Abstract:The neural dynamics generating sensory, motor, and cognitive functions are commonly understood through field theories for neural population activity. Classic neural field theories are derived from highly simplified models of individual neurons, while biological neurons are highly complex cells. Integrate-and-fire neuron models balance biophysical detail and analytical tractability. Here, we develop a statistical field theory for networks of integrate-and-fire neurons with stochastic spike emission. This reveals an exact mapping to a self-consistent renewal process and a new mean field theory for the activity in these networks. The mean field theory has a rate-dependent leak, approximating the spike-driven resets of the membrane voltage. This gives rise to bistability between quiescent and active states in homogenous and excitatory-inhibitory pulse-coupled networks. The field-theoretic framework also exposes fluctuation corrections to the mean field theory. We find that due to the spike reset, fluctuations suppress activity. We then examine the roles of spike resets and recurrent inhibition in stabilizing network activity. We calculate the phase diagram for inhibitory stabilization and find that an inhibition-stabilized regime occurs in wide regions of parameter space, consistent with experimental reports of inhibitory stabilization in diverse brain regions. Fluctuations narrow the region of inhibitory stabilization, consistent with their role in suppressing activity through spike resets.

Abstract:We analyze a large system of heterogeneous quadratic integrate-and-fire (QIF) neurons with time delayed, all-to-all synaptic coupling. The model is exactly reduced to a system of firing rate equations that is exploited to investigate the existence, stability and bifurcations of fully synchronous, partially synchronous, and incoherent states. In conjunction with this analysis we perform extensive numerical simulations of the original network of QIF neurons, and determine the relation between the macroscopic and microscopic states for partially synchronous states. The results are summarized in two phase diagrams, for homogeneous and heterogeneous populations, which are obtained analytically to a large extent. For excitatory coupling, the phase diagram is remarkably similar to that of the Kuramoto model with time delays, although here the stability boundaries extend to regions in parameter space where the neurons are not self-sustained oscillators. In contrast, the structure of the boundaries for inhibitory coupling is different, and already for homogeneous networks unveils the presence of various partially synchronized states not present in the Kuramoto model: Collective chaos, quasiperiodic partial synchronization (QPS), and a novel state which we call modulated-QPS (M-QPS). In the presence of heterogeneity partially synchronized states reminiscent to collective chaos, QPS and M-QPS persist. In addition, the presence of heterogeneity greatly amplifies the differences between the incoherence stability boundaries of excitation and inhibition. Finally, we compare our results with those of a traditional (Wilson Cowan-type) firing rate model with time delays. The oscillatory instabilities of the traditional firing rate model qualitatively agree with our results only for the case of inhibitory coupling with strong heterogeneity.

Analytically Tractable Studies of Traveling Waves of Activity in Integrate-and-fire Neural Networks

Abstract:In contrast to other large-scale network models for propagation of electrical activity in neural tissue that have no analytical solutions for their dynamics, we show that for a specific class of integrate and fire neural networks the acceleration depends quadratically on the instantaneous speed of the activity propagation. We use this property to analytically compute the network spike dynamics and to highlight the emergence of a natural time scale for the evolution of the traveling waves. These results allow us to examine other applications of this model such as the effect that a nonconductive gap of tissue has on further activity propagation. Furthermore we show that activity propagation also depends on local conditions for other more general connectivity functions, by converting the evolution equations for network dynamics into a low-dimensional system of ordinary differential equations. This approach greatly enhances our intuition into the mechanisms of the traveling waves evolution and significantly reduces the simulation time for this class of models.

Abstract:OBJECTIVES We have previously shown that dexamethasone increases serum leptin in fed but not in fasted human subjects. We hypothesized that insulin and/or glucose mediated the effect of food intake. The primary aim of this study was to determine whether the administration of a pulse of insulin with dexamethasone was sufficient to increase serum leptin in vivo in fasted human subjects. Whether the presence of transient hyperglycemia and the dose of insulin were important was tested as a secondary aim. METHODS Twenty-nine normal subjects were studied. In experiment 1 (meal-like), a pulse of insulin (0.03 U/kg s.c.) and of dexamethasone (2 mg i.v.) was given, and the blood glucose transiently elevated to 50 mg/dl above baseline for the first 2 h. In experiments 2 and 3 (dose-response), the effect of two doses of insulin (0.03 U/kg in experiment 2 and 0.06 U/kg in experiment 3) was tested in combination with dexamethasone, this time without transient hyperglycemia. Nine subjects were studied under fasting conditions, with or without dexamethasone, as a control experiment. RESULTS A meal-like transient hyperinsulinemia and hyperglycemia, with a pulse of dexamethasone, increased serum leptin levels from baseline by 54+/-21% at 9 h (P=0.038). In the absence of transient hyperglycemia, leptin increased significantly after doses of both insulin and dexamethasone. The effect of insulin was dose-dependent, with a larger increment of serum leptin at 9 h after the highest dose of insulin (75.2+/-15.7% vs 21.3+/-8.5%, P=0.013). Fasting, with or without dexamethasone, resulted in a significant 20% decrease in leptin from morning basal levels. Conversely, the administration of a pulse of insulin and glucose, in the absence of dexamethasone, prevented the drop in serum leptin observed during fasting, regardless of the insulin dose or the serum glucose elevation. CONCLUSIONS With the permissive effect of dexamethasone, a single pulse of insulin triggered a rise in serum leptin in humans, even in the absence of transient hyperglycemia. A single pulse of insulin with glucose can prevent the drop in serum leptin normally observed during fasting.

Abstract:Low-dimensional descriptions of neural network dynamics are an effective tool for bridging different scales of organization of brain structure and function. Recent advances in deriving mean-field descriptions for networks of coupled oscillators have sparked the development of a new generation of neural mass models. Of notable interest are mean-field descriptions of all-to-all coupled quadratic integrate-and-fire (QIF) neurons, which have already seen numerous extensions and applications. These extensions include different forms of short-term adaptation (STA) considered to play an important role in generating and sustaining dynamic regimes of interest in the brain. It is an open question, however, whether the incorporation of pre-synaptic forms of synaptic plasticity driven by single neuron activity would still permit the derivation of mean-field equations using the same method. Here, we discuss this problem using an established model of short-term synaptic plasticity at the single neuron level, for which we present two different approaches for the derivation of the mean-field equations. We compare these models with a recently proposed mean-field approximation that assumes stochastic spike timings. In general, the latter fails to accurately reproduce the macroscopic activity in networks of deterministic QIF neurons with distributed parameters. We show that the mean-field models we propose provide a more accurate description of the network dynamics, although they are mathematically more involved. Using bifurcation analysis, we find that QIF networks with pre-synaptic short-term plasticity can express regimes of periodic bursting activity as well as bi-stable regimes. Together, we provide novel insight into the macroscopic effects of short-term synaptic plasticity in spiking neural networks, as well as two different mean-field descriptions for future investigations of such networks.

Abstract:We consider a ring network of quadratic integrate-and-fire neurons with nonlocal synaptic and gap junction coupling. The corresponding neural field model supports solutions such as standing and travelling waves, and also lurching waves. We show that many of these solutions satisfy self-consistency equations which can be used to follow them as parameters are varied. We perform numerical bifurcation analysis of the neural field model, concentrating on the effects of varying gap junction coupling strength. Our methods are generally applicable to a wide variety of networks of quadratic integrate-and-fire neurons.

Abstract:We consider a large network of Izhikevich neurons. Each neuron has a quadratic integrate-and-fire type model with a recovery variable modelling spike frequency adaptation (SFA). We introduce a biologically motivated synaptic current expression and a delay in the synaptic transmission. Following the Ott-Antonsen theory, we reduce the network model to a mean-field system of delayed differential equations. Numerical bifurcation analysis allows us to locate higher-codimension bifurcations and to identify the regions in the parameter space where the network exhibits changes in the macroscopic dynamics, including transitions between states where the individual neurons exhibit asynchronous tonic firing and different types of synchronous bursting. We investigate the impact of the heterogeneity of the quenched input current, the SFA mechanism and the synaptic delay on macroscopic dynamics. In the limit that the heterogeneity goes to zero, our perturbation and bifurcation analysis shows that the behaviour of the mean-field model remains consistent, although this limit breaks an assumption of the model reduction. For a single population of neurons with SFA, the synaptic delay has little effect on the generation of COs for weak coupling, but favours their emergence beyond that, and even induces new macroscopic dynamics. In particular, Torus bifurcations may occur, and these are a crucial mechanism for the emergence of population bursting with two nested frequencies. We discuss how these solutions may relate to cross-frequency coupling which is potentially relevant for understanding healthy and pathological brain functions.

Abstract:We investigate numerically the collective dynamical behavior of pulse-coupled non-leaky integrate-and-fire-neurons that are arranged on a two-dimensional small-world network. To ensure ongoing activity, we impose a probability for spontaneous firing for each neuron. We study network dynamics evolving from different sets of initial conditions in dependence on coupling strength and rewiring probability. Beside a homogeneous equilibrium state for low coupling strength, we observe different local patterns including cyclic waves, spiral waves, and turbulent-like patterns, which -- depending on network parameters -- interfere with the global collective firing of the neurons. We attribute the various network dynamics to distinct regimes in the parameter space. For the same network parameters different network dynamics can be observed depending on the set of initial conditions only. Such a multistable behavior and the interplay between local pattern formation and global collective firing may be attributable to the spatiotemporal dynamics of biological networks.

Scale Free Avalanches in Excitatory-Inhibitory Populations of Spiking Neurons with Conductance Based Synaptic Currents

Abstract:We investigate spontaneous critical dynamics of excitatory and inhibitory (EI) sparsely connected populations of spiking leaky integrate-and-fire neurons with conductance-based synapses. We use a bottom-up approach to derive a single neuron gain function and a linear Poisson neuron approximation which we use to study mean-field dynamics of the EI population and its bifurcations. In the low firing rate regime, the quiescent state loses stability due to saddle-node or Hopf bifurcations. In particular, at the Bogdanov-Takens (BT) bifurcation point which is the intersection of the Hopf bifurcation and the saddle-node bifurcation lines of the 2D dynamical system, the network shows avalanche dynamics with power-law avalanche size and duration distributions. This matches the characteristics of low firing spontaneous activity in the cortex. By linearizing gain functions and excitatory and inhibitory nullclines, we can approximate the location of the BT bifurcation point. This point in the control parameter phase space corresponds to the internal balance of excitation and inhibition and a slight excess of external excitatory input to the excitatory population. Due to the tight balance of average excitation and inhibition currents, the firing of the individual cells is fluctuation-driven. Around the BT point, the spiking of neurons is a Poisson process and the population average membrane potential of neurons is approximately at the middle of the operating interval $[V_{Rest}, V_{th}]$. Moreover, the EI network is close to both oscillatory and active-inactive phase transition regimes.

Abstract:The dynamic features of interspike interval sequences and structures of spatiotemporal patterns of firing in a coupled noisy neural network are investigated. The system displays complex dynamics under periodic external stimuli. The dynamics is modulated by a periodic impulse-like synaptic current which relates to a global coupling interaction between neurons. The firings of the stimulated neurons are phase-locked to this current. In addition, the interspike interval histograms are studied for the case of frozen noise which does not change its value within a time interval once it has been distributed onto the network. It is found that the peaks in these histograms are located at integer multiples of the period of the external stimulus, and the heights of these peaks decay exponentially, which corresponds to the experimental results.

Abstract:We study the mean-field limit of a model of biological neuron networks based on the so-called stochastic integrate-and-fire (IF) dynamics. Our approach allows to derive a continuous limit for the macroscopic behavior of the system, the 1-particle distribution, for a large number of neurons with no structural assumptions on the connection map outside of a generalized mean-field scaling. We propose a novel notion of observables that naturally extends the notion of marginals to systems with non-identical or non-exchangeable agents. Our new observables satisfy a complex approximate hierarchy, essentially a tree-indexed extension of the classical BBGKY hierarchy. We are able to pass to the limit in this hierarchy as the number of neurons increases through novel quantitative stability estimates in some adapted weak norm. While we require non-vanishing diffusion, this approach notably addresses the challenges of sparse interacting graphs/matrices and singular interactions from Poisson jumps, and requires no additional regularity on the initial distribution.

Abstract:In the mean field integrate-and-fire model, the dynamics of a typical neuron within a large network is modeled as a diffusion-jump stochastic process whose jump takes place once the voltage reaches a threshold. In this work, the main goal is to establish the convergence relationship between the regularized process and the original one where in the regularized process, the jump mechanism is replaced by a Poisson dynamic, and jump intensity within the classically forbidden domain goes to infinity as the regularization parameter vanishes. On the macroscopic level, the Fokker-Planck equation for the process with random discharges (i.e. Poisson jumps) are defined on the whole space, while the equation for the limit process is on the half space. However, with the iteration scheme, the difficulty due to the domain differences has been greatly mitigated and the convergence for the stochastic process and the firing rates can be established. Moreover, we find a polynomial-order convergence for the distribution by a re-normalization argument in probability theory. Finally, by numerical experiments, we quantitatively explore the rate and the asymptotic behavior of the convergence for both linear and nonlinear models.
Desynchronization of large-scale neural networks by stabilizing unknown unstable incoherent equilibrium states

Abstract:In large-scale neural networks, coherent limit cycle oscillations usually coexist with unstable incoherent equilibrium states, which are not observed experimentally. We implement a first-order dynamic controller to stabilize unknown equilibrium states and suppress coherent oscillations. The stabilization of incoherent equilibria associated with unstable focus and saddle is considered. The algorithm is demonstrated for networks composed of quadratic integrate-and-fire (QIF) neurons and Hindmarsh-Rose neurons. The microscopic equations of an infinitely large QIF neural network can be reduced to an exact low-dimensional system of mean-field equations, which makes it possible to study the control problem analytically.

Abstract:We analyze a large ensemble of Quadratic Integrate-and-Fire (QIF) neurons with heterogeneous input currents and adaptation variables. For a particular class of adaptation, which we call quadratic spike-frequency adaptation (QSFA), the high-dimensional system admits an exact reduction to a low-dimensional system of ordinary differential equations for three mean-field variables: the population's firing rate, the mean membrane potential, and a mean adaptation variable. These low-dimensional firing rate equations (FRE) reveal an important generic feature of heterogeneous networks with spike frequency adaptation: Both the center and the width of the distribution of the neurons' firing frequencies are reduced, and this largely favors the emergence of collective synchronization in the network. These results are confirmed by the bifurcation analysis of the FRE, which faithfully describe the collective dynamics of the spiking neuron network, including collective oscillations, bursting, and macroscopic chaos.

Abstract:For infinitely large sparse networks of spiking neurons mean field theory shows that a balanced state of highly irregular activity arises under various conditions. Here we analytically investigate the microscopic irregular dynamics in finite networks of arbitrary connectivity, keeping track of all individual spike times. For delayed, purely inhibitory interactions we demonstrate that the irregular dynamics is not chaotic but rather stable and convergent towards periodic orbits. Moreover, every generic periodic orbit of these dynamical systems is stable. These results highlight that chaotic and stable dynamics are equally capable of generating irregular activity.

Abstract:Contemporary modeling approaches to the dynamics of neural networks include two important classes of models: biologically grounded spiking neuron models and functionally inspired rate-based units. We present a unified simulation framework that supports the combination of the two for multi-scale modeling, enables the quantitative validation of mean-field approaches by spiking network simulations, and provides an increase in reliability by usage of the same simulation code and the same network model specifications for both model classes. While most spiking simulations rely on the communication of discrete events, rate models require time-continuous interactions between neurons. Exploiting the conceptual similarity to the inclusion of gap junctions in spiking network simulations, we arrive at a reference implementation of instantaneous and delayed interactions between rate-based models in a spiking network simulator. The separation of rate dynamics from the general connection and communication infrastructure ensures flexibility of the framework. In addition to the standard implementation we present an iterative approach based on waveform-relaxation techniques to reduce communication and increase performance for large-scale simulations of rate-based models with instantaneous interactions. Finally we demonstrate the broad applicability of the framework by considering various examples from the literature, ranging from random networks to neural-field models. The study provides the prerequisite for interactions between rate-based and spiking models in a joint simulation.

Abstract:The dynamics of synaptic interactions within spiking neuron networks play a fundamental role in shaping emergent collective behavior. This paper studies a finite-size network of quadratic integrate-and-fire neurons interconnected via a general synaptic function that accounts for synaptic dynamics and time delays. Through asymptotic analysis, we transform this integrate-and-fire network into the Kuramoto-Sakaguchi model, whose parameters are explicitly expressed via synaptic function characteristics. This reduction yields analytical conditions on synaptic activation rates and time delays determining whether the synaptic coupling is attractive or repulsive. Our analysis reveals alternating stability regions for synchronous and partially synchronous firing, dependent on slow synaptic activation and time delay. We also demonstrate that the reduced microscopic model predicts the emergence of synchronization, weakly stable cyclops states, and non-stationary regimes remarkably well in the original integrate-and-fire network and its theta neuron counterpart. Our reduction approach promises to open the door to rigorous analysis of rhythmogenesis in networks with synaptic adaptation and plasticity.

The distinct timescales of synaptic plasticity and neural activity dynamics play an important role in the brain's learning and memory systems. Activity-dependent plasticity reshapes neural circuit architecture, determining spontaneous and stimulus-encoding spatiotemporal patterns of neural activity. Neural activity bumps maintain short term memories of continuous parameter values, emerging in spatially organized models with short-range excitation and long-range inhibition. Previously, we demonstrated nonlinear Langevin equations derived using an interface method which accurately describe the dynamics of bumps in continuum neural fields with separate excitatory/inhibitory populations. Here we extend this analysis to incorporate effects of short term plasticity that dynamically modifies connectivity described by an integral kernel. Linear stability analysis adapted to these piecewise smooth models with Heaviside firing rates further indicates how plasticity shapes the bumps' local dynamics. Facilitation (depression), which strengthens (weakens) synaptic connectivity originating from active neurons, tends to increase (decrease) stability of bumps when acting on excitatory synapses. The relationship is inverted when plasticity acts on inhibitory synapses. Multiscale approximations of the stochastic dynamics of bumps perturbed by weak noise reveal that the plasticity variables evolve to slowly diffusing and blurred versions of their stationary profiles. Nonlinear Langevin equations associated with bump positions or interfaces coupled to slowly evolving projections of plasticity variables accurately describe how these smoothed synaptic efficacy profiles can tether or repel wandering bumps.
