes were detected using fluorescence, ultra-violet and circular dichroism spectra. The addition of 11S globulin increased the contents of α-helix, β-turn and random coil, but decreased β-sheet content, and the change in secondary structure was correlated with particle size. Moreover, exposure of hydrophobic groups and formation of disulfide bonds occurred in the complexes. Molecular dynamics simulation verified these experimental results through analyses of root mean square deviation and fluctuation, hydrogen bond, secondary structure, and binding free energy analysis. This study contributes to expounding the interaction mechanism of protein and protein from the molecular level.
The Role of Indica Starch in the Mechanism of Formation of Fresh Rice Noodles

Abstract:Starch is the largest component in frequently consumed fresh rice noodles (FRN). The effects of morphology, relative crystallinity, molecular weight distribution and physicochemical (rheological, water hydration and gel texture) properties of starch from five rice cultivars with different amylose contents on the formation of FRN were investigated. FRN made from NR rice cultivar, used for commercial rice noodles making, showed excellent FRN texture. NR starch exhibited larger G ' and smaller tan delta values. The morphology of FRN from GM03 cultivar was closest to NR, but had higher hardness and lower springiness, resilience and cohesiveness. Its starch granules morphology was relatively small and round. The extracted starch from Zhongzao 35, Zao 87 and BX02 rice cultivars, with higher swelling power or lower proportions of amylose (FrI) and higher proportions of amylopectin (FrII), could not produce a dense and consistent FRN structure. It was observed that relative crystallinity of the starch samples had no influence on the formation of FRN. Nevertheless, the gel texture of starch played a role in the formation of FRN. The formation of FRN is the common effect of structure and physicochemical properties of rice starch. The finding of this research would be useful in understanding the role of starch in producing FRN of high quality.

Abstract:Dengue is the most prevalent mosquito-borne viral infection. Recent outbreaks in the southern United States illustrate the risk of reemergence. The first autochthonous cases since 1934 in Key West, FL, occurred in 2009-2010. We conducted a survey in 2012 with decision makers instrumental to the control of the outbreak to 1) determine their awareness of the multiple strategies used to control the outbreak and 2) assess their perceptions of the relative effectiveness of these strategies. An online survey was delivered to a predefined list of decision makers from multiple sectors to better understand dengue preparedness and response. Thirty-six out of 45 surveys were returned for an 80% response rate. Results indicate the need to focus prevention strategies on educational campaigns designed to increase population awareness of transmission risk. Respondents remain concerned about future dengue transmission risk in Key West and lack of resources to respond.
Formation kinetics and fractal characteristics of regenerated silk fibroin alcogel developed from nanofibrillar network

Abstract:The formation mechanism of RSF alcogel was elucidated from morphological observations using TEM, AFM and confocal laser scanning microscopy (CLSM) as well as the kinetic studies by Thioflavin T fluorescence and rheological methods. An intriguing dual-structure-a nanofibrillar network on the nanoscale and a floc-like network on the microscale-was observed within the RSF alcogel. Based on our kinetic study, the fractal dimension of the ramified fibrillar aggregate was found to be 2.4-2.8 depending on RSF concentration. Meanwhile, through a scaling analysis of the mechanical properties of RSF alcogels, the fractal dimension of the floc-linked network was found to be 2.2. Furthermore, our results showed that fibrillarization of RSF was triggered by a dramatic change in the interaction between RSF and cosolvent, and the gelation kinetics could be tuned by adjusting the concentration of ethanol as well as RSF. The controllable dual-structure and kinetics make such biopolymer gel a potential candidate for applications in nanotechnology.
Stability, Structure, Rheological Properties, and Tribology of Flaxseed Gum Filled with Rice Bran Oil Bodies

Abstract:In this study, rice bran oil bodies (RBOBs) were filled with varying concentrations of flaxseed gum (FG) to construct an RBOB-FG emulsion-filled gel system. The particle size distribution, zeta potential, physical stability, and microstructure were measured and observed. The molecular interaction of RBOBs and FG was studied by Fourier transform infrared spectroscopy (FTIR). In addition, the rheological and the tribology properties of the RBOB-FG emulsion-filled gels were evaluated. We found that the dispersibility and stability of the RBOB droplets was improved by FG hydrogel, and the electrostatic repulsion of the system was enhanced. FTIR analysis indicated that the hydrogen bonds and intermolecular forces were the major driving forces in the formation of RBOB-FG emulsion-filled gel. An emulsion-filled gel-like structure was formed, which further improved the rheological properties, with increased firmness, storage modulus values, and viscoelasticity, forming thermally stable networks. In the tribological test, with increased FG concentration, the friction coefficient (μ) decreased. The elasticity of RBOB-FG emulsion-filled gels and the ball-bearing effect led to a minimum boundary friction coefficient (μ). These results might contribute to the development of oil-body-based functional ingredients for applications in plant-based foods as fat replacements and delivery systems.

Abstract:The effects of stirring speed (0, 250, 500, 750, 1000, 1250, and 1500 rpm) on the rice glutelin hydrocolloids (1 %, w / v ) during the acidified process were investigated. As the stirring speed was increased to 750 rpm, the hydration diameter of the rice glutelin was significantly decreased, but higher stirring speeds had no significant effect on size. The highest and lowest solubility were recorded for the samples treated at 750 and 0 rpm stirring speeds, respectively. The surface hydrophobicity and molecular weight increased first and then decreased, both the minimum value was recorded at 750 rpm sample. The principal component analysis (PCA) was employed to detect patterns between changes in various properties (solubility, particle size, β-sheet content, surface hydrophobicity, and ζ-potential) and stirring treatment. To conclude, the various properties of rice glutelin refold during acidification are drastically affected by employing different stirring speeds. Choosing a suitable stirring speed is important for quality control in protein hydrocolloid production.

Abstract:Glutinous rice is extensively consumed due to its nutritious content and wonderful flavor. However, glutinous rice flour has a high glycemic index, and the storage deterioration of sweet dumplingsissevere. Transglutaminase (TG) was used to cross-link glutinous rice protein and improve the characteristics of glutinous rice products. The findings demonstrated that TG significantly catalysed protein cross-linking to form a dense protein network, reduced the viscosity of glutinous rice paste and improved the thermal stability. The protein network may physically block the access of starch granules to digestive enzymes to lower the digestion rate of starch, and attenuate the damage of ice crystal molecules to the starch structure to improve the freezing stability of starch gels. The cracking rate and water loss of sweet dumplings prepared using glutinous rice flour with TG treated for 60 min reduced significantly. In conclusion, this study broadened the application of TG in starch products.

The formation of γ-glutamyl-lysine bonds in RNS-COM and RNS-MTG was shown by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Scanning electron microscope showed that compared to others, the structure of RNS-COM was denser, smoother with extensive apparent interconnectivity of aggregates. The optimum cooking time was in the order: YAN > RNS-COM > RNS-MTG > RNS-GDL > RN (rice flour noodles); tensile strength was in the order: YAN > RNS-COM > RNS-MTG > RNS-GDL > RN; and elasticity were in the order: YAN > RNS-COM > RNS-MTG, RNS-GDL > RN. Overall, RNS-COM showed similar textural and structural breakdown parameters as compared to those of YAN. Changes in microstructures and improvement of RNS-COM in certain properties were likely due to enhanced crosslinking of proteins attributed to MTG- and GDL-induced cold gelation of proteins at reduced pH value. It is possible to use the combination of MTG and GDL to improve textural and mechanical properties of RNS comparable to those of YAN. PRACTICAL APPLICATION: Combined MTG and GDL yield rice flour noodles with improved textural properties. The restructured rice flour noodles have the potential to replace yellow alkaline noodles.
