Abstract:The neural dynamics generating sensory, motor, and cognitive functions are commonly understood through field theories for neural population activity. Classic neural field theories are derived from highly simplified models of individual neurons, while biological neurons are highly complex cells. Integrate-and-fire neuron models balance biophysical detail and analytical tractability. Here, we develop a statistical field theory for networks of integrate-and-fire neurons with stochastic spike emission. This reveals an exact mapping to a self-consistent renewal process and a new mean field theory for the activity in these networks. The mean field theory has a rate-dependent leak, approximating the spike-driven resets of the membrane voltage. This gives rise to bistability between quiescent and active states in homogenous and excitatory-inhibitory pulse-coupled networks. The field-theoretic framework also exposes fluctuation corrections to the mean field theory. We find that due to the spike reset, fluctuations suppress activity. We then examine the roles of spike resets and recurrent inhibition in stabilizing network activity. We calculate the phase diagram for inhibitory stabilization and find that an inhibition-stabilized regime occurs in wide regions of parameter space, consistent with experimental reports of inhibitory stabilization in diverse brain regions. Fluctuations narrow the region of inhibitory stabilization, consistent with their role in suppressing activity through spike resets.

Abstract:Low-dimensional descriptions of neural network dynamics are an effective tool for bridging different scales of organization of brain structure and function. Recent advances in deriving mean-field descriptions for networks of coupled oscillators have sparked the development of a new generation of neural mass models. Of notable interest are mean-field descriptions of all-to-all coupled quadratic integrate-and-fire (QIF) neurons, which have already seen numerous extensions and applications. These extensions include different forms of short-term adaptation (STA) considered to play an important role in generating and sustaining dynamic regimes of interest in the brain. It is an open question, however, whether the incorporation of pre-synaptic forms of synaptic plasticity driven by single neuron activity would still permit the derivation of mean-field equations using the same method. Here, we discuss this problem using an established model of short-term synaptic plasticity at the single neuron level, for which we present two different approaches for the derivation of the mean-field equations. We compare these models with a recently proposed mean-field approximation that assumes stochastic spike timings. In general, the latter fails to accurately reproduce the macroscopic activity in networks of deterministic QIF neurons with distributed parameters. We show that the mean-field models we propose provide a more accurate description of the network dynamics, although they are mathematically more involved. Using bifurcation analysis, we find that QIF networks with pre-synaptic short-term plasticity can express regimes of periodic bursting activity as well as bi-stable regimes. Together, we provide novel insight into the macroscopic effects of short-term synaptic plasticity in spiking neural networks, as well as two different mean-field descriptions for future investigations of such networks.

Non-exchangeable networks of integrate-and-fire neurons: spatially-extended mean-field limit of the empirical measure

Abstract:The dynamics of exchangeable or spatially-structured networks of $N$ interacting stochastic neurons can be described by deterministic population equations in the mean-field limit $N\to\infty$, when synaptic weights scale as $O(1/N)$. This asymptotic behavior has been proven in several works but a general question has remained unanswered: does the $O(1/N)$ scaling of synaptic weights, by itself, suffice to guarantee the convergence of network dynamics to a deterministic population equation, even when networks are not assumed to be exchangeable or spatially structured? In this work, we consider networks of stochastic integrate-and-fire neurons with arbitrary synaptic weights satisfying only a $O(1/N)$ scaling condition. Borrowing results from the theory of dense graph limits (graphons), we prove that, as $N\to\infty$, and up to the extraction of a subsequence, the empirical measure of the neurons' membrane potentials converges to the solution of a spatially-extended mean-field partial differential equation (PDE). Our proof requires analytical techniques that go beyond standard propagation of chaos methods. In particular, we introduce a weak metric that depends on the dense graph limit kernel and we show how the weak convergence of the initial data can be obtained by propagating the regularity of the limit kernel along the dual-backward equation associated with the spatially-extended mean-field PDE. Overall, this result invites us to re-interpret spatially-extended population equations as universal mean-field limits of networks of neurons with $O(1/N)$ synaptic weight scaling.

Abstract:Abstract Coarse-graining microscopic models of biological neural networks to obtain mesoscopic models of neural activities is an essential step towards multi-scale models of the brain. Here, we extend a recent theory for mesoscopic population dynamics with static synapses to the case of dynamic synapses exhibiting short-term plasticity (STP). The extended theory offers an approximate mean-field dynamics for the synaptic input currents arising from populations of spiking neurons and synapses undergoing Tsodyks–Markram STP. The approximate mean-field dynamics accounts for both finite number of synapses and correlation between the two synaptic variables of the model (utilization and available resources) and its numerical implementation is simple. Comparisons with Monte Carlo simulations of the microscopic model show that in both feedforward and recurrent networks, the mesoscopic mean-field model accurately reproduces the first- and second-order statistics of the total synaptic input into a postsynaptic neuron and accounts for stochastic switches between Up and Down states and for population spikes. The extended mesoscopic population theory of spiking neural networks with STP may be useful for a systematic reduction of detailed biophysical models of cortical microcircuits to numerically efficient and mathematically tractable mean-field models.

Abstract:This paper studies a stochastic neural field model that is extended from our previous paper [14]. The neural field model consists of many heterogeneous local populations of neurons. Rigorous results on the stochastic stability are proved, which further imply the well-definedness of quantities including mean firing rate and spike count correlation. Then we devote to address two main topics: the comparison with mean-field approximations and the spatial correlation of spike count. We showed that partial synchronization of spiking activities is a main cause for discrepancies of mean-field approximations. Furthermore, the spike count correlation between local populations are studied. We find that the spike count correlation decays quickly with the distance between corresponding local populations. Some mathematical justifications of the mechanism of this phenomenon is also provided.
Finite size effects in the correlation structure of stochastic neural networks: analysis of different connectivity matrices and failure of the mean-field theory

Abstract:We quantify the finite size effects in a stochastic network made up of rate neurons, for several kinds of recurrent connectivity matrices. This analysis is performed by means of a perturbative expansion of the neural equations, where the perturbative parameters are the intensities of the sources of randomness in the system. In detail, these parameters are the variances of the background or input noise, of the initial conditions and of the distribution of the synaptic weights. The technique developed in this article can be used to study systems which are invariant under the exchange of the neural indices and it allows us to quantify the correlation structure of the network, in terms of pairwise and higher order correlations between the neurons. We also determine the relation between the correlation and the external input of the network, showing that strong signals coming from the environment reduce significantly the amount of correlation between the neurons. Moreover we prove that in general the phenomenon of propagation of chaos does not occur, even in the thermodynamic limit, due to the correlation structure of the 3 sources of randomness considered in the model. Furthermore, we show that the propagation of chaos does not depend only on the number of neurons in the network, but also and mainly on the number of incoming connections per neuron. To conclude, we prove that for special values of the parameters of the system the neurons become perfectly correlated, a phenomenon that we have called stochastic synchronization. These discoveries clearly prevent the use of the mean-field theory in the description of the neural network.

Mean-field approximations for coupled populations of generalized linear model spiking neurons with Markov refractoriness

Abstract:There has recently been a great deal of interest in inferring network connectivity from the spike trains in populations of neurons. One class of useful models that can be fit easily to spiking data is based on generalized linear point process models from statistics. Once the parameters for these models are fit, the analyst is left with a nonlinear spiking network model with delays, which in general may be very difficult to understand analytically. Here we develop mean-field methods for approximating the stimulus-driven firing rates (in both the time-varying and steady-state cases), auto- and cross-correlations, and stimulus-dependent filtering properties of these networks. These approximations are valid when the contributions of individual network coupling terms are small and, hence, the total input to a neuron is approximately gaussian. These approximations lead to deterministic ordinary differential equations that are much easier to solve and analyze than direct Monte Carlo simulation of the network activity. These approximations also provide an analytical way to evaluate the linear input-output filter of neurons and how the filters are modulated by network interactions and some stimulus feature. Finally, in the case of strong refractory effects, the mean-field approximations in the generalized linear model become inaccurate; therefore, we introduce a model that captures strong refractoriness, retains all of the easy fitting properties of the standard generalized linear model, and leads to much more accurate approximations of mean firing rates and cross-correlations that retain fine temporal behaviors.

Abstract:Dynamics of an ensemble of $N$-unit FitzHugh-Nagumo (FN) neurons subject to white noises has been studied by using a semi-analytical dynamical mean-field (DMF) theory in which the original $2 N$-dimensional {\it stochastic} differential equations are replaced by 8-dimensional {\it deterministic} differential equations expressed in terms of moments of local and global variables. Our DMF theory, which assumes weak noises and the Gaussian distribution of state variables, goes beyond weak couplings among constituent neurons. By using the expression for the firing probability due to an applied single spike, we have discussed effects of noises, synaptic couplings and the size of the ensemble on the spike timing precision, which is shown to be improved by increasing the size of the neuron ensemble, even when there are no couplings among neurons. When the coupling is introduced, neurons in ensembles respond to an input spike with a partial synchronization. DMF theory is extended to a large cluster which can be divided into multiple sub-clusters according to their functions. A model calculation has shown that when the noise intensity is moderate, the spike propagation with a fairly precise timing is possible among noisy sub-clusters with feed-forward couplings, as in the synfire chain. Results calculated by our DMF theory are nicely compared to those obtained by direct simulations. A comparison of DMF theory with the conventional moment method is also discussed.

Abstract:Finite-sized populations of spiking elements are fundamental to brain function but also are used in many areas of physics. Here we present a theory of the dynamics of finite-sized populations of spiking units, based on a quasirenewal description of neurons with adaptation. We derive an integral equation with colored noise that governs the stochastic dynamics of the population activity in response to time-dependent stimulation and calculate the spectral density in the asynchronous state. We show that systems of coupled populations with adaptation can generate a frequency band in which sensory information is preferentially encoded. The theory is applicable to fully as well as randomly connected networks and to leaky integrate-and-fire as well as to generalized spiking neurons with adaptation on multiple time scales.

Abstract:The paper deals with non-linear Poisson neuron network models with bounded memory dynamics, that can include both Hebbian learning mechanisms and refractory periods. The state of a network is described by the times elapsed since its neurons fired within the post-synaptic transfer kernel memory span, and the current strengths of synaptic connections, the state spaces of our models being hierarchies of finite-dimensional components. We establish ergodicity of the stochastic processes describing the behaviour of the networks and prove the existence of continuously differentiable stationary distribution densities (with respect to the Lebesgue measures of corresponding dimensionality) on the components of the state space and find upper bounds for them. For the density components, we derive a system of differential equations that can be solved in a few simplest cases only. Approaches to approximate computation of the stationary density are discussed. One is to reduce the dimensionality of the problem by modifying the network so that each neuron cannot fire if the number of spikes it emitted within the post-synaptic transfer kernel memory span reaches a given threshold. We show that the stationary distribution of this `truncated' network converges to that of the unrestricted one as the threshold increases, and that the convergence is at a super-exponential rate. A complementary approach uses discrete Markov chain approximations to the network process. We derive linear systems for the stationary distributions of these Markov chains and prove that these distributions converge weakly to the stationary laws for the original processes.

Abstract:Populations of spiking neuron models have densities of their microscopic variables (e.g., single-cell membrane potentials) whose evolution fully capture the collective dynamics of biological networks, even outside equilibrium. Despite its general applicability, the Fokker-Planck equation governing such evolution is mainly studied within the borders of the linear response theory, although alternative spectral expansion approaches offer some advantages in the study of the out-of-equilibrium dynamics. This is mainly due to the difficulty in computing the state-dependent coefficients of the expanded system of differential equations. Here, we address this issue by deriving analytic expressions for such coefficients by pairing perturbative solutions of the Fokker-Planck approach with their counterparts from the spectral expansion. A tight relationship emerges between several of these coefficients and the Laplace transform of the interspike interval density (i.e., the distribution of first-passage times). "Coefficients" like the current-to-rate gain function, the eigenvalues of the Fokker-Planck operator and its eigenfunctions at the boundaries are derived without resorting to integral expressions. Abstract:Neural population equations such as neural mass or field models are widely used to study brain activity on a large scale. However, the relation of these models to the properties of single neurons is unclear. Here we derive an equation for several interacting populations at the mesoscopic scale starting from a microscopic model of randomly connected generalized integrate-and-fire neuron models. Each population consists of 50-2000 neurons of the same type but different populations account for different neuron types. The stochastic population equations that we find reveal how spike-history effects in single-neuron dynamics such as refractoriness and adaptation interact with finite-size fluctuations on the population level. Efficient integration of the stochastic mesoscopic equations reproduces the statistical behavior of the population activities obtained from microscopic simulations of a full spiking neural network model. The theory describes nonlinear emergent dynamics such as finite-size-induced stochastic transitions in multistable networks and synchronization in balanced networks of excitatory and inhibitory neurons. The mesoscopic equations are employed to rapidly integrate a model of a cortical microcircuit consisting of eight neuron types, which allows us to predict spontaneous population activities as well as evoked responses to thalamic input. Our theory establishes a general framework for modeling finite-size neural population dynamics based on single cell and synapse parameters and offers an efficient approach to analyzing cortical circuits and computations.

Elucidating the neurophysiological mechanisms underlying neural pattern formation remains an outstanding challenge in Computational Neuroscience. In this paper, we address the issue of understanding the emergence of neural patterns by considering a network of renewal neurons, a well-established class of spiking cells. Taking the thermodynamics limit, the network's dynamics can be accurately represented by a partial differential equation coupled with a nonlocal differential equation. The stationary state of the nonlocal system is determined, and a perturbation analysis is performed to analytically characterize the conditions for the occurrence of Turing instabilities. Considering neural network parameters, such as the synaptic coupling and the external drive, we numerically obtain the bifurcation line that separates the asynchronous regime from the emergence of patterns. Our theoretical findings provide a new and insightful perspective on the emergence of Turing patterns in spiking neural networks. In the long term, our formalism will enable the study of neural patterns while maintaining the connections between microscopic cellular properties, network coupling, and the emergence of Turing instabilities.

Abstract:In this study, we consider limit theorems for microscopic stochastic models of neural fields. We show that the Wilson–Cowan equation can be obtained as the limit in uniform convergence on compacts in probability for a sequence of microscopic models when the number of neuron populations distributed in space and the number of neurons per population tend to infinity. This result also allows to obtain limits for qualitatively different stochastic convergence concepts, e.g., convergence in the mean. Further, we present a central limit theorem for the martingale part of the microscopic models which, suitably re-scaled, converges to a centred Gaussian process with independent increments. These two results provide the basis for presenting the neural field Langevin equation, a stochastic differential equation taking values in a Hilbert space, which is the infinite-dimensional analogue of the chemical Langevin equation in the present setting. On a technical level, we apply recently developed law of large numbers and central limit theorems for piecewise deterministic processes taking values in Hilbert spaces to a master equation formulation of stochastic neuronal network models. Dynamic Mean-Field Theory for Continuous Random Networks

Abstract:This article studies the dynamics of the mean-field approximation of continuous random networks. These networks are stochastic integrodifferential equations driven by Gaussian noise. The kernels in the integral operators are realizations of generalized Gaussian random variables. The equation controls the time evolution of a macroscopic state interpreted as neural activity, which depends on position and time. The position is an element of a measurable space. Such a network corresponds to a statistical field theory (STF) given by a momenta-generating functional. Discrete versions of the mentioned networks appeared in spin glasses and as models of artificial neural networks (NNs). Each of these discrete networks corresponds to a lattice SFT, where the action contains a finite number of neurons and two scalar fields for each neuron. Recently, it has been proposed that these networks can be used as models for deep learning. In this application, the number of neurons is astronomical; consequently, continuous models are required. In this article, we develop mathematically rigorous, continuous versions of the mean-field theory approximation and the double-copy system that allow us to derive a condition for the criticality of continuous stochastic networks via the largest Lyapunov exponent. We use two basic architectures; in the first one, the space of neurons is the real line, and then the neurons are organized in one layer; in the second one, the space of neurons is the p-adic line, and then the neurons are organized in an infinite, fractal, tree-like structure. Networks with both types of architectures exhibit a type edge of the chaos organization.

The mean-field Limit of sparse networks of integrate and fire neurons

Abstract:We study the mean-field limit of a model of biological neuron networks based on the so-called stochastic integrate-and-fire (IF) dynamics. Our approach allows to derive a continuous limit for the macroscopic behavior of the system, the 1-particle distribution, for a large number of neurons with no structural assumptions on the connection map outside of a generalized mean-field scaling. We propose a novel notion of observables that naturally extends the notion of marginals to systems with non-identical or non-exchangeable agents. Our new observables satisfy a complex approximate hierarchy, essentially a tree-indexed extension of the classical BBGKY hierarchy. We are able to pass to the limit in this hierarchy as the number of neurons increases through novel quantitative stability estimates in some adapted weak norm. While we require non-vanishing diffusion, this approach notably addresses the challenges of sparse interacting graphs/matrices and singular interactions from Poisson jumps, and requires no additional regularity on the initial distribution.

Abstract:We continue the work of a series of previous studies of a mathematical model that describes the mean-field limit behavior of a homogeneous network of excitatory point spiking neurons. Contrary to other models, here noise is intrinsic to the neurons through a membrane-potential dependent spiking rate probability, that we assume to be given by a power law. This allows one to collapse several sources of neural noise into a single function, which aids for a more treatable mathematical and comptuational analysis. In particular, we give pseudo-analytical expressions for the invariant distributions of membrane potentials across the population and study their stability computationally. The neurons are assumed to be connected both by chemical and possibly electrical (gap junction) synapses and can also undergo a leakage of ions with the extracellular medium. The distributions are of compact support whenever leakage or gap junction rates are non-zero and an infinite discontinuity appears at the maximum potential in the population. This happens invariably for the leakage (with no gap junctions) and gap junction (without leakage) cases. However, these discontinuous distributions might only be stable for linear spiking rate functions. It was recently shown how the network can present highly synchronous states of global oscillations in its activity when leakage is not present and gap junctions are strong enough. Here we confirm how oscillations persist also when weak leakage is considered. Thus, these dendritic (rather than axonal) gap junctions could play a role in the high levels of neural synchrony necessary for the development of neural systems at young ages. The model, thus, presents a rich phenomenology and capability to reproduce several biological features of neural networks despite of its mathematical simplicity, hence presenting a powerful tool for high performance computing.

Abstract:Three international stakeholder meetings were organized by the Netherlands‐based “Kluyver Center for Genomics of Industrial Fermentation” with the objective to identify the future societal issues in the field of industrial biotechnology and to develop a coordinated strategy for public dialogue. The meetings resulted in five unanimous recommendations: (i) that science, industry and the European Commission in conjunction with other stakeholders create a comprehensive roadmap towards a bio‐based economy; (ii) that the European Commission initiate a series of round‐table meetings to further articulate the views, interests and responsibilities of the relevant stakeholders and to define policy; (iii) that the development of new innovative communication activities is stimulated to increase public engagement and to discuss the ways that we do or do not want technologies to shape our common future; (iv) that further social studies are undertaken on public attitudes and behaviors to the bio‐based economy and that novel methods are developed to assess public views of future technological developments; and (v) that the concept of sustainability is further operationalized and taken as a core value driving research and development and policy making.
Neural Fields and Noise-Induced Patterns in Neurons on Large Disordered Networks

Abstract:We study pattern formation in class of a large-dimensional neural networks posed on random graphs and subject to spatio-temporal stochastic forcing. Under generic conditions on coupling and nodal dynamics, we prove that the network admits a rigorous mean-field limit, resembling a Wilson-Cowan neural field equation. The state variables of the limiting systems are the mean and variance of neuronal activity. We select networks whose mean-field equations are tractable and we perform a bifurcation analysis using as control parameter the diffusivity strength of the afferent white noise on each neuron. We find conditions for Turing-like bifurcations in a system where the cortex is modelled as a ring, and we produce numerical evidence of noise-induced spiral waves in models with a two-dimensional cortex. We provide numerical evidence that solutions of the finite-size network converge weakly to solutions of the mean-field model.
