Abstract:This study aimed to evaluate the gelation characteristics of soybean protein hydrolysate (SPH) extracted by enzyme-assisted aqueous extraction. Specifically, the changes in gelation behaviors for heat-induced (95°C, 20 min) SPH dispersions treated with pH (pH 3, 5, 9; pH 7 as control) and ultrasound (U; 240 W, 30 min) were investigated. The results showed that typical gel behavior with high elastic nature in the viscoelasticity and network structures were observed during the heating process, where the disulfide bond played a dominant role in the gel network formation of all the samples. Notably, the heat-induced aggregation in the SPH gels was mainly formed by the association of the basic B polypeptide in 11S and β subunit in 7S. The most superior SPH gel was formed at pH 7 when assisted by ultrasonication during the heating process. This as-synthesized gel showed a uniform filamentous structure and exhibited the more excellent textural, rheological and thermal properties than those of the samples formed under acidic and alkaline conditions. These results are of great value in revealing the gelation mechanism of SPH.

Abstract:Native soy proteins have limited surface properties, but fortunately, it is expected that their foaming behaviors can be improved through the enzymatic cross-linking method. In this paper, by using the cross-linking method with microbial transglutaminase (mTG) as the catalyst, soy protein isolate (SPI), soy β-conglycinin (7S), and soy glycinin (11S) nanoparticles (NPs) were prepared in the molten state of the protein induced by dithiothreitol (DTT). We compared the structural characteristics of these nanoparticles, including particle sizes, morphologies, zeta potentials, surface hydrophobicities, changes of spatial structure, and cross-linking degrees. In addition, their foaming properties, including foamabilities, foam stabilities, and bubble diameters, were also tested. The results show that there are no significant differences in the foamabilities between the native soy proteins and their nanoparticles with similar initial foam heights. However, it is found that 11S NPs have enhanced foam stabilities with longer half-lives and smaller changes in bubble size, which is closely related to their high wettability and cross-linking degree. This study may provide references for the application of soy protein nanoparticles as new foam stabilizers to replace the traditional foaming agents in the food industry.
Analysis of the gel properties, microstructural characteristics, and intermolecular forces of soybean protein isolate gel induced by transglutaminase

Abstract:Soybean protein isolate (SPI) is a high-quality plant protein that is primarily used to process various soybean products coagulated by transglutaminase (TGase). In this study, the degree of hydrolysis (DH), sulfhydryl content (SH), surface hydrophobicity (H0 ), secondary structural constitution, and microstructure of TGase-treated soybean protein (SPI, 7S, and 11S) were determined, as well as the effects of NaCl, urea, and SDS on the properties and intermolecular forces of SPI gel were analyzed. The results show that the H0 and SH content of SPI, 7S, and 11S decreased significantly with TGase treatment time (p < .05), while the DH gradually increased and reached its highest value (3.72%, 7.41%, and 1.27%, respectively) at 30 min. As the concentration of these two secondary structures exhibited an inverse relationship, the degradation of β-turns resulted in the increase in β-sheets. The microstructures of SPI and 11S gels were similar, being denser and more ordered than 7S gel. The low concentration of NaCl solution (0.2 mol/L) enhanced gel properties and intermolecular forces, promoting the formation of SPI gel, whereas a high concentration (0.4-0.8 mol/L) had a significant inhibitory effect. Urea and SDS solutions substantially inhibited the formation of SPI gel, leading to significant decreases in the water holding capacity and hardness as well as a considerable increase in the coagulation time (p < .05). The results revealed that hydrogen bonds and hydrophobic interactions were the main intermolecular forces responsible for the gel formation. This study provides adequate technical support and a theoretical basis for soybean protein gel products.

Abstract:Abstract The limited enzymatic hydrolysis gelation method was investigated using peanut protein isolate (PPI) without any coagulators. A peanut protein gel could be formed by enzyme treatment with Alcalase at low temperature (50–70 °C). The influence of enzymatic hydrolysis on the rheological and physicochemical properties was investigated. Structural changes in the PPI were characterized by analyzing the subunits, chemical forces, surface hydrophobicity, fluorescence spectra, and circular dichroism (CD) spectra. The results revealed that enzymatic hydrolysis significantly affected the conarachin II protein of PPI, and had little influence on conarachin I and the basic subunits of arachin. Hydrophobic interaction was the main chemical force active in the peanut protein gel. An increase in the surface hydrophobicity coupled with red-shifts of the fluorescence spectra indicated that inner hydrophobic regions were exposed after hydrolysis, resulting in gel formation via hydrophobic interactions. The CD spectra showed that significant changes occurred in the secondary structure of PPI, where the ordered PPI structure formed a more open structure after enzymatic hydrolysis.

Abstract:Soy glycinin could self-assemble into nanoparticles, microgels or hollow microcapsules depending on the extrinsic conditions. This research compares ion- and/or pH-induced particle formation through simple heat treatment of soy glycinin (80 °C, 5 min and 600 r/min), and the difference of zeta potential, particle size, hydrophobicity and their relevance to particle morphology were investigated. The results showed that only a combination of monovalent cation (K+ or Na+) and monovalent anion (Cl−) promoted the formation of hollow microcapsules with coexistence of microgels (protein solution pH: 7.2), whereas a reduce in pH inhibited hollow microcapsule formation upon heating, either in the presence of electrolytes or not. Strategies for size reduction of microparticles were also studied, either by increasing the shear stress during heating or through ultrasound pre-treatment of soy glycinin. In general, native soy glycinin was favorable for the formation of hollow microcapsules than the partially denatured, and the hollow microcapsule formation was highly sensitive to the extrinsic conditions.

Abstract:This study investigated the impact of different preheat treatments on the emulsifying and gel textural properties of soy protein with varying 11S/7S ratios. A mixture of 7S and 11S globulins, obtained from defatted soybean meal, was prepared at different ratios. The mixed proteins were subjected to preheating (75 °C, 85 °C, and 95 °C for 5 min) or non-preheating, followed by spray drying or non-spray drying. The solubility of protein mixtures rich in the 7S fraction tended to decrease significantly after heating at 85 °C, while protein mixtures rich in the 11S fraction showed a significant decrease after heating at 95 °C. Surprisingly, the emulsion stability index (ESI) of protein mixtures rich in the 7S fraction significantly improved twofold during processing at 75 °C. This study revealed a negative correlation between the emulsifying ability of soy protein and the 11S/7S ratio. For protein mixtures rich in either the 7S or the 11S fractions, gelling proprieties as well as emulsion activity index (EAI) and ESI showed no significant changes after spray drying; however, surface hydrophobicity was significantly enhanced following heating at 85 °C post-spray drying treatment. These findings provide insights into the alterations in gelling and emulsifying properties during various heating processes, offering great potential for producing soy protein ingredients with enhanced emulsifying ability and gelling property. They also contribute to establishing a theoretical basis for the standardized production of soy protein isolate with specific functional characteristics.

Abstract:Soy protein isolate (SPI) is widely used in the food industry as a gelling agent due to its high water-holding capacity and gelation. As a key in elucidating the structure-function relationships of SPI, subunit composition can explain many functional properties of soy protein. In this study, to explore the influence of α and α′ subunits on the gel properties of soy protein, the gel strength and water holding capacity of the heat-induced gel with missing subunits were determined. Texture Profile Analysis (TPA) showed that gel strength from large to small was α′-lack > (α and α′)-lack > native > α-lack, and the lack of α′ subunit significantly enhanced (p < 0.05) the gel strength and water-holding capacity of the resultant gels. In the gel formation process, microstructure observation of the gels revealed that the α′-lacking gel had a more uniform and denser network structure with higher storage modulus, promoting more water molecules to be bound in the gel structure as immobilized water and exhibiting higher thermal stability. Subsequently, the lack of α′ subunit increased the zeta potential, surface hydrophobicity (H0), and fluorescence intensity of the gel. The deletion of the α′ subunit promoted the exposure of hydrophobic groups with more hydrophobic amino acids and promoted the aggregation of the subunits after heat treatment. These changes in the physicochemical properties of the gel affected their heat-induced gelation. In addition, gel solubility results signify that hydrophobic and disulfide bond interactions are the main forces in the gel formation process. Thus, the deletion of the α′ subunit will facilitate the development and application of soy protein products in food processing and improve their gel properties. This study will benefit the application of soy protein at the subunit level in traditional dairy products based on heated-induced gelation.

Physicochemical properties of soya bean protein gel prepared by microbial transglutaminase in the presence of okara

Abstract:The influences of okara on the gelling and secondary protein structure of soya protein isolate (SPI) were investigated. Okara was ultra‐refined by high‐pressure homogenisation. The resulting particle sizes and microscopic morphologies were determined using a laser particle size analyser and a confocal laser scanning microscope, respectively. After okara homogenisation, surface area average particle size (D32) decreased from 49.68 to 23.29 μm and volume average particle size (D43) decreased from 118.94 to 55.08 μm. Dynamic rheological measurements showed that the storage (G′) and loss (G′′) moduli of SPI gelled in the presence of okara were mostly higher than those of SPI gels alone and were increased as the okara amounts increased. Linear law analysis revealed that SPI–okara gels were weak gels and frequency independent. Fourier transform infrared spectroscopy showed that okara and transglutaminase influenced the prevalence of α‐helices, β‐turns, β‐sheet and random coils. Soy protein isolate and homogenized okara can form gels using TGase. Dynamic rheological measurements showed the gels are weak gels and frequency independent, and Fourier‐transform infrared spectroscopy showed that okara influenced protein secondary structures.

Abstract:The effects of glycosylation followed by limited hydrolysis on the structural and functional properties of soybean protein isolate (SPI) were investigated. SPI was glycosylated by transglutaminase (TGase) in the presence of oligochitosan to generate a glycosylated soybean protein isolate (GSPI) and then hydrolyzed using Alcalase to obtain hydrolysates with degree of hydrolysis (DH) of 1%, 2% and 4%. Structural properties were characterized using estimate of molecular weight distribution and circular dichroism (CD) spectra. Electrophoresis analysis revealed that GSPI contains peptide polymers, and its hydrolysates exhibit broad molecular weight distributions. CD analysis indicated more flexible structure for GSPI compared to SPI, and the following limited hydrolysis of GSPI induces even more flexible structure on its hydrolysates. GSPI showed significant increase in the apparent viscosity at the shear rate of 0.1–10/s. Emulsion stability of GSPI also increased by 33.9% than that of SPI. Hydrolysates from GSPI, especially for DH of 4%, showed significantly improved emulsifying activity index, increasing by 39.2% compared to SPI. The hydrolysates also showed notably enhanced antioxidant activities, namely, DPPH radical-scavenging ability and reducing power. These results revealed that TGase-induced glycosylation and further limited hydrolysis may have potential for creating food ingredients with improved functional properties.

Freeze-thaw stability of transglutaminase-induced soy protein-maltose emulsion gel: Focusing on morphology, texture properties, and rheological characteristics

Abstract:In this study, soy protein isolate (SPI) and maltose (M) were employed as materials for the synthesis of a covalent compound denoted as SPI-M. The emulsion gel was prepared by transglutaminase (TGase) as catalyst, and its freeze-thaw stability was investigated. The occurrence of Maillard reaction was substantiated through SDS-PAGE. The analysis of spectroscopy showed that the structure of the modified protein was more stretched, changed in the direction of freeze-thaw stability. After three freeze-thaw cycles (FTC), it was observed that the water holding capacity of SPI-M, SPI/M mixture (SPI+M) and SPI emulsion gels exhibited reductions of 8.49 %, 16.85 %, and 20.26 %, respectively. Moreover, the soluble protein content also diminished by 13.92 %, 23.43 %, and 35.31 %, respectively. In comparison to unmodified SPI, SPI-M exhibited increase in gel hardness by 160 %, while elasticity, viscosity, chewability, and cohesion demonstrated reductions of 17.7 %, 23.3 %, 33.3 %, and 6.76 %, respectively. Concurrently, the SPI-M emulsion gel exhibited the most rapid gel formation kinetics. After FTCs, the gel elastic modulus (G') and viscosity modulus (G″) of SPI-M emulsion were the largest. DSC analysis underscored the more compact structure and heightened thermal stability of the SPI-M emulsion gel. SEM demonstrated that the SPI-M emulsion gel suffered the least damage following FTCs.

Abstract:The gelation behaviour of two different pea protein isolates and one soy protein isolate were investigated with a focus on the role of the protein properties. Protein solubility was the lowest in pH 3 citrate-phosphate buffer (<10% w/w), increased in pH 7.4 phosphate-buffered saline (12–21% w/w), and was the highest in pH 7.6 MilliQ water (∼20–40% w/w). Heat-induced gelation conditions for the protein sources were sensitive to both the soluble and the insoluble fractions as obtained during extraction. At low protein concentrations (≤5% w/v), the proteins started to lose their viscoelastic behaviour and exhibited predominantly viscous properties. Fitting of the fractional Kelvin-Voigt model to the frequency sweeps showed an increase in the fractal gel strength with increasing protein concentration. Secondary structures of the soluble species showed mostly unordered proteins, suggesting that the proteins were denatured during the commercial extraction process although gelation has to date been suggested to be highly dependent on the denaturation of soluble proteins. Synchrotron Radiation Circular Dichroism measurements of the insoluble proteins showed a significant amount of ordered protein structures. SEM imaging of the gels also suggested a new gelation pathway in which insoluble proteins act as dispersed fillers within a continuous matrix of soluble proteins. This research elucidated the role of different protein fractions, globulins and albumins, and their secondary structure in the formation of a gel network and how this affects their viscoelastic behaviour.

Effects of heat treatments on the properties of soymilks and glucono- δ - Lactone induced tofu gels

Abstract:Optimal heat treatment of the soymilks is important to the production of tofu. In this study, soymilk with protein concentration of 40 mg/mL were heated at different temperatures for the fixed 50 s and were characterized by surface hydrophobicity, disulfide linked protein species determined by non-reducing SDS-PAGE and protein structural elements determined by the circular dichroism (CD). Tofu gels were prepared by acidifying the heated soymilks at 60 ℃ and 80 ℃ respectively and gelation time, gel mechanical properties as well as gel viscoelastic properties were determined by rheological analysis. The results showed that most soymilks' properties except surface hydrophobicity changed rapidly when heating temperature was higher than 80 ℃. Gelation time, storage modulus (G') at the end of acidifying and cooling processes as well as retardation time (λ) and recovery rate of tofu gels were affected by the heat treatments of soymilks. The distances between the standardized data describing heated soymilks and tofu gels respectively were calculated and compared. It was found that gelation time, G' and λ were most closely related to disulfide bond linked polymer, [CD]222 and surface hydrophobicity respectively. This study will provide useful information to the improvement of tofu processing.

Abstract:A microbial transglutaminase was used to cross-link soybean protein isolates in the presence of gelatin to prepare a cross-linked composite soybean protein-gelatin. Cross-linking conditions, such as total proteins, the ratio of soybean protein isolates to gelatin, and original pH, were fixed at 5% (w/v), 4:1 (w/w), and 7.5, respectively. The 4-hydroxyproline content in the modified product was measured and used as an index to select suitable transglutaminase addition, reaction temperature, and time by single factor experiments, which were found to be 20 U/g proteins, 40°C, and 2 h, respectively. Under the identified conditions, a modified product with 4-hydroxyproline content of about 36 mg/g proteins was prepared, and its functional properties were evaluated. SDS-PAGE analysis showed that several protein polymers existed in the modified product. Compared to the original soybean protein isolates or a cross-linked soybean protein isolates, the modified product had a better emulsifying stability and water holding capacity but a poor enzymatic digestibility in vitro, emulsifying activity and oil binding capacity; meanwhile, the dispersions formed by the modified product exhibited the highest apparent viscosity, storage modulus, and viscous modulus.

Effects of endogenous small molecular compounds on the rheological properties, texture and microstructure of soymilk coagulum: Removal of phytate using ultrafiltration

Abstract:This study aims to clarify the roles played by endogenous small molecular components in soymilk coagulation process and the properties of gels. Soymilk samples with decreasing levels of small molecules were prepared by ultrafiltration, to reduce the amount of phytate and salts. CaSO4-induced coagulation process was analyzed using rheological methods. Results showed that removal of free small molecules decreased the activation energy of protein coagulation, resulting in accelerated reaction and increased gel strength. However, too fast a reaction led to the drop in storage modulus (G'). Microscopic observation suggested that accelerated coagulation generated a coarse and non-uniform gel network with large pores. This network could not hold much water, leading to serious syneresis. Endogenous small molecules in soymilk were vital in the fine gel structure. Coagulation rate could be controlled by adjusting the amount of small molecules to obtain tofu products with the optimal texture.
Effects of Sequential Enzymolysis and Glycosylation on the Structural Properties and Antioxidant Activity of Soybean Protein Isolate

Abstract:The effects of limited hydrolysis following glycosylation with dextran on the structural properties and antioxidant activity of the soybean protein isolate (SPI) were investigated. Three SPI hydrolysate (SPIH) fractions, F30 (>30 kDa), F30-10 (10–30 kDa), and F10 (<10 kDa), were confirmed using gel penetration chromatography. The results demonstrated that the glycosylation of F30 was faster than that of F30-10 or F10. The enzymolysis caused the unfolding of the SPI to expose the internal hydrophobic cores, which was further promoted by the grafting of dextran, making the obtained conjugates have a loose spatial structure, strong molecular flexibility, and enhanced thermal stability. The grafting of dextran significantly enhanced the DPPH- or •OH-scavenging activity and the ferrous reducing power of the SPI or SPIH fractions with different change profiles due to their different molecular structures. The limited enzymolysis following glycosylation was proven to be a promising way to obtain SPI-based food ingredients with enhanced functionalities.

Abstract:Glycated conjugates were prepared by heating aqueous dispersions of soy protein isolate (SPI) and maltodextrin (MD) after ultrasonic treatment (138 W/cm2, 20 kHz) for 5, 15, and 25 min. The effect of the ultrasonic pretreatment on the structure, physicochemical characteristics, and acid-induced gelation properties of the resulting SPI–MD conjugates was investigated. The analyses of free amino groups, degree of graft, intermediates, and browning intensity confirmed the formation of SPI–MD conjugates. The ultrasonic treatment promoted the glycation reaction, which led to the increase in the β-type ordered secondary structure and less compact tertiary conformation of conjugates. The ultrasonic treatment for 5 min increased the isoelectric point, surface hydrophobicity (H0), free sulfhydryl (-SH) groups, and particle size of the conjugates. These changes in the structural and physicochemical properties of the conjugates affected their acid-induced gelation. Glucono-δ-lactone was used to form acid gels at 25 °C. Although the glycation with MD deteriorated the acid-induced gelation properties of SPI, the 5 min ultrasonic conjugates formed gels faster than the untreated conjugate, and their gels exhibited higher storage modulus (G′), gel strength, and water holding capacity (WHC), as well as denser microstructure, than the untreated conjugate gels. However, prolonged ultrasonic treatment (15 or 25 min) weakened the gelation properties of the conjugates by promoting the glycation reaction and reducing the H0. Thus, an optimum ultrasonic pretreatment can be used to modify the structure of soy protein glycation conjugates to control their gelation properties.

Transglutaminase-induced soybean protein isolate cold-set gels treated with combination of ultrasound and high pressure: Physicochemical properties and structural characterization

Abstract:Soybean protein isolate (SPI) was treated by the combined exposure to ultrasound and high pressure and then subjected to transglutaminase (TGase)-catalyzed cross-linking to prepare SPI cold-set gels. The effects of combined treatments on physicochemical and structural properties of TGase-induced SPI cold-set gels were investigated. The combination of ultrasound and high pressure promoted the covalent disulfide bonds and ε-(γ-glutaminyl) lysine isopeptide bonds as well as non-covalent hydrophobic interactions, which further improved the gelation properties of SPI compared to ultrasound or high pressure alone. In particular, the 480 W ultrasound followed by high pressure treatment of gels led to higher strength (120.53 g), water holding capacity (95.39 %), immobilized water (93.92 %), lightness (42.18), whiteness (51.03), and elasticity (G′ = 407 Pa), as well as more uniform and compact microstructure, thus resulting in the improved gel network structure. The combination of two treatments produced more flexible secondary structure, tighter tertiary conformation and higher denaturation degree of protein in the gels, leading to more stable gel structure. The structural modifications of SPI contributed to the improvement of its gelation properties. Therefore, the combined application of ultrasound and high pressure can be an effective method for improving the structure and properties of TGase-induced SPI cold-set gels.

Abstract:Crowded environments, commonly found in the food system, are utilized to enhance the properties of soybean proteins. Despite their widespread application, little information exists regarding the impact of crowded environments on the denaturation behaviors of soybean proteins. In this study, we investigated how crowding agents with varying molecular weights, functional groups, and topology affect the denaturation behavior of glycinin under crowded conditions. The results reveal that thermal stability in PEG crowded environments is mainly influenced by both preferential hydration and binding. The stabilization is primarily enthalpy-driven, with aggregation contributing additional entropic stabilization. Specifically, ethylene glycol and diethylene glycol exhibit temperature-dependent, bilateral effects on glycinin stability. At the denaturation temperature, hydrophobic interactions play a predominant role, decreasing glycinin's thermal stability. However, at a molecular weight of 200 g/mol, there is a delicate balance between destabilizing and stabilizing effects, leading to no significant change in thermal stability. With the addition of PEG 400, 1000, and 2000, besides preferential hydration, additional hard-core repulsions between glycinin molecules enhance thermal stability. Methylation modification experiments demonstrated that 2-methoxyethyl ether exerted a more pronounced denaturing effect. Additionally, the cyclization of PEG 1000 decreased its stabilizing effect.

Abstract:The effect of high-pressure treatment on structural and rheological properties of soybean protein dispersions was studied. A sequential high-pressure/thermal treatment was also analyzed. Dissimilar effects on soy protein isolate (SPI) and the enriched soybean protein fractions: β-conglycinin (βCEF) and glycinin (GEF) were observed. High pressure (600MPa) promoted βCEF gelation, but did not modify the rheological properties of GEF in spite of its complete denaturation. Pressure treatment also induced the establishment of hydrophobic interactions and disulfide bonds that allowed the formation of soluble high molecular mass aggregates from the different polypeptides of both β-conglycinin and glycinin. Protein strands formation was detected in matrix microstructure of HP-treated SPI and βCEF dispersions in accordance with their rheological behavior of weak gels. In the case of GEF modifications induced by HP in the microstructure (apparition of large granules) were not accompanied by rheological changes.Heating process after HP treatment induced protein gelation. A decrease in the temperature of onset of matrix formation of SPI and βCEF samples was observed. The magnitude of this effect was proportional to the intensity of HP treatment. Contrarily, HP provoked a delay in gelation process of GEF dispersions. During the thermal cycle, previous HP treatment diminished the ability of both soybean globulins to establish hydrophobic interactions on heating and hydrogen bonds on cooling, thus obtaining gels with small elastic modulus.
